      摘要:Objective:To develop an HPLC method for quantitave determination of paeoniflorin in Tongren Niuhuang Qingxin Tablet. Method:The chromatographic column was Symmetry ShieldTM Rp18 (3.9 mm×150 mm,5 μm),the mobile phase was methanol-0.05 mol·L-1 potassium dihydrogen phosphate(25∶75) with a flow velocity of 0.8 mL·min-1. The detection wave-length was 230 nm,and the column temperature was 30 ℃. Results:It was showed that paeoniflorin had a good linear relationship between the concentrations and peak areas in (0.102 6~0.513 0)μg,and the average recovery was 98.94%(RSD=2.05%). Conclusion:This procedure is highly sensitive,and easy to be performed,which can be used in the quantitative determination of paeoniflorin in Tongren Niuhuang Qingxin Tablet.

      摘要:Objective:To develop an HPLC-ELSD method for the determination of verticine and verticinone in Wanbei Zhike capsule. Methods:The analysis was carried out on a Dikma C18 analytical column with the mobile phase consisting of acetonitrile-0.1% diethylamine (70∶30). The detector drift tube temperature was set at 93 ℃ and the nitrogen flow rate was 2.2 mL·min-1.Results:The recoveries for verticine and verticinone were 97.35% and 97.46% with RSD 1.24%and1.27%(n=5). Conclusion:The developed method is simple,rapid,replicable,and can be used for the quality control of the preparation.

      摘要:Objective:To optimize the extraction process of Fructus Citri Sarcodactylis(FCS) Formula Granule. Methods:Taking extraction yields of solids,volatile oil and 5,7-dimethoxy-coumaring as investigation index,the best extraction process was screened by orthogonal experimental design. Results:The optimum condition of extraction process was that the decoction pieces were decocted two times in 12-fold water,one hour each time. Conclusion:The optimum extraction process was simple,reasonable,stable and useful for further development.

      摘要:Objective:To optimize the technical parameters of enrichment and purification of the total glycosides in tree peony bark with macroporous resin. Methods:HPLC method was used to analyze the content of total glycosides,the adsorption and desorption performance of D101,D201,D301,D401,AB-8,NKA-9 were investigated,and the purification of conditions were inspected. Results:The best macroporous resin for purified extractive of total glycosides in tree peony bark was D101,the concentration of Paeoniflorin was 1.83 mg·mL-1 with pH 6.5,and with the elution flow rate of 1 BV·h-1. Purification of the conditions were as follows:10 BV water and 4 BV 70% ethanol were used as eluant of paeoniflorin,respectively. Portions of 70% ethanol were collected,concentrated,and dried. Conclusion:D101 resin can be used to refine the total glycosides in the water extracts of tree peony bark.

      摘要:Objective:By observed the effection in volatile oil of San Ao Tang and its Serial decoctions on ovalbumin induced asthma model in guinea pig,their action on asthma is preliminary compared. Methods:Guinea pigs were sensitized intraperitoneally with 10% ovalbumin(OVA). Two weeks after sensitization they were challenged with 1% OVA aerosol. The delitescence,scores on ethology,percentages of eosinophils(EOS) in bronchoalveolar lavage fluid(BALF),levels of interleukin(IL)-4,IL-5 and interferon gamma(IFN-γ) in serum were detected by enzyme-linked immunoassay(ELISA). Results:Volatile oil of San Ao Tang DecoctionⅠand Ⅱcould improve the percentages of EOS in BALF,IL-4,5 and IFN-γ in serum,and scores of lung tissue pathology. Volatile oil of San Ao Tang Decoction Ⅲ can significantly improve the percentages of EOS in BALF and pathological scores of lung tissue(P<0.01). Volatile oil of San Ao Tang can improve the percentages of EOS in BALF,and IL-4,IFN-γ in serum. Conclusion:Volatile oil of San Ao Tang DecoctionⅠandⅡcould regulate the immune function of the body,in aspects of regulating the cytokines,suppressing the inflammation in respiratory,and may directly inhibit the role of tracheal smooth muscle spasm,reaching its role of suppression in asthma symptoms. Volatile oil of San Ao Tang Decoction Ⅲ has the significant effects in inhibiting the production and aggregation of the EOS in respiratory.

      摘要:Objective:To study the effect of Shenqi Fuzheng Injection on the balance of Th1/Th2 in S 180-bearing mice with chemotherapy. Methods:45 of 57 tumor-bearing mice were given cyclophosphamide twice by intraperitoneal injection,and then randomly divided into four groups:the model group、the high dose group (8 g·kg-1)、the clinical equivalent dose group(4 g·kg-1) and the low dose group (2 g·kg-1) of Shenqi Fuzheng Injection. The model group and the 12 tumor-bearing mice without cyclophosphamide injection were given physiological saline by intraperitoneal injection once a day,while the others were given Shenqi Fuzheng Injection in the same way. 9 days later,the eyes and 0.5 mL blood were obtained before each mouse was sacrificed.The blood was for cytometric bead aray(CAB)test.The tumors were weighed,and the tumor index was calculated.Results:Shenqi Fuzheng Injection,in the clinical equivalent dose,could significantly depress the weights of tumors and the tumor indexes(P<0.01),and elevate the contents of TNF-α and IL-2 (P<0.01~0.05) in S180-bearing mice with chemotherapy. Conclusion:In clinical equivalent dose(4 g·kg-1),Shenqi Fuzheng Injection could make Th1 cells to be predominant to Th2 cells in S180-bearing Mice with chemotherapy.

      摘要:Objective:To evaluate the effects of pre-treatment of Chinese Medicine with functions of cooling and promoting blood circulation on the pathological changes of rats with radio-pulmonary lesion,and validate its control efficiency on the TCM syndrome. Methods:160 female wistar rats were divided randomly into the model group(A),the Chinese medicine group(B),the Chinese medicine control group(C) and the blank control group(D) with 40 rats in each group. The rats in group A and B received X-ray radiation at 3Gy×2/W×(3~5W). 8 rats randomly selected from each group were sacrificed at the end of 3th,5th,8th,12th,26th week respectively. The developmental characteristics of TCM were analyzed in radio-pulmonary lesion during exposure by dynamic examination and comparison. Results:In group A,in early stage of radiation exposure,signs and symptoms as "dominance of heat in the interior" syndrome were observed and signs as "collaterals damaged by heat" syndrome were observed by microscope; in later stage of radiation exposure,signs and symptoms as "emphraxis of collaterals by blood stasis" syndrome were observed by microscope. All the signs and symptoms in group B showed less serious than group A; and no abnormal changes were observed in group C and D. Conclusion:Pre-treatment with Chinese medicine of cooling and promoting blood can effectively prevent radio-pulmonary lesion and control the evolution of TCM syndrome. Symptomatological and developmental characteristics of radiation-induced lung injury can be revealed in animal experiments at each levels. The study provides theoretical and experimental basis for pre-treatment with Chinese medicine of cooling and promoting blood to prevent and control radio-pulmonary lesion.

      摘要:Objective:To explore the toxicity of Angelica sinensis injection (ASI) and Astragalus membranaceus injection (AMI) in the chick embryo chorioallantoic membrane (CAM) model. Methods:Fertilized eggs were incubated for 7 days at 37.8℃,and randomly divided into 6 groups,and then were respectively treated with NS (negative control),ASI,AMI,and different ratio of ASI and AMI.After another 3 days of incubation,the number of living chick embryos were counted. Results:The survival of chick embryos could not be inhibited by ASI and AMI,and our assay showed that no difference between therapy groups and negative control (P>0.05). Conclusion:ASI and AMI could be used to study angiogenesis by CAM.

      摘要:Objective:To study anti-complementary effects of the polysacchride obtained from Herba Scutellariae Barbatae. Methods:The polysacchride was extracted and analyzed with the hemolysis assay. Results:The polysaccharide inhibited complement activation on both the classic and alternative pathways with CH50 value of(1.2±0.1)mg·mL-1 and AP50 value of (4.9±0.2)mg·mL-1. Conclusions:The polysacchride obtained from Herba Scutellariae Barbatae shows significant inhibitory effect on complement activation.

      摘要:Objective:To investigate the effects of Panaxdiols Saponin (PDS)on hemorheology and PGF1a,TXB2 in rat model of acute blood stasis. Methods:Sixty rats were divided at random into six groups:normal control,model control,positive control,5 mg·kg-1 PDS,10 mg·kg-1 PDS and 20 mg·kg-1 PDS groups.The blood stasis model was made by adrenaline administration and coldness stimulation.The effects of PDS(iv.) on hemorheological parameters and PGF1a,TXB2 in rat model were observed. Results:PDS can decrease the whole blood viscosity and TXB2 in heart and lung,increase PGF1a in heart and lung. Conclusions:PDS improves significantly abnormal changes of hemorheological parameters in rat model of acute blood stasis.PDS can adjust the balance between PGF1a and TXB2.

      摘要:Objective:To explore the effects of Xingyi Jiangtangling on the of parameters of platelet and P-selectin in diabetic rats. Methods:The model of diabetic rat was established by subabdominal injection with streptozotocin 65 mg·kg-1 and then concomitant hypodemic injection with thyroxine,once a day,from 1st to 7th day. After treating with Xingyi Jiangtangling for 21 days,the content of P-selectin in the experimental rats was detected by radioimmunoassay. Result:Compared with normal group,the parameters of platelet and P-selectin in the diabetic rats were significantly increased (P<0.01).However,that of the treated group was significantly declined,compared with model group(P<0.05). Conclusion:Xingyi Jiangtangling could decline the parameters of platelet and P-selectin in the diabetic rat.Its molecular mechanism might be related to restaining the activation of platelet and mitigating hypercoagulation in microcirculation.

      摘要:Objective:To study the in-vitro inhibitory of limax polysaccharide on hepatitis B virus. Methods:The cell toxicity of limax polysaccharide was determined with MTT method,and the effect of limax polysaccharide on HBsAg and HBeAg secreted by HepG2.2.15 cells was evaluated with ELISA method on the ninth day of incubation. real-time PCR was used for determining secrected HBV-DNA in the culture medium. Results:limax polysaccharide had no obvious cell toxicity at the concentration less than 1 mg·mL-1 and depressed duplication on HBV-DNA. Limax polysaccharide had obvious restrain effect on HBsAg and HBeAg with maximum inhibition ratio addig up to 56.2%and 58.1%. The therapeutic index of limax polysaccharide was 19.98% for HBsAg and 22.78% for HBeAg. There was certain inhibition on replication of HBV-DNA(P<0.05). Conclusion:Limax polysaccharid has no obvious immediate cell toxicity on the HepG 2.2.15 cell and can inhibit duplication of hepatitis B virus in vitro.

      摘要:Objective:To investigate the mechanism of Yiqi Tongbi Mixture on anti-rheumatoid in arthritic rats. Methods:Arthritis of rat induced by type Ⅱ collage and complete Freund's adjuvant was used as rheumatoid arthritis model. The therapeutic effect of Yiqi Tongbi Mixture was observed by investigating CIA rats' joint pathological changes,right metapedes radiology and the active expression of VEGF,matrixmetalloproteinase-1,2,3,9(MMPs) in serum. Results:1.The new vessels in membrana synovialis of Yiqi Tongbi Mixture-treated rats were all conspicuously decreased compared with model group (P<0.05); 2.The radiology change of joint in treated group were improved. 3.The active expression of VEGF and MMPs in serum of model group was obviously stepped up,while the expression of Yiqi Tongbi Mixture-treated group was down-regulated.Conclusion:Yiqi Tongbi Mixture can improve CIA rats' joint pathological changes and radiological changes,and to restrain the angiogenesis of CIA rats' membrana synovialis by decreasing the expression of VEGF and MMPs in serum.

      摘要:Objective:To review the chemical studies progress on medicinal plants in genus Forsythiae.Methods:To review the types of compounds,quantities and their structures from the reference.Results:20 phenylethanoid glucosides,18 lignans,C6-C2 natural alcohols,13 terpenoids,5 flavonoids and some other compounds were isolated and identified from medicinal plants in genus Forsythiae.Conclusion:By the system summary of the chemical constituents in medicinal plants in genus Forsythiae,it is very significant for other medicinal phants in genus Forsythiae.

      摘要:The Caco-2 cell model is an in vitro model for screening drug oral absorption characteristics. It has been used widely for evaluating drug absorption characteristics and transportation mechanism.The authors describ the Caco-2 cells culture and how to establish and verify the cell monolayer model.The mechanism of its application in the drug absorption and transportation study is investigated.The applications for passive transportation of Chinese material medica,vector mediated active transportation,efflux mechanism and the drug-drug interaction in the absorption process are discussed in detail.The efficiency on the absorption and transportation mechanism research of Chinese material medica for the Caco-2 cell is evaluated.Finally,the prospect of the cell model in future application is put forward.

      摘要:In a formula,the dosage of an ingredient is closely related with its effect and toxicity. Any change in the total weight of a formula,weight of the main herbs or the ratios between herbs can change not only the power of the recipe but also its actions and indications. That is why proper dosages can make the toxic herb a remedial one,whereas wrong dosages can make a remedial herb toxic.To use the minimum dose and get the maximum effect,more studies should be made as to the Chinese herbs,especially the dose-response relationship among dosage,effect and toxicity. The spectra of poisoning and the safety of a compound preparation should be evaluated as well. In addition,attenton should be paid to the collection and analyses of the clinic reports.
