摘要:Objective:To develop the analytical method of five anthraquinones contained in rhubarb produced in Quanshui Village.To investigated the relativity between the contents of five constituents and the appearance.Method:The HPLC method is as follows:Kromasil-C18 column(4.6 mm×150 mm,particle diameter 5 μm),detective wavelength is 254 nm,methanol-0.1% phosphonic acid solution(85∶15) as mobile phase.Results:Compared with the high-grade rhubarb,the content of five anthraquinones in inferior ones descented about 30%.Compared with the high-grade part,the content of five constituents in the inferior part of the same root tuber descented about 70~80%.The contents of five anthraquinones in pale yellow rhubarb were lower than that in yellow ones.Conclusion: The method with fine repeatability is suitable to evaluate the contents of anthraquinones in rhubarb and the results indicate the contents of five constituents are associated with the appearance.

摘要:Objective:To establish a method for HPLC determination of icariin in Xianling gubao Dispersibie Tables.Methods: The condition for HPLC was C18 column(250 mm×4.6 mm,5 μm),Agilent the detection wavelength was 270 nm.Results: The standard curve was liner in the range of 0.368~1.84 μg and the correlation coefficient was(0.999 9.)The coverage recovery was 99.84% with RSD 1.59%.Conclusion: The method shows good separating result,accuracy and reproducibility,can be used for the quality control of Xianling gubao Dispersibie Tables.
摘要:Objective:To search for the micronization impact to the pill preparation,we carry out a series of comparative study on the micro characteristic、TLC characteristic、dissolution results of Ultro-micro and Normal powder of Zhizhu pill.Method:Using Fructus Aurantii Immaturus and Rhizoma Atractylodis Macrocephalae,the TLC information can be identified.Taking Hesperidin as the index component,the dissolution character were compared.The Weibull distribution analysis system were also applied.Result:We can see the almost similar TLC characteristic in the two pills,however the significant differences can be identified in the dissolution study on the Ultro-micro and the normal pills.Obviously,the dissolution rate and the total quantity of Hesperidin in the ultro-micro pills were increaseed.Conclusion:The active component of the pills can not be destructed in the process of micronization.Moreover,this behavior truly increase the vitro-utilization of the preparation.

摘要:Objective:An HPLC method was established for the determination of Rutin.Methods:Using Merck Lichrospher C18(5 μm,250 mm×4.6 mm) with the mobile phase consisiting of methanol-1% glacial acetic acid(32∶68);The detective wavelength was 257 nm;The flow rate was 1 mL·min-1.Results:The standard curve was liner with the range of 0.27~7.11 μg(n=6,RSD=0.91%),r=(0.999 5). The average recovery was 99.54%,RSD 0.91%.Conclusion:The method is simple and rapid which can be used for the quality control of Zhiyanxiao Keli.
摘要:Objective:To investigate the chemical constituents in the Panzeria alaschanica Kupr.Methods:The constituents were isolated by column chromatography and their structures were determined on the basis of chemical and spectral data.Results:Seven compounds were isolated and identified as β-sitosterol(1),stigmasterol(2),5-hydroxyl-7,4′-dimethoxy flavone(3),kaempferol-3-O-β-D-glucopyranoside(4),β-sitostero-3-O-glucoside(5),Rutin(6),Quercetin(7).Conclusions:All these Compounds except Compound(6) were isolated from Panzeria alaschanica Kupr.for the first time.
摘要:Objective:To optimize the extraction condition of total alkaloids from Rhizoma Coptidis and Cortex Phellodendri Chinensis.Methods:The extraction conditions of ratio of liquid to solid,extraction time and times were optimized by response surface method with the total alkaloids as the selective marker.The purification condition of total alkaloids was also studied.Results: The optimum extraction conditions were as follows: extraction repeated 4times,1 hour every time and ratio of liquid to solid was 18.125:1.The yield of total alkaloids under this condition was 66.44 mg·g-1 The total alkaloids were concentrated by acetic ether extraction under pH 12,with the ratio 1:1 of solvent to extract, also the extraction was repeated 6 times,then the content of total alkaloids was 66.130%.Conclusion:The experiment provides a basis for improvement of extraction condition of total alkaloids from Rhizoma Coptidis and Cortex Phellodendri Chinensis.
摘要:Objective:To probe the correlation between the effect of icariin on learning and memory and the level of estadiol(E2) in serum in senescence accelerated mouse prone/8(SAMP8).Methods:10 six-mon-old senescence accelerated mouse resistance 1(SAMR1) and 40 the same age SAMP8 were selected.SAMP8 were divided randomly into 4 groups(model,75 and 150 mg·kg-1 icariin ig,and 0.2 mg·kg-1 diethylstilbestrol ig).Then,the effects of icariin administration by oral gavage for 3 months on learning and memory deficits in SAMP8 and SAMR1 were investigated by the Morris water maze test;the level of E2 in serum in SAMP8 and SAMR1 was determined by radioimmunoassay kit.The mice were sacrificed after ethology test,and the uterus coefficient was calculated. Results: The administration of icariin orally could enhance the learning and memory ability of SAMP8 in Morris water maze test;increase the level of E2 in serum in SAMP8;create non-significant difference in the uterus coefficient of SAMP8.There was a significant negative correlation between the latency in Morris water maze test and the level of E2 in serum in SAMP8 analyzed by Pearson correlation coefficient.Conclusions: Icariin could improve the cognitive impairments of SAMP8,and its mechanism may be related to the increase in the level of estrogen in serum in SAMP8.

摘要:Objective:To explore the changes of rat serum proteins related with syndrome of Qi-deficiency and blood stasis and to clarify the molecular biomarkers of the syndrome.Methods: The rat models with Qi-deficiency and blood stasis were established by combined methods.The changes of serum protein expressing pattern were obeserved by two dimensional gel electrophoresis(2-DE)、Image Master 2D Elite 6.01 analysis software and MALDI-TOF-MS.Results:By MALDI-TOF-MS and then with the search engine MASCOT,compared with the normal group,6 proteins were identified,in which CD5 antigen-like was down-regulated,other 5 proteins including haptoglobin,complement component C3,etc.were up-regulated.Conclusion:The functions and features in these proteins had relevance to the pathological process of inflammation,immune regulation,which is consistent with the known occurrence and development of the syndrome of Qi-deficiency and blood stasis.

摘要:Objective:To discuss the clinical effective of Busuishengxue granules to chronic aplastic anemia(CAA) patients,and to observe the difference in expression level of myelo-cytokine c-kit/CD117 before and after treatment.Methods:This experiment employed reverse transcription-polymerase chain reaction (RT-PCR) method to investigate the change of expression level of bone marrow mononuclear cell(BMMC) c-kit receptor mRNA and its c-kit receptor protein CD117 with flow cytometer method(FCM).Results:The expression level of BMMC c-kit receptor mRNA and its receptor protein CD117 in CAA patients were higher than that in normal control group.And after treatment both of the expression levels descended to different degree.Moreover,the descent in Busuishengxue Granule group was superior to that in zaizhangshengxue Tablet group.Conclusions: There was abnormal expression of bone marrow SCF receptor c-kit/CD117 in CAA patients.Busuishengxue Granules by regulating the bone marrow c-kitmRNA and the expression of CD117 improved the hematopoietic function of bone marrow in CAA.
摘要:Objective: To investigate the chemical constituents and anticoagulant activity of Qin-Lian-Si-Wu decoction.Methods: Qin-Lian-Si-Wu decoction was extracted by hot-water,and then concentrated by decompression,and added ethanol till the concentration. of ethanol reached 80%.The upper solution was concentrated at(40 ℃,) dissolved by distilled water,separated by gradient elution with different concentration ethanol from macroporius adsorptive resins D101,then obtained the different fraction.Its constituents were isolated by silica gel column chromatographic method,and their structures were identified through UV,IR,NMR and MS spectrum.These compounds were prepared into aqueous solution,and then their effects on the blood plasma thrombin time in vitro in rabbits were determined.Result: Five flavonoids,one alkaloid,three aromatic acids,one sesquiterpene and two other components were isolated and identified from Qin-Lian-Si-Wu decoction.The results of thrombin time test showed that flavonoids,aromatic acids,alkaloid,sesquiterpenes could prolong the blood plasma thrombin time in vitro in rabbits remarkably.Conclusion: 12 compounds were isolated and identified from Qin-Lian-Si-Wu decoction for the first time.Nine compounds,including oroxylin A,wogonin,6-dimethoxy wogonin,berberine,baicalein,ferulic acid,vanillic acid,caffeic acid,rehmapicrogenin showed the anticoagulant activity at a certain dosage.
摘要:Objective:To observe the dynamic expression of L-FABP in rat nonalcoholic fatty liver disease(NAFLD) model and to discuss the role of L-FABP in the rat NAFLD.Methods:Fourty Wistar rats(200 g~220 g) were randomly divided into control group and model group.control group fed with standard diet;model group fed with high-fat diet(85% of the standard diet,13% of lard and 2% cholesterol).The model group rat was put to death in 4,8,12 weeks seperately.Alanine aminotransferase in serum(ALT),aspartate aminotransferase in serum(AST),total cholesterol(TC),triglyeride(TG);mensurate fasting blood glucose(FBG),Fasting insulin(FINS) were measured,and insulin sensitivity index(ISI) was calculated.HE staining was used to observe pathological changes in liver tissue;semi-quantitative reverse transcription polymerase Chain reaction(RTPCR) method was used to determine the expression of L-FABPmRNA changes.in rat liver tissue.Results:The expression of L-FABPmRNA was gradually increased in the rat liver of model group in 4,8 and 12 weeks(P<0.01),the expression of L-FABP was gradually increased compared with the control group significantly(P<0.01).In model group,the leral of serum ALT,AST,TC,TG was significantly higher than those in the control group(P<0.01),while ISI decreased significantly(P<0.01).Conclusion:Continuous 12 weeks high-fat diet can successfully replicate NAFLD rat model,and with extending the diet feeding time the model caused the expression of L-FABP increases gradually.
摘要:Objective:To investigate the therapeutic effect of Sanpi Decotion in ulcerative colitis(UC) mice and discuss its therapeutic mechanism.Methods:Forty BALB/C mice were divided randomly and equally into normal group,model group,sulfasalazine(SASP) group,and Sanpi Decotion group(ten mice in each group).After set up the oxazolone colitis,SASP group and Sanpi Decotion group were treated with sulfasalazine and sanpi decotion respectively,to observe the body weight,stool,food and drink situation of the mice every day.Killed all the mice on the tenth day,separated the spleen and weighed it,got the colon and evaluated the histological changes of the damaged colonic tissues,and measured the IL-2 and IL-10 concentrations in serum by using the enzyme-linked immunosorbent assay.Results: The symptom of Sanpi Decotion group and SASP group mice had great amelioration compared to model group mice,and compared to model group,the weight of spleen in Sanpi Decotion group and SASP group was increased,the concentration of IL-2 in Sanpi Decotion group was higher than that in model group(P<0.01),and the concentration of IL-10 was lower than that in model group(P<0.01).Conclusion:Sanpi Decotion is effective for treatment of UC and it can increase the concentration of IL-2 and decrease the IL-10 concentration,regulate the balance of the cytokines,improve the function of immunity,control the inflammation in the colon.
摘要:Objective:To study the effect of "Weichangshu" and medical interfering on blood flow and gastro smooth muscle cells in rats of diabetic gastroparesis model.Methods: One hundred thirty-one male SD rats(SPF class) were randomly divided into the control group(10 rats),model group(22 rats),ethamsylate group(33 rats),lumbrukinase group(33 rats) and WCS group(33 rats).Rats were intraperitoneally injected streptozotocin(STZ) solution(60 mg·kg-1).After seven days,we established blood sugar more than or equal to 16.7 mmol·L-1 as experimental standard.Rats in the WCS group were treated by traditional Chinese medicine "WCS". So rats in the ethamsylate group treated by ethamsylate injection,rats in the lumbrukinase group treated by lumbrukinase enteric-coated capsules,rats in the control group and rats in the model group were treated by distilled water one time per day.After having been observed for 30 weeks,blood sugar,FINS,C-P and gastric fluid emptying rate were determined.Subsequently,samples of sinus ventriculi were made HE staining,Mallory staining and immunohistichemistry staining.Pathological lesion of gastric smooth muscle cells(SMC) was examined microscopically.Other sample of sinus ventriculi was made up electron microscopic specimen so as to know pathological lesion of ultrastructure of SMC through transmission electron microscope.Results: After 30 weeks,in the model group,endochylema of gastro smooth muscle cells were degenerated.The apoptotic index of SMC in rats of the model group was(24.33±2.34)% determined by TUNEL method,there was significantly difference compared with rats of the control group(P<0.01).Protein expression of Bcl-2 in SMC was reduced in rats of the model group,but protein expression of Fas was overexpression,there was significantly difference compared with rats of the control group(P<0.01).However,pathological lesion of gastro smooth muscle was obviously improved in rats of the WCS group and in rats of the lumbrukinase group.But in rats of the ethamsylate group pathological lesion of gastro smooth muscle was more serious.Conclusion:There were pathological Changes in gastric smooth muscle cells(SMC)in rats of diabetic gastroparesis model.Medicinal interfering in blood flow had the effect on gastric SMC pathology in rats of diabetic gastroparesis model."WCS" could decrease blood sugar and improve gastric SMC pathology and accelerate gastrointestinal motility.

摘要:Objective:to discuss the effect of Panax notoginoside(PNS) on the neurologic impairment and( ) 2008-06-18( 陈志刚,)Tel:(010)67689776;E-mail:zgchen01@sina.com inflammatory factors IL-6,MMP-9 of rats with acute intracerebral hemorrhage.Method:The SD rats were divided into 3 groups randomly: sham group,model group and Panax notoginoside group.An animal model of ICH was edtablished by injecting collagenase Ⅶ into the caudate nucleus,and then Zea longa score,ICH mortality and the contents of IL-6 and MMP-9 in rat brain were measured.Results:Longa score of PNS group are lower than that of model group at the 5th day after ICH model establishment(P<0.05).The improvement of neurologic impairment of PNS group at the 5th day compared with the 1st day are greater than that of model group(P<0.01).The ICH mortality rate between PNS group and the model group had no significant difference.The contents of IL-6 and MMP-9 in brain tissues around hematoma of model group were significantly elevated at the 48th hour(P<0.01 and P<0.05),and continued to rise till the 7th day(P<0.05 and P<0.01).The inhibitive effect of PNS on the expressions of IL-6(P<0.05),MMP-9(P<0.05)in brain on the 7th day was obvious.Conclusion:Panax notoginoside can promote recovery of neurologic impairment after ICH. One of the mechanisms may be that PNS could effectively inhibit IL-6 and MMP9.
摘要:Objective: To study the pharmacological effect of Zhenhuang Pills.Methods:The anti-bacteria,anti-inflammatory,antipyretic,analgetic,immunologic and the Huafu Shengji actions of Zhenhuang Pills were studied on the corresponding pharmacological models. Results: Zhenhuang Pills had strong inhibition on many kinds of inflammatory models and it showed analgetic effects and could obviously enhance the macrophage phagocytosis of mice and antisepsis effects.It also showed a certain degree of antipyretic effect in Escherichia coli endotoxin-induced temperature increment and accelerated the removal of the rotten tissue.Conclusion: Zhenhuang Pills have obvious anti-inflammatory,analgetic effects.it can increase physical immunity and has antisepsis effects.
摘要:Objective:To observe the effects of the principle of restraining the hyperactivity of the liver yang on CD62p and ICAM-1 expressions in the cerebral vascular endothelium of renovascular hypertensive rats(RHR).Methods:50 rats were divided into control group(n=10) and renovascular hypertensive rats group(n=40).40 renovascular hypertensive rat were divided into four groups: model group,aspirin group,principle of restraining the hyperactivity of the liver yang group,principle of restraining the hyperactivity of the liver yang and aspirin group.The treatments Lasted 4 weeks after the establishment of the model.The rats were sacrificed and the tissues of the frontal lobe were slice into 5.0μm-thick frozen sections from the bregma section underwent in situ hybridization of ICAM-1 and CD62p mRNA expressions,the other five rats were examined by histochemistry under electron microscope and light microscope.Results:After treating,the plasma levels of CD62p and ICAM-1 in the principle of restraining the hyperactivity of the liver yang or aspirin group were obviously lower than that in the model group,the Aspirin group and the principle restraining the hyperactivity of the liver yang group.ICAM-1 and CD62p mRNA expressions and immunohistochemistry expressions in the principle of restraining the hyperactivity of the liveryang and aspirin group were obviously lower than that in the aspirin group and the principle restraining the hyperactivity of the liver yang group.Conclusions:Expressions of CD62p and ICAM-1 in plasma are significantly high in RHR at 12 weeks after operation.principle of restraining the hyperactivity of the liver yang obviously repress the expressions of CD62p and ICAM-1 in cerebral vascular endothelium of renovascular hypertensive rats,and the effect of treatment with principle of restraining the hyperactivity of the liver yang and aspirin in the RHR is obviously improved.

摘要:Flavonoids,terpene lactones and polyprenols are bioactivites actions of Ginkgo biloba L.Rencent progress on chemical constituents of Ginkgo biloba L were briefly reviewed,and structure of flavonoids,terpene lactones and polyprenols were detailed reviewed in this paper.
