目的:研究健脾消癌方对大肠癌肝转移裸鼠模型肝组织中磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B (Akt)通路相关蛋白表达的影响,探讨其拮抗大肠癌肝转移的作用机制。方法:随机抽取10只BALB/C-nu裸鼠作为空白组,其余裸鼠建立人大肠癌细胞肝转移裸鼠模型,建模3 d后取存活裸鼠30只,随机分为模型组,健脾消癌方组(41.6 g · kg-1),人参皂苷Rg3组(5.2 mg · kg-1),各组裸鼠灌胃相应药物及生理盐水4周后脱颈处死,观察各组裸鼠肝转移情况,采用蛋白免疫印迹法(Western blot)测定肝组织第10号染色体同源缺失性磷酸酶和张力蛋白基因(PTEN),磷酸化第10号染色体同源缺失性磷酸酶和张力蛋白基因(p-PTEN),Akt,磷酸化蛋白激酶(p-Akt)蛋白表达情况。结果:健脾消癌方及人参皂苷Rg3组肝转移率明显低于模型组(P<0.05);与空白组比较,模型组中p-PTEN,p-Akt蛋白表达明显升高(P<0.05),与模型组比较,健脾消癌方组与人参皂苷Rg3组肝组织中p-PTEN,p-Akt蛋白表达明显降低(P<0.05);健脾消癌方组与人参皂苷Rg3组p-PTEN,p-Akt表达差异无统计学意义。与空白组比较,模型组PTEN,Akt蛋白表达明显降低(P<0.05);与模型组比较,健脾消癌方组与人参皂苷Rg3组PTEN,Akt蛋白表达量明显升高(P<0.05);健脾消癌方组与人参皂苷Rg3组PTEN,Akt蛋白表达差异无统计学意义。结论:健脾消癌方可拮抗大肠癌肝转移,其作用机制可能与升高PTEN蛋白表达,降低p-PTEN,p-Akt蛋白表达有关。
