摘要:Objective:To develop a method for the determination of scopoletin in Qinghao.Method:Sample was extracted with methanol.Scopoletin in Qinghao was determined by HPLC with ODS column.The mobile phase was methanol-0.08% H3PO4(4∶6)solution.The detection wavelength was 340nm.Result:Scopoletin in sample solution was well separated.Linearity of scopoletin was good(r=(0.999 9))in range of 0.021~0.42μg.The average recovery was 99.3%,RSD was 2.0%.Conclusion:The method is simple,accurate,repeatable and suitable for the determination of scopoletin in Qinghao.
摘要:Objective:To develop a Capillary GC method to determine the seven components,α-pinene,eucalyptol,camphor,menthol,methyl salicylate,cinnamia aldehyde and eucalyptal in Huoluo oil.Method:A fused silica capillary column was used.The intial temperature of column was kept at 90℃ for 1 min,then raised to 180℃ at the rate of 6℃·min-1,finally to 250℃ at the rate of 25℃·min-1 and maintained the temperature for 5 min,the carry gas was nitrogen.The injector and detector temperature was 220℃ and 260℃.Injections were performed in the splitless mode and an FID was used as the detector.Results:The standards curves of α-pinene,eucalyptol,camphor,menthol,methyl salicylate,cinnamia aldehyde,eucalyptal are linear within the range of 6.94~41.64μg·mL-1,1.00~6.01 μg·mL-1,6.86~41.1 μg·mL-1,24.1~144.5 μg·mL-1,24.09~144.55μg·mL-1,24.16~144.98μg·mL-1,1.03~6.17μg·mL-1,1.04~6.25μg·mL-1,and RSD was 1.8,1.1,0.9,0.7,1.1,1.7,1.1%.Conclusion:This method is simple and accurate,and can be used to control of Huoluo oil.
摘要:Objective:To investigate the influence of Guizhi Tang to secreting inflammatory factors in RAW264.7.Methods:RAW264.7 cell line was stimulated by Lipopolysaccharide and treated with the serum of containing Guizhi Tang at the same time.24 hours later,collected the supernatant and measured the inflammatory factors such as NO、TNF-α and IFN-β.Results:Guizhi Tang can decrease the amount of NO、TNF-α and IFN-β,and shows dose-dependent relationship significantly.Conclusion:Guizhi Tang may produce the anti-inflammatory effect by decreasing the amount of NO、TNF-α and IFN-β.
摘要:Objective:To increase the diversity of hirudin-shuffled libraries by DNA family shuffling method.Methods: PCR products of HV1,HV2 and HV3 genes were combined and digested with DNase Ⅰ.Then random fragments about 50bp were purified and reassembled to the same size of hirudin genes by two amplifications of PCR with or without primers. Pools of hirudin mutant genes were ligated into phagemid pCANTAB-5E vector and three randomly picked clones were sequenced to observe the efficiency of shuffling.Results: The hirudin-shuffled libraries contained 1.2×106 different clones and the three clones were hybrids of three parent genes.Conclusion:These results show that a highly shuffled library of hirudin genes has been obtained which lay a substantial foundation for screening hirudin molecules with high bioactivity.
摘要:Objective:To observe the influence of sedum sarmentosum tricin-glucoside on proliferation of spleen T and B lymphocytes of normal mice.Methods:Method of mixturing()3H-TdR to mice spteen cell was applied.After stimulation with canavaline(ConA)or lipid polysaccharides(LPS),the radioactive relative quantity(cpm) of()3H-TdR in DNA in spleen cell.Results:The dosages in 100,10,1μg/mL(final concentration)of sedum sarmentosum tricin-ghcoside had highly significant inhibition to cpm number of T lymphocyte(P<0.001);the dosage in 100μg/mL of sedum sarmentosum tricin-glucoside had highly significant inhibition to cpm number of B lymphocyte (P<0.01).Conchsions:Sedum sarmentosum tricin-glucoside can obviously inhibit proliferation of spleen T and B lymphocytes of normal mice.
摘要:Objective:To observe gastroenteric hormones content changes of blood in model rat of spleen-qi deficiency during different modelling time points.Methods:Adopting excess fatigue(by swimming) and out of constant diet on rats for constant three weeks and stopping doing on them in the 4w.Results:Compared with the control group,the model rats distinctly appear diet reduction,feces rarefaction,reduction activities,accidie and extrados,etc in modelling 2~3w;weight significantly failed,and it has yet to come back in the 4w;urine D-xylose excretion rate significantly falled during 2~4w;GAS and MTL level of serum failed in the 3w and significantly came back during natural recovery period;SS level markedly lower in the 1w,and no significant changes in the remaining time points;VIP level markedly rised in the 2w,and significantly failed in 3w;SP level clearly failed in the 3w;NPY level clearly lower in the 2~3w,and β-EP level clearly declined in the 4w.Conclusion:The model rat made by excess fatigue and out of constant diet for constant three weeks can appear diet reduction,feces rarefaction,reduction activities,etc;and abnormal changes of weight,urine D-xylose excretion rate,and gastroenteric hormones of Blood.

摘要:Objective:To observe the protective effect of(-)-Catechin gallate and(+)-Epicatechin on Xanthine-Xanthine Oxidase System injury in cultured cardiomyocytes.Methods:The injury model of cultured neonatal rat cardiomyocytes was induced by Xanthine-Xanthine Oxidase System.Cell proliferation was detected by MTT assay,lactate dehydrogenase(LDH) activity,Maleic Dialdehyde(MDA) and Superoxide Dismutase(SOD) level in supernatant were measured with colorimetric assay.Results:Compared with model group,(-)-CG,(+)-EC can obviously increase survival rate of cardiomyocytes,decrease LDH activity and MDA level,increase SOD level in supernatant.Conclusion:(-)-CG、(+)-EC can protect cultured cardiomyocytes against Xanthine-Xanthine Oxidase System damage.
摘要:Objective:The effects of Danggui Buxue Tang(Chinese Angelica decoction for replenishing blood;DBT) on the erythrocyte functions were studied in the acute hypoxia mice.Methods:The mice were divided into five groups:control,model,and DBT with three dosages(20,10,5g/kg·d).Water or DBT were administrated for 10 days.Acute hypoxia model was made by putting the mice into hermetical specimen bottles for 10 minutes.Then the erythrocyte membrane fluidity,the erythrocyte deformability and the erythrocyte immunity was tested.Results:DBT(5g/kg·d) could prevent the erythrocyte membrane fluidity from decreasing.The same actions were observed for the erythrocyte deformability and the erythrocyte immunity function in the acute hypoxia mice.DBT 10g/kg·d could prevent the erythrocyte membrane fluidity and the erythrocyte deformability from decreasing.However,effects were not observed with DBT 20g/kg.Conclusion:The above study shows that small and middle dose DBT can improve the function of red blood cell,which may be the mechanism of invigorating qi to promote blood of DBT.
摘要:Objective:To discuss the mechanism of "Xiao Geng Yan Jing" capsule for treating menopausal syndrome.Methods:Extirpation of ovaries of rats was cartied out.After 30 days of oral administration,serum was collected to determine the contents of estradiol(E2)、luteinizing hormone(LH)、follicle-stimulation hormone(FSH),The contents of 5-hydroxytryptamine(5-HT)、norepinephrine(NE)、dopamine(DA) and 5-hydroxyindoleacetic acid(5-HIAA) in brain was determined.The uterus was weighed.Results:The experiment showed that "Xiao Geng Yan Jing" capsule could decrease the levels of FSH and LH obviously,and increase the level of E2.For the brain of castrated rats,it could decrease 5-HT、5-HIAA contents and 5-HT/NE.But for NE、DA contents and 5-HIAA/5-HT it have no influence.Moreover,this medicine can increase the weight of the castrated rat and their uterus.Conclusion:"Xiao Geng Yah Jing" capsule can improve and treat menopausal syndrome though the mechanism of adjusting sexual gland and nervous function.
摘要:Objective:To research the protective effect and mechanism of FBD on cerebral ischemia reperfusion.Methods:Rat model of cerebral ischemia was established by applying middle cerebral artery occlusion(MCAO).Effect of FBD on cerebral functions,infarction percentage,water-contained percentage,biochemical indices and histology of the rat brains after MCAO were in vestigated.Effect of FBD on the blood brain barrier permeability of mice after cerebral ischemia reperfusion were also investigated.Results:FBD can obviously decrease the cerebral function score,cerebral infarct size and water content,improve the activities of Na+-K+-ATPase and Ca2+-ATPase.reduce the content of MDA and NO of rat brains(P<0.05~0.01),repair pathological injury of ischemia-reperfusion rat.FBD can remarkably decrease blood brain barrier permeability of mice after cerebral ischemia reperfusion(P<0.05).Conclusion:FBD has good effects on cerebral ischemia reperfusion.
摘要:Objective:To study the preventive and therapeutic effect of Chinese medicine Jiaweibiejiajian Pill(JWBJ)on rats with hepatic fibrosis induced by human serum albumin.Methods:The rats model with hepatic fibrosis was induced by multiple subcutaneous injections of serum albumin solution for 4 times.After it was confirmed that there were albumin antibody in rats serum,and then rats were divided into model-group,low-dose group of JWBJ,mediate-dose group of JWBJ and high-dose group of JWBJ.Rats were given JWBJ orally every day,and injected albumin solution into the rats caudal veins for 9 times during 4 weeks.When hepatic fibrosis was verified to come into being by pathological examination.Alanine amino transaminase(ALT),aspartate aminotranferase(AST),total protein(TP),albumin(ALB) Hyaluronic acid(HA),Laminin(LN),Collagen Type(PC) in serum of all rats were examined.Results:Compared with the model group,JWBJ could obviously decrease the concentration of serum ALT,AST,HA,LN,CIV and increase the concentration of serum ALB,TP,and increase the ratio of serum A/G of rats with hepatic fibrosis,and reduce the degree of hepatic fibrosis.Conclusion:JWBJ can effectively prevent and treat hepatic fibrosis induced by serum albumin.

摘要:Objective:To investigate the effects of Tiaozhiqinggan(TZQG) capsule on experimental hyperlipidemia in rats and fatty liver in mice.Methods:The rat hyperlipidemic model was induced by feeding caloric diet and mouse fatty liver model was induced by ethionine,and effect of different doses of TZQG capsule on reducing blood lipid and prevention of fatty liver was obsevered.Results:TZQG capsule evidently lowered the levels of CHO,TG,and LDL-C,increased the HDLC,elevated the ratio of HDL-C and CHO in blood,and decreased the liver weight and abdominal fat of hyperlipidemic rats.It significantly improved the transformation of fatty liver of model mice.Conclusion:TZQG capsule had markedly effects on reducing blood lipid and prevention of fatty liver.
摘要:Objective:To investigate the protective effects of Gankang on D-galactosamine-induced liver injury and study its possible mechanism.Methods:The model of chemical injury in liver of mouse was prepared.The level of alanine aminotransferase(ALT),aspartate aminotransferase(AST) in serum,malondiadehyde(MDA)content,nitric oxide(NO)in liver homogenate were assayed by spectrophotometer.Results:Gankang(200,400,800mg·kg-1)significantly decreased the levels of serum transaminase(ALT,AST),liver index and spleen index.Furthermore,Gankang also decreased MDA and NO level in liver homogenate.Conclusion:Gankang possesses protective action on chemical injury in liver of mice,which is related to its mechanism of ressistance to the liver cell oxidation.

摘要:The current evaluations on clinical safety of traditional Chinese medicine can not fully reflect the information of drug safety due to various reasons or shortcomings.Based on a comprehensive evaluation of the existing clinical trails,we try to define the neglected factors and to improve the methods for safety evaluation.The efforts will provide the foundation for the future safety evaluation of clinical trails for traditional Chinese medicine.
