摘要:Objective:To develop the Qualitative and Quantitative Methods of Xuesaitong dropping pills.Methods:TLC method was used for identification.Ginsenoside Rg1、Ginsenoside Rb1 and notoginseno side R1 in dropping pills were determinded by HPLC.The assay was carried out on an ODS column,eluted with acetonitrile-water in the gradient mode.The detection wavelength was set at 203 nm,the flow rate was set at 1.0 mL·min~(-1),and the column temperature was room temperature.Results: The major components were well identified.For ginsenoside Rg1 and ginsenoside Rb1,the linear ranges were from 0.2 μg to 1.0 μg,and for notoginsenoside R1,the linear range was from 0.1 μg to 0.5 μg,and total average recovery was 97.93%.Conclusion: The methods were feasible,reproducible and can be applied to the quality control of Xuesaitong dropping pills.  
摘要:Objective:To develop a capillary electrophoresis method for determination lovastatin in capsule of monascus.Methods:Capsule of monascus was extracted with methanol.Separation was carried out in an uncoated fused silica capillary(50 μm×60.0 cm).with a running buffer containing 100 mmol·L-1 Tris,0.03% ammonia in methanol.The running voltage of 27 KV and the column temperature was maintained at 20 ℃.The UV detector wavelength was set at 238 nm.Results:Regression equation of lovastatin was Y=3.08×106X-2.08×103(r=0.999 8).The linearity was obtained over the range of 14.625 μg~468 μg.The relative recovery ranged from 96.5%~99.7%.Conclusions:The method is proved to be simple,rapid,accurate and repeatable,and it can be used to determine lovastatin in capsule of monascus.  
摘要:Objective:To study the technological parameters of the purification process of hyperoside from Hypericum perforatum extrative.Methods: HPLC was used to analyze the content of hyperoside in the samples from the different technological process,and the adsorption and elution parameters were investigated.Results: The optimal parameters were as follows:the sample extract solution,whose content of hyperoside was 1.2 mg·mL-1,was passed through the packed column with a flow rate of 3.6 BV·h-1.The adsorption rate of hyperoside was 2.33 mg·g-1,and the purity of hyperoside in the eluate reached 28.96%.Conclusion: Polyamide resin can be used to purify hyperoside in the extraction of Hypericum perforatum L.  
摘要:Objective:To optimize the technological condition of Ethanol percolating for Xiaoqinglong Patch.Method: The content of Ephedrinehydrochloride and Paeoniflorin extracted,which were regarded as chemical markers,were assayed by HPLC.Furthermore basis of initial research,the general content data determined were used to optimize the percolating extraction processing conditions by orthogonal Sampling design.Result: The optimum extraction condition was that the medicinal materials was soaked with 55% alcohol for 12h,then was percolated with 10 times amount of 55% alcohol and the percolating speed was 30 mL·min-1·kg-1.Conclusion: The content of Ephedrinehydrochloride and Paeoniflorin extracted by the the extraction process,were highest,and the optimiazed process is stable and efficient.  
摘要:Objective:To investigate the effect of water extracts of Retro-Zuojin Pill(WERZP) on proliferation and apoptosis of human gastric carcinoma cells (SGC-7901),and the relationship between apoptosis and bcl-2,bax expression.Methods: The cell viability was measured by MTT;the apoptotic rate was investigated by FCM and the morphology was observed by Hoechst 33258 staining.Detection of bcl-2,bax gene and protein expression of WERZP-treated gastric cancer cells were carried out with Western blot method.Results: After treatment with WERZP for 24 h,the proliferation of SGC-7901 cells was significantly inhibited(P<0.05).Inhibited cell proliferation and induced cell death were in a dose-dependent and time-dependent manner,with apoptotic rate of(4.02±1.33),(36.10±0.23),(55.91±0.95),(56.50±1.55) and(60.27±3.58) % accordingly.All the concentrations except the lowest induced significant apoptosis compared with the control(P<0.01).The nucleuses were positively stained with fluorochrome Hoechst 33258.WERZP could reduce the bcl-2 gene expression and increase gene expression of bax.Conclusion: WERZP could significantly inhibit the growth of SGC-7901 cells and induce apoptosis,reduce the bcl-2 gene expression.The increased gene expression of bax may be involved in apoptosis mechanism.

摘要:Objective:To study the effect of active parts in Datura metel L.(APD) for psoriasis on immune function,skin keratinized and epithelial cells mitosis,to clarify the mechanism for clinical applications and developing new drug.Methods:APD's effects on immune function were measured by the content of mice's hemolysin and lymphocyte transformation stimulated by phytohemagglutinin(PHA).The influence on the activity of vaginal epithelial mitotic cells and the effect on skin scales keratosis at the back of tail were also obsevered.Results: APD showed a trend of lowering the content of hemolysin in normal mice;and had no significant impact on lymphocyte transformation stimulated by PHA.APD could significantly inhibit the mitosis of vaginal epithelial cells;and obviously increace the quantities of scaly particles layers of mice tail with a clear role in the promotion of skin keratosis.Conclusion: APD may have no obvious effect on humoral and cell-mediated immunity to the experimental animals.However,APD appears to inhibit with epithelial cell mitosis and promote the keratosis of skin scales obviously.

摘要:Objective:To investigate the absorption characteristic and the influencing factors on absorption of protein and peptide from natural hirudin administered by nasal mucosa.Method: The toxicity of hirudin to the nasal cilia was tested by the ex vivo bufo palate model.The absorption characters of natural hirudin as well as the concentrations of natural hirudin and pH values of medium were evaluated by the experiments based on Valia-Chien infiltration pool method and by the in vivo perfusion experiments in rats.Result: The experiments based on Valia-Chien infiltration pool method and the in vivo perfusion tests in rats indicated that the natural hirudin was well absorbed per nasal mucous membrane from pigs and rats.It also indicated that both the concentrations of hirudin and the pH values of the medium influenced the absorption of hirudin.Though the concentration of hirudin has no obvious linear relationship with osmotic coefficient or rate of absorption of hirudin per nasal mucous membrane,it showed that the best absorption concentration is 0.6 mg·mL-1.Conclusion: Hirudin has low cilia toxicity;it can be well absorbed per nose mucous membrane.Both of the concentrations of natural hirudin and pH values of the medium influence the absorption characteristic of hirudin.

摘要:Objective:To observe effect of Zhenwu decoction on renal function in the chronic renal failure(CRF) rats.Methods:Rat model of Chronic renal failure was established by adenine administration after unilateral nephrectomy.Zhenwu decoction group gavaged by Zhenwu decoction for 28 days.Body weight,24 h urine and 24 h urinary protein were measured in every seven days.After 4 weeks,blood samples were taken from the femoral artery.RBC,Hb,serum creatinine,blood urea nitrogen,serum calcium and phosphorus were detected. Results:Zhenwu decoction group better than model group in 24h urinary protein,serum creatinine,blood urea nitrogen, serum phosphorus,Serum Calcium.Conclusions:Zhenwu decoction has a protective effect on renal function in rats with chronic renal failure induced by adenine after unilateral nephrectomy.  
摘要:Objective:To investigate the effects of Xiaoyanlidanpian(XLP) on mice model of experimental hepatitis and acute inflammation.Methods:The experimental hepatitis model was established by injection of Propionibacterium acnes and lipopolysaccharide(P.acnes-LPS) via lateral tail vein.Hepatic function was evaluated by assessing Alanine aminotransferase level(ALT) in plasma.The effect of Xiaoyanlidanpian on allergic contact dermatitis (ACD) induced by DNFB was studied in mice to observe its effect on delayed-type-hypersensitivity(DTH).Croton oil and Carrageenan were used as well to observe its effect on acute inflammation.Results: Compared to the model group,Xiaoyanlidanpian can inhibite the activities of ALT in mice plasma.Meanwhile,Xiaoyanlidanpian could ameliorate the ear swelling in ACD model and acute inflammatory model induced by croton oil.It also reduced the thickness of the mice hind paw induced by carrageenan as well.Conclusion:Xiaoyanlidanpian exerted potential effects on experimental hepatitis.Xiaoyanlidanpian also show partial effect on acute inflammation.  
摘要:Objective:To investigate the anti-inflammatory mechanism of total glucosides of Cape Jasmine(TGCJ) on rats with adjuvant arthritis.Methods: Rats were randomly divided into the normal group,the model group,the GTW group,and the TGCJ groups of small,medium,large dosages(20,40,80 mg·kg-1).Except for the normal group,animals were modeled to form adjuvant-induced arthritis with Freund's complete adjuvant.The arthritis index(AI) and the swelling degree of paws were recorded,and the activity of IL-1β,TNF-α and the levels of NO,NOS and PGE2 in joint synovium of secondary arthritis were determined.Results: Compared with the normal group,the levels of NO,NOS,PGE2 and the activity of IL-1β and TNF-α in joint synovium of secondary arthritis were increased significantly in the model group.Compared with the model group,TGCJ showed a dramatic inhibitory effect on secondary reaction of adjuvant arthritis,and the levels of NO,NOS,PGE2 and the activity of IL-1β and TNF-α in joint synovium of secondary arthritis were significantly decreased.Conclusions: TGCJ has a remarkable treatment effect on arthritis.Inhibiting the activity of cytokines and decreasing the level of inflammation media in inflammatory region may be involved in the mechanisms of the therapeutic effects of TGCJ.  
摘要:Objective:By uric acid sodium(MUS)-induced acute gouty arthritis model,to observe Biqing Capsule treatment for acute gouty arthritis.Methods: The IL-12 content in joint fluid of ankle was measured by ELISA method.Mouse ankle COX-2 expression was determined by immunohistochemical methods.Results:Biqing Capsule obviously decreased ankle IL-12 levels and restrained rat ankle COX-2 expression.It indicated that Biqing Capsule for acute gouty arthritis had some anti-inflammatory effects.Conclusion: The Biqing Capsule's anti-inflammation and analgesia mechanism may be related to decreased ankle IL-12 level and restrained ankle COX-2 expression.  
摘要:Objective:To explore the mechanism of the anti-tumor effect of Aidi Injection.Methods:Human lung squamous cell carcinoma xenografts were transplanted into 30 nude mice.Mice were randomLy divided into 3 groups:The Aidi Injection group received Aidi Injection(12 g·kg-1),0.2 mL,peritoneal injection) every day for 20 days,the chemotherapy group received DDP(1 mg·kg-1),0.2 mL peritoneal injection) every day for 7 days,and the control group received normal sodium(0.2 mL peritoneal injection) every day for 20 days.They were sacrificed afterwards and their tumors were processed for histological examination.The weights of the tumors were measured,the vessel density of the tumor and the expression of VEGF in tumors were detected using immunohistochemical method.Results:The cancer inhibition rates of Aidi Injection and DDP are 45.6% and 78.3% accordingly.In the Aidi Injection group,the vessel density of the tumor and the expression of VEGF in tumors decreased.In the DDP group,the vessel density of the tumor decreased,while the expression of VEGF in tumors increased.Conclusion: Aidi Injection can inhibit the expression of VEGF in tumor and decrease the vessel density of the tumor.

摘要:Objective:To explore the preventive and therapeutic mechanism of BX decoction on reflux esophagitis on the basis of esophageal movement.Methods:Sixty rats were randomly divided into 4 groups: sham operation group, model group,cisapride group and BX group.Gastrointestinal reflux models in rats were made.Physiograph was used to observe the contraction of esophageal muscle strips.An RT-PCR based assay was used to detect mRNA expression for calponin and caldesmon in esophageal smooth muscle.Intracellular free calcium was measured by laser scanning confocal microscopy.Results:Compared with that in model group,contractive range of esophageal muscle strips in BX group increased greatly,mRNA expression level for calponin and caldesmon decreased obviously and intracellular free calcium increased significantly.Conclusion: The preventive and therapeutic mechanism of BX decoction on reflux esophagitis may be due to enhancement of contraction of esophageal smooth muscle by downregulating of mRNA expression for calponin and caldesmon and increasing the intracellular free calcium.  
摘要:Objective:To study the function of promoting blood circulation to remove blood stasis and treating acute soft tissue injury(ASTI) for Fufangbinghong Soft Capsule(BHSC).Methods:Rat model of blood stasis was established.The impact of BHSC on hemorheology,thrombosis and platelet adhesion rate were observed.The model of ASTI was established by hitting with a weight to observe the influence of BHSC on injurying degree of rats and histopathology.Results: BHSC could reduce plasma viscosity,blood viscosity,the length and weight of thrombus, platelet adhesion rate and improve symptoms of ASTI.Conclusion: BHSC can promote blood circulation to remove blood stasis and treat ASTI.

摘要:Objective:The purpose of this study is to explore the effects of four active components from traditional herbs including tetramethylpyrazine,puerarin,tanshinone ⅡA and paeonol on the proliferation and collagen synthesis of vascular adventitial fibroblasts(VAF) induced by angiotensin Ⅱ(ANG-Ⅱ).Methods:MTT assay was used for observing cell proliferation,and hydroxyproline chromatometry was used for detecting collagen synthesis.Results:These four herbs significantly decreased the proliferation rate and the amount of hydroxyproline of VAF components from respectively.Conclusions:Tetramethylpyrazine,puerarin,tanshinone ⅡA and paeonol could decrease the proliferation and collagen synthesis of VAF.

摘要:Objective:To observe the effect and the mechanism of visceral hypersensitivity model rats.Methods:The visceral hypersensitivity model in rat was established by intrarectal administration of acetic acid.The substance P in the colon sub-mucosa layer and 5-serotoninergic neurons/fibers were observed. 摘要:Objective:To investigate the analgesic,anti-inflammatory effects and the chemical constituents of alcohol extract of Shufu.(AVA) Methods:The analgesic effect of AVA were tested by counting body twisting induced by HAc in mice and the anti-inflammatory effects of AVA were tested by using the Colorimetric COX Inhibitor Screening Assay.The constituents were isolated and purified by chromatographic methods.Their structures were identified by physicochemical properties and spectral analysis.Results:AVA could delay the writhing time and reduce the writhing times in mice. AVA could inhibit the COX-1 and COX-2 enzymes activitres.Seven compounds were isolated and the stuctures were identified as β-Sitosterol,cholesterol,ergosterol,7-OH cholesterol,7β-OH cholesterol,octadecanoic acid and 9-octadecenoic acid.Conclusions:AVA has obvious analgesic and anti-inflammatory effects.  
摘要:Animal model of restraint stress shows some of the mental stress responses,and could be observed on entire animal level for multi-target studies.The model is widely applied in pharmacology,physiology and pathology.We introduce the history of restraint stress model,summarize the preparation and influential factors of modeling,and review the impacts induced by stress on the body as well as its mechanism.In addition,we also discuss the problems and prospect based on our experiments.
