      摘要:Objective: To evaluate the diuretic effect of different processed products of Phytolaccae Radix and explore its mechanism of diuretic action. Method: A water load rat model was used,eighty SD rats were randomly divided into the blank group,model group,esculentoside A (EsA) group,raw product group and different processed products (vinegar broiled,vinegar boiled,boiled and steamed) group,10 rats in each group.At the same time,SD rats were randomly divided into the blank group,EsA group,raw product group and different processed products (vinegar broiled,vinegar boiled,boiled and steamed) group,10 rats in each group.The urine output was observed and relative mRNA expression of aquaporin-9(AQP9) of testicles,AQP2 and AQP4 of kidneys were determined. Result: In this respect of total urine output in 6 h,EsA,raw product group and different processed products of Phytolaccae Radix all showed a diuretic effect on water load rats.For normal rats,only vinegar-boiled product showed diuretic effect.PCR experiments showed that for water load rats,AQP2 mRNA expression were significantly decreased,AQP4 and AQP9 mRNA expression did not change significantly.For normal rats,AQP9 mRNA expression in vinegar-boiled group was significantly decreased,the other groups did not change significantly. Conclusion: Phytolaccae Radix for water load rats shows a diuretic effect,but it is unapparent for normal rats.Diuretic effect of Phytolaccae Radix may be related to the decrease of AQP2 and AQP9 mRNA expression.

      摘要:Objective: To evaluate in vitro transdermal absorption properties of iontophoresis fluid of Hanbi prescription for providing basis for its clinical application. Method: UV-Vis spectrophotometry was adopted to determine contents of alkaloids and glycosides with 413 nm and 232 nm as detection wavelength,respectively.Improved Franz diffusion cell was employed,permeation efficiency of different penetration enhancer combination and testing conditions of ion penetration tester on contents of alkaloids and glycosides in iontophoresis fluid were evaluated. Result: The optimized combination of permeation enhancers was 1.2 mL of azone,1.6 g of sodium dodecyl sulfate,0.4 mL of methyl pyrrolidone,0.4 mL of propylene glycol and 1.2 mL of oleic acid;under these conditions,the average cumulative osmotic quantity of alkaloids and glycosides from iontophoresis fluid of Hanbi prescription in 24 h were 15.84,1 379.82 μg·cm-2,their increased permeation times were 1.77-fold and 2.30-fold over that without permeation enhancer,respectively.When the voltage of ion penetration tester was 2.4 V,the penetration rates of alkaloids and glycosides in iontophoresis fluid of Hanbi prescription were 20.74% and 1.24% in 30 min,and 4.20-fold and 5.17-fold over that without the permeation enhancers and iontophoresis. Conclusion: Iontophoresis can significantly improve transdermal absorption of alkaloids and glycosides in iontophoresis fluid of Hanbi prescription across skin,and it will be benefit for curing Hanbi symptom.

      摘要:Objective: To establish an accurate HPLC method for the determination of 6 components (liquiritin,liquiritigenin,isoliquritigenin,licochalcone A,glycyrrhizic acid and glycyrrhetinic acid) in manure-prepared licorice.To compare content changes of triterpenes and flavonoids in manure-prepared licorice before and after being processed and evaluate its quality. Method: The analyses were performed on a Agilent Exlipse C18 column (4.6 mm×250 mm,5 μm) with mobile phase of acetonitrile-0.05% phosphoric acid aqueous solution for gradient elution at a flow rate of 1.0 mL·min-1,the detection wavelength was set at 237 nm and the column temperature was at 30℃.Quality of manure-prepared licorice was analyzed by cluster analysis,principal component analysis and G-test. Result: The contents of these 6 components in 12 batches of manure-prepared licorice samples were in the ranges of 0.04-0.23,0.25-0.66,0.17-0.37,0.33-3.42,1.54-15.21,0.03-0.95 mg·g-1,respectively;and the average contents of these 6 components in Glycyrrhizae Radix et Rhizoma were 1.64,0.32,0.17,0.50,22.46,0 mg·g-1,respectively.Glycyrrhizae Radix et Rhizoma and manure-prepared licorice had essential difference,there were some differences among samples prepared by different methods and 12 batches of commercial samples. Conclusion: The method is simple and reproducible.Quality of manure-prepared licorice is influenced by preparation process,raw materials and ammoniated environment.

      摘要:Objective: To establish the pattern discernment model with the use of near-infrared reflectance spectroscopy (NIRS), principal component analysis (PCA) and support vector machine (SVM) algorithms for rapid identification of Benzoinum, Succinum, Myrrha, Olibanum, Colophonium and Bambusae Concretio Silicea, all of which are the traditional Chinese medicines (TCMs) containing resins and other components. Method: According to morphological identification and conventional physical and chemical analysis on the above 6 kinds of samples collected form major national medicinal materials markets, a total of 55 batches of samples were verified as genuine medicinal herbs. These samples were smashed into uniform powders, and the NIR spectra of sample powder were scanned at 4 000-12 000 cm-1. The characteristic bands at 9 000-5 400, 5 000-4 000 cm-1 were used to establish models for pre-treatment by vector normalization (VN), first derivative (FD) and second derivative (SD) respectively, and in addition, PCA dimension reduction was also conducted. The best pre-treatment method was selected according to the three-dimensional scatter diagram of principal components, and the PCA dimension reduction data after best pre-treatment method were used to establish the pattern discernment models based on the SVM algorithm. The SVM model parameters c and g were optimized by using grid search method combined with 5-fold cross validation method. By comparing the forecast accuracy of PCA-SVM models established with different number of principal components, the best number was optimized and finally, NIR rapid identification model was established for 6 Chinese herbs. Result: At 9 000-5 400, 5 000-4 000 cm-1, SD was determined as the best pre-treatment method, and after SD pre-treatment for PCA dimension reduction, the best number of principal components was determined as 3, with an accumulative contribution rate of 93.57%. According to the grid search method, c=65 536, g=512, which were the optimum parameters of the SVM model. In the established PCA-SVM models, the forecast accuracy was 100% for both training set and verification set, and the forecast accuracy was also 100% for 5-fold cross validation method. Conclusion: The NIRS identification model based on PCA-SVM had high forecast accuracy and strong prediction ability, which can be used for rapid, nondestructive and accurate identification of above 6 kinds of TCMs when it was used in combination with the advantages of NIRS.

      摘要:Objective: To establish an HPLC method for simultaneously determining seven components in SFE-CO2 extract of Chuanxiong Rhizoma and Angelicae Sinensis Radix, and investigate the stability of coniferyl ferulate. Method: The stability of coniferyl ferulate was evaluated with CH2Cl2 as the cosolvent, and methanol, acetonitrile or 5% formic acid in methanol as the solvent. The HPLC method was performed on wondasil C18-WR column (4.6 mm×250 mm, 5 μm) with acetonitrile-0.3% acetic acid solution as mobile phase for gradient elution; the flow rate was 1.0 mL·min-1; column temperature was 35℃, and the detection wavelength was 270 nm. Result: The stability of coniferyl ferulate in the two SFE-CO2 extracts was best with the acetonitrile as solven. The seven components (ferulic acid, senkyunolide I, senkyunolide H, coniferyl ferulate, senkyunolide A, ligustulide and butylidenephthalide) co-existed in the two extracts, with good linear relationship, accuracy, recovery rate, repeatability and stability. As to SFE-CO2 extract of Chuanxiong Rhizoma, their linear range was 2.41-24.14, 3.83-38.26, 0.81-8.08, 47.43-474.32, 95.49-954.91, 164.91-1 649.14, and 3.32-33.24 ng respectively for the above 7 components, with an average recovery rate of 95.04%, 98.24%, 99.03%, 98.46%, 98.13%, 96.97%and 95.20% respectively, and RSD of 2.9%, 1.2%, 1.6%, 1.2%, 1.1%, 1.4% and 1.2%. As to SFE-CO2 extract of Angelicae Sinensis Radix, their linear range was 1.31-13.15, 1.70-17.01, 0.44-4.43, 27.52-275.18, 6.46-64.60, 206.19-2 061.92 and 3.07-30.65 ng respectively for the above 7 components, with an average recovery rate of 99.03%, 99.00%, 103.15%, 101.78%, 102.12%, 100.01% and 102.49% respectively, and RSD of 2.1%, 2.2%, 2.2%, 2.1%, 2.1%, 2.5% and 2.2%. Conclusion: Coniferyl ferulate was easily decomposed into ferulic acid. The selection of acetonitrile as solvent for the two extracts can ensure the accurate content determination of original ferulic acid and coniferyl ferulate. The method is simple and rapid, which could provide references for the quality standard for the SFE-CO2 extract from Chuanxiong Rhizoma and Angelicae Sinensis Radix.

      摘要:Objective: To investigate the anti-allergic asthma constituents in Tadehagi triquetrum. Method: The fractions were enriched from T. triquerum by using medium or high pressure liquid chromatography; such active fractions as G protein-coupled receptor-35 (GPR35) agonists were screened out by using label-free dynamic mass redistribution assays; HPLC-TOF-MS was used to analyze the main components in active fractions. Result: The 95% ethanol extracts of leaves of T. triquerum were extracted with n-heptane-70% methanol; 70% methanol extraction layer was eluted with 5%, 70% and 95% methanol through C18 medium-pressure preparation column; among them, the part eluted with 70% methanol was eluted with methanol-water through XAmide high-pressure preparative chromatography column, and separated into 10 components based on chromatographic peaks. The 10 fractions were enriched from T. triquerum; fraction 4, 5, 6, 7, 9 and 10 exhibited strong activities against GPR35 receptor, suggesting the above components are anti-allergic asthma constituents of T. triquerum. HPLC-TOF-MS was used to analyze the chemical constituents of the above components and identify 12 constituents, namely ursonic acid, catechinic acid, kaempferol-3-O-α-rhamanopyranosyl(1→6)-β-glucoside, quercetin-3-O-α-rhamanopyranosyl(1→6)-β-glucoside/quercetin-3-O-α-rhamanopyranosyl(1→6)-β-galactoside, phloroglucinol-3-O-β-galactoside, triquetrumone E, triquetrumone F, kaempferol, daucosterol and apigenin. Conclusion: Fraction 4, 5, 6, 7, 9 and 10 are active fractions, and the compounds identified from active fractions might be anti-allergic asthma constituents.

      摘要:Objective: To investigate the chemical constituents of Tripterospermum cordatum. Method: The herb of T. cordatum was crushed and then respectively extracted with 90% and 60% ethanol, and the extracts were combined and concentrated under reduced pressure. The residues were dispersed in water and respectively extracted with petroleum ether, ethyl acetate and butanol to sequentially get petroleum ether, ethyl acetate and butanol extracts. Ethyl acetate and butanol extracts were repeatedly separated and purified by column chromatography with silica gel, macroporous resin, Sephadex LH-20 and ODS-HPLC. The structures of the compound were identified by1H-NMR and13 C-NMR. Result: Seven compounds were isolated and identified as 1,7-dihydroxy-3,8-dimethoxyxanthone (1),8-hydroxy-1,2,6-trime thoxyxanthone (2), lancerin (3), isovitexin (4), iso-orientin (5), morroniside (6), 6'-O-β-D-glucopyranosylmorroniside (7). Conclusion: Compounds 3, 6, 7 are isolated from the plants of T. cordatum for the first time. And these compounds have such activities of anti-bacteria, anti-inflammation, hepatoprotection, anticoagulation and platelet aggregation and protection of myocardial cell. This study provides a good reference for further studies for the pharmacodynamic material basis and the development and utilization of T. cordatum, and a certain basis for studying chemical genetic relationship of T. cordatum.

      摘要:Objective: Clematidis Radix et Rhizoma from Ranunculaceae plant is a common traditional Chinese medicine in clinical practice. It has efficacies of eliminating wind and dampness, dredging collaterals to stop pain, and eliminating bone mineral, and can be used to treat many diseases, such as rheumatism and pharyngitis. Meanwhile, it was reported to have certain side effects. In recent years, some findings showed that its extract has an inhibitory effect on the proliferation of leukemia cells. This paper aims to study the chemical constituents from Clematidis Radix et Rhizoma and reveal its material base. Method: Clematidis Radix et Rhizoma were crushed into powder and extracted with ethanol. This residues were dispersed in H2O, and then extracted successively with petroleum ester, ethyl acetate and butanol. The compounds were isolated by column chromatography on D101 Macroporous resin, silica gel, Sephadex LH-20, MCI, and RP-18, and the structures were identified through spectroscopic methods, including 1H-NMR,13 C-NMR and MS. Result: Compounds 1-9 from this plant were identified as cimigenol (1), cimigenol-3-O-β-D-xylpyranoside (2), cimigoside (3), friedelin (4), (+)-3,3'-bisdemethylpinoresinol (5), 5-O-feruloyl-2-deoxy-D-ribono-γ-lactone (6), ferulic acid (7), syringic acid (8) and β-sitosterol (9). Conclusion: Compounds 1-6 were isolated from the rhizoma of Clematidis Radix et Rhizoma for the first time. Compounds 1-3 were cycloartane triterpenoid and obtained from Clematis genus for the first time. The isolated compounds further enrich chemical structure types from Clematidis Radix et Rhizoma, and offer a new basis to study plant taxonomy, quality control and pharmacological function.

      摘要:Objective: To study the structures and cytotoxic activity of sesquiterpene from Kadsura heteroclite rhizome. Method: Dried rhizomes of K. heteroclite rhizome were smashed and extracted with 95% ethanol for three times (150 L×1 h×3). The ethanol extract was concentrated and separated with water through polyamide chromatography into 30% ethanol, 60% ethanol and 95% ethanol, respectively. The eluate of 30% ethanol was separated by MCI chromatography with water into 30% ethanol, 50% ethanol, and 95% ethanol, respectively. The 50% ethanol was enriched sesquiterpenes. With methanol and water as mobile phase, MPLC was used to separate sesquiterpenes, and TLC and HPLC were used to combine the components. The components containing sesquiterpenes were separated with HPLC into monomeric compounds, which were by ESI-MS and NMR. Result: In vitro antitumor screening suggests that 8 sesquiterpenes were isolated from the plant and identified as 1α-hydroxy-9-deoxycacalol (1), balsamiferine B (2), balsamiferine D (3), chaetopenoid A (4), chaetopenoid B (5), asterothamnone A (6), asterothamnone B (7), and 10-dehydroxy-melleoliede B (8). Compounds 2, 6, 7 exhibited a cytotoxic activity against SMMC7721, with IC50 values of 10.6, 9.5, 15.5 μmol·L-1, respectively. Conclusion: Compounds 1-8 were obtained from Kadsura for the first time.

      摘要:Objective: To develop a certified reference material (CRM) of sakuranetin according to the Work Guideline for Reference Materials (3):Reference Material-General Principles and Statistical Method for Certification (GB/T 15000.3-2008) upon the approved by Standardization Administration of China (SAC). Method: Sakuranetin was extracted from dried barks of Populus tomentosa. The structure of sakuranetin was identified by optical rotation, elemental analysis, IR, UV, high-resolution MS, NMR, and X-ray diffraction analysis. Thin layer chromatography was carried out for the identification. Sakuranetin was divided into 400 bottles, with 5 mg per bottle. The homogeneity test, stability test, and quantitative analysis were carried out by high-performance liquid chromatography-ultra violet detector. Result: This CRM is DL-5,4'-dihydroxy-7-methoxyflavanone. The homogeneity of the 95% confidence interval was good. The storage period was 24 months at 0-8℃. The certified purity value was 99.97%, with a relative expanded uncertainty of 0.02%. The CRM of sakuranetin reached the technical requirements of CRM, and was accepted by SAC. Conclusion: The CRM of sakuranetin is successfully developed, and can be used for determining content, evaluating test methods, detecting relevant products and controlling quality.

      摘要:Objective: To establish a highly sensitive and highly accurate method for simultaneous and fast multi-elements analysis, determinate the contents of 10 kinds of trace elements in Lepisorus ussuriensis which are essential to human by using this method, fill the blank of studying on the trace elements of this plant, and provide scientific basis for its quality control and further research. Method: Three batches of L. ussuriensis produced in Jilin Province were used as samples. The contents of trace elements were determined by inductively coupled plasma mass spectrometry (ICP-MS) after the samples were diluted with wet digestion in HNO3-H2O2 system; then with In, Rh as the internal standard, the contents of elements were calculated with standard curve method. Result: For all of the analyzed elements, the standard curve correlation coefficients were all over 99.98%. RSDs were less than 3.8% and the recovery rates were between 94.8%-105.6%. The detection limit was 9×10-4 μg·g-1 for Co element, and were more than 1×10-3 μg·g-1 for other 9 elements. The linear range was 0.05-150 μg·g-1 for all the 10 trace elements, and the contents of elements Mn, Zn, Cr, Ni, Cu, V, Mo, Se, Co, Sn were 213.7, 48.63, 13.89, 6.543, 6.502, 2.571, 1.231, 0.581 9, 0.598 2, and 0.295 7 μg·g-1 respectively. Conclusion: The established rapid determination method has high sensitivity, wide linear range and good accuracy. L. ussuriensis has rich contents of Mn, Zn, Cr, Ni, Cu, V and other trace elements essential to human, and they may play direct or indirect effects on the efficacy of this plant when they participate in vivo process. This paper could provide some references for further research on elemental speciation, quality standard establishment and activity screening of L. ussuriensis.

      摘要:Objective: In the presented paper, the volatile components in flower, leaf and stem of Leonurus japonicas from Guizhou were analyzed by Fourier transform infrared spectroscopy and solid phase microextraction coupled with gas chromatography-mass (SPME-GC-MS) for the understanding and application of L. japonicas from Guizhou. Method: The detectable components with over 80% matching were statistically analyzed using the peak area normalizatilon method. Result: The detectable component in flower, leaf and stem of Leonurus japonicas from Guizhou was 43,27 and 29, respectively, and 16 volatile components were found to be contained in all samples. Furthermore, volatile components in flower, leaf and stem of L. japonicas from Guizhou were much different from those reported in literatures. The most remarked result found by comparing the main volatile components (content>3%) contained in flower, leaf and stem was that their relative content (such as trans-caryophyllene, 6,10,14-trimethyl-2-pentadecanone, β-cubebene, α-humulene and caryophyllene oxide, et al.) are much variant from each another although most of the main volatile components are found in all of samples:the flower has the highest content of trans-caryophyllene (38.9%) and the amount of α-humulene in flowers are nearly double of leaf and stem; the content of β-cubebene and caryophyllene oxide in leaf are nearly twice of flowers and stems. Conclusion: Terpenoids is the most high in volatile components of L. japonicas from Guizhou, and the content of trans-caryophyllene is first in different parts.

      摘要:Objective: In this study,sterically stabilized liposomes (SSL) containing DiR-iodide fluorescent dye (DiR-SSL) was prepared,this liposome was performed on SD rats and BALB/c nude mice for in vivo targeting studies.HGRFG-5-FAM-SSL (HFL) was preparing by SSL loading Trionycis Carapax peptide containing 5-carboxylfluorescein(5-FAM) (HGRFG-5-FAM,HF),and to investigate pharmacokinetics of administration by HF and HFL in rats,respectively. Method: DiR-SSL and HFL were prepared by thin-film dispersion method.HF and HFL were injected into the tail vein of two groups of SD rats,respectively;the concentration and dose of HF were the same.Plasma samples were collected at different time points and analyzed by multi-function microplate detector,and pharmacokinetic parameters were calculated.DiR-SSL was injected intravenously in BALB/c nude mice,the mice were in vivo imaged respectively at 1,2,5,7,24 h,and fluorescence values were read.The normal SD rats were injected DiR-SSL intravenously,after 24 h,the heart,liver,spleen,lung and kidney were imaged in fluorescence imaging system and fluorescence values were read,frozen sections of liver tissue were taken for confocal laser scanning. Result: After HF being injected into rats,its T1/2 was only 0.826 h;at the same dose,T1/2 and AUC0-∞ of HFL were 16.253 times and 11.899 times that of HF,respectively.In vivo fluorescence imaging of BALB/c nude mice showed fluorescence was mainly in the liver position and weakened gradually over time.The fluorescence of DiR-SSL in the liver of SD rats among different organs was strongest,DiR-SSL remained in the liver tissue at 24 h after injection. Conclusion: SSL drug delivery system can significantly extend T1/2 of HF in SD rats,and this system has a liver targeting effect,suggesting that this system can be used as drug carriers to realize liver targeted drug delivery.

      摘要:Objective: To analyze the endogenous metabolite changes in the fecal supernatant of mice infected with H1N1 influenza virus and intervened by Mahuang Xixin Fuzi Tang (MXF),and to explore the therapeutic mechanism of MXF through metabolomics technology. Method: KM mices were randomly divided into blank group,model group and MXF group,modle mice were intranasally inoculated with H1N1 influenza virus,and then intervened by MXF (dose of 20 mL·kg-1) through orally administration.All drug treatments were administered daily for consecutive 7 days,body weight and rectal temperature were measured,the feces were collected.Ultra performance liquid chromatography coupled to Q-exactive high resolution mass spectrometry analysis was prformed on a Halo C18 column (2.1 mm×100 mm,2.7 μm) with 0.05% formic acid in water and 0.05% formic acid in acetonitrile for gradient elution as mobile phase and the mass spectral scanning range of m/z 80-1 200.LC-MS data were processed with principal components analysis (PCA) and partial least squares discriminant analysis (PLS-DA) to find out potential biomarkers. Result: Significant differences were found in the metabolic profiles among the blank group,model group and MXF group.Nine metabolites were identified as potential biomarkers,such as phosphatidylethanolamine,phosphatidylserine,phosphatidylcholine,lyso-phosphatidylcholine,L-kynurenine,oxalacetic acid,pyridoxal,succinic acid and melatonin. Conclusion: The flu induced by H1N1 virus may interfere with the typophan,vitamin B6,glycerol phospholipids and tricarboxylic acid cycle metabolic pathway according to these identified biomarkers.MXF may play a therapeutic role in a way of regulating the above disordered metabolic pathways.

      摘要:Objective: To investigate the influence of Aconiti Radix Cocta extract on percutaneous absorption of paeoniflorin and explore synergistic effect of Aconiti Radix Cocta and Paeoniae Radix Alba. Method: KM mice were randomly divided into the Paeoniae Radix Alba gel group,Paeoniae Radix Alba and Aconiti Radix Cocta gel group and Paeoniae Radix Alba-azone gel group.Microdialysis sampling was employed,the concentration of paeoniflorin in dialysate was determined by HPLC,DAS 2.0 software was used to caculate pharmacokinetic parameters of paeoniflorin,scanning electron microscope was employed to investigate the changes of mouse skin stratum corneum. Result: AUC0-t,MRT0-t,t1/2,Tmax and Cmax of Paeoniae Radix Alba gel group were (3.28±1.01) mg·L-1·h,(3.95±0.32) h,(0.92±0.44) h,(2.00±0) h and (0.72±0.24) mg·L-1;these parameters of Paeoniae Radix Alba and Aconiti Radix Cocta gel group were (10.64±1.24) mg·L-1·h,(3.35±0.08) h,(1.32±0.67) h,(1.00±0) h and (3.06±0.38) mg·L-1;these parameters of Paeoniae Radix Alba-azone gel group were (59.82±13.51) mg·L-1·h,(3.67±0.08) h,(0.89±0.16) h,(2.67±0.29) h and (13.24±4.14) mg·L-1,respectively.Scanning electron microscope results indicated that Aconiti Radix Cocta extract had the same effect on the skin surface structure as azone in exerting its permeation enhancing effect. Conclusion: Aconiti Radix Cocta extract can significantly promote transdermal absorption of paeoniflorin in herb couple of Aconiti Radix Cocta and Paeoniae Radix Alba,which may be related to the decrease of barrier function of stratum corneum and the improvement of epidermal permeability by some ingredients in Aconiti Radix Cocta.

      摘要:Objective: To develop a simple,rapid and accurate HPLC-MS/MS for the determination of sinomenine in human plasma and study on pharmacokinetics of sinomenine after oral administration of Qishe pills. Method: Human plasma samples were prepared by protein precipitation method with methanol as solvent.The separation of sinomenine and propranolol (the internal standard,IS) was performed on a ZORBAX SB-C18 column with mobile phase of methanol-water (containing 0.1% formic acid and 5 mmol·L-1 ammonium formate) for gradient elution.Electrospray ionization (ESI) mass spectrometry was operated in the positive ion mode with selected reaction monitoring (SRM) of the transitions at m/z 330.2-152.2 for sinomenine and m/z 260.2-183.1 for IS.Pharmacokinetic parameters were calculated by WinNonlin 6.3 software. Result: The linear range of sinomenine in human plasma was 5-1 000 μg·L-1 with the lower limit of quantitation (LLOQ) of 5.0 μg·L-1,the relative standard deviations of intra-batch and inter-batch precision were below 15.0%,while the deviations of assay accuracy were within ±10.0%.The extraction recovery of sinomenine was 103.3% and no obvious matrix effect was observed.The main pharmacokinetic parameters of sinomenine (Tmax,Cmax,AUC0-48 h and t1/2) after oral administration of Qishe pills were (2.5±0.8) h,(477.9±60.1) μg·L-1,(4 808.8±791.7) h·μg·L-1 and (7.5±1.3) h,respectively. Conclusion: The validated method is successfully applied to pharmacokinetics of sinomenine in human after oral administration of Qishe pills,which can provide a methodological basis and data for the future clinical studies of this preparation.

      摘要:Objective: To evaluate the effects of Quyu Huatan Tongmai granule (QHT) on the heart hemodynamics and myocardial consumption of oxygen in experimental beagle dogs. Method: In this experiment, thoracotomy was performed to observe the effects of QHT Granule on the heart hemodynamics and myocardial consumption of oxygen in experimental beagle dogs. Totally 30 beagle dogs were randomly divided into five groups:the normal group, diltiazem group (5 mg·kg-1), high-dose, medium-dose and low-dose QHT groups (2.0, 1.0, 0.5 g·kg-1), with six in each group. They were given the corresponding drugs through duodenum. Multiple physiological recorders were adopted to measure arterial blood pressure, heart rate, standard Ⅱ ECG, coronary blood flow, resistance in coronary artery, cardiac output, left ventricular pressure, the maximal rate (dp/dtmax) of rise of left ventricular pressure, oxygen contents of the coronary artery and coronary vein(venous sinus), oxygen utilization, and oxygen consumption index. Result: Compared with control group, high-dose, medium-dose and low-dose QHT could significantly increase coronary artery blood flow and reduce coronary resistance. At 15-60 min after administration, high-dose and medium-dose QHT could increase cardiac output, with statistical differences from pre-administration value and normal group; QHT could improve the function of left ventricular; 60 min later after administration, medium-dose QHT could increase venous oxygen content by about 19% and decrease myocardial oxygen consumption by 21%, with statistical differences from normal group and pre-administration value (P < 0.05, P < 0.01). QHT showed no significant impact on heart rate, arterial blood pressure, left ventricular pressure, maximum rise rate of left ventricular pressure, myocardial oxygen consumption index. Conclusion: QHT can improve the cardiac hemodynamics and myocardial oxygen consumption in anesthetized beagle dogs.

      摘要:Objective: To explore the effects of the decoction and active ingredients of Epimedii Folium combined with Ligustri Lucidi Fructus on transforming growth factor-β1 (TGF-β1)/Smads pathway in glucocorticoid-induced osteoporosis (GIOP) rats. Method: The 49 male SD rats were randomly divided into 7 groups:the normal group, the model group, the extract of Epimedii Folium combined with Ligustri Lucidi Fructus groups (200, 100 mg·kg-1), the decoction groups (7, 3.5 g·kg-1) and Caltrate calcium D3 group (0.277 3 g·kg-1), with 7 rats in each group. All of the rats were given dexamethasone (1 mg·kg-1) through intramuscular injection twice a week to establish osteoporosis model. After eight weeks, the rats were put to death, and left femurs were collected to measure weight, calculate the ratio of wet weight, dry weight and ash weight to bone volume. The protein expressions of TGF-β1, p-Smad2/3, Smad4, Smad7 were observed by immunofluorescence staining. Result: Body weight, bone mass and the expressions of TGF-β1, p-Smad2/3 and Smad4 in model group were lower than those in normal group (P<0.01), the protein levels of TGF-β1, p-Smad2/3 and Smad4 significantly decreased (P<0.01), while Smad7 protein expression were higher in femurs (P<0.01). Compared with the model group, the body weight and bone mass increased significantly in each administration group, the protein levels of TGF-β1, p-Smad2/3 and Smad4 were increased (P < 0.05,P < 0.01), and Smad7 protein expression was decreased in all Epimedii Folium and Ligustri Lucidi Fructus treatment groups (P<0.01). Conclusion: The mechanism of the combination of Epimedii Folium and Fructus Ligustri Lucidi in treating GIOP may be related to the up-regulation on the expressions of TGF-β1, p-Smad2/3 and Smad4, down-regulation on the expression of Smad7, and strengthening of the function of osteoblasts, and reduction of bone loss in GIOP rats.

      摘要:Objective: To investigate the regulation effect of Isatidis Radix-containing serum on interferon-β(IFN-β), Toll-like receptors 3(TLR3), TBK1, interferon regulatory factor 3(IRF3) in RAW264.7 cells infected with virus, in order to explore its antiviral mechanism and provide experimental basis for clinical application. Method: The experiment was divided into 6 groups:normal control group, RSV infected model group, Isatidis Radix low, medium and high dose groups (40%, 50%, 60% Isatidis Radix-containing serum), and Ribavirin group (70% Ribavirin-containing serum). RAW264.7 cells were infected with RSV, and treated with Isatidis Radix-containing serum or Ribavirin-containing serum. After 12 hours, supernatant was collected to detect the level of IFN-β with ELISA assay, cells were collected to detect the level of TLR3, TBK1, IRF3 and IFN-β mRNA with Real-time PCR, and protein expressions of TLR3, TBK1, IRF3 and p-IRF3 were detected by Western blot. Result: Compared with normal group, model group showed obvious increases in expressions of TLR3, TBK1, IFN-β mRNA and protein of TLR3, TBK1, p-IRF3 12 h later after RAW264.7 cells were infected with RSV (P<0.01), with no significant change in IRF3 mRNA and protein. Compared with model group, medium and high-dose Isatidis Radix groups showed obvious decreases in the high expressions of TLR3, TBK1, IFN-β mRNA (P < 0.05, P < 0.01) and the high expressions of protein of TLR3, TBK1, p-IRF3 (P<0.05). The down-regulation effect of Isatidis Radix-containing serum was lower than that of Ribavirin-containing serum. Both of Isatidis Radix-containing serum and Ribavirin-containing serum had no a significant effect on the expressions of protein of IRF3 and mRNA of IRF3. Conclusion: RSV can activate TLR3 signaling transduction pathway to promote the expression of IFN-β. Isatidis Radix-containing serum can down-regulate TLR3, TBK1, p-IRF3 of TLR3 signaling transduction pathway to control the proper expression of IFN-β.

      摘要:Objective: To examine the effect of Zhengan Xifeng Tang on 6-hydroxydopamine (6-OHDA)-induced apoptosis of PC12 cell, and its antioxidant activity on neuroprotection. Method: A total of 40 male Wistar rats were randomly divided into the control group and high, middle and low-dose Zhengan Xifeng Tang groups (32, 16, 8 g·kg-1). Rats were given Zhengan Xifeng Tang with intragastric administration to prepare Zhengan Xifeng Tang-containing serum. The control group was given the same volume of normal saline. The proliferation of PC12 cells were cultured and divided into the blank group, the model group and high, middle and low-dose Zhengan Xifeng Tang groups. The blank group and the model group were given blank serum, the other three groups were given 10% drug-containing serum incubated for 1 h, and added with 100 μmol·L-1 6-OHDA to co-cultured for 24 h. the cell viability was measured by MTS. Apoptosis was analyzed by flow cytometry. Measurements of lipid peroxidation (LPO) levels were performed using spectrophotometry method. Real-time PCR were used to analyze the expression of NADPH quinine oxidoreductase 1 (NQO-1) mRNA. Result: Compared with blank control, cell viability was significantly decreased, whereas apoptosis and LPO levels were significantly increased in 6-OHDA-exposed PC12 cells (P<0.05). Compared with the model group, cell viability were significantly increased, whereas apoptosis and LPO levels were significantly decreased in Zhengan Xifeng Tang-containing serum treatment groups (P<0.05). Compared with the model group, NQO-1 mRNA expression were significantly up-regulated in Zhengan Xifeng Tang-containing serum treatment groups (P<0.05), but with no significant difference in NQO-1 mRNA expression between the normal group and model group. Conclusion: Zhengan Xifeng Tang-containing serum has the neuroprotective effect against 6-OHDA-induced apoptosis, which may be attributable to its antioxidant properties through up-regulation of NQO-1 mRNA expression.

      摘要:Objective: To observe the protective effects of Zuoguiwan (ZGW) on tertiary butyl hydroperoxide-induced MC3T3-E1 cell apoptosis, in order to study the possible mechanisms of ZGW via mitochondria-dependent pathways. Method: ZGW drug-contained serum was prepared. With murine osteoblastic MC3T3 cells as the research object, the oxidative stress model was established by tertiary butyl hydroperoxide (t-BHP), and then divided into 3 groups, blank control group, t-BHP group, ZGW+t-BHP group and Estradiol Valerate+t-BHP group. After incubation for 24, 48, 72 hours, the effects of ZGW on viability and apoptosis of t-BHP-induced MC3T3-E1 cells were investigated by MTT assay. After incubation for 48 hours, cell apoptosis was measured by acidine orange/ethidium bromide (AO/EB) staining. The changes of nucleus were observed by Hoechst 33342 staining. The changes of mitochondrial membrane potential were analyzed with rhodamine 123 (Rh123) staining. Western blot analysis was used to evaluate expressions of Bcl-2, Bax and Caspase-3 protein expressions. Result: Compared with blank model group, the proliferation of MC3T3-E1 cell was significantly inhibited in t-BHP group (P < 0.05, P < 0.01), cell apoptosis increased, mitochondrial membrane potential decreased, and the protein expression of Bcl-2 was significantly down-regulated (P<0.01). Compared with t-BHP group, the survival of MC3T3-E1 cells exposed to the oxidative damage induced by t-BHP was promoted in ZGW+t-BHP group (P<0.01), particularly at 48 h, cell apoptosis reduced, mitochondrial membrane potential significantly improved, and the protein expression of Bcl-2 was increased (P<0.05), while Bax and caspase-3 expression were decreased (P<0.01). Conclusion: Drug-contained serum of ZGW can inhibite t-BHP-induced MC3T3-E1 cell apoptosis. The possible mechanism may be related to mitochondria-dependent pathways.

      摘要:Objective: To study the protective effect of Huangqisan (HQS) on cardiomyopathy of rats with type 2 diabetes mellitus (T2DM) and its mechanism. Method: Thirty-six male SD rats were randomized into 4 groups, namely normal group, model group, HQS group (2.4 g·kg-1),and Losartan treatment group (0.1 g·kg-1), with 12 rats in each group.Rats received high fat diet and streptozocin (STZ) to reproduce the model of type 2 diabetic cardiomyopathy rats, and were orally administered with drugs for 16 consecutive weeks. Fasting blood glucose (FBG),total cholesterol (TC), triglyceride(TG), high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C) and histological features of myocardium were observed. Mitsugumin 53 (MG53) and peroxisome proliferator-activated receptor-α (PPAR-α) mRNA expression were observed by Real-time PCR. Result: Compared with the normal group,the levels of FBG,TC,TG,HDL-C, LDL-C and MG53, PPAR-α mRNA expression in model groups were significantly elevated(P<0.01). Besides, disordered arrangement and different sizes of myocardial tissues, obvious fat vacuole and widened interval between fibers were found in the model group. Compared with the model group, the levels of FBG,TC,TG,HDL-C and LDL-C in HQS group were significantly reduced (P < 0.05, P < 0.01). Both of HQS group showedregular myocardial tissue, without fat vacuole and fiber dissolution. But HQS group showed scanty fat vacuole in fiber interval. HQS group showed significant reduction in MG53, PPAR-α mRNA expressions (P < 0.05, P < 0.01). Conclusion: HQS has a good therapeutic effect on diabetic cardiomyopathy rats. Its mechanism may be related to reduction in the level of blood glucose and blood lipids, regulation on MG53/PPAR-α signaling pathway and inhibition of MG53, PPAR-α mRNA expressions of myocardium.

      摘要:Objective: To investigate the effect of Ershiwuwei Ercha pill in acute gouty arthritis rats induced by sodium urate crystals. Method: A total of 80 SD rats were randomly divided into blank group, model group, colchicine group (0.3 mg·kg-1), hydrocortisone group (4 mg·kg-1) and twenty-five taste tea pill different dose group (2.4, 1.2, 0.6 g·kg-1). Except for blank group, all of the remaining groups were included into the acute gout arthritis model by injecting uric acid into the rat ankle joint. The joint swelling degree was measured after making model at 0, 3, 6, 9, 12, 15.At the same time, white blood cell(WBC) and neutrophil(NEUT) were determined by analyzingblood cell from orbital venous plexus, and the levels of tumour necrosis factor-α(TNF-α), interleukin-1β(IL-1β), IL-8, cyclooxygenase-2(COX-2) in synovial tissues were determined by ELISA, and the expression of COX-2 protein in synovial tissues was detected by Western blot. Result: Compared with the blank group, the joint swelling degree of rats in the model group was significantly increased at each time point.The numbers of WBC and NEUT were significantly increased,the level of TNF-α,IL-1β,IL-8,COX-2 were significantly increased (P < 0.05, P < 0.01),the expression of COX-2 protein were slightly increased, but with no statistical difference. Compared with the model, the joint swelling degree of rats in the high, middle and low-dose Ershiwuwei Ercha pill groups were significantly reduced, with a significant increase in inhibition ratio.The numbers of WBC and NEUT were significantly reduced. The level of TNF-α,IL-1β,IL-8,COX-2 were significantly decreased.The expression of COX-2 protein were significantly reduced(P < 0.05, P < 0.01). Conclusion: Ershiwuwei Ercha pill can reduce joint swelling degree,leukocyte, neutrophil cell number and inflammatory factors release to resist inflammatory reaction.

      摘要:Objective: To observe the effects of Youdujing preparation on the proliferation and apoptosis of cervical immortalized cells (H8 cells) and explore the possible mechanism. Method: H8 cells, which were transfected with HPV-16 gene and passed stably, were chosen to be the object of the study. Methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay was used to detect the proliferation inhibition of H8 cells after being treated with 1, 2, 3, 4, 5 g·L-1 Youdujing for 24, 48, 72 hours. Meanwhile, the blank was set up for parallel control. MTT was used to detect the inhibitory effect on cell proliferation. Apoptotic changes of cells that were intervened by 1, 2, 3, 4, 5 g·L-1 Youdujing for 24 h and stained by Hoechst 33342 were observed by fluorescence microscope. Flow cytometry was used to detect the early-stage apoptotic rate of H8 cells after being double stained with Annexin V/propidium iodide (PI). Expressions of Caspase-3 protein and Poly ADP-ribose polymerase (PARP) protein in H8 cells, which were treated with 1, 2, 3, 4, 5 g·L-1 Youdujing for 24 h, were tested by Western blot method. Result: The growth of H8 cells intervened by 1, 2, 3, 4, 5 g·L-1 Youdujing was significantly inhibited in concentration-dependent and time-dependent manners (P<0.05), compared with blank group. According to Hoechst 33342 staining, nuclear chromatin condensations were observed after being intervened by 1, 2, 3, 4, 5 g·L-1 Youdujing for 24 h, and round, bright blue apoptotic bodies increased after being treated with 3, 4, 5 g·L-1 Youdujing. The early-stage apoptotic rates of cells treated with 1, 2, 3 g·L-1 Youdujing were (23.73±5.72)%,(63.9±3.59)%,(71.53±1.59)%, respectively, according to flow cytometry, which were significantly higher than that of control group(6.53±0.85)%(P<0.01). Caspase-3 and PARP proteins were consistently down-regulated in H8 cells (P<0.05). Conclusion: Youdujing preparation can inhibit H8 cell growth and induce early-stage apoptosis in H8 cells, which may be implemented by activating Caspase-3-PARP.

      摘要:Objective: To investigate the effects of Artesunate on suppressing the proliferation of Bortezomib-resistant multiple myeloma (MM) cell line and to explore its molecular mechanism on reversal of drug resistance. Method: Human MM cell line NCI-H929 was treated with Bortezomib in a dose-dependent manner to establish Bortezomib-resistant cell line NCI-H929BR. The inhibitory role of Artesunate on NCI-H929BR and its reversal effect against Bortezomib-resistance were determined by methylthiazolyldiphenyl-tetrazolium bromide(MTT) assay. Cell apoptosis was determined by flow cytometry, and the protein expression levels of nuclear factor-κB p65 (NF-κB p65), phospho-nuclear factor-κB p65 (NF-κB p-p65), P-glycoprotein (P-gp), B-cell lymphoma/leukemia-2 (Bcl-2) protein and Bcl-2 associated X protein (Bax) were detected by Western blot assay. Result: Bortezomib-resistance index of NCI-H929BR was 20.2 times. Artesunate treatment had significant inhibitory effect on the proliferation of NCI-H929BR and the inhibitory effect was in a concentration-dependent manner. Bortezomib (50 nmol·L-1)alone had less effect on NCI-H929BR proliferation, while the inhibition rate of artesunate (12.5 mg·L-1) alone was (23.53±2.21)% (P<0.05); Bortezomib combined with artesunate treatment had greater inhibitory effect (60.71±3.43)% (P<0.01). Artesunate treatment increased NCI-H929BR apoptosis, down-regulated NF-κB p65, NF-κB p-p65, P-gp and Bcl-2 expression levels, and up-regulated Bax expression level in a concentration-dependent manner. Conclusion: Artesunate could inhibit NCI-H929BR proliferation, promote apoptosis and reverse Bortezomib-resistance, and its mechanism may be associated with down-regulating expression levels of NF-κB p65, NF-κB p-p65, P-gp, Bcl-2 and up-regulating expression level of Bax.

      摘要:Objective: To observe the synergistic protective effect of mangiferin combined with puerarin on spontaneously hypertensive rat (SHR) inflammation injury, in order to provide the experimental basis for clinical prevention and treatment of hypertensive patients with myocarditis induced by hypertension. Method: Eight rats with normal blood pressure rat (WKY) were taken as blank group (A); 72 10-week-old SHR rats were taken as the study object and randomly divided into model group (B), benazepril group (C, 10 mg·kg-1·d-1), single mangiferin group (D, 20 mg·kg-1·d-1), single puerarin group (E, 20 mg·kg-1·d-1), mangiferin (20 mg·kg-1·d-1)+puerarin (10 mg·kg-1·d-1) group (F), mangiferin (20 mg·kg-1·d-1)+puerarin (20 mg·kg-1·d-1) group (G), mangiferin (20 mg·kg-1·d-1)+puerarin (40 mg·kg-1·d-1) group (H), mangiferin (10 mg·kg-1·d-1)+puerarin (20 mg·kg-1·d-1) group (I), and mangiferin (40 mg·kg-1·d-1)+puerarin (20 mg·kg-1·d-1) group (J). The rats were orally administered with drugs (10 mL·kg-1) for consecutively two months. They were fasting for 12 h before being put to death. The morphology of myocardial tissues in SHR was observed under microscope. Immunohistochemical assay and enzymelinked immunosorbent assay (ELISA) were used to detect interleukin (IL)-6, IL-10 and tumor necrosis factor-α(TNF-α). Result: Heart tissues of rats of each group showed no significant morphological change; immunohistochemistry and ELISA results showed that compared with the model group, each drug treated group showed decreases in myocardial IL-6, IL-10, TNF-α levels to varying degrees (P < 0.05,P < 0.01). Conclusion: Mangiferin combined with puerarin have no significant effect on morphology of heart tissues of SHR, but can down-regulate abnormal elevation of inflammatory cytokines IL-6, IL-10, TNF-α in heart tissues.

      摘要:Objective: To investigate the antitumor activity of different solvent extracts of Momordica cochinchinensis Fructus in vivo and in vitro, and the action mechanism of ethyl acetate extracts of M. cochinchinensis with a significant antitumor effect. Method: In order to identify active components, M. cochinchinensis was extracted with solvents according to the sequence of polarities through the Soxhlet extraction method. This study assessed the potency and the underlying mechanisms of action of ethyl acetate extracts of M. cochinchinensis on cancer cells through animal experiments, and used modern biology experiment technologies to investigate its underlying mechanism. Result: Among various fractions, ethyl acetate extracts of M. cochinchinensis showed significant anti-proliferative effects in vivo and in vitro (P<0.05), with no obvious toxic and side effect on experimental animals. This study found for the first time that ethyl acetate extracts of M. cochinchinensis could inhibit phosphorylation of epidermal growth factor (EGFR) tyrosine kinase and protein phosphorylation (P<0.05), and expressions of phosphor-ERK1/2, phosphor-JNK and phosphor-p38 of mitogen activated protein kinase (MAPKs) in a dose-dependent manner (P<0.05). Conclusion: Ethyl acetate extracts of M. cochinchinensis could show a significant anti-cancer effect by inhibiting phosphorylation of EGFR tyrosine kinase.

      摘要:Objective: To investigate the effect of Heshen pill (HSP) on urinary protein and the expression of nephrin protein in rats with adriamycin-induced nephropathy. Method: The rats were randomly divided into four groups, namely the model group, the HSP group (1.8 g·kg-1), the benazepril group (1 mg·kg-1) and the control group. Nephropathy rat model was induced through injection of adriamycin (6 mg·kg-1). After successful modeling, the HSP group and the benazepril group were giventhe corresponding drugs, and the model group and the control group were given equivalent volume of distilled water, 1 times a day, for 28 days. At the end of the experiment, 24 h urine was collected for urine protein detection, and aortic blood was collected for albumin(Alb), serum creatine(SCr), blood urea nitrogen(BUN) detection. The morphological changes in renal tissues were assessed by hematoxylin-eosin (HE) staining, and the changes in foot process and basilar membrane of podocytes were probed under transmission electron microscope (TEM). The nephrin protein was measured by immunohistochemistry and Western blot. Result: After the treatment, the 24 h proteinuria of the HSP group was decreased significantly than that of the model group (P<0.01). SCr, BUN were decreased significantly, and Alb was increased significantly of the HSP group than those of the model group (P<0.01). Histopathological examination revealed that glomerular epithelial cell swelling of the HSP group was reduced significantly, glomerular volume was increased slightly, mesangial cells and matrix showed slight hyperplasia, and no protein cast is found in lumen. Ultrastructural examination showed that the glomerular basement membrane of the HSP group was smooth and even, with slight thickening, and foot process was clear and complete, with occasional fusion. Immunohistochemistry showed that the positive area rate of nephrin protein of the HSP group was significantly superior to that of the model group (P<0.01). Western blot demonstrated that the nephrin protein expression of the HSP group was significantly superior to that of the model group (P<0.01). Conclusion: The inhibitive effects of HSP on proteinuria are achieved by promoting nephrin expression and repairing podocyte damage.

      摘要:Objective: To study the effect of Fuzheng Sanjie prescription in inhibiting non-small cell lung cancer and regulating serum vascular endothelial growth factor (VEGF) and tumor supplied group of factors (TSGF). Method: A total of 130 cases of non-small cell lung cancer patients were divided into chemotherapy group (A group, 32 cases), comprehensive group (Fuzheng Sanjie prescription+chemotherapy group, B group, 48 cases) and Fuzheng Sanjie prescription group (C group, 50 cases). After treatment, curative effect, recurrence and metastasis, traditional Chinese medicine(TCM) symptoms, improvement in quality of life and regulation in serum VEGF and TSGF were analyzed. Result: B group's control display rate reached 79.6%, which was significantly higher than that of A, C groups (P<0.05). After treatment, metastasis in groups A, B and C were reduced by 46.8%, 75.0% and 44.0% respectively. The new metastasis rate of the Fuzheng Sanjie prescription group was significantly lower than that of the chemotherapy group (P<0.05). In the aspect of TCM symptoms, B group (79.1%) showed the most obvious improvement (P<0.05), and significant improvement in tongue nature and fur than chemotherapy group (P<0.05). And the average levels of serum VEGF and TSGF in three groups were higher than that before treatment. After treatment, the serum VEGF level of C group and B group were significantly decreased (P < 0.05, P < 0.01). The average level of TSGF in three groups slightly decreased after treatment (P<0.05). Conclusion: Fuzheng Sanjie prescription can reduce the side effects of chemotherapy, improve clinical symptoms and quality of life, decrease the levels of serum VEGF and TSGF, and reduce the occurrence of tumor metastasis.

      摘要:Objective: To study the effect of Taohong Siwu Tang on postoperative rehabilitation in patient with patellar fracture and its mechanism of action. Method: A total of 114 cases of patellar fracture treated at our hospital from January 2014 to January 2015 were reviewed. All of the patients were randomly divided into control group and treatment group, with 57 cases in each group. Patients in both groups received surgical treatment and conventional rehabilitation training. In addition, Taohong Siwu Tang was given to treatment group since the first day after operation, which lasted for 20 courses (7 days for a course). The rehabilitation effect of two groups was compared. Result: Pain scores, knee joint swelling index score and tenderness index of the treatment group were significantly lower than that in control group at 3, 7 and 10 days after treatment (P<0.05). Moreover, knee function HSS score of the treatment group at 4, 6 and 8 weeks after treatment increased significantly compared with the control group, with statistically significant differences. At the 20th week after the operation, the clinical efficacy rate in treatment group was 98.24%, which was significantly higher than that in control group (85.96%, P<0.05). In addition, bone metabolism indexes, namely bone gla protein (BGP), alkaline phosphatase (ALP), bone absorption index of urinary calcium (U-Ca2+) improved significantly. And BGP and ALP were higher in treatment group than that of control group (P<0.05); while U-Ca2+ were decreased in treatment group when compared with control group, with statistically significant differences (P<0.05). One case in treatment group suffered from joint activity disorder, and 3 cases in control group suffered from joint activity disorder, with no statistically significant difference. Conclusion: Taohong Siwu Tang can promote rehabilitation of patellar fracture after the operation, reduce pains and swelling, and promote functional recovery of knee joint. Especially, it has a significant effect on early-stage function recovery during after fracture.

      摘要:Objective: To observe the clinical efficacy of traditional Chinese medicine(TCM) of invigorating kidney and promoting blood circulation on the inflammatory factors of patients with intrauterine adhesion after surgery, and explore its mechanism. Method: The 123 patients were randomly divided into control group (61 cases) and observation group (62 cases). Patients in control group were treated by sequential application of Estrogen-progesterone, and those in observation group were also given Bushen Huoxue decoction for two weeks in addition to the therapy of control group. Renal and liver functions of both groups were monitored. Hemoglobin (HGB), partial thromboplastin time (APTT), D-dimer (DD) and serum concentration of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) before and after treatment were measured in two groups. The incidence of adhesion within three months was compared. Result: The level of DD was significantly decreased, and HGB were higher after treatment in observation group than that in control group (P<0.01). The levels of TNF-α and IL-6 were lower at the 3rd, 7th day after the operation (P<0.01). The clinical curative effect of observation group was significantly higher than that of control group (P<0.05). The incidence of adhesion within three months of observation group was also lower than control group (12.90% vs 27.87%, P<0.05). Conclusion: Bushen Huoxue decoction may improve clinical efficacy and prevent postoperative adhesion in patients by improving uterine microcirculation and inhibiting inflammatory reaction.

      摘要:Objective: To investigate the effect of Quyu Huatan decoction on ovary polycystic change, endocrine and metabolism in patients with polycystic ovary syndrome, and explore its action mechanism. Method: The 92 patients who met the inclusion criteria in the department of endocrinology in our hospital were selected and randomly divided into 2 group, 46 cases in control group and 46 cases in observation group. Patients in control group were treated with ethinylestradiol and cyproterone acetate, 1 tablet a day, and patients in treatment group received Quyu Huatan decoction based on the treatment in control group. All patients were treated for 3 menstrual cycles, and then their serological index, polycystic ovarian changes, and clinical efficacy were compared after the treatment. Result: After treatment, the levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH) and testosterone (T) were decreased, levels of estradiol (E2) were increased, levels of glucose and lipid metabolism indexes were decreased; levels of visfatin (VF), leptin (LEP) were decreased,levels of adiponectin (APN) were increased, and the number of ovarian follicles and ovarian volume were decreased in both groups (P<0.05). As compared with control group, levels of FSH, LH and T were lower, levels of E2 were higher, levels of glucose and lipid metabolism indexes were lower, levels of VF, LEP were lower, levels of APN were higher, and the number of ovarian follicles and ovarian volume were lower in treatment group after treatment (P<0.05). The effective rate in control group (73.91%) was significantly lower than that in observation group (91.31%) (P<0.05). Conclusion: Quyu Huatan decoction had clear clinical efficacy in the treatment of polycystic ovary syndrome, and it can improve ovary polycystic changes, endocrine and metabolic function.

      摘要:Objective: To discuss the effect of Wenshen Huoxue decoction in ameliorating the activity tolerance of patients with senile chronic heart failure and Yang blood stasis, and its impact on levels of N-terminal pro-B-type natriuretic peptide (NT-proBNP), peptide, cardiac troponin T (CTnT) and plasma carbohydrate antigen 125 (CA125). Method: One hundred and fifty-eight patients were randomly divided into control group and observation group. Control group adjusted their way of life, and got compound captopril tablets, 2 tablets/time, 3 times/day. Atenolol tablets, 12.5 mg/time, 2 times/day. Spironolactone tablets, 10 mg/time, 1 time/day. And Digoxin tablets when necessary, 0.125 mg/time. In addition to the therapy of control group, patients in observation group were also administered with Wenshen Huoxue decoction, 1 dose/day. The course of treatment for both of the groups was 16 weeks. Before treatment, and at the 8th and 16th week after treatment, activity tolerance was evaluated by 6 MWT, quality of life survey of heart failure in Minnesota (MLHFQ) and concise health questionnaire (SF-36) were used to evaluate the quality of life. Before and after treatment, Lee's heart failure score, NYHA cardiac function classification and Chinese medicine syndrome were graded. Left ventricular ejection fraction (LVEF), cardiac output (CO), left ventricular end systolic diameter (LVESD), and ratio of peak early diastolic blood flow (E) and peak late diastolic blood flow (A) (E/A) were recorded by echocardiography. Levels of NT-proBNP, peptide, CTnT and CA125 were detected before and after treatment. Result: The total effect rate of cardiac function in observation group was 88.73%, which was higher than 71.43% in control group (χ2=6.631, P<0.05). The total effect rate of heart failure in observation group was 85.92%, which was higher than 67.14% in control group (χ2=6.929, P<0.01). At the 8th and 16th week after treatment, 6 minutes walking distance in observation group were higher than that of control group (P<0.01). SF-36 scores in observation group were higher than those in control group (P<0.01). After treatment, Lee's heart failure, MLHFQ, Yang blood stasis syndrome and NYHA cardiac function grading score in observation group were lower than those in control group (P<0.01). LVEF, CO and ratio of E/A in observation group were higher than those in control group (P < 0.05, P < 0.01). And levels of NT-proBNP, CTnT, CA125 and peptide in observation group were lower than those in control group (P<0.01). Conclusion: Wenshen Huoxue decoction can relieve heart failure, ameliorate cardiac function, regulate levels of NT-proBNP, CTnT, CA125 and peptide, and improve activity tolerance and quality of life of patients with senile chronic heart failure with Yang blood stasis.

      摘要:Objective: To observe the clinical effect of Danggui Buxuetang and Liuwei Dihuang Tang on early hypertensive renal damage, and its impact on levels of transforming growth factor-β1(TGF-β1), plasminogen activator inhibitor-1 (PAI-1), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α). Method: Two hundred and forty patients were divided into control group (120 cases) and observation group (120 cases) by random number table. Both groups were given valsartan capsules, 80 mg/time, 1 time/day, and nifedipine controlled release tablets were additionally provided to those beyond blood pressure standard, 1 tablet/time, 1 time/day. Control group took Liuwei Dihuang Tang, 1 dose/day; Observation group took Danggui Buxuetang combined with Liuwei Dihuang Tang, 1 dose/day. The course of treatment for both of the groups was 12 weeks. And levels of β2-microglobulin (β2-MG), urinary N-acetyl-β-glucosaminidase (NAG), cystatin (CysC), microalbumin (mALB) and serum creatinine (SCr) were detected. Urinary albumin/creatinine (UACR) and glomerular filtration rate (eGFR), including eGFRMDRD and eGFRCKD-EPI, were calculated. Before and after treatment, scores of liver and kidney Yin deficiency were graded. Levels of TGF-β1, PAI-1, IL-1β and TNF-α were detected, and adverse reactions were evaluated. Result: Total effective rate of traditional Chinese medicine(TCM) syndrome in observation group was 89.09%, which was higher than 77.27% in control group, with statically significant intergroup differences (χ2=5.491, P<0.05). After treatment, levels of β2-MG, CysC, NAG, mALB, SCr and UACR in observation group were lower than those in control group (P<0.01). And levels of eGFRMDRD and eGFRCKD-EPI in observation group were higher than those in control group (P<0.01). And levels of TGF-β1, PAI-1, IL-1β and TNF-α in observation group were lower than those in control group (P<0.01). No adverse reaction was found in both of the groups. Conclusion: Danggui Buxuetang combined with Liuwei Dihuang Tang can protect kidney by relieving clinical symptoms and early kidney injury, prolonging progress in renal damage, and regulating levels of TGF-β1, PAI-1, IL-1β and TNF-α.

      摘要:Objective: To systematically evaluate the efficacy and safety of Zhengan Xifeng Tang in treating parkinson disease (PD). Method: All randomized controlled trials (RCTs) of Zhengan Xifeng Tang for treatment of PD were searched from the main Chinese and English medical databases. The assessment tool of bias risk in Cochrane Handbook 5.1.0 software was used to evaluate the bias risks in included literature, and RevMan 5.3 software was applied for statistics and Meta-analysis. In addition, the quality of evidences was evaluated by GRADE profile software. Result: Six studies involving 498 cases were included. Meta-analysis results showed the change of Unified Parkinson's Disease Rating Scale (UPDRS) scores:Subgroup analysis was performed according to the different intervention measures, Zhengan Xifeng Tang vs Madopa(MD=-9.03, 95%CI[-11.83,-6.23]), Zhengan Xifeng Tang combined with Madopa vs Madopa(MD=-5.21, 95%CI[-9.52,-0.90]); UPDRS Ⅰ+Ⅱ+Ⅲ scores:Zhengan Xifeng Tang+Madopa, Madopa+western medicine vs Madopa, Madopa+western medicine (MD=-5.21, 95%CI[-9.52,-0.90]), with statistically significant differences; UPDRS Ⅱ+Ⅲ scores (MD=-7.18, 95%CI[-15.71, 1.35]), with no statistically significant differences. Meta-analysis on the effective rate calculated by UPDRS reduction rate showed that. Zhengan Xifeng Tang combined with western medicine vs western medicine alone (RR=1.32, 95%CI[1.09, 1.61]), with statistically significant differences. Adverse reactions:the adverse events or adverse reactions were reported in 4 RCTs. The GRADE profile showed that the included RCTs had low quality of evidences. Conclusion: Zhengan Xifeng Tang or Zhengan Xifeng Tang combined with Madopa, Madopa+western medicine were more effective than Madopa, and Madopa+western medicine. But due to low methodological quality and the small sample size in this systemic review, the clinicians shall be cautious in clinical decision making.

      摘要:Objective: To explore the composing principles of prescriptions for primary osteoporosis (POP) by using Traditional Chinese Medicine (TCM) Inheritance System, and provide references for the clinical treatment of POP. Method: The literature on TCM prescriptions for primary osteoporosis in CNKI were collected, then the data were input into the TCM inheritance system to establish prescription database. The composing principles of the prescriptions were analyzed by using the methods of frequency statistics, association rules, cluster analysis and complex system entropy clustering respectively. Result: Total 82 prescriptions related to POP were screened, involving 122 kinds of Chinese herbs. The herbs with high frequent use were Eucommiae Cortex, Rehmanniae Radix Praeparata, Epimedii Folium, Angelicae Sinensis Radix, Drynariae Rhizoma, and so on.The herbal combinations with higher frequent use were Rehmanniae Radix Praeparata-Eucommiae Cortex, Epimedii Folium-Drynariae Rhizoma, and Epimedii Folium-Eucommiae Cortex, et al. Six new prescriptions were got based on entropy clustering analysis. Conclusion: This study indicated that the prescriptions for POP were mainly in the liver, spleen and kidney meridians, and those herbs mostly had the functions of nourishing liver and kidney,invigorating spleen and supplementing Qi, promoting blood circulation to remove blood stasis. New prescriptions also mainly included those invigorating herbs based on entropy hierarchical clustering algorithm.

      摘要:Objective: To explore and analyze the clinical medical records from famous Chinese medicine expert XUAN Guo-wei based on the conditional random field. Method: A new text mining model for the medical records were designed based on conditional random field. Then this model was used to automatically recognize and extract the clinical terms information in the medical records from famous Chinese medicine expert XUAN Guo-wei, and association rules algorithm was used for data mining analysis. Result: The text information mining model which was constructed based on conditional random fields, can effectively identify different clinical terms and different disease types; in the results of recognizing different types of terms, ‘traditional Chinese medicine(TCM) diagnosis terms’ had the highest F-measure value, reaching 89.87%; in the results of recognizing different disease types, ‘eczema’ had the highest F-measure value, reaching 83.77%. Based on XUAN Guo-wei's clinical experience in the treatment of urticaria:professor Xuan often used the methods of tonifying Qi and strengthening exterior, dispelling wind and arresting itching, as well as removing dampness and detoxifying for urticaria patients; his commonly used herbal pairs included Astragali Radix-Atractylodis Macrocephalae Rhizoma, Astragali Radix-Saposhnikoviae Radix, Atractylodis Macrocephalae Rhizoma-Saposhnikoviae Radix, etc, and the confidence degree and support degree of Atractylodis Macrocephalae Rhizoma-Saposhnikoviae Radix were highest, respectively reaching 1.00 and 0.83; his commonly used triple herbal combinations included Astragali Radix-Perillae Folium-Atractylodis Macrocephalae Rhizoma, etc. Common TCM-symptom association pairs included Rehmanniae Radix-wheals, Saposhnikoviae Radix-itching, etc, and the confidence degree and support degree of Rehmanniae Radix-wheals were highest, respectively reaching 1.00 and 0.97. Conclusion: A new text information mining model was constructed in this study based on conditional random fields, which was conducive to efficiently excavate and succeed the clinical medical experience and academic thoughts of famous old Chinese Medicine experts.

      摘要:Xiao Xumingtang has been a traditional Chinese prescription for treating stroke since Tang and Song dynasty.However,it was silence for hundred of years due to some reasons.Now in recent years,it has been gradually regain.The chemical composition of Xiao Xumingtang is complex,and studies on its effective substance basis and pharmacological mechanism of anti-stroke are still in the preliminary stage of development.It is highly necessary to study and explore how to monitor the complex changes,confirm the action target and the biological effect of each component on the basis of screening and analysis of the effective components in Xiao Xumingtang.This article reviews and summarizes the research progress in material basis,pharmacological research and clinical application of Xiao Xumingtang in recent years,and provides references for the modernization prescription studies of Xiaoxuming Tang.

      摘要:Chaenomeles sinensis with a wide range of medical values was widely planted in our country, and a series of studies were carried out by domestic and foreign scholars. Studies showed that C. sinensis had organic acids, flavonoids, triterpenoids, steroids, lignans and other compounds, and exhibited antitumor, hepatoprotective, immunosuppressive and antibacterial activities. It was commonly used in combination with other Chinese herbal medicines to treat rheumatoid joint pain, lumbocrural pain and limbs anesthesia in clinic application. The studies on the chemical constituents and pharmacological activities of C. sinensis were more and more deep recently, and the results showed that C. sinensis also had polysaccharides, lignin glycosides, oxylipins, biphenyls and some other new compounds, and exhibited the effects of antioxidant, analgesic, anti-inflammatory, antibacterial, hpyerglycemic, anti-cancer, antidementia, anti-influenza virus, antianaphylaxis and choline acetyl transferase activator. Although the correlations between its material basis and pharmacological activity were further studied, the studies were not comprehensive, and still needed further research and exploration. By reviewing the domestic and abroad literature in recent years, the chemical constituents and pharmacological activities of C. sinensis were summarized in order to provide certain scientific basis for further research and development on C. sinensis.

      摘要:The literature in recent years about the intervention mechanisms of Chinese herbal compounds for rheumatoid arthritis were summarized and analyzed in this paper. Wanfang, CNKI and other large Chinese databases and foreign databases were searched for the literature about the intervention mechanisms of Chinese herbal compounds for rheumatoid arthritis. The results showed that the intervention mechanisms of Chinese herbal compounds for rheumatoid arthritis were mainly through multiple-target system, referring to regulating inflammatory factor levels, neuroendocrine, immune response pathways and gene expression.The current researches indicate that Chinese herbal compounds can intervene the pathological process of rheumatoid arthritis in various methods, showing the great advantages and prospects of Chinese herbal compounds for rheumatoid arthritis; however, it remains to be explored about how to apply Chinese traditional dialectical theory for accurate syndrome differentiation and precise prescription. In addition, big-scale samples collection and data mining will become the more effective methods to accurately and precisely explore the syndrome differentiation mechanism for Chinese herbal compounds.
