      摘要:Objective:To establish UV method to determine polysaccharide in Fomitiporia robusta(P.Karst.) Fiasson.Method:The parameter of colorimetric method was optimiged through L9(34) orthogonal design with the index of glucose,by the use of phenyl hydrate-sulfuric acid,at the 488 nm.Result:The significant effects of phenyl hydrate-sulfuric acid volume on glucose content were discovered.The average of recovery was 96.6% and RSD 1.71% for glucose with in good linear relation in 3.223~12.89 μg·mL-1 with a correlation coefficient of 0.999 1.Conclution: The proposed method is sensitive,rapid and accurate.It can be used to assay polysaccharide with method indican.

      摘要:Objective:To analyst monosaccharide composition in astagalus neutral polysaccharides(ANP) by high performace liquid chromatography(HPLC).Method: Orthogonal design was used to optimize the hydrolytic condition of ANP by trifluoroacetic acid(TFA),the monosacchride composition in hydrolysate was assayed directly by HPLC-RID method or HPLC-DAD method after precolumn-derivation with 1-phenyl-3-methyl-5-pyrazolone(PMP) with six monosaccharides as control.Result: The optimum hydrolization procedure of ANP was configered with 1M TFA hydrolyzing 5 hours at 100 ℃.The results of two HPLC methods showed that the ANP consisted of glucose.Conclusion: The optimum hydrolization procedure and HPLC methods are simple and convenient and could be applied to analysis of the monosaccharide composition in ANP.

      摘要:Objective:To develop an HPLC method for the determination of geniposide and baicalin in Lanqin oral solution.Methods:Analysis was carried out at 40 ℃ on a Nova-Pak C18 column(3.9 mm×150 mm,4 μm)with methanol-0.1%TFA(25∶75)as the mobile phase,detecting at 280 nm.The results were calculated with the external standard method.Results:The average recovery was 100.8%,RSD=1.6%(n=9)for geniposide,the linear range was 0.50~2.5 μg,r=0.999 9 and the average recovery was 101.2%,RSD=2.0%(n=9)for baicalin,the linear range was 0.21~1.26 μg,r=0.999 4.Conclusion: It can be used for the quality control of Lanqin oral solution.

      摘要:Objective:To determine the contents of aloeemodin,rhein,emodin and chrysophanol in Qingyi Tablets by HPLC.Method: C18 column was used with mobile phase consisted of methanol-0.1%.Perchloric acid solution(75∶25),at a flow rate of 1.0 mL·min-1.The detecting wavelength was set at 254nm.Result: The linearity of this method was good in the ranges of (0.062 4~0.312)μg for aloeemodin(r=0.999 9);(.156~0.780 μg for rhein(r=0.999 8);(0.062 4~0.312)μg for emodin(r=0.999 2);(0.156~0.780) μg for chrysophanol(r=0.999 9).The recoveries of aloeemodin,rhein,emoclin and chrysophanol were 99.31%,99.65%,99.10% and 99.51%.RSD were 1.71%,0.56%,1.82% and 1.24%,respectively.Conclusion: The method is accurate,precise and is suitable for assaysment of aloeemodin,rhein,emodin and chrysophanol in Qingyi Tablets.

      摘要:Objective:To study the methods of identification and determination of the concent of effective constituents in Zhitejia Tablet.Method: Fructus sophorae,Fructus aurantii and Radix angelicae sinensis were identified by TLC.Rutin was assayied by HPLC.Result:The TLC spots were clear and the repeatable.The average recovery of Rutin was 100.14% with RSD 0.78%.Conclusion:The quality of Zhitejia Tablet can be controlled effectively by above methods.

      摘要:Objective:To investigate the optimum conditions for extracting the major active components in Fructus Aurantii Immaturus.Method: According to the contents of Hesperidin,Naringin and Synephrine,the use of extraction solvent of the other active components of Fructus Aurantii Immaturus in which the volatile oil were distilled was studied.And L9(3)4 orthogonal design was used to determine the optimum extraction conditions.Result: The content of hesperidin in the EtOH extraction was obviously higher than that in the water extraction,and the contents of naringin and synephrine had no significant difference.Conclusions:The optimum ethanol extraction conditions was 10 times amount of 60% alcohol,refluxed two times,1.0 hour each time.

      摘要:Objective:To determin the feasibility of the new technique to dispel Rhizoma Typhon's sting and hot irritation.Method:By the ultramicro-pulverzing technique different diameter of ultro-powder particles were used for the compared studies through pharmacologic experiments such as mouse acute toxicity test,rabbit conjunctiva stimulation and sedation tests,etc between the raw Rhizoma Typhon、the old and the new processed Rhizoma Typhon.Result:After the ultramicro-powder ing Rhizoma Typhon nearly has the same pharmacological effects as the raw and old processed Rhizoma Typhon,and its toxicity and irritation was less than old processed Rhizoma Typhon,while.Conclusion:The Rhizoma Typhon's sting and irritation was dispelled when with the sting rank of raw RT,old processed method and ultramicro-pulverying method processed by the new technique of ultramicro-pulverization.While pharmacological effects of Rhizoma Typhon can be still retained.

      摘要:Objective:To observe the effect of abnormal savda munziq on morphological structure of dominating organ cells in animal model of abnormal savda syndrome,to explore the essence of abnormal savda on morphology,to explore the working mechanism of abnormal savda munziq.Methods: Male ICR rats were fed with cold-natured diet under cool dry environment,and stimulated with electric shocks to establish animal model of abnormal savda syndrome in Uyghur medicine.Three other drug groups were given the same stimulative elements and further intervened with three different dosages(2.53,5.06,10.12 g·kg-1) of abnormal savda munziq during the whole procedure.In order to observe histological changes of these dominating organs by microscope,the brains,cardiac muscles and livers were prepared.Results: Microscopic examination of the model group showed swelling of hypothalamic cells,interstitial angiectasis in the hypothalamus.There was adipose cells infiltration and interstitial angiectasis on cardiac muscles. There were necrosis and inflammatory cells infiltration in liver tissues.However,the morphological changes of dominating organ cells in drug intervene groups improved significantly according to the concentration of drugs.Conclusion: Abnormal savda munziq can ptotect and recover the pathological damage on dominating organ cells in animal model of abnormal savda syndrome.

      摘要:Objective:To study anti-inflammatory effect of Paino powder and quantitative analysis of the composition.The Paino powder is composed of Fructus Aurantii Immaturi(FAI),Radix Paeoniae Alba(RPA) and Radix Platycodonis(RP).Methods: The animal model of acute inflammation was established in mice by acetic acid with infiltration increasing of capillary vessel.An orthogonal design was used to assign experimental groups of the combinations of components.The groups of aspirin and saline were selected as the controls.Quantitative analysis of the components and their pharmacodynamic interactions were analyzed by the optical density(OD) of infiltrated Evans Blue.Results: Paino powder and the different combinations of components showed obvious anti-inflammatory effect,and the predicted value of OD is 0.067~0.161.The best anti-inflammatory effect is the whole formulation of Paino powder,while comparing with the group of aspirin the difference was insignificant(P>0.05).The interaction of whole formulation of Paino powder showed antagonism,and the contribution to combined effect of each component: FAI>RPA>RP.Conclusion: Paino powder has significantly anti-inflammatory effect,and FAI shows the best effective.The interaction of combinations is antagonism in anti-inflammatory effect.

      摘要:Objective: To study the influence of Sini decoction at different doses and with different preparing methods on the contracting function of the isolated toad heart.Methods: The improved toad heart-intubation method were adopted,the isolated toad hearts were perfused Sini water decoction(SWD) or Sini water-decocted and alcohol-sinked liquid(SWDASL) at different doses,the contraction tension and the contraction extent of isolated toad heart were recorded.Results: The raw Fuzi SWD and SWDASL at the low doses had the excitation effect on isolated toad heart,while at high doses had the restraint effect on isolated toad heart;the fried Fuzi SWD and SWDASL at different doses had the restraint effect on isolated toad heart.The restraint effect on isolated toad heart was weakened,after Sini decoction was treated as alcohol-sinked liquid.Conclusion: The influence of Sini decoction on the isolated toad heart were different according to the alcohol-sinked treatment and the Fuzi with different processing.

      摘要:Objective:To identify pharmacokinetics of syringopicroside in rabbits.Methods:The plasma samples were collected after intravenous injection and syringopicroside was determined by HPLC after solid-phase extraction.The data were analyzed with software Multi97 and the pharmacokinetic parameters were calculated.Results:Two compartment model was shown after syringopicroside administration.The main pharmacokinetic parameters are presented respectively: T1/2(α)=2.655 min,T1/2(β)=16.54 min,AUC=272.32 μg·mL-1·min,CL=(0.037 1 mL·min-1),K12=(0.090 8 min-1),K21=0.124 min-1.Conclusion:The pharmacokinetics of syringopicroside showed a rapid distribution,the elimination was also fast,and the retention time was short.

      摘要:Objective: To study the effect of ligustri-astragali casein co-peptid(LACP)drug serum in inducing differentiation of Bel-7402 human hepatocarcinoma cell(HHC) and its influence of the telomerase activity.Methods: Rats were fed with LACP decoction to prepare the drug-serum of LACP,and the drug-serum was used for Bel-7402 HHC culture.Serum of normal rat,was taken as negative control and that of retinoic acid(RA) as the positive control.Cell proliferation was detected by MTT colorimetric assay,nucleocytoplasm ratio by image assay,α-fetoprotein(AFP) by RIA,alkaline phosphatase(ALP) activity and γ-glutamyl transpeptidase(γ-GT) by dynamic colorimetric assay and telomerase activity of Bel-7402 cell by PCR-ELISA.Results: Drug-serum of FACP could inhibit the proliferation of Bel-7402 cell,markedly reduce the nucleocytoplasm ratio and the secretion of AFP,decrease γ-GT activity,increase ALP activity and suppress the telomerase activity of Bel-7402 cell.Conclusion: LACP can induce the differentiation of Bel-7402 HHC,the main mechanism maybe throngh the suppression of telomerase activity.

      摘要:Objective: To investigate the effect of the different ratio of Angelica sinensis Injection(ASI) and Astragalus membranaceus Injection(AMI) on angiogenesis of the chick embryo chorioallantoic membrane(CAM).Methods: After fertilized eggs were incubated for 7 days at 37.8 ℃, the chorioallantoic membrane(CAM)models were made,and were respectively treated with NS(negative control),different ratio of ASI and AMI,and EGF(positive control).After incubated for another 3 days,the CAM samples were made,then counted the number of the first grade and second grade blood vessels(capillaries).Result: The proliferation of capillaries in the therapy groups was obviously increased compared with NS control(P<0.05),but the effect is lower than EGF control(P<0.05) excluding the group of ASI∶AMI=1∶5.Conclusion: The result showed that the combination of different ratio of ASI and AMI can obviously stimulate angiogenesis of the chick embryo CAM.And proliferation and migration of vascular endothelial cells can be the mechanism of angiogenesis.

      摘要:Objective:To explore the therapeutic effects and the mechanism of Ling-tong powder on dysmenorrhea.Methods: The mice model was established by giving Oxytocin and PGF2α.The number and occurrence rate of twisting reaction were observed,the effects of anti-inflammatory,analgesic,sedative,activating blood and eliminating stasis were also observed.Results: Ling-tong powder obviously reduced the twisting reaction of mice induced by oxytocin or PGF2α.It could alleviate the auricular edema in mice and lessen pains induced by hot plate.It could prolong sleeping time of mice and decrease the weight of the thrombus.Conclusion: Ling-tong Powder has marked therapeutic effects on dysmenorrhea,by anti-inflammatory,analgesic,sedative,activating blood and eliminating stasis.

      摘要:Objective:To study pharmacodynamic action of Ji-Ming-San(JMS) in animal model and acute toxicity in mice.The circadian variations of the dosing action and toxicity were approached as well,to certify the rationality about the experience of predecessors,i.e.medication of timing in administering JMS.Methods:Bliss method was used to evaluate medial lethal dose(LD50) and circadian variation on the acute mortality rate of JMS was observed with identical dosage in mice.The analgesic effects were observed by hot-plate test and body torsion test in mice.Nitrate reductase method was used to measure nitric oxide(NO) levels in blood serum and brain tissue in acetic acid caused-pain mice.The anti-inflammatory action was observed by auricle swelling test in mice and voixpedis swelling test in rats.Ultraviolet spectrophotometer method was employed to measure Prostaglandin E2(PGE2) level in voixpedis swelling rat model.Results:JMS produced marked time-dependent acute toxicity in mice with dosage of LD50) and remarkably analgesic effect on mice model with a diurnal rhythmicity.Furthermore,the results coincided with the circadian changes of decreased NO levels in blood serum and brain tissue.JMS obviously inhibited the ear edema in mice induced by dimethylbenzene and pedes intumesce in rats induced by egg white.The anti-inflammatory action of JMS showed circadian dependent change with dosing time. Furthermore,the rhythm was in line with the circadian change of PGE2 level reduced by JMS.Conclusion:JMS has obvious analgesia,anti-inflammatory effects,showing dosing time-dependent.Analgesic effect may be correlated with decreased NO levels and anti-inflammatory effect probably relates to reduced PGE2 level.

      摘要:Objective:To study the natures of different ratios of Coptis chinensis and Fructus Evodiae.Methods: The recent literatures on four natures of chinese materia,Coptis chinensis and Fructus Evodiae published were reviewed.Results: Current studies on different combinations of Coptis chinensis and Fructus Evodiae are mainly concentrated in the chemical composition of compatibility changes,pharmacological and clinical applications.The material basis for the efficacies in the molecular and cellular level and the compatibility mechanism have not been invastigated sufficiently.Conclusion: Four natures of Coptis chinensis and Fructus Evodiae need further studies.
