      摘要:Objective: Using machine vision and electronic nose technology to identify Strychni Semen pieces and its processed products with different processing degree, and to explore correlation of its color, smell and internal components. Method: Machine vision and electronic nose was employed to obtain information of color and odor of Strychni Semen and its processed products, variation in processing process was analyzed by combining chemometric methods (principal component analysis and discriminant factor analysis).HPLC was adopted to determine contents of strychnine and brucine in pieces with different processing degree, correlation between value of color, odor and composition in different samples was analyzed by correlation analysis. Result: With deepening of processing degree of Strychni Semen, statistical analysis of samples concluded its processing color change present that hue H value became smaller, saturation S became larger, brightness V value became smaller;differentiation and identification of processed products can realize by principal component analysis (PCA) and discriminant factor analysis (DFA).Smell of different processing degree of Strychni Semen had significant different and showed some variation;with increasing of processing time, contents of strychnine and brucine in Strychni Semen decreased.Correlation analysis showed that there were significant correlation between color change (hue H and brightness V) and contents of strychnine and brucine, but correlation between odor changes and contents of these two components was not obvious. Conclusion: By objective evaluation of machine vision and electronic nose technology to color and odor can achieve identification between Strychni Semen and its processed products.Color as a control index in processing of Strychni Semen has certain scientific in traditional method.

      摘要:Objective: To establish an odor fingerprint map by electronic nose, and to explore odor qualitative identification method to distinguish different fermentation time, different manufacturers and different bile proportion products of Arisaema Cum Bile. Method: Eighteen batches of different bile proportion samples and twenty four different fermentation time samples were collected. The data was analyzed by chemometrics methods such as principal component analysis (PCA) and statistical quality control analysis (SQC). Result: Samples of different fermentation time could be obviously distinguished by PCA, SQC and radar chart. Different bile proportion products could be obviously distinguished by PCA, and the PCA distinguish index was high. Conclusion: Electronic nose can be applied to identify odor qualitative, it provides reference for the rapid identification of Arisaema Cum Bile.

      摘要:In current quality standard of Chinese herbal pieces, reference crude herb is one of standard material in its quality control, identification, inspection and so on of most Chinese herbal pieces are all described as the words ofthe same as herb.However, Chinese herbal pieces as a direct applied drugs, its description, active components, functions and indications and so on are different from that of crude herbs.Using quality standards of crude herbs to control quality of Chinese herbal pieces is unreasonable, unscientific and can not meet need of quality control of Chinese herbal pieces.Aimed at problems of some unreasonable items and lack of specificity in quality standards of Chinese herbal pieces, we proposed a thread that control pieces can be used as one of effective standard materials in quality control of Chinese herbal pieces on the basis of summary these relative articles.In this paper, we put forward strategies to corresponding problems after in-depth analysis and thinking on crude medicinal materials, supplement materials, processing technologies and other relative aspects in process of preparing control pieces.Furthermore, we also expected advantages of application and research directions in the future of control pieces.

      摘要:Fermented medicine is commonly used fermented processed products of traditional Chinese medicine, due to complexity of prescription form and fermentation process, quality control and assessment of fermented medicine are common problems faced in industry.Currently, quality standards of fermented medicine is not coincide with each other, but shows confusing, incomplete and lack of evaluation index to reflect fermentation characteristics of fermented medicine, resulting in many difficulties in quality control and evaluation of fermented medicine in actual production.Based on herbal textual research of fermented medicine, this paper is arranging existing common problems of quality standards situation and research progress of quality standards, such as prescription, preparation and characters are various in each standards;identification, examination and content determination are incomplete and lacking of evaluation index to reflect fermentation characteristics of fermented medicine and so on, thinking and discussing according to these problems.We consider that prescription, preparation and characters of quality standards of fermented medicine should be standardized firstly, then identification, examination and content determination should be completed, besides, evaluation index to reflect fermentation characteristics of fermented medicine should be built, thus providing ideas and references for unification, completion and improvement of quality standards of fermented medicine.

      摘要:Objective: To identify Physalis plants in Solanaceae by internal transcribed spacer 2(ITS2) sequence. Method: ITS2 sequence of Physalis and Physaliastrum medicinal plant samples was amplified by RT-PCR and sequenced.In order to expand scope of research topic, ITS2 sequences of related species were downloaded from GenBank.All sequences were spliced by CodonCode Aligner V3.0.1, ITS2 sequences were compared and analyzed by DNAMAN software.Related data analysis and processing was performed by MEGA 5.10 software and neighbor-joining tree was constructed.ITS2 secondary structure information was predicted by ITS2 database and its differences of samples was analyzed. Result: All samples were clustered into two branches, Physaliastrum was clustered into a single branch and the same as Physalis.Except a few samples, all species of both genus were clustered into a branch where all intra-species samples were clustered to one branch respectively and obviously distinguished, which showed monophyletic.Bootstrap support rates between each two were very high. Conclusion: ITS2 sequences have great potential in terms of systematic research and species identification between related species.

      摘要:Objective: To measure effect exerted by differing microwave drying methods on rehmaionoside A, rehmaionoside D and leonuride in fresh Rehmanniae Radix. Method: Contents of rehmaionoside A, rehmaionoside D and leonuride were determined by HPLC, Aichrombond-AQ C18 column (4.6 mm×250 mm, 5 μm), a mobile phase of acetonitrile-water (5:95) was used, flow rate was 1 mL·min-1, column temperature was set at 30℃, a UV detector was applied with wavelength of 203 nm.Taken freeze drying as a reference, influence of microwave drying on contents of index components in fresh Rehmanniae Radix was investigated. Result: Contents of rehmaionoside A was the highest, which was reached by 5 kW microwave to cut fresh Rehmanniae Radix into slices with thickness of 3-4 mm (0.249 mg·g-1);maximum contents of rehmaionoside D and leonuride were 3.461, 6.857 mg·g-1, respectively, which were reached by 3 kW microwave to cut fresh Rehmanniae Radix into slices with thickness of 1-2 mm. Conclusion: Microwave drying method has the biggest effect on the content of leonuride and the smallest effect on contents of rehmaionoside D.Adopting microwave drying method in process of preparing fresh Rehmanniae Radix slices is plausible, which provides technology of concocted processing integration of Radix Rehmanniae with some room for thinking.

      摘要:Objective: To investigate influence of different drying method on contents of antitumor ingredients in fresh leaves of Paliurus ramosissimus and optimize its extraction process. Method: HPLC-ELSD was adopted to determine contents of betulinic acid and paliurusene with mobile phase of 1% formic acid-acetonitrile with gradient elution and detection wavelength of 320 nm.Take comprehensive score of contents of betulinic acid and paliurusene as index, othogonal test was adopted to optimize extraction process with ethanol concentration, ethanol comsumption and extracting time as factors. Result: It was better for conservation of betulinic acid and paliurusene by freeze drying and drying in shade.Optimal extraction conditions was as follows:extracted twice with 4 times the amount of 95% ethanol, 48 h for per time;yield of betulinic acid and paliurusene were up to 4.61, 0.0774 mg·g-1. Conclusion: Freeze drying is the best drying method for P. ramosissimus, but considering cost and mass production, drying in shade is better.This optimized extraction process is stable and reasonable with high yield of active constituents.It can provide a reference for development and utilization of P. ramosissimus.

      摘要:Objective: To identify and explore the antimicrobial activity of the fungi (Cs-ZZ23) isolated from the Cordyceps sinensis collected from Guoluo, Qinghai province. Method: The morphology and multilocus phylogenetic analysis were used for identification of the fungi. Meanwhile, 6 strains were used as indicators to test the antimicrobial activity of this fungus using agar disk diffusion method and improved agar block method. Result: The morphological characteristics of this strain were similar to those of Mortierella alpina in M. sp., and the polygenes sequence analysis showed that the sequence homology was 99% between strain Cs-ZZ23 and the multiple strains of M. alpina.Therefore, this strain was determined as M. alpina and it exhibited antibacterial activity against E. coli, Bacillus subtilis and Pythium aphanidermatum. Conclusion: The strain Cs-ZZ23 is a kind of M. alpina and its secondary metabolites deserve to be further developed.

      摘要:Objective: To provide an important scientific basis for Ferula ferulaeoides germplasm resources and quality evaluation through the study on correlation between the volatile oil and the genetic diversity of F. ferulaeoides. Method: The content of volatile oil of F. ferulaeoide from different origins was determined, and the genetic diversity of 176 samples from 8 counties in Xinjiang was studied with inter-simple sequence repeat(ISSR) molecular marker technique, and then the grey relational analysis (GRA) was used to analyze the effect of genetic diversity on the volatile oil content of F. ferulaeoides. Result: The ISSR analysis of the F. ferulaeoides samples was carried out by using 15 primers, a total of 126 clear bands were amplified and 93 of them were polymorphic. The highest level of genetic diversity among the 8 populations was in Yumin County[percentage of polymorphic bands (PPB)=29.37%, Shannon's information index (I)=0.1643, Nei's diversity index (H)=0.1104], and the lowest level was in Qinghe County (PPB=19.05%, H=0.0705, I=0.1054). The genetic distance of F. ferulaeoides was greatest Shawan County and Fuyun County in Xinjiang (0.2199), with the lowest genetic identity (0.8026), indicating that these two areas had the farthest genetic relationship and great genetic differences. The genetic diversity of F. ferulaeoides calculated with H, I and PPB showed a strong correlation to the total contents of ginsenol, camphor, γ-eudesmol and volatile oils. Conclusion: The genetic variation was 63.08% in F. ferulaeoides between different populations, and 36.92% within the same population. The geographic distribution of F. ferulaeoides was significantly correlated with its genetic diversity, and the Nei's diversity index among the three factors had a greater impact on the volatile composition and content of F. ferulaeoide.

      摘要:Objective: To evaluate the effect of honey-processing on therapeutical basis of Astragalus Radix, and systemically study the honey-processing of Astragalus Radix from its chemical composition content and estrogen receptor action and with the crude product and fried product of Astragalus Radix as the control products. Method: The content of water-soluble extracts was determined by using cold-maceration method;content of polysaccharides was determined by phenol-sulfuric acid colorimetry method;the total protein content was determined by Dimethyl quinoline method;high performance liquid chromatography-tandem-Q-time of flight mass spectrometry (HPLC-Q-TOF-MS) was used to compare the chemical profile and content of main components. Luciferase assay was used to measure the effect on estrogen receptor. Result: An increasing level of the water-soluble extracts and a reducing level of polysaccharides were observed in honey-processed Astragalus Radix. Honey-processing kept the stability of astragaloside Ⅱ and reduced the destruction on astragaloside Ⅱ content caused by heating;significantly increased the content of isoflavone glycosides (calycosin-7-O-β-D-glucoside and ononin). The effect of honey-processing on estrogen receptor was slightly higher than that of crude product and much higher than that of fried-processing. Conclusion: Honey-processing can protect the component stability and increase the content of extracts, thus enhancing the medicinal activity of Astragalus Radix.

      摘要:Objective: To study the fingerprints of Pien Tze Huang by total quantum statistical moment (TQSM) and total quantum statistical moment similarity (TQSMS) method, further verify the relevance between the previously established methods, and provide basis for the quality control of Pien Tze Huang. Method: With Notoginsenoside R1, ginsenoside Rg1, ginsenoside Rb1and sodium taurocholic acid as the control products, RP-HPLC was used to establish the chromatographic fingerprints of Pien Tze Huang, and the correlation between TQSM and TQSMS of the fingerprints was analyzed. Result: There were 14 common characteristic peaks for 10 batches of Pien Tze Huang samples. By using TQSM method, the average values of AUCT, relative AUCT, λT, relativeλT, σT, relative σTwere calculated as 7.430×106 μV·sec, 1.002, 22.540 min, 0.972, 5.391 min, 1.065 and their RSD were 5.0%, 4.3%, 1.0%, 1.0%, 2.9%, 2.9% respectively. The average similarity was 0.943 and RSD was 1.9% compared with reference fingerprint by TQSMS. The correlation coefficient of relative λTand similarity was 0.980, P<0.01, and the correlation coefficient of relative σTand similarity was -0.683, P<0.05, with statistically significant difference. Conclusion: There was correlation between TQSM and TQSMS and this method was additive, couplable and statistical. Besides, the established chromatographic fingerprint was highly precise and reproducible, thus can be applied in the quality control of Pien Tze Huang.

      摘要:Objective: To study the chemical constituents from the aqueous extract of the fruits of Euodia rutaecarpa. Method: Compounds were isolated and purified by macroporous resin, silica gel, MCI resin, Sephadex LH-20, ODS column chromatography, and semi-preparative HPLC. Their structures were identified on the basis of physicochemical properties and spectral data. Result: Eleven compounds (including five phenolic acids and their glycosides, four flavonoids, one limonin glucoside and one alkaloid) were isolated from the aqueous extract of the fruits of E. rutaecarpa and identified as chlorogenic acid (1), 5-O-caffeoylquinic acid (2), 4-O-caffeoylquinic acid (3), coniferin (4), caffeic acid (5), quercetin 3-O-α-D-arabinopyranoside (6), hyperin (7), rutin (8), quercetin (9), limonin diosphenol 17-β-D-glucopyranoside (10), dehydroevodiamine (11). Conclusion: Compounds 2~4 and 6 were isolated from this genus for the first time. As confirmed by two-dimensional nuclear magnetic spectrum, the 1H-NMR spectral data of compound 10 was totally reported for the first time. In addition, by ultra high performance liquid chromatography-mass spectrometry (UPLC-MS), chlorogenic acid (1), 5-O-caffeoylquinic acid (2) and 4-O-caffeoylquinic acid (3) were identified from E. rutaecarpa var. officinalis and E. rutaecarpa var. bodinieri for the first time, whereas, hyperin (7) was identified from E. rutaecarpa var. officinalis.

      摘要:Objective: To improve preparation process of Jinying Huanggui decoction for increasing the content of total polysaccharides in this preparation. Method: Step of petroleum ether extraction in preparation process of Jinying Huanggui decoction had been deleted, changed 70%, 75% and 80% alcohol precipitation step to 80% alcohol precipitation by one step.Monosaccharide composition in total polysaccharides from Jinying Huanggui decoction was analyzed by GC-MS.Comparison of the content of total polysaccharides before and after process improvement was determined by phenol-sulfuric acid colorimetry. Result: Monosaccharide composition in total polysaccharides was composed of rhamnose, arabinose, xylose, mannose, glucose and galactose with molar ratio of 0.08:0.08:0.02:0.03:1.00:0.04;after process improvement, this molar ratio was 0.05:0.07:0.02:0.05:1.00:0.08.Before and after process improvement, average concentrations of total polysaccharides in Jinying Huanggui decoction were 11.52, 22.69 g·L-1, respectively. Conclusion: This method is simple, accurate and reproducible, it can be used for determination of total polysaccharides of Jinying Huanggui decoction.Process improvement of this preparation is in favor of increasing of total polysaccharides content.

      摘要:Objective: To determine the content of ligustroflavone in wine Ligustri Lucidum Fructus by near infrared spectroscopy. Method: Ligustri Lucidum Fructus was processed with ricewine from three different regions. With the content of ligustroflavone determined by HPLC as reference value, quantitative model of ligustroflavone content in wine Ligustri Lucidum Fructus was established by using near infrared spectroscopy combined with partial least squares (PLS). Result: The correlation coefficients (R2) of the quantitative calibration model for ligustroflavone was 0.96789;root mean square error of calibration (RMSEC) was 0.107;root mean square error of predication (RMSEP) was 0.103;root mean square error of cross validation (RMSECV) was 0.25669, and the number of main factors was 16. Conclusion: This method is convenient and accurate. The established models can realize fast determination of ligustroflavone in wine Ligustri Lucidum Fructus.

      摘要:Objective: To establish a method for rapid determination of the content of buddleoside in Chrysanthemi Indici Flos by Near Infrared Spectroscopy (NIRS) and partial least square (PLS). Method: The near infrared spectra of Chrysanthemi Indici Flos were collected. With the content of buddleoside determined by HPLC method as a reference value, the NIR spectra were then associated with the reference values to establish the quantitative prediction model of buddleoside in Chrysanthemi Indici Flos. Finally, the model was verified by unknown samples. Result: The correlation coefficients (R2) of the quantitative analysis model was 0.99909;the root mean square error of calibration (RMSEC) was 0.0136;the root mean square error of cross validation (RMSECV) was 0.03970;the correlation coefficients of predication (r) and the root mean square error of predication (RMSEP) was 0.9976 and 0.0185 respectively. Conclusion: This method of near infrared spectroscopy is simple, fast and accurate, and could be used for the rapid determination of the content of the buddleoside in Chrysanthemi Indici Flos of Henan province.

      摘要:Objective: To establish a new method for fast determination of forsythiaside A content in different Forsythiae Fructus by using near infrared spectroscopy, and provide new ideas for quality analysis of this Chinese medicine. Method: Content of forsythoside A in Forsythiae Fructus was determined by using HPLC method prescribed in 2010Chinese Pharmacopoeia, and simultaneously the near infrared spectroscopy was combined with partial least squares (PLS) to establish the quantitative calibration model of forsythoside A in different Forsythiae Fructus. Result: The correlation coefficients (R2) of forsythoside A in the quantitative calibration model was 0.99489;the validation coefficients (R2) was 0.9970;and root mean square error of calibration (RMSEC) was 0.182.The model had well prediction results. Conclusion: This near infrared spectroscopy method is convenient and accurate and suitable for rapid determination of the content of forsythoside A in different Forsythiae Fructus.

      摘要:Objective: To conduct qualitative identification analysis of different Forsythia Fructus with use of near infrared spectroscopy combined with discriminant analysis method. Method: Near infrared spectrograms of different Forsythia Fructus were collected, then multiplicative scatter correction method (MSC) and Savitzky-Golay (SG) smoothing method were used for spectral preprocessing. Qualitative analysis model was established under the optimal modeling wave band of 4065.57-8350.90 cm-1 and optimal number (10) for principal components, and then this model was validated with unknown samples. Result: The classification results of the qualitative model were consistent with the actual conditions, and the validation results were also satisfactory. Conclusion: This method is simple, rapid, accurate, and non-polluting, with great significance for resources saving and efficient use of energy during the quality inspection in Chinese herbs, tobacco and other industries.

      摘要:Objective: To establish a method for simultaneous rapid determination of berberine content and phellodendrine content in Phellodendron Bark. Method: With the berberine content and phellodendrine content determined in high performance liquid chromatography (HPLC) as the reference values, near infrared spectra of the samples were associated with the reference values above, and then the quantitative analysis models of berberine and phellodendrine were established by using partial least squares (PLS) method. Result: The correlation coefficient (R2) was 0.99713 and 0.98923 respectively for quantitative analysis model of berberine and phellodendrine. The root mean square error of calibration (RMSEC) was 0.0823 and 0.0215 respectively. The root mean square error of predication (RMSEP) was 0.0717 and 0.0166 respectively. The performance index (PI) was 95.6 and 93.5 respectively. Conclusion: The quantitative analysis model established by using near-infrared spectroscopy is rapid and nondestructive to determine the content of berberine and phellodendrine in Phellodendron Bark.

      摘要:Objective: To establish a rapid and environmentally-friendly method to determine the moisture content in Ligustri Lucidi Fructus by using near infrared spectroscopy (NIRS) combined with partial least squares (PLS). Method: The reference values of water content in samples were obtained by oven-drying method, and the near infrared spectral data of the samples was collected by Nicolet 6700 NIR spectrograph. Then the quantitative analysis model of moisture content in Ligustri Lucidi Fructus was established by partial least square (PLS) and this model was validated by unknown samples. Result: The correlation coefficient of cross-validation (R2) of the established quantitative model for moisture content was 0.98687;the root-mean-square error of calibration (RMSEC) was 0.199;the root-mean-square error of cross-validation (RMSECV) was 0.24641.The validation set prediction correlation coefficient (R2) was 0.9566, and the root-mean-square error of prediction (RMSEP) was 0.220. Conclusion: This method is simple, rapid, accurate, and non-polluting to determine the content of moisture in Ligustri Lucidi Fructus.

      摘要:Objective: To investigate absorption characteristics of six components from anti-inflammatory effective parts of Smilacis Chinae Rhizoma. Method: Taking rat everted gut sac model, intestinal sac contents were collected at different time and absorption concentration of polydatin, astilbin, oxyreverstroal, quercitrin, engeletin and resveratrol was determined by HPLC, absorption kinetics parameters were calculated for describing their absorption characters in different intestinal segments (duodenum, jejunum and ileum). Result: Absorption of polydatin in duodenum, jejunum and ileum was no significant difference, while the other five components showed large differences.Absorption of six ingredients showed significant differences in the same intestinal site, their absorptivity at duodenum, jejunum and ileum were 0.85%-2.79%, 0.70%-2.73% and 0.90%-1.77%, respectively.Quercitrin showed the highest absorption rate in duodenum and jejunum, polydatin showed the highest absorption rate in ileum.Absorption rate of oxyreverstroal was significantly slower than the other five ingredients, it just showed obvious absorption after 90 min in duodenum. Conclusion: Absorption of polydatin, astilbin, quercitrin, engeletin and resveratrol conforms the first order kinetics, everted gut sac method can evaluate intestinal absorption characteristics of multi-component in Smilacis Chinae Rhizoma extracts.

      摘要:Objective: To develop UPLC-MS/MS for determination of 8-O-acetyl shanzhiside methyl ester and shanzhiside methyl ester in rat plasma and their pharmacokinetics study. Method: Plasma was precipitated with acetonitrile, mobile phase was consisted of acetonitrile and 0.1% formic acid solution with gradient elution pumped at a flow rate of 0.4 mL·min-1.Analysis was detected with positive electrospray ionization in multiple reaction monitoring (MRM) mode and rutin was used as internal standard. Result: Excellent linear calibration curves of 8-O-acetyl shanzhiside methyl ester and shanzhiside methyl ester were obtained in the concentration range of 1-250 μg·L-1, their lower limits of quantification were 0.2 μg·L-1.Intra-day and inter-day relative standard deviations of low, medium and high quality control samples were all less than 8.8%;Average recovery of 8-O-acetyl shanzhiside methyl ester were (103.89±9.18)%, (99.34±6.63)% and (95.88±3.69)%, while average recovery of shanzhiside methyl ester were (105.33±8.64)%, (101.55±1.22)% and (96.89±5.42)%, respectively. Conclusion: This method is simple, rapid and sensitive, which is suitable for pharmacokinetics analysis of 8-O-acetyl shanzhiside methyl ester and shanzhiside methyl ester in rat plasma.

      摘要:Objective: To extract effective components from Panax ginseng, P. quinquefolium, P. japonicas and P. notoginseng, and compare their inhibitory activities against ovarian cancer HEY cells. Method: Water and ethanol extracts of P. ginseng, P. quinquefolium, P. japonicas and P. notoginseng were prepared by reflux extraction with water and ethanol. Total saponins and their panaxadiol saponin (PDS) and panaxtrol saponin (PTS) fractions were prepared from 70% ethanol water extracts of above herbs using macroporous resin chromatography. 3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-H-tetrazolium bromide (MTT) assay was used to detect the activities of these extracts and fractions against HEY ovarian cancer cells. propidium iodide(PI) staining was used to detect the effect of extracts on cell cycle distribution, and western blot was performed to detect their effect on expression of cell cycle related proteins. In addition, scratch assay was conducted to detect the effect of extracts on cell migration. Result: All the indicated extracts showed varied activities against HEY cells. Among them, panaxadiol saponin of P. quinquefolium (PDSPQ), total saponin of P. japonicas (TSPJ), ethanol extract of P. ginseng (EEPG), total saponin of P. ginseng (TSPG), and panaxadiol saponin of of P. ginseng(PDSPG) showed relative higher inhibitory rate against HEY cells, with 45.59%, 45.78%, 50.48%, 46.98% and 64.36%, respectively. Further, PDSPQ, TSPG, and PDSPG could down-regulate the expressions of CDK4, CDK6, and cyclin D1 to different degrees, but had no significant effect on the migration of HEY cells. Conclusion: Fractions with PDS were more effective on inhibiting HEY cells growth, and the mechanism may be associated with down-regulating the protein expressions of CDK4, CDK6, and cyclin D1.

      摘要:Objective: To investigate the effect of safflower polysaccharide on proliferation and vascular endothelial growth factor (VEGF) expressions of human cervical cancer Hela cells, and discuss its molecular mechanism against cervical cancer. Method: Human cervical cancer Hela cells were cultured in vitro with different concentrations of safflower polysaccharide (0.02, 0.04, 0.08, 0.16, 0.32, 0.64, 1.28 g·L-1) for 48 hours. Their effect on Hela cells proliferation was assessed by 3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-H-tetrazolium bromide (MTT) assay. Hela cells were treated with different concentrations of safflower polysaccharide (0.16, 0.32, 0.64 g·L-1) for 48 hours, and then enzyme linked immunosorbent assay (ELISA) was used to detect the VEGF expression in cells supernatant;reverse Real-time PCR was used to detect VEGF mRNA expressions and Western blot method was used to detect VEGF protein expressions. Result: Compared with the blank group, safflower polysaccharide (0.16, 0.32, 0.64 g·L-1) could significantly inhibit the proliferation of Hela cells after 48 h treatment in a dose-dependent manner (P<0.05). Safflower polysaccharide could significant down-regulate mRNA expressions and protein expressions of VEGF of Hela cells, with significantly statistical differences (P<0.05). Conclusion: Safflower polysaccharide can inhibit proliferation of cervical cancer Hela cells and have its antitumor effect by inhibiting the expressions of VEGF.

      摘要:Objective: To optimize the hydrolytic process of antioxidant silk fibroin (SF) by pancreatin, and investigate the effects and mechanism of the resulting hydrolysates on H2O2-induced HepG2 cell injuries. Method: With the superoxide radical scavenging rate as index, the effects of reaction time, pH, temperature, substrate concentration and enzyme concentration on silk fibroin hydrolytic process were investigated by orthogonal tests. The molecular weight distribution of resulting hydrolysates, the scavenging rate of hydroxyl radicals and the inhibition rate of H2O2-induced erythrocyte hemolysis were determined. Cells were treated with different concentrations of H2O2 for 24 h, and then the optimal H2O2 concentration was selected to determine cell activity. Then cells were divided into blank group (without drug treatment), H2O2-induced group, H2O2 pretreatment+5 mmol·L-1 N-acetyl cysteine(NAC) treatment as positive control group, H2O2 pretreatment+10, 20, 30, 50 g·L-1 SF treatment groups. Changes in cells activity, malondialdehyde (MDA) content, tumor necrosis factor-α (TNF-α) content, superoxide dismutase (SOD) activity, catalase (CAT) activity and total antioxidant capacity (T-AOC) were observed. Result: Optimal conditions for SF hydrolysis in the study were as follows:enzyme concentration of 6%, reaction time of 160 min, substrate concentration of 20 g·L-1, pH of 8, and temperature 38℃. Under this hydrolysis process, the hydrolyzate's superoxide radical scavenging rate was 72.73%, and the molecular weight of hydrolyzate was below 10 kDa, with greatly increased hydroxyl radical scavenging rate and inhibition rate of H2O2-induced erythrocyte hemolysis. Cell activity was decreased and MDA content was increased after H2O2 induced cells injury (P<0.05). Cell activity was increased and MDA content was decreased in SF treatment group as compared with the H2O2-induced injury group (P<0.05). The TNF-α content was increased and the levels of SOD, CAT, and T-AOC capacity were decreased after H2O2 induced injury (P<0.05). SF treatment could reduce TNF-α content, increase SOD, CAT secretion and the T-AOC capacity(P<0.05). Conclusion: Study demonstrated that SF could improve H2O2-induced injury of HepG2 cells, and its mechanism may be associated with lowering TNF-α content and increasing autioxidant enzyme activities in cells.

      摘要:Objective: To evaluate the effects of curcumol on LoVo cells-planted xenogaft tumors in nude mice with colorectal cancer(CRC) and to investigate the possible mechanism. Method: Human CRC LoVo cells were implanted in BALB/c nude mice, then, tumor-bearing mice were randomly divided into five groups:blank group, model group, curcumol low dose group, middle dose group and high dose group (20, 40, 80 mg·kg-1, respectively). The drugs were administered by intragastric injection (21 days), then the expressions of vascular endothelial growth factors (VEGF) and cyclooxygenase-2(COX-2) in these tumor tissues were determined by reverse transcription-PCR (RT-PCR) and immunohistochemistry. Result: Compared with the model group, curcumol high dose group, middle dose group and low dose group significantly inhibited the tumor volume, tumor weight, and expressions of VEGF and COX-2 in a dose-dependent manner. The anti-tumor rate was 25.73%, 42.08%, 66.88% respectively in curcumol low dose group, middle dose group and high dose group. There was statistical difference in expression of VEGF between model group and all curcumol groups (P<0.05). There was statistically significant difference in COX-2 expression between model group and curcumol middle and high dose groups (P<0.05). Moreover, VEGF and COX-2 expressions were positively correlated with each other (r=0.874, P<0.01). Conclusion: Curcumol significantly inhibited tumor growth in CRC implanted mice, probably associated with down-regulating expressions of VEGF and COX-2, and may have a role in treating human colorectal cancer by the signal pathways of COX-2/PGE2/VEGF.

      摘要:Objective: To observe the inhibitory effect of Compound phyllanthus Urinsria Ⅱ(CPUⅡ) on formation of liver cancer in rats induced by diethylnitrosamine (DEN), and explore its anti-cancer mechanism. Method: Two hundred and forty SPF grade Wistar male rats were randomly divided into normal group, model group, CPU II high dose group (23.14 g·kg-1), low dose group (11.57 g·kg-1), and tegafur group. Rat models of liver cancer were induced by DEN for eighteen weeks, and medicine intervention was done at the same time according to the experimental design for eighteen weeks. 8 rats were sacrificed in each group after treatment for every 6 weeks, to observe the general condition, hepatic function index and nodule formation of liver cancer of the rats. Pathologic changes of liver tissues were observed under microscope. Radioimmunoassay method was used to determine the levels of interleukin(IL)-6 in serum of rats. Gene expression levels of mRNA and protein related to IL-6 signal pathway and microRNA let-7a regulatory network of the liver tissues were detected respectively by real-time fluorescent quantitative PCR and Western blot method. Result: At the end of eighteenth week, the number of nodules of liver cancer in rats of CPU II high dose group and low-dose group was significantly reduced as compared with that in the model group (P<0.05);levels of IL-6, ALT and AST in serum of rats in CPU II high dose group and low dose group were significantly decreased (P<0.05);the expression level of microRNA let-7a in liver tissues increased obviously in CPU II high dose group as compared with that in model group, while the mRNA expression levels of NF-κB-p65, IL-6, STAT3, RAS and c-myc genes were significantly decreased (P<0.05). The protein expression levels of STAT3 and p-STAT3 were also obviously reduced in CPU II high dose group and low dose group as compared with that in model group (P<0.05). Conclusion: CPUⅡcan effectively inhibit the formation of liver cancer in rats induced by DEN through inhibiting IL-6 expression and activation of IL-6/STAT3 signaling pathway, and its mechanism may be related with up-regulating microRNA let-7a expression and down-regulating NF-κB-p65 expression.

      摘要:Objective: To investigate the anti-tumor effect of Erdong Gao and study the effect of nuclear factor-κB (NF-κB) in Erdong Gao. Method: The dose-and time-related inhibitory effect of Erdong Gao in proliferation of A549 cells was determined by methyl thiazolyl tetrazolium (MTT) assay. Effect of Erdong Gao on the p65 mRNA expression and protein expression in A549 cells were detected by reverse transcription-PCR (RT-PCR) and Western blot respectively. Result: The MTT test indicated that the proliferation of A549 cells can be significantly inhibited by the drug serum of different concentrations of Erdong Gao (P<0.05), showing certain dose-dependence and time-dependence respectively. The cell proliferation was inhibited after 24 hours of drug intervention. The most obvious inhibitory effect was detected in Erdong Gao middle-dose group (3.2 g·kg-1, bid) after intervention for 48 hours. The p65 mRNA expression and p65 protein expression in A549 cells were obviously inhibited in Erdong Gao middle-dose group after intervention for 24 hours and 48 hours respectively. Conclusion: Erdong Gao can significantly inhibit the proliferation of A549 cells in human lung adenocarcinoma cells, and its anti-tumor effect might be mediated by inhibiting NF-κB.

      摘要:Objective: To investigate the effect of Notoginseng Radix et Rhizoma saponins, salidroside and effective components of Astragali Radix in mobilizing bone marrow mesenchymal stem cells in the rat model of myocardial infarction. Method: The SD rat model of myocardial infarction was established by Olivette method. The successfully modeled rats were randomly divided into:Huoxue group, Yiqi group, Yiqi and Huoxue group, model group and normal group, and then subdivided according to time points at 1, 3, 7, 14, 28 d. Huoxue group was given 8 mg·kg-1 of Xuesaitong soft capsule. Yiqi group were treated with 5 mg·kg-1 of Huangqi granule. Yiqi and Huoxue group was given 7 mg·kg-1 of Nuodikang capsule. Model group was given normal saline. Normal group received no drug. Normal group and model group were administered with equivalent normal saline by gavage once a day for 28 days. The positive cells percentage of CD54, CD106, CD105, CD117 in peripheral blood and bone marrow were detected by flow cytometry at different time points, and ELISA method was used to detect the expression of stem cell factor (SCF) in peripheral blood and bone marrow at different time points. Result: SCF level and positive cells percentage of CD105, CD117 content increased in model group since the first day, reached the peak on the 14th day, and then decreased gradually. But the increasing trend was superior in Huoxue group, Yiqi group, Yiqi and Huoxue group (P<0.05), particularly Huoxue group was the best. The prescription of promoting blood circulation to remove blood stasis showed an obvious mobilizing effect. Positive cells percentage of CD54, CD106 in peripheral blood and bone marrow in model group reached the peak on the first day, and then decreased gradually, but Huoxue group showed a superior decreasing trend in CD54, CD106 than model group in peripheral blood (P<0.05). Compared with model group, CD54 and CD106 in bone marrow had no significant change. Conclusion: Notoginseng Radix et Rhizoma saponins, salidroside and effective components of Astragali Radix can mobilize bone marrow mesenchymal stem cells, and the mobilizing effect of the prescription of promoting blood circulation to remove blood stasis is the most obvious.

Method: Hyperlipidemia animal model was set up in Wistar rats, and randomly divided into normal control group, hyperlipidemia model group, FN high, medium and low dose groups (200, 100, 50 mg·g-1), and Simvastatin butyl group (8 mg·kg-1). They were orally given drugs for six weeks. Biochemical method was used to detect totalcholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C) and high-densty lipoprotein cholesterol (HDL-C) content in rat blood and liver tissues. The steatosis model was established by exposing L-02 hepatocytes in fat emulsion, randomly divided into normal control group, model group, FN low, medium and high dose groups (100, 300, 500 μmol·L-1), and Simvastatin butyl group (100 μmol·L-1), and cultured for 24 h. The effects of FN on the contents and activities of key enzymes and proteins in the synthesis and decomposition pathways of TG and TC were investigated by biochemical method; ELISA was used to detect diacylgcerol acyltransferase(DGAT), 3-hydroxy-3-methylglutaryl-coenzyme(HMG-CoA), cholesterol 7a-hydroxylase(CYP7A) and lipase liver triglycerides(HTGL) contents. Result: In the rat experiment, compared with the normal group, the model group showed a higher blood lipid(P<0.01); compared with the hyperlipidemia model group, FN middle dose and high dose (300, 500 μmol·L-1) groups inhibited rat liver weight (P<0.05), reduced TC, TG, LDL-C content in serum (P<0.05, P<0.01), increased HDL-C content (P<0.05, P<0.01), and inhibited concentrations of TC and TG in liver (P<0.05). In the cell experiment, FN middle dose and high dose groups can significantly reduced the levels of TC, TG (P<0.05, P<0.01) and DGAT, HMG-CoA activity (P<0.01), and increased CYP7A, HTGL activity (P<0.05, P<0.01). Conclusion: FN can reduce blood lipid, which may be correlated with decrease in DGAT, HMG-CoA activity and increase in CYP7A, HTGL activity.

      摘要:Objective: To observe the comprehensive regulating effects and mechanism of Yiqi Huoxue recipe on the p38 mitogen-activated protein kinases(p38MAPK) signal pathway and impaired endothelial function of diabetic rats. Method: High fat and high glucose diet and streptozotocin (streptozotocin, STZ) were used to establish the model of diabetic rats. The rats were randomly divided into 7 groups:normal group, model group, Danzhi Jiangtang capsules high dose group, middle dose group and low dose group (1.08, 0.72, 0.54 g·kg-1·d-1), pioglitazone hydrochloride capsules group(10 mg·kg-1·d-1), Danzhi Jiangtang capsules+pioglitazone hHydrochloride capsules group (1.08 g·kg-1·d-1+10 mg·kg-1·d-1). The diabetic model rats were given with corresponding dose medicine by intragastric administration, once a day, for 8 consecutive weeks. Blood samples were collected from abdominal aorta for detection. Western blot assay was used to detect p38MAPK, mitogen activated protein kinase kinasse3/6(MKK3/6), cAMP response element-bingding protein1(CREB1) and mitogen-activated protein kinase phosphatase-1(MKP-1) protein expressions in endothelial cells. qPCR was used to detect the mRNA expression of monocyte chmotactic protein 1(MCP-1) and vascular endothelial cells Resistin in blood vessel endothelium. Result: Compared with the normal group, p38MAPK, MEK3/6, CREB1 protein expression and MCP-1, vascular endothelial cells Rresistin mRNA expression were significantly increased in the model group (P<0.01), while MKP-1 protein expression was significantly reduced (P<0.01). Compared with the model group, p38MAPK, MEK3/6, CREB1 protein expression and MCP-1, vascular endothelial cells Resistin mRNA expression were significantly reduced in various treatment groups, while MKP-1 protein expression was significantly increased, with statistically significant difference (P<0.05). Conclusion: Yiqi Huoxue recipe can reduce the vascular endothelium injury by regulating the expression of p38MAPK signal transduction protein blood vessels of diabetic rats.

      摘要:Objective: To investigate the protective effect of Shenxiong Glucose Injection (SGI) on H2O2-induced oxidative damage of H9c2 cells. Method: The in vitro oxidative stress damage model was established by treating H9c2 cells with H2O2(300 μmol·mL-1) for 0.5 h. SGI-pretreated group:pretreated H9c2 cells with SGI (100, 200, 400 μmol·mL-1) for 6 h. Blank group and model group were treated additionally with H2O2 for 0.5 h. The intracellular reactive oxygen species (ROS) and mitochondrial membrane potential (MMP) were detected by flow cytometry. The cell viability was detected by MTS assay;ELISA methods was used to determine LDH release, MDA content, SOD, CAT, GSH-PX activity. The expression of apoptosis-related genes Caspase-3, Bax and Bcl-2 was detected by western blotting technology. Result: After H9C2 cells was treated with H2O2(300 μmol·mL-1) for 0.5 h, the survival rate of the cells decreased to about 50%. The experimental results were so appropriate and repeatable that the subsequent oxidative damage model was established under the conditions. Compared with the control group, the cells survival was significantly increased (P<0.05, P<0.01) by pretreating with SGI for 6 h. SGI not only decreased the release of LDH (P<0.05, P<0.01), MDA (P<0.05, P<0.01), but also increased the activity of SOD, GSH-PX, CAT (P<0.01). In addition, the content of ROS significantly decreased (P<0.01), and MMP was increased (P<0.01). Western blot showed SGI can significantly enhance the expression of Bcl-2 and weaken the expression of Caspase-3(P<0.05, P<0.01). Conclusion: SGI shows the protective effect on H2O2-induced oxidative damage of H9c2 cells. Its mechanism may be correlated with increase in ROS removal ability and inhibition of apoptosis.

      摘要:Objective: To observe the effect of Banxia Baizhu Tianma Tang (BBT) on Mitogen-activated protein kinase (MAPK) signaling pathway of myocardial hypertrophy of hypertensive rats with the syndrome of excessive accumulation of phlegm-dampness (EAPD), and explore the pathological mechanism of alleviating myocardial hypertrophy. Method: Spontaneous hypertensive rats (SHRs) were fed with high-fat diet for 20 weeks to establish hypertensive EAPD rat models (HEAPD). Other 10 SHR and WKY rats were included into control groups. Both groups were fed for 20 weeks. After 20 weeks, the modeled rats were divided into high dosage BBT decoction group (13.8 g·kg-1·d-1), low dosage BBT decoction group (6.90 g·kg-1·d-1), telmisartan group (6.33 g·kg-1·d-1) and model group (equivalent normal saline). SHR and WKY control groups were intragastrically administrated with equivalent normal saline for 12 weeks. Body weight and blood pressure were measured every week. Left ventricular end diastolic diameter (LVEDd), left ventricular posterior wall diameter (LVPWd), interventricular septal diameter (IVSd), left ventricular ejection fraction (LVEF) were observed by echocardiography every two weeks. Left ventricular mass (LVM) and left ventricular mass index (LVMI) were calculated. AngiotensinⅡ (AngⅡ), tumor necrosis factor alpha (TNF-α), interleukin 6(IL-6), nitric oxide (NO) in myocardial homogenate were determined by ELISA. Morphological changes of left ventricular observed by HE staining. The distributions of angiotensin II type 1 receptor (AT1) were detected by immuonhistochemical staining assay. Levels of mRNA expression of angiotensin converting enzyme(ACE), AT1, proto-oncogene serine/threonine-protein kinase(c-Raf), extracellular regulated protein kinases 1(ERK 1), ERK-2 were determined by real-time PCR. The expression of p-c-Raf and p-ERK1/2 were observed by Western blot. Result: EAPD rat models were established successfully after high fat diet for 20 weeks. After drug administration for 12 weeks, compared with SHR model group, weight of both high and low dosage BBT groups and telmisartan group was significantly lower (P<0.05), blood pressure of high dosage BBT group and telmisartan group was decreased significantly (P<0.05) in the fourth week, while blood pressure of low dosage BBT group was decreased significantly (P<0.05) in the eighth week. IVSd, LVPWd, LVM, LVMI of high dosage BBT group and telmisartan group were decreased significantly, while LVEDd increased (P<0.05);AngⅡ, TNF-α, IL-6 in myocardium of high dosage BBT group and telmisartan group were decreased significantly (P<0.05), while NO increased significantly (P<0.05). NO of low dosage BBT group increased significantly (P<0.05). The myocardial morphology of high dosage BBT group and telmisartan group was improved, and the distribution of AT1 was decreased in different degrees after drug treatment. Levels of mRNA expression of ACE, AT1, c-Raf, ERK1 and ERK2 of high dosage BBT group and telmisartan group were decreased significantly (P<0.05). The protein expression of p-c-Raf and p-ERK1/2 of both BBT high and low dosage groups and telmisartan group were decreased significantly (P<0.05). Conclusion: BBT decoction can alleviate cardiac hypertrophy in HEAPD. The possible mechanism may be correlated with the down-regulation of MAPK pathway and the inhibition of RASs.

      摘要:Objective: To investigate the protective effect of QingyiⅡ (QY) on the bile-duct ligation (BDL)-induced cholestatic liver injury. Method: Kunming mice were randomly divided into the sham operation group, model group, QY-0.5 group (0.5 g·kg-1) and QY-1.0 group (1.0 g·kg-1), and orally administered for 7 days. On the 7th day, mice received BDL to prepare the cholestatic liver injury model. Livers and blood were collected 72 hours later after BDL. Serum enzyme biochemical indexes, histopathological changes and genes expression were determined. Result: Compared with sham operation group, there were significant increases in serum alanine aminotransferase (ALT) and alkaline phosphatase (ALP), increased total bile acid (TBA) and total bilirubin (TBIL) in model group. According to hepatic tissue pathological indexes, neutrophile infiltration and hepatocyte necrosis were apparent, the expression of cholesterol-7α-hydroxylase (Cyp7a1), 12α-hydroxylase (Cyp8b1), farnesoid X receptor (FXR), small heterodimer partner (SHP) and Ntcp mRNA expression were decreased (P<0.05). Compared with model group, QY decreased ALT, ALP activities and TBA, TBIL levels, alleviated the pathological injury, and up-regulated the expression of above genes (P<0.05). Conclusion: QY II could protect against cholestatic liver injury, and maintain the stability of bile acid by regulating gene expressions related to bile acid synthesis and transport in liver.

      摘要:Objective: To investigate the protective effect of Prunellae Spica extract on the myocardium and its effect on the expression of stromal cell derived factor-1(SDF-1) in acute heart failure (AHF) rat models and discuss its possible mechanism. Method: Wistar rats were used to establish AHF models and randomly divided into four groups, such as AHF model group (AHF group) with equal volume of normal saline by intragastric administration;Prunellae Spica low-dose group, 0.5 g·kg-1 Prunellae Spica extract by intragastric administration;Prunellae Spica middle-dose group, 1.0 g·kg-1 Prunellae Spica extract by intragastric administration;and Prunellae Spica high-dose group, 2.0 g·kg-1 Prunellae Spica extract by intragastric administration. Another sham operation group (Sham group) was set to receive equal volume of normal saline by intragastric administration. After 6 weeks, hemorheology examination and ultrasonic cardiogram were taken. The positive rate of CD34+ cells and microvessel density (MVD) were measured by flow cytometry and HE staining respectively. Western blot and qPCR were further used to detect the protein and mRNA expressions of SDF-1. Result: Prunellae Spica extract can effectively improve left ventricular diastolic function, reduce ventricular wall thickness and promote formation of CD34+ cell mobilization and angiogenesis in AHF rat models, with statistically significant difference as compared with AHF group(P<0.05, P<0.01). Prunellae Spica extract can significantly increase the expressions of SDF-1 protein and SDF-1mRNA, with statistically significant difference as compared with AHF group (P<0.05, P<0.01). Conclusion: Prunellae Spica extract has obvious protective effect on heart failure, and the mechanism may be related to promoting angiogenesis, increasing SDF-1 expression levels and mobilizing the release of CD34+ cells to peripheral blood.

      摘要:Objective: To investigate the effect of forsythiaside on scopolamine-induced learning and memory impairment in mice, and explored the possible mechanism. Method: Kunming mice were randomly divided into 4 groups (n=12 per group):normal group, scopolamine model group (3 mg·kg-1), positive control group (donepezil, 3 mg·kg-1), and forsythiaside group (200 mg·kg-1). All groups were administered with drugs by gastric gavage for 2 weeks. On the 14th day, scopolamine model group, donepezil group and forsythiaside group were intraperitoneally injected with scopolamine (3 mg·kg-1). 20 min later, the step-down passive avoidance test was performed to evaluate the effect of forsythiaside on AchE and CAMP-extracellular signal-regulated kinase pathway. For another experiment, Kunming mice were randomly divided into 4 groups (n=8 per group):normal group, scopolamine model group (3 mg·kg-1), positive control group (Vit E, 100 mg·kg-1), forsythiaside group (200 mg·kg-1). After 14 days, all mice were beheaded to detect the effect of forsythiaside on superoxide dismutase(SOD), malondialdehyde(MDA) and monoamine oxidase(MAO). Result: Significant effects were observed in the step-down passive avoidance test in the acquisition and consolidation periods. Forsythiaside can increase the reduced safe platform time ratio in the scopolamine group (P<0.05). Forsythiaside can decrease the AchE activity in the cortex and hippocampus, increase the expression of p-ERK in hippocampus (P<0.05), with statistically significant differences compared with model group. Compared with the model group, scopolamine can reduce SOD, MDA and MAO content in cortex and hippocampus (P<0.05). Forsythiaside can significantly increase SOD, MDA and MAO content in cortex and hippocampus, with significant differences compared with scopolamine group (P<0.05). Conclusion: Forsythiaside ameliorated scopolamine-induced learning and memory impairment by modulating AchE activity, cAMP expression and p-ERK and resisting oxidation.

      摘要:Objective: To investigate the clinical efficacy of Zishen Roujing decoction in improving non-motor symptoms liver-kidney Yin deficiency syndrome in patients with Parkinson's disease, and explore its effect on adiponectin (APN), uric acid(UA), tumor necrosis factor (TNF)-α, and interleukin(IL)-6 levels. Method: One hundred and eight eligible cases were selected and randomly divided into control group and treatment group according to random number table, 54 cases in each group. Patients in control group were treated with madopar tablets, 1 tablet/time and tid. Based on the treatment in control group. Patients in treatment group also received Zishen Roujing decoction, 1 dose/d and bid. The treatment course was 12 weeks for all patients. Scores of traditional Chinese medicine (TCM) symptoms, scores of non-motor symptoms questionnaire (NMSQuest), scores of parkinson disease sleep scale (PDSS), and scores of hamilton depression scale (HAMD) were compared between two groups. Clinical efficacy was analyzed between two groups after 12 weeks' treatment. The levels of APN, UA, TNF-α, and IL-6 in serum were detected in both groups. Result: After treatment, TCM symptom scores in treatment group were significantly lower than those of control group (P<0.01). Total effective rate was 87.04% in treatment group, significantly higher than 68.52% in control group (P<0.05). Scores of NMSQuest and HAMD in treatment group were significantly lower than those of control group, and PDSS scores were significantly higher than those of control group after treatment (P<0.01). After 6 and 12 weeks' treatment, Levels of APN and UA in serum of treatment group were higher, while TNF-α and IL-6 levels were significantly lower than those of control group (P<0.01). Conclusion: Based on western medicine, Zishen Roujing decoction in improving non-motor symptoms liver-kidney Yin deficiency syndrome in patients with Parkinson's disease could remarkably improve TCM symptoms and non-motor symptoms, and enhance clinical efficacy. Its mechanism may be associated with up-regulating APN and UA levels and down-regulating TNF-α, and IL-6 levels in serum.

      摘要:Objective: To investigate the application effect of Tongqiao Huoxue Tang combined with aspirin for the patients with ischemic stroke. Method: The 80 cases of patients with ischemic stroke in our hospital from September 2013 to September 2015 were randomly divided into control group (40 cases) and observation group (40 cases) according to random number table. Patients in control group received oral application of aspirin tablets and other conventional western treatment. Patients in treatment group received Tongqiao Huoxue Tang combined with the aspirin treatment in the control group. The clinical efficacy, adverse reaction and hemorheology, platelet aggregation rate, National Institute of Health stroke scale(NIHSS) scores, Barthel indexes before and after treatment of two groups were compared. Result: The 4 weeks after the treatment, the total effective rate of treatment group was significantly higher than that of the control group(P<0.05). Platelet aggregation rate and NIHSS scores of the treatment group were all significantly lower than those of the control group, but the Barthel index was significantly higher than that of the control group after treatment(P<0.05). Conclusion: Combination of Tongqiao Huoxue Tang and aspirin for the patients with ischemic stroke is helpful to improve the clinical efficacy and daily life ability of the patients, and is helpful to improve the blood theology and nervous functional defects, worthy of wide application.

      摘要:Objective: To observe the clinical effect of Qingfei Chengqi Decoction in the adjuvant therapy of severe pneumonia with syndrome of phlegm-heat obstructing lung and investigate its influences on levels of serum receptor for advanced glycation end products (RAGE), tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6). Method: Ninety-six patients with severe pneumonia in ICU from our hospital were randomly divided into treatment group and control group (48 cases in each group) according to randomized digital table. The control group received routine treatments, with intravenous drip of linezolid injection (iv, gtt/12 h, 600 mg/time). On the basis of treatment in control group, the treatment group also received Qingfei Chengqi decoction (1 dose/d, bid). The treatment course was 2 weeks for both groups. Traditional Chinese medicine (TCM) symptom scores, the changes of arterial blood gas and sores of acute physiology and chronic health evaluation scoring system (APACHE)Ⅱ were compared pre-post treatment in 2 groups. The clinical effects were evaluated after treatment in both groups. The levels of RAGE, TNF-α and IL-6 in serum were detected pre-post treatment in 2 groups. Result: Scores of fever, cough, accumulating-phlegm and breath shortness in TCM symptom scores of treatment group were significantly lower than those of control group after treatment (P<0.01). The total effective rate was 95.83% in treatment group, superior to 79.17% in control group (P<0.05). Arterial partial pressure of carbon dioxide (PaCO2) and APACHEⅡ scores in treatment group were significantly lower than those of control group, while oxyhemoglobin saturation (SaO2) and partial pressure of blood oxygen (PaO2) were significantly higher than those of control group after treatment (P<0.01). The levels of RAGE, TNF-α and IL-6 in serum of treatment group were significantly lower than those of control group after treatment, with statistically significant differences(P<0.01). Conclusion: Qingfei Chengqi decoction combined with western medicine in the adjuvant therapy of severe pneumonia with the syndrome of phlegm-heat obstructing lung could significantly improve TCM symptom scores, APACHEⅡscores, arterial blood gas and clinical effect, and reducing the levels of RAGE, TNF-α, and IL-6 in serum may be one of its action mechanisms.

      摘要:Objective: To observe the efficacy and anticoagulant effect of Yiqi Tongluo decoction in patients with Qi-deficency and blood-stasis syndrome after cardiac valve replacement. Method: Eighty eight patients with cardiac valve replacement were selected and randomly divided into control group (44 cases) and observation group (44 cases) according to number table method. Patients of control group were treated with warfarin sodium tablets from day one to day three with dose of 3-4 mg·d-1, and after day three, with a maintenance dose of 2.5 mg·d-1. According to the international normalized ratio (INR), dose of warfarin sodium tablets was added or subtracted each time to make the INR value reach 2-3.Patients of observation group were treated with Yiqi Tongluo decoction boiled conventionally in morning and evening, bid. Patients of both groups were observed for three months continuously. Score of traditional Chinese medicine (TCM) symptoms and thrombin time (TT), activated partial thrombin time (APTT), prothrombin time (PT), and plasma fibrinogen(FIB) were compared between two groups. Levels of hemorheology indexes were detected in two groups 1 d and 3 months after the operation. Complications and deaths were recorded in two groups. Result: Three months after operation, score of TCM symptoms in observation group were significantly lower than those of control group (P<0.01). Three months after operation, There was no statistically significant difference in TT, APTT, PT and FIB between two groups. During the 1 year of follow-up, postoperative complications occurred in 3 cases (6.82%) of observation group, and 12 cases (27.27%) of control group, with statistically significant differences (P<0.05). Three months after operation, there were no statistically significant differences in various levels of hemorheology indexes between two groups. During 1 year of follow-up, death occurred in 1 case (2.27%) of observation group, and 2 cases (4.55%) in control group, with no statistically significant differences. Conclusion: Yiqi Tongluo decoction showed definite anticoagulant effect and was safe in patients with Qi-deficency and blood-stasis syndrome after cardiac valve replacement. It is worthy of clinical application.

      摘要:Objective: To observe clinical effect of the best decoction time of Yinqiao San and influence to peripheral blood T lymphocytes subpopulations in treating acute upper respiratory infection. Method: One hundred and twenty-two patients were randomly divided into control group (62 cases) and experimental group (65 cases) by random number table, patients in control group received Yinqiao San for 12 minutes' boiling, patients in experimental group received Yinqiao San for 6 minutes' boiling, 200 mL/dose, and according to state of an illness, 100 mL/time, 2-4 times/day. Courses of treatment were 3 days. And onset time of hypothermia and tiem of fever were observed. Before and after treatment, scores of traditional Chinese medicine(TCM) syndrome was graded. Levels of T lymphocytes subpopulations were also detected for safety evaluation. Result: The revealing rate of observation group was 69.23%, which was higher than that was 51.61% in control group (P<0.05). At the third and sixth hours after taking drugs, the temperature of observation group was lower than that in control group (P<0.05). The onset time and antipyretic time in observation group were both shorter than these data in control group (P<0.05). After treatment, Scores of sore throat, head pain, stuffy nose, bad wind in observation group were inferior to the scores in control group (P<0.01). The levels of CD3+ CD4+, and CD4+/CD8+ in observation group were superior to the levels in control group (P<0.05). While the CD3+ CD8+ standards in observation group were lower than these in control group (P<0.05). Conclusion: At the sixth minute after boiling, the Yinqiao San can treat acute upper respiratory infection (the wind heat invading syndrome), improve symptoms and clinical effects. Its immunoregulation was superior to the Yinqiao San which was at the twelfth minute after boiling. Thus, the best time for Yinqiao San boiling was about six minutes after elixation.

      摘要:Objective: To observe the short-term effect amd safety of Yangzheng Xiaoji capsule in the treatment of cancer cachexia, and provide theory basis for its clinical treatment. Method: Totally 70 patients with cancer cachexia were randomly divided into observation group and control group, 35 cases in each group. Patients of both groups received diet guidance and general nutrition support. Patients in observation group also orally took Yangzheng Xiaoji capsules, 4 tablets/time, 3 times/d;while the patients in control group orally took megestrol acetate tablets, 160 mg/time, 1 time/d. Observation period lasted for 1 month. Traditional Chinese medicine (TCM) symptom scores, food-intake and middle upper arm circumference of both groups were observed before and after treatment. Quality of life (QOL) was graded, and the level of serum albumin, hemoglobin, T lymphocyte subsets and adverse reactions were also detected. Result: After treatment, the TCM symptom efficacy and food-intake in the observation group were superior to those in the control group (P<0.05). After treatment, middle upper arm circumference of patients in the observation group and control group were (24.58±2.21) score and (24.01±2.29) score respectively, with an increase as compared with those before treatment, but there was no statistically significant difference between two groups. The QOL score was (36.52±3.39)score and (34.80±3.46)score for observation group and control group respectively, with an increase as compared with those before treatment, but the improvement of QOL score in observation group was more significant than that of control group (P<0.05). The levels of serum albumin, hemoglobin and CD3+, CD4+, CD8+, CD4+/CD8+ cells were significantly improved after treatment in both groups (P<0.01, P<0.05). However, the improvement in the observation group was more significant than that in control group after treatment (P<0.05).There was no adverse reaction in the observation group. Conclusion: Yangzheng Xiaoji capsule could effectively improve TCM symptoms for patients with cancer cachexia, increase food-intake, improve quality of life and significantly improve serum albumin, hemoglobin and T lymphocyte subsets level. It is much more better than megestrol acetate in clinical efficacy and safety. Yangzheng Xiaoji capsule deserves much more clinical research and application.

      摘要:Objective: To systemically evaluate the effectiveness and safety of Cinobufacini injection for lung cancer. Method: We searched The Cochrane Central Register of Controlled Trials (CCTR), PubMed, CNKI, CBM, VIP databases up to Dce. 2014. The methodological quality assessment and data extraction of the included studies were conducted by two reviewers independently according to the inclusion and exclusion criteria. Meta-analysis was performed for homogeneous studies using RevMan 4.2.10 software after subgroup analysis of the measurement indicators and intervention measures. Result: A total of 17 studies involving 1279 patients were included. None of the studies mentioned allocation concealment method and no blinding method was performed. Conclusion: In the treatment of lung cancer, the combination of cinobufacini injection and western medicine is superior to western medicine alone for the long-term survival rate, quality of life and short-term efficacy.

      摘要:Objective: To systematically review the effectiveness of Yiqi Fumai injection combined with western medicine for heart failure. Method: Databases such as PubMed, CENTRAL, EMbase, VIP, CNKI, CBM and Wanfang were electronically searched to collect the randomized controlled trials (RCTs). After document screening and quality evaluation, Meta-analysis was conducted for valid data. Result: Thirteen RCTs involving 1001 patients were included. The results of meta-analysis showed that the combination of Yiqi Fumai injection and western medicine was superior to the western medicine alone in improving clinical comprehensive efficacy[OR=3.09, 95%CI (2.05, 4.67), P<0.00001], left ventricular ejection fraction[MD=4.84, 95%CI (3.56, 6.13), P<0.00001], cardiac output[MD=0.30, 95%CI (0.19, 0.42), P<0.00001], 6 minutes walking distance[MD=39.03, 95%CI (25.06, 53.00), P<0.00001], reducing NT-proBNP[MD=-67.72, 95%CI (-110.30, -25.13), P=0.002], and shortening left ventricular end systolic dimension (LVESD)[MD=-2.94, 95%CI (-4.86, -1.02), P=0.0003], with statistically significant differences between two groups. But there was no significant difference between two groups in shortening left ventricular end diastolic dimension (LVEDD)[MD=-2.06, 95%CI (-4.70, 0.58), P=0.13]and improving E/A[MD=0.06, 95%CI (-0.03, 0.15), P=0.21]. Conclusion: Combination of Yiqi Fumai injection and western medicine has higher clinical efficacy than western medicine alone for the treatment of heart failure.

      摘要:Objective: To analyze the medication rules given by professor LI Ji-ren for treatment of active rheumatoid arthritis (RA), and provide reference for traditional Chinese medicine (TCM) treatment in active RA. Method: Prescriptions used for active RA in January 2013-December 2014 given by professor LI Ji-ren were collected. Then the data was entered into TCM inheritance support system to build a database, and drug frequency, drug combination, association rules and core combinations were obtained by using data mining methods such as association rule, to analyze professor LI Ji-ren's medication rules for the treatment of RA. Result: By analyzing 84 screened prescriptions based on TCM inheritance support system, high frequency herbs for the activity RA included the clearing heat and promoting diuresis herbs such as Scolopendra, Sophorae Flavescentis Radix, Dioscoreae Hypoglaucae Rhizoma;tonifying Qi and invigorating blood circulation herbs such as Astragall Radix, Angellcae Sinensis Radix, Spatholobi Caulis, Sargentodoxae Caulis, Olibanum, Myrrha;expelling wind, dredging collateral and pain reliving herbs such as Scorpio, Scolopendra;warming and activating meridian and pain relieving herbs such as Aconitti Radix Cocta, Aocniti Kusnezoffii Radix Cocta, and Cinnamomi Ramulus. Meanwhile, 35 core combinations and 9 new prescriptions were dig out. Conclusion: Professor LI Ji-ren is experienced in treatment of active RA. He believes that the pathogenic wind-cold damp-heat is of great significance in the activity of rheumatoid arthritis etiology. In the syndrome differentiation, the ‘cold, hot’ should be emphasized. Mildly regulating cold and heat, relieving both primary and secondary symptoms and protecting spleen and stomach shall act as the treatment principles, Qingluoyin as the main composition, with compatibility of cold and heat drugs based on the bias and complex transformation of wind-cold damp-heat. The herbs can be added or subtracted according to the symptoms, so that the cold herbs would not hurt Yangqi and hot herbs would not hurt Yin essence.

      摘要:Objective: To systemically evaluate the safety and efficacy of Shenqi Fuzheng injection combined with GP treatment for non-small cell lung cancer. Method: Wanfang, CNKI, CBM, VIP and US national library of medicine were searched for the clinical randomized controlled trials of Shenqi Fuzheng injection combined with GP treatment for non-small cell lung cancer from the database establishment to August 2015.The articles were selected according to inclusion and exclusion criteria, and the data extraction and quality assessment were done for the included studies, and then Revman 5.3 software was used for Meta-analysis. Result: The 12 randomized controlled trials were included in the systematic review, covering a total of 928 patients. All of them were low-quality studies with Jadad score ≤3. Meta analysis showed that:compared with GP treatment alone, Shenqi Fuzheng injection combined with GP treatment for non-small cell lung cancer could significantly increase the efficiency[OR=1.72, 95% CI (1.32, 2.25), P<0.0001], improve the quality of life for patients[OR=3.35, 95% CI (2.50, 4.49), P<0.00001], reduce leucopenia[OR=0.30, 95% CI (0.21, 0.42), P<0.00001]and hemoglobin decline[OR=0.37, 95% CI (0.22, 0.62), P=0.0002], and reduce gastrointestinal reactions[OR=0.47, 95% CI (0.33, 0.68), P<0.0001], with statistically significant difference. Conclusion: Shenqi Fuzheng injection combined with GP treatment for non-small cell lung cancer can significantly improve the efficiency and quality of life for patients and reduce side effects of chemotherapy. However, the existing small-sample randomized controlled trials have low methodological and reporting quality, so further rigorous clinical trials with high quality and large sample are still required for validation.

      摘要:Myocardial ischemia-reperfusion injury is a kind of pathological process, which would further aggravate ischemic myocardial injury, because of oxidative stress reaction hyperthyroidism and occur irreversible damage, seriously threating to people's life.Rhodiolae Crenulatae Radix et Rhizoma has potential to protect myocardial ischemia reperfusion injury, there are many related research reports about protecting myocardial ischemia reperfusion injury of this herb.We have a review about researches of Rhodiolae Crenulatae Radix et Rhizoma protecting myocardial ischemia reperfusion injury function from its mechanism, aiming at providing a reference about Rhodiolae Crenulatae Radix et Rhizoma in myocardial ischemia-reperfusion injury protection mechanism of related research for people in the future.
