      摘要:Objective: To improve matrix composition of Qufeng Zhitong ointments and check its skin safety. Method: Taking appearance,ductility,viscosity,pH value and centrifugal stability as comprehensive evaluation index,response surface methodology was employed to optimize matrix formulation with ratio of oil phase to water phase,amounts of stearic acid,glycerin and 72/721 emulsifier as factors.Then its skin irritation,skin allergy and acute skin toxicity was investigated. Result: The best matrix formulation of Qufeng Zhitong was as followings:glycerin of 16.5 g,72/721 emulsifier of 25.5 g,stearic acid of 26 g,oil phase-water phase (1:7.8).The ointments showed no skin irritation,allergy and toxicity. Conclusion: After being improved,Qufeng Zhitong ointments has good preparation,stable performance and no-toxic for external use.

      摘要:Objective: To prepare shortstalk monkshood root total alkaloids microemulsion. Method: Formulation of shortstalk monkshood root total alkaloids microemulsion was selected and prepared by pseudo-ternary phase diagram method with different oil phase,emulsifier and co-emulsifier,and preliminarily study its stability and physicochemical properties.HPLC was adopted to determine the content of bullatine A. Result: The best prescription was total alkaloids in Aconitum brachypodum of 2%,isopropyl myristate of 3.3%,polyoxylethylene castor oil ether 35 of 19.8%,absolute ethanol of 9.9%,water of 65.0%.The microemulsion was clear,transparent and spherical with particle size among 10-100 nm.Storage at 4℃ and 25℃ for six months is stable.The content of bullatine A was 2.28 mg·g-1. Conclusion: The optimized microemulsion formulation can achieve expected requirements.It can lay a foundation for further study of shortstalk monkshood root total alkaloids microemulsion.

      摘要:Objective: Preparation and evalution of baicalin molecularly imprinted polymer prepared by precipitation polymerization. Method: The molecularly imprinted polymers (MIPs) microsphere which using baicalin as template molecule, acrylamide (AM) as function monomer, ethyleneglycol dimethacrylate (EGDMA) as cross-linker were synthesized by precipitation polymerization. The surface morphologies and characteristics of the MIPs were characterized by transmission electron microscope (TEM) and infrared spectroscopy (IR). The adsorption properties of the polymers for the template molecule, baicalin, were determined respectively by the static adsorption equilibrium experiments, dynamic adsorption equilibrium experiments and selective experiments. Result: The results of above experiments show that dispersivity which using the precipitation polymerization of polymer molecules is excellent, the morphology have a high quality, and the particle size distribution is uniform. On the other hand, the experiment shows that molecular imprinted polymers have higher selective ability and stronger binding capacity for baicalin by precipitation polymerization. Conclusion: The molecular imprinting technology (MIT) provide a new way for the efficient separation and enrichment from the natural product of baicalin.

      摘要:Objective: To analyze factors affecting formulation of quercetin liquid proliposomes and evaluate its quality. Method: Quercetin liquid proliposomes was prepared.Taking particle size and encapsulation efficiency as indexes,formulation of quercetin liquid proliposomes was optimized by single factor experiment and orthogonal test.Free quercetin and quercetin proliposomes was separated by dialysis method,the content of quercetin was determined by HPLC,stability of quercetin proliposomes in artificial gastric juice and artificial intestinal juice was investigated. Result: Optimum formulation of quercetin liquid proliposomes was as follows:lecithin as lipid membrane material,cremorphor RH40 as surfactant,drug phospholipids ratio of 1:20,cremorphor RH40 amount of 20% to total content of phospholipid and main drug,the concentration of quercetin of 10 g·L-1.The particle size,Zeta potential and encapsulation efficiency of the proliposomes after hydration were (228.7±2.61) nm,(-21.2±1.47) mV and (93.12±1.18)% with good stability within 12 h and good repeatability.It was suitable for oral administration. Conclusion: Quercetin liquid proliposomes has good in vitro properties.Within 12 h,its particle size do not change significantly in each aqueous medium.

      摘要:Objective: To optimize purification process of total saponins in fermentation products of Notoginseng Radix et Rhizoma by anion exchange resin. Method: Single factor tests were conducted on resin type,diameter-height ratio of resin column,flow rate,absorption-influencing factors and elution-influencing factors,in order to ensure purity of total saponins in fermentation products of Notoginseng Radix et Rhizoma. Result: D301 anion resin was selected,its optimum purification parameters were as follows:diameter-height ratio of 1:8.3,dry weight of resin-fermentation products of 1:0.64,sample solution concentration of 0.25 g·mL-1,sample solution pH of 9,flow rate of 2 BV·h-1,adsorption time of 15 minutes.In addition,impurities were removed by 5 BV of distilled water before 4 BV of 60% ethanol was used to elute target compounds.Purity of total saponins had been successfully purified to more than 90%. Conclusion: This study achieves a simple,efficient and repeatable process with excellent purification performances for total saponins in fermentation products of Notoginseng Radix et Rhizoma,based on which industrial production can be fulfilled.It provides a theoretical support for new drug and related products development of total saponins in fermentation products of Notoginseng Radix et Rhizoma.

      摘要:Objective: To observe contents variation of emodin,chrysophanol and physcion from Rhei Radix et Rhizoma in Huangxiong Kangshuan capsules before and after compatibility. Method: According to prescription,herbs were extracted separately in different proportion and separate extraction were combined partly.Contents of emodin,chrysophanol and physcion were determined by UPLC. Result: Contents of emodin,chrysophanol and physcion increased significantly when herbs extracting separately,which reached to 2.437,17.040,2.720 mg·g-1,respectively;then declined to 1.914,13.236,2.155 mg·g-1 in combined samples;decreased significantly when herbs extracting together,which reached to 1.308,6.478,1.169 mg·g-1,respectively. Conclusion: Great influence of combination of Rhei Radix et Rhizoma with other herbs in this prescription on contents of main chemical constituents is mainly carried on in extraction process of Huangxiong Kangshuan capsules.

      摘要:Objective: To realize quality control of water extracting process,near-infrared (NIR) spectroscopy was used to establish a rapid quantitative analysis model for the content of gastrodin and total solid content in the second water extracting process of Tianshu capsules,which could timely reflect its state. Method: Samples of the second water extracting process of Tianshu capsules were collected,after rejection of abnormal samples,pretreatment of spectra and choice of corresponding wave band,multivariate calibration models were respectively established by partial least squares (PLS) algorithm between spectra and the content of gastrodin or total solid content.Samples in the concentrating process,which did not participate in modeling were used to test performance of models. Result: Correlation coefficients of the content of gastrodin and total solid content were 0.9680 and 0.9772;predicted coefficients of them were 0.9659 and 0.9786,root mean square error of calibration (RMSEC) were 4.686 and 0.053,root mean square error of prediction (RMSEP) were 3.698 and 0.034,and the relative standard errors of predictions (RSEP) were 4.85% and 3.07%,respectively. Conclusion: The method mentioned above is proved to be convenient,rapid,accurate and applicable for fast analysis and monitoring of active components in the second water extraction process of Tianshu capsules.

      摘要:Objective: To set up HPLC fingerprints of Rehmannia Radix Praeparata processed by Jiangxi Jianchang medicinal Band and provide basis for quality evaluation and quality control of processed Rehmannia Radix Praeparata. Method: The fingerprints of Rehmannia Radix Praeparata processed by Jiangxi Jianchang medicinal Band were established by HPLC. Data analysis was conducted according to "2004A similarity software of Chinese medicine fingerprints" of the Chinese Pharmacopoeia Commission. Result: Thirteen peaks were identified in HPLC fingerprint of ten batches samples of Rehmannia Radix Praeparata processed by Jiangxi Jianchang medicinal Band, and the overall similarity was good according to the similarity calculation. Conclusion: This method is stable and reproducible, and can be used for quality evaluation and quality control of Rehmannia Radix Praeparata processed by Jiangxi Jianchang medicinal Band.

      摘要:Objective: To establish HPLC method for simultaneous determination of hydroxysafflor yellow A and salvianolic acid B in Baidianfeng pills. Method: The determination was performed on a Welchrom C18 column with acetonitrile-0.1% phosphoric acid as the mobile phase in gradient elution mode at the flow rate of 1.0 mL·min-1. The detection wavelength was 280 nm. Result: Hydroxysafflor yellow A presented a good linearity in the concentration range of 4.91-36.86 mg·L-1 (r=0.9995). The average rate of recovery was 99.12% with RSD 0.9%. Salvianolic acid B presented a good linearity in the concentration range of 14.99-112.42 mg·L-1 (r=0.9997). The average rate of recovery was 98.79% with RSD of 1.0%. Conclusion: This method is simple, accurate and reproducible, and it can be used for quality control of Baidianfeng pills.

      摘要:Objective: To established an UPLC method for simultaneously determination of two indicative components of Cinnamomum (Lauraceae), namely procyanidin B-2 and procyanidin C-1, and to complete the determination of 17 batches of cinnamon bark samples by the established method. Method: The UPLC analysis was performed on an shiseido capcell pak C18 column (2.0 mm×100 mm, 2μm) with acetonitrile(A)-0.2% acetic acid solution(B) as mobile phase for gradient elution(0-8 min,11%-20%A;8-10 min,20%-25%A;10-12 min, 25%-75% A;12-13 min,75%A). The detection wavelength was set at 280 nm. The flow rate was 0.35 mL·min-1. The column temperature was set at 25℃. Result: Two indicative components showed a wide linear range and a good linear relationship. The content of the two components by UPLC is moderately correlated with the content of total polyphenol by Ultraviolet-visible Spectrophotometry in the 17 batches of cinnamon bark samples from market. Conclusion: The established method is simple, accurate and reproducible, and can be applied to determine the content of water-soluble oligomers procyanidin in cinnamon bark.

      摘要:Objective: To establish HPLC method for simultaneously determining 11 constituents in Yinchenhao Tang composed by decoction pieces from 10 hospitals. Method: HPLC was used on Symmetry C18 column (4.6 mm×250 mm, 5μm), with acetonitrile-0.1% phosphoric acid solution as a mobile phase in gradient elution, at a flow rate of 1.0 mL·min-1.The column temperature was 30℃ and detection wave length was set at 254 nm. Result: The mass of gallic acid, deacetylasperulosidic acid methyl ester, geniposide, aloe-emodin-8-O-glucopyranoside, chrysophanol-1-glucoside, emodin-8-glucoside, aloe-emodin, rhein, emodin, chrysophanol and physcion was between 0.142-1.016,0.518-1.256,9.021-17.214,0.088-0.793,0.097-0.990,0.187-1.587,0.057-0.578,0.147-1.062,0.114-0.730,0.380-1.898,0.375-1.958 mg·g-1, respectively. Conclusion: The content of active constituents in Yinchenhao Tang composed by different decoction pieces has significant differences.

      摘要:Objective: To establish a HPLC method for determination ferulic acid,liquiritin,naringin,hesperidin,neohesperidin,ammonium glycyrrhizinate,emodin,chrysophanol and physcion in Runchangtong pills. Method: HPLC was applied on a Agilent C18 column(ZORBAX SB-AQ, 4.6 mm×250 mm,5μm)with acetonitrile -0.2% formic acid water as mobile phase by gradient elution.The flow rate was 1.0 mL·min-1, and multiple detection wavelength of 235 nm (liquiritin,ammonium glycyrrhizinate), 285 nm (naringin,hesperidin,neohesperidin,emodin,chrysophanol,physcion),320 nm (ferulic acid). Result: The methodology of 9 components met the requirments with no interference of negatives,the average recovery rates were 97.0%-102.4%,RSD was 0.9%-1.9%. Conclusion: This eastablished method eastablished is easy,accurate with high specificity.This study provides scientific basis for the quality evaluation of Runchangtong pills and preparations of the same kind.

      摘要:Objective: To assay the volatile constituents of Paederia scandens. Method: The volatiles were analyzed by head-space solid micro-extraction, coupled with GC/MS and Kovats index. Result: There were 18 kinds of volatile constituents in P. scandens, 95.28% of the total essential constituents. The contents of thymol (30.97%), dimethyl disulfide(9.78%),quinoline (5.74%), n-hexadecanoic acid (5.53%), dibutyl phthalate (5.07%), dimethyl phthalate (3.67%), nonanal (3.65%), and diisobutyl phthalate (3.18%) were higher. Conclusion: The major volatile constituents of P. scandens were thymol and phthalate esters.

      摘要:Objective: To study the chemical constituents in the stem bark of Pterostyrax psilophyllus. Method: The chemical constituents were separated and purified by chromatographic methods combined with spectroscopic analyses. Result: Twelve compounds were isolated from the stem bark of P. psilophyllus and identified as following:methyl-2,4-dihydroxy-3,6-dimethylbenzoate(1), lupeol(2), prunetin(3), genistein(4), 2-hydroxy-4-methoxy-3,6-dimethyl benzoic acid(5), genistein-4'-O-β-glucoside(6), amyrin(7), catechin(8), epicatechin(9), vanillic acid(10), β-sitosterol(11), daucosterol(12). Conclusion: All twelve compounds were isolated from the plants of P. psilophyllus for the first time.

      摘要:Objective: To study the dynamic changes of chemical compositions in different organs of cultivated Arctium lappa from Ningxia during different growth periods,the method of HPLC with switching wavelengths was used to analyze the contents of arctiin,arctigenin,and chlorogenic acid in different organs of cultivated A. lappa from Ningxia during different growth periods. Method: The HPLC analysis was carried out on Agilent C18(4.6 mm×250 mm,5μm)column at the temperature of 30℃;the mobile phase composed of acetonitrile and 0.1%phosphoric acid with gradient elution at a flow rate of 1.0 mL·min-1, the detection wavelength was 280 nm shifted to 328 nm;the sample loading volume was 10μL. Result: The contents of lignans and phenol chemicals were found to have larger variations in different organs and during different growth perioda;the fruit has the highest contents of the rctiin and arctigenin,and the content of chlorogenic acid is the highest in the leaves. Conclusion: This study has provided theoretical basis for determining the rational utilization and suitable harvest periods of arctium A-lappa,in Ningxia,as wellas for evaluating the product quality.

      摘要:Objective: To investigate chemical constituents from flowers of Pueraria lobata. Method: Flowers of P. lobata was carried out with silica column chromatography,Sephadex LH-20 and preparative HPLC. The structures were identified on the basis of physicochemical properties and spectroscopic data. Result: Five isoflavone glycosides were isolated and identified as 6"-O-xylosyl glycitin(1) 5,6,7,4'-tetrahydroxyisoflavone-6,7-di-O-β-D-glucopyra noside(2),glycitin(3),6"-O-xylosyltectoridin(4) and tectoridin(5). Conclusion: The compound 1 and 3 were isolated from flowers of P. lobata for the first time in domestic. The compound 1 and 2 were not report from other plants in domestic.

      摘要:Objective: To establish the HPLC fingerprint chromatography of Lemai granules and determine the content of six compounds. Method: HPLC-DAD fingerprints of Lemai granules were determined with high-performance liquid chromatography on Phenomenex Luna C18 column (4.6 mm×250 mm, 5μm) with the gradient solvent system consisting of methanol (A) and water (B).The flow rate was 1.0 mL·min-1;the detection wavelength was set at 210 nm, and the column temperature was maintained at 30℃. Result: Fingerprints of Lemai granules were obtained. A total of 16 chromatographic peaks were common peaks, and the fingerprints of 10 batches of Lemai granules were identified with high similarity of fingerprints. Six chemical components were identified and their contents were determined. Conclusion: The establishment of HPLC fingerprints provide a scientific basis for specifying medicinal resources and quality evaluation of this formula.

      摘要:Objective: To investigate metabolism of rat intestinal flora on pulchinenoside B3,so as to provide scientific basis for metabolism characteristic analysis of saponins in intestinal flora. Method: With phillyrin as internal standard,UPLC-ESI-Q-TOF-MS/MS with negative ion and multiple reaction monitoring (MRM) mode was applied to analyze metabolites of pulchinenoside B3 in rat intestinal flora. Result: Pulchinenoside B3 prototype and other eight metabolites were detected in rat intestinal flora.Various types of metabolitic reactions happen to pulchinenoside B3 in intestinal flora,including 3-side chain sugar-remove,hydroxylation,methylation, and methoxylation on the mother nucleus of aglycone. Conclusion: Pulchinenoside B3 can be effective metabolism by rat intestinal flora under these experimental conditions,which shows these eight metabolites are active ingredients of pulchinenoside B3.

      摘要:Objective: To investigate abnormal changes of plasma endogenous metabolic in rats with W256 sarcocarcinoma and regulation of Xiaoai Jiedu prescription (XJP) on metabolic disorder of transplanted tumor rats.To explore mechanisms of XJP on anti-tumor based on metabonomics. Method: W256-transplanted tumor rats model were established and were divided into the control group,the model group and XJR treatment groups.Plasma smaples were collected on the 1st,6th and 11th day after administration.Endogenous metabolites of all groups were determined by GC-MS.Partial least squares-discriminant analysis (PLS-DA) was applied to analyze multivariate data and established model,metabolite identification was adopted database,t test was used in significant statistical analysis and metabolic pathways. Result: Compared with the control group,rats in the model group on the 11th day,30 endogenous metabolites significantly changed,10 endogenous metabolites got differently restoration to the normal group after treated XJR. Conclusion: Thirty endogenous metabolites may be potential metabolic biomarkers.Abnormal metabolite levels in plasma can be partly recovered by XJR,these 10 endogenous metabolites may be biomarker of XJP working.And potential antitumor effects of XJR may be contributed to regulation of related metabolic pathways.

      摘要:Objective: To explore pharmacokinetic characteristics of the new calcium sensitizer-M6 in rat plasma,and establish a fast,convenient and effective HPLC for determination of M6 in rat plasma. Method: Jugular vein haemospasia was adopted,M6 was injected by tail vein in rats,plasma concentration was determined by HPLC after 1,3,5,10,15,30 min and 1,2,4,6,8 h,mobile phase of 1% acetic acid solution-acetonitrile (85:15),detection wavelength was set at 280 nm.Plasma concentration-time curve was drawn,and pharmacokinetic parameters were calculated by PKSolover software. Result: Linear range of M6 was 3.125-200 mg·L-1,the limit of quantitation was 0.6 mg·L-1,the intra-day,inter-day precision and stabilization were less than 10%,recovery of method was more than 85%.These main pharmacokinetic parameters of M6 were as followed:tmax of 1 min,Cmax of 6.67 mg·L-1,t1/2 of 21.36 min,MRT of 30.83 min,concentration-time curve showed double peaks. Conclusion: Results suggest that t1/2 of M6 is longer than t1/2 of its prototype drug-PPTA.This HPLC is sensitive,rapid and accurate,it can be empolyed to determine concentration of M6 in rat plasma.

      摘要:Objective: To analyze the effect of Xiangsha Liujun pills on serum metabolites in rats with diabetic gastroparesis. Method: SD rats were randomly divided into normal group, diabetic gastroparesis model group, and Xiangsha Liujun pill group. The changes of metabolites in serum were detected in both groups using gas chromatography and mass spectrometry detector. PLS-DA and OPLS-DA were used respectively to compare the difference in small molecule metabolites between model group and Xiangsha Liujun pills group. Result: There were statistically significant differences in small molecule metabolites in serum between normal group, diabetic gastroparesis model group, and Xiangsha Liujun pill group. Conclusion: Xiangsha Liujun pills could change the serum metabolites of Diabetic Gastroparesis rats, and its mechanism maybe relevant to regulation effect on amino acid metabolism.

      摘要:Objective: To observe the effect of traditional Chinese herbal compound Jinlida particles (JLD) combined Tongxinluo capsule (TXL) on the ultrastructure and function of islet β-cells and the protein expressions of Beclin-1, Atg7 and p62 of pancreasin diabetic in type 2 diabetes mellitus (T2DM) mice. Method: T2DM mice were established by 6 weeks of high-fat diet combined with intraperitoneal injection of streptozotocin (STZ). Fifteen healthy male mice were set as the normal group. Another 75 T2DM mice were randomly divided into 5 groups:the model group, the JLD group (3.0 mg·kg-1·d-1), the TXL group (0.4 g·kg-1·d-1), the combination group (JT, 3.0 g·kg-1·d-1+0.4 g·kg-1·d-1) and the pioglitazone (PLZ) group (10 mg·kg-1·d-1). At the 6th week, levels of serum glucose and insulin of each group were measured and the index of β-cell function in HOMA (HOMA-β) was calculated. The organelles injury of islet β-cells was investigated by transmission electron microscopy. Beclin-1, Atg7, p62 proteins were detected by Western blotting. Result: Compared with the normal group, the injury on islet β-cells was slight, the index of β-cell function in HOMA-β was decreased, the protein expressions of Beclin-1 and Atg7 decreased and the p62 protein expression increased in the model group (P<0.01). Compared with model group, the index of β-cell function in HOMA-β was increased, the protein expressions of Beclin-1 and Atg7 increased and the p62 protein expression decreased in the treatment groups (P<0.01). Moreover, the results were better in the JT group (P<0.05). Conclusion: JLD and TXL could improve the islet β-cells function, the effect may be related to inducing islet autophagy and maintaining the pancreas level of autophagy. The later may be the crucial measures on the time delay of diabetes progression.

      摘要:Objective: To explore the effects of the leaves of Nauclea officinalis tableton on acute pharyngitis of rat models. Method: Rats were randomly divided into 6 groups,normal group,model group,aspirin group(0.20 g·kg-1),the leaves of N. officinalis tablets groups(25.8,12.9,6.5 g·kg-1).Each drug was administrated to mice orally twice a day for 3 days.Their commonly living conditions and bacterial colony were observed, the pharyngeal tissues were pathologically examined,the nitric oxide(NO) and myeloperoxidase(MPO)of the pharyngeal tissues and serum were determined,and the temperature were tested by directly spraying ammonia water and applying Staphylococcus aureus into pharyngeal mucosa. Result: The leaves of N. officinalis tablet could be decrease the content of NO and MPO in the pharyngeal tissues and serum,and also decrease the temperature;pathologic changes of rats with actue pharyngitis were improved in different degree;the leaves of N. officinalis tablets restrain the growth of the bacteria. Conclusion: The leaves of N. officinalis tablet has a good curative effect on acute pharyngitis.

      摘要:Objective: To study the protective effect of the compounds of taurine, epigallocatechin-3-gallate (EGCG) and genistein on chronic alcohol-induced hepatic fibrosis, and to explore its underlying mechanism. Method: Wistar male rats were randomly divided into six groups:normal group, model group, colchicine group, combination therapy low(taurine 12.5 mg·kg-1+EGCG 1.875 mg·kg-1+genistein 1.25 mg·kg-1),middle(taurine 25 mg·kg-1+EGCG 3.75 mg·kg-1+genistein 2.5 mg·kg-1)and high (taurine 50 mg·kg-1+EGCG 7.5 mg·kg-1+genistein 5 mg·kg-1) dose group. The rats except normal group were tablished by intragastric administration of alcohol (5.0-9.5 g·kg-1) once a day for 24 weeks and the normal group were given normal saline.Twenty-four hours after the last administration of drugs, all rats were killed, and the hepatic tissue were taken. The contents of hydroxyproline(Hyp), superoxide dismutase(SOD), malondialdehyde(MDA)in hepatic tissue and serum biochemical indicators were measured, as well as the related key cytokines/proteins. The damage of liver tissues was observed by light microscope. Result: Compared with normal group, the activities of alanine aminotransferase(ALT), aspartate aminotransferase(AST), alkaline phosphatase(ALP), gamma glutamyl transpeptidase(GGT), MDA, Hyp, hyaluronic acid(HA), laminin(LN), collagen type Ⅲ (PCⅢ), α-smooth muscle actin(α-SMA), transforming growth factor-β1(TGF-β1) and mothers against decapentaplegic homolog 3(Smad3) increased (P<0.05) and SOD, glutathione peroxidase (GSH-Px) decreased significantly(P<0.05) in model group;compared with model group, the levels of ALT, AST, ALP, GGT, MDA, Hyp, HA, LN,PCⅢ, α-SMA,TGF-β1 and Smad3 decreased(P<0.05) and SOD, GSH-Px increased significantly(P<0.05) in combination therapy group. Conclusion: The compounds of taurine, EGCG and genistein can relieve chronic hepatic fibrosis induced by alcohol. Its mechanism may involve scavenging free radica, alleviating the lipid peroxidation, inhibiting the expression of α-SMA,TGF-β1 and Smad3.

      摘要:Objective: To investigate the effect of apigenin on proliferationof human hepatocellular carcinoma Hepg2 cells and glucose uptake. Method: Human hepatocellular carcinoma Hepg2 cells were used as study objects and apigeninof different concentrations (0.625-10μmol·L-1) acted on the cells.The cells were inoculated at 6×103/mL in 96-well plate, 5 duplicate wells/group. After culture for 24, 48, 72 h, cell proliferation was detected by MTT. Glucose uptake was detected by glucose test kit. The cells were inoculated at 106/mL in 6-well plate, and after culture for 48 h, the changes in glucose transporter protein 1(GLUT1) was detected by Western blot. Result: The proliferation of human hepatocellular carcinoma Hepg2 cells could be inhibited by apigenin compared with the control group, and the inhibitory effect was in a time and concentration dependent manner, with statistically significant difference (P<0.05, P<0.01).Apigeninof different concentrations could significantly reduce the intake of glucose by Hepg2 cells (P<0.05, P<0.01). Compared with the control group, GLUT1 expression was decreased with the increase of apigenin concentration (P<0.05, P<0.01). Conclusion: Apigenin of different concentrations could inhibit the proliferation of human hepatocellular carcinoma Hepg2 cells and glucose uptake.

      摘要:Objective: To study the effect of Zhenju Jiangya tablet (ZJT) and its Chinese medicine and western medicine components on central nervous system, cardiovascular system, and respiratory system, and then to compare and search for the efficacy of synergism and attenuation. Method: ICR mice were randomly divided into ZJT low-dose, middle-dose and high-dose groups (280, 1120, 4480 mg·kg-1), ZJT Chinese medicine part (ZJTZ) low-lose, middle-dose and high-dose groups (274, 1095, 4380 mg·kg-1), ZJT western medicine part (ZJTX) low-lose, middle-dose and high-dose groups (6.2, 24.9, 99.5 mg·kg-1), normal group, and another diazepam group. Beagle dogs were divided into ZJT low-dose, middle-dose and high-dose groups (60, 240, 960 mg·kg-1), ZJTZ low-lose, middle-dose and high-dose groups (58.7, 235, 939 mg·kg-1), ZJTX low-lose, middle-dose and high-dose groups (1.3, 5.3, 21.3 mg·kg-1), and normal group. ZJT and its traditional Chinese medicine or western medicine part were respectively ig administered to ICR mice or orally administered to Beagle dogs.Spontaneous activity after the treatment in mice, and effect of synergistic subthreshold hypnotic dose of sodium pentobarbital on hypnosis were observed. Changes of systolic pressure, diastolic blood pressure, heart rate, PR interval, QRS complex, QT interval, T wave form, rhythm of heart, breathing rate and breathing depth were recorded and compared before and 0.5, 1, 2, 4, 8, 24 h after treatment in Beagle dogs. Result: Compared with the normal group, spontaneous activity was reduced in tested mice at doses of 280, 1120, 4480 mg·kg-1with ig ZJT, with synergistic hypnotic effect with subthreshold hypnotic dose of sodium pentobarbital, shortened sleep latency and prolonged sleep time(P<0.05,P<0.01). The oral administration at dose of 960 mg·kg-1in Beagle dogs reduced systolic pressure, heart rate, breathing rate and breathing depth in tested animals, where the blood pressure began to decrease at 1 h and lasted for 24 h. Prescription studies showed that oral administration of large-dose ZJT could reduce systolic blood pressure, heart rate, breathing rate and breathing depth, with significant sedation effect, mainly due to the western medicine part of the formula. Administration of ZJT could significantly enhance antihypertensive activity, prolong antihypertensive time and prevent PR interval lengthening (P<0.05,P<0.01). Conclusion: The oral administration of high-dose ZJT could decrease systolic blood pressure in tested animals whose extent and dose are positively correlated, and with a certain inhibitory effect on heart rate and respiration. In addition, ZJT has significant sedation effect. The above effect of ZJT mainly come from western medicine part of the formula, and the Chinese medicine part could enhance antihypertensive activity, prolong antihypertensive time and prevent PR interval lengthening, indicating that ZJT characterized by Chinese and western medicine compatibility has certain synergy and attenuation effect.

      摘要:Objective: To observe the effect of Huatan Tongluo formulae on expression of C/EBP homologous protein(GADD153/CHOP) and C-Jun NH2-terminal Rinases(JNK1) in apoptosis pathway of endoplasmic reticulum stress in rat reconstructive tissue plasminogen activator(rt-PA) after the acute cerebral infarction. Method: The 160 healthy male SD rats were randomly divided into sham group, model group, rt-PA group, rt-PA-Huatan Tongluo group (Chinese medicine group for short). Own embolus method was used to establish rat models of middle cerebral artery embolism (MCAO). The rats in rt-PA group were injected with rt-PA (concentration of 5.67 mg·kg-1), and those of Chinese medicine group were given with rt-PA and Chinese medicine Huatan Tongluo formulae (7.2 g·kg-1), 2 times a day. RT-PCR was used in four time phases of 6 hours, 1 day, 3, 7 days to detect the expression of GADD153/CHOP and JNK1 mRNA in cerebral cortex infarction tissues in rats. TUNEL method was used to detect brain neurons apoptosis in rats. Result: Compared with sham group, the gene expression of GADD153/CHOP and JNK1 in 4 time phases was significantly higher in model group (P<0.05). Compared with sham group within the same time phase, neurons apoptosis in model group was significantly increased (P<0.05). Compared with the model group, the expression of JNK1 mRNA was decreased in 4 time phases in both rt-PA group and Chinese medicine group (P<0.05), and rt-PA group could decrease GADD153/CHOP mRNA expression on 1 d while Chinese medicine group could decrease the expression on 6 h and 1 d, with significant difference (P<0.05). Neurons apoptosis in rt-PA group on 1 and 7 d was significantly lower than that in the model group (P<0.05), while Chinese medicine group decreased neurons apoptosis in various time phases (P<0.05). However, there was no significant difference between rt-PA group and Chinese medicine group. Conclusion: Huatan Tongluo formulae can prevent and treat partially reperfusion injury after acute cerebra accepted thrombolysis by reducing GADD153/CHOP and JNK1 mRNA expression in apoptosis pathway of endoplasmic reticulum stress, and partially inhibiting the apoptosis of nerve cells.

      摘要:Objective: To evaluate the inhibitory activity of α-glucosidase from the endophytes of Tripterygium hypoglaucum. Method: With 98 endophytes isolated from T. hypoglaucum as the research objects and acarbose as positive medicine, α-glucosidase in vitro screening models were established to conduct activity screening of n-butanol extracts from 98 endophytes fermentation liquor of T. hypoglaucum. The active strain ThF-63 was identified based on fungal ITS sequence. Result: In the substrate, under the conditions where 4-ntropheny-α-D-glucopy-ranoside(PNPG) concentration was of 2.5 mmol·L-1, enzyme concentration of 0.8 U·mL-1 and response time of 15 min, n-butanol extracts from 15 strains of 98 endophytes showed significant α-glucosidase inhibitory activity (IC50<30 g·L-1). ThF-63 was one of the most significant strain against α-glucosidase (IC50 1.034 g·L-1), with concentration dependence relations. The endophyte ThF-63 was identified as Alternaria by ITS method. Conclusion: Alternaria with α-glucosidase inhibitory activity were screened from the endophytes of T. hypoglaucum.

      摘要:Objective: To establish atherosclerosis rabbit models, study the effect of Danlou tablet on atherosclerosis and the expression of endoplasmic reticulum stress (ERS) related genes immunoglobulin heavy chain binding protein (BIP), and provide experimental basis for clinical application of Danlou tablet. Method: The 24 male big-eared Japanese rabbits were randomly divided into normal group, model group, Rosuvastatin group, Danlou tablet+Rosuvastatin group, with 6 rabbits in each group. Normal group was fed with standard diet, while in model group, 2% cholesterol and 0.02% methionine were added in standard diet.In Rosuvastatin group and Danlou tablet+Rosuvastatin group, 2% cholesterol, 0.02% methionine and the corresponding control drug were added in the standard diet to establish atherosclerosis rabbit models. Before medication and after 9 weeks of treatment, cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), apolipoprotein A (ApoA) and apolipoprotein B (ApoB) values were detected. After 9 weeks of treatment, thoracic aorta segment was taken from the experiment animals to observe morphological changes by HE. In situ hybridization was used to detect BIP gene expression. Result: Compared with the normal group, TC, TG, LDL-C and ApoB levels in model group were significantly higher, while HDL-C and ApoA levels were significantly lower (P<0.05). Compared with the model group, TC, TG, LDL-C and ApoB levels in Rosuvastatin group and Danlou tablet+Rosuvastatin group were significantly lower, while HDL-C and ApoA levels were significantly higher (P<0.01). HE staining showed normal endangium morphology in normal group, severe hyperplasia of endometrium and endothelial cell damages in model group, mild hyperplasia of endometrium in Rosuvastatin group, and close to normal endangium morphology in Danlou tablet+Rosuvastatin group. In situ hybridization results showed that BIP protein expression was significantly increased in model group, and Rosuvastatin group and Danlou tablet+Rosuvastatin group could down-regulate BIP protein expressions. Conclusion: Danlou tablet combined with Rosuvastatin can delay the development of atherosclerosis and have significant therapeutic effect.

      摘要:Objective: To observe the synergistic antitumor effect and possible mechanism of extract from Periplaneta americana(EPA) and cyclophosphamide (CTX) on mice bearing-H22. Method: KM mice were inoculated subcutaneously with H22 cells to form solid tumor models and then were treated for 10 days with high dose, middle dose and low dose (50, 100, 200 mg·kg-1) EPA, 30 mg·kg-1 CTX or received combination therapy respectively. 10 days later, the tumor weight, liver index, and spleen index were measured;and the levels of malondialdehyde (MDA) and superoxide dismutase (SOD) in liver tissues were measured to detect the antioxidant activity of drugs;the contents of tumor necrosis factor-α (TNF-α), interleukin 2(IL-2), and the number of white blood cells were detected to evaluate the synergistic antitumor effect of EPA and CTX. Result: Compared with the model group, all the treatment groups could inhibit the tumor growth (P<0.05), most significantly in combination group of 50 mg·kg-1 and CTX, with the tumor inhibitory rate up to 61.96%. EPA alone and combination of EPA and CTX could increase SOD level in liver tissues, reduce MDA level, and significantly increase serum TNF-α and IL-2 levels (P<0.05, P<0.01). The various doses of EPA could improve the spleen index and thymus index of tumor-bearing mice, but combination groups had no significant effect on the organs of tumor-bearing mice, but could enhance the amount of white blood cells in the peripheral blood. Conclusion: Combined application of EPA and CTX has significant antitumor effect in mice bearing-H22, and the mechanism may be associated with antioxidant activity and enhanced immunity.

      摘要:Objective: To investigate the effect of Buyang Huanwu Tang(BYHWT) on the expression of phosphatase and tensin homolog deleted on chromosome ten (PTEN) mRNA and platelet activation in rat models with cerebral ischemia-reperfusion injury. Method: Sixty specific pathogen free (SPF) Sprague-Dawley rats (weighing 250-300 g) were randomly divided into 5 groups as following:sham-operated group, clopidogrel group, high-dose BYHWT group, low-dose BYHWT group, and model group, 12 rats in each group. The high-dose BYHWT group and the low-dose BYHWT group were ig administered with 26, 6.5 g·kg-1·d-1 BYHWT respectively while the clopidogrel group was ig administered with 6.8 mg·kg-1·d-1. Another two groups were ig administered with normal saline. After treatment of the animals for 14 days,middle cerebral artery occlusion method was used to establish models of transient focal cerebral ischemia and reperfusion, and corresponding ig treatment was provided 2 h before modeling. The plugging wire was removed after modeling for 60 min and reperfusion for 12 h before samples collection for determination of the content of P-selection(CD62P) in plasma and expression of PTEN mRNA in hippocampus. Result: The neurological score in model group was significantly higher than that in sham-operated group, and the neurological scores in various treatment groups were significantly lower than that in model group.The expression of CD62P in the model group was significantly higher than that in sham-operated group (P<0.05). BYHWT groups and clopidogrel group can significantly inhibit the expression of CD62P (P<0.05), but without significant difference between BYHWT groups and clopidogrel group. PTEN mRNA expression in model group was significantly higher than that in sham-operated group. PTEN mRNA expression in various treatment groups were significantly lower than that in model group (P<0.05), mostly significant in BYHWT high dose group. Conclusion: The neuroprotective mechanism of BYHWT on acute cerebral ischemia model may relate with down-regulating the expression of PTEN gene over-expression and inhibiting expression of CD62P.

      摘要:Objective: To compare the effects and possible mechanism of Siwu Tong(SWT), Sijunzi Tang(SJZT), and Bazhen Tang(BZT) in anemia (blood deficiency syndrome) rats induced by 5-fluorouracil (5-FU) and preliminarily explore the effect difference and action mechanism of blood replenishing prescription, qi tonifying prescription as well as blood and Qi double replenishing prescription. Method: SD rats were randomly divided into normal group, model group, Bazhen Tang group, Siwu Tang group and Sijunzi Tang group (2.3, 3.0, 1.2 g·mL-1), 8 rats for each group. Rats in other 4 groups except normal group were modeled by intravenous injection at tail at 150 mg·kg-1 5-FU. Effect of the three prescriptions on appearance of anemia rats, spleen index and thymus index, red blood cells (RBC), hemoglobin (HGB), platelet (PLT), white blood cells (WBC) count, reticulocyte and myelogram was observed, and reverse transcriptase-polymerase chain reaction (RT-PCR) was applied for determining the expression of EPO mRNA in kidney and liver. Result: Compared with normal group, spleen index and thymus index were significantly reduced in model group, RBC, HGB, PLT, and WBC counts were significantly reduced (P<0.01), bone marrow proliferation ability was reduced, and EPO mRNA expression was reduced. Compared with the model group, BZT group and SWT group could significantly improve the appearance of anemia rats, increase spleen index and thymus index, increase RBC, HGB, PLT and WBC counts (P<0.05, P<0.01), improve the ability of bone marrow proliferation, and up-regulate EPO mRNA expressions. Conclusion: Blood and Qi double replenishing prescription (Bazhen Tang) among these three prescriptions has the best curative effect. Blood replenishing prescription (Siwu Tang) has better effect than Qi tonifying group (Sijunzi Tang). The experimental results provide experimental basis for the treatment of blood deficiency diseases by Qi tonifying therapy, blood replenishing therapy as well as blood and Qi double replenishing therapy, illustrating the rationality of dialectical theory in clinical treatment of anemia (blood deficiency syndrome) to some extent.

      摘要:Objective: To observe the effect of Tonglian decoction (TD) on histomorphology of transplanted tumor and tumor necrosis factor-α (TNF-α), interleukin(IL)-6, and IL-8 in peripheral blood of nude mice inoculated with gastric cancer MGC803 cells. Method: All mice were inoculated subcutaneously with MGC803 cells in the right forelimbs and were randomly divided into the following groups,the model group (saline solution, 10 mL·kg-1), the Xiaoaiping group (1.8 g·kg-1) and high-, middle-, low-dose TD groups (1.5, 0.75, 0.37 g·kg-1). After 3-week administration, they were observed for another 3 weeks continuously. The hematoxylin and eosin (HE) stain of the tumor tissue was made followed the observation on micro-structure by fluorescence microscope. The concentrations of serum TNF-α, IL-6 and IL-8 were examined by enzyme linked immunosorbent assay (ELISA) method. Result: In the high-dose TD group, HE pathological changes showed large necrotic area in slice center, accompanied with stained tumor cells in light color and few cellular nuclear division. The high-dose TD could down-regulate the contents of TNF-α, IL-6 and IL-8 in peripheral serum of nude mice with the values of (33.82±14.43), (45.49±15.61),(132.09±29.51) ng·L-1, respectively, showing statistical difference when compared with the model group (P<0.05, P<0.01). Conclusion: High-dose TD could promote the tumor necrosis of mice inoculated with MGC803 cells. The mechanism may be achieved by down-regulating the contents of TNF-α, IL-6 and IL-8 in peripheral serum and strengthening the immune ability.

      摘要:Objective: To observe the effect of Xianyuquan granules (JYQG) on the contents of aminobutyric acid (GABA) and glutamic acid (Glu) in hippocampus of rats with epilepsy caused by pentetrazol, and explore the mechanism of JYQG in treatment of epilepsy. Method: Seventy healthy male SD rats were randomized into normal group, model group, valproic acid (VPA) group (200 mg·kg-1), low-dose and high-dose JYQG groups (586, 1172 mg·kg-1). Subconvulsant dose of pentetrazol (PTZ) was ip administered to establish epileptic rat models.The others were administrated with their medicine respectively for twenty-eight days. Hippocampus tissues were taken after behavior observation. The contents of GABA and Glu in hippocampus of the rats were measured on HPLC-MS/MSQ-TRAP. Result: Fifty-nine rats were analyzed finally. Compared with the model group, the incubation period of the VPA treated group and JYQG treated group was prolonged and the convulsion duration was shorter (P<0.01), convulsion duration in high dose group was shorter than that in low dose group (P<0.05). Glu content in the hippocampus of model group was significantly higher than that of the normal group and the GABA content was significantly less than that of the normal group (P<0.01). Glu content in rats administrated with corresponding medicines was significantly less than that of model group while GABA content was higher than that of model group (P<0.05, P<0.01). Conclusion: The application of subconvulsant-dose pentetrazol can establish the epileptic model. The anti-epileptic effect of JYQG may be associated with decreasing the content of Glu and increasing the content of GABA in the hippocampus.

      摘要:Objective: To observe the intervention effect of ethyl acetate extract of Punica granatum fruit (PGFEAE) on blood glucose and blood lipid in rats with insulin resistance (IR) and normal rats, and evaluate its pharmacological effects on improving insulin resistance. Method: High-sugar and high-fat diet plus intraperitoneal injection of streptozocin(STZ) twice (30 mg·kg-1) to establish rat models of insulin resistance. After 4 weeks of ig administering 100, 200, 300 mg·kg-1 PGFEAE respectively, fasting blood glucose (FBG), fasting insulin (FINS), insulin sensitivity index (ISI), serum free fatty acid (FFA) concentration, and triglyceride (TG) concentration were measured. Result: Compared with the normal group, FBG, FINS, FFA, and TG levels were significantly higher and ISI level was lower in model group (P<0.05,P<0.01), after 4 weeks of modeling, FBG, FINS, and TG levels were 5.94, 30.71, 0.57 mmol·L-1. 13.05, 42.36, 1.62 mmol·L-1 respectively in normal group and model group. After 4 weeks of treatment, various dose groups of PGFEAE can effectively reduce FBG, FINS, FFA, and TG levels and increase ISI level in rats with insulin resistance compared with the model group (P<0.05, P<0.01). Conclusion: PGFEAE can effectively improve the symptoms of high blood glucose and blood lipid in rats with insulin resistance.

      摘要:Objective: To investigate the clinical effect of Angong Niuhuang Wan plus Fengyin decoction in treating severe hand-foot-mouth disease (terrific heat and dynamic wind type) as well as its influence on serum S-100β protein (S-100β) and brain batriuretic peptide (BNP). Method: One hundred and forty-two patients with severe hand-foot-and-mouth disease were randomly divided into control group and treatment group, with 71 cases in each group according to hospital admission sequence. Control group received routine treatment and symptomatic treatment referring to treatment guide of hand-foot-and-mouth disease (2010 year). Based on the treatment in control group, patients in treatment group also received retention enema of Angong Niuhuang Wan (1 wan/time, 1/4 wan/time for child of 3 years old,and 1/2 wan/time for child of 4-6 years old), 1 time/day;combined with oral administration or nasal feeding of Fengyin decoction (1 dose/d and bid in convention decoction form, 50 mL/time for child of 3-5 years old, 10-40 mL/time for child<3 years old, 3-5 times/d). Course of treatment was 10 days in both groups. Clinical symptoms and signs were observed and length of stay was recorded for two groups. PaO2, SPO2, S-100β, and BNP were detected in both groups. Result: The total efficacy of treatment group was 88.73%, which was superior to 73.23% in control group (P<0.05). Extinction time of erythra, bleb, skittishness and limb tremor, as well as temperature recovery time and length of stay in treatment group were superior to control group (P<0.01). PaO2 and SPO2 of treatment group were significantly higher than those of control group (P<0.01). After treatment, serum S-100β and BNP in treatment group were significantly lower than those in control group with statistical difference (P<0.01). Conclusion: Based on western medicine conventional therapy, Angong Niuhuang Wan combined with Fengyin decoction could relieve clinical symptoms and signs of severe hand-foot-mouth disease (terrific heat and dynamic wind type), improve oxygen saturation of blood and partial pressure of oxygen, and shorten length of stay. Reduction of serum S-100β and BNP levels may be involved in above-mentioned regulation.

      摘要:Objective: To observe the clinical efficacy of Jiannao Bushen pills combined with Ginkgo Leaves tablet (GLT) in treating mild cognitive impairment (MCI) and its influence on oxidative stress and acetylcholinesterase (AChE). Method: Sixty cases MCI patients were randomly divided into the control group and treatment group according to the method of random digits table, 30 patients for each group. Besides, another 30 healthy volunteers were selected as the normal group. The control group received Donepezil hydrochloride tablets, 2.5 mg·d-1after meals. The treatment group received Jiannao Bushen pills(15 pieces/time, 2 times/d) combined with GLT (2 tablets/time, 3 times/d), with the treatment course of 3 months for both groups. Changes of scores of all indexes of clinical memory scale and memory quotient (MQ) before and after treatment were observed for both groups, and clinical therapeutic effect was evaluated by using MQ increasing values. Change of malondialdehyde (MDA), superoxide dismutase (SOD), and AChE levels before and after treatment were observed. Result: All indexes of clinical memory scale and MQ in MIC patients were significantly lower than those in normal group (volunteers) (P<0.01). After treatment, the satisfaction rate was 46.67% in the control group and 76.67% in the treatment group, with significant difference between the two groups (P<0.05). After treatment, the scores of all indexes of clinical memory scale of two groups except associated memory were significantly increased (P<0.05, P<0.01) in both groups, while the scores in direction memory, picture free memory, associated memory of human picture characteristics, and MQ in the treatment group were higher than those in the control group (P<0.05). Serum MDA and AChE levels were decreased and SOD was increased in both groups after treatment (P<0.05, P<0.01). Serum MDA and AChE levels in the treatment group were lower than those in control group after treatment, while SOD level in the treatment group was higher than that in the control group after treatment (P<0.05). After treatment, there was no statistical difference in MDA, AChE, and SOD levels between treatment group and normal group. Conclusion: Jiannao Bushen pils combined with GLT can effectively improve the memorial function of MCI patients, and its therapeutic mechanism may be associated with alleviating the damage of oxygen free radicals and depressing the activity of AChE.

      摘要:Objective: To observe the clinical efficacy of modified Sanren Tang in rescue treatment after failed eradication of helicobacter pylori (Hp) and its effect on nuclear factor kappa B (NF-κB) and heat shock protein70(HSP70). Method: One hundred and thirty-five patients with previous failed eradication of Hp were randomly divided into control group (67 cases) and treatment group (68 cases) according to visiting sequence. Control group received quadruple regimen treatment, including Colloidal bismuth pectincapsule 400 mg (bid), orally taken after meals.Sodium Rabeprazole Enteric-coated Tablets, 10 mg (bid), orally taken before meals.furazolidone tablets 100 mg (bid), and amoxicillin capsule 1000 mg (bid). Based on control group, the treatment group also received modified Sanren Tang treatment, 1 dose/d and bid, in conventional decoction form in the morning and evening. The treatment course was 10 days for both groups. Hp eradication rate was detected in both groups. Scores of traditional Chinese medicine (TCM) syndrome and clinical efficacy were compared between two groups. Serum levels of NF-κB and HSP70 were measured in both groups. Result: Hp eradication rate was 87.69% in treatment group, which was superior to 72.31% in control group (P<0.05). The total effective rate was 93.85% in treatment group and 80% in control group, and there was statistical difference between two groups (P<0.05). After treatment, scores of epigastric pain, epigastric distention, epigastric burning, fetid oris, nausea and vomiting as well as the total scores in treatment group were obviously lower than those of control group (P<0.01). After treatment, serum level of NF-κB in treatment group was significantly lower than that in control group, while HSP70 was significantly higher than that in control group, with statistically significant difference between two groups (P<0.01). Conclusion: Based on quadruple regimen treatment, modified Sanren Tang in treatment of failed eradication of Hp could obviously enhance Hp eradication rate and clinical efficacy, improve TCM syndrome, and its mechanism may be associated with decreased NF-κB level and increased HSP70 level.

      摘要:Objective: To observe the clinical efficacy of Yougui wan in treating spleen and kidney deficiency type chronic functional constipation (CFC) and its influence on superoxide dismutase (SOD) and malondialdehyde (MDA) levels. Method: One hundred and twenty CFC patients were randomly divided into control group (60 cases) and treatment group (60 cases) according to random number table. Patients of control group were treated according to Guideline of Diagnosis and Treatment of Chronic Constipation in China, including adjustment of life-style, cathartic of phenolphthalein tablet (100 mg/time, bid), and impetus drug of domperidone tablets (1 tablet/time, tid, taking ante cibum 30 min). Based on the treatment in control group, patients in treatment group also received Yougui Wan (1 dose/d, tid). Course of treatment was 4 weeks for both groups. Main CFC symptoms were assessed before and after treatment between both groups. Quality of life for two groups was evaluated according to short-form 36 health survey scale (SF-36). Serum SOD and MDA levels were measured in all cases pre-post treatment. Result: The total clinical efficacy of treatment group was 98.33%, which was superior to 85% in control group, with statistical difference (P<0.01). After treatment, CFC symptom scores in treatment group were lower than those in control group (P<0.01). Score in each item of SF-36 in treatment group was significantly higher than that of control group after treatment (P<0.05). After treatment, serum SOD level in treatment group was significantly higher than that of control group while MDA level was lower than that in control group with statistical difference (P<0.01). Conclusion: Based on control group, Yougui Wan may improve clinical symptoms and clinical efficacy in treating spleen and kidney deficiency type CFC, and its effect may be relevant to rising SOD level and reducing MDA levels in serum.

      摘要:Objective: To observe the effect of addition and subtraction Dihuang Yinzi on language function of aphasia from apoplexy with syndrome of both liver and kidney deficiency. Method: Thirty patients were randomly divided into control group and treatment group by random number table method, with 15 cases in each group. The conventional therapy was given to all patients in both groups. Patients in control group were treated with Schuell language training, 30 min/time, 3 times/week. Based on the treatment in control group, the patients in treatment group also received addition and subtraction Dihuang Yinzi treatment, 1 dose/d, tid with warm water. The treatment course was 2 months for all cases. Speaking function of the patients in two groups was evaluated by China rehabilitation research center aphasia examination (CRRCAE), Chinese functional communication profile (CFCP), aphasia quotient (AQ), and Boston diagnostic aphasia examination (BDAE). Result: The markedly clinical effective rate was 73.33% in treatment group, which was significantly superior to 26.67% in control group (P<0.05). After treatment, score in each item of CRRCAE in treatment group was significantly higher than that in control group (P<0.01). Scores of CFCP and AQ were higher than those in control group after treatment with statistical significance (P<0.01). After treatment, BDAE grading in treatment group was superior to that in control group (P<0.01). Conclusion: On the basis of the conventional therapy in control group, addition and subtraction Dihuang Yinzi plus Schuell's language training in treatment group improves language function of patients with aphasia from apoplexy with syndrome of both liver and kidney deficiency, increases scores of CRRCAE, CFCP,and AQ, and decreases BDAE score.

      摘要:Objective: To investigate the effect of Huangqi injection on oxidative stress and inflammatory response in patients with viral myocarditis. Method: One hundred and seventy-eight patients with viral myocarditis hospitalized in our cardiology department from May 2011 to December 2013 were selected. Cases in conventional group were given with comprehensive western medicine treatment, while the cases in combined group were given with Huangqi injection intravenously on the basis of the conventional group. 12 h after hospitalization and two courses of treatment, the levels of superoxide dismutase (SOD), malondialdehyde (MDA), glutathione S-transferase (GST) and nitric oxide (NO), as well as the levels of interleukin-6(IL-6), interleukin-8(IL-8), tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ) were detected. Result: The total effective rate of combined group was 88.8%, much higher than 71.9% of conventional group. The difference was statistically significant (P<0.05). After treatment, SOD and GST in the two groups were (123.6±12.4) U·mL-1 and (20.4±5.7) U·mL-1, which were increased than those before treatment, while MDA and NO were (3.3±0.2) μmol·L-1 and (68.4±10.9) mmol·L-1 which were reduced. The differences were statistically significant (P<0.05). After treatment, IL-6, IL-8, TNF-α and IFN-γ levels were higher than those before treatment in both groups (P<0.05). After treatment, IL-6, IL-8, TNF-α and IFN-γ levels were (43.5±20.3), (16.8±1.9), (8.9±1.5), (273.8±79.6) ng·L-1respectively, lower than those in convention group. The differences were statistically significant (P<0.05). Conclusion: Huangqi injection could effectively improve the clinical treatment for viral myocarditis, reduce the effects of oxidative stress and inflammatory response on myocardial cells injury.

      摘要:Objective: To probe into the clinical application value of Reduning injection combined with salmeterol and fluticasone propionate inhalation powder for patients with bronchial asthma. Method: From January 2013 to June 2014, 110 patients with bronchial asthma were selected in our hospital. The patients were divided into treatment group (n=55) and control group (n=55) with random number table. The patients with bronchial asthma in control group were treated salmeterol and fluticasone propionate inhalation powder, 1 suction/time, 2 times/d, 14 d/treatment. The patients with bronchial asthma in treatment group were treated combined with Redunning injection therapy based on control group, 20 mL/second, 1/d, 14 d/treatment. The clinical treatment, changes of pulmonary function, expression of inflammatory cytokines, cell-mediated immunity factor of the patients in two groups were compared and analyzed. Result: Compared with control group, the patients in treatment group, efficiency ratio was 47.27% (26/55) and total efficiency ratio was 92.73% (51/55), there were increased significantly, the differences were significant statistically (P<0.05). Compared with before treatment, the patients in two groups after treatment, the indexes of lung function, included forced vital capacity (FVC), the first second forced expiratory volume (FEV1) and forced expiratory flow rate (FEF) were increased significantly, the differences were significant statistically (P<0.05). Compared with control group, the patients in treatment group after treatment, the indexes of lung function, included FVC was (3.01±0.17) L, FEV1 was (2.86±0.51) L and PEF (4.96±1.10) L·s-1, there were increased significantly, the differences were significant statistically (P<0.05). Compared with before treatment, the patients in two groups after treatment, the levels of inflammatory cytokines interferon-γ(IFN-γ) were increased significantly, and the levels of interleukin-4(IL-4)were decreased significantly, the differences were significant statistically (P<0.05). Compared with control group, the patients in treatment group after treatment, the level of inflammatory cytokines IFN-γ was (21.69±3.14) ng·L-1, there was increased significantly, and the level of IL-4 was (8.97±1.67) ng·L-1, there was decreased significantly, the differences were significant statistically (P<0.05). Conclusion: Reduning injection combined with salmeterol and fluticasone propionate powder has very important significance for improving the clinical effect, lung function and inflammatory cytokines expression of patients with bronchial asthma.

      摘要:Objective: To observe the effect of Kushen injection combined with GP regimen on the clinical effect, quality of life and immune function of patients with non-small cell lung cancer. Method: From January 2013 to December 2014, 120 cases of non small lung cancer patients treated by respiratory department of internal medicine were selected. The patients were randomly divided into control group and experiment group with random number table method. Control group (gemcitabine+cisplatin) received GP regimen chemotherapy. Injection of gemcitabine was dissolved in 250 mL normal saline at a proportion of 1000 mg·m-3on day 1 and day 8 respectively,with intravenous drip for 30 min. Cisplatinswas was administered by intravenous drip at 25 mg·m-3 on day 2, day 3 and day 4.In the experimental group, compound Kushen injection was also used in addition to GP regimen, 20 mL, once a day, for continuous 10 days, and with 21 days as 1 cycle. After 2 cycles, short-term curative effect, life quality and immune function were compared between two groups. Result: In the experimental group, the total effective rate was 75.0%, higher than 56.67% in control group, with statistically significant difference (P<0.05). The effective rate of life quality in the experimental group was higher than that of the control group. CD3+, CD4+, CD4+/CD8+levels in the control group were lower than those before treatment, while CD3+, CD4+, CD4+/CD8+levels in the experimental group were hihjer than those before treatment, with statistically significant difference (P<0.05). The change of NK cells before and after treatment was not statistically significant in two groups. IgG, IgM, IgA in patients in the control group were lower than those, before treatment, while immunoglobulin in the experimental group were higher than those before treatment, with statistically significant difference (P<0.05). Conclusion: Kushen injection combined with GP regimen has clear clinical efficacy for patients with non-small cell lung cancer. It can improve the quality of life and improve their immunity,for patients with lung cancer. It is worthy of clinical application.

      摘要:Objective: To summarize Professor FAN Rui-hong's experience of Tong-bu method on the treatment of chronic heart failure. To observe the clinical thereapeutic effect and safety on chronic heart failure(CHF) treated with self-made Tongbu Xinbao decoction of traditional Chinese medicine(TCM) of having the efficacy of Tong-bu. Method: Totally 76 cases of chronic heart failure were randomized into treatment group of 38 cases and control group of 38 cases.The control group was given routine western therapy and the treatment group was added with Tongbu Xinbao decoction of TCM.One treatment course lasted for 28 days.The heart function and the Chinese medicine syndrome scores and left ventricular ejection fraction and the distance in 6 minutes walking test and N-terminal pro-brain natriuretic peptide (NT-proBNP) were observed in both groups before and after treatment.The side effect was observed. Result: After treatment, the total effective rate of the heart function of the treatment group and the control group was 78.95% and 60.53% respectively, with significant difference between groups (P<0.05).The total effective rate of the Chinese medicine syndrome scores was significantly higher in treatment group (81.58%) than in control group (55.26%, P<0.01). The treatment group was better than the control group with improving the left ventricular ejection fraction and the distance in 6 minutes walking test and decreasing the level of NT-proBNP(P<0.05). There was no serious adverse event in two groups. Conclusion: The effect of Tong-bu method on the treatment of chronic heart failure is satisfaction, the treatment of integrated Tongbu Xinbao decoction of TCM and western medicine on chronic heart failure is superior to routine western therapy.

      摘要:Objective: To explore the Xihuang capsule in combination with radiotherapy clinical outcomes in patients with nasopharyngeal carcinoma. Method: From April 2012 to April 2014 in our hospital 80 patients with nasopharyngeal carcinoma were analyzed data, the patients were randomly divided into a control group method and the experimental group and the control group using a single radiation treatment, the experimental group combined with the control group on the basis of Xihuang capsule treatment were compared treatment. Result: 55% of the treatment group, the results are satisfactory, the control group (42.5%) (P<0.05). The experimental group was 82.5% higher than clinical benefit (P<0.05). two groups of patients before treatment serum T cell subsets group level, such as:indexes were CD3+, CD4+, CD8+ and CD4+/CD8+, which are not significant. The experimental group after treatment CD3+, CD4+, CD8+ and CD4+/CD8+ other indicators significantly lower than the control group (P<0.05). The adverse reactions in the experimental group was 32.5%, lower than in the control group (42.5%) (P<0.05), ADL score in the experimental group, physical function score, mental function scores, social function scores were higher (P<0.05). Conclusion: Capsule NPC patients in radiotherapy combined with western treatment based on the desired therapeutic effect, can improve clinical outcomes and reduce the incidence of adverse reactions, should be introduced.

      摘要:Objective: To analyze the composition principles of prescriptions for emesis in Prescription of Traditional Chinese Medicine Dictionary. Method: The prescriptions for emesis in Prescription of Traditional Chinese Medicine Dictionary were selected, and traditional Chinese medicine inheritance support system (TCMISS) V2.5 was used to enter information and build the database. The commonly used herbs, combination principles, core combinations and new combinations in the prescriptions for emesis were analyzed with data mining methods, such as association rules, improved mutual information method, complex system entropy clustering and unsupervised hierarchical clustering. Result: Total 174 herbs were involved in the 305 prescriptions for emesis, including, 49 herbs with the frequency of more than 10 times, forty-five commonly used herb pairs and combinations, fifty-eight potential core combinations and nine newly developed prescriptions. Conclusion: The composition principles of prescriptions for emesis in Prescription of Traditional Chinese Medicine Dictionary are defined,which provides references for clinical medication and new drug development. The commonly used herbs are mainly citrus, pinellia, ginger and some tonics.

      摘要:Objective: To analyze the composing principles of herbs in the prescriptions for treating polycystic ovary syndrome by using Traditional Chinese Medicine Inheritance System, and make predictions of new formulations according to the composing principles. Method: The literatures on prescriptions for experimentally treating polycystic ovary syndrome in China national knowledge infrastructure (CNKI) were collected and a database was set up, then traditional Chinese medicine (TCM) inheritance system was used to analyze the composing principles of herbs in the prescriptions. Result: The 166 literatures on the prescriptions in the database were screened to achieve 38 commonly used herbs, 11 common composing principles, 22 core combinations of new prescriptions and 11 new prescriptions. Conclusion: Composing principles of herbs in the prescriptions for treating polycystic ovary syndrome be showed by TCM inheritance system.Analysis shows that the drugs for polycystic ovary syndrome are mainly kidney invigorating, blood circulating and phlegm removing drugs. The new prescriptions obtained by cluster analysis have similar treatment effect with the common prescriptions such as Guizhi Fuling capsule, Guishao Dihuang Tang combined with Yueju Wan, Jiawei Daotan decoction for polycystic ovary syndrome, but the new prescriptions also have spleen invigorating and liver soothing herbs to achieve spleen and liver regulating effect. Its compatibility combinations conform to the main treatment ideas of kidney invigorating, blood circulating and phlegm removing, helpful to guide clinical practice, and provide certain basis for clinical development and use of new drugs.

      摘要:Objective: To evaluate the effectiveness of Qingyi decoction in the treatment of severe acute panereatitis in a systematic review. Method: The inclusion and exclusion criteria and retrieval strategies were made using Meta-analysis methods, with computer retrieval of CNKI (1979-2014), Wanfang Data Knowledge Service Platform (1990-2014), Chinese Biomedical Database (1978-2014), pubmed (1966-2014) and other Chinese and English databases.The quality of the literatures was assessed according to Cochrane review criteria. Result: The 19 studies were included. Compared with control group, the adjunctive therapy with Qingyi decoction group had significant differences in the time of first defecation[WMD=-2.098, 95%CI(-2.739,-1.456), P<0.01)], the time of stomach ache relief[WMD=-3.008, 95%CI(-3.689,-2.326), P<0.01], hospital stay (days)[WMD=-7.633, 95%CI(-8.449,-6.756), P<0.01], case fatality rate[OR=2.719, 95%CI (1.717, 4.306), P<0.01]. Conclusion: The adjunctive therapy with Qingyi decoction in severe acute pancreatitis can significantly decrease the case fatality rate, shorten the first time of defecation, stomachache relief time and hospital stay.

      摘要:Objective: To analyze and excavate the medication rule in herbal prescriptions for diabetes by using Traditional Chinese Medicine in Heritance Support System (TCMISS). Method: Herbal prescriptions for diabetes in recent 10 years were collected from papers, and Apriori algorithm was used to analyze correlation between herbs, which was based on improved mutual information method. And according to the relevant constraints and punishment degree, core combination of herbs was analyzed by complex system entropy clustering method based on the correlation analysis between drugs. New prescriptions were acquired by using unsupervised clustering algorithm. The network diagrams of medicine association rules and core combination herbs in the new prescriptions were drawn based on the analysis results. The above operations were completed besed on TCMISS V1.1. Result: The frequency of each herb and the association rules among the herbs were found out, 16 core combinations and 8 new prescriptions were excavated from the database about the analysis of 110 medicine prescriptions of diabetes. Conclusion: The data mining method based on Apriori algorithm and cluster analysis is suitable for mining, analysis and summary of the medication rules in herbal prescriptions. Herbs of nourishing qi and yin, combined with herbs for heat-clearing and activating blood circulation in accordance with different types is important to improve the curative effect on diabetes.

      摘要:Chronic pelvic inflammatory disease is a common disease in gynecology department, especially among middle-aged females. The lesions are mainly related to the female internal genital organs, pelvic peritoneum and connective tissues. The long duration of treatment and easily migrated and recurrent symptoms are the main features of the disease. The inflammatory exudates accumulate in the pelvic cavity due to recurrence of chronic pelvic inflammatory disease, so pelvic fluid effusion is also common in the patients of this disease, which increases the difficulty of treatment. Classical prescription has a thousand history of traditional Chinese medicine, characterized by refined medication, direct to lesions and curative effect, with preferred effect in the treatment of internal medicine and stubborn diseases. This paper would systemically analyze the application value of classical prescription in the treatment of chronic pelvic inflammatory disease combined with pelvic fluid effusion. 1 specific clinical case would be used for analysis, to give inspiration for the treatment of such diseases.

      摘要:Acanthopanacis Senticosi Radix et Rhizoma Seu Caulis is a frequently used herb, which contain saponins, flavonoids, polysaccharides, phenolic acids and other chemical components, and protective cardiovascular,antitumor, anti-inflammatory, antioxidant, blood sugar, such as a wide range of pharmacological effects. Clinical commonly used parts for root and rhizome part, but as a result of resource irrefragable, studying the reproducibility of Acanthopanacis Senticosi Radix et Rhizoma Seu Caulis leaf has the vital significance. On the basis of literature review,this paper about the pharmacological action, mechanism of action and therapeutic effects of material basis of Acanthopanacis Senticosi Radix et Rhizoma Seu Caulis at home and abroad in recent years leaf were summarized,finding leaves of pharmacological effect research main concentrated in saponins, but other chemical components research reported less,and Acanthopanacis Senticosi Radix et Rhizoma Seu Caulis leaf can instead of acanthopanax root and rhizome applied to clinical, how to establish quality standards, these problems limited the application of acanthopanax leaf resources. In order to more comprehensive and scientific development and utilization of acanthopanax leaf resources, has yet to be further research.

      摘要:High methylation of DNA is closely related to occurrence of tumor.Reversing aberrant methylation of DNA of tumor cells,restoring expression of tumor suppressor gene or DNA repair gene become new hot spots in development and research of anti-tumor drugs.Compared to single target point and resistance of chemical drugs,traditional Chinese medicine (TCM) has advantages of comprehensive and multiple targets for treatment.However,there are few reports on effect of TCM on DNA demethylation.In this paper,anti-tumor TCM and composition in TCM on DNA demethylation are combed.All these TCM were classified into four types based on their efficacy,including heat-clearing,activating blood circulation to remove stasis,anti-rheumatism and others.Demethylation effects of these Chinese herbs on different target genes in different tumors were described,and prospects of this study were also discussed.

      摘要:Fermentation is a distinctive and important processing technology of traditional Chinese medicine (TCM),its inheritance and innovation should be highly regarded within the industry.Traditional fermentation of TCM is a natural fermentation,due to processing is controlled by experience and is vulnerable to changes of environment and season,so it is difficult to control quality and inherit.Modern fermentation technology of TCM,which based on traditional fermentation technology of TCM,combined with fermentation engineering and other modern biotechnology,has been gradually developed,and scholars have carried out some related research.However,currently some common problems are faced in research of fermentation technology of TCM,such as scientific interpretation of fermentation mechanism,regulation fermentation processing,establishing quality standards suitable for fermentation characteristics of TCM and so on.This paper is based on review of traditional and modern research on fermentation technology of TCM,analyzing and summarizing key technical problems impacting inheritance and development of fermentation technology,and discussing from both inheritance and innovation.The paper proposes that inheriting fermentation technology of TCM need to correctly understand fermentation theory,excavate fermentation essence and establish fermentation database;innovating fermentation technology of TCM need to scientifically interpret fermentation mechanism,objectively inherit fermentation experience,regulate and innovate fermentation processing,complete quality standards and develop new varieties of fermentation of TCM.This paper provides ideas and references for development of fermentation technology of TCM.
