      摘要:Objective: To establish a standard for different dosage forms of Ginseng Radix et Rhizoma and provide reference for production practice,based on the study of some threshold values of standard decoction of Ginseng Radix et Rhizoma (SDRG). Method: Twelve batches of SDRG were prepared from representative samples through standard process,then the paste-forming rate,index components content along with their transfer rate,and high performance liquid chromatographic (HPLC) fingerprints similarity were analyzed,compound identification of the common peaks was employed UPLC-Q-TOF/MS;the difference among the slices,the standard decoction and the granule,all from the same batch,were comparatively analyzed to reveal the mass transfer rule in the process. Result: The content of ginsenoside Rb1 and the total amount of ginsenoside Rg1,Re should be not less than 0.282 g·L-1 and 0.529 g·L-1,respectively.Extract rate should be not less than 40%,the fingerprint similarity between batches should be not less than 0.905,and it should be not less than 0.966 when compared with the reference fingerprint R.Ten kinds of saponins were identified in 12 common peaks,the process from slices to standard decoction was the qualitative transfer,while standard decoction to granule was the quantitive change. Conclusion: The standard decoction can act as standard reference object in the production process of formula granule,to guarantee the consistent of final productions through batch charging and process optimization.As to the different forms of Chinese herbal pieces,whether traditional forms or modern ones such as ultramicro piece and formula granule,are almost all dispersed in water in clinical practice;in consideration of the same material basis between the water liquid used and the standard decoction, the standard decoction and its concentration can act as reference material in the aspect of identification and quality control.

      摘要:Objective: To establish quality control methods for standard decoction of Carthami Flos. Method: Standard decoction of Carthami Flos was prepared,its dry extract rate,transfer rate of index components and pH were calculated to evaluate the stability of the process.A quality control method containing determination of hydroxysafflor yellow A and UPLC-UV fingerprint was established,the main common peaks in fingerprint were identified to clarify the main chemical constituents in standard decoction of Carthami Flos. Result: The concentration of hydroxysafflor yellow A in the standard decoction was 3.6 g·L-1.The similarities of fingerprints of samples were >0.9.The dry extract rate of the standard decoction was 32.6%,the transfer rate of hydroxysafflor yellow A was 61.2%,pH was 4.1. Conclusion: Methods for evaluating the quality of standard decoction of Carthami Flos is presented,they can provide reference for the quality control of products which are stemmed from the water extract of Carthami Flos.

      摘要:Objective: To establish quality control methods for standard decoction of Angelicae Sinensis Radix. Method: Standard decoction of Angelicae Sinensis Radix was prepared,a quality control method containing UPLC fingerprint and determination of ferulic acid was established,the main common peaks in its fingerprint were identified to clarify the main chemical constituents in standard decoction of Angelicae Sinensis Radix and evaluate the stability of the process by calculating parameters,such as dry extract rate,transfer rate of index component. Result: The concentration of ferulic acid in standard decoction of Angelicae Sinensis Radix was 0.077%.The similarities of fingerprints of standard decoction of Angelicae Sinensis Radix were >0.9.There were 11 major common peaks in the fingerprint of standard decoction of Angelicae Sinensis Radix,including organic acids,volatile oils and nucleosides. The dry extract rate was 51%,the transfer rate of ferulic acid was 78.5%. Conclusion: Methods for evaluating the quality of standard decoction of Angelicae Sinensis Radix is presented,it can provide reference for the quality control of products which are stemmed from the water extract of Angelicae Sinensis Radix.

      摘要:Objective: To establish quality control methods for standard decoction of Codonopsis Radix and evaluate its stability and main influencing factors. Method: Standard decoction of Codonopsis Radix was prepared,its quality control method containing the determination of lobetyolin and UPLC fingerprint was established.The main common peaks in fingerprint were identified to clarify the main chemical constituents in the standard decoction and evaluate the stability of process by calculating the parameters,such as dry extract rate,transfer rate of index components and pH of the standard decoction. Result: The concentration of lobetyolin in the standard decoction was 0.02%,the similarities of its fingerprints were >0.9;there were 12 major common peaks in the fingerprint.Dry extract rate of standard decoction of Codonopsis Radix was 45.4%,the transfer rate of lobetyolin was 58.1%. Conclusion: Methods for evaluating the quality of standard decoction of Codonopsis Radix is presented,and thickness of decoction pieces of Codonopsis Radix is the main factor affecting dry extract rate of the standard decoction.

      摘要:Objective: To establish quality control methods for standard decoction of Gardeniae Fructus. Method: Fifteen batches of standard decoction of Gardeniae Fructus were prepared and its dry extract rate,transfer rate of index components and pH were calculated.Fingerprints of standard decoction of Gardeniae Fructus were established by UPLC and the main chromatographic peaks were identified by mass spectrometry. Result: The average dry extract rate of standard decoction of Gardeniae Fructus was 28.4%,the transfer rate of geniposide was 77.7% and the pH was 4.The similarities of fingerprints of the standard decoction were >0.99,main component of standard decoction of Gardeniae Fructus was geniposide. Conclusion: Methods for evaluating the quality of standard decoction of Gardeniae Fructus is stable and accurate for quality control of preparations including Gardeniae Fructus water extract.

      摘要:Objective: To prepare standard decoction of Scutellariae Radix and establish its quality standard. Method: According to the standard of Chinese herbal medicine decoction preparation principle,standard decoction of Scutellariae Radix was prepared to calculate its transfer rate of baicalin and the dry extract rate.In addition,the quality standard of standard decoction of Scutellariae Radix was also established. Result: Transfer rate of baicalin ranged from 55.1% to 82.2%,dry extract rate was at the range of 28.6%-43.8% and pH value was at the range of 4.5-5.5.The fingerprint similarities of 15 batches of standard decoction of Scutellariae Radix with reference fingerprint were >0.9. Conclusion: The preparation method established in this study is standard and the quality standard is perfect,it is suitable for evaluating the quality of standard decoction of Scutellariae Radix.

      摘要:Objective: To design and establish UPLC fingerprint on several varieties of Pueraria from Sichuan, and provide basis for its medicinal materials identification and quality control. Method: The UPLC fingerprint was performed on Agilent Zorbax SB-C18 (2.1 mm×5 mm, 1.8 μm), with acetonitrile -0.1% acetic acid aqueous solution as mobile phase; the column temperature was 30℃; the flow rate was 0.4 mL·min-1 and injection volume was 0.5 μL; the detection wavelength was set at 250 nm. In addition, similarity analysis and cluster analysis were conducted. Result: The UPLC fingerprint of P. lobata, P. thomsonii and P. omeiensis were established, and respectively confirmed 29, 18 and 16 peaks. Seven peaks in P. lobata and six peaks in P. thomsonii and P. omeiensis were identified by comparing the retention time of reference substance and chromatographic peak. The main differences between three varieties were peaks 2, 13, 14, 16, 23, 25, 26 and 29, which were not detected in P. thomsonii and P. omeiensis; the main differences between P. thomsonii and P. omeiensis were peaks 6 and 14, which were not detected in P. omeiensis. The correlation results obtained in clustering analysis were close to similarity analysis results. Conclusion: UPLC fingerprint technology is fast and its result is reliable and accurate, so it can be used as a reliable method for study on this kind of medicine fingerprint.

      摘要:Objective: Comparison and correlation analysis of polysaccharose contents in Polygonati Odorati Rhizoma from different provenances to provide the basis for superior provenance selection of Polygonati Odorati Rhizoma. Method: The 21 pieces of wild provenance resources of Polygonati Odorati Rhizoma were collected from the main distribution area of China. Their agronomic traits, longitude and latitude, height were measured and recorded. Polysaccharose content was detected by phenol-sulfuric acid method, and then the obtained data were analyzed by Excel, SPSS and DPS software. Result: There were extremely significant difference in agronomic traits and polysaccharose contents between Polygonati Odorati Rhizoma from different provenances (P<0.01). The polysaccharose contents in Hunan and Sichuan provenance were higher, followed by Zhejiang provenance. Polysaccharose contents had low correlations with width of leaves, diameter of rhizome and heights, however, the polysaccharose contents were higher in the lower longitude and latitude provenances from which the plants presented larger area and ratio of length and width of leaves, thicker stem and larger diameter of fruits. What's more, 12 characters tested were simplified to 5 principle components, with a cumulative contribution rate of 85.44%. 21 provenances could be divided into 4 groups and the comprehensive characters of the first group showed the best performance. Conclusion: The 8 superior provenances have been selected and more attention should be paid on Hunan and Sichuan Pingchang provenance, followed by Zhejiang Lin'an, Zhejiang Pan'an and Jilin Fusong provenances for superior variety breeding.

      摘要:Objective: To establish the HPLC fingerprints of different polarity fractions from Henan Forsythia Fructus, compare their chemical compositions between the different polarity fractions, and provide a reference for comprehensive evaluation of the quality of Henan Forsythia Fructus. Method: Henan Forsythia Fructus was extracted by 75% ethanol to obtain the total extract, and then it was extracted by petroleum ether, ethyl acetate, chloroform, n-butanol, and water respectively; concentrated to extractum, and dried under reduced pressure to obtain powder samples. The samples were analyzed by HPLC fingerprints, and the similarity evaluation software was used for data analysis. Result: Eighteen common peaks were determined in HPLC fingerprints of petroleum ether fraction, and 3 peaks of them were identified; 25 common peaks were determined in HPLC fingerprints of chloroform extract fraction, and 4 peaks of them were identified; 20 common peaks were determined in the HPLC fingerprints of ethyl acetate fraction, and 3 peaks of them were identified; 17 common peaks were determined in HPLC fingerprint of n-butyl alcohol fraction, and 3 peaks of them were identified; 17 common peaks were determined in HPLC fingerprints of water extract, and 2 peaks of them were identified. The fingerprint similarity of the same polarity fractions of Forsythia Fructus was high, with significant differences in chemical compositions between different polarity fractions. Conclusion: The experimental method is accurate, reliable, and highly repeatable, and can fully reflect the distribution of the chemical composition of Forsythia Fructus, providing a reference for the comprehensive quality evaluation of Forsythia Fructus decoction pieces.

      摘要:Objective: To study the chemical constituents of Tibetan medicine Polygonum sibiricum. Method: The air-dried whole plants of P. sibiricum were extracted with 70% ethyl alcohol. The extract was condensed to a small amount of volume and then was subjected to macroporous resin chromatography with an eluent of ethyl alcohol(0%, 30%,50%,90%) to get four fractions. The compounds from 90% ethyl alcohol fraction were isolated and purified by column chromatography. Result: Eight compounds were isolated from P. sibiricum, and were identified as quercetin (1), kaempferol (2), luteolin (3), guajavarin (4), o-hydroxybenzoic acid (5) and oleanolic acid (6), daucosterol (7), and β-sitosterol (8). Conclusion: Compounds 1-8 were isolated from P. sibiricum for the first time.

      摘要:Objective: To study the secondary metabolites of an endophytic fungus Alterraria sp. isolated from Saussurea medusa. Method: Alterraria sp. was isolated from the medical plant S. medusa.The strain was cultured in a large-scale after the culture conditions had been optimized. The secondary metabolites were isolated by chromatography methods such as silica gel, Sephadex LH-20 columns, ODS flash column and HPLC, and the analysis of LC-MS. The structures of these compounds were identified by theirphysico-chemical constants and the NMR and MS data. Result: Nine compounds were isolated and identified as spirotryprostatin A (1), 4-hydroxybenzoic aldehyde(2), 4-hydroxyphenyl-2'-hydroxypropanoate (3), dihydro-p-methoxycinnamic acid(4),4-methoxybenzoic acid(5), p-hydroxymethylphenylacetate(6), 4-hydroxyphenyl ethanol(7), 4-hydroxybenzoic acid (8), 3-phenylpropanoic acid(9). All compounds were tested for their inhibitory activities against human lung adenocarcinoma strain A-549.Compound 1 exhibited strong cytotoxic activity against A549 with IC50 value of 2.0 mg·L-1. Conclusion: All the compounds were isolated from Alterraria for the first time.

      摘要:Objective: To study on gliotoxin metabolites from endophytic Penicillium sp. of Astragali Radix and investigate their antimicrobial activity to Erysiphe pisi. Method: The compounds were isolated and purified by repeated silica gel column chromatography, Sephadex LH-20 gel chromatography, and HPLC method; their chemical structures were identified by chemical methods combined with nuclear magnetic resonance analysis and mass spectrometry. The MIC values for E. pisi. were determined as sensitivity to the compounds in biological evaluation. Result: Seven gliotoxins were obtained and elucidated as gliotoxin (1), bisdethiobis(methylthio) gliotoxin (2), dehydroxybisdethiobis(methylthio) gliotoxin (3), dehydrogliotoxin (4), 6-deoxy-5a,6-didehydrogliotoxin (5), didehydrobisdethiobis (methylthio) gliotoxin (6), and bis-N-norgliovietin (7). Bacteriostasic activity test results showed that compounds 1, 4 and 5 had strong inhibitory activity for E. pisi, MIC values of 1.56, 0.78 and 6.25 mg·L-1. Conclusion: All compounds were isolated from the endophytic fungi of Astragali Radix for the first time, and the endophytic fungus Aspergillus sp. from the stems of Astragali Radix, is a promising source for gliotoxins.

      摘要:Objective: To establish an analysis method for direct detection of monosaccharide compositions of Ephedrae Radix et Rhizoma polysaccharide by HILIC-LC-MS/MS. Method: The separation was performed on ACQUITY UPLC BEH Amide column (2.1 mm×100 mm, 1.7 μm) with 95%acetonitrile (A)-95% water (B) as the mobile phase for gradient elution (0-10 min, 92%-70% A; 10-10.1 min, 70%-92% A; 10.1-15 min, 92%A) at a flow rate of 0.2 mL·min-1. The injection volume was 2 μL, and the column temperature was 60℃. In mass spectrometry, electrospray ion source was used:multiple reaction ion monitoring mode (MRM) under the negative ion. Turbo V ion source parameters were common to all analytes, as follows:the capillary voltage was -4.5 kV, and the source temperature was at 550℃. The curtain gas and collision gas setting were 30 psi and 10 psi, respectively. The pressure for nebulization gas and vaporization gas setting were 55 psi. The entrance potential (EP) and collision cell exit (CXP) were all -10 V. Result: Eight monosaccharide components were separated clearly within 12 min. The RSD values of precision, stability and reproducibility were all less than 2.1%. The average recoveries were 99.3%-102.5%, with RSD of 1.5%-2.9%. The developed method has been successfully applied to determine the major saccharides in 5 Ephedra Radixet Rhizoma polysaccharide samples. It was clear that the predominant composition monosaccharide in Ephedra Radixet Rhizoma polysaccharides were Rha, Ara, Man, Gal and GalUA. Conclusion: The established method for determination of the eight monosaccharides is stable, reliable, accurate, and reproducible, its sample pre-treatment is simple, without derivatization, and can be used for rapid detection of monosaccharide compositions in plant polysaccharide.

      摘要:Objective: To establish a UPLC method for simultaneous determination of 7 compounds (epidemin A1, epidemin A, epidemin B, epidemin C, icariin, 2-O-rhamnosyl-icariside Ⅱ and baohuoside I) in Yinyanghuo Zonghuangtong capsule (YZC) by quantitative analysis of multi-components with a single-marker (QAMS) and to evaluate the adaptation and application of QAMS method in the quality control of YZC. Method: Icariin was used as the internal reference substances and the relative correlation factors (f) of epimedin A1, epimedin A, epimedin B, epimedin C, rha-icariside Ⅱ and baohuoside Ⅰwere calculated. Result: The relative correlation factors (f) were 1.413, 1.231, 1.375, 1.166, 0.962 and 0.689 for epimedin A1, epimedin A, epimedin B, epimedin C, rha-icariside Ⅱ and baohuoside Ⅰrespectively. The determination results were compared with those obtained by the external standard method. The reproducibility of the established method was satisfying and there was no significant difference in assay results between the QAMS method and the external standard method. Conclusion: The QAMS method was reliable and could be used for quality control of YZC.

      摘要:Objective: To establish a HILIC-UPLC-MS/MS for determination of 11 nucleosides and nucleobases in Ziziphi Spinosae Semen. Method: Acquity UPLC BEH Amide (2.1 mm×100 mm, 1.7 μm) was adopted as chromatographic column, with Amide 1.7 μm VanGuard as the guard column; the mobile phase consisted of acetonitrile (containing 0.1% acetic acid) and water (containing 0.8% acetic acid and 10 mmol·L-1 ammonium acetate) at a flow rate of 0.3 mL·min-1; the temperature was set at 35℃; sample size was 1 μL. Waters XevoTM TQ worked in MRM mode. Result: The Tested components were separated in 7 min. All calibration curves were linear (r>0.992 2) within the tested range between 0.002 32-8.66 mg·L-1. The average recoveries ranged from 92.15% to 108.23%, with RSD value below 7%. The contents of these batches of nucleosides and nucleobases were obviously different. The mass fraction of all of the target compounds ranged between 24.5-48.48 μg·g-1. Sample No. 10 had the lowest content, while sample No. 2 had the highest content. Uridine was found to be the highest content in all samples (mean:14.84 μg·g-1). Furthermore, the content of thymidine was observed to be lower than others in all markers (mean:0.14 μg·g-1). Conclusion: The method was simple and fast, with a high precision, sensitivity and repeatability, which can be used for qualitative and quantitative analysis for nucleosides and nucleobases in Semen Ziziphi Spinosae.

      摘要:Objective: To establish a method for quality control of the Ershiwuwei Zhenzhu Wan. Method: Qualitative analysis was conducted by wavelength dispersive X ray fluorescence spectrometer (WD-XRF), and inductively coupled plasma-optical emission spectrometer (ICP-OES) was used to determine the contents of Al, Ca, Fe, K, Mg, Na in the 4 types of Ershiwuwei Zhenzhu Wan and 27 kinds of prescription materials. Then the source of the mineral elements was traced from Tibetan materials according to the prescription compositions. Result: Mineral elements were mainly derived from pearl, nacre mother of pearl, lapis micae aureus, travertine, crab, calculus bovis, ajowan, cluster mallow fruit, musk and other materials. All the medicinal materials complemented and were compatible with each other. Calcium content was (6.64±0.03)%, which could fully reflect the main effective components in Tibetan medicine and could be used as an index for controlling the quality of Ershiwuwei Zhenzhu Wan.The regression equation of calcium was Y=6.30X-1.02 (r=0.999 9), and its linear range was 12.5-5 000 μg, with an average recovery rate of 98.1%(RSD of 1.7%). Conclusion: The method could be applied for the quality control of the Ershiwuwei Zhenzhu Wan to ensure its effectiveness, providing a new research method for the quality control of Tibetan medicine.

      摘要:Objective: To establish a method of UPLC-Q-TOF/MS for determination of tumulosic acid and dehydro-tumulosic acid in rat plasma after oral administration of Poria triterpenoids extract powder and soft capsules content,differences of pharmacokinetics of tumulosic acid and dehydro-tumulosic acid in two forms were compared. Method: Liquid-liquid extraction was adopted to extract plasma samples with ethyl acetate,taking glycyrrhetic acid as internal standard,good chromatographic separation was achieved with the mobile phase consisting of 0.1% acetic acid in acetonitrile and 0.1% acetic acid in water for gradient elution.Mass conditions were as following:positive sensitive mode,capillary voltage of 3 kV,ion source temperature of 120℃,desolvent temperature of 500℃ and desolvation gas flow of 800 L·h-1. Result: Cmax and AUC0-t of tumulosic acid and dehydro-tumulosic acid in soft capsules content were greater than those of powder.Tmax of tumulosic acid in soft capsules content was longer than that of the powder,but Tmax of dehydro-tumulosic acid was lower than that of powder. Conclusion: These results indicate that vegetable oil can promote the absorption of tumulosic acid and dehydro-tumulosic acid.It is probably that the different form of Poria triterpenoids extract lead to the change of absorption rate due to the different chemical structure of tumulosic acid and dehydro-tumulosic acid.

      摘要:Objective: To investigate the protein and mRNA expression levels of mitochondrial respiratory chain complex Ⅳ A(COX VA) in gastric tissues of functional dyspepsia (FD) rats with spleen deficiency and explore the effect of Pixu I recipe intervention. Method: Seventy 10-day-old male SD rats were randomly divided into normal group, FD model group (single model group), spleen deficiency type FD model group (dual model group), Domperidone group, Pixu I recipe low dose group, medium dose group, and high dose group. The rats in FD model group and spleen deficiency type FD model group were gavaged with 0.2 mL of 0.1% IA in 2% sucrose for 6 days. Normal feeding was given to the rats in spleen deficiency type FD model group till 6-week-old, and then modified multiple platform method (MMPM) was used for 14 days. After successful modeling, they were gavaged with 10 mL·kg-1·d-1 distilled water, 3.125 mg·kg-1·d-1 Domperidone, 1.275, 2.55, 5.1 g·kg-1·d-1 Pixu I recipe for 14 days. COX VA mRNA and protein expression levels in liver tissues were detected by immunohistochemistry, Western blot and Real-time PCR. Result: As compared with the normal group, the mean optical density value of COX VA proteins was increased in high-dose Pixu I recipe group (P<0.05); COX VA protein expression levels were decreased to different degrees in Domperidone group, low-dose and medium-dose Pixu I recipe groups (P<0.01), while the COX VA protein expression level in high-dose Pixu I recipe group was increased; the increase in COX VA mRNA expression was most obvious in single model group (P<0.01). As compared with the dual model group, COX VA protein expression levels were decreased to different degrees in Domperidone group, low-dose and medium-dose Pixu I recipe groups (P<0.01), and the mRNA expression levels were also decreased while the levels in high dose group and single model group were increased without statistically significant differences. Conclusion: COX VA mRNA and protein expression levels were decreased in spleen deficiency type FD rats, and Pixu I recipe could increase the COX VA protein expression level and improve energy metabolism.

      摘要:Objective: To investigate the effect of matrine on the apoptosi of human colon cancer cell line SW480, and to explore its underlying mechanism. Method: The effect of matrine on SW480 cell line proliferation was detected by methylthiazolyldiphenyl-tetrazolium bromide(MTT) assay, its effect on SW480 cell apoptosis was detected by Hoechst33258 staining and flow cytometry was used to determine the apoptosis rate. Protein levels of proteinkinase B (Akt), p-Akt, B-cell lymphoma-2 (Bcl-2), and Baxin SW480 cells were detected by Western blot. Result: Matrine(0.25, 0.5, 1.0, 1.5, 2.0 g·L-1) inhibited SW480 cell proliferation in time and dose-dependent manners. The half maximal inhibitory concentration(IC50) was 1.72, 1.08, 0.67 g·L-1 respectively for 24, 48, 72 h. Hoechst33258 staining and flow cytometry showed that matrine could significantly induce apoptosis of SW480(P<0.01). The apoptosis rate was gradually increased with the increase of drug concentration. Western blot results showed that after adding matrine in the SW480 cells, the expression levels of p-Akt and Bcl-2 were down-regulated (P<0.05), while the expression level of Bax was up-regulated and the expression level of total Akt wasn't affected. Conclusion: Matrine could induce apoptosis in SW480 cells, which may be related to the inhibition of Akt Signaling Pathway and the related downstream targets.

      摘要:Objective: To study the preventive mechanism of modified Buyang Huanwu Tang on formation of atherosclerosis. Method: Thirty male ApoE-/- mice were randomly divided into four groups:ApoE-/- mice blank group, model group, modified Buyang Huanwu Tang group and simvastatin group. Normal male C57BL/6J mice were used as a normal group. High-fat diet was given for modeling in model group, modified Buyang Huanwu Tang group and simvastatin group. 4 weeks later, the mice in Buyang Huanwu Tang group or simvastatin group were intragastrically administrated with Buyang Huanwu Tang (20 g·kg-1)+leech powder (0.46 g·kg-1) or simvastatin(3 mg·kg-1). After 9 weeks, the total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) in serum were detected. In addition, aorta segment was taken from the experiment animals to observe morphological changes by HE staining,measure fiber-cap thickness and vascular intima-media thickness. Western blot was used to determine the expression levels of aortic p38 mitogen activated protein kinase(p38MAPK) and extracellular signal-regulated kinase 5(ERK5). Result: As compared with the normal group, TC, TG and LDL-C levels were significantly increased in the ApoE-/-mice blank group and model group, while HDL-C level was significantly decreased (P<0.01). As compared with the model group, TC, TG and LDL-C levels were decreased in the modified Buyang Huanwu Tang group and simvastatin group, while HDL-C level was significantly increased (P<0.05, P<0.01). HE staining showed no aortic plaque formation in C57BL/6J mice normal group, few aortic plaque formation and slightly increased vascular intima-media thickness in ApoE-/- mice blank group, significant aortic plaque formation and increased vascular intima-media thickness in model group. As compared with the model group, aortic plaque formation and vascular intima-media thickness was reduced in both modified Buyang Huanwu Tang group and simvastatin group. The protein expression level of aortic p38MAPK in model group was higher while protein expression level of ERK5 was lower than those in C57BL/6J normal group (P<0.01). In addition, the protein expression level of aortic p38MAPK was significantly decreased while the protein expression level of ERK5 was increased in both modified Buyang Huanwu Tang group and simvastatin group as compared with model group (P<0.01, P<0.01). Conclusion: Both modified Buyang Huanwu Tang and simvastatin have an effect in preventing the development of atherosclerosis, and the potential mechanism could be related to intervening p38MAPK and ERK5 in MAPK signal pathway.

      摘要:Objective: To explore the mechanism of icariin in inhibiting the osteoclast differentiation via detecting the mRNA expression levels of estrogen receptor α(ERα) and receptor activator of NF-κB (RANK) in osteoclast induced from RAW264.7. Method: RAW264.7 cells were induced into osteoblasts with receptor activator of nuclear factor κB ligand (RANKL) (50 μg·L-1), and were identified by tartrate resistant acid phosphatase(TRAP) staining and bone resorption pit formation. After RAW264.7 cells were treated with different concentrations (1×10-5, 1×10-6, 1×10-7 mol·L-1) of icariin for 5 days and 9 days, TRAP staining, and bone resorption pit formation analysis were conducted. After treatment for 6 days, the gene expression levels of(MMP-9), cathepsin-K (CK), TRAP, RANK and ERα in osteoclasts were detected by RT-PCR. ICI182780, an estrogen receptor antagonist, was used to invalidate the regulatory effect of icariin on ERα/RANK pathway. Result: Icariin could significantly inhibit the number of osteoclast formation and number of bone resorption pits(P<0.05), significantly down-regulate the gene expression levels of MMP-9, CK, TRAP and RANK in the osteoclasts(P<0.05), and up-regulate the gene expression of ERα.ICI182780 could inhibit the icariin in up-regulating the gene expression level of ERα and down-regulating the gene expression level of RANK. Conclusion: Icariin could inhibit osteoclast differentiation and bone absorption function, which may be achieved via increasing ERα mRNA expression and decreasing RANK mRNA expression.

      摘要:Objective: To study the protective effects of Curcuma zanthorrhiza rhizome on liver injury induced by concanavalin A(Con A) in mice, and investigate its possible mechanism. Method: SPF mice were randomly divided into normal group, model group, biphenyl group, C. zanthorrhiza rhizome high dose group, middle dose group and low dose group. All the mice except those in normal group received Con A (30 mg·kg-1) by injection via tail vein to induce mice liver injury. The drugs were given for 7 days by gavage administration, and the mice in normal group received the equal volume of normal saline. After fasting (but water was given) for 8 h, their body weight was measured; blood was taken from eyeball, and the mice were killed by taking off the cervical vertebrae. Then their liver index, spleen index, contents of alanine aminotransferase (ALT), aspartate amino transferase (AST), and lactate dehydrogenase (LDH) in serum and total superoxide dismutase (T-SOD), malondialdehyde (MDA), nitric oxide (NO) in liver homogenates were measured. Hepatic tissue was observed by hematoxylin-eosin staining. Furthermore, real-time fluorescence quantitative PCR (Real-time PCR) and Western blot assay were used to detect mRNA and protein expression levels of liver tramsforming gravth factor-β1(TGF-β1), Smad3 and Smad7. Result: As compared with the normal group, the liver index, spleen index, levels of ALT, AST and LDH in serum as well as MDA and NO levels in liver homogenates were significantly increased in model group; T-SOD level in liver homogenates was significantly reduced; mRNA and protein expression levels of liver TGF-β1, Smad3 were significantly increased, while mRNA and protein expression levels of Smad3 were significantly reduced (P<0.01), with more obvious liver lesions in model group. As compared with the model group, both C. zanthorrhiza rhizome and biphenyl significantly reduced the liver index and spleen index; decreased the levels of ALT, AST, LDH in serum and MDA, NO levels in liver homogenates; and significantly increased T-SOD level in liver homogenates. The liver lesions were significantly relieved in C. zanthorrhiza rhizome high dose group, mRNA and protein expression levels of liver TGF-β1, Smad3 were reduced, while mRNA and protein expression levels of Smad3 were increased (P<0.05, P<0.01). Conclusion: Curcuma zanthorrhiza rhizome had significant protective effect for the liver injury induced by Con A, and its mechanism to block the progression of liver injury may be associated with multi-site affecting TGF-β1/Smad pathways in liver tissues.

      摘要:Objective: To observe the effect of Yiqi Huoxue Lishui decoction on necdin-Wnt signal pathway in rats with hepatic fibrosis, and explore its mechanismin treating hepatic fibrosis. Method: The 12 SD rats were selected from 60 SD rats as normal group, and 45 successfully modeled SD rats were randomly divided into Yiqi Huoxue Lishui decoction standard dose group(standard group, n=12), Yiqi Huoxue Lishui decoction high dose group(high dose group, n=11), colchicine group (Col group, n=11) and model group(n=11). The rats innormal group and model group received intragastric administration of normal saline, rats in standard group received intragastric administration of 9.6 g·kg-1·d-1Yiqi Huoxue Lishui decoction, rats in high dose group received intragastric administration of 14.4 g·kg-1·d-1Yiqi Huoxue Lishui decoction, and the rats in Col group received intragastric administration of 0.1 mg·kg-1·d-1colchicine. After ten weeks, the level of hyaluronic acid (HA), laminin (LN), type Ⅲ Procollagen(PC Ⅲ),and type Ⅳ collagen (C Ⅳ) in serum were observed in all the groups.Hematoxylin-eosin staining(HE) was used to observe their pathologic manifestation, and Masson staining was used to observe the collagen changes.The protein and mRNA expression levels of necdin, β-catenin, and peroxisome proliferator activated receptor γ(PPARγ) were observed. Result: As compared with the normal group, the levels of HA, LN, PCⅢ and CⅣ in serum, Metavir scores in fibrosis stage, content of collagen fibers, levels of necdin and β-catenin protein and mRNA expression levels were significantly higher in model group (P<0.01). As compared with the model group, the levels of HA, LN, PCⅢ and CⅣ, Metavir scores, content of collagen fibers, levels of necdin and β-catenin proteinand mRNA expression levels of necdin and β-catenin in Col group were significantly lower (P<0.01). As compared with the Col group, the levels of HA, LN, PCⅢ and CⅣ, Metavir scores, content of collagen fibers, levelsof necdin and β-catenin protein and mRNA expression levels were significantly lower in standard group(P<0.01). As compared with standard group, the levels of HA, LN, PCⅢ and CⅣ, Metavir scores, content of collagen fibers, levels of necdin and β-catenin protein and mRNA expression levels were significantly lower in high dose group(P<0.05,P<0.01). As compared with normal group, the PPARγ protein and mRNA expression levels were significantly lower in model group (P<0.01). As compared with the model group, the PPARγ protein and mRNA expression levels were significantly higher in Col group(P<0.01). As compared with Col group, the PPARγ protein and mRNA expression levels were significantly higher in standard group(P<0.01). As compared with the standard group, the PPARγ protein and mRNA expression levels were significantly higher in high dose group(P<0.05, P<0.01). Conclusion: The Yiqi Huoxue Lishui decoction can inhibit the activity of necdin-Wnt/β-catenin pathway, relieve the inhibition of Wnt/β-catenin on PPARγ, and have the effect of antihepatic fibrosis.

      摘要:Objective: To study the protective effect and mechanism of methyl helicterate isolated from Helicteres Radix on chronic liver injury induced by carbon tetrachloride (CCl4) in rats. Method: The rats were randomly divided into 5 groups, including normal control group, model group, silymarin positive control group(50 mg·kg-1), and high and low-dose methyl helicterate (200, 50 mg·kg-1) groups. Except for the normal control group, all of the other groups were intragastrically administrated with 20% CCl4 for 8 weeks,twice a week. Meanwhile, the animals in the positive control group and methyl helicterate-treated groups were given silymarin (50 mg·kg-1) and methyl helicterate (200, 50 mg·kg-1) by gavage once a day for 8 weeks. After the experiment, the levels of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin (ALB) and plasma interleukin-6 (IL-6), tumor necrosis factor -α (TNF-α) were detected by enzyme-linked immuno sorbent assay(ELISA) kits. The hepatic pathological changes were observed by HE staining. And the levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GSH-Rd), NF-κB p65 in liver tissues were detected by ELISA kits. Western blot assay was performed to detect the expression of nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), Caspase-3, Caspase-8 Fas and FasL in liver tissues. Result: Methyl helicterate significantly alliviated CCl4-induced serious liver injury, decreased the levels of serum ALT, AST and ALB, while increased the activities of hepatic SOD, GSH-Px and GSH-Rd. Methyl helicterate also reduced the contents of inflammatory cytokines TNF-α and IL-6 in liver tissues. The further study showed that methyl helicterate significantly inhibited the vitality of NF-κB,iNOS and COX-2, which played a crucial role in regulat inginflammatory response.In addition, methyl helicterate significantly suppressed the expressions of Caspase-3, Caspase-8, Fas and FasL, and reduced the hepatocytes apoptosis. Conclusion: Methyl helicterate alleviates liver injury induced by CCl4 in rats, the mechanism may be associated with the inhibition of NF-κB signaling pathway and the decrease in cell apoptosis.

      摘要:Objective: To analyze the compositions of Chinese herbal prescription Jiuwei Tongqiao decoction and its pharmacodynamic in vitro, explore its poaaile mechanism and provide experimental basis for its effectiveness. Method: Jiuwei Tongqiao decoction was separated into seven prescriptions based on the principles of Jun, Chen, Zuo, Shi. After alcohol extract enrichment, U251 cells of anthropogenic brain glioma were used inmethylthiazolyldiphenyl tetrazolium bromide (MTT) assay to determine the effect of different prescriptions on cell survival. Flow cytometry instrument was used to compare the effect of optimal prescription Ⅳ (10, 4,1 g·L-1) and whole prescription on cells apoptosis. Western blot was used to detect apoptosis-related protein B-cell lymphoma-2 (Bcl-2) and p53 expression differences between the optimal prescription Ⅳ and the whole prescription. Alcohol extraction of the optimal prescription Ⅳ was concentrated with petroleum ether, chloroform and ethyl acetate, and then MTT was applied to determine the effect of various extraction parts on cell survival. Result: Prescription Ⅳ had the most obvious inhibition effect on U251 cells in a concentration-dependent manner. The apoptosis ratesinprescription Ⅳ treatment group (10, 4,1 g·L-1)and whole prescription were all significantly higher than those in blank group after 24 h (P<0.01). The Bcl-2 protein expression levels of prescription Ⅳ group and the whole prescription group were significantly lower than those in blank group (P<0.01), and p53 protein expression levels were significantly higher(P<0.01). The directive ingredients in part Ⅲ of prescription Ⅳ were more comprehensive after gradient extraction. Conclusion: Prescription Ⅳ is the optimal prescription, and part Ⅲ is the most effective part with the anti-proliferative effect on U251 glioma cells.

      摘要:Objective: To investigate the effect of Miao medicine Jinwu Jiangu decoction on the expression levels of nuclear transcription factor-κB (NF-κB) p65, IκB kinase-α (IKK-α) and IκB kinase-β (IKK-β)in the fibroblast-like synoviocyte of rheumatoid arthritis (FLS of RA) and explore its mechanism. Method: FLS of RA models were established in vitro through culturing, and the second to third generation cells were randomly divided into 6 groups:blank group, tripterygium wilfordii group, prednisolone group, Jinwu Jiangu decoction high, middle and low dose treatment groups(28.8, 9.6, 3.2 g·kg-1). After treatment for 24 h, the morphological changes were observed in all groups. Then flow cytometry was adopted to detect the apoptosis rate of the processed fibroblast-like synoviocyte of RA and Western blot was used to detect the expression levels of NF-κB p65, IKK-α and IKK-β of the fibroblast-like synoviocyte of rheumatoid arthritis. Result: As compared with the control group, the apoptosis rate of the fibroblast-like synoviocyte of rheumatoid arthritis was gradually increased with the higher concentration of Jinwu Jiangu decoction (P<0.05), and the expression levels of NF-κB p65, IKK-α and IKK-β were significantly decreased (P<0.05). Conclusion: Miao medicine Jinwu Jiangu decoction can induce the apoptosis of fibroblast-like synoviocyte of rheumatoid arthritis, and reduce the expression levels of NF-κB p65, IKK-α and IKK-β in fibroblast-like synoviocyte of rheumatoid arthritis.

      摘要:Objective: To study the effect of Huangqi Guizhi Wuwu Tang on CD4+/CD8+and related cytokines in rats with Yang deficiency cold coagulation type osteoarthritis. Method: The 50 SD rats were randomly divided into normal group, model group, celecoxib group, Huangqi Guizhi Wuwu Tang 7 days group and Huangqi Guizhi Wuwu Tang 14 days group, n=10 in each group. All the rats except those in the normal group received frozen and physical stimulation methods for 6 weeks to establish Yang deficiency cold coagulation type osteoarthritis models. Before successful modeling, the rats in all groupsreceived gastric administration of corresponding drugs or saline:7 days for Huangqi Guizhi Wuwu Tang 7 days group, 14 days for Huangqi Guizhi Wuwu Tang 14 days group (30 g·kg-1), and 7 days for Celecoxib group (20.82 mg·kg-1). In addition, the rats in normal group and model group received normal saline for 7 days equally. After modeling, the expression levels of (iNOS), collagen type Ⅱand (TGF-β1) in knee joints were detected by immunohistochemical method. Blood was taken to test the levels of CD4+/CD8+by using flow cytometry, and detect the levels of interleukin-4(IL-4), C-reactive protein(CRP), insulinlike growth factors(IGF) and hypoxia inducible factor-1α (HIF-1α) by using ELISA method. Result: As compared with the normal group, the ratio of CD4+/CD8+in model group was decreased (P<0.05). As compared with the model group, the expression levels of CD4+and the ratio of CD4+/CD8+were increased in Huangqi Guizhi Wuwu Tang 14 days group and celecoxib group (P<0.05). As compared with the normal group, the expression levels of IL-4, CRP, iNOS and HIF-1α were increased in model group (P<0.05), while the expression levels of Collagen type Ⅱand TGF-β1 were decreased (P<0.05). As compared with the model group, the expression levels of Collagen type Ⅱand TGF-β1 were increased in various treatment groups (P<0.05), while the level of IL-4 was decreased (P<0.05); meanwhile, the expression levels of CRP, iNOS and HIF-1α were decreased in Huangqi Guizhi Wuwu Tang 14 days group (P<0.05), and the expression levels of CRP and iNOS in celecoxib group were also decreased (P<0.05). As compared with celecoxib group, the expression level of collagen type Ⅱwas decreased in Huangqi Guizhi Wuwu Tang 7 days and 14 days groups (P<0.05), and the expression level of iNOS in Haungqi Guizhi Wuwu Tang 7 days group was increased (P<0.05). There was no significant difference in IGF level among 5 groups. Conclusion: Huangqi Guizhi Wuwu Tang can regulate the immunosuppressed state and has positive function in repairing the broken chondrocyte to osteoarthritis through the methods of regulating the changes of IL-4 and its downstream cytokines iNOS, HIF-1α and TGF-β1.

      摘要:Objective: On evaluate the effectiveness and safety of compound Xiaoyao oral,compound Dihuang oral in the treatment of female precocious puberty patients. Method: The 140 girls who were were diagnosed with hyperactivity of liver-fire and Yin deficiency and Huo exuberance treated at out-patient of our hospital from Sep. 2013 to Aug. 2015.Hyperactivity of liver-fire group were randomly assigned to two groups,the control group was given to Xiaoyaowan,the experimental group was given to compound Xiaoyao oral which was our hospital self-made traditional Chinese medicine. Yin deficiency and Huo exuberance were also randomly assigned to two groups,the control group was given to Zhibai Dihuang Wan,the experimental group was given to compound Dihuang oral. After 3 months treated,all patients were observed the breast changes,uteruses,ovarians volume change and the level of luteinising hormone(LH),follicle-stimulating hormone(FSH),estradiol(E2),sex hormone binding globulin(SHBG)to evaluate its efficacy. At the end day,recording side effect of drug events; blood routine,liver function,renal function to evaluate its safety. Result: The treatment group was better than control group,the breast diameter and the reduction of uterine and ovary volume were changed significantly(P<0.05). In both of two groups the incidence of adverse reactions and no side effect of drug events were found. Conclusion: The compound Xiaoyao oral,compound Dihuang oral treatment of idiopathic true precocious puberty were significant clinical effect and safety reliable.

      摘要:Objective: To investigate the effect of Kangliu Zengxiao formula combined with gefitinib in treating elderly non-small cell lung cancer and on inflammation factors, level of T cell subsets and serum tumor markers. Method: The 84 cases of elderly non-small cell lung cancer were selected, and the patients or their family members signed the informed consent toactively cooperate with the study. They were divided into 2 groups. The 42 patients in control group were treated with gefitinib alone, while the 42 patients in treatment group received Kangliu Zengxiao formula. Quality of life, inflammatory cytokines and T lymphocyte subsets, and the clinical efficacy and adverse reactions were observed and compared. Result: The control group was inferior to treatment group in effective rate(P<0.05). Compared with before treatment,survival quality, inflammatory cytokines and T cell subsets level changed in both groups. Compared with the control group, body function, role function, social function, emotional function score, levels of interleukin-2 (IL-2),interleukin-12(IL-12) and interferon-γ(IFN-γ) were higher in treatment group, with lower CD8+ level, and CD4+ and CD4+/CD8+ levels were higher in control group(P<0.05). After treatment, vascular endothelial growth factor(VEGF) increase dinthe control group, whereas treatment group showed decreased VEGF (P<0.05). Compared with before treatment, the two groups showed no statistically significant difference in carcino embryonic antigen (CEA), carbohydrate antigen-125(CA125) and cytokeratin 19 fragments(CYFRA21-1), with no inter-group statistical significance. Adverse reactions in two groups were mild, with no adverse drug reaction. Conclusion: Kangliu Zengxiao formula combined with gefitinib in the treatment of elderly non-small cell lung cancer was effective, and can improve the life quality and immune function.

      摘要:Objective: To discuss the clinical efficacy of Compound Zhenzhu Anchuang Tablets Combined with modified Diandao powder in treating adolescent facial acne with spleen-stomach damp-heat syndrome, and its effect on facial skin. Method: One hundred and sixty-four patients were randomly divided into control group (81 cases) and observation group (83 cases) by random number table. Both groups got grading treatment according guidance, grade I:adapalene gel for external use, 2 times/day, grade II and Ⅲ:added benzoyl peroxide gel, 2 times/day, grade Ⅲ:added doxycycline hydrochloride tablets, 100 mg/time, 2 times/day. Patients in observation group took compound Zhenzhu Anchuang tablets, 4 tablets/day, 3 times/day. In addition to the therapy of control group, the control group took modified Diandao powder mask, 1 time/day for 15 day, which later changed to 1 time for every two days. The courses of treatment were 8 weeks. And both groups got guidance of health education during the treatment. Before and after treatment, the numbers of facial acne, papules and pustules were recorded. Water content of cuticle, sebum secretion and pH were detected by VISIA skin detecting instrument. Degree of erythema and chromatosis were evaluated. And quality of life before and after treatment was evaluated by specific quality of life scale. Result: After treatment, the total effect rate in observation group was 93.98%, which was higher than 82.72% in control group, with statistically significant difference (χ2=5.074, P<0.05). After treatment, sebum secretions at middle part of brow, right cheek, right side of the nose and jaw in the two groups decreased (P<0.01), which was more obvious in observation group (P<0.01). pH at the above four skin areas of the two group increased, particularly in observation group (P<0.01). And water content of cuticle at the above four skin areas of the two group decreased, and water content of cuticle at cheeks and nose of observation group were lower than those in control group (P<0.01). After treatment, skin lesion score in observation group was lower than that in control group (P<0.01). After treatment, scores of self-perception, emotive function, acne symptoms, social function and quality of life were increased and higher than those in control group (P<0.01). Scores of erythema and chromatosis in observation group were lower than those in control group (P<0.01). The incidence of facial irritant reaction in control group was 59.26%, which was higher than 36.14% in observation group (χ2=8.781, P<0.01). According to Ridit analysis, skin irritation reaction in observation group was lower than that in control group (P<0.05). Conclusion: In addition to dietary guidance and emotional regulations, compound Zhenzhu Anchuang tablets combined with modified Diandao powder had a good clinical effect and safety, and can promote resolution of erythema and pigmentation, improve skin texture and recovery of facial skin, and thus being worth clinical promotion.

      摘要:Objective: To discuss the efficacy of Yiqi Yangyin Tongbi prescription for rheumatoid arthritis and pulmonary interstitial lesions (RA-ILD) and investigate its effects on serum matrix metalloproteinases-9 (MMP-9), MMP-1, transforming growth factor-β1 (TGF-β1) and surface antigen of Ⅱ type alveolar table cells (KL-6). Method: One hundred and twenty patients were randomly divided into control group (60 cases) and observation group (60 cases) by random number table and randomized block design. Patients in control group orally took celecoxib capsules, 0.2 g/time, 2 times/day.Cyclophosphamide tablets, 100 mg/time, 1 time/every 2 days.Prednisolone tablets, 30 mg/time, 1 time/day on every morning, and the dose was decreased after taking effect. Based on the treatment of control group, patients in observation group added Yiqi Yangyin Tongbi prescription, 1 dose/day. The treatment course was 6 months for both groups. Before and after treatment, high resolution-CT (HRCT) was conducted and graded, and symptoms and signs were graded. Lung function, total lung capacity (TLC), forced vital capacity (FVC), forced vital capacity per minute (FEV1), maximal voluntary ventilation per minute (MVV) and carbon monoxide diffusion capacity (DLCO) were detected before and after treatment. Levels of MMP-9, MMP-1, TGF-β1 and KL-6 were detected before and after treatment. Result: After treatment, scores of HRCT, symptoms and signs of patients in observation group were lower than those in control group (P<0.01). Levels of TLC, FVC, FEV1%, MVV and DLCO in observation group were higher than those in control group after treatment (P<0.05, P<0.01).The levels of MMP-9, TGF-β1 and KL-6 were lower than those in control group, and level of MMP-1 was higher than that in control group (P<0.01). Conclusion: Yiqi Yangyin Tongbi prescription based on the immunosuppressive agents and hormone therapy can reduce score of HRCT, reduce area and degree of interstitial lung disease, ameliorate patients lung function, and relieve clinical symptoms. The mechanism may be associated with regulating levels of MMP-9, TGF-β1, KL-6 and MMP-1.

      摘要:Objective: To analyze the prescription rules of formulae containing Notoginseng Radix et Rhizoma in Chinese Pharmacopoeia and Pharmaceutical Standard of Ministry of Public Healthof the People's Republic of China-Chinese Patent Drug (hereinafter referred to as Chinese Patent Drug). Method: Through TCMISS(V2.5),formulae containing Notoginseng Radix et Rhizoma in Chinese Pharmacopoeia and Chinese Patent Drug were selected and collected to build the database. Frequency count, association rules and other data mining methods were used to analyze symptoms, syndromes and combination rules, and relevant in-depth analysis was made for high-frequency medicine pairs and syndromes. Result: There were a total of 197 prescriptions containing Notoginseng Radix et Rhizoma, involving 391 kinds of traditional Chinese medicines, 67 commonly used medicine pairs, mainly 14 symptoms and seven syndromes.According to the analysis for high-frequency medicine pairs of Notoginseng Radix et Rhizoma and Angelicae Sinensis Radix and Notoginseng Radix et Rhizoma and Carthami Flos and high-frequency syndromes of ‘Qi-stagnation and blood stasis’ and ‘Qi-deficiency and blood stasis’,Notoginseng Radix et Rhizoma had almost the same effect in different medicine pairs, but different effects to varied syndromes. Conclusion: The formulae containing Notoginseng Radix et Rhizoma in Chinese Pharmacopoeia and and Chinese Patent Drug have the effect of promoting the blood stasis and relieving the pains, and is usually combined with medcines for activating blood, reinforcing and regulating Qi. The prescription rules and characteristics provide basis for clinical R&D of new drugs. And drug combinations for ‘Qi-stagnation and blood stasis’ and ‘Qi-deficiency and blood stasis’ provides theoretical basis for studying the activating and tonifying effects of Notoginseng Radix et Rhizoma.

      摘要:Objective: Analyzing the ‘disease-syndrome-treatment-medicine’ experience of professor Ding for diabetes and its chronic complications by using Traditional Chinese Medicine(TCM) Inheritance Support System(V2.5). By analyzing the data, we summarize the Professor Ding's academic experience. Method: Through collecting the standard clinical cases,combined with the software analysis system, we analyze the composing principles by using datamining methods such as frequency statistics, revised mutual information, association rules analysis, cluster analysis, improvement of mutual information, complex system entropy clustering, unsupervised hierarchical clustering entropy and so on. Result: Characteristics of simple diabetes. Based on the analysis of 39 cases of simple diabetic ‘Yin deficiency and heat Syndrome’, the highest-frequency used drugs were Lycii Cortex,Rhizoma Coptidis, Dioscoreae Rhizoma, Rehmanniae Radix, Anemarrhenae Rhizoma and so on. The highest-frequency used drug combinations were Mori Folium, Rhizoma Coptidis;Mori Cortex, Rhizoma Coptidis; Rehmanniae Radix, Mori Folium, Rhizoma Coptidis and so on. Based on the analysis of 29 cases of ‘Qi and Yin deficiency syndrome’, the highest-frequency used drugs were Rhizoma Coptidis, Dioscoreae Rhizoma, Rehmanniae Radix, Lycii Cortex, Anemarrhenae Rhizoma and so on. The highest-frequency used drug combinations were Rehmanniae Radix, Lycii Cortex; Rehmanniae radix, Lycii Cortex, Rhizoma Coptidis; Lycii Cortex, Dioscoreae Rhizoma,Rhizoma Coptidisand so on. Conclusion: Treating simple diabetes focus on the ‘asthenia in origin and asthenia in superficiality’,‘remove heat and nourish Yin is important’, we often use herbs which aresweet-flavored and cold-natured.

      摘要:Autophagy is an energy metabolism and self-update mechanism for maintaining the intracellular homeostasis. It is involved in many physiological and pathological processes in cells. The recent studies showed that autophagy is closely related to tumorigenesis and development. It participates in the regulation of tumor formation, proliferation, metastasis and energy metabolism. Autophagy's regulatory effect on tumors is a complex dynamic process. Autophagy can impede early cancer development, while promoting advanced tumor progress. In recent years, autophagy has become a new hotspot in the field of cancer research. At present, more and more studies showed that Chinese herbal medicines and their effective components could regulate autophagy in cancer cells. In response to the complex role of autophagy in cancer development, traditional Chinese medicines and their derived anti-cancer compounds also have multiple effects in autophagy regulation. Some of them can cause autophagic cell death, while others may inhibit autophagy metastasis or enhance tumor sensitivity to anti-cancer drugs by restraining autophagy process. They may show distinct effects in autophagic death or protective autophagy in different types of tumor. At present, studies on anticancer compounds with regulatory effect on tumor autophagy mainly focus on saponins, alkaloids, polyphenols, flavone and lactone. Their antitumor effects can be achieved by inducing autophagic death or autophagy suppression. In this paper, the effect and mechanisms of anti-cancer herbal medicines in regulating tumor cell autophagy were reviewed to help develop antitumor herbal drugs targeting autophagy and provide a new direction for anticancer drug development.

      摘要:The syndrome of Qi deficiency and blood stasis is a common syndrome of coronary heart disease in clinic. The basic treatment principle is to supplement Qi and activate blood circulation. Qi supplementing and blood circulation activating recipes, like Buyang Huanwutang, Bazhentang, Qishen Yiqi pills, are commonly used in clinic, with a good clinical curative effect. Yiqi Huoxue decoction has multiple-level, multiple-target and multiple-link advantages in treating coronary heart disease, with flexible clinical composition and small side effect. Current literature researches about treating coronary heart disease with Qi deficiency and blood stasis mainly focus on summarization of therapeutic methods, curative effect observation and prescriptions, without in-depth research and summarization of mechanisms of prescriptions. In order to clarify the targets of Yiqi Huoxue decoction, we collected relevant domestic and foreign literatures in ten years, and summarized mechanisms of Yiqi Huoxue decoction in treating coronary heart disease with Qi deficiency and blood stasis syndrome as follows:improving heart function, resisting platelet aggregation, improving blood rheology, protecting vascular endothelium, reducing inflammatory reaction, regulating blood lipid and stablizing plaque, reducing oxidative stress, increasing blood flow of coronary artery. The definition of targets of traditional Chinese medicine(TCM) is not only beneficial to guide clinical medicine, but also useful to advance the development of TCM. This paper is a systematic review of Yiqi Huoxue decoction's mechanism in treating coronary heart disease with Qi deficiency and blood stasis syndrome, and combined the characteristics of TCM with the modern pharmacological researches to elucidate the mechanism of Yiqi Huoxue decoction. This will provide some references for further studies for therapies for coronary heart disease and their mechanisms. Strengthening of researches on the mechanism of TCM prescriptions may be an important direction for future researches for Yiqi Huoxue decoction in the treatment of coronary heart disease with Qi deficiency and blood stasis syndrome.

      摘要:Buzhong Yiqitang, which was created by LI Dong-yuan, has functions of tonifying middle-energizer and Qi, ascending up spleen-Qi and Yang, removing heat with sweet-warm herbs. In clinic, the decoction was used to treat spleen stomach Qi deficiency, middle-energizer sinking and other diseases related to spleen-stomach dysfunction. Modern researchers have expounded its chemical composition, pharmacological actions, mechanisms of action, quality standards and compatibility. There are a large number of papers on the compatibility discipline, but without any review summarization. This review aimed to summarize modern researches of compatibility discipline of Buzhong Yiqitang in the aspects of compatibility principle and chemical composition. The results showed the rational composition of Buzhong Yiqitang. That is to say, the lack of any single herb or herbal pair could impact the efficacy of the original prescription. In terms of the impact on chemical composition, both of the sovereign medicine and the minister medicine played a dominant role in increasing the content of hesperidin and the assistant medicine reduced the content of glycyrrhizic acid; with respect to the impact of pharmacological effectiveness, the sovereign medicine and the drug combination with tonifying Qi played the role in modulating immune, while the assistant medicines Cimicifugae Rhizoma and Buplenri Radix played a synergistic effect. Meanwhile, the drug combination with medicines for tonifying Qi and raising Yang played an irreplaceable role in modulating pathological state and metabolism of animals with insufficiency of the spleen. These results fully prove the basic theory of TCM and the theory of dosimetry. The study for compatibility principles of classical prescriptions is conducive to better interpreting the theory of the TCM and guiding clinical application and scientific researchers.

      摘要:To systematically collect the literature on Pheretima and its compound, and sort their pharmacological effects and mechanism on diabetic nephropathy. It was found that pheretima and its compound could improve the prothrombotic state in patients with diabetic nephropathy by inhibiting platelet activation, repairing vascular endothelial injure induced by high glucose, and inhibiting the formation of blood clots with the mechanism of anticoagulation, thrombolysis and fibrinolytic system balance adjustment. Secondly, in the early stage of diabetes, Pheretima could improve the tissue blood circulation and inhibit the occurrence and development of diabetic nephropathy by correcting abnormal blood rheology, reducing the viscosity of whole blood and plasma, reducing the content of fibrinogen in blood, inhibiting blood sedimentation and erythrocyte aggregation, and enhancing the capacity of deformation of red blood cells. Furthermore, Pheretima could improve microcirculation by adjusting blood lipid metabolic disorder, inhibiting the increase of triglycerides, cholesterol, low density lipoprotein cholesterol and very low density lipoprotein, and inhibiting the reduction of high density lipoprotein, thereby inhibiting diabetic microvascular disease-occurrence and development of diabetes nephropathy. More importantly, Pheretima and its compound could inhibit the proliferation of mesangial cells, degrade extracellular matrix, and prevent the occurrence and development of glomerulosclerosis by the ways of antioxidant and regulating cytokines associated with diabetic nephropathy (such as transforming growth factor-β1, connective tissue growth factor, interleukin-6, tumor necrosis factor-α). Conclusively, Pheretima has broad prospects as a potential drug therapy for diabetic nephropathy.
