      摘要:Objective: To establish high performance liquid chromatography (HPLC) fingerprints of aqueous extract of unripe Forsythiae Fructus and its different polar parts,to clarify the contribution of fingerprints peaks in different parts of unripe Forsythiae Fructus to inhibit inflammation,and to reveal the pharmacodynamic material basis. Method: HPLC was introduced to analyze fingerprint of different parts of unripe Forsythiae Fructus with the mobile phase consisted of acetonitrile and water including 0.1% formic acid for gradient elution.The anti-inflammatory activity of different polar parts of unripe Forsythiae Fructus was evaluated via the variety of tumor necrosis factor-α (TNF-α),interleukin-6 (IL-6) and IL-1β concentrations of the lipopolysaccharide-induced acute lung injury model in rats.Grey relational analysis and bivariate correlation analysis were used to establish the spectrum-effect relationship. Result: Seven components with greater contribution rate of effect were obtained by grey relational analysis,the different peaks ranked as No.23,25,31,29,24,26 and 30.Bivariate correlation analysis showed that there was a significant negative correlation between the No.23 peak content and TNF-α,between the No.18 and 13 peaks contents with IL-6,between No.22 peak content with IL-1β,between the No.7,13 and 6 peaks contents with IL-1β.That was to say,the higher contents of chemical components of these peaks,the lower of TNF-α,IL-6 and IL-1β,and the better was the anti-inflammatory effect. Conclusion: According to the results of two kinds of analysis,it is concluded that No.23,13,25,31 and other peaks have the greatest contribution to the anti-inflammatory effect of unripe Forsythiae Fructus.The established HPLC fingerprint method is simple and reproducible.The anti-inflammatory substances of unripe Forsythiae Fructus mainly concentrate in ethyl acetate part,which is the middle polar composition.Moreover,the anti-inflammatory effect is the combined action of these components.

      摘要:Objective: To discuss effect of compatibility of Aristolochia manshuriensis with processed Aconiti Lateralis Radix Praeparata and Zingiberis Rhizoma on the content of aristolochic acid A,diuretic action of rats,acute renal injury of rats and in vitro renal cytotoxicity. Method: Different proportion of processed Aconiti Lateralis Radix Praeparata and Zingiberis Rhizoma compatibility with A. manshuriensis was chosen according the theory of traditional Chinese medicine (TCM).The content of aristolochic acid A after compatibility was determined by HPLC.The change of diuretic effect after compatibility was compared by rat water-load test,the change of rat acute renal injury after compatibility was compared,the change of human renal tububar epithelial cell toxicity after compatibility was compared inverted microscope observation and CCK-8. Result: After compatibility of processed Aconiti Lateralis Radix Praeparata-Zingiberis Rhizoma-A. manshuriensis (3: 2: 1),the content of aristolochic acid A depressed markedly with reduction percentage of 19.18%,diuretic effect of A. manshuriensis on rats had no change,its acute renal injury in rats was significantly lower than the crude product,its cell activity was better than the crude product. Conclusion: Compatibility can reduce the toxicity and save the effect of A. manshuriensis,it has a certain guiding significance to solve current clinical applications of TCM containing aristolochic acid A.

      摘要:Objective: Clinical curative effect is closely related to the specifications and quality of Chinese medicine decoction pieces.Chemical fingerprint methods and DNA molecular identification technology were used to evaluate the quality of precise powder decoction pieces (PPDP) of Notoginseng Radix et Rhizoma by comparing with the traditional commercial slices. Method: Different specifications of PPDP of Notoginseng Radix et Rhizoma were prepared,their dry extract yields were in contrast with those of commercial slices;three batches of commercial slices were collected,the content uniformity,fingerprint and similarity evaluation before and after the mixing and pulverization were studied by HPLC-DAD and DNA sequence alignment. Result: Dry extract yield of PPDP of Notoginseng Radix et Rhizoma were remarkablely higher than that of the traditional commercial slices.Dissolution rates of notoginsenoside R1,ginsenoside Rg1 and Rb1 in PPDP were higher than those of traditional commercial slices.RSDs of inter-assay dissolution of the three components of commercial slices were 13.1%,32.7% and 24.5%,respectively,which could be reduced to 3.6%,2.6% and 3.5% after mixing and preparing into PPDP.After comminution,the similarity of fingerprints was improved,14 peaks were identified and their peak areas were improved. Conclusion: PPDP and traditional commercial slices of Notoginseng Radix et Rhizoma share consistent properties,while PPDP can apparently improve dry extract yield,dissolution rate and quality uniformity.

      摘要:Objective: To investigate the rationality and superiority of precise powder decoction pieces system,Salviae Miltiorrhizae Radix et Rhizoma was employed in this study. Method: The second internal transcribed spacer (ITS2) sequence was used as a DNA barcode to identify Salviae Miltiorrhizae Radix et Rhizoma.The chemical composition of the medicinal materials was characterized by HPLC fingerprints,similarity between precise powder decoction pieces and traditional commercial slices were both evaluated.The concents of salvianolic acid B and rosmarinic acid were determined by HPLC,and the characteristic common peaks were identified.In addition,the extraction efficiency was also compared by standard decoction method. Result: Salviae Miltiorrhizae Radix et Rhizoma could be accurately identified by ITS2 sequence.HPLC fingerprints were very similarity between precise powder decoction pieces and traditional commercial slices.However,the extraction efficiency of precise powder decoction pieces was significantly improved with >30% increased dry extract yield and approximately 20%-120% increased dissolution of chemical components.Moreover,the relative standard deviation was also decreased between batches. Conclusion: Precise powder decoction pieces can improve the extraction efficiency and uniformity of traditional commercial slices.

      摘要:Objective: To establish the commodity specification and grade standard of Citri Grandis Exocarpium for guiding the market liquidity of this herb to standardize and scientize,and offering the reference for its quality control and identification. Method: The 35 batches of Citri Grandis Exocarpium samples were measured in quantitative characteristics of appearance and the content of index components,such as naringin,rhoifolin and total flavonoids.The data were evaluated by correlation analysis,principal component analysis and cluster analysis.So that the commodity specification and grade standard of Citri Grandis Exocarpium was formulated. Result: Diameter,weight and the density of villi were chosen as three indexes of appearance characteristics,and the content of naringin,rhoifolin and total flavonoids as the internal indexes.The grade of Citri Grandis Exocarpium was divided into four ranks,such as the first-grade,second-grade,third-grade and four-grade. Conclusion: According to comprehensive analysis with the appearance characteristics,such as diameter,weight and the density of villi,and internal indexes of Citri Grandis Exocarpium,combinding with results of market survey,these established standards are basically the same as the traditional classification.

      摘要:Objective: To discuss the effect of thermal moxibustion emplastrum with Jianchangbang Cyperi Rhizoma on hemorheology of dysmenorrhea rats caused by oxytocin and to investigate its in vitro transdermal absorption properties. Method: SD rats were randomly divided into 4 groups,such as the blank group,model group,thermal moxibustion emplastrum with Jianchangbang Cyperi Rhizoma group and petroleum ether extract of Jianchangbang Cyperi Rhizoma group.The dysmenorrhea rats model were made by oxytocin and diethylstilbestrol,then were observed the times and incidence of wrings within 30 minutes.The blood viscosity and plasma viscosity of rats in all groups was tested by automatic blood rheometer.The percutaneous permeation of α-cyperone and cyperotundone in the thermal moxibustion emplastrum with Jianchangbang Cyperi Rhizoma was studied by in vitro transdermal delivery experiment with Franz diffusion cells and their contents were determined by HPLC. Result: Thermal moxibustion emplastrum group and petroleum ether extract group could significantly reduce the times and incidence of wrings within 30 minutes,and the thermal moxibustion emplastrum group was better than the petroleum ether-extract group.Compared with the blank group,the model group significantly increased the whole blood viscosity and plasma viscosity,but the thermal moxibustion emplastrum group and the petroleum ether-extract group could significantly reduce them of model rats,also the thermal moxibustion emplastrum was better.The permeation rates of α-cyperone and cyperotundone from the thermal moxibustion emplastrum with Jianchangbang Cyperi Rhizoma were 45.42,58.58 μg·cm-2·h-1,and their transdermal behavior conformed to Higuchi equation. Conclusion: Thermal moxibustion emplastrum with Jianchangbang Cyperi Rhizoma can significantly improve the drug release properties of active ingredients with auxiliary heating materials,it can effectively relieve oxytocin-induced pain in rats and improve hemorheology of dysmenorrhea rats.

      摘要:Objective: To obtain the key enzyme genes involved in cardiac glycosides biosynthesis pathway, acetyl-CoA C-acetyltransferase (AACT) gene, from leaves of Lepidium apetalum seedlings, and conduct sequence analysis and prokaryotic expression analysis. Method: Based on the transcriptome data of L. apetalum, specific primers were designed to obtain cDNA sequence of LaAACT gene, construct prokaryotic expression vector pET-32a-LaAACT and induce the expression of recombinant LaAACT protein. Result: LaAACT gene had ORF full length of 1 212 bp, which encoded a protein of 403 amino acid residues. Sequence analysis indicated that LaAACT protein located in cytoplasm had no transmembrane domain and signal peptide, and the conserved domains of thiolase super family were detected. Phylogenetic analysis indicated that LaAACT protein had the closest relationship with AACT protein from cruciferous plant. The recombinant LaAACT protein was successfully expressed in E. coli BL21 (DE3) cells and finally, the recombinant LaAACT protein was purified through Ni2+ affinity chromatography. Conclusion: LaAACT gene was isolated from leaves of L. apetalum seedlings for the first time, and the stable prokaryotic expression system of pET-32a-LaAACT was constructed. This study could provide fundamental information for antibody preparation of LaAACT protein and provide basis for functional researches of LaAACT gene in cardiac glycosides biosynthesis pathway.

      摘要:Objective: To compare the differences in ITS sequence of Salviae Miltiorrhizae Radix et Rhizoma from different origins, analyze the relationship between ITS sequence, effective composition content and origins by detecting its effective composition content,and lay a foundation for exploring the quality differences mechanism of Salviae Miltiorrhizae Radix et Rhizoma from different regions. Method: DNA was extracted from the Salviae Miltiorrhizae Radix et Rhizoma from different origins, and PCR-sequencing technology was adopted, then the sequence analysis software was used to analyze the gene sequence differences between different origins. UPLC method was used to measure the effective composition content in Salviae Miltiorrhizae Radix et Rhizoma from different origins. Result: It was found that there were some differences in ITS sequences among different origins, then the ITS sequences after being induced by the elicitor showed two types of obvious mutations (A and G); in addition, the effective composition content also showed obvious differences between different origins. Conclusion: It was concluded that the mutations at 152 bp of Salvia Miltiorrhizae Radix et Rhizoma induced by different elicitors could lead to more ‘G’-shaped transformation. The water-soluble constituents of Salvia Miltiorrhizae Radix et Rhizoma were increased after being processed by chitosan and salicylic acid respectively. No significant effect on the increase of the active ingredients was found after combined induction by chitosan and salicylic acid.

      摘要:Objective: Accuracy of NIR model is restricted by complex matrix and overlapping bands of traditional Chinese medicine (TCM). To solve the above problem, the application of competitive adaptive reweighted sampling (CARS) in quantitative analysis of Chinese medicinal materials, TCM extract and patent medicines was discussed in this paper. Method: Near-infrared diffuse reflectance spectra of Puerariae Lobatae Radix, Puerarin Lobata Radix extract and Yufeng Ningxin dripping pills were collected to determine the contents of puerarin. Then the spectra were pre-processed with optimized methods to exclude uncommon samples, and CARS method was used to screen relevant variables and establish partial least squares (PLS) correction models. Result: Root mean square error of cross-validation (RMSECV) was 0.35%, 1.76% and 0.54%, respectively in models for raw material, semi-product and finished product. As compared with the models based on the full spectra, increased accuracy was obtained from the CARS-PLS models. Conclusion: CARS could increase prediction accuracy of the models and simplify calibration, which would offer a new approach for rapid and non-destructive determination of TCM.

      摘要:Objective: By measuring dissolution contents of three kinds of toxic components (triptolide,tripterine, and wilforlide) in simplified Qingluo Tongbi decoction, Tripterygium wilfordii(TW), TW +Notoginseng Radix et Rhizoma, as well as TW+Rehmanniae Radix, explore ‘Diversity Restriction’ mechanism of detoxicity from the perspective of the chemical basis composition, and provide experimental basis for clinical compatibility application of TW for detoxicity effect. Method: Highly selective and sensitive triple quadrupole mass spectrometry (LC-MS/MS) were used at the same time to detect three active ingredients-triptolide, tripterine and wilforlide, and then their dissolution changes were compared between TW, TW+ Notoginseng Radix et Rhizoma, TW+Rehmanniae Radix, as well as Simplified Qingluo Tongbi decoction. Result: In TW medicinal water extract, triptolide had the highest content, followed by wilforlide, and the content of tripterine was lowest. When the amount of medicine TW was same, the dissolution trend of the above three active ingredients was consistent under different compatibility environments, and the dissolution order was as follows: TW> TW+Rehmanniae Radix> TW+Notoginseng Radix et Rhizoma >Simplified Qingluo Tongbi decoction. Conclusion: In the compatibility, Notoginseng Radix et Rhizoma had a significant inhibitory effect on dissolution of three active ingredients of TW, while Rehmanniae Radix had a weak inhibitory effect. In addition, the dissolution of three active ingredients was significantly reduced in Simplified Qingluo Tongbi decoction. The results provided an experimental basis for clinical ‘Diversity Restriction’ detoxicity mechanism of TW.

      摘要:Objective: To analyze chemical ingredients of oils from Periplaneta australasiae and P. americana, in order to provide experimental basis for reasonable utilization of animal medicines of blattaria. Method: Ethanol reflux extraction method was used to extract medicinal materials of blattaria, and petroleum ether was used to extract fat-soluble components. Its oil components were analyzed by GC-MS technique and identified by NIST11.L standard mass spectra library. HP-5MS fused-silica capillary column was adopted, the carrier gas was high-purity helium, electron impact ionization source was at 230 ℃, and quality scanning range was between m/z 12-550.Their relative contents were calculated by area normalization. Result: Thirty-seven compounds were identified in the oil of P. australasiae, accounting for 90.99% of the total weight. The compounds with higher content level included linoleic acid (29.86%), oleic acid (26.30%), hexacosane (10.57%), stearic acid (3.94%), and heptadecane (3.78%). Besides, a small amount of arachidonic acid was found in the oil of P. australasiae. Thirty-five compounds were identified in the oil of P. americana, accounting for 94.14% of the total weight. The compounds with higher content level included oleic acid (44.09%), palmitic acid (16.03%), stearic acid (6.32%), and 3-eicosyne (5.31%). Conclusion: Esters, fatty acids and hydrocarbon compounds were the main compounds in the oils of P. australasiae and P. americana, but there were varying degrees of differences in the varieties and relative content. Totally 21 common constituents were identified in the oils from the two kinds of cockroaches, but there were great differences in the relative content.

      摘要:Objective: To determine and compare the contents of congmunoside TTP and congmunoside V in Aralia elata leaves from different habitats and gathering periods. Method: HPLC-ELSD was carried out on a Diamonsil C18 (2) column (4.6 mm×250 mm, 5 μm) at a column temperature of 30 ℃. The mobile phase was acetonitrile and water (31: 69) at flow rate of 1.0 mL·min-1. The detector was ELSD, with the temperature of drift tube set at 110 ℃, and the flow rate of carrier gas (N2) was 3.2 mL·min-1. Result: The linear ranges of congmunoside TTP and congmunoside V were 0.51-2.55 μg (r=0.999 8) and 0.50-4.00 μg (r=0.999 2), respectively. According to the results, the contents of congmunoside TTP and congmunoside V in A. elata leaves from different habitats and gathering periods were different. This may be related to distribution areas, growing seasons and physiological state of leaves. On the basis of the experimental results, the content of two kinds of ingredients was high mostly in July and August, and partially in September. Conclusion: The method is simple, rapid and accurate for determining simultaneously the contents of congmunoside TTP and congmunoside V in A. elata leaves, and can provide a basis for the quality control of A. elata leaves.

      摘要:Objective: To establish a UPLC method for the simultaneous determination of aesculin, salidroside, aesculetin, syringin, fraxin, fraxetin and scopoletin in the Fraxini Cortex formula granule, in order to compare the difference of the seven chemical constituents in various Fraxini Cortex formula granule samples and provide an effective method to ensure the scientific quality of Fraxini Cortex formula granule. Method: The separation was performed on a Waters ACQUITY UPLC BEH C18 column (2.1 mm×50 mm, 1.7 μm), and eluted with mobile phase consisting of methanol-0.1% phosphoric acid solution in a gradient mode (0-1 min, 5%-10%A; 1-10 min, 10%-12%A) at a flow rate of 0.4 mL·min-1. The sample size was 2 μL. The detection wavelength was 220 nm, and the column temperature was 38 ℃. Result: Peak area and concentration for aesculin, salidroside, aesculetin, syringing, fraxin, fraxetin and scopoletin showed a good linearity within their ranges, the average recoveries of the chemical constituents were 100.0% (RSD 2.0%), 99.5% (RSD 2.1%), 99.9% (RSD 1.5%), 99.7% (RSD 2.2%), 99.4% (RSD 1.4%), 100.8% (RSD 1.8%), 99.4% (RSD 2.6%), respectively. Conclusion: The established method is simple and accurate, and can be used to simultaneously determine seven chemical constituents in Fraxini Cortex formula granule, and the analysis time was significantly shortened. It's the first time to make a quantitative determination for many ingredients except for aesculin and the aesculetin in the Fraxini Cortex formula granule. There are certain differences in the contents of chemical compositions in the Fraxini Cortex formula granule of various batches and manufacturers.

      摘要:Objective: To establish a UPLC method for the simultaneous determination of eight compounds in Forsythiae Fructus from different habitats. Method: Eight compounds in Forsythiae Fructus were simultaneously determined by UPLC with BEH C18 column (2.1 mm×50 mm, 1.7 μm), with methanol (A)-water (B) (consisting of 0.1%formic acid) as the mobile phase for gradient elution (0-2 min, 10%-20%A; 2-3 min, 20%-30%A; 3-6 min, 30%-60%A; 6-6.01 min, 60%-10%A; 6.01-9 min, 10%A); at the flow rate of 0.4 mL·min-1; the determination wavelength was 235 nm; and the injection volume was 2 μL. Result: Eight components were separated clearly within 6 min. The RSD values of precision, stability and reproducibility were all less than 3.0%. The average recoveries were 98.8%-101.4%with RSD of 1.0%-1.9%. Conclusion: The method is simple, rapid, accurate and highly sensitive, providing a scientific basis for quality assessment of Forsythiae Fructus. PCA results showed that different production place, harvesting seasons and processing method played an important role in quality of Forsythiae Fructus.

      摘要:Objective: To examine the content of linderane and norisoboldine in different parts of Linderae Radix, and make a discriminant analysis by fourier transformation infrared spectroscopy. Method: Linderae Radix was divided into three groups, namely skin group, peeling group and original group. According to the chromatographic conditions for linderane and norisoboldine of Linderae Radix in Chinese Pharmacopoeia, HPLC was used to examine the content of linderane and norisoboldine in different parts of Linderae Radix. Near-infrared spectra of skin group, peeling group and original group samples were analyzed through principal component analysis. The principal components analysis model was developed based on the spectrum band of 5 053.85-4 115.54 cm-1 with ‘SNV+spectrum+F-G’. Result: Three samples from the three parts of Linderae Radix were divided into three categories by the model of FT-NIR qualitative discriminant analysis, and there was a clear boundary between the different parts. According to the findings of HPLC analysis, the sequence of the content of linderane and norisoboldine in different parts from high to low was skin group > original group > peeling group. Conclusion: The results of HPLC analysis showed that the content of linderane and norisoboldine in Linderae Radix skin group was higher, and that of the peeling group was the lowest. FT-NIR analysis further proved the differences in the quality of different part Linderae Radix. The two analysis methods provide the scientific basis for weather Linderae Radix shall be peeled or not during processing.

      摘要:Objective: To establish a HPLC method to simultaneously determine the content of caffeic acid, acteoside, isoacteoside and pogostone in Pogostemonis Herba formula granules, in order to provide basis for studying its quality standards. Method: An HPLC method was developed. Wondasil C18 column (4.6 mm×250 mm, 5 μm) was used and eluted with mobile gradient of 0.1% phosphoric acid-acetonitrile at the flow rate of 1.0 mL·min-1. The wavelength were set at 325 nm (0-18 min, caffeic acid), 330 nm (18-30 min, acteoside and isoacteoside), and 311 nm (30-55 min, pogostone). The column temperature was 30 ℃. Result: The good linear relationships between the concentration and peak area were between 0.021 8-0.218 μg (r=0.999 8) for caffeic acid, 0.032 6-0.326 μg (r=0.999 8) for acteoside, 0.099 4-0.994 μg (r=0.999 7) for isoacteoside acid and 0.174 2-1.742 μg for pogostone (r=0.999 9), respectively. The four tested indicators of sample solutions were steady within 24 h. The average recoveries were 98.85%, 99.53%, 99.37%, 100.58%, with RSD of 1.9%, 1.1%, 1.4%, 2.0%, respectively. Conclusion: Caffeic acid, acteoside, isoacteoside and pogostone were determined by this method simultaneously. The method is feasible and accurate with a good reproducibility, and can provide a reference for quality control.

      摘要:Objective: To establish a HLC fingerprint of the compounds in Fuke Qianjin tablets and make a multi-component quantitative determination,in order to provide a reference for quality control. Method: Sunfire C18 column (4.6 mm × 250 mm,5 μm) was used,with a mobile phase of 0.1% phosphoric acid and acetonitrile gradient elution. The flow rate was 1.0 mL·min-1. The detection wavelength was 326 nm from 0 to 12 min,316 nm from 12 to 18 min,225 nm from 18 to 24 min and 254 nm from 24 to 50 min. The column temperature was 30 ℃,and the sample size was 10 μL. Similarity evaluation and cluster analysis (CA) were performed to evaluate the fingerprint of the 11 batches of Fuke Qianjin tablets. Result: The common mode of the fingerprint included 14 common peaks,and 5 of them were identified by comparison with reference substances as chlorogenic acid,ferulic acid,andrographolide,berberine hydrochloride and dehydroandrographolide. The similar degrees of the 11 batches of samples were more than 0.90. Conclusion: The established method has a high sensitivity and specificity,and can be used for quality control of Fukeqianjin tablets.

      摘要:Objective: To investigate the major metabolites of ginkgolide A (GA),ginkgolide B (GB) and ginkgolide K (GK) from ginkgo diterpene lactone meglumine injection in human urine. Method: The urine samples of the volunteers after a single intravenous infusion administration of ginkgo diterpene lactone meglumine injection were collected for 0-6 h,then the samples were prepared with or without acidification treatment and analyzed by LC-MS/MS.The potential metabolites of GA,GB and GK were analyzed through comparing the total ion current (TIC) and extract ion current (EIC) chromatogram of the test samples with the blank urine and analyzing the quasi-molecular ion and secondary fragment ion of all chromatograms. Result: In addition to the prototype drugs,twelve metabolites were found in human urine containing five phase Ⅰ metabolites and one phase Ⅱ metabolite from GA,one phase Ⅰ metabolite and three phase Ⅱ metabolites from GB,one phase Ⅰ metabolite and one phase Ⅱ metabolite from GK. Conclusion: The major metabolic pathways of GA in human are hydroxylation and lactone ring hydrolysis,then hydrolyzation is followed by conjugation with sulfuric acid.The major metabolic pathways of GB are hydroxylation and lactone ring hydrolysis,then hydrolyzation is followed by conjugation with sulfuric acid,glucuronic acid,and glutathione,respectively.The major metabolic pathways of GK is methyl glucuronic acid binding reaction.

      摘要:Objective: To study the effects of different extracts from Wikstroemia indica root on the hepatotoxicity in rats. Method: Seventy-two SD rats were randomly divided into blank control group, ethanol extract group, petroleum ether extract group, ethyl acetate extract group, n-butanol extract group and water extract group, with 12 rats in each group. Different drug groups were intragastrically administrated with drugs for two weeks, and the rats' body weight was measured once every two days. On the fifteenth day, six rats were randomly selected from each group, and anaesthetized with 3% pentobarbital sodium, and their blood was collected from aorta abdominalis. After blood centrifugation, the alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total bilirubin (T-BIL), total protein (TP), albumin (ALB), blood glucose (GLU), total cholesterol (CHOL), and creatinine (Cr) in serum were detected. Their livers were collected and weighed, and the organ index was calculated. The hepatic histopathology was examined. Then the remaining six rats in each group were given no drug but fed with normal food and water for two weeks to observe their recovery. The above indexes were detected on the twenty-eighth day. Result: Compared with control group, the liver coefficients of ethanol extract group and ethyl acetate extract group increased significantly after two weeks (P<0.05). The serum levels of ALT, AST, GLB were increased, and A/G, TP and ALB were decreased (P<0.05) in ethanol extract group. The serum levels of ALP, T-BIL, GLU were increased, and A/G, TP and ALB were decreased (P<0.05) in ethyl acetate extract group. The serum levels of AST, AST/ALT, ALP, GLU were increased, T-BIL, ALB, TP and A/G were decreased in petroleum ether extract group (P<0.05,P<0.01). The serum levels of AST/ALT, T-BIL were increased and A/G decreased (P<0.05,P<0.01) in extract of butanol. Only A/G decreased in the serum of water extract group (P<0.05). The rat liver tissues of ethanol extract group, ethyl acetate extract group and petroleum ether extract group showed different degrees of inflammatory cell infiltration, certain fatty degeneration of liver cells and focal necrosis of liver cells, which were rare in other rare group. After the drug withdrawal for two weeks, the liver coefficient of each group was close to that of blank control group. The biochemical indexes of liver function in each group were recovered to the level of blank group, and there was no significant difference. The liver tissue cells of each group showed a clear structure, with rare inflammatory cells infiltration and no fatty degeneration and necrosis of liver cells. Conclusion: The ethanol extract group and the ethyl acetate extracted group of Miao medicine W. indica root have a greater damage to the liver, with obvious hepatotoxicity. The petroleum ether extract group also shows a certain injury to liver, which however is lower than that of the ethanol extract group and the ethyl acetate extracted group. The n-butanol extract group shows a lower toxicity, and the water extract group shows no toxicity. After recovery for two weeks, the biochemical indexes of liver function of rats in each group were almost recovered to the level of the blank group, and the liver injury of rats was significantly reduced, which indicated that the hepatotoxicity of W. indica root had a certain reversibility.

      摘要:Objective: To explore the curative effect of Tongxie Yaofang on ulcerative colitis (UC) in rat hypothalamus cytokine interleukin-6 (IL-6) and interleukin-6 receptors (IL-6R) gene and protein expressions. Method: After the ulcerative colitis model was established by the composite method. SPF Wistar rats were randomly divided into 5 groups: model group, low, middle, high-dose Tongxie Yaofang groups (2.75, 5.5, 11 g·kg-1), and 5-ASA group. After modeling, rats were given drugs for 21 days. Then, colonic mucosa damage was observed. The levels of IL-6 in the serum tissues were detected by enzyme linked immunosorbent assay (ELISA). The proteins expressions of IL-6 and IL-6R in rat hypothalamus tissues were detected by immunohistochemistry, and their gene expressions were measured by Real-time PCR method. Result: ELISA test results show that compared with control group, model group showed significant increase in content of IL-6 in serum of rats. After drug treatment, the serum IL-6 in drug group were decreased, with the most obvious effect in high-dose Tongxie Yaofang group (P<0.05). Real-time PCR results showed that model group's hypothalamic IL-6 and IL-6R gene expressions were higher than those of blank group (P<0.05). After treatment with drugs, drug group's IL-6 and IL-6R gene expressions were reduced, and 5-ASA and high-dose Tongxie Yaofang groups were superior to the other groups (P<0.05). Immunohistochemical staining results showed that compared with control group, IL-6 and IL-6R protein expressions of ulcerative colitis rats increased significantly (P<0.05), drug treatment can reduce their hypothalamic IL-6 and IL-6R protein expressions, and 5-ASA and high-dose Tongxie Yaofang groups were superior to the other groups (P<0.05). Conclusion: Tongxie Yaofang can obviously inhibit IL-6 and IL-6R gene and protein expressions in hypothalamic tissues of the rat UC model by TNBS/ethanol method, suggesting Tongxie Yaofang's mechanism in the treatment of ulcerative colitis may be correlated with inhibition of abnormal activation of IL-6/IL-6R signaling pathway in hypothalamus.

      摘要:Objective: To investigate the efficacy and its mechanism of rhein on dextran sulphate sodium(DSS )- induced ulcerative colitis in mice. Method: C57BL/6 male mice were randomly divided into 6 groups: normal control group, model group, mesalazine treatment group, low-dose rhein group, middle-dose rhein group and the high-dose rhein group (6.25, 12.5, 25 mg·kg-1), n=10 in each group. The mice in normal control group received distilled water, while all the other mice were treated with 3% dextra sulfate sodium for 7 days to induce the ulcerative colitis model. From the first day of modeling, mesalazine and different concentrations of rhein were given by gavage for 14 days continuously, and equal volume of distilled water was given in normal group and model group. The body weight, defecation condition and hematochezia of the mice were observed and recorded everyday; scores of disease activity index (DAI) were calculated and evaluated; colon was taken and its length was measured; colon paraffin slices were prepared and their pathological changes were observed under microscope after HE staining. Peripheral blood was taken and the serum was collected to detect the level of anti-flagellin antibody by using ELISA kit. The expression levels of Toll like receptor 5(TLR5) and nuclear factor (NF )-κB p65 in colon were measured by Western blot. Result: As compared with the normal control group, DAI scores were significantly increased in model group (P<0.01); the colon length was significantly shortened(P<0.01); meanwhile, epithelium and intestinal gland disappeared, with a large number of lymphocyte infiltration and local vascular dilation in submucous layer. Above symptoms were ameliorated after giving mesalazine and different concentrations of rhein (P<0.05, P<0.01). The anti-flagellin antibody in serum, the expression levels of TLR5 and NF-κB p65 were significantly increased in model group (P<0.01) as compared with the normal group; while these levels were decreased after treatment by mesalazine and different concentrations of rhein (P<0.05, P<0.01). Conclusion: Rhein could treat DSS-induced ulcerative colitis in mice, and its mechanism may be related to inhibiting the expression of TLR5/NF-κB p65 pathway.

      摘要:Objective: To observe the effect of Huangqi Liuyi Tang on diabetic rats' blood glucose, glycosylated hemoglobin, blood lipid and the renal tubular epithelial reabsorption of glucose transporter 2(SGLT2), and explore its efficacy and possible mechanismt in the treatment of diabetes. Method: The 77 SPF rats were randomly divided into 6 groups by weight: normal control group, model group, metformin group (0.15 g·kg-1), Huangqi Liuyi Tang low, middle and high dose groups (3.15, 6.30,12.60 g·kg-1). Diabetes models were established by using high fat and high glucose diet for 28 days and then with intraperitoneal injection of streptozotocin at the dose of 40 mg·kg-1 body weight. 7 days after STZ injection, corresponding drugs were given by gavage. Equivalent volume of water was given in the normal control group and model control group by the same method. Gavage medication lasted for six weeks, once a week. Venous blood from the tails was collected to measure blood glycose after 6-hour fasting; after fasting 12-hour at the last time of gavage, the venous blood from the tails was taken for blood glycose and then the rats were anaesthetized to collect the abdominal venous blood. The kidney and pancreas were taken for histopathological examination after the rats were sacrificed. The blood glucose, glycosylated hemoglobin, blood lipid, body weight, food intake, and the expression of renal tissue SGLT2 were observed. Result: The blood glucose levels in modeling groups were higher than those in normal control group (P<0.01), with a blood glucose value>13.0 mmol·L-1. After six weeks self-healing, the blood glucose level in model group was still significantly higher than that of normal control group (P<0.01); the blood glucose levels in metformin group, Huangqi Liuyi Tang in middle and high dose groups were lower than that in model group (P<0.05). Glycosylated hemoglobin level was increased in model group and higher than that in the normal control group (P<0.01). As compared with the model group, glycosylated hemoglobin level was decreased in metformin group and the Huangqi Liuyi Tang high dose group (P<0.05). In the pancreas pathological HE staining in model rats, complete pancreas islet structure was seen, indicating that islet was not destroyed completely in this experiment. Renal pathological slices showed that the structures of renal glomerulus, collections, tubular, proximal and distal tubule were complete in all groups, with no obvious pathological damages. After treatment, SGLT2 protein expression levels were reduced in various treatment groups (P<0.05, P<0.01). Conclusion: High sugar and high fat diet with intraperitoneal injection of STZ increased SGLT2 protein expression in kidney tissues of diabetic models. Huangqi Liuyi Tang can reduce blood glucose, glycosylated hemoglobin, and lower SGLT2 protein expression in diabetes model rats. Its treatment efficacy for diabetes may be associated with partial inhibition of renal tubular epithelial reabsorption of glucose.

      摘要:Objective: To observe the effect of Coriopsis extract on the levels of blood sugar, insulin secretion, and insulin receptor substrate-1/phosphatidyl inositol kinase(3)/glucose transporter type 4(IRS-1/PI3K/GLUT4) pathway of signal transduction in streptozocin(STZ)-induced diabetic rats. Method: Total 35 male STZ-induced diabetic Wistar rats were randomly divided into five groups: model group, Coriopsis extract low, medium and high dose groups, and positive control group (n=7 in each group); another 7 SPF male Wistar rats were used as normal controls. Normal saline was given in normal group and model group; the rats in Coriopsis extract low, medium and high dose groups received 100, 200, 400 mg·kg-1·d-1 extract respectively by intragastric administration; the rats in positive control group were administered with pioglitazone 10 mg·kg-1·d-1. After four weeks of treatment, pericardial blood was taken and serum was taken by centrifugation; in addition, pancreatic tissues were taken. ELISA assay was used to test the insulin (INS) level in rats; Western blot assay was used to test the protein expression levels of IRS-1,PI3K and GLUT4 in skeletal muscle; GLUT4 mRNA expression level was tested by Real-time PCR . Result: After 4 weeks of treatment, the effect of Coriopsis extract on reducing the body weight of diabetic rats was not obvious, but it could reduce the fasting blood glucose, postprandial 2 h blood glucose obviously, promote insulin secretion to a certain extent, and promote the expression levels of regulatory factors IRS-1,PI3K and GLUT4, with significant difference as compared with model group (P<0.05). Conclusion: Coriopsis extract can reduce blood sugar in type 2 diabetic rats, promotes insulin secretion, improve IRS-1,PI3K, GLUT4 expression levels in skeletal muscle of rats, and improve insulin resistance.

      摘要:Objective: To investigate the protective effect of hyperoside on H2O2-induced oxidative damage of A549 cells. Method: In vitro oxidative stress damage model was established by treating A549 cells with H2O2 (400 μmol·L-1) for 24 h. Cells were divided into blank group, model group (400 μmol·L-1 H2O2), positive drug group (N-acetyl cysteine 100 μmol·L-1), hyperoside groups (1, 10, 100 μmol·L-1). Cell viability was determined by thiazolyl blue(MTT) assay. Contents of reactive oxygen(ROS) and malondialdehyde(MDA), lactic dehydrogenase(LDH), catalase(CAT), glutathion peroxidase(GSH-Px) and superoxide dismutase(SOD) activity were detected by enzyme-linked immunosorbent assay(ELISA) method. The membrane potential changes of mitochondria were detected by Rhodamine 123 staining, and the protein expression levels of sirtuin 3(Sirt3), isocitrate dehydrogenase 2(IDH2) and MnSOD were determined by Western blot technology. Result: As compared with model group, hyperoside could significantly enhance the cell viability (P<0.01), decrease LDH activity, contents of ROS and MDA, and increase GSH-Px and SOD activities (P<0.05,P<0.01). Moreover, hyperoside markedly improved mitochondrial membrane potential, and Western blot showed that hyperoside could significantly up-regulate the protein expression levels of Sirt3, IDH2 and MnSOD (P<0.05,P<0.01). Conclusion: Hyperoside could protect H2O2-induced oxidative damage of A549 cells, and its protective effects may be related with increase in ROS removal ability and regulation of Sirt3 mitochondrial pathway.

      摘要:Objective: To investigate the effects and potential mechanisms of icariin (ICA) on acute gouty arthritis(GA) induced by sodium urate in rats. Method: Seventy-two Sprague Dawley rats were randomly divided into normal control group, model group, ICA groups (20, 40, 80 mg·kg-1) and colchicine group (1.5 mg·kg-1), 6 female and 6 male rats in each group. All rats were treated with ICA or colchicine or equal volume of normal saline by intragastric administration for 7 days according to the groups. At the 5th day, the gouty arthritis model was established by injection of 3% sodium urate (100 μL) into the right ankle joint cavity of rats. The circumference of the right ankle joint and score of gait were measured during 48 h treatment after model establishment. At 48 h after GA model establishment, all rats were anesthetized, and leukocyte infiltration was observed in arthroedema and synovial tissues by hematoxylin and eosin staining; the contents of interleukin (IL)-1β, IL-6, tumor necrosis factor(TNF )-α and prostaglandin E2(PGE2 ) were determined by ELISA kit; cyclooxygenase (COX-2) protein level was measured by Western blot in joint capsule and synovial tissues. Result: In model group, the tissue around joint was swelling; gait was limping; a large number of leukocytes were infiltrating in arthroedema and synovial tissues; the contents of IL-1β, IL-6, TNF-α were significantly higher than those in normal control group (P<0.05); the PGE2 and COX-2 protein levels were also significantly increased (P<0.05). As compared with model group, all of ICA low dose, middle dose and high dose treatment significantly attenuated the joint swelling and abnormal gait, and significantly decreased leukocyte infiltration and contents of IL-1β, IL-6, TNF-α and PGE2. Conclusion: ICA can attenuate the symptoms of acute GA in rats by inhibiting the inflammatory reaction.

      摘要:Objective: To investigate the anxiolytic effect of Bentuntang(BTT) in mice with chronic restraint stress and explore its possible mechanism from the perspective of the central neurotransmitter. Method: The 72 mice were randomly divided into 6 groups: saline group, model group, BTT low dose (3.51 g·kg-1), BTT middle dose (10.53 g·kg-1), BTT high dose group (31.59 g·kg-1) and diazepam group (4 mg·kg-1). All the drugs were administrated by intragastric administration for 30 days. Chronic restraint stress was used to induce mice anxiety models; open field test, locomotion activity test, elevated plus maze tests and light/dark box test were used to analyze the protective effect of BTT in stressed mice from the behavioral prospective; high performance liquid chromatography (HPLC) with fluorescence detection were used for detection of glutamate (Glu) and gamma aminobutyric acid(GABA) contents;the expression levels of GABAa-Rα5, GABA transporter (GAT-1) and GAT-3 in hippocampus were detected by Western blot. Result: The experimental results showed that as compared with the model group, high dose BTT could significantly improve the time in the central region in open field test(P<0.05), increase the entries and time in open arms in elevated plus maze tests (P<0.05); increase the level of GABA in the hippocampus (P<0.05), and decrease glutamate level (P<0.05). In addition, the protein expression levels of GAT-1 and GAT-3 in hippocampus were decreased. Conclusion: BTT could improve the chronic restraint stress-induced anxiety in mice, possibly by regulating the contents of amino acid neurotransmitters via decreasing GAT-1 and GAT-3 protein expression levels in the central nervous system .

      摘要:Objective: To investigate the effects of ginsenoside-Rg1 pretreatment on the PI3K-Akt-eNOS signaling pathway in the myocardium of rats with isoproterenol-induced acute myocardial ischemia. Method: SD rat models of acute myocardial ischemia were established with isoproterenol. 50 rats were randomly divided into normal control group, model group, puerarin group, Rg1 high dose group (20 mg·kg-1) and Rg1 low dose group (10 mg·kg-1). The blood flow on heart surface was observed by using Moor laser blood flow imaging system in all groups; the levels of creatine kinase (CK), lactate dehydrogenase (LDH) and nitric oxide (NO) in rat serum and the levels of superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) in rat myocardium were measured by ELISA assay; the mRNA expression level of eNOS in rat myocardium was detected by Real-time PCR; and the protein expression levels of PI3K, Akt and p-Akt in rat myocardium were detected by Western blot. Result: As compared with the normal group, average blood flow on heart surface was significantly reduced in model group, with decreased NO level and increased CK and LDH levels in serum; their myocardial MDA level was increased; GSH-Px level was reduced; eNOS mRNA level was significantly decreased and the protein expression of PI3K/Akt pathway was reduced (P<0.05, P<0.01). As compared with the model group, blood flow on heart surface was significantly increased in the ginsenosides Rg1 high-dose group and low dose group; NO level was increased, whereas the levels of CK and LDH in serum were decreased; the myocardial MDA level was reduced and the GSH-Px level was increased; the mRNA expression of eNOS was increased and the protein expression levels of PI3K expression/Akt pathway were increased (P<0.05, P<0.01). Conclusion: Ginsenoside-Rg1 can improve the heart condition in rats with acute myocardial ischemia through regulating PI3K-Akt-eNOS signaling pathway to prevent and treat cardiovascular disease.

      摘要:Objective: To explore the effect and mechanism of Shanjing capsule on energy metabolism, oxyradical and apoptosis-related genes in myocardial ischemia rats. Method: The chronic myocardial ischemia models were established with ligating coronary artery. Then the modeled rats were randomly divided into Shanjing capsule low dose, middle dose and high dose groups (283.5, 850.5,1 701.0 mg·kg-1), positive drug group (324.0 mg·kg-1). The rats in sham operation group and model group received equivalent amount of 0.9% Sodium Chloride Injection for 10 days. Then ultraviolet visible spectrophotometer was used to detect the content and activity of lactic acid (LD), free fatty acids (NEFA), malondialdehyde (MDA), total superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), NO and ATP in serum and tissues. The expression levels of nuclear transcription factor-κB (NF-κB), Caspase-3, B-cell lymphoma/leukemia-2(Bcl-2) and Bcl-2 associated X protein(Bax) in myocardial tissues were detected by Western blot and RT-PCR. Result: As compared with the sham operation group, contents of LD, NEFA, MDA were increased (P<0.05); levels of NO, Na+-K+-ATP, Ca2+-ATP enzyme, SOD and GSH-Px were decreased (P<0.05); the mRNA and protein expression of NF-κB, Caspase-3 and Bax in cardiomyocytes were up-regulated significantly (P<0.05), while the mRNA and protein expression of Bcl-2 was decreased (P<0.05) in model group. As compared with the model group, Shanjing capsules could decrease the contents of LD, NEFA, MDA (P<0.05), increase the levels of NO, Na+-K+-ATP, Ca2+-ATP enzyme, SOD and GSH-Px (P<0.05), significantly down-regulate the mRNA and protein expression levels of NF-κB, Caspase-3, Bax in cardiomyocytes (P<0.05), and up-regulate the mRNA and protein expression of Bcl-2 (P<0.05). Conclusion: Shanjing capsule could increase the ability of antioxidant and oxygen free radical scavenging in myocardial ischemia rats, improve energy metabolism of cardiomyocyte, regulate the expression of apoptosis related protein and genes by regulating the NF-κB signal pathway, which plays a role in myocardial preservation by inhibiting cardiomyocyte apoptosis and decreasing pathological lesion under cardiomyocyte ischemia and anoxia.

      摘要:Objective: To find out the hazard factor of toxic heat syndrome of acute myocardial infarction (AMI) and the causes. Method: Clinical data of the patients who were diagnosed as acute myocardial infarction at our hospital were collected retrospectively. We collected the patients' traditional Chinese medicine syndrome and divided the patients into toxic heat group and non-heat toxin group. We analyzed the general clinical data and the hospitalization prognosis of both groups. Logistic regression analysis was made to find out the hazard factor of toxic heat syndrome of acute myocardial infarction. Result: Totally 322 patients were enrolled in this study, including 42.20% of heat toxin patients (136 patients) and 57.8% of non-heat toxin patients (186 patients). In terms of the index of the culprit vessel and left ventricular ejection fraction and left ventricular diastolic volume, the heat toxin group was severer than the non-heat toxin group (P<0.05), but there was no significant difference in the hospitalization cardiovascular death between the two groups. According to the univariate analysis, toxic heat syndrome in the AMI patients were the multiple cardiovascular risk factors for people with smoking, spicy and high-fat diet, diabetes, hyperlipidemia (P<0.05). Smoking, low-density lipoprotein and triglyceride were the independent risk factors according to the logistic regression analysis, and the odds ratios were 1.755, 1.637, 1.483. Conclusion: The causes of the toxic heat syndrome on acute myocardial infarction include smoking and hyperlipidemia. There is no significant difference in the hospitalization prognosis between the two groups, but the heat toxin group is severer than the non-heat toxin group in the degree of ventricular remodeling.

      摘要:Objective: To observe the effect of Zhigancao Tang on heart rate variability and inflammatory factors in patients with cardiac arrhythmia with Qi-Yin deficiency. Method: A total of 90 patients with definite arrhythmia were randomly divided into treatment group (47 cases) and control group (43 cases). The control group received routine Western medicine plus amiodarone tablets, and treatment group was treated with Zhigancao Tang in addition to the therapy of control group. Both groups were treated for 8 weeks, and their arrhythmia efficacy, traditional Chinese medicine (TCM) symptom score and efficacy, heart rate variability (HRV), interleukin-6 (IL-6), and high sensitivity C-reactive protein (hs-CRP) indicators were compare. Result: After treatment, the therapeutic effect of arrhythmia in treatment group was better than that in control group (P<0.05). According to the comparison of TCM syndromes between two groups after treatment, the total effective rate in treatment group was higher than that in control group (P<0.05). Compared with before treatment, both groups could improve TCM symptom scores, and the score of improvement in treatment group were better than that in control group (P<0.01). After treatment, HRV in both groups was increased (P<0.05, P<0.01), while IL-6 and hs-CRP were decreased in both groups (P<0.05, P<0.01), the improvements of HRV, IL-6 and hs-CRP in treatment group were better than that in control group (P<0.01). Conclusion: Zhigancao Tang can improve the clinical efficacy of arrhythmia, regulate heart rate variability, inhibit inflammatory cytokines, improve clinical symptoms and repair cardiac autonomic neuropathy and inflammatory injury in patients with arrhythmia.

      摘要:Objective: To investigate the efficacy of anti-tumor synergistic formula combined with tegafur on advanced colorectal cancer and the effect on quality of life and serum tumor markers, and explore their mechanism of action. Method: A total of 100 advanced colorectal cancer (ACC) patients diagnosed at our hospital between January 2013 and December 2015 were selected and randomly divided into two groups, with 50 cases in each group. The control group was treated with Gimeracil and Oteracil Porassium capsules, and the experiment group was treated with anti-tumor synergistic formula in addition to the therapy of control group. Peripheral blood T lymphocytes and related tumor markers were detected, the quality of life was assessed, and follow-up visits were made to record their respective survival time and compare the treatment effect and survival rate. Result: Compared with before treatment, CD3+, CD4+ and CD4+/CD8+ levels were lower (P<0.01), and the CD8+ level was higher (P<0.01) in control group, and the CD3+, CD4+ and CD4+/CD8+ levels were higher (P<0.01), and the CD8+ level was lower (P<0.01) in experiment group. Carcinoembryonic antigen (CEA), carbohydrate antigen 242 (CA242) and carbohydrate antigen 19-9 (CA19-9) levels were lower in two groups (P<0.01), the Quality of Life Questionnaire-Core 30 (QLQ-C30) scale function domain score was increased (P<0.01), and the QLQ-C30 symptom domain score was lower (P<0.01). Compared with control group, the CD3+, CD4+ and CD4+/CD8+ levels were higher (P<0.01), the CEA, CA242 and CA19-9 levels were lower (P<0.01), the QLQ-C30 scale function domain score was increased (P<0.01), and the QLQ-C30 symptom domain score was lower (P<0.01), the objective response rate (ORR) was higher (P<0.05), and the follow-up survival rate was higher (P<0.05) in experiment group. Conclusion: Anti-tumor synergistic formula combined with tegafur can improve the quality of life and survival of ACC, shows an ideal treatment effect, and is worth promotion.

      摘要:Objective: To observe the curative effect of Ruangan Huaxian decoction combined with Entecavir in treating hepatic fibrosis of chronic hepatitis B patients with syndromes of liver depression, spleen deficiency and blood stasis obstruction and its effect on their quality of life. Method: A total of 100 patients with chronic hepatitis B were randomly divided into treatment group and control group according to parallel experimental design and randomized controlled single blind test. The treatment group (50 cases) was given Entecavir and Ruangan Huaxian decoction, and control group (50 cases) was given Entecavir and traditional Chinese medicine placebo. The course of treatment was 6 months. The changes in liver function, fibrosis index and hepatitis B virus etiology and quality of life were observed and compared before and after treatment. Before and after treatment, Medical Outcome Study 36-itenl Short Form Health Survey (SF-36) and Chronic Liver Disease Questionnaire (CLDQ) scale were used to measure patient's quality of life in both groups. Result: After treatment, alanine transaminase (ALT) normalization rate in observation group was significantly higher than that of control group (P<0.05). There were significant differences in hyaluronan (HA), laminin(LN), Ⅳ-C and PCⅢ between two groups after treatment (P<0.05, P<0.01). After treatment, the liver fibrosis indexes in treatment group were significantly lower than those in control group (P<0.05, P<0.01). After treatment, there was no significant difference in HBV-DNA negative rate, HBsAg negative rate, HBeAg negative rate and HBeAg conversion rate between two groups. Compared with before treatment, the oblique diameter and the diameter of portal vein of liver, and the diameter of splenic vein and the thickness of spleen in treatment group were significantly lower after treatment (P<0.05). After treatment, the treatment group was significantly lower than control group, but with no statistical significance. The total effective rate was 92% in treatment group and 74% in control group. There was a significant difference between the treatment group and the control group (P<0.05). After treatment, two groups showed higher CLDQ scale abdominal symptoms (AS), fatigue (FA), systemic symptoms (SS) and activity (AC) scores than those before treatment, and the treatment group was significantly higher than control group (P<0.05). After treatment, two groups showed higher SF-36 scale physical functioning (PF), role physical (RP), vitality (VT), body pain (BP) and general health (GH) than before treatment, and the treatment group was significantly higher than the control group (P < 0.05). Conclusion: Ruangan Huaxian decoction combined with Entecavir for chronic hepatitis B liver fibrosis with syndromes of liver depression, spleen deficiency and blood stasis obstruction is safe, reliable and effective. It can significantly improve the quality of life of CHB patients.

      摘要:Objective: To explore the clinical efficacy of improved Tongqiao Huoxuetang combined with valproate for manic episodes in bipolar disorder patients and investigate its effect on biochemical indicators. Method: The 104 patients with manic episodes of bipolar disorder admitted to our hospital from February 2014-August 2015 were randomly divided into observation group (52 cases) and control group (52 cases). Patients in control group received sodium valproate treatment, and patients in observation group received improved Tongqiao Huoxuetang treatment on the basis of treatment in control group. The course of treatment was 8 weeks in both groups. Result: The total effective rate(94.23%)and effective rates of different syndromes(96.00%,93.33%,91.67%)in observation group were higher than those in control group (73.08%,73.91%,77.78%, and 72.73%) (P<0.05). Hamilton Depression Scale(HAMD) total scores and each factor scores were decreased significantly in both groups after treatment (P<0.05); the above scores in observation group were significantly lower than those in control group after treatment (P<0.05).After treatment, Bech-Rafaelsen Mania Rating Scale(BRMS) scores and Generic Quality of Life Inventory(GQL-LI) scores were decreased significantly in both groups(P<0.05); and the above scores in observation group were significantly lower than those in control group after treatment (P<0.05). The serum levels of interleukin-1β(IL-1β), interleukin-4(IL-4) and tumor necrosis factor-α (TNF-α) were significantly decreased after treatment in both groups (P<0.05); and the above levels in observation group were lower than those in control group after treatment(P<0.05). Plasma albumin and total bilirub in levels were increased in both groups after treatment (P<0.05); and the above levels in observation group were higher than those in control group after treatment(P<0.05). The adverse reaction rate in observation group (7.69%) was lower than that in control group (26.92%) (P<0.05). Conclusion: The improved Tongqiao Huoxuetang combined with valproate is safe and reliable with obvious efficacy formanic episodes in bipolar disorder patients, which could decrease the serum IL-1β, IL-4, TNF-α levels and improves the plasma albumin, total bilirubin levels.

      摘要:Objective: To discuss the clinical effect of modified therapy of Zhuye Shigao Tang combined with Qingqi Huatan pills in treating chronic obstructive pulmonary disease at acute exacerbation stage (AECOPD) with syndrome of phlegm-heat obstructing lung and investigate its effect on levels of hypersensitive C-reactive protein (hs-CRP), copeptin (CPP), serum amyloid A (SAA), nuclear factor-κB (NF-κB) and procalcitonin (PCT). Method: One hundred and sixty-six AECOPD patients were randomly divided into control group (83 cases) and observation group (83 cases) by random number table. Patients in control group took oxygen therapy by referring to the guide. Received ceftezole sodium injection by intravenous infusion, 2 g/time, 2 times/day by oral administration. Prednisolone tablets, 10 mg/time, 3 times/day. And ipratropium bromide aerosol, 3-4 times/day, 2 sprays/time. Based on the treatment in control group, the patients in observation group also received modified therapy of Zhuye Shigao Tang combined with Qingqi Huatan pills. The treatment course was 14 days for both groups. The main outcome measures were COPD patients self-assessment test questionnaire (CAT), modified version of the British Medical Research Council's Respiratory Questionnaire (mMRC), and scores of phlegm-heat obstructing lung syndrome. The secondary indicators were arterial blood gas analysis [blood oxygen (PaO2), carbon dioxide partial pressure (PaCO2), oxygen saturation (SaO2) and pH value] and pulmonary function [forced expiratory volume in 1 second (FEV1) and 1 second forced expiratory volume in percent predicted (FEV1%)]. In addition, the levels of hs-CRP, CPP, SAA, NF-κB and PCT were detected. All of the above indicators were detected once before and after treatment. Result: The total clinical effective rate was 94.87% in observation group, higher than 83.54% in control group (χ2=4.721, P<0.05). After treatment, scores of CAT, mMRC and syndrome of phlegm-heat obstructing lung in observation group were all lower than those in control group (P<0.01). Levels of PaCO2,hs-CRP, CPP, SAA, NF-κB and PCT were lower than control group. And the levels of PaO2, SaO2, FEV1 and FEV1% were higher than those in control group (P<0.01), but there was no statistically significant difference in pH value between two groups. Conclusion: Based on the basic western medicine treatment, modified therapy of Zhuye Shigao Tang combined with Qingqi Huatan pills can control the symptoms and infection, ameliorate pulmonary function, reduce inflammation, and improve the clinical efficacy in patients with AECOPD and syndrome of phlegm-heat obstructing lung.

      摘要:Objective: To observe the effects of Bazhen decoction modified fatigue state, quality of life and immune function in patients with cancer-related fatigue (CRF) and Qi and blood deficiency after colorectal cancer surgery. Method: One hundred and sixty patients were divided into control group (80 cases) and observation group (80 cases) by complete computer software balanced grouping method. The patients in both groups received comprehensive symptomatic treatment such as relieving pain, antiemetic, nutritional support, appropriate aerobic exercise, immunomodulatory, improving sleeping, psychological adjustment and nursing. Patients in control group received Buzhong Yiqi mixture, 10 mL/time, 3 times/day. Patients in observation group received Bazhen decoction modified, 2 times/day, 1 dose/day. The course of treatment was 3 months in both groups. Before and after treatment, Piper fatigue revision scale (PFS-R), Qi and blood deficiency and quality of life measurement for cancer patients (EORTC QLQ-C30) scale were graded, and levels of T lymphocyte subsets (CD3+, CD4+, CD8+, CD4+/CD8+), natural killer cells (NK), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interferon-γ (IFN-γ) were detected. Result: After treatment, scores of PFS-R and Qi and blood deficiency in observation group were lower than those in control group (P<0.01). Ridit analysis showed that, degree of fatigue in observation group was lighter than that in control after treatment (P<0.01).Scores of somatic function, role function, emotional function, cognitive function, social function and overall quality of life were higher than those in control (P<0.01). And scores of fatigue, nausea and vomiting, sleep difficulties, loss of appetite, constipation and diarrhea were lower than those in control group (P<0.01). Levels of CD3+, CD4+, CD4+/CD8+, NK and IFN-γ in observation group were higher than those in control group (P<0.01). While the levels of TNF-α and IL-1β were lower than those in control group (P<0.01). Conclusion: Bazhen decoction modified can regulate inflammatory factors, improve immune function, and ameliorate symptoms caused by CRF and improve their quality of life.

      摘要:Objective: To analyze the composition principles of Chinese patent medicine for treating vertigo in Chinese Pharmacopeia (2015 edition, volume I) and the People's Republic of Chinese Ministry of Public Health-Chinese Patent Preparations (Chinese Patent Preparations for short). Method: Through Traditional Chinese Medicine Inheritance Support System (TCMISS, V2.5), Chinese patent medicines for treating vertigo set forth in Chinese Pharmacopeia and Chinese Patent Preparation were selected, their information was entered to build a database. The frequently used syndromes and drugs, combination principles and new Chinese prescriptions for vertigo were analyzed. Result: By analyzing 329 Chinese patent medicines for treating vertigo, vertigo could be classified into excess syndrome and deficiency syndrome. ① The vertigo of excess syndrome totally involved 147 Chinese patent medicines, the main syndromes were liver-Yang hyperactivity and obstruction of collaterals by blood stasis; The pathogenesis included wind, fire, phlegm and blood stasis. The drugs were dominated by heat-clearing drugs, blood-activating drugs and calming-liver and suppressing liver-yang drugs. The main drug combinations were Scutellariae Radix, Coptidis Rhizoma, Phellodendri Chinensis Cortex, Gardeniae Fructus; Pinellia Rhizoma, Citri Reticulatae Pericarpium, Poria, Glycyrrhizae Radix et Rhizoma, Atractylodis Macrocephalae Rhizoma and Gastrodiae Rhizoma, Chuanxiong Rhizoma, Salviae Miltiorrhizae Radix et Rhizoma, Puerariae Radix. Five new prescriptions for excess syndrome were obtained: wind-dispelling prescription, wind-calming prescription, fire-purging prescription, phlegm-eliminating prescription and blood-activating prescription. ②The vertigo of deficiency syndrome totally involved 182 Chinese patent medicines, the main syndromes were deficiency of Qi and blood and deficiency of liver and kidney, the drugs were based on kidney-reinforcing drugs, blood-tonifying drugs and Qi-invigorating drugs, the main combinations were Rehmanniae Radix Praeparata,Dioscoreae Rhizoma,Poria,Lycii Fructus,Schisandrae Chinensis Fructus;Ginseng Radix et Rhizoma,Ophiopogonis Radix,Schisandrae Chinensis Fructus,Poria and Angelicae Sinensis Radix,Astragali Radix,Atractylodis Macrocephalae Rhizoma,Dioscoreae Rhizoma,and got 3 new prescriptions according to deficiency syndrome 's pathogenesis: reinforcing kidney prescription, nourishing liver and kidney prescription, reinforcing Qi and blood prescription. Conclusion: The pathogenesis of vertigo was complex, including wind, fire, phlegm, blood stasis and deficiency. We should pay attention to all of them in the treatment, determine etiologic factors based on differentiation, give consideration to both excess and deficiency, and treat both the symptoms and the causes of disease, in order to improve the clinical efficacy.

      摘要:Objective: To analyze regularities of medicinal diet recipes in Chinese Medicinal Diet Dictionary for phlegm-dampness constitution based on Traditional Chinese Medicine Inheritance Support System (TCMISS) (V2.5), in order to provide references for diet therapy for adjusting phlegm-dampness constitution. Method: TCMISS was used to build a database of medicinal diet recipes for adjusting phlegm-dampness constitution. The software integration data mining method was used to analyze the regularities of prescriptions. Result: By analyzing frequencies of 1 393 medicated diet recipes, we found 367 kinds of Chinese materia medica, 523 foods and 68 condiments. The three most commonly used medicines were Zingiberis Rhizoma Recens, Jujubae Fructus and Dioscoreae Rhizoma. And the three most commonly used foods weresticky rice, egg and carp. At the same time, 35 common combinations were achieved through association rules, and 16 new prescriptions were mined based on complex networks entropy method. Conclusion: This study is the first time to analyze the regularities of medicinal diet recipes composed of medicines and foods through TCMISS, and a pioneering attempt compared with the previous analysis on prescriptions consisting of only traditional Chinese medicines.

      摘要:Objective: To dig up the medication rules used by famous doctors in Lingnan bone injury. Method: The database of Lingnan bone injury prescriptions were established through collecting and analyzing famous doctors' works, manuscripts, papers and other literature. Frequency analysis, Latent Dirichlet Allocation (LDA) thematic model and association rules were used to analyze their medication rules. Result: The literature of lingnan bone injury involved 802 prescriptions and 811 kinds of traditional Chinese medicine (TCM) herbs. The warm drugs were used most, accounting for 28.3%. The drugs categories were dominated by dissipating blood stasis drugs, tonifying deficiency drugs, Qi-regulating drugs, heat-clearing drugs, removing wind-dampness drugs, and interior-warming drugs. Five core prescriptions were obtained from the LDA thematic model, and the through association rules analysis, a total of 21 herbal pairs and herbal group association rules. The core herbs included Angelicae Sinensis Radix, Chuanxiong Rhizoma, Paeoniae Radix Alba, Paeoniae Radix Rubar and so on. Conclusion: The famous doctors of Lingnan bone injury emphasized the treatment of activating blood and resolving stasis. On this basis, the drugs showed obvious early, middle and late stages of drugs use for bone injury: in early stage, clearing heat and detoxicating, or emphasizing hemostasis, or opening orifice and waking up;in middle stage, both attacking and tonifying, or both tonifying liver and kidney;in late stage, mostly tonifying (tonifying Qi, blood and spleen), combined with activating Qi, blood, and nourishing liver and kidney. On this basis, the medication also reflected the importance of invigorating spleen and eliminating dampness in characteristic treatment in Lingnan bone injury.

      摘要:Objective: To analyze the prescription rule of traditional Chinese medicine (TCM) medication for bradyarrhythmias by means of data mining. Method: The clinical research literature on TCM for bradyarrhythmias from 1975 to 2015 was searched in CNKI,a database of TCM decoctions for bradyarrhythmias was established.The frequency,dosage and association rule of the herbs were analyzed. Result: Based on the analysis of 102 herbs in 56 prescriptions,the frequency and dosage of each herb and association rules among the herbs were computed,6 main herbs and 2 core combinations were mined from the database. Conclusion: The core herbs of TCM for bradyarrhythmias are Aconiti Lateralis Radix Praeparata,Cinnamomi Ramulus,Astragali Radix and so on,Astragali Radix should be as a monarch herb in prescription.

      摘要:Dendrobii Officinalis Caulis is a traditional medicinal plant used for thousands of years in the daily health care and clinical treatment. It has attached more and more attention by domestic and foreign scholars. Based on the previous studies, the author collected, analyzed and summarized the literature on Dendrobii Officinalis Caulis at home and abroad in recent 10 years, and systemically reviewed the research advances in chemical constituents and pharmacological activities. The research results showed that the chemical compositions of Dendrobii Officinalis Caulis were complicated, including polysaccharides, stilbenes, amino acids, mineral elements, alkaloids, volatile oil and so on, mainly of polysaccharides and stilbenes. According to modern pharmacological researches, this herb medicine has many activities, such as antioxidant, anti-tumor, immune regulation, relieving fatigue and treating diabetes, mainly of antioxidant and anti-tumor effects. In this paper, the chemical constituents and pharmacological activities of Dendrobii Officinalis Caulis were summarized based on the studies at home and abroad, hoping to provide a theoretical reference for further research, reasonable exploitation and utilization of the species.
