      摘要:Objective: Infering and validating the pyrolysis reaction mechanism of fried charcoal about Rhei Radix et Rhizoma,Sanguisorbae Radix and Moutan Cortex,and further improve theory of fried charcoal of the shamisen traditional Chinese medicines(TCMs). Method: The pyrolysis reaction mechanism of fried charcoal about Rhei Radix et Rhizoma,Sanguisorbae Radix and Moutan Cortex was infered with indefinite temperature method of single scan rate(Coats-Redfern model and Achar model) that belonged to mode-fitting method.The pyrolysis reaction mechanism was validated by comparing thermal analysis curves about the theoretical value,experimental value of conversion rate(α) and 1/T. Result: The pyrolysis mechanism functions of fried charcoal about the shamisen TCMs were as follows: No. 9 mechanism function in integral form of g(α)=[(1-α)-1/3-1]2 for Rhei Radix et Rhizoma and Moutan Cortex,and No. 2 mechanism function in integral form of g(α)=α+(1-α)ln(1-α) for Sanguisorbae Radix. Conclusion: Pyrolysis reaction mechanism of fried charcoal about the shamisen TCMs all belong to reaction diffusion,there are a certain differences between the theoretical value and the experimental value of α.

      摘要:Objective: To prepare nanosuspension-based gel of curcumin (Cur NS-gel) and investigate its therapeutic effect on rat selenium-induced cataract. Method: Cur-loaded nanosuspensions (Cur NS) was prepared by high pressure homogenization method.Carbopol 940 was added as the matrix for Cur NS-gel.The morphology,drug loading,particle size,Zeta potential,gelling buffer ability,dropping ability and viscosity were determined.The in vitro release profiles of Cur was determined by flow-through cell method.The rats were divided into blank group,model group,Cur suspension group,Cur NS group and Cur NS-gel group,the rat selenium-induced cataract model was established with dose of 0.25 μL · g-1.The lens opacity and relative biomarkers were detected. Result: Cur NS had spherical-like shape with particle size of (298.2±14.9) nm and Zeta potential of (30.7±2.12) mV.The physical properties were not changed significantly after the Cur NS-gel was obtained,together with the elevated viscosity increased to (802±15.9) mPa · s.The in vitro cumulative release in 6 h for Cur NS and Cur NS-gel were 26.2 times and 18.3 times of that in Cur suspension,respectively;and lag time phenomenon appeared in Cur NS-gel.In vivo pharmacodynamic study indicated that Cur NS-gel could significantly delay the development of lens opacity.The total superoxide dismutase activity of Cur NS-gel was elevated by comparing with model group,Cur suspension group and Cur NS group.The content of glutathione in Cur NS-gel group was 2.1 times and 1.5 times of that in Cur suspension group and Cur NS group,respectively(P<0.05). Conclusion: Nanosuspension-based gel can be used as effective carriers of Cur,which can further improve its therapeutic effect on rat selenium-induced cataract.

      摘要:Objective: To confirm the existence of boswellic acids in essential oil and water extract of vinegar-processed Olibanum,and to determine its optimum extraction method by comparing the anti-inflammatory activity and the content of main boswellic acids. Method: Water extraction,ethanol extraction and steam distillation-ethanol continuous extraction was adopted,contents of six boswellic acids in water extract,ethanol extract and steam distillation-ethanol continuous extract were determined by HPLC,the anti-inflammatory effect was evaluated by in vitro model of RAW264.7 macrophages induced by lipopolysaccharide(LPS). Result: Four boswellic acids can be detected in essential oil with low extraction rate of 1.73%-13.02% and all of six boswellic acids can be extracted by water.In the same extraction conditions,efficiency of ethanol extraction was optimum with extraction rate of 81.09%-88.38%.Optimum ethanol extraction process was as follows:refluxing extracted twice with 8 times the amount of 95% ethanol for 30 min per time;yield of six boswellic acids were >90%.The content of tumor necrosis factor-α(TNF-α) was (1.933 2±0.422 9) μg · L-1 for ethanol extraction,(2.248 5±0.332 2) μg · L-1 for water extraction and (2.148 3±0.534 3) μg · L-1 for steam distillation-ethanol continuous extraction;their interleukin-1β(IL-1β) contents were (23.22±3.24),(25.53±3.23),(24.32±4.32) ng · L-1,respectively. Conclusion: The existence of boswellic acids in essential oil and water extract of vinegar-processed Olibanum is confirmed.Ethanol extraction is superior to other extraction methods with the strongest anti-inflammatory activity.

      摘要:Objective: To compare the dryness in different processed products of Aurantii Fructus,and investigate their effect on gastrointestinal function of functional dyspepsia(FD) rats. Method: Taking water intake,urinary output,aquaporin 3(AQP3) content in kidney and serum cyclic adenosine monophosphate(cAMP)/serum cyclic guanosine monophosphate(cGMP) as indexes,effect of crude and processed products of Aurantii Fructus with different doses on the water metabolism of healthy rats was observed.Taking gastric mucosal damage index and contents of serum tumor necrosis factor-α (TNF-α),interleukin-6(IL-6),IL-8 as indexes,effect of crude and processed products of Aurantii Fructus with different doses on gastric mucosal injury of healthy rats was observed.Taking gastric remnant rate,intestinal propulsive rate and contents of serum motilin(MTL),vasoactive intestinal peptide(VIP),calcitonin gene-related peptide(CGRP) as indexes,effect of crude and processed products of Aurantii Fructus with different doses on gastrointestinal function of functional dyspepsia rats was observed. Result: In the aspect of water metabolism,the same dose of medicine compared with the blank group,difference was in the order of Aurantii Fructus > Aurantii Fructus fried with bran > Aurantii Fructus fried with honey and bran,and the high dose group is higher than the low dose group.In indexes of gastric mucosal injury,there was no significant difference between processed products of Aurantii Fructus and the blank group.In the aspect of gastrointestinal motility,the same dose of medicine compared with the model group,difference was in the order of Aurantii Fructus fried with bran > Aurantii Fructus > Aurantii Fructus fried with honey and bran,and the low dose group > the high dose group. Conclusion: The dryness of Aurantii Fructus is mainly performance for body fluid injury,the degree is related to the drug dosage,and it has no obvious stimulation of stomach.After being processed,Aurantii Fructus can relax the body fluid injury to a certain extent,and Aurantii Fructus fried with honey and bran has the best effect to ease the adverse reaction.Different processed products of Aurantii Fructus all can promote the recovery of gastrointestinal function of FD rats,and the treatment of Aurantii Fructus fried with bran is stronger than the crude and honey-bran products.

      摘要:Objective: To study on the scientificity of theory of reducing toxicity by processing for Euphorbia fischeriana dry roots,and understand its pharmacodynamic changes before and after processing,and develope a RP-HPLC for the simultaneous determination of jolkinolide B and 2,4-dihydroxy-6-methoxy-3-methylacetophenone in E.fischeriana dry roots and its processed products. Method: HPLC-Q-TOF/MS was employed to compare composition differences of E.fischeriana dry roots before and after processing,the acute toxicity and diarrhea of E.fischeriana dry roots on the normal mice before and after processing was compared,and RP-HPLC was adopted to determine the contents of jolkinolide B and 2,4-dihydroxy-6-methoxy-3-methylacetophenone in this herb and its different processed products. Result: Compared with the raw products,the contents of 30,20,21 peaks were decreased in milchigs products,Chebulae Fructus soup products and wine products;LD50 of raw products,milchigs products,Chebulae Fructus soup products and wine products were 2.694,3.642,4.309,3.461 g · kg-1,respectively.When the dosages of drug powder were 0.53,0.133 g · kg-1, the raw products and its processing products all could significantly improve intestinal propulsive rate.Jolkinolide B and 2,4-dihydroxy-6-methoxy-3-methylacetophenone were linear in the ranges of 0.060 72-1.214 4,0.060 12-1.202 4 μg,their average recoveries were 103.03% with RSD of 3.6% and 107.55% with RSD of 4.7%. Conclusion: The processing mechanism of E.fischeriana dry roots may be related to the changes of chemical components after processing.The determination of two components in this herb and its processed products is accurate and reliable with good reproducibility,it can be used for the quality control of E.fischeriana dry roots and its processed products.

      摘要:Objective: According to previous study,macromolecular compounds like starch can influence the dissolution of small molecule compounds in the decoction.This study was in order to investigate the effect of processing on starch by determining the basic physicochemical properties of starch in Pinelliae Rhizoma and Pinelliae Rhizoma Praeparatum. Method: The effect of processing on starch was evaluated by detecting the swelling degree,solubility and water binding capacity,crystal structure and micromorphology of starch in Pinelliae Rhizoma and Pinelliae Rhizoma Praeparatum. Result: There was no significant difference in microstructure between starch in Pinelliae Rhizoma and Pinelliae Rhizoma Praeparatum.Compared with starch in Pinelliae Rhizoma,swelling degree,solubility and water binding capacity of starch in Pinelliae Rhizoma Praeparatum were obviously decreased from 30.80%,17.22% and 10.56 to 13.93%,10.13% and 7.82,respectively;but the crystallinity of starch in Pinelliae Rhizoma Praeparatum was significantly increased from 31.77% to 55.11%. Conclusion: Processing can influence the physicochemical properties of starch in Pinelliae Rhizoma.Combined with our previous study,we speculate that the change of starch in Pinelliae Rhizoma is one of the reasons influencing the efficacy during processing.

      摘要:Objective: To establish a near infrared spectral qualitative model of Fluoritum for rapidly identifying raw,calcined,vinegar calcined products and its counterfeits based on multi-reference correlation coefficient method(MRCC) and back-propagation artificial neural networks(BP-ANN). Method: The near infrared spectra of 4 kinds of Fluoritum samples were collected,including raw products,calcined products,vinegar calcined products and counterfeit products.The second derivative and 9 points smoothing were used to calculate multiple correlation coefficients.Multiple correlation coefficients were used as input data in Matlab 2014a software,and a rapid identification model for 4 different types of Fluoritum samples was established by BP-ANN. Result: BP-ANN identification model of near infrared spectroscopy of Fluoritum was established.The prediction results of 14 batches of samples were correct,only 1 batch of samples was predicted error,the accuracy rate was 93.33%. Conclusion: The BP-ANN identification model of near infrared spectroscopy of Fluoritum can be used to distinguish among 4 different types of samples by a one-time integrated operation,and the result is accurate and reliable.In addition,based on the correlation coefficient method of near infrared spectroscopy,the multi-group correlation coefficient calculated by the multiple reference spectra is used as the input data of the network characteristics,the compression of spectral data is realized.

      摘要:Objective: To establish a simple and efficient method for the preparation of bakuchiol,and study on its antiosteoporotic activity. Method: The powder of Psoraleae Fructus was extracted with ethanol to get the crude extract of bakuchiol,and bakuchiol was purified with D101 macroporous resin.Moreover,antiosteoporotic activity of bakuchiol on the osteoporosis model was evaluated by zebrafish in vivo. Result: Optimum process for the purification of bakuchiol was as follows,the powder of Psoraleae Fructus was soaked in 85% ethanol overnight and the ratio of ethanol to sample was 1∶ 8,it was refluxed thrice for 2 h each time.The crude extract was concentrated by a vacuum rotary evaporator,and it was dissolved with 0.2% NaOH solution,it was adsorbed by D101 macroporous resin column with a flow rate of 1 BV · h-1,washed by 6 BV of water after 4 h and 2 BV of 60% ethanol,4 BV of 95% ethanol was used to wash the column at last,and eluent was concentrated to get bakuchiol.The yield and purity of bakuchiol prepared by the method were 10% and 80% at least,respectively.In addition,the bioassay indicated that it could prevent bone loss of zebrafish induced by prednisolone,and accelerate growth of zebrafish bone. Conclusion: The method combines the advantages of generality,rapidity and simplicity with high purity.Bakuchiol can be developed into drugs and foods for preventing or treating osteoporosis.

      摘要:Objective: To grade the Citrus reticulate ‘Chachi’ commercial specifications and formulate the new draft of C. reticulate ‘Chachi’ commercial specifications and grades for the market application. Method: Experienced experts in the study field of Chinese medicinal materials commercial specifications and grades were selected, and the C. reticulate ‘Chachi’ medicinal materials were evaluated by a two-round blind investigation based on Delphi method. The experts evaluated the grade of C. reticulate ‘Chachi’ commercial specifications by observing their appearance. Then the statistical analysis was conducted to screen out the core indexes of the sensory evaluation of C. reticulate ‘Chachi’ commercial specifications, sort out their importance, and calculate the individual reproducibility and consistency among experts group in sensory evaluation. Result: The main indexes of sensory evaluation of C. reticulate ‘Chachi’ commercial specifications were smell, oil cavity, taste, thickness and surface color, etc., and their degree of importance was weakened successively. The average individual reproducibility of two blind evaluations was 81.4%, and the average reproducibility among experts group was 75.0%. Conclusion: The experts experience in sensory evaluation of the C. reticulate ‘Chachi’ commercial specifications and grades was objective and reproducible, and Delphi method can be used as one of methods to study the traditional sensory evaluation of Chinese medicinal materials objectively and normatively. By combining the experimental results and the market actual situations, the new draft of C. reticulate ‘Chachi’ commercial specifications and grades was formulated in this study, providing a reference and theoretical basis for the ultimate draft of the grades standards.

      摘要:Objective: To determine the contents of alcohol-soluble extractive,total saponins and total polysaccharides in different grades of Asparagi Radix,and to explore the correlation between the effective components and their grades for investigating the rationality of grading method of Asparagi Radix. Method: The content of alcohol-soluble extraction of Asparagi Radix with different grade was determined according to the 2015 edition of Chinese Pharmaeopoeia.Ultraviolet visible spectrophotometry was used to determine the content of total saponins in Asparagi Radix samples with detection wavelength at 408 nm.The content of total polysaccharides was quantified by phenol-sulfuric acid method.The correlation analysis and linear regression analysis were used to analyze the correlation between the grades and quantitative indexes. Result: The contents of alcohol-soluble extractive and total saponins existed significant difference (P<0.05) in different grades of Asparagi Radix. The content of total polysaccharides was significantly different (P<0.05) between the first and the third grade,but the difference was not significant between other grades.The correlation analysis indicated that the grades showed extremely significant positive correlation with alcohol-soluble extractive content and total saponins content(P<0.01),and showed significant positive correlation with total polysaccharides content(P<0.05). Conclusion: The contents of total saponins and alcohol-soluble extractive in Asparagi Radix are consistent with the classification method by taking diameter of the middle part of medicine as the main standard,which indicates that the grading method of Asparagi Radix is still reasonable.Quality evaluation of Asparagi Radix is recommended to determine the content of total saponins by taking contents of alcohol-soluble extractive and total polysaccharides as the secondary indicators.

      摘要:Objective: To synthesize bezohpenone derivatives, characterize their structures and study their antifungal activities. Method: Benzophenone derivatives were synthesized by Friedel-Crafts acylation reaction with benzoyl chloride or phthalic annhydride as the raw materials. The structures of these compounds were elucidated by 1H-NMR, 13C-NMR and MS-ESI. Their effects on minimum inhibitory concentration (MIC) of four fungi were investigated with antimicrobial susceptibility test. Result: The results showed that 13 benzophenone derivatives had antifungal activity in varying degrees, and the carbonyl group was a key to the antifungal activity of benzophenone derivatives. Different substituent groups had significant differences in the effect on their antifungal activities and the diphenyl ketone derivatives with multiple chlorine atoms showed the most excellent antifungal activity. Conclusion: 13 benzophenone derivatives had antifungal activity. The results of this experiment offered a method and reference for structure modification of natural benzophenone compounds with antifungal activity.

      摘要:Objective: To study the chemical constituents of the whole plant of Liparis nervosa. Method: The herb L. nervosa was crushed into powders and then extracted with 95% EtOH at 85 ℃. After removal of EtOH under reduced pressure, the solvent was recovered to obtain total extract. The extract was then suspended in distilled water and partitioned successively with petroleum ether (PE), EtOAc, and n-BuOH to get corresponding fractions. Silica gel column chromatography (CC), Sephadex LH-20 gel CC and semi-preparation HPLC were used for the separation and purification of PE and EtOAc fractions. The structures of the obtained compounds were identified by physicochemical properties and spectra data. Result: Twelve compounds were isolated from PE and EtOAc extracts of L. nervosa and their structures were identified as p-hydroxybenzoic acid (1), protocatechuate (2), 3-(4-hydroxyphenyl)-2-propenoic acid (3), (E)-ferulic acid docosyl ester (4), (Z)-ferulic acid docosyl ester (5), stigmast-4-ene-3-one (6), meranzin hydrate (7), xanthotoxol (8), auraptenol (9),stigmasterol-3-O-β-D-glucoside (10), vanillic acid (11) and p-hydroxybenzaldehyde (12). Conclusion: Compounds 1-10 were obtained from the genus Liparis for the first time, and compound 11 was obtained from L. nervosa for the first time.

      摘要:Objective: To analyze and identify the chemical constituents of Xuanmai Ganjie granules by ultra-performance liquid chromatography time-of-flight mass spectrometry (UPLC-Q-TOF-MS). Method: The separation was performed on an XB-Bridge BEH C18 column (2.1 mm×100 mm, 2.5 μm) with acetonitrile-0.1% formic acid solution as mobile phase for gradient elution. The flow rate was 0.3 mL · min-1. Electrospray ionization source was used to scan and collect data in positive and negative ion modes. Result: According to the high resolution MS spectra data, fragmentation ion information and retention time, 33 chemical constituents of Xuanmai Ganjie granules were identified including 9 iridoid glycosides, 20 flavonoids, and 4 saponins by comparison with literature reports and MS cleavage mechanism. Conclusion: The established rapid analysis method based on UPLC-Q-TOF-MS could provide technical support for studying the chemical constituents of Xuanmai Ganjie granules and further studies on its material basis.

      摘要:Objective: To establish a method to determine the total polysaccharides content in Hedysari Radix and Astragali Radix from different areas of Gansu province, compare their contents of total polysaccharides from different habitats, and explore the effect of different growth years (1, 2 years) on the total polysaccharides content. Method: Total polysaccharides of Hedysari Radix and Astragali Radix from different areas of Gansu province were determined by using the phenol sulfuric acid method at 490 nm, with glucose as reference substance. Result: A good linear relationship of glucose was achieved within the range of 16.5-165 μg (r=0.999 9), the average recoveries of Hedysari Radix and Astragali Radix were 97.43% (RSD 2.1%, n=6) and 100.33% (RSD 2.3%, n=6).For one-year-old plant, the average content of total polysaccharides in Hedysari Radix from Dangchang, Wudu, Longxi and Gansu were 11.45%,5.96%,10.97% and 9.35%, respectively. The average contents of total polysaccharides in Astragali Radix from Dangchang, Wudu, Longxi and and Gansu were 3.69%, 1.93%, 7.06% and 4.02%, respectively. For two-year-old plant, the average contents of total polysaccharides in Hedysari Radix from Dangchang, Wudu, Longxi and Gansu were 6.28%, 5.82%, 6.69% and 6.23%, respectively. The average contents of total polysaccharides in Astragali Radix from Dangchang, Wudu, Longxi and Gansu were 7.91%, 8.03%, 7.12% and 7.73%, respectively. Conclusion: The method is simple, accurate and reproducible, and can be used for the determination of total polysaccharides in Hedysari Radix and Astragali Radix. There was significant differences in varieties and years between the content of total polysaccharides in Hedysari Radix and Astragali Radix from 5 counties and 1 city in Gansu province (P<0.01). According to the analysis of the content of total polysaccharides, Hedysari Radix was not suitable to substitute Astragali Radix. The total polysaccharide content of one-year-old Hedysari Radix was higher than that of Astragali Radix, and the result of two-year-old plants was lower than that of Astragali Radix. Hedysari Radix in Dangchang is better, and the one-year-old Hedysari Radix is the best. One-year-old Astragali Radix in Longxi, Minxian is better, and two-year-old Astragali Radix in Wudu is better.

      摘要:Objective: To establish the quality control standard for Huoluo Zhitong gel. Method: Thin layer chromatography(TLC) was used for qualitative identification of Cinnamomi Cortex, Aconiti Kusnezoffii Radix and Corydalis Rhizoma in Huoluo Zhitong gel. Gas Chromatograph-Mass Spectrometer-computer(GC-MS)was used to qualitatively identify the volatile oil in the prescription, and high performance liquid chromatography(HPLC) was used to determine the contents of cinnamaldehyde, mesaconitine, hypaconitine and aconitine. Result: The characteristic TLC spots of the testing samples were the same as the spots of the standard samples without any interference from the negative reference substance. Cinnamic aldehyde was the main component in volatile oil, followed by eugenol. Mesaconitine showed a good linear relationship with peak area within the injection amount of 0.48-2.88 μg(r=1), hypaconitine within the injection amount of 0.16-0.96 μg(r=1), aconitine within the injection amount of 0.34-2.04 μg(r=1), and cinnamaldehyde within the injection amount of 0.268-1.608 μg(r=0.999 1). Conclusion: The method has a good separation and it is simple and accurate, which could be used in the quality control of Huoluo Zhitong gel.

      摘要:Objective: To explore the effect of Jianpi Yichang powder (JP) on protein and mRNA expression levels of heat shock protein 70 (HSP70) in colon tissues of rats with ulcerative colitis (UC). Method: The 60 healthy SPF male SD rats were randomly divided into normal group and model group. The UC models were established by trinitro-benzene-sulfonic acid(TNBS)/ethanol method. Then the successfully modeled rats were randomly divided into the model group, sulfasalazine group (0.3 g ·kg-1), and JP high, middle and low dose groups (204, 136 and 68 g ·kg-1) with 10 rats in each group. After ig administration for 21 days, the general condition and the colonic mucosa injury of rats were observed. Immunohistochemistry, Western blot and Real-time PCR were used to detect the protein and mRNA expression levels of HSP70 in colon tissues of rats. Result: As compared with the normal group, the colon mucosal injury score was increased significantly (P<0.01), and the protein and mRNA expression levels of HSP70 were significantly decreased (P<0.01) in model group. As compared with the model group, the colon mucosal injury scores in each drug group were significantly lower (P<0.01), and the protein and mRNA expression levels of HSP70 were significantly increased (P<0.05, P<0.01). The effect was most obvious in JP high dose group (P<0.05, P<0.01). Conclusion: JP may promote the expression of HSP70 to protect intestinal mucosal immunity in UC rats, so as to play a therapeutic role.

      摘要:Objective: To observe the effect of Astragali Radix polysaccharides on the proliferation of colon cancer cell line SW620, and to explore its anti-tumor mechanism. Method: SW620 cells were cultured with different concentrations of Astragali Radix polysaccharides (0.1,0.2,0.4,0.6,0.8,1.0 g ·L-1) in vitro for 48 h, and then treated with thiazole blue colorimetry (MTT) to detect the contents of Astragali Radix polysaccharides on the proliferation of human colon cancer cells SW620. Propidium iodide (PI) was used to detect the tumor cell cycle after treatment, and Annexin V/PI double staining was used to detect the apoptotic rate of tumor cells, while SW620 cells were cultured with Astragali Radix polysaccharides (1.0 g ·L-1) for 48 h in vitro. The protein expressions of Caspase-3, Caspase-9, cytochrome C, Bax and Bcl-2 were detected by Western blot, while SW620 cells were cultured with Astragali Radix polysaccharides (0.25, 0.5, 1.0 g ·L-1) for 48 h in vitro. Result: The growth of SW620 cells was significantly inhibited by APS in a dose-dependent manner (P<0.05, P<0.01). Compared with control group, the ratio of G2/M phase in treatment group of Astragali Radix polysaccharide 1.0 g ·L-1 was increased significantly (P<0.05, P<0.01). The early apoptosis rates, the late apoptosis rates and the total apoptosis rates were significantly higher than those in control group (P<0.05, P<0.01). Bax/Bcl-2 radio was significantly increased, and the expression of Caspase-9 was significantly decreased (P<0.05, P<0.01). The expressions of activated Caspase-3, activated Caspase-9 and cytochrome C were significantly increased (P<0.05, P<0.01). Conclusion: Astragali Radix polysaccharide could inhibit the proliferation of human colon cancer SW620 cells and induce the apoptosis of SW620 cells through mitochondrial pathway.

      摘要:Objective: To observe the effect of conditioned medium of MCF-7 cells stimulated by jolkinolide B (JB) on human umbilical vein endothelial cells (HUVECs), and analyze the mechanism of JB. Method: The 25, 55, 85 mg ·L-1 of JB were used to stimulate MCF-7 human breast cancer cells, respectively. The cells stimulated by different concentrations of JB were taken as the JB groups, and the normal cultured MCF-7 cells were taken as control group. The supernatants of tumor cells stimulated by different concentrations of JB were collected as the conditioned medium (CM) of the corresponding concentrations. According to the corresponding concentrations of JB, the conditioned medium was used to culture HUVECs, and these cells were taken as 25-CM group, 55-CM group and 85-CM group, respectively. The normally cultured HUVECs were taken as the non-conditioned medium group (NCM). Methylthiazolyldiphenyl-tetrazoliumbromide (MTT) assay, Annexin V-FITC, cell scratch and transwell chamber experiment were separately used to detect the proliferation, apoptosis and migration of HUVEC in the above groups. The Western blot assay was used to detect the mechanism of JB on MCF-7 cells, and the enzyme linked immunosorbent assay (ELISA) was used to analyze the changes in the content of vascular endothelial growth factor (VEGF) in the conditioned medium of each group. Result: Compared with NCM group, the apoptosis rate of HUVEC in 25-CM group, 55-CM group and 85-CM group were gradually increased(P<0.01), and the A values of HUVEC proliferation in the above groups were significantly decreased (P<0.01); compared with NCM group, the closure rate and the migration rate of HUVEC in 25-CM group, 55-CM group and 85-CM group were significantly decreased (P<0.01). There were significant differences in apoptosis rate, the A value of cell proliferation, the scratch closure rate, and cell migration rate among 25-CM group, 55-CM group and 85-CM group (P<0.01). Compared with control group, both JB with the concentrations of 25, 55, 85 mg ·L-1 can inhibit the expression of Akt/STAT3/mTOR signaling pathway in MCF-7 cells, and reduce the expression of VEGF in MCF-7 cells. Conclusion: JB can reduce the secretion of VEGF in MCF-7 cells by inhibiting the Akt/STAT3/mTOR signaling pathway, so as to inhibite the proliferation and migration of HUVEC.

      摘要:Objective: To observe the intervention effect and mechanism of Yifei Jiedu decoction on the invasion and metastasis of lung cancer adenocarcinoma A549 cells. Method: A549 cells were cultured and treated with 150 μmol · L-1 CoCl2, 150 μmol · L-1 CoCl2+15 g · L-1 Yifei Jiedu decoction, and 15 g · L-1 Yifei Jiedu decoction, respectively. Untreated cells were used for normal control. Morphological changes were observed under inverted microscope. Wound scratch assay were performed to further identify if Yifei Jiedu decoction could reduce the migration ability of A549 cells. Transwell was used to explore the intervention effect of Yifei Jiedu decoction on the invasion and movement of A549 cells. Expressions of E-cadherin, Vimentin, Snail, β-catenin and p-gsk-3β were examined by Western blot. Result: CoCl2 can induce morphological changes in A549 cells, which were accompanied by up-regulation of E-cadherin expression and down-regulation of Vimentin. Wound scratch assay indicated that Yifei Jiedu decoction could reduce the migration ability of A549 cells. Transwell assay showed that Yifei Jiedu decoction could inhibit the invasion of A549 cells(P<0.05). Western blot showed that Yifei Jiedu decoction could up-regulate the expressions of E-cadherin,p-gsk-3β protein and down-regulate the expressions of Vimentin, Snail and β-catenin protein respectively(P<0.05). Conclusion: Yifei Jiedu decoction could inhibit the migration and invasion of human adenocarinoma A549 cells via regulation of β-catenin expression.

      摘要:Objective: To observe the effect and mechanism of Sixiaosan on wound tissue healing of diabetic foot ulcer in rats. Method: SPF SD rats were randomly divided into 4 groups: blank group (normal rats), model control group, metronidazole group (5 mL · kg-1 · d-1) and Sixiaosan group (100 mg · kg-1 · d-1). Metronidazole injection was applied in metronidazole group for external use, Sixiaosan was provided to Sixiaosan group for external use. At 14 d after treatment, serum epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) were detected. Immunohistochemistry was used to detect the numbers of fiber cells and newborn blood capillaries in wound granulation tissues; hematoxylin-eosin (HE) staining was used to observe histomorphological changes. Result: In metronidazole and Sixiaosan groups, bFGF and EGF in serum of rats were significantly increased (P<0.01). Specifically, bFGF and EGF in Sixiaosan group increased higher than those of metronidazole groups (P<0.01). In metronidazole and Sixiaosan groups, the number and density of newborn blood capillaries and the number of fibroblasts were significantly increased (P<0.05). In Sixiaosan group, the number and density of newborn blood capillaries and the number of fibroblasts increased higher than those of metronidazole groups (P<0.05). Conclusion: Sixiaosan is able to improve the levels of bFGF and EGF in serum of rats, and conducive to the proliferation of fibroblasts and newborn blood capillaries, and the growth of wound granulation tissues, so as to promote the healing of diabetic foot ulcer.

      摘要:Objective: To study the analgesic effect and central mechanism of CQ compound in rat models with tibia metastatic cancer pain. Method: Female Wistar rats were randomly divided into sham operation group and surgery group, with operation control and tibia metastatic cancer pain operation for Walker-256 breast cancer cells respectively. The 40 successfully modeled rats were randomly divided into model group, CQ high dose (CQ-H) group (200 mg · kg-1), CQ middle dose (CQ-M) group (150 mg · kg-1), CQ low dose (CQ-L) group (100 mg · kg-1) and gabapentin group (100 mg · kg-1), 8 rats in every group. Mechanical withdrawal threshold (MWT) measured by von frey hairs fiber filament was used to evaluate rats' pain behavior. Microdialysis was used to detect glutamate (Glu) level of cerebrospinal fluid; radioimmunoassay was used to detect substance P (SP) and β-endorphin (β-EP) contents in L4-L6 spinal cord sections; and Aim Plex Multiple Immunoassay was used to detect the, interleukin-12 (IL-12-P70) and β-nerve growth factor (β-NGF) levels of tumor local tissues. Result: As compared with the model group, CQ compound groups and gabapentin group significantly increased MWT of rats; MWT of rats in CQ compound groups began to rise in 30 min after administration, and peaked in 60 min (P<0.05, P<0.01). Threshold elevation in CQ-H, CQ-M, and CQ-L groups showed dose effect relationship, and the analgesic effect of three consecutive days administration was stronger than that of single dose administration; CQ compound and gabapentin reduced the level of glutamate, SP and β-EP (P<0.05), and raise the level of IL-12-P70 (P<0.05). Conclusion: CQ compound can relieve the bone cancer pain of rat models, and its effect intensity is similar to that of gabapentin. Its analgesic mechanism may be associated with suppressing the central excitatory amino acids, pain-associated neuromodulator and peripheral nerve growth factor and promoting IL-12-P70 in immunological reaction.

      摘要:Objective: To discuss the mechanism of the method of supplementing Qi, nourishing Yin, activating blood circulation and dredging collaterals in regulating transforming growth factor-β1 (TGF-β1)/Smads signaling pathway in lung fibroblasts. Method: A total of 30 SPF male rats were divided into three groups by the method of random number table, namely traditional Chinese medicine (TCM) serum group, western medicine serum group and normal serum group. Serums were extracted at the end of the fourth day after administration. Bleomycin was intratracheally injected to make animal model of pulmonary fibrosis in rats, and the lung fibroblasts were cultured by trypsin digestion method. The cells were divided into model group, TCM group, western medicine group and inhibitor group. Lung fibroblasts were identified by immunofluorescence, and materials were collected at 24, 48, 72 h. Reverse transcription-polymerase chain reaction was used to detect the mRNA expressions of TGF-β1, Smad2, Smad4, and Smad7, and flow cytometry was used to detect apoptosis rate. Result: The results of 24, 48, 72 h showed that compared with model group, the mRNA expressions of TGF-β1, Smad2, and Smad4 in the TCM group, western medicine group and inhibitor group were obviously lower (P<0.01). The mRNA expressions of TGF-β1 in 24 h western medicine group was lower than that of TCM group (P<0.05). The results of 48, 72 h showed that compared with the TCM group, the mRNA expressions of TGF-β1 in the western medicine group and the inhibitor group were all obviously lower (P<0.05, P<0.01). The results of 24, 48 h showed that compared with the model group, the Smad2 mRNA expression in TCM group, western medicine group and inhibitor group were obviously lower (P<0.01). The expressions of TCM group, western medicine group and inhibitor group were similar, with no statistical significance. The results of 72 h showed that compared with the TCM group, the Smad2 mRNA expressions in the western medicine group and the inhibitor group were all obviously lower (P<0.05). The results of 24, 48, 72 h showed that compared with the TCM group, the Smad7 mRNA expressions in the western medicine group and the inhibitor group were all obviously lower (P<0.01). The results of 24, 48, 72 h showed that compared with the model group, the apoptosis rates of TCM group, western medicine group and inhibitor group were obviously increased (P<0.01). The apoptosis rate of TCM group was higher than that of inhibitor group (P<0.01). Conclusion: The method of supplementing Qi, nourishing Yin, activating blood circulation and dredging collaterals may delay the progress of idiopathic pulmonary fibrosis by decreasing the content of TGF-β1, Smad2 and Smad4, increasing the content of Smad7 and regulating the TGF-β1/Smads signaling pathway.

      摘要:Objective: To explore the effect of Xuefu Zhuyu capsule (XZC) in regulating Jagged1/Notch signal in angiogenesis. Method: XZC-containing serum and blank serum were used respectively to treat human microvascular endothelial cell-1 (HMEC-1). The mRNA expression of Jagged1 was detected by Real-time quantitative (PCR) at 12, 24, 36, 48 h, and the protein expression of Jagged1 was detected by Western blot at 12, 24, 48 h. The γ-secretase inhibitor DAPT was used to block the Notch signaling pathway under the optimal condition of promoting angiogesis by XZC. The cells were randomly divided into 3 groups, namely DAPT processing group, solvent control group and blank control group. The angiogenesis effect was tested by an in vitro angiogenesis assay. Result: XZC up-regulated the mRNA expression of Jagged1 after culturing HMEC-1 cells for 12 h (P<0.01), and the mRNA and protein expressions of Jagged1 were up-regulated at 24 h (P<0.05). The ability of angiogenesis in vitro was inhibited after the Notch signaling pathway was blocked by DAPT. Conclusion: XZC can regulate the expression of Jagged1, and Jagged1/Notch signaling pathway may be correlated with XZC's effect on regulating angiogenesis.

      摘要:Objective: To analyze the pharmacological effect and action mechanism of Shuangyi prescription (SYP) in the treatment of type 2 diabetes rats. Method: High fat diet plus 25 mg · kg-1 streptozotocin were intraperitoneally injected to establish type 2 diabetic rats model. The model animals were randomly divided into model group, metformin group (ig, 85 mg · kg-1), SYP high dose group (ig, 2 000 mg · kg-1), SYP middle dose group (ig, 1 000 mg · kg-1), and SYP low dose group (ig, 500 mg · kg-1). Body weight, food intake, water intake, urine, stool, blood glucose, oral glucose tolerance test (OGTT), hemoglobin A1c (HbA1c), glycosylated serum protein (GSP), and insulin of type 2 diabetes rats were studied; Western blot was used to detect protein tyrosine phosphatase 1B (PTP1B), Akt, p-Akt, glycogen synthase kinase-3β (GSK-3β), p-GSK-3β expressions in liver tissues; Western blot to detect insulin receptor substrate-1 (IRS-1), p-IRS-1, glucose transporter 4 (Glut4) expressions in skeletal muscle tissues. Result: SYP can reduce fasting blood glucose (FBG), HbA1c level and OGTT of type 2 diabetes rats, reduce insulin resistance index. Compared with the model group, SYP can alleviate polydipsia, polyphagia, polyuria, and weight loss conditions in type 2 diabetic rats in each dose group (P<0.05, P<0.01), SY can reduce insulin, glycated serum protein and random blood glucose (P<0.05, P<0.01), the effect of SYP was better in the high-dose group; SYP can significantly increase p-IRS-1 and GLUT4 expressions (P<0.01,P<0.05), reduce the expression of PTP1B (P<0.01, P<0.05) in skeletal muscles of type 2 diabetic model rats; SY can significantly increase p-Akt expression, and significantly reduce PTP1B, p-GSK-3β expressions (P<0.01, P<0.05) in liver of type 2 diabetic model rats. Conclusion: SYP can effectively regulate glycometabolic disorders in type 2 diabetes rats, improve insulin resistance. The regulation of skeletal muscle PI3K/Akt and liver Akt/GSK-3β signaling pathway may be the mechanism of SY in the treatment of type 2 diabetes.

      摘要:Objective: To study the effect of Chuankezhi injection on lipopolysaccharide (LPS)-induced acute lung injury (ALI) in mice and its mechanism. Method: The 48 SPF grade female C57BL/6J mice were randomly divided into normal group, model group, dexamethasone group, and Chuankezhi group, a total of 4 groups, 12 mice in each group. Each mouse was intraperitoneally injected with 0.2 mL · d-1 in Chuankezhi group, once a day for 7 days. The mice in the dexamethasone group were injected intraperitoneally at 1 mg · kg-1 per day for 3 days before modeling. The mice in model group were injected with equal dose of normal saline while the normal group had no treatment. Except in the normal group, LPS 0.1 mL (0.5 g · L-1) was injected intraperitoneally on the 8 day, and the mice received 25 μL (0.5 g · L-1) by nasal drops after ether inhalation for anesthesia to establish ALI models. Detection was conducted after 24 h. The expression levels of tumor necrosis factor (TNF)-α, interleukin (IL)-1β and IL-18 were detected by enzyme-linked immunosorbent assay (ELISA). The expression level of nuclear factor-kappa B (NF-κB) p65 protein was detected by immunohistochemistry. Western blot was used to detect the expression levels of p65, Toll-like receptor4 (TLR4), Nod-like receptor protein 3 (NLRP3) and cysteine aspartate protease-1 (Caspase-1) protein in lung tissues, and Real-time polymerase chain reaction (Real-time PCR) was used to detect the expression of p65 and TLR4 mRNA in lung tissues. Result: As compared with the normal group, the levels of TNF-α, IL-1β and IL-18 in serum in the model group were increased (P<0.01); the expression of p65, TLR4, NLRP3 and Caspase-1 were increased; p65 and TLR4 mRNA were increased (P<0.05). As compared with the model group, the levels of TNF-α, IL-1β and IL-18 in serum in Chuankezhi group were significantly decreased (P<0.01), and the levels of p65, TLR4, NLRP3 and Caspase-1 protein and expression levels of p65 and TLR4 mRNA were also decreased (P<0.05). Conclusion: Chuankezhi injection can effectively improve the injury of lung tissues induced by LPS in mice with acute lung injury. The mechanism may be associated with inhibiting TLR4/NF-κB/NLRP3 signaling pathway to protect the lung tissues.

      摘要:Objective: To study the antitussive and antiasthmatic effect of Lingbei Zhike syrup and conduct the experimental study on reducing lung tissue's pathological apoptosis to reach the antiasthmatic effect. Method: The 48 guinea pigs were randomly divided into model group, positive control group (Chuanbei Xueli syrup, Dextromethorphan Hydrobromide group), Lingbei Zhike syrup low, medium and high dose groups (1.27, 2.55, 5.1 g · kg-1), n=8 in each group. Citric acid-induced cough experiment in guinea pig was used to investigate the antitussive effect of Lingbei Zhike syrup. The 60 guinea pigs were randomly divided into model control group, positive control group (Chuanbei Xueli syrup, aminophylline group), Lingbei Zhike syrup low, medium and high dose groups (1.27, 2.55, 5.1 g · kg-1), n=10 in each group. Acetylcholine-histamine phosphate-induced asthma experiment was used to investigate the antiasthmatic effect of Lingbei Zhike syrup. In 68 healthy mice, 50 g tobacco leaves and wood shavings were fumigated once respectively in the morning and evening, 30 min/time for continuous 7 days to establish bronchitis models. Hematoxylin and eosin (HE) staining was used to determine whether the models were successfully established. After successful modeling, immunofluorescence histochemistry (IHC) method was used to compare the blank control group, model control group, Chuanbei Xueli syrup group, and Lingbei Zhike syrup low, middle and high dose groups (2.14, 4.28, 8.56 g · kg-1) in lung tissue pathological apoptosis related protein B cell lymphoma/leukemia-2 (Bcl-2) and integral optical density value of factorassociatedsuicide (Fas), so as to investigate the effects of Lingbei Zhike syrup on apoptosis related protein Bcl-2 and Fas protein expression levels in lung tissues of bronchitis models. Result: Lingbei Zhike syrup could significantly prolong cough incubation period of guinea pigs and decrease the frequency of cough. HE staining showed that the mouse models of acute bronchitis were successfully established. Immunohistochemistry results showed that Lingbei Zhike syrup could significantly increase the expression level of Bcl-2 protein in lung tissues with inflammation and reduce the expression of Fas protein. Conclusion: Lingbei Zhike syrup could inhibit lung tissue's pathological apoptosis through regulating the expression of apoptosis-related proteins Bcl-2 and Fas, thus helping to restore the normal tissue structure and reduce lung tissue inflammation in playing antiasthmatic effects.

      摘要:Objective: To investigate the effect of Zuoguiwan on the neuro-endocrine-immune function of kidney-Yin deficiency rats. Method: Total 60 healthy Wistar male rats were randomly divided into normal group (same volume of normal saline), model group (same volume of normal saline), prevention group (at the same time of modeling, 1 g · kg-1 Zuoguiwan) and the low (after modeling, 0.5 g · kg-1 Zuoguiwan), middle (after modeling, 1 g · kg-1 Zuoguiwan) and high dose (after modeling, 2 g · kg-1 Zuoguiwan) groups of Zuoguiwan. Thyroid tablets were given by ig administration to establish kidney-Yin deficiency models; all groups received ig administration for continuous 21 days. Then their spleen, thymus, urine and blood samples were collected to calculate the organ index; the levels of corticotropin releasing hormone (CRH), adrenal corticosteroids hormone (ACTH), cortisol (CORT), thyroid stimulating hormone (TSH), triiodothyronine (T3), tetraiodothyronine (T4), cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), 17-hydroxycorticosteroids(17-OHCS), interleukin-6(IL-6), and interferon-γ (IFN-γ) of rats were detected by enzyme linked immunosorbent assay. Result: As compared with the normal group, T3, T4, cAMP and cGMP, IL-6, and IFN-γ were increased significantly; TSH, cAMP/cGMP, spleen index and thymus index were decreased in model group, with significant differences (P<0.05, P<0.01); all the indexes in prevention group had no significant difference. As compared with the model group, T3, T4, cAMP, cGMP, IL-6 and IFN-γ of each dose group of Zuoguiwan were significantly reduced, and TSH, cAMP/cGMP, spleen index, and thymus index were significantly increased, with significant difference (P<0.05,P<0.01). T3, T4, CRH, ACTH, CORT, cAMP, cGMP, 17-OHCS, spleen index, thymoid index in prevention group were significantly increased (P<0.01), while TSH, cAMP/cGMP, IL-6, IFN-γ was significantly reduced (P<0.01). As compared with the prevention group, T3, CRH, ACTH of each dose group of Zuoguiwan were higher (P<0.05, P<0.01). Conclusion: Zuoguiwan can adjust hypothalamus-pituitary-adrenal axis and hypothalamus-pituitary-thyroid axis function in rats with hyperthyroidism type kidney-Yin deficiency, and improve immune function. This could be one of the mechanisms for treating hyperthyroidism type kidney-Yin deficiency.

      摘要:Objective: To explore the protective effect of essential oil from fructus Alpinia zerumbet(EOFAZ) on endothelial-mesenchymal transition (EndMT) in human umbilical vein endothelial cells(HUVECs) induced by transforming growth factor-β1(TGF-β1). Method: HUVECs were cultured in vitro and the EndMT cell model was reproduced by 10 μg · L-1 TGF-β1 incubated for 48 h.The experiment was divided into four groups,such as normal group,model group(10 μg · L-1 TGF-β1),EOFAZ high dose group (10 μg · L-1 TGF-β1+4 μg · L-1 EOFAZ) and EOFAZ low dose group (10 μg · L-1 TGF-β1+2 μg · L-1 EOFAZ).After pre-incubation with EOFAZ (4,2 μg · L-1) for 2 h,the EndMT cell model was reproduced.The cell morphology was observed under inverted microscope,and the cell migration ability was analyzed by scratch test.Western blot was used to detect the specific protein expression of vascular endothelial cadherin(VE-cadherin),α-smooth muscle actin(α-SMA),zinc finger transcription factor(Snail) and nuclear transcription factor-κB(NF-κB) p-p65. Result: After incubation with TGF-β1,the cell morphology of HUVECs was long spindle shape with visible cells of filamentous pseudopodia,the close connection between cells were reduced,and the cell migration ability of the model group was significantly increased(P<0.01),and the protein expression of VE-cadherin were down-regulated significantly(P<0.01).On the contrary,the expression of α-SMA,Snail and NF-κB p-p65 was significantly increased(P<0.01).Pretreatment with high dose of EOFAZ,the cell migration ability was significantly inhibited;meanwhile,the expression of VE-cadherin was significantly increased(P<0.01),and expression of α-SMA,Snail and NF-κB p-p65 protein were significantly decreased(P<0.01). Conclusion: EOFAZ has protective effect on EndMT in HUVECs induced by TGF-β1,and the mechanism is related to inhibition of phosphorylation of NF-κB p65.

      摘要:Objective: To explore the clinical effect of modified Juanbitang on patients with wind-cold-wetness type rheumatoid arthritis (RA) and its effect on inflammatory factors and quality of life. Method: The 134 cases with RA treated at our hospital from January 2015 to January 2017 were selected as research objectives, and randomly divided into two groups. The same nonpharmacological treatment was provided to both groups. The control group was additionally given meloxicam+methotrexate, and the observation group was treated with modified Juanbitang. Two groups' traditional Chinese medicine (TCM) syndrome, physical and chemical indicators before and after treatment, synovial fluid inflammatory factor level, quality of life, serum matrix metalloproteinase-3 (MMP-3), bone protection element osteoprotegerin (OPG) and receptor activator for nuclear factor-κB ligand (RANKL) expression were recorded and compared. Adverse reactions were also observed. Result: After treatment for 12 weeks, the overall effective rate of TCM syndrome in observation group was 94.0%, which was significantly higher than 82.1% in control group. The observation group was superior to control group. The two group's erythrocyte sedimentation rate erythrocyte sedimentation rate (ESR), serum C-reactive protein (CRP), rheumatoid factor (RF) after treatment for 12 weeks were significant lower than those before treatment (P<0.05), and those in observation group were decreased more significantly (P<0.05). Two groups' joint fluid tumor necrosis factor (TNF)-α, and interleukin interleukin-17 (IL-17) after treatment for 12 weeks were significant lower than those before treatment (P<0.05), and those in observation group were decreased more significantly (P<0.05). Two groups' physical component summary (PCS) and mental component summary (MCS) after treatment for 12 weeks were significant higher than those before treatment (P<0.05), and those in observation group were increased more significantly (P<0.05). Compared with before treatment, MMP-3 and RANKL levels of two groups reduced, while OPG increased after treatment (P<0.05). Compared with control group, RANKL level of treatment group was significantly decreased after treatment, with statistically significant differences (P<0.05). Conclusion: Modified Juanbitang can relieve or eliminate the symptoms and signs of patients with wind-cold-wetness type RA, reduce the systemic inflammatory response, delay the disease progression, and improve the life quality, with a good clinical effect.

      摘要:Objective: To study the clinical efficacy of Liu Junzitang and its effect on lung function, exercise tolerance and blood gas analysis in patients with chronic obstructive pulmonary disease (COPD) with symptoms of lung and spleen deficiency at stable stage. Method: A total of 106 cases of patients with COPD at stable stage treated in our hospital were selected and divided into two groups by the random number table, with 53 cases in each group. Patients in both groups were treated with salmeterol and fluticasone propionate powder for inhalation, while treatment group was additionally given Liu Junzitang. The course of treatment was 12 weeks. Then, the changes in lung function, exercise tolerance, respiratory muscle fatigue, values of serum-BNP and inflammatory mediators in two groups were compared. Result: In terms of clinical efficacy, compared with control group (83.02%), treatment group showed the total effective rate of 96.23%. There was a significant increase (P<0.05). In effect on lung function, forced expiratory volume in the first second (FEV1), forced vital capacity (FVC) and FEV1/FVC showed no difference between two groups. There were significant differences between two groups of patients before and after 4, 12 weeks treatment (P<0.05) in FEV1, FVC and FEV1/FVC improvement. And the values of FEV1, FVC and FEV1/FVC in treatment group is superior to those of control group (P<0.05). Regarding exercise tolerance (6 mWD), the value of 6 mWD before treatment between two groups showed no difference. There were significant differences between two groups of patients before and after 4, 12 weeks treatment (P<0.05). And the value of 6 mWD in treatment group was superior to that of control group (P<0.05). In terms of blood gas analysis (PaO2, SaO2, PaCO2), there were significant differences between two groups of patients before and after 4,12 weeks treatment (P<0.05). And the values of PaO2, SaO2, PaCO2 in treatment group were superior to those of control group (P<0.05). The value of serum-BNP before treatment between two groups showed no difference. There were significant differences between two groups of patients before and after 4, 12 weeks treatment (P<0.05). And the value of serum-BNP in control group was superior to that in treatment group. In terms of inflammatory mediators, values of IL-6, IL-8 and hs-CRP before treatment between two groups showed no difference. There were significant differences between two groups of patients before and after 4 and 12 weeks treatment (P<0.05). And the values of IL-6, IL-8 and hs-CRP in control group were superior to those in treatment group. Conclusion: Liu Junzitang has a good therapeutic effect on COPD with symptoms of lung and spleen deficiency at the stable stage. Liu Junzitang can significantly improve lung function and exercise tolerance, and relieve inflammatory reaction.

      摘要:Objective: To study the effect of Maxing Shigan Tang combined with azithromycin on anti-inflammatory and pro-inflammatory cytokines and immune function of children with mycoplasmal pneumoniae pneumonia (MPP). Method: A total of 165 cases of children with MPP treated at our hospital from November 2014 to April 2016 were selected and divided into two groups according to the random number table, with 83 cases in clinical group and 82 in control group. Patients in control group were treated with azithromycin sequential therapy, while clinical group was given Maxing Shigan Tang combined with azithromycin sequential therapy. Result: The total effective rate of clinical group was 93.98%, which was 81.71% in control group, indicating that clinical group was superior to control group (P<0.05). Compared with before treatment, interleukin (IL)-4 and IL-10 of two groups were decreased, while IL-2 was increased after treatment (P<0.05). Compared with control group, the levels of IL-4 and IL-10 in patients of clinical group were lower, while IL-2 was higher after treatment (P<0.05). The levels of IL-6, tumor necrosis factor-α(TNF-α) and interferon-γ (IFN-γ) of two groups showed significant differences before and after treatment. The level of inflammatory factor in the clinical group was lower than in the control group (P<0.05). Compared with before treatment, CD8+and RBC-ICR of two groups were decreased, while CD3+, CD4+and RBC-C3bR were increased after treatment (P<0.05). Compared with control group, the levels of CD8+and RBC-ICR in patients of clinical group were lower, while CD3+, CD4+and RBC-C3bR were higher after treatment (P<0.05). Compared with control group, TCM syndrome score of clinical group was lower, with statistically significant differences (P<0.05). Conclusion: Maxing Shigan Tang is very effective in treating MPP children. It can significantly improve anti-inflammatory factors, reduce pro-inflammatory cytokines, and improve the immune function.

      摘要:Objective: To discuss the clinical effect of Bufei Jianpi Yishen decoction ointment on chronic obstructive pulmonary disease at stable period, and its effect on nutritional status, immune function and inflammatory factors. Method: One hundred and thirty-six patients with COPD were randomly divided into control group and observation group by random number table. Patients in control group got ipratropium bromide aerosol, 40-80 μg/time, 2-4 times/day. And Salmeterol inhalation, 1 suck/time, 2 times/day. In addition to the therapy of control group, patients in observation group was also given Bufei Jianpi Yishen decoction, 15 g/time, 3 times/day. And 24 weeks were a course of treatment. The symptoms were evaluated by Modified British Medical Research Council (mMRC), chronic obstructive pulmonary disease self-assessment test (CAT) and traditional Chinese medicine (TCM) syndrome scale before treatment, and at the 12th and 24th. And before and after treatment, forced expiratory volume in the first second (FEV1), forced vital capacity (FVC) and proportion of FEV1 to expected value (FEV1%) were detected. And scores of St. George's Respiratory Questionnaire (SGRQ), 6 minute walk test (6 mWD) and mini nutritional assessment (MNA) were evaluated. Besides, T lymphocyte subsets (CD4+, CD8+ and CD4+/CD8+), immunoglobulins (IgG, IgM and IgA), inflammatory factors [interleukin-8 (IL-8), IL-4, tumor necrosis factor-α (TNF-α)], serum total protein (TB), albumin (ALB), prealbumin (PAB) and Hemoglobin (HB) were detected, 1 times were detected before and after treatment. Result: The total effect rate of TCM symptoms in observation group was 93.85%, which was higher than 80.65% in control group (χ2=5.022, P<0.05). At the 12th and 24th week, scores of mMRC, CAT and TCM symptoms in observation group were lower than those in control group (P<0.01). After treatment, FEV1%, FEV1 and FVC were all higher than those in control group (P<0.05). Breathing symptoms, scores of activity ability, disease effects and total score of SGRQ were all lower than those in control group, and 6 mWD was longer than that in control group (P<0.01). After treatment, score of MNA was higher than that in control group, and levels of TB, ALB and PAB and CD3+, CD4+, CD4+/CD8+ were higher than those in control group, and levels of CD8+ and IL-8, IL-4 and TNF-α were lower than that in control group (P<0.01). After treatment, levels of IgG, IgM and IgA in observation group increased, which were higher than those in control group (P<0.05). Conclusion: Bufei Jianpi Yishen decoction can ameliorate nutritional status, improve lung function and immune function, relieve inflammation and clinical symptoms, increase the quality of life of the patients and exercise capability, with a better clinical effect than western medicine therapy.

      摘要:Objective: To observe the clinical effect of modified Guizhitang on children with vitiligo (Qi-blood circulation) andits effect onlevels of T lymphocyte subsets, immunoglobulin and alexin. Method: One hundred and thirty-four patients were randomly divided into control group 67 cases and observation group 67 cases by random number table. Patients in control group were given 0.03% tacrolimus ointment, 2 times/day. In addition to the therapy of control group, patients in observation group were also given modified Guizhitang, 1 dose/day. And 3 months were a course of treatment. Before and after treatment, vitiligo area score index (VASI), Melanin content index (MCI), survey scale of quality of life in skin disorder (DIQL), and score of Qi-blood circulation were graded, and color recovery was recorded. Levels of CD3+, CD4+, CD4+/CD8+ and CD8+ for T lymphocyte subsets, C3,C4 foralexin, IgG, IgM and IgA for immunoglobulin were detected. Result: The total effect rate in observation group was 84.13%,which washigher than 63.49% in control group (χ2=6.938, P<0.01). After treatment, VASI, scores of Qi-blood circulation and DIQL in observation group were lower than those in control group (P<0.01), and MCI was higher than that in control group (P<0.01). Rate of color recovery was (62.83±11.47), which was higher than (53.78±10.29) in control group (P<0.01). Compared with healthy control group,among children with vitiligo, levels of IgG and IgA, CD3+, CD4+ and CD4+/CD8+ decreased, level of CD8+ increased (P<0.01).With no statistical significance among levels of IgM, C3 and C4.After treatment, levels of IgG and IgA in observation group increased, which were higher than those in control group (P<0.01). And levels of CD3+, CD4+, CD4+/CD8+ in observation group increased(P<0.01), which were higher than those in control group (P<0.05). And level of CD8+ decreased, whichwas lower than that in control group (P<0.05). Conclusion: Modified Guizhitang can reduce the area of skin lesion, increase the white melanin content,rate of color recovery rate and ameliorate quality of life of patients, with a significant clinical efficacy. Its mechanism may be correlated with the regulation of autoimmune imbalance, and thus is worthy for further research.

      摘要:Objective: To explore the potential mechanism of Dracaenea Cochinchinensis Resina in the treatment of ischemic stroke. Method: Online software of Arrowsmith based on the theory of Swanson's non-correlated literature-bases knowledge discovery was applied to explore the underlying mechanism by analyzing the intermediate words in the literature on Dracaenea Cochinchinensis Resina and ischemic stroke. Result: The preliminary investigation results by Arrowsmith software showed that Dracaenea Cochinchinensis Resina and its active components might have certain ability to regulate the expression levels of cyclooxygenase-2 (COX-2), transient receptor potential vanilloid 1 (TRPV1), nitric oxide synthase (NOS), voltage-gated potassium channel 1.3 (Kv1.3) and transforming growth factor-β1 (TGF-β1); and these above indexes had abnormal expression in ischemic stroke. Conclusion: The mechanism may be associated with down-regulating the expression of COX-2 and iNOS, blocking TRPV1 and Kv1.3 channel, and promoting the expression of eNOS and TGF-β1.This can provide strong reference for the laboratory to conduct further mechanism verification studies.

      摘要:The toxicity of traditional Chinese medicine (TCM) is an objective existence. At present, most of the toxic mechanism of TCM is unclear, and the research is still at the phase of exploration and accumulation at home and abroad. To meet the modern development of TCM, it is very important to establish the objective experimental data of toxic grades and the safety evaluation model. Genomics, proteomics and metabolomics have been increasingly applied in identifying TCM toxic markers and toxic mechanism. Besides, in vitro cytotoxicity evaluation, as an alternative method for evaluating traditional toxicity with animal models, has been widely studied and applied. It also has a distinct advantage in diagnozing early, slight, hidden toxicity. Besides, the quantitative automation high throughput detection and screening based on human cells or cell lines have helped improve the toxicology detection efficiency. The preponderance, particularity, evaluation index and detection methods for in vitro cytotoxicity tests are reviewed to discuss the existing problem, with the aim to develop and improve the research system of TCM toxicity.

      摘要:Chronic complications of diabetes are mainly macrovascular disease and microvascular disease, in which microvascular disease is a diabetic-specific complication, with microcirculation disturbance and microvascular basement membrane thickening as the typical lesions. Specifically, heart microvascular disease and myocardial metabolic disorders can cause extensive myocardial necrosis, which is known as diabetic cardiomyopathy. The pathogenesis of diabetic cardiomyopathy is very complex, involving many aspects, including myocardial fibrosis, myocardial glucose and lipid metabolism, oxidative stress, cytokine abnormalities, insulin resistance, mitogen-activated protein kinase (AMPK) protein kinase and its associated protease signaling pathway, mammalian target of rapamycin (mTOR) signaling pathway, and NF-E2-related factor 2-antioxidant response element (Nrf2-ARE) signaling pathway. Traditional Chinese medicine(TCM) doctors mainly consider in the aspects of activating blood and unblocking collaterals, and focus on single herb and TCM compounds. A large number of animal experiments and clinical data show that TCMs have a good effect in the treatment of this disease. It is a very meaningful to bring TCM's characteristic of treatment based on syndrome differentiation, deeply dig Chinese herbal medicine resources, and apply its characteristics of composition and multiple dosage forms in the treatment of diabetic cardiomyopathy. This article reviews the pathogenesis of diabetic cardiomyopathy and the studies for prevention and treatment of TCM, and provides a reference for clinical prevention and treatment of this disease.

      摘要:Apocynaceae Melodinus plants have a long history in folk use in China, and their chemical compositions and bioactivities are concerned by domestic and abroad scholars. As the important effective ingredients, more than 200 alkaloids with novel and complex structures have been isolated from Melodinus genus. According to the modern pharmacological studies, alkaloids of the Melodinus genus show good antitumor, antibacterial and antifertility activities. The studies on alkaloids of Melodinus genus were summarized according to the domestic and foreign literature in the past 30 years. The indole and quinoline alkaloids and their bioactivities were reviewed from M. hemsleyanus, M. tenuicaudatus, M. fusiformis, M. henryi, M. axillaris, M. khasianus, M. yunnanensis and M. suaveolens respectively to provide reference for further development of Melodinus plants.

      摘要:Hederagenin is a pentacyclic triterpenoid isolated from plants. It is widely distributed in a variety of medicinal plants and is the main active ingredient of many natural products. The latest research progress on resource distribution, structural modification and pharmacological action of saponins of hederagenin was reviewed by integrating and analyzing the related literature at home and abroad, which provided the reference for further study on the structural modification and pharmacological action of hederagenin. Studies have shown that hederagenin is widely distributed in Dipsacaceae, Caprifoliaceae, Ranunculaceae, Araliaceae, Patrinia and other medicinal plants, rich in the natural resources. Hederagenin has a poor solubility, low bioavailability, poor oral administration effects and other characteristics, so that the clinical application of hederagenin is limited. Therefore, domestic and foreign scholars have made structural modifications which mainly occur in its C-28 position, C-3 position, C-23 position, C-12 position and C-13 position. The structural modification is more in C-28 position. Pharmacological activity studies have shown that hederagenin has anti-tumor, anti-depression, anti-bacterial, anti-inflammatory, anti-diabetes and other pharmacological effects, but its related mechanism is still lack of in-depth study. Through the systematic review of the resource distribution, structural modification and pharmacological action of the hederagenin, it can provide a reference for the deep research of hederagenin and provide a scientific basis for the safe and effective application of hederagenin and its derivatives.
