摘要:Objective: To study and determine the Water-soluble substances of Salvia chinensis.Methods: The constituents of the n-Butanol-soluble portion of the water extractive were isolated and purified by means of Sephadex LH-20 column chromatographic methods.HPLC was used to determine the contents.Results: Two compounds were isolated and identified as danshensu and rosmarinic acid.The linear range of danshensu was 1.48~7.40 μg.The average recovery was 102.8% and RSD was 1.04%;The linear range of protocatechualdehyde was 0.05~0.26μg.The average recovery was 101.5% and RSD was 0.72%.Conclusion: The method can provide useful references to quality control of Herba Salvia chinensis.Danshensu was firstly isolated from Herba Salvia chinensis.

摘要:Objective:To develop an HPLC method for determination of aloe-emodin,rhein,emodin,chrysophanol and physcion in QING RE JIE DU entract.Methods: The analysis was carried out on a Kromasil C18(250 mm×4.6 mm,5 μm)column eluted with a mobile phase of methanal-water containing 0.2% phosphoric acid solution(V∶V=85∶15).The flow rate was 1.0 mL·min-1 and column temperature was 35 ℃.The detection wavelength was set at 254 nm.Results: The calibration curve was linear within the range of 1.012 5~16.2,1.025~16.4,1.025~16.4,1.025~16.4 and 0.625~10 μg for aloe-emodin,rhein,emodin,chrysophanol and physcion,respectively.The average recoveries were 101.5%,99.2%,101.9%,101.5%,101.4% for the abore five conponents,respectively.Conclusion: This method is convenient,accurate and reproducible for determination of aloe-emodin,rhein,emodin,chrysophanol and phascion in QING RE JIE DU extraet.

摘要:Objective:To establish a method for the determination of effective components in Lingzhi Jiangtang Capsule.Methods: The contents of ginsenoside Rg1、Re、Rb1 and notoginsenoside R1 in Lingzhi Jiangtang Capsule were determined by HPLC.Results: The resolutions of the determined peaks were fine.The linearity ranges for ginsenoside Rg1,ginsenoside Re,ginsenoside Rb1 and notoginsenoside R1 were 3.86~23.16 μg(r=0.999 9),0.628~3.768 μg(r=0.999 9),3.74~22.44 μg(r=0.999 9) and 0.764~4.584 μg(r=0.999 9) respectively.The average recoveries were 100.18% with RSD 1.52%,100.22% with RSD 0.41%,100.51% with RSD 0.74%,and 99.05% with RSD 1.10% for ginsenoside Rg1,ginsenoside Re,ginsenoside Rb1,notoginsenoside R1 respectively.Conclusion: The quantitative method for determining the ingredient of Lingzhi Jiangtang Capsule is simple,feasible and repeatable,and is beneficial to quality control of Lingzhi Jiangtang Capsule.

摘要:Objective:To establish the qualitative and quantitative methods for Gubaoxinshen Tablets.Method: Soy bean,and epimedium herb were identified by TLC,and the content of genistein and icarrin were determined by HPLC.Result: The developed TLC sports were was fairly clear,the HPLC method showed good repeatability.The average recovery of genistein was 98.9% with RSD 0.9%,and the average recovery of icarrin the was 98.0% with RSD 1.0%.Conclusion: The method is simple,accurate and can becauseofon control quality of Gubaoxinshen Tablets.
摘要:Objective:To study the qualitative and quantitative methods of Shenbai concentrated pills.Methods: Danshen root and lightyellow sophora root in Shenbai concentrated pills were identified with the method of TLC.The contents of astragaloside Ⅳ and paeoniflorin were determined with the method of HPLC.Results: For TLC,the spots of the two herbal drugs were well separated and without interference.Quantitative analysis of HPLC showed that the average recovery of astragaloside Ⅳ was 100.82%(RSD1.73%) with the RSD values of precision,repeatability and stability tests were 2.79%,1.48% and 1.44%,respectively;and the average recovery of paeoniflorin was 99.83%(RSD2.34%) with the RSD values of precision,repeataibility and stability tests were 2.25%,2.16% and 1.60%,respectively.Conclusions: The methods set up by this study are accurate and easy to perform with the merits of good resolution,specificity and reproducibility.It could effectively control the quality of Shenbai concentrated pills.
摘要:Objective: To denvolop HPLC an method for the determination of the content of three saponins in Zhixiao suppositories.Methods: Analysis carriedout was performed on a Kromasil C18 column(4.6 mm×250 mm.,5 μm)at 23 ℃.The mobile phases were methanol and 0.2% phosphoricacid solution in a gradient elution.The flow rate was set at 1.0 mL·min-1 and the ultraviolet detector was operated at 203 nm.Results: The linear ranges of notoginsenoside R1,ginsenoside Rg1 and ginsenoside Rb1 were 0.368~2.76 μg,0.676~5.07 μg and 0.812~6.09 μg,respectively.The average recoveries of the three saponins were 98.53%,99.20% and 99.35%,respectively.The RSD(n=5)were 2.11%,1.88% and 2.38%,respectively.Conclusion: The separation is a good and the results are accurate and reproducible.It can be used for the quality control of Zhixiao suppositories.
摘要:Objectives:To observe the changes of the tongues in rats with blood-stasis syndrome induced mainly by cold.To explore the evidence that tongues were the target of blood stasis syndrome and the target of medication.Methods: Rats with blood-stasis syndrome induced by cold(BSC group) were dipped into ice water(0 ℃) for 5 minutes every day and lasted 20d individually.Different drugs were given orally after the model establishment.And then we took photos of tongues of all rats by digital camera,analyzed the gray scale value of all rats’ tongues using image analysis software,and observed the capillaries in tongues by electron microscope.Results: The tongues of rats which had been frozen in ice water for 20 d(once a day) were dark purple,the same as that of models induced by chemical materials.While as for the normal rats,the tongues were lustrous and ruddy.The changes of color of the tongues persisted about 1 week and were great obvious at the 3rd day after the model establishment among different time-points.After given different drugs,the degree of dark purple tongue degraded,showed significant difference(P<0.01) from that of model rats.The capillary stegnosis and nucleus turgescence of vascular endothelial cell were observed in tongues in BSC group by using electron microscope.After medication,the above changes recovered.Conclusion:The degree of dark purple tongue indicates the degree of diseases with blood-stasis syndrome.The tongue probably is a target of drug treatment.
摘要:Objective:To observe the therapeutic effects of effective compounds which were extracted from TongMai.Methods: SD rats experimential model with MsPGN were induced by subcutaneously injection rabbit anti-rat thymocyte serum(ATS) and forced swim.The rats were respectively ig with different dose(1.2,0.6,0.3 g·kg-1) of effective compound of TongMai for 4 weeks.Then the proliferative degree of mesangium was examined by HE stain,PAS stain and immunfluorescent respectively.Mesagium area proportion in glomerular capillaries area was calculated using image analyzing system.The expression of PCNA in the glomeruli was examined by immunohis to chemistry.Results: The effective compounds of TongMai can control the hyperplasia of the glomerular mesangial cells(GMC)and the mainifold of the mesangial matrix,and prevent the deposit of IgG in mesangium.The proportions in glomerular capillaries area in model control group were significantly higher than that in the treatment groups;The express of proliferative cell nuclear antigen(PCNA) were ascended either.Conclusion: The effective compound of TongMai can improve the damage s of pathology in glomerular.

摘要:Objective:To observe the changes of glycemia,urine protein,serum creatinine,blood urea nitrogen and glycosylated hemoglobin in diabetic nephropathy rats and treating effects of Xiaokekang.Methods: Streptozotocin(STZ) was abdominally administered and was feeded by high lipid diet to establish diabetic nephropathy model in rats.Animals were divided into three groups: model group,Xiaokekang treatment group and normal group.Changes in glycemia,serum lipids,serum creatinine,blood urea nitrogen were measured at the 8th,12th and 16th week after STZ injection.Glycosylated hemoglobin was measured at the 16th weeks after STZ injection.Results: Glycemia,serum lipids,serum creatinine were higher at the 8th,12th and 16th week compared with normal group.Serum creatinine was higher at the 12th and 16th week,and glycosylated hemoglobin was also highter than that of the normal group.Xiaokekang reduced these changes.Conclusion: Xiaokekang can reduce glycemia,serum lipids and protect renal function.
摘要:Objective:To determine the appropriate time of early administration of dopamine(DA) before nitroglycerin(GTN) for establishing rat model with vasomotor abnormalities.Methods: Rats were divided into 5 groups: normal group,DA Group and other 3 groups in accordance with DA at different time points(10,30,45 min) plus GTN.Common carotid artery(CCA) blood flow velocity was dynamically measured with pulsed Doppler flow probe to evaluate the influence of DA,GTN on blood flow velocity rate(BFVR).Results:The blood flow velocity rate induced by DA of All medication administration was different from the normal group within 10 minutes(P<0.05).However,the differences were not statistically significant after 11 min(P>0.05).The BFVR of 10min interval group had differences with the DA group at 45min,50min,55min,60min,65min and 70min(P<0.05 or P<0.01).CCA BFVR of 45 min interval group was statistically significant different from the DA group at 80,85,90,95 min and 100 min(P<0.05).No statistically significant differences existed between CCA BFVR of 30min interval group and DA group(P>0.05).Conclusion: Subcutaneous injection of DA,45min after injection of GTN can induce vasomotor abnormalities in rat with phase characters.The model may be useful for vasoactive screening of anti-migraine drugs.
摘要:Objective:To investigate the effect of astragalus on the serum levels of hepatocyte growth factor(HGF),renal function and the damage of renal morphology in ischemic acute renal failure(IARF).To study the protective mechanism of astragalus in IARF.Methods: The rats were pretreated with astragalus or vehicle for 3 days.IARF was induced by left renal artery clipping for 60 min,after the right kidney was removed.Biochemical parameters for serum HGF,blood urea nitrogen(BUN),creatinine(Cr) were measured and histological analyses of the kidneys were performed by tubular grading.Results: There have no significant difference between the serum HGF levels of I/R controls and the levels observed in sham-operated group(P>0.05).Serum HGF levels in astragalus treated group was significantly increased than those of I/R control group(P<0.01).After 4 h of I/R,serum levels of BUN and Cr were increased in I/R control rats compare with Sham-operated rats(P<0.01).Astragalus treatment reduced the levels elevation(P<0.05).Simultaneously,I/R-induced severe damage in renal morphology were significantly ameliorated by astragalus treatment(P<0.01).Conclusions: Astragalus pretreatment can alleviate the damage of renal function in IARF.It is may be one of the mechanisms of astragalus for protecting renal tissue in IARF that astragalus can induce the expression of HGF.
摘要:Objective:To study the effect and mechanism of Rhodobryum giganteum(Schwaegr.) Par.’s anti-atherosclerotic effect.Methods:Vascular endothelial cells were cultivated and the H2O2 were used to induce the oxidative stress injury of human umbilical vascular endothelial cells(HUVEC).Water extract of Rhodobryum giganteum(Schwaegr.) Par.was added to cultivated HUVEC,and the activity of the cells was carefully determined by OD value with MTT method.The Griess Reagent was used to detect the NO concentration of different groups.At the same time,the NO fluorescence analysis probe,DAF-FM DA(3-amino,4-aminomethyl-2′,7′-difluorescein,diacetate),was used to determine the activity of NO synthase.Results:The most suitable stimulating concentration of H2O2 on HUVEC is 12.5 mmol·L-1.Water extract of Rhodobryum giganteum(Schwaegr.)Par.was co-cultured with HUVEC damaged by H2O2.The OD values indicate that 3.33,2.50 mg·mL-1 water extract groups increase activity of the cells significantly compared with the model group(P<0.05),and 3.33 mg·mL-1 is much better than 2.50 mg·mL-1 dose group.To determine the content of NO and NOS,the 2.50 mg·mL-1 dose has significant effect(P<0.05).Conclusions: The H2O2 concentration of 12.5 mmol·L-1 could be used to establish the injured model of endothelial cell successfully.2.50 and 3.33 mg·mL-1 water extract of Rhodobryum giganteum(Schwaegr.) Par.have protective effect on ECV304 injured by H2O2.2.50 mg·mL-1water extract of Rhodobryum giganteum(Schw aegr.) Par could increase the activity of NOS and promote the synthesis and secretion of NO.
摘要:Objective: To observe the anti-tumor effect of Phellinus Linteus and Coriolus Versicolor Capsules(PLCVC) in S180 sarcoma and H22 hepatoma animal models in mice.Methods: The sarcoma S180 and hepatoma H22 models were established in mice.After 12 days of treatment,the animals were killed,and the subcutaneous sarcoma were separated and weighted.The levels of vascular endothelial growth factor(VEGF),CD4 and CD8 of S180 tumor tissue were investigated by immunohistochemical method.KM mice were intraperitoneal injected with H22 hepatoma cells,and treated with different experimental drugs.The survival time was observed and recorded,and life-prolongation rate was calculated.Result: PLCVC could inhibit the growth of S180 and H22 tumor,and inhibit the expression of VEGF,improve the expression of CD4 and CD8.The survival time of the mice treated by PLCVC were significantly longer than the untreated group.Conclusion: PLCVC can inhibit the growth of tumour,the mechanism is partially related to inhibiting angiogenesis and improving the immunological function.
