      摘要:Objective: To investigate feasibility of application of zebrafish neuronal injury model on quality control for Yangxue Qingnao granules and to provide a new quality control method for this preparation. Method: Biological activity effect and stability of Yangxue Qingnao granules were quantitatively assessed in a zebrafish neuronal injury model which was established by mycophenolate mofetil (MMF). Result: Central nerve injury zebrafish model as a biological quality control standard for Yangxue Qingnao granules nerve cell protection exhibited excellent precision (intra-day RSD of 2.2%-3.7%, inter-day RSD of 1.2%), repeatability (RSD of 8.4%) and stability (RSD of 4.9%).In the range of 0.1-1.0 g·L-1, there was a significant linear correlation;this method met requirements of biological activity determination of guiding principles. Conclusion: It can be turned out that methodology which established in dissertation is good at determining neuroprotective efficacy of Yangxue Qingnao granules, it is reliable, simple and repeatable, this method also provides a reference for development and improvement of bioassay of Chinese medicines.

      摘要:Objective:To study on antioxidant activity of effective components in Acanthopanacis Senticosi Radix et Rhizoma Seu Caulis roots. Method: Contents of total saponins and total flavonoids were determined by UV.Antioxidant activity of effective components in Acanthopanacis Senticosi Radix et Rhizoma Seu Caulis roots were conducted with superoxide anion free radical scavenging method, hydroxyl free radical scavenging method, DPPH scavenging method and FRAP method. Result: Different effective components in a certain concentration range had some antioxidant activities.When the concentration increased, antioxidant ability enhanced.Clearance of 30% ethanol eluted site on O2-·was(91.61±2.59)% at the concentration of 20 g·L-1.Clearance of 30% ethanol eluted site on·OH was(90.29±2.06)% at the concentration of 1.0 g·L-1.Clearance of 30% ethanol eluted site on DPPH was(84.45±0.14)% at the concentration of 0.7 g·L-1.FRAP value of 30% ethanol eluted site was 3474±36.84 at the concentration of 1.0 g·L-1. Conclusion: All the effective components of Acanthopanacis Senticosi Radix et Rhizoma Seu Caulis roots have a certain antioxidant activity.Antioxidant activity of 30% ethanol eluted site is the highest.

      摘要:Objective: To investigate in vitro transdermal permeability of salidroside phospholipid complex. Method: Taking isolated mice skin as permeability barrier, in vitro permeation test was performed by modified Franz diffusion pool.HPLC was adopted to determine the content of salidroside with mobile phase of acetonitrile-water-phosphoric acid (15:85:0.05) and detection wavelength at 275 nm. Result: Cumulative permeation amounts of salidroside phospholipid complex and salidroside alcohol solution in 8 h were (14.8±0.42) μg·cm-2 and (3.01±0.41) μg·cm-2, retention of salidroside in isolated mice skin were 0.95 μg·cm-2 and 0.02 μg·cm-2, enhancement permeation coefficient of salidroside phospholipid complex was 52 times the amount of salidroside alcohol solution. Conclusion: Ability of permeation is strengthened after salidroside being formed into phospholipid complex.

      摘要:Objective: To analyze sources and residue accumulation dynamic change of SO2 in Ophiopogonis Radix for providing a theoretical basis to solute problem that SO2 residue in Ophiopogonis Radix exceeded the standard. Method: Twenty-eight medicinal materials of Ophiopogonis Radix were collected, which were processed with different methods in main producing area of Sichuan.Residue of SO2 in samples were measured.Based on measurement results and after processed by coal fire baking and smouldering sulphur, residue accumulation dynamic change of SO2 in Ophiopogonis Radix were investigated. Result: Residue of SO2 in 28 samples exceeded the standard with rate of 35.71%.Sources of SO2 residue were coal fire baking and smouldering sulphur.SO2 residue was in direct proportional to baking time.Marginal time of SO2 residue out of the limit was 8-10 h.SO2 residue was also in direct proportional to smoked sulfur time and material moisture content.SO2 residue reached up to 1 017.69 mg·kg-1 when fresh Ophiopogonis Radix was processed with smouldering sulphur for only 1 h. Conclusion: Consider of quality and medicine safety of Ophiopogonis Radix, it should be forbidden with smouldering sulphur and control baking time strictly in processing of Ophiopogonis Radix.

      摘要:Objective: To evaluate the quality of Gansu merchandise Wen Codonopsis Radix with the fuzzy matter-element model. Method: Content of five main components in the samples of Wen Codonopsis Radix with different commercial grades from different origins were determined. Then the quality of Wen Codonopsis Radix was evaluated by the fuzzy matter-element model based on variation coefficient weight. Result: According to the value of Euclid approach degree, there were 18 batches of samples higher than 0.450, with higher evaluation degree, mainly coming from Wen county and Wudu district in Gansu. The samples of commercial grade two were generally at the very top in ranking. The comprehensive ranking was not depending on the commercial grade, but also greatly affected by place of origin. Conclusion: The method and model can be used to objectively evaluate the quality of Wen Codonopsis Radix, and can be used for the quality evaluation of other medicinal materials.

      摘要:Objective: Gentiana lhassica was compared with G. macrophylla in their pharmacognosy. Method: Quality of G. lhassica from some aspects of micromorphology was evaluated, on morphological and physicochemical identification by pharmacognostic determination.HPLC was employed to determine contents of swertiamarin and gentiopicroside with mobile phase of methanol-water for gradient elution and detection wavelength at 254 nm. Result: Cork cells of G. lhassica had only one layer in the inner peripheral skin and peripheral skin.The content of gentiopicroside was 6.24%, higher than standard of the 2015 edition of Chinese Pharmacopoeia.The content of swertiamarin was 0.26%. Conclusion: There are some differences between G. lhassica and G. macrophylla in pharmacognostic character.This study provides important reference for exploitation and utilization of G. lhassica.

      摘要:Objective: To analyze and identify the chemical compounds in Mori Folium aqueous extract and the components in serum of rats after oral administration. Method: HPLC was employed to analyze the compounds in Mori Folium aqueous extract as well as in rat serum after intragastric administration. LTQ-Orbitrap XL high-resolution mass spectrometer was used for online ions detection and secondary ions scanning to identify the compounds in Mori Folium aqueous extract and its metabolites in blood samples. Result: The 11 chemical compounds were identified in the Mori Folium aqueous extract, including isochlorogenic acid(1), 5, 7-dlhydroxycommarin-7-O-β-D-glucopyranoside(2), scopolin(3), chlorogenic acid(4), kaempferol-3, 7-dl-O-β-D-glucopyranoside(5), 4-caffeoylquinic acid methyl ester(6), rutin(7), hyperoside(8), isoquercitrin(9), astragalin(10), and isorhamnetin-3-O-glucoside(11). The 30 compounds were preliminarily identified in the blood, and major of, them were aglycone metabolites. The above 11 original compounds were not found in serum. Conclusion: The present method can be used for the identification of the compounds in Mori Folium aqueous extract and blood of rats after oral administration, providing reference for further efficacy study and secondary development of Mori Folium.

      摘要:Objective: To study the chemical components of Artemisiae Argyi Folium and their anti-hepatitis B virus(HBV) activities. Method: The chemical components were isolated and purified by silica gel column chromatography, and their structures were identified according to their physicochemical properties, nuclear magnetic resonance spectroscopy (NMR), mass-spectrography (MS) and literature analysis. Furthermore, the toxicity of these chemical components to HepG 2.2.15 cells was measured by MTT method and their effects on HBeAg and HBsAg secretion in HepG 2.2.15 cell supernatant were measured by ELISA method. Result: Five compounds were isolated from Artemisiae Argyi Folium and their structures were identified as stigmasterol(1), 5, 4'-dyhydroxyl-6, 7, 3'-trimethoxyflavone (3'-methoxythistle flavin) (2), dammaradienylacetate(3), 5, 7, 4'-trihydroxy-3', 8-dimethoxyflavone(4), and hexadecanoic dihydro ypropyleste(5). Conclusion: Compounds 2-4 were isolated from Artemisiae Argyi Folium for the first time and 3'-methoxythistle flavin showed inhibitory effect on HBsAg and HBeAg lines with IC50values of 8.09 mg·L-1 and less than 2.5 mg·L-1respectively.

      摘要:Objective: To extract the volatile oil and small polar components of the Hui formula Muxiang oil by steam distillation (SD) and supercritical CO2fluid extraction (SFE), respectively;analyze and identify the components and their contents with Gas Chromatography-Mass Spectromet (GC-MS), and compare the differences in types of components between different extraction methods. Method: The mass spectrometric conditions were set as follows:Shimadzu QP2010 plus GC-MS, Rxi-5Sil MS quartz capillary column (0.25 mm×30 m, 0.25 μm), the starting temperature of 50℃, maintained for 1 min, heated to 150℃ with 8℃·min-1, maintained for 3 min, then heated to 210℃ with 3℃·min-1, maintained for 3 min, and then heated to 280℃ with 15℃·min-1, maintained for 15 min until completion of analysis;helium as the carrier gas, column flow rate at 1.33 mL·min-1, split ratio of 25:1, inlet temperature of 250℃, EI ionization source of 70 eV, ion source temperature of 230℃, and scan range of m/z 35-500. Result: Yield of volatile oil was 0.8% and 2.91% respectively in SD and SFE methods;25 compounds were identified, accounting for 96.28% of the total peak area of volatile oil in SD method;36 compounds were identified, accounting for 97.98% of the total peak area of volatile oil in SFE method. Conclusion: There was significant difference in types of components contained in the volatile oil between SD and SFE methods, and GC-MS could be used to identify the small polar components of the medicine, providing experimental basis for further development and utilization of Hui medicine.

      摘要:Objective: To establish a HPLC method for simultaneous determinationof the content of epicatechin, vitexin, isovitexin, kaemferol-3-O-rutinoside, isorhamnetin-3-O-β-D-glucoside, and narcissi in extract of Microctis Folium. Method: The HPLC analysis was carried out on Agilent HC-C18column (4.6 mm×250 mm, 5 μm) with amethanol-0.05% phosphoric acid solution as the mobile phase for gradient elution at a flow rate of 0.8 mL·min-1.The detection wavelength was 288 nm, and the column temperature was 35℃. Result: Good linear relationship were found in the the range of 131.200-1 312.000 ng for epicatechin, 149.800-1 348.200 ng for vitexin, 131.400-1 051.200 ng for isovitexin, 99.175-595.050 ng for kaemferol-3-O-rutinoside, 109.955-659.730 ng for isorhamnetin-3-O-β-D-glucoside, and 267.000-1 348.200 ng for narcissi. Their average recoveries were 98.13%, 97.41%, 96.93%, 98.22%, 97.37%, 97.82%, respectively. Conclusion: The method is accurate, reliable, and can be used for the quality control and evaluationof total flavonoid extracts of Microctis Folium.

      摘要:Objective: To establish the self-organization competitive neural network method for distinguishing the fingerprints of Canarii Fructus from different producing areas, and lay the foundation for quality evaluation of Canarii Fructus. Method: The fingerprints of Canarii Fructus from different producing areas of Canarii Fructus were established by HPLC on Phenomenex Luna C18 column (2) 100A (4.6 mm×250 mm, 5 μm), with acetonitrile-1% methanoic acid as the mobile phase;the detection wavelength was 270 nm;the flow rate was 0.8 mL·min-1 and the column temperature was 20℃. Then the self-organization competitive neural network model was used to distinguish the Canarii Fructus from different producing areas, with 3 competition layer neurons, a learning rate of 0.01, and 690 convergence times. Result: The self-organization competitive neural network for distinguishing Canarii Fructus from different producing areas was established, but the rate of error for HPLC fingerprint extract was 39.13% in average. Conclusion: The self-organizing competitive neural network can not be used to effectively distinguish the Canarii Fructus from different producing areas, as the differences in types and contents of chemical compositions of Canarii Fructus from different producing areas are not evident.

      摘要:Objective: To establish an UPLC method for simultaneously determining content of 6 major compounds (chlorogenic acid, myricitrin, hyperoside, isoquercitrin, kaempferitrin, quercitrin) in Vicia amoena var. angusta. Method: An Eclipse Plus C18 RRHD column (2.1 mm×50 mm, 1.8 μm) was adopted, with acetonitrile (B)-0.1% formic acid aqueous solution (A) as mobile phase for gradient elution (0-7 min, 5%-10%B;7-11 min, 10%-12%B;11-20 min, 12%B;20-25 min, 12%-19%B;25-30 min, 19%-24%B). The flow rate was 0.2 mL·min-1. The detection wavelength was set at 340 nm and the column temperature was at 30℃. Result: Six components of V. amoena var. angusta showed good linear relationship in the linear range(r> 0.999 9) and the average recoveries were between 99.15% to 101.98% with the RSD of 0.7%-1.7%. Conclusion: The developed method is convenient, rapid and accurate with high repeatability and stability, suitable for simultaneously determining contents of 6 compounds in V. amoena var. angusta and can provide basis for the quality control of V. amoena var. angusta.

      摘要:Objective: To establish the HPLC fingerprints for Bawei Chenxiang pills, and provide scientific basis for further improving the quality standard. Method: Waters Xbridge Shield RP18 chromatographic column (4.6 mm×250 mm, 5 μm) was used, with methanol-0.05% phosphoric acid solution as the mobile phase for gradient elution, at a flow-rate of 1.0 mL·min-1;the detection wavelength was set at 220 nm and column temperature was 25℃, with injection volume of 10 μL. Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine (2004A) was used to evaluate the HPLC fingerprints for 10 batches of Bawei Chenxiang pills. Result: Thirty-one common peaks were identified, and 4 common peaks were determined as gallic acid, costunolide, dehydrocostus lactone and dehydrodiisoeugenol. The similarity of 10 batches of Bawei Chenxiang pills was al ≥0.90. Conclusion: This method was proved simple, stable and reproducible, and can provide reliable basis for establishment of quality standards of Bawei Chenxiang pills.

      摘要:Objective: To establish an UPLC method for the simultaneous determination of geniposide, albiflorin, paeoniflorin, liquiritin, benzoylpaeoniflorin, and glycyrrhizic acid in extract of modified Xiaoyao extract. Method: UPLC was performed on UPLCTM HSS T3 column (2.1 mm×100 mm, 1.8 μm) with acetonitrile-0.05% phosphoric acid solution as the mobile phase for gradient elution. The detection wavelength was set at 230 nm. The flow rate was 0.4 mL·min-1 and column temperature was 30℃. Result: The six components were well separated and showed good linearity in their respective concentration ranges with R2≥0.999 4. The average recovery was 98.29%-101.67% with RSD less than 3.8%. Conclusion: The established method is simple and stable with satisfactory separation degree. It can be used for the quality control of modified Xiaoyao extract.

      摘要:Objective: To establish HPLC fingerprint of Danggui Liuhuang Tang, analyze chromatographic data and analyze main components in Danggui Liuhuang Tang by high performance liquid chromatography and mass spectrometry (LC-MS) method. Method: HPLC was used on Agela Venusil XBP-C18(4.6 mm×250 mm, 5.0 μm) column, with acetonitrile (A)-0.3% formic acid (B) as mobile phase for gradient elution (0-10 min, 5%-15%A;10-15 min, 15%-20%A;15-35 min, 20%-25%A;35-50 min, 25%-31%A;50-62 min, 31%-55%A;62-70 min, 55%-60%A;70-75 min, 60%-70%A);with column temperature of 30℃, and volume flow rate of 0.8 mL·min-1;electrospray ionization was used for mass spectrometry, and full scan testing was done under cationic-anionic mode. Compounds were estimated through [M-H]-, [M+H]+ plasma information. Result: HPLC analysis and fingerprint were established for Danggui Liuhuang Tang;LC-MS spectra were established for Danggui Liuhuang Tang;19 characteristic chromatographic peaks of the Tang were identified. Conclusion: HPLC fingerprint analysis method of the Danggui Liuhuang Tang is stable and reliable, with good repeatability;the main components of Danggui Liuhuang Tang can be effectively identified through the establishment of LC-MS spectra.

      摘要:Objective: To establish a method for determining contents of three major compounds in human plasma, namely paeoniflorin, paeonol and α-cyperone from Baixiangdan capsules, and to determine plasma protein binding rate. Method: Equilibrium dialysis method was carried to determine plasma protein binding rate.Samples were extracted by liquid-liquid extraction with ethyl acetate, LC-MS/MS was employed with a Phenomenex Luna C18 column (2.0 mm×100 mm, 3 μm), mobile phase of methanol-0.1% formic acid solution (80:20), flow rate was 0.2 mL·min-1.Quantitation was performed on a triple quadrupole mass spectrometer employing electrospray ionization (ESI) technique, operating in multiple reaction monitoring (MRM) and positive ion mode. Result: Within low, middle and high concentration (56.2, 112.5, 168.8 mg·L-1) of Baixiangdan capsules, plasma protein binding rates of paeoniflorin were (24.82±3.91)%, (20.88±2.69)% and (22.40±3.33)%;binding rates of paeonol were (88.62±1.68)%, (86.16±2.36)% and (88.73±0.80)%;binding rates of α-cyperone were (80.69±1.12)%, (78.10±2.28)% and (72.57±0.30)%. Conclusion: Human protein binding rates of three major compounds in Baixiangdan capsules were decreased in the order of paeonol > α-cyperone > paeoniflorin.

      摘要:Objective: To establish a determination for docetaxel that derives from mice plasma and tissue fluid samples, and evaluate effectiveness for this method. Method: Samples were purified by methyl tert-butyl ether with paclitaxel as internal standard.Chromatographic separation was performed on a Hypersil BDS C18 column using a mobile phase composed of methanol-0.1% formic acid(80:20), column temperature was 40℃, flow rate was 0.3 mL·min-1.Detection was performed on atriple-quadrupole tandem mass spectrometer with multiple reaction monitoring (MRM) mode via electrospray ionization (ESI) source, ESI voltage was 3.5 kV, capillary temperature was 320℃, sheath gas pressure was 35 Pa, auxiliary gas pressure was 10 Pa, collision energy was 30 V, CID pressure was 12 V, detection ion for docetaxel was m/z 876.3-549.1, detection ion for paclitaxel was m/z 830.3-307.9. Result: Docetaxel in mice plasma, heart, liver, spleen, lung and kidney showed good linear relationship within 0.013-13, 0.012-12.937, 0.013-13.068, 0.012-13.001, 0.013-13.133, 0.011-12.965 mg·L-1.RSD of intra-day and inter-day assays were less than 15%.Recovery rates of docetaxel with low, middle and high concentration in plasma were 98.24%, 97.51% and 93.70%, its recovery rates in heart, liver, spleen, lung and kidney were 87.87%-98.90%. Conclusion: Compared with HPLC, LC-MS/MS is more suitable for quantitative analysis of docetaxel in mice plasma and tissue fluid samples.

      摘要:Objective: To study on pharmacokinetic characteristics of ellagic acid microspheres in rabbits by oral administration. Method: Orifice method was used to prepare ellagic acid microspheres, HPLC was employed to determine the concentration of ellagic acid with acetonitrile-0.1% phosphoric acid solution (23:77) at a flow rate of 0.8 mL·min-1, detection wavelength was set at 254 nm, column temperature was controlled at 30℃.Pharmacokinetic parameters differences of ellagic acid microspheres and ellagic acid tablets were analyzed by DAS 2.0. Result: The linear range of ellagic acid was within 0.01-1.50 mg·L-1 with r of 0.999 4, quantitation limit was 0.01 mg·L-1.Plasma concentration-time curve of ellagic acid showed double peak, there may be enterohepatic circulation in rabbits.Pharmacokinetic of ellagic acid microspheres could be evaluated by two-compartment model.Main pharmacokinetic parameters were shown as t1/2β of (7.683±2.762) h, t1/2α of (7.161±1.275) h, tmax of (3.00±0) h and Cmax of (1.121±0.259) mg·L-1. Conclusion: By comparing with ellagic acid tablets, ellagic acid microspheres exhibits higher bioavailability and longer action duration in rabbits.

      摘要:Objective: To observe the antioxidant effects of modified Bufei Tang (MBT) combined with methylprednisolone on bleomycin-induced idiopathic pulmonary fibrosis (IPF) in rats, and investigate the mechanism. Method: Sixty Sprague-Dawley rats were randomly divided into normal group, model group, methylprednisolone group (10 mg·kg-1·d-1), MBT group (100 mg·kg-1·d-1), combined treatment group 1, and combined treatment group 2, n=10 in each group. The IPF models were reproduced by intratracheal injection of bleomycin (1 mg for each rat). 24 hours after modeling, the rats were treated with daily intragastric administration or intramuscular injection until one day before sacrificing. The rats in model group were treated with the same volume of normal saline. Lung coefficient was calculated, and lung tissue morphology changes were observed in IPF rats. The hydroxyproline (Hyp), malondialdehyde (MDA) and superoxide dismutase (SOD) levels in lung homogenates of rats were measured. Hematoxylin-eosin staining was applied for fixed lung tissues. Result: As compared with the normal group, degree of alveolar inflammatory cells infiltration and pulmonary fibrosis was obvious;lung coefficient was increased significantly;Hyp and MDA levels in liver tissues were increased significantly;and SOD activity in liver tissues was decreased significantly in the rats of model group (P<0.05, P<0.01). Various treatment groups could improve the living state of IPF rats;methylprednisolone group, MBT group, and combined treatment group 1 could significantly improve the degree of alveolar inflammatory cells infiltration and pulmonary interstitial fibrosis of IPF rats, significantly reduce lung coefficient (P<0.01, P<0.05), reduce Hyp and MDA levels in the lung tissues and increase SOD activity in the lung tissues. Conclusion: Combined treatment group 1 has superior effect than single treatment in reducing liver coefficient, Hyp and MDA levels and increasing SOD activity in the lung tissues of IPF rats. The mechanism might be related to decreasing the level of lipid peroxide to achieve antioxidant effects.

      摘要:Objective: To clarify the medicinal properties of chemical components in Daturae Flos, and provide scientific basis for the objectivity of new hypothesis theory "One flavor and one property for Chinese medicine, X flavors and Y properties for one medicine (Y≤X)". Method: Male Kunming mice and Wistar rats were randomly divided into normal group, Daturae Flos decoction group, alkaloid components high dose group, low dose group, macroporous resin 50% ethanol elution components high dose group and low dose group. The mice were treated by intragastric administration and the mice in normal group received same volume of normal saline, 1 time a day for consecutive 10 days. The effects of Daturae Flos and its chemical components on heart rate, breathing rate, blood pressures, the amount of salivary secretory and body temperature relative to autonomic nervous system were observed as part of the medicinal properties evaluation of Daturae Flos for further research. Result: Compared with the normal group, the Daturae Flos and its alkaloid components had certain effect or effect trend in increasing the heart rate of mice, accelerating the breathing rate of rats, increasing the mean arterial blood pressure in rats, inhibiting salivary secretory in mice and increasing body temperature of rats (P<0.05, P<0.01). Macroporous resin 50% ethanol elution components could promote salivary secretory in mice, and have certain effect on reducing heart rate of mice and reducing breathing rate of rats (P<0.05, P<0.01), but with no significant effect on mean arterial blood pressure or body temperature in rats. There was certain difference or statistically significant difference between macroporous resin 50% ethanol elution components and alkaloid components in effects on heart rate of mice, breathing rate of rats, mean arterial blood pressure in rats, salivary secretory in mice and body temperature of rats (P<0.05, P<0.01). Conclusion: Daturae Flos and its alkaloid components have warm medicinal properties, while macroporous resin 50% ethanol elution components have cold medicinal properties based on the results and previous research results.

      摘要:Objective: To investigate the effect of Guizhi Shaoyao Zhimu Tang(GT) on inflammatory signal expression in synovial tissues of joint of rats with monosodium urate crystal-induced gouty arthritis(GA). Method: Totally 180 male SD rats were randomLy divided into 3 experiments[the Immunohistochemistry, enzyme linked immunosorbent assay(ELISA), Western blot experiment] with 60 rats each experiment divided into 6 groups with 10 rats each group according to weight.The high, medium and low dose group of GT (4, 8, 16 g·kg-1)and colchicine group (3×10-4 g·kg-1) were treated with medicine by gastric administration, the normal and model group were given equal volume of distilled water. Medicine or distilled water was given once daily for seven consecutive days throughout the experiment.On the fifth day, GA model was made by injection of monosodium urate in the ankle joint cavity of rat.The synovial tissues of the rats were taken and expression of nod-like receptor protein 3(NLRP3) inflammasomes were detected with immunohistochemistry and integrated absorbance(IA) was measuring with Image-Pro Plus 6.0 analysis system. The expression levels of apoptosis-associated speck-like protein containing a CARD(ASC), Caspase-1 signaling proteins were detected by Western blot. Expression of inflammatory cytokine such as interleukin-1 beta (IL-1β) and interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-α), nuclear factor-κB (NF-κB) were detected with ELISA. Result: Compared with normal group after 72 hours, the expression of NLRP3 inflammasomes, ASC, Caspase-1 and IL-1β, IL-6, TNF-α, NF-κB in synovial tissues of the joint of GA rats significantly increased(P<0.05), whereas Caspase-12 significantly decreased(P<0.05). the expression of NLRP3 inflammasomes, ASC in medium and high dose group of GT and Caspase-1 in all group of GT significantly decreased than the model group(P<0.05), whereas Caspase-12 increased(P<0.05) and there was significant decreasing change in IL-1β, IL-6, TNF-α, NF-κB in all dose group of GT. Conclusion: The acion mechanism of GT on GA in rats is related to decreasing the expression levels of NLRP3, ASC and Caspase-1 and increasing the expression of Caspase-12, and accordingly inhibiting the maturation of IL-1β and the activation of NF-κB, reducing the expression of inflammatory factors in NLRP3 inflammasomes signaling pathway.

      摘要:Objective: To observe the therapeutic effect and mechanism of Mongolian Dongqingye-7 capsule for the pulmonary fibrosis induced by pingyangmycin in rats. Method: Totally 90 healthy SD rats were randomly divided into sham-operation group and model group after 1 week of adaptive breeding. Gap injection of pingyangmycin was done by using tracheal cartilage ring to establish the pulmonary fibrosis models in rats. Postoperative day 7, the rats were divided into Mongolian Dongqingye-7 capsule groups (120, 240, 480 mg·kg-1), dexamethasone group (0.3 mg·kg-1), sham-operation group and model group with same amount of normal saline. 7, 14, 28 days after the treatment, blood was taken to detect total antioxidant capacity (T-AOC);right lung was taken to detect transforming growth factor-β1 (TGF-β1) and hydoroxyproline(Hyp) content, while the left lung was fixed. Result: On day 7, 14, 28, as compared with the sham-operation group, the TGF-β1 protein expressions were significantly increased (P<0.05, P<0.01);lung coefficient was increased significantly (P<0.01);Hyp content in lung tissues was increased significantly (P<0.05, P<0.01);and total antioxidant capacity in serum was decreased significantly (P<0.05, P<0.01) in the model group at each observation point. As compared with the model group, TGF-β1 protein expression was decreased significantly in Mongolian drug groups (P<0.05, P<0.01), and significantly decreased on day 14 and day 28 in hormone group (P<0.05);lung coefficient was significantly decreased in both Mongolian drug groups and hormone group (P<0.05, P<0.01);Hyp content in liver tissues was significantly decreased on days 7, 14, 28 in Mongolian drug high dose group and the hormone group (P<0.05, P<0.01), and significantly decreased on days 14 and 28 days in Mongolian drug low dose group and mid dose group (P<0.05, P<0.01);total antioxidant capacity in serum was significantly increased on day 14 and day 28 in Mongolian drug mid dose group, high dose group and hormone group (P<0.05, P<0.01), and significantly increased on day 28 in Mongolian drug low dose group (P<0.01). Conclusion: Mongolian Dongqingye-7 capsule has certain therapeutic effects for the pulmonary fibrosis induced by pingyangmycin in rats, and its preliminary analysis shows that the mechanism may be related to the expression of TGF-β1.

      摘要:Objective: To analyze the types and main potential causes of Diemailing (DML) allergic reactions, and explore the underlying mechanisms of such allergic reactions. Method: According to the Guideline for Immunotoxicity Research (Allergy and Photoallergic Reaction) on Chinese Medicine and Natural Medicine, the active systemic anaphylaxis (ASA) test and passive cutaneous anaphylaxis (PCA) test were carried out for DML ixeis sonchifolia hance injection. Serum IgE, C3, C4, interleukin(IL)-4, IL-10, IL-2, prostaglandin D2(PGD2), leukotriene C4 (LTC4), tumor necrosis factor (TNF)-α and interferon (IFN)-γ levels were determined by using ELISA. Result: ASA allergic symptoms, after stimulation, the ovalbumin group experienced the strongest positive symptom (death). The allergic symptoms of the non-ultrafiltrated DML group were stronger than those of the sodium chloride injection group and the ultrafiltrated DML group (P<0.01). Changes in serum immunological parameters:Compared with the sodium chloride injection group, some indexes in serum of the non-ultrafiltrated DML group were increased significantly such as IgE, IL-10, LTC4, TNF-α, IFN-γ and IL-2 (P<0.05, P<0.01), and the levels of PGD2, TNF-α and IFN-γ in ultrafiltrated DML group were also increased significantly (P<0.05, P<0.01). Compared with the ultrafiltrated DML group, Levels of serum IgE and IL-4 of the non-ultrafiltrated DML group were significantly increased (P<0.05, P<0.01). PCA:Positive rate of blue spots is zero in the sodium chloride injection group. Compared with the ovalbumin group, the diameters of blue spots were significantly decreased in both non-ultrafiltrated DML group and ultrafiltrated DML group (P<0.05, P<0.01). Compared with the sodium chloride injection group and ultrafiltrated DML group, the absorbance (A) of the non-ultrafiltrated DML group was significantly increased (P<0.01). The A of the ovalbumin group was significantly increased than that of the ultrafiltrated DML group (P<0.01). Conclusion: Type I allergy mediated by IgE is the main type of DML injection's allergic reactions. The macromolecular substance in DML can release various inflammatory mediators through IgE mediated immune stimulation. This kind of immunity reaction may be the main mechanism of allergic reactions in DML, so reducing macromolecular substances may be an important way to prevent allergic reactions of DML.

      摘要:Objective: To investigate effect of total flavonoids from Fagopyrum tataricum on expression of nicotinamide adenine dinucleotidephosphateoxidase-4 (NOX4) in EA. hy926 cell induced by palmitic acid, and to discuss mechanism of total flavonoids from F. tataricum in signal transduction pathway of insulin resistance. Method: EA. hy 926 cells were divided into blank group, insulin resistance model group, total flavonoids of F. tataricum group (31.25, 62.5, 125 mg·L-1) and metformin group.Every groups were combined insulin with 50 nmol·L-1.Compared with the blank group, other groups were combined 600 μmol·L-1 palmitic acid.Total flavonoids of F. tataricum group was combined 125 mg·L-1 total flavonoids of F. tataricum.Metformin group was combined 2 mmol·L-1 metformin.The content of asymmetric dimethylarginine (ADMA) was tested by double antibody sandwich method.The content of NO was tested by nitrate reductase method.NOX4 protein expression levels were determined by Western blot. Result: Compared with model group, contents of ADMA in the middle and high dose groups of total flavonoids were significantly reduced, contents of NO were increased significantly.Compared with model group, expression levels of NOX4 protein were reduced significantly in high dose groups of total flavonoids. Conclusion: Total flavonoids from F. tataricum can promote synthesis of NO by inhibiting synthesis of NOX4 and ADMA in EA. hy926 cell induced by palmitic acid, then it may restrain insulin resistance.

      摘要:Objective: To observe the effect of Naokang capsule on vascular dementia and hippocampal apoptosis genes in rats. Method: Vascular dementia models were established by permanent bilateral common carotid arteries occlusion in healthy male sprague-dawley rats, then were randomly divided into sham-operation group, model group, Naokang capsule group and ginkgo biloba extract (EGB) group. Morris water maze (MWM) was used to evaluate the learning and memory ability of rats after 4 weeks of intragastric administration. Expression levels of superoxide dismutase (SOD) and malonaldehyde (MDA) in hippocampal tissues of rats were detected by enzyme linked immunosorbent assay (ELISA). The mRNA and protein expression of Bcl-2 and Bax as well as Bcl-2/Bax ratio in hippocampus were detected by Real-time quantitative PCR and Western blot. Result: Compared with sham-operation group, the escape latency of model group was significantly prolonged;frequency of crossing platform and swimming distance were obviously shortened, SOD activity was decreased significantly and MDA activity was increased significantly, mRNA and protein expression levels of Bax were significantly increased and the levels of Bcl-2 were significantly decreased, resulting in a decreased Bcl-2/Bax ratio in hippocampus. Compared with the model group, the escape latency of the rats in Naokang group was significantly shortened, frequency of the crossing platform and swimming distance were obviously increased, SOD activity was increased significantly and MDA activity was decreased significantly, mRNA and protein expression levels of Bax were significantly decreased and the levels of Bcl-2 were significantly increased, resulting in an increased Bcl-2/Bax ratio. Conclusion: Naokang capsule could significantly improve the learning and memory ability of vascular dementia rats and the mechanism may be related to antioxidant stress and inhibiting hippocampal apoptosis genes expression.

      摘要:Objective: To investigate the effect of Jiangu tablets on bone metabolism, microarchitecture and stiffiness in an ovariectomized rat model. Method: Totally 50 Sprague-Dawley rats were randomly divided into the sham group (n=10), the model group (n=10), the estradiol valerate tablets group (n=10), the low-dose Jiangu tablets group (n=9) and the high-dose Jiangu tablets group (n=9). Ovaries were removed by surgery in each group to establish an ovariectomized rat model except sham group. Different groups were treated with different drugs or water. Estradiol valerate tablets group was administrated with estradiol valerate tablets at 0.1 mg·kg-1·d-1, low-dose Jiangu tablets group was administrated with Jiangu tablets at 0.35 g·kg-1·d-1, high-dose Jiangu tablets group was administrated with Jiangu tablets at 1.05 g·kg-1·d-1. The treatment maintained for 12 weeks. The body weight of rats was detected every week. Serum concentration of osteocalcin (OC), alkaline phosphatase (ALP), tartrate-resistant acid phosphatase (StrACP), calcium (Ca), and phosphorus (P) were determined after treated. Bone microarchitecture and bone mineral content (BMC), tissue mineral content (TMC), bone mineral density (BMD), tissue mineral density (TMD), bone volume fraction(BV/TV), trabecula number (TbN), trabecular separation (TbSp), trabecular thickness (TbTh) were detected by Micro-CT. The femur maximum load and stiffness were detected by a three-point bending testing. Result: Compared with the sham group, the levels of OC and ALP were increased significantly, the levels of Ca and P were decreased significantly (P<0.05, P<0.01), the trabecula of distal and middle femur in model group became rare, BMC, TMC, BMD, TMD, BV/TV decreased significantly (P<0.05, P<0.01), femur maximum load decreased significantly (P<0.05, P<0.01) in the model group. Compared with the model group, the levels of OC and ALP were significantly decreased, the levels of Ca was significantly increased in the the low-and high-dose Jiangu tablets group, and the level of P was significantly increased in the high-dose Jiangu tablets group (P<0.05, P<0.01). The levels of femur BMC, TMC, BMD, BV/TV were significantly increased in the low-and high-dose Jiangu tablets group(P<0.05, P<0.01), and the level of TbSp was significantly decreased in the high-dose Jiangu tablets group(P<0.05). The maximum load and stiffness of femur were significantly increased in the low-and high-dose Jiangu tablets group (P<0.05, P<0.01). Conclusion: Jiangu tablets influenced the bone metabolism, ameliorated the bone microarchitecture and increased the bone stiffness in an ovariectomized rat model.

      摘要:Objective: To investigate the effect of combination of paeonol (Pae) and Notoginseng Radix et Rhizoma saponins (NRS) on expressions of type Ⅰ and Ⅲ collagen in proliferation of cardiac fibroblasts induced by angiotensin Ⅱ(AngⅡ). Method: Primary cultured cardiac fibroblasts (CFs) of neonatal rats were randomly divided into six groups:blank group, model group, Pae group (45 mg·L-1), NRS group (50 mg·L-1), combined Pae and NRS high-dose and low-dose groups (Pae 45, 30 mg·L-1+NRS 50, 30 mg·L-1). The mRNA expressions of type Ⅰ and Ⅲ collagen were detected by RT-PCR after drug intervention for 48 h. The protein expressions of type Ⅰ and Ⅲ collagen were determined by Western blot. Result: As compared with the blank group, the mRNA expressions and protein expressions of type Ⅰ and Ⅲ collagen were significantly increased in model group (P<0.01). Compared with the model group, the proliferation of CFs were stimulated by AngⅡ, and the mRNA and protein expressions of type Ⅰ and Ⅲ collagen were inhibited by Pae group, NRS group, and the combined Pae and NRS high-dose and low-dose groups (P<0.05). The combined groups had better efffect than other groups, and the combined high-dose group had the best inhibitory effect. Conclusion: The combination of Pae and NRS can inhibit the gene expressions of type Ⅰ and Ⅲ collagen in cardiac fibroblasts induced by AngⅡ, and shows significant synergistic effects.

      摘要:Objective: To investigate the effects of Shenxiong glucose injection (SGI) on the activities and mRNA expressions of hepatic cytochrome P450 2E1 (CYP2E1), 3A11 (CYP3A11), 1A2(CYP1A2) and 2D22 (CYP2D22) in mice. Method: Kunming mice were randomly divided into normal group, hepatic microsomal enzyme inducer group, and SGI low dose, middle dose and high dose (13.0, 19.5, 26.0 mL·kg-1) groups. The mice were killed after medicines administration once a day for consecutive 7 days by tail intravenous injection. Liver microsomes were prepared for in vitro metabolism experiment to determine the enzyme activities of CYP2E1, CYP2D22, CYP1A2 and CYP3A, and quantitative real-time PCR was employed to examine the mRNA expressions of these four CYP450 enzymes in liver tissues of mice. Result: There was no significant difference in the liver indexes between various treatment groups and normal group. In vitro metabolism experiment showed that, as compared with the normal group, CYP2E1 activity was decreased by 0.45 times in the SGI high dose group (P<0.05), and CYP1A2 activities were increased by 1.20-1.23 times in the SGI middle dose and high dose groups (P<0.05). SGI was found to have no significant effect on the activities of CYP3A and CYP2D22. The results of fluorescence quantitative PCR showed that, as compared with the normal group, CYP2E1 mRNA expression was decreased by 0.45 times in the SGI high dose group (P<0.01), and CYP1A2 mRNA expressions were increased by 1.7 times and 2.6 times respectively in SGI middle dose and high dose groups (P<0.05). There was no significant difference between SGI groups and normal group in the mRNA expressions of CYP3A11 and CYP2D22. Conclusion: Middle dose and high dose SGI could up-regulate the hepatic CYP1A2 mRNA expressions and induce the enzyme activity of CYP1A2 in mice;high dose SGI could down-regulate the hepatic CYP1A2 mRNA expressions and inhibit enzyme activity of CYP2E1 in mice.

      摘要:Objective: To investigate the efficacy of Aidi injection combined with doctaxel combined with cisplatin(DP) chemotherapy regimen in female patients with advanced non-small cell lung cancer (NSCLC), and observe its effect on cellular immune function and quality of life. Method: We retrospectively analyzed clinical data of seventy-nine patients with advanced NSCLC. According to treatment regimens, they were divided into treatment group (41 cases) and control group (38 cases). Patients in control group were treated with DP regimen only, while patients in treatment group were treated with Aidi injection combined with DP chemotherapy regimen. Treatment for 21 days was regarded as one cycle, and evaluation was conducted after two cycles. Result: Efficacy was 51.2% in treatment group and 39.5% in control group. Disease control rate was 82.1% in treatment group and 66.7% in control group, with statistically significant difference between two groups (P<0.05). In treatment group, counts of CD3+ T cells, CD4+ T cells and CD4+/CD8+ after treatment were significantly higher than those before treatment and those in control group, while the count of CD8+ T cells was decreased (P<0.05, P<0.01). In treatment group, scores of domains in physiological status, social and family status and total scores after treatment were improved than those before treatment and those in control group (P<0.05, P<0.01). Major adverse effects included bone marrow suppression and gastrointestinal reactions. Incidences of leukopenia of Ⅲ/Ⅳ degree and decreased hemoglobin in treatment group were lower than those in control group, with statistically significant difference between two groups. Conclusion: Aidi injection combined with DP regimen could improve the efficacy, cellular immune function and quality of life in female patients with advanced NSCLC. Furthermore, it could decrease toxic and adverse effects, so it is worthy of further clinical application and promotion.

      摘要:Objective: To observe the effect of Tianma Gouteng decoction on the levels of blood lipid and catalase (CAT) in patients with wind-Yang nuisance type acute cerebral infarction. Method: Sixty-eight patients with wind-Yang nuisance type acute cerebral infarction were randomly divided into treatment group (34 cases) and control group (34 cases). The patients in control group received conventional cerebral infarction treatment(bayaspirin 100 mg qd and lipitor 20 mg qn), and the patients in the treatment group also received Tianma Goutengyin decoction on the basis of conventional treatment in control group, for a total of 2 treatment courses, with 7 days as one course of treatment.The changes of blood lipid, CAT, neurological function defect scores, and traditional Chinese medicine (TCM) symptom scores were detected before and after treatment. Result: Compared with those of the control group, the levels of total cholesterol(TC), triglyceride(TG), low-density lipoprotein cholesterol(LDL-C) and apolipoprotein-B(Apo-B) in the treatment group were reduced(4.08±0.67), (1.20±0.65), (1.98±0.24)mmol·L-1, (1.03±0.41)g·L-1, while the level of CAT was increased(20.07±5.83)U·L-1, there was statistical significance between two groups after treatment (P<0.05, P<0.01).The neural function defect score and TCM score were reduced in treatment group(3.65±2.15), (9.27±7.11), there was statistical significance between two groups after treatment (P<0.05).The total effective rate was 88.2% in the treatment group and 79.4% in the control group. The treatment group was better than the control group in clinical efficacy (P<0.05). Conclusion: Tianma Gouteng decoction can effectively reduce the levels of blood lipid and increase the CAT level, so that improve the antioxidant capacity in patients with wind-Yang nuisance type acute cerebral infarction.

      摘要:Objective: To observe the curative efficacy of Kanli granule in treating coronary artery disease (CAD) patients with syndrome of heart and kidney Yang deficiency, investigate its effect on function and structure of artery, and explore the possible mechanism of these effects. Method: Totally 62 CAD patients with syndrome of heart and kidney Yang deficiency were randomly divided into western medicine group (31 cases) and Kanli granule group (31 cases). Changes in heart function and arterial elasticity indexes cardio ankle vascular index (CAVI), ankle brachial index (ABI) were measured, aortic flow shear rate (SRs) and arterial remodeling conditions (intima-media thickness) were observed;indexes related to serum endothelial functions (NO, ET-1) were measured, levels of type I collagen metabolites procollagen I N-terminal peptide (PINP) and type I carboxy-terminal peptide (ICTP) were measured. The possible mechanism of Kanli granule effect on artery was investigated by observing the blood flow mediated changes in endothelial function and arterial wall structure. Result: After treatment with Kanli granule, left ventricular ejection fraction (LVEF) was significantly increased in CAD patients with syndrome of heart and kidney Yang deficiency (P<0.05). After treatments, ABI of arterial elasticity indexes was significantly improved in Kanli granule group (P<0.05), and the improvement was more obvious than that of western medicine group (P<0.05). SRs in western medicine group was significantly decreased after treatment(P<0.05). In serum indexes, iNOS was significantly reduced in western medicine group after treatment (P<0.05). Conclusion: Kanli granule can improve left ventricular systolic function and significantly improve arterial elasticity function in the patients with stable coronary artery disease and syndrome of both heart and kidney yang deficiency. Its mechanism may be associated with endothelial protection by raising SRs and reducing NO level activated by iNOS.

      摘要:Objective: To investigate the effect and mechanism of Jianpi Yishen method on improving the cancer-related fatigue in patients with advanced colorectal cancer, and to provide objective evidence for the traditional Chinese medicine (TCM) treatment of cancer-related fatigue. Method: Totally 150 cases of colorectal cancer patients with the spleen and kidney deficiency were collected and randomly divided into Chinese medicine treatment group and control group by using random number table method. They were asked to fill out a spleen and kidney deficiency symptoms rating table, Piper Fatigue Scale and Karnofsky (KPS) scale before and after treatment. The serum cytokines content was detected, then the relationship between cytokines and the cancer-related fatigue was investigated. The degree of fatigue, TCM clinical symptoms, quality of life, serum cytokines changes, and the degree of adverse reactions were compared between the two groups before and after treatment. Result: There were significantly positive correlations between the fatigue scores and symptom scores, and these two items were negatively correlated to the levels of interferon-γ (IFN-γ), interleukin-2 (IL-2), and Th1/Th2 in serum, but were positively correlated to IL-4 and IL-5 levels in colorectal cancer patients (P<0.05). No obvious adverse reactions were found in patients of both groups during the treatment, and the improvement in fatigue and the quality of life in the Chinese medicine treatment group was significantly better than that in the control group (P<0.05). The levels of Th1/Th2, IL-2 and IFN-γ in serum were significantly increased after treatment in two groups, and the increase degree in the Chinese medicine treatment group was higher than that in the control group (P<0.05);however the levels of IL-4 and IL-5 in serum were significantly decreased after treatment in both groups and the decrease degree in Chinese medicine treatment group was more obvious than that in the control group (P<0.05). Conclusion: The Jianpi Yishen method can redress the imbalance of Th1/Th2, and effectively improve the symptoms and the quality of life in the patients with advanced colorectal cancer.

      摘要:Objective: To discuss the correlation between cytokines, occurrence, development and prognosis of radioactive dermatitis, and investigate the effect and mechanism of Chinese herbal compound ulcer oil in prevention and treatment of radiation dermatitis. Method: The 80 eligible patients who received radiotherapy in head, neck and chest were prospectively enrolled in this study and randomly divided into observation group (ulcer oil group) and control group (western medicine group). Efficacy was observed, then changes in epidermal growth factor (EGF), tumor necrosis factor (TNF)-α, interleukin (IL)-6, IL-1, IL-8 and IL-10 in peripheral blood of the patients were detected to investigate action mechanism of ulcer oil in prevention and treatment of radiation dermatitis. Result: Ulcer oil can significantly delay occurrence of radiation dermatitis, reduce grade of radiation dermatitis, and accelerate healing rate of radiation dermatitis (P<0.05). At 30 Gy, EGF levels were significantly lower than those before radiotherapy in both groups (P<0.01). At 60 Gy, EGF levels of two groups were increased compared with 30 Gy, but the increase degree in observation group was significantly higher than that of control group (P<0.01). At 30 Gy, both groups showed significant increases in IL-6, TNF-α, IL-1 and IL-8 levels of peripheral blood (P<0.01). At 60 Gy, IL-6, TNF-α, IL-1 and IL-8 levels were decreased in both groups. In comparison between two groups, increase degree in IL-6, TNF-α, IL-1 and IL-8 levels in observation group was lower than that of control group at 30 Gy (P<0.05). At 60 Gy, IL-10 levels were increased in both groups, decrease degree in observation group was significantly lower than that of the control group (P<0.01). At 60 Gy, IL-10 level of the observation group was higher than that of the control group (P<0.05). Conclusion: Compound ulcer oil may delay occurrence of skin inflammation, and increase its recovery speed by inhibiting production and release of IL-6, TNF-α, IL-1 and IL-8, reducing decline speed of IL-10 at early stage, and promoting production of EGF at advanced stage.

      摘要:Objective: To investigate the clinical effects of traditional Chinese medicine (TCM) experienced prescription in assisting western medicine for premature ovarian failure. Method: The 120 patients with premature ovarian failure were chosen in the period from March 2014 to March 2015 in Hangzhou TCM Hospital and randomly divided into control group (60 cases) with estrogenic, and progesterone sequential therapy alone and TCM group (60 cases) with TCM experienced prescription (from Professor XIA Gui-cheng) on the basis of control group. Clinical efficacy, Kupperman scores and serum hormone levels before and after treatment of both groups were compared. Result: The total effective rate was 60.00% and 81.67% respectively in control group and TCM group, clinical efficacy of TCM group was significantly better than that of control group (P<0.05). Kupperman score after treatment in TCM group was significantly superior to that before treatment and that in control group (P<0.05). Levels of follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2) and anti-Mullerian hormon (AMH) in TCM group after treatment were significantly better than those before treatment and those in control group (P<0.05). Conclusion: TCM experienced prescription in assisting western medicine for premature ovarian failure can efficiently alleviate clinical symptoms and signs, promote the recovery of menstrual cycle and is helpful to improve serum hormone levels.

      摘要:Objective: To investigate efficacy of Erxian Bushen decoction combined with warm acupuncture in treating post-menopause osteoporosis(PMOP) with syndrome of kidney-Yang deficiency and its influences on levels of serum peroxidase body growth activated receptor γ synergy stimulating factor 1-α (PGC-1α), steroid receptor auxiliary activation factor 3 (SRC-3), estrogen-related receptor α(ERRα), and osteopontin (OPN) levels. Method: One hundred and twenty-four eligible cases were randomly divided into control group and treatment group (62 cases of each group) by random number table. Patients of control group were treated with alendronate sodium tablets (70 mg/time and 1 time/week) and calcium carbonate D3 tablets (qd). Based on the treatment of control group, patients in treatment group were also treated with Erxian Bushen decoction (1 dose/d and bid) combined with warm acupuncture (1 time/3 d). The treatment course was 6 months for both groups. Traditional Chinese medicine (TCM) symptom scores, bone mineral density (BMD) of right femoral neck and anteroposterior position of lumbar spine (L2-4) were compared between two groups. Levels of PGC-1α, SRC-3, ERRα, and OPN in serum were detected in two groups. Result: The TCM symptom scores in treatment group were lower than those of control group after treatment (P<0.01). Clinical total effective rate was 91.94% in treatment group, significantly higher than 77.42% in control group (P<0.05). BMD levels of right femoral neck and L2-4 in treatment group were significantly higher than those of control group after treatment (P<0.01). Levels of PGC-1α and SRC-3 in serum of treatment group were significantly higher than those of control group after treatment, while ERRα and OPN levels were significantly lower, with statistical differences (P<0.01). Conclusion: On the basis of western medicine routine treatment, Erxian Bushen decoction combined with warm acupuncture in treating PMOP with syndrome of kidney-Yang deficiency could obviously improve clinical symptoms, increase BMD levels of right femoral neck and L2-4, and improve clinical efficacy, which may be related with increasing the levels of PGC-1α and SRC-3 in serum and inhibiting levels of ERRα and OPN.

      摘要:Objective: To investigate the clinical efficacy of modified Buyang Huanwu detection in treating traumatic peripheral nerve injury with arteries and veins stasis and its influence on levels of nuclear factor-κB (NF-κB) and tumor necrosis factor-α(TNF-α) in serum. Method: Ninety-two cases with traumatic peripheral nerve injury were randomly divided into control group (46 cases) and treatment group (46 cases) by random number table. Patients in control group were treated with mecobalamin tablets by orally taking (1 tablet/time, tid) and electrical stimulation therapy (25 min/time, qd, and 6 times/week). Based on treatment in control group, patients of treatment group were also treated with modified Buyang Huanwu detection, 1dose/d and bid. Treatment course was 3 months for both groups. Traditional Chinese medicine(TCM) symptom scores were compared between two groups pre-post treatment. Motor nerve conduction velocity (MNCV) and sensory nerve conduction velocity (SNCV) were detected. Levels of NF-κB and TNF-α in serum were detected in two groups pre-post treatment. Result: TCM symptom scores in treatment group were significantly lower than those of control group after treatment (P<0.01). Cure-remarkable-effectiveness rate was 80.43% in treatment group, significantly higher than 56.52% in control group, with statistical differences (P<0.05). MNCV and SNCV levels of median nerve, ulnar nerve, radial nerve, nervus tibialis, and nervus peroneus communis in treatment group were higher than those of control group (P<0.01). After treatment, the levels of NF-κB and TNF-α in serum of treatment group were significantly lower than those of control group, with statistical differences (P<0.01). Conclusion: On the basis of western medicine treatment, modified Buyang Huanwu detection in treating traumatic peripheral nerve injury with arteries and veins stasis could obviously improve clinical symptoms and increase the conduction velocity of motor nerve and sensory nerve, with better clinical efficacy than western medicine alone. Inhibiting the levels of NF-κB and TNF-α in serum may be one of its mechanisms.

      摘要:Objective: To discuss the curative effect of Longdan Xiegan Tang combined with Ditan Tang in treatment of epilepsy attack with syndrome of phlegm-fire disturbing spirit and investigate its influences on anti-oxidative stress of patients. Method: One hundred and seven cases were included in this study and were randomly divided into treatment group (52 cases) and control group(55 cases) according to random number table. Patients in both groups received therapy of the recommended drugs with reference to The Clinical Diagnosis and Treatment Guidelines of Epilepsy. The patients in treatment group also received Longdan Xiegan Tang combined with Ditan Tang (1 dose/d, bid) on the basis of treatment in control group. The treatment course was 12 weeks for both groups. Epileptic seizure frequency and time of duration were recorded in two groups pre-post treatment. Quality of life was compared between two groups with reference to Quality of Life in Epilepsy (QOLIE-31). Total superoxide dismutase (SOD), reduced glutathione (GSH), glutathione peroxidase(GSH-Px) and malondialdehyde (MDA) levels in serum were detected in two groups. Result: Epileptic seizure frequency and time of duration in treatment group were significantly lower than those of control group after treatment (P<0.01). The total effective rate 88.46% in treatment group, significantly higher than 70.91% in control group (P<0.05). Every index score of QOLIE-31 in treatment group was obviously higher than that of control one after treatment (P<0.01). The levels of SOD, GSH and GSH-Px in serum of treatment group were significantly higher than those of control group, while MDA level was significantly lower than that of control group after treatment, with statistically significant differences (P<0.01). Conclusion: Longdan Xiegan Tang combined with Ditan Tang based on western medicine in treatment of epilepsy attack with syndrome of phlegm-fire disturbing spirit could evidently reduce epileptic seizure frequency and time of duration, improve quality of life and curative effect for the patients. The effect may be related to increasing anti-oxidative stress capacity.

      摘要:Objective: To investigate the efficacy of Fuzi Lizhong Tang plus Sishen Wan in treating diarrhea type irritable bowel syndrome (IBS-D) with spleen and kidney deficiency syndrome and observe its effect on protease activated receptor 2 (PAR2)and PAR4 of mucous membrane of rectum. Method: One hundred and twenty IBS-D patients were randomly divided into treatment group (60 cases) and control group (60 cases) according to random number table. All cases were given with non-drug therapy referring to Diagnosis and Treatment Plan of Integrated Chinese and Western Medicine for Irritable Bowel Syndrome. Patients of control group also orally received bacillus subtilis and enterococcus faecium enteric-coated capsules, 2 tablets/time and bid. While the patients of treatment group were also given with Fuzi Lizhong Tang plus Sishen Wan, 1dose/d and bid. The treatment course was 6 weeks for both groups. The clinical symptom scores, stool property score, and defecation were compared between two groups. PAR2 and PAR4 levels of mucous membrane of rectum were detected for two groups. Result: The clinical symptom scores in treatment group were significantly lower than those in control group after treatment (P<0.01). The clinical efficacy was 96.67% and 83.33% respectively in treatment group and control group, with statistically significant difference between two groups (P<0.05). Scores of stool property and days for defecation urgency within 10 days in treatment group were significantly decreased after treatment (P<0.01). After treatment, PAR2 and PAR4 levels of mucous membrane of rectum in treatment group were lower than those in control group, with statistically significant difference (P<0.01). Conclusion: Fuzi Lizhong Tang plus Sishen Wan in treatment of IBS-D with spleen and kidney deficiency can decrease symptom scores, improve defecation, enhance clinical efficacy, and down-regulate PAR2 and PAR4 levels of mucous membrane of rectum.

      摘要:Objective: To observe the efficacy of integrated Chinese and western medicine on knee osteoarthritis (KOA) with syndrome of cold-dampness blocking collaterals and investigate its effect on stromal cell derived factor 1(SDF-l) and Irisin. Method: One hundred and fourteen cases with KOA patients were divided into control group (56 cases) and treatment group (58 cases) according to visiting sequence referring to random number table. Patients in control group were given with glucosamine hydrochloride tablets (2 tablets/time, 3 times/d) and self management training (1 time/week and 2 h/time). Based on the treatment of control group, the patients in treatment group were also given with mustard seed powder point application (3-5 h/time and 1 time/week) and manipulative therapy (30 min/time and 1 time/d). All patients were treated for 6 weeks. Scores of the Western Ontario and McMaster Universities (WOMAC), qualify of life, and clinical efficacy were compared between two groups. SDF-1 and Irisin levels in serum and synovial fluid were detected in both groups. Result: Scores of various indexes of WOMAC scale and total score in treatment group were significantly lower than those in control group after treatment (P<0.01). Scores of all indexes of SF-36 scale in treatment group were higher than those in control group (P<0.01). Ridit analysis showed that the treatment group was superior to control group in clinical efficacy (P<0.05). After treatment, SDF-l levels in serum and synovial fluid in treatment group were significantly lower than those in control group, while Irisin level was significantly higher that of control group (P<0.01). Conclusion: Efficacy of integrated Chinese and western medicine on knee osteoarthritis with syndrome of cold-dampness blocking collaterals is exact, and the mechanism may be related to inhibiting SDF-1 level in serum and synovial fluid and up-regulation of Irisin.

      摘要:Objective: To explore the molecular mechanisms of effective or toxic components of Kansui Radix and Glycyrrhizae Radix et Rhizoma by using network pharmacology method, and to investigate the correlations between multi-compounds, multi-targets and multi-pathways. Method: The main effective or toxic components of Kansui Radix and Glycyrrhizae Radix et Rhizoma, including kansuinine A, kansuinine B, kansuinine C, gycyrrhizic acid, liquiritin, isoliquiritin, formononetin were collected to build multi-components-proteins networks based on PharmMappear database. The obtained targets information was used to obtain corresponding pathways by using Boao MAS 3.0 system, and then Cytoscape software was used to construct components-targets-pathways network model. Result: The network analysis showed that, action of the effective or toxic components of Kansui Radix and Glycyrrhizae Radix et Rhizoma involved 61 pathways related to nerve-immune-endocrine-urinary. Contrary to their efficacy in adjusting water and electrolyte excretion, they were related with renin-angiotensin system, steroids biosynthesis and other pathways. Kansui Radix and Glycyrrhizae Radix et Rhizoma had synergistic effects in anti-tumor, anti-inflammatory and immune regulation, covering 33 pathways. Compatibility of Kansui Radix and Glycyrrhizae Radix et Rhizoma would enhance toxicity concerned to inhibition or induction of hepatic drug enzyme. Conclusion: Compatibility of Kansui Radix and Glycyrrhizae Radix et Rhizoma could result in decreased or synergistic effect and would change the toxicity. This reflected the compatibility mode of multi-compounds, multi-targets and multi-pathways for traditional Chinese medicines. It may provide reference for deeper study on molecular mechanism of compatibility of Kansui Radix and Glycyrrhizae Radix et Rhizoma.

      摘要:Objective: To analyze the composition principles in traditional Chinese medicine (TCM) prescriptions for postpartum lochiorrhea based on the TCM Inheritance Support System (TCMISS). Method: The prescriptions for postpartum lochiorrhea in Dictionary of Chinese Medicine Prescription were collected and their composition principles were analyzed after data entry into the TCMISS. Result: Based on the analysis of 111 prescriptions including 215 herbs for postpartum lochiorrhea, there were 45 herbs with frequency≥6, 48 herbal combinations with support≥10, 13 association rules with support≥10 and confidence coefficient≥0.85. 18 new core combinations and 4 new prescriptions were obtained from the databases. Conclusion: In Dictionary of Chinese Medicine Prescription, herbs for enriching blood and herbs for invigorating blood circulation and eliminating stasis were used as the core compositions in prescriptions for postpartum lochiorrhea, often combined with Angelicae Sinensis Radix and Chuanxiong Rhizoma, and supplemented by herbs of reinforcing Qi, interior-warming herbs and the herbs of clearing heat and cooling blood.

      摘要:Objective: To investigate the prescription experiences of professor ZHOU Zhong-ying in treating rheumatoid arthritis (RA) based on complex networks analysis. Method: Totally 166 RA cases (845 encounters) were selected as the clinical cases, and their diagnosis and treatment information was transformed into structured electronic medical record system.Then the complex network method was used with ‘Prestigious Traditional Chinese Medicine Specialists Clinical Information Collection System’, to dig out the core herbs and prescription rules of professor ZHOU Zhong-ying for treating RA. Result: The core herbs used by professor ZHOU Zhong-ying for RA included Sinomenii Caulis, Rehmanniae Radix, Astragali Radix, Euonymus Alatus, Stephaniae Tetrandrae Radix, Paeoniae Radix Rubra, Bombyx Batryticatus, Scorpio, Arisaema Cum Bile, Cinnamomi Ramulus, Phellodendri Chinensis Cortex, etc. The main therapy for RA was based on dispelling wind and eliminating dampness, clearing heat and dredging collaterals, warming channel and expelling cold, dissipating phlegm and removing blood stasis, strengthening the body resistance and tonifying deficiency. Conclusion: The results of complex network analysis fully reflected professor ZHOU Zhong-ying's clinical perceive of pathogenesis, syndrome differentiation and medication features of RA.

      摘要:Objective: To systemically evaluate the efficacy and safety of Yanhuning injection for the children with viral pneumonia. Method: We searched CBM, CNKI, VIP, Wanfang and PubMed for the randomized controlled trials on Yanhuning injection treatment for children with viral pneumonia. The studies were screened according to the inclusion and exclusion criteria, and the trials were evaluated according to Jadad quality assessment criteria, then Revman 5.3 software was used for Meta-analysis. Result: A total of 10 randomized controlled trials were included in the systematic review. Meta analysis showed that:the total clinical effective rate [OR=4.54, 95%CI (2.73, 7.56), P<0.000 01], fever abatement time [MD=-1.15, 95%CI (-1.58, -0.71), P<0.000 01], cough disappearing time [MD=-2.07, 95%CI (-3.08, -1.07), P<0.000 1], and pulmonary rales disappearing time [MD=-2.24, 95%CI (-2.53, -1.94), P<0.000 01] in Yanhuning injection group were better than those in control group in the treatment of children with viral pneumonia, with statistically significant difference. Conclusion: Yanhuning injection in the treatment of viral pneumonia in children can significantly improve efficiency and shorten the duration of fever, relieve cough symptoms in children, and accelerate the disappearance of pulmonary rales, without significant adverse reactions. However, the existing small sample randomized controlled trials had low methodological and reporting quality, so further high-quality, large-scale clinical studies are required for confirmation.

      摘要:Objective: To systemically assess the efficacy and safety of Tongxinluo capsule combined with clopidogrel in treatment of angina pectoris. Method: Such databases as CBM, CNKI and Wanfang data were electronically searched to collect relevant randomized controlled trials (RCTs). According to the inclusion and exclusion criteria, two reviewers independently screened literature, extracted data, and assessed the methodological quality of included studies. Then, Meta-analysis was conducted using RevMan 5.2.6 software. Result: The 10 RCTs with 867 patients were included. Tongxinluo capsule combined with clopidogrel had a better efficacy on angina pectoris than clopidogrel alone [OR=4.01, 95%CI (2.68, 5.99)], with statistically significant difference between the two groups (P<0.000 01). There was no statistically significant difference in electrocardiography (ECG) efficacy between Tongxinluo capsule combined with clopidogrel and clopidogrel alone [OR=1.67, 95%CI (0.94, 2.98), P=0.08]. The subgroup analyses based on regimen in the control group showed that, compared with clopidogrel alone group, the Tongxinluo capsule combined with clopidogrel group had a higher efficacy on angina pectoris, with statistically significant difference between the two groups [OR=6.95, 95%CI (3.87, 12.69), P<0.000 01]. Compared with clopidogrel combined with inosine group, the Tongxinluo capsule combined with clopidogrel group had a higher efficacy on angina pectoris, with statistically significant difference between the two groups [OR=2.61, 95%CI (1.06, 6.40), P<0.05]. Indirect treatment comparison showed that there was no significant difference in efficacy on angina pectoris between clopidogrel alone group and Tongxinluo capsule alone group [OR=1.19, 95%CI (0.42, 3.39), P>0.05]. Conclusion: Existing evidences suggest that Tongxinluo capsule combined with clopidogrel is effective and safe in improving angina pectoris. However, the quality of RCTs is not high, so more randomized double-blind clinical trials with high quality are still needed for further verification.

      摘要:Objective: To analyze the Chinese medication regularity and experience of professor YU De-zhao for treating the chronic gastritis in children induced by helicobacter pylori, and provide more ideas for clinical treatment of chronic gastritis in children induced by helicobacter pylori. Method: Total 77 pediatric outpatient cases including 243 electronic medical records for chronic gastritis treated by professor Yu from June 2014 to June 2015 were collected. Absolute frequency analysis, relative frequency analysis, and Hierarchical Cluster analysis were used to analyze and evaluate the Chinese medicine regularity. Result: The 9 herbs were used with a frequency over 50.00%, 11 herbs were used with a frequency between 20.00% and 50%, and 13 herbs were used with a frequency below 20.00%. Moreover, theses 33 herbs could be divided into 7 types by Hierarchical Cluster analysis. Conclusion: The regularity of Chinese medication of professor Yu for treating the chronic gastritis in children induced by helicobacter pylori was based on the physiological characteristics of spleen and stomach in children, focused on the lung-regulating and spleen-tonifying therapy with combination of liver soothing and Qi regulating. At the same time, professor Yu also paid attention to the damp-and-hot accumulate knot, and usually adjusted measures to differing conditions, with local characteristics of Lingnan area.

      摘要:Chinese herbal compound formula (CHCF) are widely used in clinic with development of evidence-based medicine, it has put forward a new challenge on CHCF.It becomes a strategic planning to further research and develop CHCF, provide sufficient curative evidence for promoting modernization of traditional Chinese medicine.This paper puts forward CHCF evidence-based biosystem research strategy (CHCF-EB-BRS), which conducts multi-level study of CHCF from ancient medical books, system evaluation and analysis, quality control, material basis and so on.In order to reveal material basis for efficacy of CHCF and its related mechanism, solve bottleneck of CHCF.CHCF clinical efficacy is verified multiply from ancient medical books, system evaluation and clinical study, furthermore, taking a good quality control of CHCF for laboratory and clinical research.Various research levels of the strategy has been widely implemented in basic and clinical research, but it still lacks of a systematic study of CHCF.CHCF-EB-BRS is helpful to urther research on development of traditional Chinese medicine.

      摘要:The major clinical manifestations of vascular dementia (VaD) include cerebrovascular accidents and cognitive dysfunction, whole occurrence and development are closely related to the cerebral blood flow and neuron survival state. Studies have shown that the phosphoinositide 3-kinase (PI3K)/protein kinase B (PKB/Akt) signaling pathway widely exist in various cells. In brain microvascular endothelial cells (BMEC), it can regulate the vasomotor function and cerebral blood flow through downstream endothelial nitric oxide synthase (eNOS)/nitric oxide (NO) pathway;in neurons, it can promote neuronal survival against neuronal apoptosis, which is one of the important parts in the occurrence and development of VaD. So far, some studies have shown that traditional Chinese medicine, active ingredients and compounds can play the neuroprotective effect through the activation of PI3K/Akt signaling pathway. In order to provide reference for further in-depth study in the future, it is necessary to make a review on the research progress in this filed.

      摘要:Objective: Obesity has become one of the crucial social health problems.Yet there are many controversies, it is extensively accepted that simple obesity is based on energy absorption greater than consumption, leading to a lot of fat accumulation.Reasons for obesity formation are quite complex, intestinal flora imbalance, energy metabolic disorders, low level of inflammatory and immune dysfunction are the main causes.The present work reviews current advances on exploration of relationships of immune dysfunction with nutritional obesity, and cross interaction among immune, endocrine and metabolism.We conclude that obesity is closely related to body's immune status.Our group will continue to verify feasibility of this theory in the next experiment, in order to find new targets and methods for obesity treatment based on intestinal mucosal immune, it can provide novel thought for treatment of obesity and related metabolic diseases.
