      摘要:Objective: To discuss the influence of frying and pounding on contents and decocting rates of active components in Cassiae Semen. Method: HPLC was employed to simultaneously determine contents and decocting rates of three naphthopyrone glycosides (rubrofusarin gentiobioside,cassiaside and cassiaside C) and three anthraquinones (aurantio-obtusin,methyl-obtusifolin and obtusifolin) in Cassiae Semen before and after processing,acetonitrile-tetrahydrofuran-1% acetic acid for gradient elution was used as the mobile phase,detection wavelength was set at 278 nm. Result: The content of water soluble extract of Cassiae Semen increased 1.7% after frying,this index of raw products and fried products raised 13.3% and 13.1% after pounding,respectively.Contents of three naphthopyrone glycosides in decoction were dropped after frying,but three anthraquinones were increased.The above results were similar to raw products after pounding by comparing with raw products without pounding.After pounding,decocting rates of three naphthopyrone glycosides in fried products increased by comparing with fried products without pounding,but these of three anthraquinones did not change significantly.Decocting rates of five compounds were dropped and one compound had no abvious change in fried products without pounding;but three naphthopyrone glycosides went up and three anthraquinones dropped in fried products after pounding. Conclusion: Both of frying and pounding have a great impact on contents and decocting rates of active compounds in Cassiae Semen,especially the latter.And in frying and decocting process,glycosidic enzymes contained in Cassiae Semen are involved in promoting the transformation of these active ingredients in this herb.

      摘要:Objective: pH responsive polymer micelles loaded with cantharidin were prepared to increase the solubility of cantharidin in water.The pH responsive property of the micelles was verified by the controlled drug release under tumoral mimic mild acidic environment. Method: Cantharidin micelles were prepared by thin film hydration method with amphiphilic methoxy poly(ethylene glycol)-b-polylactide (PELA) block copolymer and methoxy poly(ethylene glycol)-b-polylactide-b-poly(β-amino ester) (PELA-PBAE) as carriers.The structure and morphology of the micelles was characterized by transmission electron microscopy.The particle size of the micelles was measured by laser light scattering analyzer.HPLC was employed to determine drug loading and encapsulation efficiency of the micelles.In vitro release of the micelles was performed in phosphate buffered saline (pH of 7.4, 6.5 and 5.5,respectively) by permeation diffusion method. Result: The micelles of PELA-cantharidin (PELA-C) and PELA-PBAE-C exhibited spherical shape with particle size of 21.6 nm and 21.2 nm,respectively;they were at least stable within 5 d.Drug loading and encapsulation efficiency of PELA-C micelles were 1.68% and 92.0%,while these of PELA-PBAE-C micelles were 1.72% and 88.8%,respectively.The cumulative release rates of PELA-C micelles and PELA-PBAE-C micelles in 8 h were 24.7% and 79.1% under pH of 5.5,13.6% and 50.0% under pH of 6.5,10.2% and 13.9% under pH of 7.4,respectively. Conclusion: The solubility of cantharidin is significantly improved with PELA-PBAE as nanocarrier,in vitro release of PELA-PBAE-C micelles has significant pH responsive performance.

      摘要:Objective: To compare the content of volatile ingredients components between Scutellariae Radix and charred Scutellariae Radix. Method: Volatile components were extracted by steam distillation.Volatile components in these two products were analyzed by GC-MS and identified by NIST-05 standard mass spectra library. Result: The contents of volatile components in Scutellariae Radix and charred Scutellariae Radix were 1.3,0.8 μL·g-1,respectively.Fifty kinds of volatile components were identified,including 25 kinds in raw products and 31 in processed products.Based on the peak area normalization method,25 kinds of volatile components consisted in Scutellariae Radix accounting for 100%,31 kinds of volatile components in charred Scutellariae Radix accounting for 99.93%. Conclusion: After being processed into charred Scutellariae Radix,the composition and content of volatile components in Scutellariae Radix change a lot,while there are still some volatile components exist in charred Scutellariae Radix.This research can provide a scientific basis for elucidating the mechanism of Scutellariae Radix nature partly preserves after being charred.

      摘要:Objective: To optimize the formulation of self-microemulsion containing total flavonoids from Folium Kaki by simplex lattice method. Method: Based on screening of preparation process,taking rates of oil phase,emulsifier and co-emulsifier as factors,simplex lattice method was adopted to optimize formulation with particle size,polydispersity index and drug loading as indexes.Physicochemical properties of this self-microemulsion were determined,such as viscosity,dissolution and so on. Result: Optimum formulation was 10% of median chain triglyceride oil,30% of polyoxyethylene hydrogenated castor oil,60% of diethylene glycol ethyl ether.Average particle size,polydispersity index and drug loading of self-microemulsion containing total flavonoids from Folium Kaki were 32.7 nm,0.163 and 22.42 mg·g-1;physicochemical properties was stable,cumulative dissolution at 20 min was 89.09%. Conclusion: Simplex lattice method can build a mathematical models with good prediction and objective analysis,and it can provide a new idea for optimization of self-croemusion formulation.

      摘要:Objective: To optimize separation and purification technology of iridoid glycosides from Lonicerae Japonicae Flos by macroporous resin. Method: The content of loganic acid,7-epi-loganin,sweroside and secoxyloganin was determined by UPLC,static adsorption-elution test was adopted to screen type of macroporous resin,separation and purification process conditions of iridoid glycoside were optimized by single factor tests. Result: H-103 macroporous resin was selected,optimum technology was:sample solution concentration of 100 g·L-1,adsorption flow rate of 2.0 BV·h-1,sample liquid volume of 2.5 BV,eluted by 2.5 BV of water and 3.0 BV of 30% ethanol at elution flow rate of 2.0 BV·h-1.Under these conditions,elution rate and purity of iridoid glycosides were 90.0% and 25.6%,respectively. Conclusion: Iridoid glycosides from Lonicerae Japonicae Flos are purified effectively with H-103 macroporous resin,this optimized process is simple,stable and feasible.

      摘要:Objective: To select the best semi-bionic extraction method of Zhizi Bopi Tang, and provide the scientific basis for extracting process of Zhizi Bopi decoction. Method: The herbs in the prescription were arranged in different combinations based on ‘Jun(A), Chen(B), Zuo(C) and Shi(D)’ principle. There were 5 combinations in total, and semi-bionic extraction method was used for extraction. The levels of gardenoside, berberine hydrochloride, glycyrrhetinic acid, total alkaloids and extracts with molecular weight≤1 000 Da were used as the indexes. The comprehensive evaluation values were acquired by using weighted summation of the five indexes after standardized treatment. Those with larger comprehensive evaluation values were better than others. Result: The comprehensive evaluation value Y was B+AC >A+B+C >C+AB >ABC >A+BC respectively for the 5 indexes, and B+AC was the biggest value. Conclusion: The semi-bionic extraction method of Zhizi Bopi Tang is decocting Cortex Phellodendri Chinensis solely first, then decocting Gardeniae Fructus and Glycyrrhizae Radix together, and finally combining the above extract Tang.

      摘要:Objective: To investigate the germplasm distribution of genus Epimedium in Hubei province, systemically evaluate the quality and achieve sustainable utilization and development of resources. Method: The germplasm of Epimedium in Hubei province was determined by reviewing the literature, specimen and monograph, as well as field investigation. Population samples collected at full-bloom stage were used for the subsequent quality evaluation based on epimedin A, epimedin B, epimedin C and icariin. Result: According to the record of relative literature, specimens and our field investigation, 24 species and 1 variety of Epimedium may be expected in Hubei. However, due to the frequent revision and description of this genus, and some endemic species were easily affected by habitat origins, only 11 species were collected in field investigation. The quality evaluation showed that E.sagittatum was the first class, followed by E.mikiorii, E.dolichostemon and E.wushanense. E.sutchuenense had low concentration of the above four ingredients. While the four ingredients were not detected in E.epsteinii, E. franchetii, E. zhushanense, E.leptorrhizum, E.baojingense and E.brachyrrhizum. In addition, there was significant difference between populations in both E.dolichostemon and E. sagittatum. Conclusion: Due to the changes of taxonomic treatment and natural habitat, it is time to update the germplasm information. Hubei province is rich in natural Epimedium resources. Most species that were not recorded in Chinese pharmacopoeia have inferior quality, but E. mikiorii and E. dolichostemon have better quality, whose excellent population may be used as candidate germplasm for introduction and acclimatization.

      摘要:Objective: To carry out the pharmacognostic identification of Toddalia asiatica root bark including plant tissue cross section and fine powder microscopic identification,its hemostatic activity and sample preparation method. Method: Through making horizontal slices of full root of T.asiatica,water loading slides,permeabilizing slides and staining slides of the root bark and the root cores respectively,the tissue section characteristics of full root of T. asiatica were improved in traditional means of pharmacognostic identification,fine powder microscopic characteristics between the root bark and the root cores of T. asiatica were also investigated.For hemostatic activity of T. asiatica root bark,bleeding time (BT),amount of bleeding (BA) and clotting time (CT) were chosen as indexes,different extraction methods and polarity fractions were studied by typical mouse tail-cutting method and glass capillary tube method. Result: T. asiatica root bark contained a lot of stone cells,calcium oxalate crystals,cork cells, oil cells,et al.Meanwhile,the root cores had wood fibers,calcium oxalate crystals,textured pit catheters,et al.Both 95% ethanol cold maceration method and 50% ethanol reflux method were suitable for extraction of T. asiatica root bark.The ethyl acetate fraction of cold maceration extract showed the best hemostatic activity by comparing with petroleum ether fraction and n-butanol fraction.Under administration of 1.50 g·kg-1 ethyl acetate fraction of cold maceration extract,average values of BT,BA and CT were (59.67±12.31) s,(4.42±1.67) mg and (79.67±5.57) s. Conclusion: Stone cells,cork cells and oil cells are characteristic items of T. asiatica root bark,wood fibers only existes in T. asiatica root cores relatively.Combination of cold maceration method and ethanol reflux method is reasonable for extraction of T. asiatica root bark.T. asiatica root bark has excellent hemostatic and coagulation effects,and hemostatic effect of ethyl acetate fraction of cold maceration extract is the best.

      摘要:Objective: To analyze the distribution rules of SSR loci in Curcuma kwangsiensis EST, develop C. kwangsiensis EST-SSR primers, and explore the feasibility of EST-SSR for C. kwangsiensis species genetic diversity. Method: The 12 678 items of Curcuma EST (expressed sequence tag) sequences were downloaded from NCBI database, and MISA software was used to find SSR loci and select qualified sequences. Primer 5.0 software was used to design EST-SSR primers; polyacrylamide gel electrophoresis (PAGE) was used to analyze the PCR amplification characteristics of these EST-SSR primers, and further verify the rationality and validity of the developed results. Result: From 12 678 downloaded EST sequences, 926 sequences contained SSR loci, accounting for 7.30% of the entire EST database. The largest proportion for SSR loci was in dinucleotide repeat sequences, followed by trinucleotide and tetranucleotide sequences, 623 (50.90%), 388 (31.70%) and 125 (10.21%) respectively. A total of 165 EST-SSR primer pairs were designed according to the microsatellite sequences, and 24 of them with more than 92 points were selected for synthesis. PCR detection showed that 21 primer pairs (87.50%) could amplify stable and clear bands; in 6 different Germplasms in C. kwangsiensis, 13 pairs of EST-SSR primers showed polymorphism, accounting for 54.17% in the primer design. 13 pairs of EST-SSR primers were used to verify the phylogenetic relationship for 20 C. kwangsiensis varieties. Conclusion: C. kwangsiensis EST-SSR markers have a higher development efficiency, so it is one of the important measures for development of C. kwangsiensis SSR markers, with vital significance for the identification of C. kwangsiensis variety, genetic polymorphism analysis and breeding.

      摘要:Objective: To identify the steroidalsaponins in the tubers of Ophiopogonis Radix by ultra-performance liquid chromatography tandem with time-of-fight mass spectrometry (UPLC-Q-TOF-MSE)combined with the relative retention time (RRT) of furostanolsaponins in reverse chromatography. Method: The retention time of several furostanolsaponins was tested on two kinds of reversed-phase column, Acquity UPLC HSS T3 and Agelavenusil XBP C18-2,using water (0.1% formic acid)-acetonitrile (0.1% formic acid) and acetone-water, respectively. The mass spectrometric data of Ophiopogonis Radix were obtained by UPLC-Q-TOF-MSE. Result: The RRT of furostanolsaponins was consistently in three chromatographic conditions. When the sugar parts were identical, the RRT was shorter alone with the increase number of hydroxyl; location of hydroxyl affected the RRT, the orders were asfollow:1-hydroxyl< 17-hydroxyl< 14 -hydroxyl. When only the sugar chain at C-26 was different, the RRT was as follow:glucosyl-(1→6)-glucosylEcombined with RRT, a total of 21 compounds were identifiedincluding 14 furostanolsaponins and 7 spirostanolsaponins. Conclusion: The RRT of furostanolsaponins provide supportive data for rapidly identify isomers of steroidalsaponins from Ophiopogonis Radix by UPLC-Q-TOF-MSE.

      摘要:Objective: To study the secondary metabolites from endophytic fungus AR08 of Astragalus membranaceus. Method: The compounds were isolated and purified by silica gel column chromatography, Sephadex LH-20, and HPLC. Their structures were identified by the spectral analysis. Result: Six fumiquinazoline alkaloids were obtained and elucidated as fumiquinazoline J (1), fumiquinazoline I (2), fumiquinazoline C (3), fumiquinazoline H (4), fumiquinazoline D (5), and fumiquinazoline B (6). Conclusion: Compounds 2 and 4 were obtained from the plant endophytic fungus for the first time.

      摘要:Objective: To extract and isolate the chemical constituents from the sporophore of Phellinus lonicerinus and identify their structures. Method: The ethyl acetate extracts of the sporophore of P. lonicerinus were separated and purified by silica gel, RP-18, and Sephadex LH-20 column chromatography. Their structures were identified by spectroscopic analysis. Result: Ten compounds were isolated and identified as methyl linoleate (1), (22E,24R)-ergosta-4,6,8(14),22-tetraen-3-one (2), (22E,24R)-5α,8α-epidiory-ergosta-6,22-dien-3β-ol (3), (22E,24R)-ergosta-5,7,22-trien-3β-ol (4), 4-hydroxyphenethyl alcohol (5), furan-2,5-diyldimethanol (6), O-hydroxyphenylacetic acid (7), cyclo-(Leu-Val) (8), (22E,24R)-ergosta-7,22-dien-3β,5α,6β-triol (9), and cerebroside D (10). Conclusion: All the compounds except compounds 3 and 4 were isolated from P. lonicerinus for the first time.

      摘要:Objective: To obtain the information about the antioxidant and antimicrobial activities of components from the fresh flower bud of Zingiber mioga. Method: The flower bud of Z. mioga was extracted and concentrated with methanol, and after extraction with petroleum ether, dichloromethane, ethyl acetate, n-butanol and water, the five fractions were evaluated for antioxidant activities by DPPH radical scavenging method and antibacterial activities on four strains (Escherichia coli, Salmonella lignieres, Pseudomonas aeruginosa and Staphylococcus aureus) by agar disc diffusion method. Result: The extraction rate of Z. mioga flower bud was 3.1% in heating reflux method; the ethyl acetate extract showed the highest scavenging capacity on DPPH radical with IC50 of (26.22±1.12) mg·L-1, and its inhibition zone on Pseudomonas aeruginosa was (10±1) mm; petroleum ether fraction showed highest antibacterial activities, with minimal inhibitory concentration(MIC) of 25.0 mg·L-1 on Staphylococcus aureus. Conclusion: All of fractions of the methanol extract from Z. mioga flower bud showed antioxidant abilities. Their potency of antibacterial activity was as follows:petroleum ether fraction > dichloromethane fraction > ethyl acetate fraction > methanol extract, providing theoretical basis for the utilization and development of Z. mioga flower bud antioxidant and bacteriostatic products.

      摘要:Objective: To study the correlation between the color of Celosiae Cristatae Flos powder and its chemical components. Method: Colorimeter was used to determined L*, a* and b* values of Celosiae Cristatae Flos powder, RP-HPLC was used to determine the content of the four active components, spectrophotometer was adopted to determined total flavonoids in Celosiae Cristatae Flos (based on rutin), and SPSS 19.0 software was used to analyze the correlation between color values and content of active component in this study. Result: In Celosiae Cristatae Flos, L* and luteolin, quercetin, kaempferol, isorhamnetin were significantly negatively correlated; a* and luteolin, quercetin, kaempferol were significantly positively correlated; and b* and luteolin, quercetin, kaempferol were significantly negatively correlated. a*, b* and isorhamnetin were uncorrelated, and L*, a*, b* and total flavonoids (based on rutin) were significantly negatively correlated. Conclusion: There was a significant correlation between the color values and the content of the four effective components. It is energy-saving and convenient to assess the quality control of Celosiae Cristatae Flos by determining the color of powder. Although the new method can be applied to assess the quality of Celosiae Cristatae Flos and provide reference, some problems still shall be further studied.

Method: The separation was performed on UHPLC Welch C18 column (2.1 mm×100 mm,1.7 μm), with a mobile phase using water containing 0.1% formic acid (A) and acetonitrile (B) for gradient elution, the flow rate was 0.35 mL·min-1. Electrospray ion (ESI) source were applied for the analysis under the positive/negative ion mode, and the running time was 30 min. Result: Based on the standards of MS/MS and compared with the reference results, 32 compounds, containing coumarins, flavonoids and organic acids were identified. Conclusion: The efficient separation ability of UHPLC and high sensitive detection of MS are used in this study, which will provide the evidences for evaluating the quality of Violae Herba, stabilizing the curative effect in clinic, and explaining the mechanism.

      摘要:Objective: To establish the verified method of gas chromatography (GC) characteristic chromatogram of volatile components in Zhengtian pills,and provide a reference for the quality control of this preparation. Method: Volatile components in 16 batches of Zhengtian pills were extracted by steam distillation and analyzed by GC,taking relative retention time as index,GC characteristic chromatogram of volatile components in Zhengtian pills was established,difference of characteristic chromatogram between Zhengtian pills and 14 batches of intermediate was investigated. Result: Twenty common peaks were selected as characteristic peaks to assess similarities of different samples from different origins,and three of them were confirmed.Relative standard deviations (RSDs) of precision,reproducibility,stability were all less than 0.1%.RSDs of relative retention time of common peaks in intermediate were less than 0.1%,and threre was little difference between intermediate and Zhengtian pills. Conclusion: This method is accurate and reliable with good repeatability,which can be used for quality control of volatile components in Zhengtian pills.Preparation technology of Zhengtian pills is stable and its consistency between batches is good.

      摘要:Objective: To establish the quantification method of water-soluble polysaccharide and alkali-soluble polysaccharide in Poria by near infrared spectroscopy (NIR). Method: Optimization of the extract method about water-soluble polysaccharide and alkali-soluble polysaccharide was carried out. Meanwhile quantification method with phenol-concentrated sulfuric acid colorimetric reagent was established and the contents of water-soluble polysaccharide and alkali-soluble polysaccharide of ninety batches of samples were obtained. Near-infrared diffused reflected spectroscopy was implemented to obtain the NIR spectra of Poria. The NIR method was established by referring to the obtained contents with partial least squares regression (PLS) to pretreat the NIR spectra and with the program named synergy interval partial least squares regression (SIPLS) over Metlab R2012a platform to select sensitive wavelength number. And parameters was optimized by investigating the root-mean-square error of prediction (RMSEP) the correlation coefficient of calibration (Rc2), the root-mean-squares error of cross-validation (RMSECV) and the correlation coefficient of prediction (Rp2). Result: The RMSEC value of water-soluble polysaccharide is 0.270, the Rc2 value is 0.905 9, the RMSEP value is 0.119, the value of Rp2 is 0.913 8.Regarding to alkali-soluble polysaccharide, the RMSEC value is 0.242, the Rc2 value is 0.999 6, the RMSEP value is 0.243,and the value of Rp2 is 0.998 9. Conclusion: The established NIR method is convenience and accurate and satisfies the requirement of the rapid quality evaluation of Poria.

      摘要:Objective: To establish quality standards of Tongmai granules. Method: Ginseng Radix et Rhizoma Rubra, Astragali Radix, Notoginseng Radix et Rhizoma and Paeoniae Radix Rubra in Tongmai granules were identified by TLC; the contents of notoginsenoside R1, ginseng saponins Rg1, ginseng saponins Re and ginseng saponins Rb1 were determined by HPLC. Result: TLC spots were clear and specific in good repeatability. Linear ranges of notoginsenoside R1, ginseng saponins Rg1, ginseng saponins Re and ginseng saponins Rb1 were 0.058 8-0.588 8, 0.487 9-4.879, 0.104 9-1.049 and 0.308 3-3.083 μg,average recoveries were 95.34% (RSD 1.8%), 95.42% (RSD 2.0%), 98.99% (RSD 1.8%) and 102.13% (RSD 1.8%), respectively. Conclusion: This method is accurate, sensitive, simple and specific in good repeatability, which is suitable for quality control of Tongmai granules.

      摘要:Objective: An ultrafast liquid chromatography-hybrid triple quadrupole/linear ion trap mass spectrometry (UFLC-MS/MS) combined with statistical analysis method was used to investigate the changes of endogenous small molecule compounds and nephrotoxic mechanism in the rat serum induced by Radix et Rhizoma Tripterygii and to explore biomarkers associated with renal toxicity as well as metabolism pathways. Method: After intragastric administration for 14 days,serum biochemical indexes was detected by kit method and nephridial tissue section was observed.The serum samples were added acetonitrile for protein precipitation and analyzed by UFLC-MS/MS with full scan mode under positive ion scan mode,then principle component analysis (PCA) and principle component analysis discriminant analysis (PCA-DA) were used for data processing. Result: Compared with the blank group,serum biochemical indexes[creatinine (Cr) and blood urea nitrogen (BUN)] of Radix et Rhizoma Tripterygii ethanol extract group increased significantly and histopathological section damaged obviously which showed that the animal module prepared well.Nine significant different metabolites, such as dimethylglycine,lysine,phenylalanine,lysolecithin,phosphatidyl choline,phosphatidylserine and diglyceride, were identified as biomarkers of nephrotoxicity induced by Radix et Rhizoma Tripterygii. Conclusion: Nephrotoxic mechanism induced by Radix et Rhizoma Tripterygii may be related to the metabolism of amino acid and phospholipids,and it provides reference relating to the early clinical toxicity prevention of Radix et Rhizoma Tripterygii.

      摘要:Objective: The concents of aconitine,mesaconitine and hypaconitine in rat plasma were determined by UPLC-MS/MS after oral administration of Shiwuwei Rupeng pills,and pharmacokinetics of these three alkaloids in rats was investigated. Method: The separation was carried out on Welch UPLC XB-C18 column (2.0 mm×50 mm,1.7 μm),mobile phase was acetonitrile-0.1% formic acid in water for gradient elution,flow rate was 0.35 mL·min-1.Electrospray ionization (ESI) source was applied and operated in the positive ion mode. Result: The linear ranges of aconitine,mesaconitine and hypaconitine were 126.56-10 125,18.75-1 500,16.32-1 305.5 ng·L-1,respectively.The main pharmacokinetic parameters of aconitine,mesaconitine and hypaconitine were obtained as follows:Cmax of (0.75±0.14),(0.23±0.03),(0.17±0.02) μg·L-1,t1/2 of (2.93±1.72),(3.02±1.60),(3.70±2.63) h,AUC0-t of (5.03±1.37),(1.51±0.26),(1.14±0.21) h·μg·L-1. Conclusion: This method is rapid,accurate and sensitive,and it can be used for pharmacokinetic study of such alkaloids,which provides pharmacokinetic evidence for in vivo study of Shiwuwei Rupeng pills.

      摘要:Objective: To explore the effect of Xiaofu Tongjie recipe on VIP-cAMP-PKA-AQP3 signaling pathway in colon tissues of rats with functional constipation in rats. Method: Totally 150 SD male rats were randomly divided into normal group, model group, Xiaofu Tongjie recipe high dose, middle dose and low dose groups (380, 190, 95 g·kg-1), n=30 in each group. Compound diphenoxylate was applied by ig to induce functional constipation SD rat models, and then different doses of Xiaofu Tongjie recipe were given by ig administration for 4 weeks. The excretion time of first black defecating feces and moisture content in the feces were detected. The Real-time PCR and Western bolt were used respectively to detect the mRNA and protein expression levels of vasoactive intestinal peptide (VIP), cyclic adenosine monophosphate (cAMP), protein kinase A (PKA) and aquaporin 3 (AQP3) at different time points after treatment of 1, 2, 3 weeks. Result: As compared with the normal group, the excretion time of first black defecating feces was significantly prolonged in model group; moisture content in the feces was significantly reduced (P<0.01); the mRNA and protein expression levels of VIP, cAMP, PKA and AQP3 in colon tissues were significantly reduced (P<0.05, P<0.01). As compared with the model group, the excretion time of first black defecating feces was significantly shortened in various Xiaofu Tongjie recipe groups, and the moisture content in the feces was significantly increased (P<0.05, P<0.01); the mRNA and protein expression levels of VIP, cAMP, PKA and AQP3 in colon tissues were increased, and the increase was more obvious in high and middle dose groups; the changes of the indexes at week 2 and week 3 were more obvious than those of week 1 (P<0.05, P<0.01). Conclusion: Xiaofu Tongjie recipe can regulate the intestinal peristalsis and intestinal fluid metabolism and cure the functional constipation in a time and dose-dependent manner, which may be achieved by intervention on VIP-cAMP-PKA-AQP3 signaling pathway.

      摘要:Objective: To compare the effects of Yiqi Yangxue Tang containing Radix Astragali or Radix Hedyseri on the immune function of Senescence accelerated mouse P8(SAMP8) mice. Method: Ten Kunming mice were taken as normal group. 40 SAMP8 mice were randomly divided into normal saline group, thymosins group,Yiqi Yangxue Astragali Radix group and Yiqi Yangxue Hedyseri Radix group. The normal saline and thymosins were intragastrically administrated at 4.6 mg·kg-1 daily, and the mice in the Yiqi Yangxue Astragali Radix group and the Yiqi Yangxue Hedyseri Radix group received treatment at the dose of 6 g·kg-1 by ig, once a day for fifteen times. Eyeball was taken for blood 2 h after the last ig administration; then spleen was also taken. The contents of interleukins-2(IL-2) and γ-interferon (IFN-γ) in the serum were detected by ELISA; the expression levels of CD3, CD4, CD8, CD28 and CD152 on splenic lymphocyte surface were detected by using flow cytometry; mRNA and protein expression levels of p38MAPK were detected by Real-time PCR and Western blot respectively. Result: As compared with the normal group, the contents of IL-2 and IFN-γ were reduced; the expression of CD8 and CD28 molecules were decreased; CD4 molecule expression and CD28CD152 molecular double positive expression were increased; p38MAPK mRNA and protein expression levels were decreased in the normal saline group (P<0.05). As compared with the normal saline group, IL-2 and IFN-γ contents in the serum of SAMP8 mice were increased in thymosins group and Yiqi Yangxue groups; the contents in the thymosins group were higher than those in Yiqi Yangxue groups; the level of IL-2 in Hedyseri Radix group was higher than that in the Astragali Radix group; the expression level of CD28 was increased and the double positive expression level of CD28, CD152 was reduced in both thymosins group and Yiqi Yangxue groups; the double positive expression rate in the thymosins group and Astragali Radix group was lower than that in Hedyseri Radix group. Yiqi Yangxue groups increased the expression levels of CD8 molecules and the expression in Astragali Radix group was higher than that in Hedyseri Radix group. The expression level of CD4 was decreased in both thymosins group and Astragali Radix group. The p38MAPK mRNA expression level was increased in both thymosins group and Yiqi Yangxue groups, and the level in thymosins group was higher than that in Yiqi Yangxue groups; the level in Astragali Radix group was higher than that in Hedyseri Radix. p38MAPK protein content and protein positive rate were increased in both thymosins group and Yiqi Yangxue groups groups, and the effect in thymosins group was higher than that in Yiqi Yangxue groups (P<0.05). Conclusion: The Yiqi Yangxue Tang containing Astragali Radix or Hedyseri Radix could improve the immunosenescence performances of SAMP8 mice, which may be associated with up-regulating p38MAPK expression levels.

      摘要:Objective: To investigate the effects of Dingxin recipe (DXR) on phosphatase and tensin homologue deleted on chromosome 10 (PTEN) and the possible anti-atherosclerotic mechanism of DXR in ApoE knockout(ApoE-/-) mice. Method: Totally 50 ApoE-/-mice were randomly divided into model group,low-dose DXR group (9.25 g·kg-1·d-1), medium-dose DXR group (18.59 g·kg-1·d-1), high-dose DXR group (37.18 g·kg-1·d-1),and simvastatin group (0.005 g·kg-1·d-1), n=10 in each group. Another 10 C57/BL6J mice were selected as normal group. After feeding with high-fat diet for 16 weeks, all the mice were sacrificed and the atherosclerotic lesions in aortas were examined by HE staining. The levels of blood lipid and malondialdehyde (MDA), total superoxide dismutase (T-SOD), glutathione peroxidase (GSH-Px) and total antioxidant capacciyt (T-AOC) were detected. PTEN protein expression in the aorta was identified by Western blot. Result: As compared with the normal group,the levels of blood lipid and MDA were increased in model group,while the levels of T-SOD,GSH-Px,T-AOC were reduced, and the protein expression of PTEN was significantly reduced (P<0.05). As compared with the model group, blood lipid level was significantly reduced, while the levels of T-SOD,GSH-Px, and T-AOC were increased, and MDA level was reduced in medium and high-dose DXR groups (P<0.05); in addition, the medium and high-dose groups could up-regulate the expression level of PTEN in ApoE-/-mice (P<0.05). As compared with the model group, plaque size was significantly reduced in all DXR groups.(P<0.05). Conclusion: DXR can attenuate atherosclerosis in ApoE-/-miceby regulating blood lipids, suppressingoxidative stress,and up-regulating the protein expression of PTEN.

      摘要:Objective: To observe the effects of water extract from cortex of Berberis dictyophylla (BDCE) on expressions of retinal protein kinaseC-β(PKC-β), hypoxia inducing factor-1α(HIF-1α), vascular endothelial growth factor(VEGF) in spontaneous type 2 diabetic db/db mice, in order to explore its mechanism in preventing and treating diabetic retinopathy (DR). Method: Eighteen-week-old db/db mice were selected, with their thin db/m mice for normal control group. The db/db mices were randomly divided into 6 groups:model control (db/db) group, high, middle and low-dose BDCE group (1.50, 0.75, 0.38 g·kg-1), calcium-dobesilate group (0.23 g·kg-1), and berberine hydrochloride group (0.135 g·kg-1). The mice were given relevant drugs for consecutively two months. At the end of the two-month treatment, all the mice were put to death. The immunohistochemical method was used to observe the expressions of the three factors on the retina. Real-time quantitative PCR and western blot methods were used to detect genes and proteins expressions level of PKC-β, HIF-1α and VEGF in retina. Result: Compared to db/m group, the immunohistochemical results showed positive reaction of PKC-β and VEGF in retinal tissues of db/db group, with significant increase in expressions (P<0.01); positive reaction of HIF-1α, with no significant increase in expressions of db/db group; mRNA and protein expressions of PKC-β, HIF-1α, VEGF increased significantly in db/db group (P<0.01). Compared to db/db group, the immunohistochemical results showed positive reaction of PKC-β on retinal tissues in calcium-dobesilate group, with significant decrease of expression (P<0.05); positive reaction of PKC-β on retinal tissues in high-dosage BDCE group, with significant decrease of expression (P<0.01); a downward trend in PKC-β expression in other treatment groups compared with the db/db group, with no statistical significance in difference; positive reaction of HIF-1α on retinal tissue in all treatment groups, with no significant decrease of expression; positive reaction of VEGF on retinal tissues in berberine hydrochloride group and low-dose BDCE group, with significant decrease of expression (P<0.01); and positive reaction of VEGF on retinal tissue in all other treatment groups, with no significant decrease of expression. Compared with db/db group, real-time quantitative PCR results showed significant decreased mRNA expressions of HIF-1α in calcium-dobesilate group (P<0.05), decreased PKC-β, HIF-1α, VEGF mRNA expressions in other treatment groups, with no significant decrease. Compared with db/db group, Western blot results showed significant decreased protein expressions of PKC-β in calcium-dobesilate group, berberine hydrochloride group, and high and low-dose BDCE groups (P<0.01), and slightly decreased protein expressions of PKC-β in middle-dose BDCE group, with no significant decrease. Western blot results showed significant decreased protein expressions of HIF-1α in calcium-dobesilate group, berberine hydrochloride group, high and middle-dose BDCE groups (P<0.01), slightly decreased protein expressions of HIF-1α in BDCE low-dose group, with no significant decrease; Western blot results showed significant decreased protein expressions of HIF-1α in all treatment groups (P<0.01). Conclusion: BDCE was an efficient Tibetan medicine to prevent and treat DR, whose mechanism may be correlated with inhibition the expressions of PKC-β, HIF-1α, VEGF.

      摘要:Objective: To investigate the effect of Tongluo Tangtai power (TLTTP) on JNK signaling pathway in rats of diabetic peripheral neuropathy (DPN). Method: Totally 70 GK rats were selected to set up the rat model of spontaneous DPN induced by high fat diet. the 50 successfully modeled DPN rats were randomly divided into five groups, namely model group, TLTTP high-dose group, TLTTP group, TLTTP Low-dose group, and metformin+methycobal group, with 10 rats in each group; another 10 male GK rats were selected in normal group. All of the rats were detected for the expressions of p-JNK and Caspase-3 protein,JNK and Insulin-like growth factor 1(IGF-1) mRNA, sciatic nerve conduction velocity, and the levels of tumor necrosis factor-α(TNF-α),interleukins-6(IL-6),C-reactive protein(CRP),myelin basic protein(MBP) in serum after 4 weeks of intragastric administration. Result: Compared with the normal group, the model group showed significant increase in p-JNK and Caspase-3 proteins, JNK mRNA expression, and serum TNF-α, IL-6, CRP and MBP levels, and significant decrease in IGF-1 mRNA expression (P<0.05); TLTTP could down-regulate the expression of p-JNK,Caspase3,JNK mRNA, increase the expression of IGF-1 mRNA, significantly improve sciatic nerve conduction velocity, and significantly reduce the levels of TNF-α,IL-6,CRP,MBP in serum. Conclusion: TLTTP may prevent the development of DPN by inhibiting JNK signaling pathway, mitigating ell apoptosis, IR, inflammation reaction and abnormal expression of downstream target genes.

      摘要:Objective: To detect the molecular target of copper transport pathway in hepatocytes of TX mice with Gandou decoction (GDD) treatment, and explore its corresponding regulatory metabolism. Method: Totally 20 rabbits were randomly divided into normal group (normal saline) and GDD group (4.5 g·kg-1·d-1) after adaptive feeding for 3 days. Treatment was given by ig, bid, for 10 days to obtain drug serum. Hepatocytes of DL and TX mice were isolated and extracted by 2 step perfusion method; atomic absorption method was used to detect the concentration of microelement in the hepatocytes of TX mice after treatment with rabbit serum containing different concentrations of GDD; and another blank group was set up. Western blot was used to detect the expression levels of adenosine triphosphate7b (ATP7b), anti-oxidant1 (ATOX1), copper transporter1 (CTR1), metallothionein (MT), copper chaperone of superoxide dismutase (CCS), and cytochrome-coxidase17 (COX17) in the hepatocytes of TX mice after being treated with rabbit serum containing GDD. Result: As compared with blank group, the contents of copper and iron were increased; the content of zinc was reduced; the protein expression levels of ATP7b, ATOX1 CCS, COX17 were significantly reduced, and the protein expression levels of MT, CTR1 were significantly increased in hepatocytes of model group TX mice (P<0.01). As compared with the model group, GDD could reduce the contents of copper and iron in the hepatocytes of TX mice, and increase the content of zinc in a dose-dependent manner(P<0.05,P<0.01). Western blot results indicated that GDD could significantly increases the protein expression levels of ATP7b, ATOX1 CCS, COX17 (P<0.05, P<0.01)and decrease the protein expression levels of MT, CTR1 in the hepatocytes of TX mice (P<0.01). Conclusion: GDD may increases the copper excretion in hepatocytes of TX mice by up-regulating the protein expression levels of ATP7b, ATOX1, CCS, COX17 and down-regulating the expression levels of CTR1 and MT. GDD could reduce copper contents in hepatocytes through a multi-target and multi-channel regulation of copper metabolism pathway.

      摘要:Objective: To investigate the onset time and duration of action for the antidepressant effect of total saponins from Bupleuri Radix, and observe their effects on corticosterone damage-induced depression behavior and learning and memory impairment in mice. Method: Male ICR mice were randomly divided into normal group (distilled water),total saponins from Bupleuri Radix low, middle and high dose groups (12.5,25,50 mg·kg-1).They were weighed every week, and field experiment was conducted after drug administration for 13 days.Total saikosaponins was given intragastrically once a day for 14 consecutive days. The effects of total saponins from Bupleuri Radix on motionless time in forced-swimming test were observed after the last administration at different time (1 h, 3 d, 6 d). Male c57BL/6J mice were randomly divided into normal group, corticosterone (CORT) group, citalopram group (10 mg·kg-1) and total saponins from Bupleuri Radix group (25 mg·kg-1).Corticosterone (35 mg·L-1, equivalent to 5 mg·kg-1·d-1) was delivered for 5 weeks in drinking water in CORT group, citalopram group and total saponins from Bupleuri Radix group.3 weeks after modeling,total saponins from Bupleuri Radix were given by ig administration for 2 weeks until week 5; citalopram was also given by ig administration for 2 weeks. 1 day after drug administration, forced swimming test and water maze test were conducted. After the behavioral experiment, 3 mice were taken from each group. They were sacrificed then,and hippocampus was taken to detect the expression levels of brain derived neurotrophic factor (BDNF) and tyrosine kinase B (TrkB)by Western blot. Result: As compared with the normal group, total saponins from Bupleuri Radix had no effects on weight and moving distance and velocity in field experiment after 14 days administration, but could decrease the motionless time in forced swimming test at different time (1 h and 3 d after drug with drawal) (P<0.05, P<0.01). 6 days after drug withdrawal, total saponins from Bupleuri Radix 25 and 50 mg·kg-1 could still shorten the motionless time in forced swimming test (P<0.05). Two weeks treatment with total saponins from Bupleuri Radix at dose of 25 mg·kg decreased the motionless time in forced swimming test (P<0.01) and shorten the latency to reach the platform, and increase the number of platform crossing times in Morris Water Maze (P<0.05); in addition, they could increase the BDNF and Trk B levels in the hippocampus of mice with corticosterone damage (P<0.01). Conclusion: Total saponins from Bupleuri Radix could have a rapid and sustained antidepressant activity and reverse the depression-like behavior as well as learning and memory impairment induced by corticosterone damage. The mechanism may be associated withup-regulating BDNF and Trk B expression levels in the hippocampus of mice.

      摘要:Objective: To observe the effect of Qingzao Jiufei Tang(QJT) on tumor weight, tumor index, nuclear transcription factor kappa B (NF-κB), intercellular cell adhesion molecule-1 (ICAM-1), janus kinase 1(JAK1), epidermal growth factor receptor (EGFR) and signal transducers and activators of transcription 1 (STAT1) protein phosphorylation of Lewis lung cancer mice. Method: Totally 50 male C57BL/6J mice were randomly divided into model group, chemotherapy group[50 mg·kg-1·(2 d)-1], and high, medium and low-dose QJT groups (15.2, 7.6, 3.8 g·kg-1·d-1), with 10 in each group. The animal model was induced through the axillary injection with Lewis cells in the right arm of mice. QJT groups were intragastrically administered with medicines two weeks before modeling, chemotherapy group was intragastrically administered with cyclophosvnamide(CTX) at the corresponding dose, once every other day, and model group was intragastrically administered with the same volume of saline. At 2 weeks after modeling, the mice were put to death to weigh the tumor index, detect the expression of NF-κB by immunohistochemical method, the expression of ICAM-1 by Real-time PCR method, and JAK1, pEGFR and pSTAT1 by Western blot method. Result: Tumor weight and tumor index of high and medium-dose QJT groups and CTX group were significantly decreased, compared with the model group, with significant differences(P<0.01). CTX group, and high and medium-dose QJT groups showed significant decreases in the expression of NF-κB and EGFR protein expression (P<0.05, P<0.01), high and medium-dose QJT groups showed significant decrease in the expressions of ICAM-1 and pJAK1 (P<0.05, P<0.01), high, medium and low-dose QJT groups showed significant decrease in phosphorylation of STAT1, compared with the model group, with significant differences (P<0.01), high and medium-dose QJT groups were superior to CTX group (P<0.01). Conclusion: QJT can significantly inhibit the proliferation of Lewis lung cancer cells. It may inhibit lung cancer by reducing the protein expressions of NF-κB, ICAM-1, pJAK1, EGFR, phosphorylation of STAT1 protein.

      摘要:Objective: To investigate effects of Astragali Radix extracts on glucose absorption in Caco-2 cell and its relevant regulation mechanism. Method: All extracts of Astragali Radix were extracted by using systematical solvent separation method. Except for the blank group, Caco-2 cells were treated with Astragali Radix extracts, and then divided into four extraction groups (ethanol, petroleum ether, ethyl acetate, and n-butanol). The glucose absorption was detected by using the fluorescent D-glucose analog, with 2-deoxyglucose (2-NBDG) as the probe indicator. After the efficient extracts for promoting glucose absorption in Caco-2 cell was extracted from Astragali Radix, its effective concentration was studied. The expression of sodium-dependent glucose transporter 1 (SGLT1) and glucose transporter 2 (GLUT2) were detected by Western blot. Result: Astragali Radix n-butanol extract could improve glucose absorption in Caco-2 cell. Compared with control group, Astragali Radix n-butanol extract 100, 150 mg·L-1 group showed significant increase in glucose content in Caco-2 cells (P<0.05, P<0.01). N-butyl alcohol extracts of Astragali Radix could up-regulate GLUT2 and SGLT1 of Caco-2 cells (P<0.05, P<0.01). Conclusion: Astragali Radix n-butanol extract can promote the glucose absorption in Caco-2 cells. Its mechanism of improving glucose absorption may be correlated with regulation of SGLT1 and GLUT2.

      摘要:Objective: To observe the effect of Zhizhu granules on the rat model with chronic atrophic gastritis (CAG). Method: The rat model with chronic atrophic gastritis was established, and divided into model group, positive control group (Weifuchun,0.86 g·kg-1), and Zhizhu granules high, medium and low-dose(20,10,5 g·kg-1)groups. The mRNA and protein expressions of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) were detected by Western blot, immunohistochemical (IHC) and Real-time PCR method. Result: Compared with normal group, model group showed incomplete gastric mucosa gland cavity structure, obvious inflammatory cell infiltration, significant decreasing in weight growth rate, and significant increase in COX-2 and VEGF protein and mRNA expressions (P<0.01). Compared with model group, Zhizhu granules high, medium and low-dose groups and Weifuchun group showed significant increase in CAG rats' body weight growth rate (P<0.01), decrease in COX-2, VEGF protein and mRNA expressions in rat gastric tissues (P<0.01). Compared with Weifuchun group, Zhizhu granules high, medium and low-dose groups showed significant increase in CAG rats' body weight growth rate (P<0.01), high-dose group could decrease the expression of COX-2, VEGF protein and mRNA expressions in rat gastric tissues, with no significant difference. Conclusion: Zhizhu granules can increase the growth of the body weight of rats, and also alleviate pathological morphology in stomach tissues. Zhizhu granules show the preventive and therapeutic effects by down-regulating the expressions of COX-2 and VEGF.

      摘要:Objective: To observe the effect of Tanzhixiao recipe (TZXR) on signal transduction of ovarian insulin in rats with letrozole and high-fat diet induced insulin resistance in polycystic ovary syndrome (PCOS-IR). Method: Female SD rats were fed with letrozole by gavage and high-fat diet to induce PCOS-IR models. Then the rats were randomly divided into model group, TZXR group (35 g·kg-1·d-1) and the metformin group (200 mg·kg-1·d-1). The drugs were given by gavage administration for 20 days, and another normal blank group was established. After the end of the experiment, abdominal aortic blood was taken to determine fasting plasma glucose (FPG), and fasting insulin (FINS), and insulin resistance index was calculated by homeostasis model of assessment for insulin resistance index (HOMA-IR). The mRNA expression levels of insulin receptor (INSR), insulin receptor substrate-1 (IRS-1), phosphatidylinositol-3-kinase (PI3K), and protein kinase B (Akt) were detected by Real-time PCR. In addition, the expression levels of INSR, IRS, PI3K, Akt protein and their corresponding phosphorylated forms were detected by Western blot. Result: As compared with the blank group, the levels of FINS and HOMA-IR were increased in model group (P<0.01), but there was no significant increase in FPG level, mRNA expression levels of INSR, IRS, PI3K and Akt were significantly decreased (P<0.05), protein expression levels of INSR, p-INSR, IRS, p-IRS, PI3K, p-PI3K, Akt and p-AKT were significantly decreased (P<0.05), the phosphorylation ratios of INSR, IRS, PI3K, and Akt were significantly decreased in the model group (P<0.05). As compared with the model group, the levels of FINS and HOMA-IR were significantly decreased (P<0.01), mRNA expression levels of INSR, IRS, PI3K and Akt were significantly increased (P<0.05),protein expression levels of INSR, p-INSR, IRS, p-IRS, PI3K, p-PI3K, Akt and p-Akt were significantly increased (P<0.05), and phosphorylation ratios of INSR, IRS, PI3K, and Akt were significantly increased (P<0.05) in the TZXR group and metformin group. Conclusion: IR and abnormal signal transduction of PI3K/Akt pathway existed in PCOS model rats. TZXR could improve IR of PCOS rats, and the mechanism may be associated with regulating key protein expression levels of insulin signal trasduction pathway.

      摘要:Objective: To investigate the effects of serum containing Jianpi Yiqi decoction (JYD) on MHCC-97H cell apoptosis, and the changes in Vimentin and Caspase-3 in the process of apoptosis and the significance. Method: By using the serologic pharmacological method, Vimentin breaker and Caspase inhibitor were adopted to observe the effect of serum containing Jianpi Yiqi decoction at concentrations of 7.5%, 15% and 30% on proliferation, invasion and apoptosis of liver tumor cells, and protein expressions of Vimentin, Caspase-3 and poly (ADP-ribose) polymerase-1 (PARP-1). Result: Compared with the blank serum group, decreased protein expression of Vimentin and PARP-1(P<0.01), and increased protein expression of Caspase-3 were observed in Jianpi Yiqi decoction groups (with concentrations of 15% and 30%) and Vimentin-breaker group. The down-regulation degree of Vimentin and PARP-1 were consistent with the proliferation inhibition rate, invasion inhibition rate and apoptosis rate of MHCC-97H cells. The proliferation and invasion inhibition rates and apoptosis rate of the Vimentin-breaker group cells were the highest among all experimental groups. However, the Caspase-inhibitor group showed the lowest proliferation and invasion inhibition rates, and apoptosis rate among all experimental groups. Compared with the blank serum group, medium and high-dose JYD group and Caspase-inhibitor group showed up-regulation of Caspase-3 protein expression (P<0.01), down-regulation in Vimentin and PARP-1 expressions (P<0.01), and fragments in PARP-1 protein, the Caspase-inhibitor group showed down-regulation in Caspase-3 expression (P<0.01), with no statistically significant difference in Vimentin and PARP-1 protein expressions. Conclusion: The expression of Vimentin in MHCC-97H cells was down-regulated by serum containing Jianpi Yiqi decoction, especially at the concentration of 15%. Jianpi Yiqi decoction may inhibit liver cancer cell proliferation and invasive by down-regulating Vimentin via Caspase-3, while inducing liver cancer cell apoptosis by using positive feedback apoptosis-promoting apoptosis signal formed by Caspase-3/Vimentin.

      摘要:Objective: To discuss the effect of Compound Cortex Phellodendri fluid on inflammatory factors in promoting the healing process of diabetic foot ulcers, and observe its effect in promoting wound tissue healing. Method: Totally 120 patients with diabetic foot ulcers of toxic heat syndrome diagnosed in Dongzhimen Hospital between May 2014 and June 2015 were selected and randomly divided into treatment and control groups (with 60 cases in each group). Treatment group was locally administered with Compound Cortex Phellodendri fluid, while control group was locally administered with Kangfuxin solution, with once dressing change per day. At 0 d before administration and 7, 14, 21 and 28 d after administration, venous blood was collected to observe dynamic changes in high sensitive C reactive protections (hs-CRP), tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6) and advanced glycation end-products (AGEs). Four weeks after treatment, improved clinical symptoms and dynamic changes in inflammatory factors of AGEs of both groups were analyzed statistically. Result: Treatment group showed the total efficiency of 81.3%, while control group showed the total efficiency of 68.8%, indicating total treatment group was superior to control group. At early stage of inflammatory reaction, both groups showed significant increases in hs-CRP, TNF-α and IL-6 (P<0.05), at 7, 14, 21 and 28 d after administration, treatment group and control group showed significant decreases in hs-CRP, TNF-α and IL-6(P<0.05), with a better efficacy in treatment group than control group (P<0.05); compared with control group, at 7, 14, 21 and 28 d after administration, treatment group and control group showed a declining trend in AGEs, with the lower AGEs in treatment group than control group (P<0.05). According to the dynamic analysis, AGEs were positively related to inflammatory factors, treatment group was superior to control group in principal syndrome integral and secondary syndrome integral (P<0.05). Conclusion: Compound Cortex Phellodendri fluid has a therapeutic effect in treating diabetic foot ulcer with toxic heat syndrome, and is worth clinical promotion.

      摘要:Objective: To discuss the clinical effects of Zhuweigao combined with massage on anorexia in children, and observed weight, body mass index (BMI), serum trace elements, calcium, iron, zinc, hemoglobin, Ghrelin/Leptin, Y/Neuropeptide Y (NPY), traditional Chinese medicine clinical syndrome integral change and clinical efficacy. Method: Totally 94 patients aged between one and seven years old were selected and randomly divided into two groups. The control group was given Jianwei Xiaoshi oral liquid according to age, specifically 5 mL/time for children aged below one year old, bid, 10 mL/time for children aged above five years old, tid, and 10 mL/time for the remaining children, bid. The treatment group was treated by Zhuweigao combined with massage, one pill/time, tid, for two weeks for a course. Before and after treatment, weight, body mass index (BMI), serum trace elements, calcium, iron, zinc, hemoglobin, Ghrelin/Leptin, traditional Chinese medicine clinical syndrome integral change and clinical efficacy of the two groups were observed. Result: The total effective rate of treatment group was 87%, which was higher than 70% in control group, with significant difference (P<0.05). Zhuweigao combined with massage can effectively improve weight, BMI, serum trace elements, hemoglobin, Ghrelin/Leptin and NPY, with a better clinical efficacy (P<0.05). Conclusion: Zhuweigao combined with massage can effectively improve appetite, with a better efficacy than routine Chinese patent medicines.

      摘要:Objective: To evaluate the clinical efficacy of Gongliuning capsules combined with mifepristone in treating hysteromyoma (stagnation of Qi and blood) and their effect on vascularendothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9), transforming growth factor-β1 (TGF-β1) and epidermal growth factor (EGF). Method: One hundred and forty-six patients were divided into control group (73 cases) and combination group (73 cases) by random number table. Patients in control group received mifepristone tablets in the morning, menstruation begins first days,25 mg/day. In addition to the therapy of control group, patients in combination group were also given Gongliuning capsule, 6 grains/time, tid. The courses of treatment were 3 months. Before and after treatment, sizes of fibroid and uterus were detected by color doppler ultrasound. Scores of stagnation of Qi and blood were evaluated. Levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2), progesterone (P), VEGF, MMP-9, TGF-β1 and EGF were detected. Recurrence was recorded during the 6-month follow-up visit. Result: The total clinical therapy in observation group was 92.31%, which was superior to 78.13% in control group (P<0.05). After treatment, sizes of fibroid and uterus in combination group were smaller than those in control group, and scores of stagnation of Qi and blood were lower than those in control group (P<0.01). Levels of E2, P, FSH, LH, VEGF, MMP-9, TGF-β1 and EGF were lower than those in control group (P<0.01). And recurrence rate in combination group was 24.62%, which was lower than 48.44% in control group (P<0.01). Conclusion: The effect of Gongliuning capsules combined with mifepristone was better than mifepristone in treating hysteromyoma (stagnation of Qi and blood), ameliorating symptoms, improving clinical effect, and reducing recurrence rate, and the mechanism of action may be correlated with decrease in levels of sex hormone, and regulation of the expressions of VEGF, MMP-9, TGF-β1 and EGF factors.

      摘要:Objective: To discuss the efficacy of Bailing capsules combined with Bufei Huoxue capsules in treating chronic obstructive pulmonary diseases at stationary phase, and their effect on immunologic function, interleukin-10 (IL-10) and interleukin-17 (IL-17). Method: One hundred and sixteen-eight patients were divided into control group (83 cases) and observation group (85 cases) by random number table. Patients in control group got salmeterol and fluticasone propionate inhalation 25 μg/time, bid, as well as nourishment, oxygen therapy and guidance of health education for disease. In addition to the therapy of control group, patients in observation group was also given Bailing capsules, 3 gains/time, tid, and Bufei Huoxue capsules, 4 gains/time, tid. A follow-up visit was made once every 2 weeks, and the courses of treatment were 24 weeks. Frequency of acute exacerbation COPD and duration were recorded within 24 weeks. And frequency and duration of cold attacks were recorded within 24 weeks. Symptoms, signs, syndrome of qi deficiency of lung and kidney, improved version of the British Medical Research Council respiratory questionnaire (mMRC), self-assessment test questionnaire for patients with chronic obstructive pulmonary disease and St George's respiratory questionnaire(SGRQ)were scored. Before and after treatment, lung function was detected. Forced vital capacity (FVC), forced expiratory volume for 1 second (FEV1) and FEV1% were recorded. Levels of IgG, IgM, IgA, alexin C3 and C4 and IL-17 and IL-10 were detected. Result: Through analysis on Ridit, the total rate of TCM symptoms in observation group was superior to that in control group (P<0.05). Scores of cough, sputum expectoration, puffing, chest distress, short breathe, weakness, cyanosis, syndrome of qi deficiency of lung and kidney, mMRC and CAT in observation group were lower than those in control group (P<0.01). After treatment, FVC, FEV1 and FEV1% in observation group were higher than those in control group (P<0.05). SGRQ scale's three scores scale and total score in observation group were lower than those in control group (P<0.01). Level of IL-17 in observation group was lower than that in control group, and level of IL-10, IgG, IgM and IgA in observation group were higher than that in control group (P<0.01). And there was no statistical significance in the differences of alexin C3 and C4 between the two groups. During the 24-week observation, both of frequency and duration of acute exacerbation COPD and cold in control group were lower than those in control group (P<0.01). Conclusion: Bailing capsules combined with Bufei Huoxue capsules can control symptoms, ameliorate lung function, improve quality of life and immune function of organism, relieve inflammatory reaction, stabilize condition and promote recovery of lung for patients of chronic obstructive pulmonary diseases at stationary phase with syndrome of qi deficiency of lung and kidney.

      摘要:Objective: To evaluate the anti-platelet aggregation activities of the Chinese patent medicines (CPM) for promoting blood circulation and removing blood stasis, explore the medication rules, and provide certain experimental basis for the development of anti platelet drugs. Method: High-throughput microwell turbidimetric method was used to detect drug response to platelet aggregation induced by adenosine diphosphate (ADP) and thrombin. Result: The 41 kinds of tested Chinese patent medicines (CPM) showed antagonistic effect in different degrees (inhibitory rate of 30%~98%) on platelet aggregation induced by ADP or thrombin. 18 kinds of CPM among them showed selectivity for the inducers. Formulation analysis revealed that highest anti-platelet aggregation activities were demonstrated by the CPM containing ingredients of Salviae Miltiorrhizae Radix et Rhizoma, Chuanxiong Rhizoma, Carthami Flos, Notoginseng Radix et Rhizoma, and Borneolum Syntheticum. Furthermore, the CPMs with promoting blood circulation character were more frequently combined with those of multi-tonic medicine, resuscitation medicine and hemostatic medicine to achieve more potent inhibitory effects on platelet aggregation. Conclusion: The compound CPMs for promoting blood circulation and removing blood stasis can achieve anti-platelet aggregation activity by anti-ADP and antithrombin means. The medicines with ‘Qi-regulating’,‘Qi-tonifying’ and ‘resuscitation’ efficacy may strengthen the activity of anti-platelet aggregation.

      摘要:Objective: To analyze the medication experience of professor ZHONG Meng-liang's prescriptions for treating arthromyodynia by using Traditional Chinese Medicine (TCM) Inheritance Support System (V2.5). Method: Professor ZHONG's prescriptions for arthromyodynia cases were collected, then the data were logged into the TCM inheritance support system by using data mining methods such as Apriori algorithm of association rules, complex system entropy cluster and unsupervised hierarchical cluster, then they were combined with professor ZHONG's personal proofreading to analyze the using frequency of herbs, compatibility and composition rules. Result: Based on the analysis of 138 prescriptions, there were 20 kinds of most commonly used herbs such as Astragali Radix, Aconiti Lateralis Radix Praeparaia and Chaenomelis Fructus. 22 core combinations and 11 new prescriptions were obtained. Conclusion: Professor ZHONG had a wealth of experience in the treatment of arthromyodynia. Professor ZHONG, and was good at using the drugs with supplementing Qi and warming Yang, expelling wind and removing damness, activating meridians and relieving pain to treat arthralgia syndrome.

      摘要:Objective: To analyse and evaluate the patterns and features of Lingnan medicine,in the areas of formation of treatment formulae for diseases caused by dampness. Method: Traditional Chinese medicine (TCM) inheritance system was adopted to organize the works of Lingnan's renowned physicians and establish a database. Result: A total of 161 Chinese medicine prescriptions were screened,a total of 211 kinds of drugs,arranged in descending order according to the frequency of drug used,a total of 20 kinds of core combination of drugs extracted from the new prescription.A total of 10 new prescriptions that has been evolved. Conclusion: According to the multiple perspectives analysis by TCM inheritance system,it is suggested that the treatment of diseases caused by dampness in Lingnan should be based on removing dampness through different methods involving drying,warming,regulating the spleen-Qi and adjusting Qixue as the main principles.
