      摘要:Objective: To prepare Ershiwuwei Feibing pellets with high drug loading for improve compliance and absorption of this Tibeten medicine,and investigate its properties. Method: Extrusion-spheronization method was employed to prepare pellets.Taking content ratio of craw drug and microcrystalline cellulose,water content,extrusion velocity and rolling speed as factors,yield and spherical degree of pellets as indexes,central composite design-response surface methodology was used to optimize preparation process.Micromeritic properties and in vitro dissolution of this pellets were determined. Result: Optimum formulation was as following:medicine materials powder-microcrystalline cellulose(2.7:1),water content of 34%,extrusion velocity of 14.7 Hz,rolling speed of 39.5 Hz and rolling time of 5 min.Cumulative release rate of hydroxysafflor yellow A was up to 80% in 60 min,cumulative release rate of alantolactone was four times as high as that of the commercial products. Conclusion: Extrusion-spheronization technique can prepare Ershiwuwei Feibing pellets with excellent quality,it has good roundness and high yield with impressive in vitro dissolution of two index components.

      摘要:Objective: Through to observe the medicinal difference changes in Coptidis Rhizome(CR) processed by different ginger juice through ultrastructural tongue scanning electron microscopy(SEM) in SD rats. Method: The rats were randomly divided into normal group, raw CR group, CR processed by fresh ginger group(FGRC), RC processed by dried ginger group(RCDG), fresh ginger juice group(FG) and dry ginger group(DG), respectively took corresponding solution for three weeks. Their tongues were cut off along tongue roots, and processed according to the conventional SEM sample methods, and their ultrastructure was observed by SEM. Result: Compared with the normal group, there was extremely significant differences in numerical lowering between CR group, FGRC group and DGRC group(P<0.01). Keratinization and spalling degree of tain mastoid and filamentous mastoid were not as obvious as normal group. DG group had significant difference in the value increase(P<0.05), and their spalling and keratinization of filamentous mastoid and tain mastoid were also the most obvious. Compared with CR group, there was significant difference in value increase in FGRC group and DGRC group(P<0.05). There was extremely significant difference in value increase in DG group and FG group(P<0.01), and the keratinization and spalling degree of tain mastoid and filamentous mastoid were obvious. Compared with FGRC group, the value of filamentous mastoid was in a rise trend in DGRC, and its mastoid keratinization and spalling degree were more obvious. Compared with DG group, the value of filamentous mastoid was in a decline trend in FG group, and its mastoid spalling and keratinization degree were not so obvious. Conclusion: DGRC there was significant difference in tongue microstructure between CR processed by ginger groups and raw CR group. The results suggest that the cold property of CR can be relieved after processing with ginger, in line with the 'processing' theory of traditional Chinese medicine, and the changes in medicinal cold and hot properties can be reflected from the microstructure of tongue in SEM. At the same time, DG and FG have hot properties, which can be also reflected from the microstructure changes in SEM, laying a solid foundation for the processing mechanism of ginger-processed CR.

      摘要:Objective: To investigate compatibility between Panacis Quinquefolii Radix extract and different pharmaceutical excipient,in order to provide a reference for basic formulation design of solid preparations of Panacis Quinquefolii Radix extract. Method: HPLC was employed to determine contents of ginsenoside Rg1,Re,Rb1,Rc,Rb2,Rb3,Rd with mobile phase of acetonitrile-water for gradient elution and detection wavelength at 203 nm.Taking appearance and total content of index components as indexes,single factor tests were used to investigate compatibility of extract and different pharmaceutical excipient. Result: Contents of ginsenosides decreased significantly under high humidity condition and appearance changed,in other conditions,these indexes relatively stable. Conclusion: Lactose,microcrystalline cellulose,sodium carboxymethyl starch sodium and magnesium stearate can be used as suitable material for solid preparation of Panacis Quinquefolii Radix extract.High humidity condition will affect stability of preparation of Panacis Quinquefolii Radix extract,so it should be kept closed in order to prevent moisture absorption.

      摘要:Objective: To investigate anti-inflammatory effect of Jinqiao Reduqing granules and spray drying powder with different drying process. Method: Eighty-four mice were randomly divided into model group, aspirin group(0.2 g·kg-1),Reduping group(21.84 g·kg-1),Jinqiao Reduqing granules group(11.7 g·kg-1) and Jinqiao Reduqing spray drying powder group(5.85,11.7,23.4 g·kg-1),twelve for each group;then with xylene-induced inflammation,mouse ear swelling was measured.In addition,an alternative 84 mice were divided into groups and treated with the same method;inflammation was induced by traperitoneal injection of acetic acid,and absorbance of peritoneal fluid was measured at 590 nm.Anti-inflammtory effect was evaluated by reducing xylene-induced ear swelling level and glacial acetic acid-induced abdominal capillary permeability. Result: Mouse ear swelling of each group were(18.5±3.2),(9.3±2.0),(14.6±4.2),(14.8±2.3),(14.8±3.6),(13.9±3.2),(13.4±2.2) mg,respectively.Compared with the model group,swelling and swelling rate of each drug administration groups had significantly decreased.Absorbance values of each group were 0.677±0.121,0.321±0.094,0.558±0.107,0.537±0.065,0.583±0.195,0.523±0.137 and 0.512±0.099, respectively. Conclusion: Jinqiao Reduqing spray drying powder and granules has a certain anti-inflammatory effect,they can obviously decrease mouse ear swelling and capillary permeability in abdominalcavity.But at the same dose,anti-inflammatory effect of Jinqiao Reduqing spray drying powder was more powerful than that of Jinqiao Reduqing granules,and under the same doses of equal clinical multiple,anti-inflammatory effect of Reduping granules was slightly weaker than that of Jinqiao Reduqing spray drying powder.

      摘要:Objective: To investigate activation mechanism of AMPK[adenosine 5'-monophosphate(AMP)-activated protein kinase] by ligustroflavone,and explore effect of ligustroflavone on assembly of adiponectin. Method: 293T cells were treated with ligustroflavone.AMPK phosphorylation and evel of cellular AMP and ATP were examined by western blotting and high performance liquid chromatography(HPLC),respectively.3T3-L1 adipocyte was treated by ligustroflavone and its effect on assembly of adiponectin was detected. Result: Ligustroflavone can activate AMPK and increase ratio of AMP/ATP in 293T cells.Ligustroflavone also can promote ssembly of adiponectin multimers. Conclusion: Ligustroflavone activates AMPK by increasing ratio of AMP/ATP and promotes adiponectin multimerization by activating AMPK.

      摘要:Objective: To analyze and evaluate polysaccharides in Elaphuri Davidiani Cornu(EDC) and Cervi Cornu(CC). Method: Glycosaminoglycan, neutral and acidic polysaccharides in EDC and CC were determined and compared by using Elson-Morgan, sulfuric acid carbazole colorimetry and phenol-sulphate acid method, with glucosamine, glucuronic acid and glucose as control drugs. Result: The contents of the three polysaccharides from different ages or sections of samples varied obviously. The highest total content of the three polysaccharides was found in 2-year-old EDC, which was followed by 5, 6-8, 3 and 4-year-old samples. The highest average content of three polysaccharides in the three sections were found in 2-year-old EDC, and the glycosaminoglycan and neutral polysaccharides contents were close to that in CC, but the acidic polysaccharides content was lower than CC. The neutral polysaccharides were the highest in all the EDC samples of various ages, which was followed by glycosaminoglycan and acidic polysaccharides. Conclusion: The study can provide a scientific basis for rational utilization of EDC resources of and its industrialization.

      摘要:Objective: To study the characteristic chromatogram of herbs in Wuji Wan and the change rules of the chemical components. Method: HPLC and HPCE method were used to study different separated prescriptions. The conditions for the capillary zone electrophoresis(CZE) method were 50 mmol·L-1 Borax buffer-Methanol(2:1), constant voltage of 25 kV, pressure sampling under 5 kPa×15 s, electrolyte sealing under 5 kPa×10 s and column temperature at 15℃. Chromatographic experiments were performed on Agelapromosil C18 column(4.6 mm×250 mm,5μm), with the mobile phase was acetomitrile(A)-0.05 mol·L-1 potassium dihydrogen phosphate(including 0.1% phosphoric acid)(B) by a gradient elution;the detection wavelength was 230 nm, column temperature was 25℃, the flow rate was 0.8 mL·min-1. The withdrawal analytical method was used for a qualitative and quantitative evaluation on Wuji Wan, in order to study the changes in chemical components in different prescriptions. Result: HPCE and HPLC were used to confirm 9 and 27 characteristic fingerprint peaks. Coptidis Rhizoma was the main herb of the prescriptions, with great changes in alkaloids in different prescriptions and small changes in Paeoniae Alba Radix. Conclusion: The fingerprint technology and the qualitative and quantitative methods are combined to guide the development of new compound prescriptions.

      摘要:Objective: To establish a HPLC method for the simultaneous determination of tanshinol, protocatechuic aldehyde, paeoniflorin, rosmarinic acid, lithospermic acid, salvianolic acid B, and paeonol in Shuangdan capsules. Method: An Agilent Zorbax SB-C18 analytical column(4.6 mm×250 mm, 5μm) was used, with the mobile phase of acetonitrile-0.1% formic acid solution for gradient elution at the detection wavelength of 235 nm, the flow rate was 1.0 mL·min-1, and the column temperature was 30℃. Result: The linear range of the seven components were 54.43-544.3, 0.96-9.6, 2.68-26.8, 0.92-9.2, 2.16-21.6, 9.36-93.6, 9.16-91.6 mg·L-1 respectively, and the correlation coefficients were 0.9997, 0.9996, 0.9995, 0.9993, 0.9992, 0.9997, 0.9995.The average recoveries were 98.6%(RSD 1.8%), 100.2%(RSD 1.9%), 100.4%(RSD 2.0%), 99.1%(RSD 2.5%), 98.8%(RSD 2.3%), 100.6%(RSD 1.9%), 99.1%(RSD 1.8%). Conclusion: This analysis method is accurate, reliable and highly reproducible, and can provide a scientific basis for controlling the quality of Shuangdan capsules.

      摘要:Objective: To establish the fingerprint of Fufang Jinqiancao granules for evaluating scientifically, controlling the quality and ensuring its production stability. Method: The RP-HPLC was performed on an ODS-BP column with acetonitrile-0.2% phosphoric acid solution as mobile phase, the flow rate was 1 mL·min-1, injection volume was 20μL, the wavelength was set at 350 nm. The temperature of column was maintained at 20℃. Fingerprint Similarity Evaluation Software(edition 2012.130723) of Chinese Pharmacopoeia Commission was used to evaluate the similarity of the 10 batches of Fufang Jinqiancao granules, and to analyze the correlations of 4 ingredients in Fufang Jinqiancao granules. Result: Mangiferin was used as the reference peak, the common mode for the HPLC fingerprints was set up, with the similarities of the 10 batches ranging between 0.961 and 0.997.Altogether 20 common peaks in the chromatograms were found, of which 19 peaks were assigned to herbs. Conclusion: The method is sensitive, accurate and simple, and can be used for the quality control of Fufang Jinqiancao granules.

      摘要:Objective: To establish a RP-HPLC method for simultaneously determining the contents of chlorogenic acid, caffeic acid forsythin, andrographalide and dehydroandrographalide in Fufang Shuanghua Jujuepian. Method: RP-HPLC was carried out on a hypersil C18(4.6 mm×250 mm, 5μm). The mobile phase was A(acetonitrile)-B(0.2% formic acid) in the gradient elution mode(0-8 min, 4%-13%A;8-24 min, 13%-14%A;24-25 min, 14%-26%A;25-65 min, 26%-33%A;65-70 min, 33%-50%A). The flow rate was 1.0 mL·min-1 and the detection wavelength was set at 240 nm. The column temperature was maintained at 30℃. Result: The calibration curves of chlorogenic acid, caffeic acid, forsythin, andrographolide and dehydroandrographolide were in good linearity within the range of 0.2825-2.8250μg(r=0.9995), 0.0868-0.8675μg(r=0.9997), 0.4800-4.8000μg(r=0.9995), 0.1588-1.5875μg(r=0.9995), 0.1488-1.4875μg(r=0.9994) and the average recoveries were 97.3%(RSD of 1.2%), 97.8%(RSD of 1.1%), 98.9%(RSD of 1.4%), 97.6%(RSD of 1.0%), 99.0%(RSD of 1.5%). Conclusion: The method is simple, reliable and specific, and can be applied for quality control of Fufang Shuanghua Jujuepian.

      摘要:Objective: To isolate and identify the chemical constituents from the pulp extract of melon. Method: The constituents were isolated with the combination of silica-gel column chromatography and Sephadex LH-20 column chromatography. The structures were identified on the basis of 1H and 13C-NMR, ESI-MS spectroscopic analysis as well as chemical properties with the comparison of reported compounds. Result: Eight compounds were obtained and identified as 3,5-dimethoxyl-benzyl alcohol-4-O-β-D-glucoside(1), 2,6-dimethoxyl-4-hydroxyl-phenol-1-O-β-D-glucoside(2),1-O-p-coumaroylglycerol(3),coniferin(4),cucumegastigmanes II(5),linarionoside C(6),δ-viniferin(7),kaempferol-3-O-glu-rha-glu(8). Conclusion: Compounds 1-8 are all isolated from this plant for the first time.

      摘要:Objective: To study four different methods for the content determination of anthocyanin in the petroleum ether part, the ethyl acetate part, the n-butyl alcohol part and the water part in Lonicera caerulea var. edulis and its antioxidant activity. Method: L.caerulea var edulis is was extracted by 95% ethanol, dissolved in water after solvent recovery, processed with petroleum ether, ethyl acetate and n-butanol to get four parts of L.caerulea var edulis. Color value method, extinction coefficient method, pH differential method and NaNO2-Al(NO3)3-NaOH colorimetric method were used to determine content of anthocyanin and polyphenols, and the DPPH was used to determine the antioxidant ability of different part in L.caerulea var. edulis, with the detection wavelength at 517 nm. Result: Color value method, extinction coefficient method and pH differential method were used to show the highest anthocyanin content in the part of n-butyl alcohol. The result of NaNO2-Al(NO3)3-NaOH colorimetric method showed that the maximum content of polyphenolsin in the ethyl acetate was 29.45%. The highest antioxidant activity in vitro was found in the ethyl acetate part;the antioxidant activities of n-butyl alcohol part and the water part were similar;the antioxidant activity of the petroleum ether part was the lowest. Conclusion: In L.caerulea var edulis, anthocyanin has a certain antioxidant activity, while polyphenols also showed a high antioxidant activity.

      摘要:Objective: To establish the HPLC fingerprint spectrum of flavonoids in Gynostemma pentaphyllum. Method: HPLC was performed on ZORBAX SB-C18 column(4.6 mm×150 mm, 5μm), eluting in a gradient mode with the mobile phase of acetonitrile(A)-0.2%H3PO4 as(B) at a flow rate of 1.0 mL·min-1. The column temperature was 30℃. The detection wavelength was 360 nm. Injection volume was 10μL. Result: Eleven common characteristic peaks were identified as fingerprint peaks, the methodological study was consistent with the technical requirements. Conclusion: The established method is stable, reliable and highly accurate, and can provide reference for quality control of Gynostemma pentaphyllum.

      摘要:Objective: To conduct a qualitative analysis for the quality of Shengxuebao mixture by HPLC, in order to achieve the quality control of the overall accuracy of Shengxuebao mixture, and establish a multi-component fingerprint chromatogram of Shenxuebao mixture by HPLC. Method: Thermo Syncronis-C18 column(4.6 mm×150 mm, 5μm) was eluted with the mobile phase of acetonitrile(A) and 0.1% phosphoric acid(B) in a gradient mode(0-15 min, 2%A;15-30 min, 2%-15%A;30-50 min, 15%-25%A;50-60 min, 25%A). Detection wavelength was set at 230 nm;column temperature was 30℃;the flow rate was 0.8-1.0 mL·min-1 and analytic time was 60 min. Injection volume was 5μL. Result: Under the same chromatogram condition, paeoniflorin and other condittuents were determined, and 14 peaks were identified on the HPLC fingerprint. The methodological results were satisfactory. The similarity of the 10 batches of Shenxuebao mixture was above 0.9. Conclusion: The method is simple and highly reproducible, and can provide scientific basis for the quality control of Shenxuebao mixture.

      摘要:Objective: To optimize the technological parameters in the purification process of active materiomic from Cassia Semen by HPLC fingerprint technique. Method: Using the established HPLC fingerprint of Cassia Semen, the macroporous resin type, loaded concentrations, and elution conditions were optimized using the similarity and recovery rates of five components in fingerprints as indexes. Result: D-101 type resin possessed the highest recovery rates of five components. The optimized purification process was as follows:the loaded concentration was 3 mL, the amount of resin was 10 mL, 50 mL of 75% ethanol was used as eluent, respectively. After being purified, the recovery rates of five components were all over 85%, and the similarity of fingerprints was higher than 0.98.These components could get balanced recovery. Conclusion: Based on HPLC fingerprint technique, the optimized purification process of active materiomic from Cassiae Semen with macroporous resin can ensure that the main effective components are consistent with the crude extract of Cassiae Semen.

      摘要:Objective: To identify Schisandrae Chinensis Fructus and Schisandrae Sphenantherae Fructus by ITS2 sequences. Method: ITS2 sequences of Schisandrae Chinensis Fructus and Schisandrae Sphenantherae Fructus were amplificated by polymerase chain reaction(PCR) and bidirectionally sequenced.ITS2 sequences of related 10Schisandra species and 2 out-group specie samples were downloaded from GenBank.CodonCode Aligner was used for sequence splicing and MEGA 5.10 was utilized for related data calculating.Then neighbour-joining tree(NJ tree) was built based on K2P model,and Blast method was used to identify specie of samples in this study.Differences between ITS2 secondary structures which were prodicted using ITS2 web server were analysed. Result: Lengths of sequences of Schisandrae Chinensis Fructus and Schisandrae Sphenantherae Fructus were 231,225-227 bp.Blast identification of samples were Schisandrae Chinensis Fructus and Schisandrae Sphenantherae Fructus. K2P genetic distance between two species(0.0104-0.0157) was greater than intraspecific genetic variation.In NJ tree,Schisandrae Chinensis Fructus and Schisandrae Sphenantherae Fructus were clustered into a single branch respectively,which showed monophyletic,and could be distinguished between other species of S. with a high Bootstrap approval rate. Conclusion: DNA barcoding based on ITS2 sequence is a powerful and efficient tool for identification of Schisandrae Chinensis Fructus and Schisandrae Sphenantherae Fructus.

      摘要:Objective: To investigate effect of full illumination in autumn and salicylic acid on physio-biochemistry and quality of Ginseng Radix et Rhizoma. Method: Full illumination coupled with spraying different concentration of salicylic acid(SA),6-benzylaminopurine(6-BA) and brassinolide(Br) in autumn,activities of superoxide dismutase(SOD),catalase(CAT),peroxidase(POD) and malondialdehyde(MDA) were determined,contents of ginsenosides were determined by HPLC. Result: Full illumination increased capacity of photosynthesis to some degree,but it weakened physio-biochemistry,which may damage Ginseng Radix et Rhizoma.After sprayed three kinds of hormones under full illumination,ability of antioxidant and net photosynthesis(Pn) improved,SA was better than 6-BA,6-BA better than Br.Contrasted with blank group,0.2 mmol·L-1 and 1.0 mmol·L-1 SA increased by 39.9% and 43.2% for SOD,43.1% and 57.2% for CAT,63.5% and 64.5% for POD,17.1% and 16.3% for Pn;contrasted with control group,increased by 1.1% and 3.5% for SOD,40.6% and 54.5% for CAT,42.0% and 42.8% for POD,33.2% and 32.3% for Pn.1.0 mmol·L-1 SA group significantly improved contents of ginsenoside Rg1 and ginsenoside Re by comparing with the blank and control group. Conclusion: Full illumination destroys structure of Ginseng Radix et Rhizoma leaves,reduces its quality.0.2-1.0 mmol·L-1 SA possesses anti-aging,it can improve quality of Ginseng Radix et Rhizoma.

      摘要:Objective: To compare contents of total saponins and platycodin-D in Platycodonis Radix from different origin. Method: Contents of total saponins and platycodin-D in 36 batches of Platycodonis Radix samples from different origin were determined by methods in the 2005 edition and the 2010 edition of Chinese Pharmacopoeia,respectively. Result: Content distribution range of total saponins in 10 batches of samples from Chifeng was 5.02%-9.20%,content of platycodin-D was 0.1%-0.175%,both of content distribution range was relative narrow,which meant their quality was more stable than others.Contents of total saponins and platycodin-D from other districts had a large scale distribution,which meant quality of Platycodonis Radix between Chifeng and other districts was very different. Conclusion: Chifeng obtains a continental climate,and could be developed into an important production base to make quality of Platycodonis Radix consistently.

      摘要:Objective: To explore tissue distribution of major constitutes form Dachengqi Tang in acute pancreatitis(AP) rats. Method: Male SD rats were randomly divided into shamp operation group,model group,normal administration group and AP administration group.AP model was established with 3.5% sodium taurocholate retrograding cholangiopancreatography injection.Rats were orally administered Dachengqi Tang with 10 g·kg-1 after 4 hours following model being established.Heart,liver,spleen,lung,kidney,large intestine,pancreas and blood were then collected 24 hours later.Components and their distribution in various tissues and blood plasma were detected by HPLC-MS/MS,and penetration ratio(PR) was analyzed respectively. Result: Concentrations of rhein and honokiol[(105.5±57.8),(40.8±6.7)μg·L-1,respectively] in the pancreas of AP rats was higher than that of the normal group[(31.9±19.4),(14.5±10.4)μg·L-1,respectively].Compared to normal rats,PR of blood-to-tissue of main components in AP group was less than that in the normal group. Conclusion: Tissue distribution of main components in Dachengqi Tang are inhibited under AP condition,except for rhein and honokiol,which shows these two components targeted distribution in pancreas might be potential pharmacological substances of Dachengqi Tang for AP.

      摘要:Objective: To investigate the bone repair effect of Bushen formula on rats with steroid-induced osteonecrosis of the femoral head(SONFH). Method: Male Wistar rats were randomly divided them into 6 groups:control group, model group, high-dose Bushen formula, mid-dose Bushen, low-dose Bushen groups, and Jiangusheng pills group. Then gluteal injection with methylprednisolone sodium succinate by 21 mg·kg-1 into all rat groups except for control group, once a day, for 3 consecutive days was conducted to establish the model of SONFH. Since the fourth day, low-dose Bushen, Mid-dose Bushen, High-dose Bushen groups and Jiangusheng group were give 5.3, 10.6, 21.2 g·kg-1 Bushen formula and 1.68 g·kg-1 Jiangusheng pills respectively. Six weeks later, their blood and femoral head were collected to observe the histopathological changes of femoral head by HE staining. Micro-CT scanning was used to observe femoral bone morphogenetic changes and analyze the bone morphometry. Bone metabolism markers osteocalcin(bone gla-protein, BGP) and calcitonin(CT) content were detected by ELISA. Result: The femoral head of model group rats showed many histopathological changes, such as more empty lacunae, more fat cells, narrowed or broken bone trabecula, the bone marrow cavity cytopenia, massive necrosis zones. Besides, femoral head collapse, osteopenia, trabecular thinning and other imagological changes can also be observed. According to the bone metrological parameters, bone volume fraction, trabecular thickness, trabecular bone pattern factor, trabecular number and bone density decreased, but trabecular separation increased in model group rats. BGP and CT content in serum significantly reduced in model group rats. Compared with the model group, mid-dose and high-dose Bushen can significantly improve the pathological changes in femoral head of rats. The three dose Bushen groups showed significant inhibition of bone necrosis. Mid-dose and high-dose Bushen can remarkably increase BGP and CT content in serum. The Jiangusheng group had a similar effect with mid-dose Bushen group. Conclusion: Bushen formula can significantly improve the histopathological and imagological changes of rats with SONFH and increase BGP and CT content in serum. This effect in promoting bone repair may be one of its mechanisms of preventing steroid-induced femoral head osteonecrosis.

      摘要:Objective: To investigate the effect of diabetes on the expression of transforming growth factor-β1(TGF-β1), transforming growth factor-β receptors Ⅱ antibodies(TGF-β RⅡ) and signal transduction molecules 7(Smad7) in rats myocardium and the intervention effects of Didang Tang and its split recipes. Method: The 12 healthy male SD rats were chosen randomly as normal group, and other 105 SD male rats were injected intraperitoneally with STZ to induce diabetes(DM) rat models, which were randomized into model group, Didang Tang group, leech and mang worm group, peach kernel and rheum officinale group, and valsartan group, receiving corresponding intervention. At the end of 8-week treatment, fasting blood glucose(FBG) was detected by full-automatic biochemical analyzer,pathomorphological change of myocardium was observed through hematoxylin-eosin staining,and the protein expression of TGF-β1, TGF-β RⅡ and Smad7 were assessed by Western blot. Result: Compared with the normal group,cell hypertrophy was observed in myocardial tissues, with obvious infiltration of inflammatory cells, significant increase in TGF-β1, TGF-β RⅡexpression and significant decrease in Smad7 expression(P<0.01). Compared with the model group, Didang Tang could significantly down-regulate TGF-β1 and TGF-β RⅡexpression and significantly down-regulate Smad7 expression(P<0.05, P<0.01). Compared with the diabetic rat models, the split recipe groups could down-regulate TGF-β1 expression and improve myocardial damages, but the effect was inferior to that of whole prescription group. Conclusion: Didang Tang could relieve cardiac fibrosis of diabetic rats and its mechanism may be associated with down-regulating TGF-β1 and TGF-β RⅡexpression in myocardium of diabetic rats and up-regulating Smad7 expression. In addition, the prophylactic and therapeutic effect of Didang Tang was better than that of its split recipes.The treatment of 'expelling heat, removing blood stasis and dredging collaterals' was associated with inhibiting TGF-β1/Smad7 signal transduction pathway.

      摘要:Objective: To study the anti-inflammatory activity of Huanglian Jiedu Tang(HLJDT) and its fractions as well as its material basis. Method: Xylene-induced ear-swelling and capillary permeability experiments were carried out to build the animal model, in order to determine the effective dose of HLJDT and then calculate the dose of each fractions of HLJDT. The animals were randomly divided into the blank group, the model group, the dexamethasone group, the HLJDT group, the HLJDT 50% alcohol-treated supernatant group, the water-treated precipitation group and the 50% alcohol-treated precipitation group, and administrated with drugs according to the traditional way for 3 days. Evans blue(EB) was intravenously injected 60 min later after the last administration, and then the mouse xylene induced inflammation experiment was immediately carried out. The anti-inflammatory effect of HLJDT and its fractions was researched with high performance liquid chromatography(HPLC), and inter-group differences were compared. Result: Compared with the model group, dexamethasone, HLJDT, HLJDT 50% alcohol-treated supernatant and 50% alcohol-treated precipitation could effectively reduce the swelling degree and showed significant difference(P<0.01). The content of EB in the three groups mentioned above decreased to(11.8±1.8),(23.4±3.8),(25.2±3.2) ng·g-1, the swelling inhibition rates were 26.44%, 29.89%, 19.54%. HLJDT 50% alcohol-treated supernatant showed a more significant effect on anti-inflammatory than 50% alcohol-treated precipitation. Conclusion: HLJDT, HLJDT 50% alcohol-treated supernatant and 50% alcohol-treated precipitation could effectively reduce the swelling degree. On the basis of the result of HPLC, it can be inferred that the flavonoids interacted with alkaloids to reduce the side effect of alkaloids, increase the iridoids and showed a significant anti-inflammatory effect. Therefore, they are the effective anti-inflammatory fractions.

      摘要:Objective: To observe the effect of Liuwei Dihuang Wan drug serum on the cells proliferation and expression of monocyte chemoattractant protein-1(MCP-1) and intercellular adhesion molecule excluded-1(ICAM-1) in mesangial cells of rats cultured in high glucose conditions. Method: Different doses of Liuwei Dihuang Wan drug serum were prepared, and rat mesangial cells were cultured under high glucose. grouping:normal group(NG), high glucose(HG) group, HG+control serum group(10% control serum),HG+low dose group(2.5% Liuwei Dihuang Wan serum+7.5% control serum), HG+middle dose group(5% Liuwei Dihuang Wan serum+5% control serum),HG+high dose group(10% Liuwei Dihuang Wan serum). Following treatment with Liuwei Dihuang Wan drug serum for 24, 48 h, mesangial cells activity and proliferation changes were observed with methyl thiazolyl tetrazolium and Brdu incorporation assay. MCP-1 and ICAM-1 secretion in cell culture supernatants were detected by ELISA. MCP-1 and ICAM-1 mRNA expression were measured by Real Time-PCR. Result: Compared with normal group, the cells proliferative activity, MCP-I and ICAM-1 content in supernatant, and the expression of MCP-1 and ICAM-1 mRNA were significantly up-regulated in high glucose group after 24,48 h culture(P<0.01). Compared with the high glucose group, treatment with 5% or 10% Liuwei Dihuang Wan drug serum significantly reversed these above changes(P<0.05), and the inhibition was in a time and dose-dependent manner. Conclusion: High glucose can induce up-regulation of cells proliferation, MCP-1 and ICAM-1 expression in rat mesangial cells. Liuwei Dihuang Wan may reduce the proliferation activity, decrease MCP-1 and ICAM 1-secretion, and down-regulate MCP-1 and ICAM-1 mRNA expression, thus delaying glomerulosclerosis in diabetic nephropathy.

      摘要:Objective: To study the effect of charred Arteisia frigida on the change of mRNA expressions of vascular endothelial growth factors(VEGF), basic fibroblast growth factors(bFGF) and their receptors in ulcer tissues of stress gastric ulcer model rats, and investigate the molecular mechanism for charred A.frigida to promote the healing of gastric ulcers. Method: 84 SD rats were randomly divided into 7 groups, named control group, model group, Yunnanbaiyao group(Positive drug, 0.18 g·kg-1), A.frigida crude drug group(1.35 g·kg-1), low dose charred A.frigida group, middle dose charred A.frigida group and high dose charred A.frigida group(0.9, 1.35, 1.8 g·kg-1). Water-immersion stress method was used in all other groups except normal group to establish rat models of cute gastric ulcer. After intragastric administration for one continuous week, stomach ulcer tissues were scissored from the conventionally sacrificed animals. Western blot and RT-PCR technique were used to detect mRNA and protein expressions of VEGF and its receptor VEGFR, bFGF and its receptor bFGFR. Result: Compared with the normal group, ulcer tissues in rats with acute stress ulcer had significantly decreased bFGF(P<0.01) and bFGFR(P<0.05) protein expressions. Compared with the model group, charred A.frigida low dose group significantly increased VEGF mRNA expression(P<0.05), VEGF protein expression VEGFR protein expression(P<0.05), but without dose dependence; no difference was found between the other groups and the model group. charred A.frigida middle dose group, high dose group and Yunnanbaiyao group significantly increased bFGF protein expression(P<0.01), and charred A.frigida high dose group also increased bFGF mRNA expression, bFGFR mRNA expression(P<0.01) and bFGFR protein expression(P<0.05). Conclusion: The molecular mechanism for charred A.frigida to promote the healing of gastric ulcers may be associated with up-regulation of VEGF and VEGFR protein expressions, bFGF and bFGFR protein expressions. The regulation of VEGF, bFGF, and bFGFR is at the level of transcription, and the regulation of VEGFR is at the post-transcriptional levels.

      摘要:Objective: To observe the protective effect of baicalin on cerebral ischemia/reperfusion in rats. Method: Fifty adult Sprague-Dawley rats were randomly divided into five groups as following:sham group, model group, 135 mg·kg-1 baicalin group, 45 mg·kg-1 baicalin group, 15 mg·kg-1 baicalin group, with 10 in each group. The sham and model groups received distilled water ig at the same volume and time points as the baicalin groups. Drug or distilled water was administered once a day for 7 d. One hour later after the last administration with baicalin, the middle cerebral artery occlusion was adopted to establish the focal cerebral ischemia/reperfusion model. After 24 h of ischemia/reperfusion, the neurological deficit scores were evaluated. Histopathology changes were observed by hematoxylin and easin(HE) and TdT-mediated dUTP nick end labeling(TUNEL) staining. The levels of superoxide dismutase(SOD), malondialdehyde(MDA), tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) in cerebral tissue were measured by commercial kit. Result: Compared with the sham group, the model group showed severe pathomorphological damages, significantly higher neurological deficit scores and neural apoptosis, MDA, TNF-α and IL-1β(P<0.01), but remarkably inactive(P<0.01). Compared with the model group, baicalin could improve the histopathology injury, decrease neural apoptosis, baicalin high dose group significantly decreased neurological deficit scores(P<0.01), baicalin middle and high dose groups significantly increased the activities of SOD(P<0.05) and decreased the levels of MDA, TNF-α and IL-1β(P<0.01). Baicalin low dose group also significantly decreased the levels of IL-1β(P<0.01). Conclusion: The baicalin had the cerebral protective effect on cerebral ischemia/reperfusion rats, the mechanism may be related to its characteristics of anti-apoptosis, reduction of free radical damage and suppression of inflammation.

      摘要:Objective: To observe the effects of Rupeng 15 powder(RPP15) on cyclooxygenase-2(COX-2), prostaglandin E2(PGE2) protein and mRNA expression levels in the knee joint fluid and synovium tissues,and explore the therapeutic mechanism of RPP15 in rats with monosodiu murate(MSU)crystals-induced gouty arthritis. Method: The rats were dividied into normal group, model group, RPP15 low-dose, middle-dose and high dose groups(0.4, 0.8, 1.2 g·kg-1),indomethacin group(3 mg·kg-1). The normal group and model group were ig administered with normal saline;various treatment groups were ig administered with corresponding doses for continuous 7 days. One hour after the last administering, 50μL sterile 1% PBS was injected in bilateral knee joint cavity in normal group, and 50μL MSU was injected in other groups. Twenty-four h after modeling, enzyme linked immunosorbant assay(ELISA) method and real-time fluorescent quantitative PCR method were used to detect COX-2, PGE2 protein and mRNA expression levels in the knee joint fluid and synovium tissues. Result: Data from our study showed that rat with gouty arthritis induced by MSU crystal demonstrated an elevation in COX-2, PGE2 in knee joint fluid and mRNA expression levels in synovium tissues(P<0.01).Compared with the model group, RPP15(0.4, 0.8, 1.2 g·kg-1) groups and indomethacin group had significantly lower COX-2 and PGE2 protein levels in knee joint fluid COX-2 mRNA level in synovium tissues(P<0.05, P<0.01),but PGE2 mRNA was no statistically significant difference in synovium tissues. Conclusion: MSU can increase the expression of COX-2 and PGE2 in knee joint fluid and synovium tissues. RPP15 may have the effect on MSU-induced gouty arthritis by decreasing COX-2 expression, COX-2 level and PGE2 level.

      摘要:Objective: To investigate the effect of drug serum by Chinese herbal compound prescription in Buyang Huanwu Tang(BHT), Xuefu Zhuyu Tang(XZT) and Sijunzi Tang(ST)on the expression of Toll-like receptor-4(TLR4) and main components of downstream signal transduction pathways, and to study their possible anti-atherosclerotic mechanism. Method: New Zealand rabbits were divided into 4 groups in random:normal group, BHT group, XZT group, and ST group, with 5 rabbits in each group. Rabbits in normal group were given gastric perfusion of 20 mL normal saline, rabbits in three Chinese herbal compound groups respectively received gastric perfusion of 20 mL BHT(6.7 g·kg-1), XZT(3.6 g·kg-1) and ST(1.6 g·kg-1), for 7 continuous days. Blood was drawn from rabbit's heart 2 h after the last gastric perfusion, and then the drug serum was prepared after centrifugation. Vascular endothelial cells were randomly divided into 6 groups:blank group, model group, Atorvastatin group(10μmol·L-1), BHT group, XZT group and ST group. After stimulation with LPS(1 mg·L-1) for 2 h, drug serum containing 10% control western medicine and drug serum containing Chinese herbal compound were added separately for intervention. Cells were collected after 24 hours. The gene expression of TLR4, myeloid differentiation factor 88(MyD88), tumor necrosis factor-associated factor 6(TRAF-6) and nuclear factor-κ B(NF-κ B) were measured with RT-qPCR. Western blotting method was used to analyze the expression of TLR4 and NF-κ B protein. Result: expressions of TLR4, MyD88, TRAF-6 and NF-κ B in model group were increased significantly after LPS stimulation, with statistical difference compared with blank group(P<0.01). Intervention by BHT and XZD could significantly decrease the expression of TLR4, MyD88, TRAF-6 and NF-κ B, with statistical difference compared with model group(P<0.05). However, ST group had no significant changes in various indicators, with no statistical difference compared with model group. Conclusion: BHT and XZD can inhibit the expression of TLR4 and main components of downstream signal transduction pathways, which may be one of its action mechanisms for anti-atherosclerosis.

      摘要:Objective: To study the antithrombotic effect of Persicae Semen extract(PSE) in blood stasis rats, its relationship with platelet adhesion molecule 31(CD31), nuclear transcription factor-κ B(NF-κ B) and its inhibitor κ B(Iκ B) expressions and the antithrombotic mechanism of Semen Persicae against blood stasis syndrome. Method: Sixty SD rats were randomly divided into the normal group, the model group, the PSE low-, middle-, high-dose groups(2, 4, 8 g·kg-1) and Danshen group(14 g·kg-1). The stasis model was established through ip with carrageenan and dry yeast liquid. All of treatment groups were given drugs by oral administration. The stasis symptom, blood viscosity, plasma viscosity, fibrinogen, micro-thrombosis and renal damage were observed by blood viscometer and histopathological method. CD31, NF-κ B, Iκ B protein and mRNA expression absorbance of vascular endothelial cells were detected by immunohistochemistry method and in situ hybridization technique. Result: Compared with the normal group, the model group had some stasis symptoms, such as congestion, swelling, cyanosis under skin mucosa;its blood viscosity, micro-thrombosis and renal injury increased, CD31, nuclear NF-κ B and total-Iκ B protein of endothelial increased, but NF-κ B total protein decreased(P<0.01). Compared with the model group, the PSE high dose group showed relieved stasis symptoms, lower blood viscosity, plasma viscosity, micro-thrombosis and relieved kidney damage(P<0.01), decreased CD31 protein of endothelial, but higher NF-κ B, IκB total protein and mRNA(P<0.05, P<0.01). Conclusion: PSE can significantly improve blood stasis symptoms, lower blood viscosity, inhibit thrombosis and protect renal. It might show the anti-micro thrombosis effect by promoting NF-κ B and its inhibitor Iκ B gene transcription and translation, or inhibiting its degradation, as well as reducing the CD31 protein.

      摘要:Objective: To explore intervention effect of Shiquan Dabu Tang-medicated serum on proteins of Wnt/β-catenin signal transduction pathway in lung adenocarcinoma cisplatin-resistant cell lines of A549/DDP. Method: SD rats were randomly divided into 4 groups:high dose(65 g·kg-1, 10 mL·kg-1) group, medium dose(32.5 g·kg-1, 10 mL·kg-1) group, low dose(16.25 g·kg-1, 10 mL·kg-1) group and blank control group(normal saline, 10 mL·kg-1). A549/DDP cells were divided into blank serum group, high, medium and low dose groups of drug-medicated serum. MTT assay was used to determine 50% inhibition concentraction(IC50)values of cisplatin. Western blotting was adopted to detect the protein expression of glycogen synthase kinase 3β(GSK-3β), p-GSK-3βser9, β-catenin and survivin. Result: With the increase in the concentration of medicated serum, IC50 values of cisplatin(DDP) in A549/DDP cells decreased, especially in mediummedium and high groups(P<0.01). Shiquan Dabu Tang-medicated serum had no effect on GSK-3β protein expression in A549/DDP cytoplasm, but can significantly down-regulate p-GSK-3βser9protein expression and total β-catenin and survivin protein expression in cytoplasm, particularly in medium and high groups(P<0.01). Conclusion: Shiquan Dabu Tang can lower the expression levels of cytoplasmic p-GSK-3βser9, total β-catenin and surviving proteins, so as to influence Wnt/β-catenin signal transduction pathway and sensitize cisplatin resistance.

      摘要:Objective: To investigate the effect of forsythiaside A(FA) on body temperature, heat-regulating center and temperature-sensitive transient receptor potential(TRP) A1 expression of mice with yeast induced fever, so as to elucidate the mechanism of the antipyretic effect and potential mechanism of FA. Method: Sixty male C57BL/6 mice were randomly divided into six groups:normal group, model group, different dosages(2, 4, 8 mg·kg-1) of FA-treated group, aspirin group(as the positive control, 90 mg·kg-1), with 10 mice in each group. The pyrexia model was induced by injection of yeast suspension subcutaneously. Rectal temperature was recorded before and 17-22 h after pyrexia induction. At the 22 h after injection of yeast, the mice were sacrificed to collect their hypothalamus and dorsal root ganglion(DRG). The expression of TRPA1 was detected by immunohistochemistry and Western blot. Result: Compared with the normal group, the rectal temperature of the mice in the model group notably increased after injection of yeast, reaching the peak at 17 h(P<0.01). In the meanwhile, the TRPA1 level in hypothalamus and DRG of the model group significantly reduced. Compared with the model group, FA(4, 8 mg·kg-1) group showed significant decrease in the elevated temperature of mice induced by yeast, and up-regulation in the expression of TRPA1 in paraventricular nuclei(PVN), supraoptic nucleus(SO) and DRG(P<0.05,P<0.01). Conclusion: FA can reduce the temperature of fevered mice induced by yeast, which may be related to their effective regulation of TRPA1 in hypothalamus and DRG.

      摘要:Objective: To investigate the effect of Danlong Xingnao decoction on the angiogenesis and the angiopoitin 1(Ang-1) protein expression of rats with cerebral ischemia-reperfusion. Method: A rat model of the middle cerebral artery occlusion/reperfusion(MCAO) was created by using the suture method, and the blood reperfusion was performed 2 hours later. The rats were randomly divided into six groups, high-dose, medium-dose and low-dose(14.76, 7.38, 3.69 g·kg-1) Danlong Xingnao decoction groups, nimodipine(10.8 mg·kg-1) group, model group and sham group. The rats were given by gavage once a day for 7 days. The microvessel vascular density(MVD) were observed and the expression of Ang-1 at protein level were studied by the immuneohistochemical method. Result: Compared with the sham group, the microvessel density, Ang-1 in model group were higher significantly(P<0.05). Compared with the model group, Danlong Xingnao decoction groups showed significantly higher levels of MVD and the protein expression of Ang-1(P<0.05, P<0.01). Conclusion: Danlong Xingnao decoction could promote angiogenesis and protect ischemic brain tissue in rats. Its mechanism may be correlated with the up-regulation of the expression of Ang-1.

      摘要:Objective: To study the protective effect and its mechanism of Shenxiong drop pills(SDP) on the acute myocardial ischemia(AMI) induced by isoprenaline(ISO). Method: Sprogue-Dawley rats were divided randomly into seven groups, with 10 in each group. They were normal group, model group, Danshen group, metoprolol group, high, medium and low-dose SDP groups(11.2, 5.6, 2.8 g·kg-1). The AMI rat model was established by injection with ISO. Five days later, the effect of SDP on the electrocardiogram(ECG) at different time points were observed, after the continuos administration for another two days, the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in myocardial tissue, the levels of endothelin-1(ET-1), nitric oxide(NO), thromboxane B2(TXB2), 6-keto-prostaglandin F1a(6-keto-PGF1α) and the activities of nitric oxide synthase(NOS) in serum were detected. Meanwhile, Ca2+-Mg2+-ATP enzyme, Na+-K+-ATP enzymatic activity and ATP content in rat myocardial tissues were determined. Result: SDP inhibited the J spot downward of ECG induced by ISO during myocardial ischemia, decreased the levels of ET-1, TXB2 in serum, and increased the levels of NO, 6-Keto-PGFlα and the activities of NOS in serum to varying degrees, but with no significant effect on ATP content in myocardial tissues and ATP enzymatic activity. Conclusion: The SDP has the apparent protective effect on AMI injury induced by ISO, its mechanism may be correlated with protecting endothelial cell and inhibiting platelet aggregation.

      摘要:Objective: To study the effect of β-sitosterol on gastric mucosal injury induced by aspirin and its possible mechanism, and explore the effect of β-sitosterol on pharmacological functions of aspirin. Method: Gastric mucosa injury modelsinduced by aspirinin SD rats was established by intragastric administration of different dosesof β-sitosterol(50,150,250 mg·kg-1) for seven days, and ulcer area was assessed to analyze the effect of β-sitosterol on gastric mucosal injury induced by aspirin in rats. Serum superoxide dismutase(SOD), tumor necrosis factor-α(TNF-α), prostaglandin E2(PGE2), malondialdehyde(MDA) and nitric oxide(NO) were measured to analyze the possible mechanism. Rat paw edema test, mouse ear swelling, rat yeast-induced fever test and mouse hot plate test, and platelet aggregation in rats were used to examine the pharmacological effect of different doses of β-sitosterol on anti-inflammatory, antipyretic, analgesic, and inhibition of platelet aggregation induced by aspirin. Result: Compared with aspirin-treated group, gastric ulcer area of β-sitosterol group rats was significantly reduced(P<0.05);gastric ulcer area in the high and middle dose β-sitosterol group rats was significantly smaller than that in β-sitosterol low dose group(P<0.05). Compared with aspirin-treated group, NO and SOD in serum of β-sitosterol rats increased significantly(P<0.05), TNF-α, MDA significantly decreased(P<0.05), and PGE2 level was not significantly different. Compared with the model group,toe swelling of rats and ear swelling of micein various treatment groups were significantly reduced(P<0.05);compared with the aspirin-treated group, toe swelling of rats and ear swelling of mice in β-sitosterol group were significantly reduced(P<0.05), but there is no significant differences between the β-sitosterol groups. Compared with aspirin-treated group, the change in the value of the rectal temperature and pain threshold in mice and platelet aggregation rate in rat of the corresponding β-sitosterol group were no significant difference. Conclusion: β-sitosterol can reduce the aspirin-mediated gastric mucosal injury, and its mechanism may be related to enhancing the clearance capacity of oxygen free radicals(OFR) of body, improving levels of NO, inhibiting aggregation and release of inflammatory cytokines TNF-α;β-sitosterol can enhance the anti-inflammatory effects of aspirin and have no significant effect on antipyretic, analgesic, and inhibition of platelet aggregation.

      摘要:Objective: To investigate the effect of Guanxin Zhitong capsule(GXZT) in atherosclerosis(AS) model rats. Method: SD rats were randomly divided into 5 groups:normal group, model control group, GXZT high and low dose groups and Atorvastatin Calcium tablets control group. The normal group received normal pellet feed. Model group and GXZT groups were injected with Vitamin D3(VD3) based on high-fat fedding and stimulated immune response method was used to establish AS models. Each group was given the corresponding drugs for 90 days. Blood serum were analyzed for total cholesterol(TC), triglyceride(TG), low density lipoprotein cholesterol(LDL-C) and high density lipoprotein cholesterol(HDL-C), calcium(Ca2+), superoxide dismutase(SOD), malondialdehyde(MDA), nitric oxide(NO), nitric oxide synthase(NOS),levels of endothelin(ET), interleukin-6(IL-6), tumor necrosisfactor-α(TNF-α), interleukin-18(IL-18), adiponectin(ADP), and soluble CD40L(sCD40L).Visceral index and arteriosclerosis index(AI) were calculated;the pathological observation of aorta was carried out by HE staining. Result: The liver index, AI, TC, LDL-C, Ca2+, MDA, ET, TNF-α, IL-18 and sCD40L were significantly higher(P<0.05) while the SOD, NO, NOS and ADP levels were lower in model group than those in normal group. Compared with those in model group, the liver index, AI, TC, and LDL-C in GXZT low and high dose groups were lower(P<0.05), while the serum NO and NOS levels were significantly increased(P<0.05). GXZT high dose group could significantly reduce serum ET, TNF-α, IL-6, IL-18 and sCD40L levels(P<0.05). HE staining showed aortic endothelial injury and atherosclerotic plaque formation in model group, and various treatment groups can significantly inhibit aortic lesions. Conclusion: GXZT has intervention effect on atherosclerotic plaque formation of model animals at multiple targets by improving metabolism of lipid in model rats, reducing the secretion or release of inflammatory cytokines, and regulating vascular active substances, but it has no significant effect on serum SOD and MDA levels.

      摘要:Objective: To conduct a comparative study on anti-inflammatory effect and its mechanism of Rheum tanguticum cultivated in Ganan state(RT-GN). Method: The SPF KM mice and SPF male Wistar rats were selected, and with the methods of xylene-induced mice ear swelling, formaldehyde-induced mice foot swelling and carrageenan-induced rats foot swelling, they were compared with high and low doses R. tanguticum cultivated in south of Gansu(4.5, 1.5 g·kg-1 for mice, and 3.0, 1.0 g·kg-1 for rats), high and low doses R. tanguticum cultivated in Xining(4.5, 1.5 g·kg-1 for mice, and 3.0, 1.0 g·kg-1 for rats) and high and low doses R. palmatum cultivated in Lixian County(4.5, 1.5 g·kg-1 for mice, and 3.0, 1.0 g·kg-1 for rats) for anti-inflammatory effects. Mice airbag synovitis was replicated, and the determination of total protein and leukotriene B4(LTB4) in the inflammatory exudate was done to comparatively investigate the anti-inflammatory mechanism of high and low dose rhubarb. Normal group was also set in the experiment. Result: Compared with the normal group, R. tanguticum cultivated in south of Gansu(RT-GN), R. tanguticum cultivated in Xining(RT-XN), and R. palmatum cultivated in Lixian(RP-LX) could remit degree of auricular swelling in mice and foot swelling in mice and rats(1.0, 3.0 g·kg-1)(P<0.05, P<0.01). Compared with the model group, 1.5, 4.5 g·kg-1RT-GN, RT-XN, and RP-LX could significantly reduce total protein and LTB4 in airbag synovitis exudates, and the effects of RT-GN and RT-XN on total protein and LTB4 in inflammatory exudates were superior to that of RP-LX(P<0.05, P<0.01). Conclusion: Three kinds of cultivated Rhubarb have obvious anti-inflammatory effect, part anti-inflammatory effect of R. Tanguticum was superior to that of R. palmatum, but there was no significant difference in anti-inflammatory effect between RT-GN and RT-XN, and one of their mechanisms is to inhibit the production of LTB4.

      摘要:Objective: To observe the time-toxicity relationship of geniposide on hepatotoxicity and nephrotoxicity in normal rats, and provide scientific basis for the clinical application of gardenia. Method: The 110 Wistar rats were randomly divided into normal group, different time groups(0.5, 1, 2, 4, 8, 12, 24, 48, 72 h group). The rats in normal group received normal saline by gavage, and the rats in other groups received 1.2 g·kg-1 geniposide by gavage. Orbital venous blood was taken at corresponding time points after gavage, and the serum was taken by centrifugation. aspartate amino transferase(AST), alkaline phosphatase(ALP), alkaline phosphatase(ALT), total bilirubin(TBIL), urea nitrogen(BUN), and creatinine(Cr) activities in serum were detected to indicate the toxicity of kidney and liver injury. Result: 12 h after administration of geniposide, AST, ALP,ALT, TBIL, BUN, and Cr levels were significantly higher than those in normal group(P<0.05,P<0.01). 24 h and 48 h after administration of geniposide, AST, ALP, ALT, TBIL, BUN, and Cr levels came to peak values. 72 h after administration of geniposide, AST, ALP, ALT, TBIL, BUN, and Cr levels began to significantly decrease. 240 h after administration, the values basically returned to normal levels. In histopathologic examination, different degree's infiltration of inflammatory cells in portal area, necrosis of liver cells, mild hyperplasia of bile duct in portal area, proliferation of fibrous tissue and other pathological changes were observe. Conclusion: Geniposide at dose of 1.2 g·kg-1 can cause acute liver and kidney injury on rats and show certain time-toxicity relationship.

      摘要:Objective: To investigate the clinical effect of Yiqi Guben Tiaomian decoction in throat-oriented treatment of IgA nephropathy patients with Qi and Yin deficiency syndrome. Method: Seventy-nine patients with IgA nephropathy were randomly divided into treatment group(39 cases, Yiqi Guben Tiaomian decoction) and control group(40 cases, Guben Tongluo decoction),with a two-month course of therapies. Result: ① Scores of throat in treatment group were significantly reduced(P<0.01), while there was no significant improvement in control group, with significantly statistical difference(P<0.01). ②Red blood cell count of urine and 24 h urinary protein excretion were reduced in control group(P<0.05), and significantly reduced in treatment group(P<0.01), with significantly statistical difference between two groups(P<0.01).The total effective rate was 84.6% in treatment group and 57.5% in control group, with significantly statistical difference between two groups(P<0.01). ③The positive relationship between the scores of throat and red blood cell count of urine(P<0.01) as well as the level of 24 h urinary protein excretion(P<0.05) was found. Conclusion: Yiqi Guben Tiaomian decoction is effective to throat-oriented treatment of IgA nephropathy with Qi and Yin deficiency syndrome and improve the symptoms of IgA nephropathy by clearing heat and relieving sore-throat, which may provide new insight for the therapy of IgA nephropathy.

      摘要:Objective: To observe the curative effect of Huazhuo granule prepared with traditional Chinese medicine(TCM) turbidity toxin theory on type 2-diabetes mellitus(T2DM) combined with nonalcoholic fatty liver(phlegm-dampness accumulating spleen type). Method: The 90 cases were randomly divided into treatment group(45 cases) and control group(45 cases). Both groups received basic therapy plus western medicine hypoglycemic therapy, while treatment group also received Huazhuo granule for 2 treatment courses, 3 months a course. Changes in TCM clinical symptoms before and after treatment in two groups, and changes in blood glucose, glycosylated hemoglobin(HbA1c), blood lipid, liver enzymes, fasting insulin(FINS) release level, insulin resistance index(HOMA IR), body mass index(BMI) and other indicators were observed. Result: Comparison of clinical effect between two groups:the total effective rate was 87.80% in treatment group and 53.84% in control group(P<0.05). Comparison of TCM symptoms:the total effective rate was 78.04% in treatment group and 64.10% in control group(P<0.05). Comparison of the conditions before and after treatment:P<0.05 for fasting plasma glucose(FPG), 2-hour postprandial blood glucose(2 hPBG), and HbA1c;P<0.01 for fasting insulin(FINS), HOMA-IR;P<0.05 for cholesterol(TC), triglyceride(TG), low density lipoprotein cholesterol(LDL-C), high-density lipoprotein cholesterol(HDL-C);P<0.01 for alanine aminotransferase(ALT), aspartate amino transferase(AST) in treatment group;P<0.05 for ALT and AST in control group;P<0.05 for body mass index(BMI). Comparison between treatment group and control group after treatment:P<0.05 for FPG and HbA1c,P<0.05 for FINS and HOMA-IR,P<0.05 for ALT and AST,P<0.05 for BMI. Conclusion: Compared with the treatment with western medicine alone, treatment with Huazhuo granule combined with western hypoglycemic medicine achieves better efficacy for T2DM of phlegm-dampness accumulating spleen type combined with nonalcoholic fatty liver.

      摘要:Objective: To observe the effect of Heidihuang Wan on spleen and stomach pattern, nutritive status and gastrointestinal hormone in chronic renal failure(CRF). Method: The CRF patients were randomly divided into two groups, 59 cases in observation group and 59 cases in control group. On the basis of general treatment, the patients in observation group were also treated with Heidihuang Wan, 9 g/time, 3 times/day. The patients in control group were treated with Niaoduqing granules 9 g/time, 3 times/day. The treatment course was 8 weeks in both groups. The scores of spleen and stomach pattern before and after treatment were graded;changes in the index of hemoglobin(Hb), albumin(ALb), prealbumi(PA), total cholesterol(TC), the level of gastrin(GAS), motilin(MTL), somatostatin(SS), Leptin and neuropeptide Y(NPY) before and after treatment were detected. Result: The total effective rate of spleen and stomach pattern in the observation group was 81.35%, superior to 67.80% in the control group(P<0.05). After treatment, the level of Hb was significantly higher(P<0.05), and the result in the observation group was better than that in the control group(P<0.05). After treatment, ALb and PA levels in both groups were increased obviously(P<0.05) and the increase in the observation group was more significant(P<0.05). After treatment, levels of GAS and MTL were decreased in both groups(P<0.05). After treatment, SS level was significantly increased in the observation group(P<0.05), which was more significant than that in the control group(P<0.05). After treatment, levels of Leptin and NPY were decreased in the observation group(P<0.05), which was more significant than that in the control group(P<0.05). Conclusion: Heidihuang Wan can well relieve TCM pattern of the spleen and stomach, and benefit the nutrient state of the patients with CRF. Its mechanism may be related to improving the gastrointestinal function disturbance.The methods of reinforcing spleen(stomach) has clear short-term effect in the treatment of CRF.

      摘要:Objective: To discuss the clinical efficacy of Fuzheng Yiliu decoction with ultrasound introduction as an adjuvant treatment for primary liver cancer and its influence on serum golgi glycoprotein-73(GP-73) and glypican-3(GPC-3). Method: One hundred and twenty primary liver cancer patients were included in the study. According to different therapeutic methods, patients were randomly divided into the control group and treatment group, 60 cases in each group. In the control group, patients were treated with hepatic arterial chemoembolization. Based on the treatment of the control group, patients in treatment group were treated with additional Fuzheng Yiliu decoction with ultrasound introduction(1 time/d) 3 days after surgery in control group, for 12 continuous weeks. Changes of main symptoms, liver function indexes and T lymphoid subgroups were compared before and after treatment. Levels of GP-73 and GPC3 in the serum of both groups were detected. Result: The total effective rate of the treatment group was 86.67%, significantly higher than 58.33% in the control group after treatment(P<0.05). Main symptoms and liver function indexes alanine aminotransferase(ALT), aspartate amino transferase(AST), γ-glutamine(γ-GT) and total bilirubin(TBIL) in the treatment group were significantly lower than those of the control group after treatment(P<0.01). The levels of CD3+, CD4+and CD4+/CD8+ in the treatment group were higher than those of the control group after treatment, while the level of CD8+ in the treatment group was lower than that in control group(P<0.01). After treatment, the levels of GP-73 and GPC3 in the serum of the treatment group were significantly lower than those in the control group, with statistical significance(P<0.01). Conclusion: Fuzheng Yiliu decoction with ultrasound introduction as an adjuvant treatment for primary liver cancer could evidently improve the clinical symptoms and quality of life, inhibit serum levels of GP-73 and GPC3, and improve immune and liver functions.

      摘要:Objective: To observe the clinical effect of Chushi Tongbi decoction combined with traction treatment in lumbar disc herniation(LDH)(moist heat arthralgia spasm syndrome) and observe the life quality. Method: One hundred and nine eligible patients were randomly divided into control group(54 cases) and observation group(55 cases) by visiting sequence. Patients in control group received etoricoxib tablets, 1tablet/time, 1 time/day, and combined with traction treatment. Patients in observation group received Chushi Tongbi decoction combined with traction treatment. Courses of treatment were 3 weeks in both groups. Pain was evaluated by McGill pain questionnaire(MPQ),function was evaluated by Oswestry disability index(ODI), activities of lumbar joints were measured;scores of activities of daily living(ADL) and main symptoms and sings were graded. The above indexes were evaluated once before and after treatment. Result: The total effective rate in observation group was 94.55%, superior to 81.48% in control group(P<0.05). After treatment, scores of pain feeling(PRI A), pain emotion(PRI S) and total scores of pain(PRI T), visual analogue scale(VAS) and present knee pain condition(PPI) in observation group were all lower than those in control group(P<0.01). After treatment, ODI total scores, scores of pain, single item scores and activities of daily living in observation group were all lower than those in control group(P<0.01). Compared with the conditions before treatment, both groups' activities of flexion and extension of lumbar joints were increased(P<0.01), which was more significant in observation group(P<0.01). After treatment, score of ADL in observation group was higher than that in control group, and the scores of main symptoms and signs were lower than those in control group(P<0.01). Conclusion: Chushi Tongbi decoction combined with traction treatment can relieve patients' symptoms, ameliorate activities of lumbar joints, and improve patients' quality of life, with significant clinical effect.

      摘要:Objective: To observe the therapeutic efficacy of Tongfu Xingshen decoction on the recovery of nerve function after minimally invasive surgery for hypertensive cerebral hemorrhage(phlegm-heat fu organ sthenia and wind phlegm blocking) and observe its influence on the serum level of interieukin-6(IL-6), tumor necrosis factor-α(TNF-α), and matrix-metalloproteinase-9(MMP-9). Method: Ninety-six patients with hypertensive cerebral hemorrhage surgery were randomly divided into control group and treatment group(48 cases in each group) by admission sequence. Based on routine therapy, patients of control group received edaravone therapy,ivgtt. Based on the treatment of control group, patients in treatment group were also treated with additional Tongfu Xingshen decoction. Course of treatment was 6 weeks for both groups. The Glasgow coma scales(GCS), neurological deficit score, and Barthel index were compared between two groups. Serum IL-6, TNF-α, and MMP-9 levels before and after treatment were detected. Result: The clinical efficacy of treatment group was superior to that in control group by ridit analysis(P<0.05). After treatment of 1 and 2 weeks, GCS scores of both groups were continuously increased(P<0.01), and GCS scores in treatment group were significantly higher than those in control group(P<0.01). After treatment, Barthel Index scores in treatment group were higher than those in control group, and National Institute of Health Stroke Scale(NIHSS) scores were lower than those in control group(P<0.01). After treatment, serum levels of IL-6, TNF-α and MMP-9 in treatment group were lower than those in control group(P<0.01). Conclusion: Tongfu Xingshen decoction combined with intravenous drip of edaravone has good effect on the recovery of nerve function after minimally invasive surgery for hypertensive cerebral hemorrhage(phlegm-heat fu organ sthenia and wind phlegm blocking), and its mechanism may be related with inhibiting the inflammatory response and MMP-9.

      摘要:Objective: Angelicae Sinensis Radix(ASR) is commonly used in compatibility with many other herbs, reflecting good synergism and restriction functions, thus forming a series of herb pairs. This study is aimed to investigate the potential rules of herb pairs containing ASR. Method: We retrieved 16529 formulaes which contain ASR from the 'Dictionary of Traditional Chinese Medicine Formulae', then built the database according to the frequency statistical method, and analyzed the compatibility proportion, corresponding indicated diseases and departments. Result: There were 170 herbs(compatibility frequency>200 with ASR) and even 22 herbs(compatibility frequency>1500). Further analysis of these 22 ASR series prescriptions showed that their compatibility proportion 1:1 was the most common, followed by 1:2 and 3:2.In addition, they were most frequently used for the treatment of the diseases in internal medicine and gynecology. Conclusion: In this paper we first formulate the data of series of herb pairs containing ASR in traditional Chinese medicine clinical application and find some regular characteristics of herb pairs containing ASR in clinical prescription, which provide the guidance and reference for the modern research and application of relative herb pairs and formulae.

      摘要:Objective: To technically analyze the prescription rules in Uighur medical treatment of the coronary heart disease by means of Data mining for high-frequency medicine pairs and efficacy compatibility, reveal the inherent rules, explore the theory of prescription composition,guide proper clinical application, and provide prescriptions clues for new drug research and development. Method: With ancient prescriptions of physicians of past generations as the clue, and with the help of the modern data processing and statistical computer technology, the Microsoft Excel database of coronary heart disease(CHD) was established. By advanced filtering functions, statistical analysis was taken for the high-frequency herbal pairs and efficacy compatibility. Result: Known and unknown synergized and attenuatedherbal combinations were found through data mining and analysis,high-frequency herbal pairs included Anchusae Herba-Dracocephali Moldavicae Herba,Anchusae Herba-Serica Gluma/Rosa Rugosa Thunb,Anchusae Herba-Santali Albi Lignum.High-frequency 3-herbs groups included Anchusae Herba-Dracocephali Moldavicae Herba-Serica Gluma,Anchusae Herba-Dracocephali Moldavicae Herba-Santali Albi Lignum/Rosa Rugosa Thunb,Anchusae Herba-Serica Gluma-Santali Albi Lignum.High-frequency 4-herbs groups included Anchusae Herba-Dracocephali Moldavicae Herba-Serica Gluma-Santali Albi Lignum,Anchusae Herba-Dracocephali Moldavicae Herba-Succinum-Moschus.High-frequency 5-herbs groups included Serica Gluma-Margarita-Succinum/Moschus-Limonium Gmelinii-Centaurea Behen,Anchusae Herba-Dracocephali Moldavicae Herba-Serica Gluma/Margarita-Limonium Gmelinii-Centaurea Behen.The best combinations of efficacy compatibility were explored. Conclusion: The results suggest that cardiotonic drug was used as the main drug,with compatibility of brain tonifying,natural force boosting drug,tendon strengthening drug,kidney-reinforcing drug,stomach-invigorating drug and corresponding Munziq of Abnormal body fluids in the prescriptions for CHD,Basic prescriptions were constituted Serica Gluma,Santali Albi Lignum,Margarita,Moschus,Succinum,Limonium Gmelinii,Centaurea Behen,Silver Foil,Gold Foil,Croci Stigma,Anchusae Herba,Dracocephali Moldavicae Herba,Caryophylli Flos,Curcumae Radix,Doronicum Hookarii,and Bambusae Concretio Silicea.The deduction Provides a reference base for clinical making prescriptions and new drug research and development.

      摘要:Objective: To analyze and summarize the prescription rules of professor XUE Bo-yu in treating patients with chronic hepatitis C. Method: All 149 cases of chronic hepatitis C treated by professor XUE Bo-yu were collected. The herbs meeting the inclusion criteria in the selected cases were recorded into Medcase V3.2 item by item. Association rules and cluster analysis were used to determine the use frequency of herbs in the prescriptions, common herbs combinations and prescription features, and explore the academic thoughts of professor XUE Bo-yu in treating patients with chronic hepatitis C. Result: The 149 medical cases were included in the study, involving 176 herbs. 26 kinds of herbs such as Salviae Miltiorrhizae Radix et Rhizoma, Rehmanniae Radix, Paeoniae Radix Rubra and Bubali Cornu were commonly used. According to association rules, the commonly used herbs combinations were ①Bubali Cornu, Salviae Miltiorrhizae Radix et Rhizoma, ②Rehmanniae Radix, Paeoniae Radix Rubra ③Moutan Cortex, Salviae Miltiorrhizae Radix et Rhizoma, Bubali Cornu. Xijiao Dihuang decoction could be used as a basic prescription to treat chronic hepatitis C. Cluster analysis showed that the herbs commonly used were for clearing heat and detoxifying, activating blood and dissolving stasis, and considering other accompanying symptoms at the same time. Conclusion: Prescription rules of professor XUE Bo-yu in treating patients with chronic hepatitis C can be found out by data mining technique. It can provide a reference and objective basis for future clinical treatment.

      摘要:Compound solubility and permeability play an important role in evaluating oral absorption of drug candidates as well as their Biopharmaceutics Classification System(BCS). Based on the characteristics of multicomponent of traditional Chinese medicine(TCM), the BCS of Chinese materia medica(CMMBCS) is more suitable for TCM research. The popularization of the spectral correlation technique provides a new method for finding the active ingredients of TCM. Taking intestinal absorption spectrum as the breakthrough point, it avoids the problems of the complex distribution of serum spectrum, as well as the metabolic process, and is not easy to omit the non-absorbent effective components, which is more convenient and more suitable for the study. The single-pass intestinal perfusion(SPIP) model is generally recognized as the model for in situ drug absorption study, which is able to correctly simulate the in situ absorption conditions of oral drugs. Combined with mesenteric blood taking method, it can reflect the actual composition absorption into the blood, more suitable for the study of CMMBCS and spectrum efficiency. However, SPIP actually reflects the apparent absorption of each component in compound, and a single composition of SPIP and in vivo absorption research should be combined for further absorption mechanism. It is necessary to study the absorption characteristics in multicomponent environment on the basics of single component research, and then return to TCM or compound for the whole research. In recent years, SPIP has been widely used in TCM research. In the literatures of recent 20 years, more than 200 articles were analyzed to summarize its applications in TCM, and combined with the experience of research team, it could be used to summarize the effective components, single herb, pair-herb compatibility, application in Chinese medicine compound, and the notice items in use, in order to provide reference for further application in TCM research.

      摘要:Objective: Stroke can, to a certain extent, promote neurogenesis, but this effect is limited. Promoting neurogenesis by drugs is an important way for treatment of stroke. In recent years, tetramethylpyrazine(TMP) has received the attention because of its neuroprotection effect, and widely been applied in the prevention and treatment of cardiovascular and cerebrovascular diseases, especially stroke. The effect of TMP on neurogenesis after cerebral ischemia was accessed from Databases, such as Pubmed. In this article, we summarized, analyzed and reviewed the literature. The results showed that TMP significantly promoted neurogenesis by increasing the proliferation, differentiation, especially migration of neural stem cells after cerebral ischemia. This article summarized the possible mechanism of TMP in the prevention and treatment of stroke, also dprovide some useful hints for expand the application of TMP in other neurogenesis disorder related diseases.

      摘要:This article uses literature summary method to analyze relationship between mitochondrial structure,membrane permeability,oxidative damage,respiratory function,enzyme activity,mitochondria biosynthesis and skeletal muscle aging,it explores influence of Bushen Yiqi prescription on mitochondria structure and function of ageing skeletal muscle,in order to provide theory basis for taking appropriate traditional Chinese medicine to retard skeletal muscle ageing.It is found that,as body gradually aging,mitochondrial structure in skeletal muscle became disorder,permeability of mitochondrial membrane changed,mitochondria suffers oxidative damaged,respiratory function,enzyme activity and mitochondria biosynthesis decreased,Bushen Yiqi prescription can retard skeletal muscle aging through improving mitochondrial structure and function of aging skeletal muscle,but its deep mechanism of action was not clear.How to combine modern experimental technology,under guidance of theory system of traditional Chinese medicine,multi-angle and multi-level to ascertain mechanism and structure-activity relationship of Bushen Yiqi prescription in retarding skeletal muscle aging is the key issue to be solved in process of developing this kind of herbs and formulae.

      摘要:To sort out and analyze the relevant literature of Hui medical treatment for stroke. With Hui medicine, Hui drug and stroke as the keywords, CNKI and Wanfang Database were searched for the recent 10 years literature. More than one hundred literature of the stroke were sorted out and analyzed for understanding and treatment advance. The theory of Hui medicine for stroke, treatment, herbal prescriptions and modern medicine research, including clinical studies, animal experiments and single herb research were introduced. Clinical studies included nasal drops, drug-oxygen therapy, Habi Faer Feirong pill and so on. Animal research is currently limited to Zhali Nusi formula and Mijian Changpu formula. Single herb research was focused on the aromatic drug research. It was to explore the unique advantages of Hui medicine in the treatment of stroke, and provide reference for the future research and application of hui medicine in treating stroke. In the treatment of ischemic stroke and its sequelae, Hui medicine showed its unique advantages in improving blood supply to the brain tissues, protecting neurons and regulating the blood-brain barrier and other functions, but its protective effect on the brain mechanism remained to need more in-depth research. Further discussion of Hui medicine's effect on protecting the brain tissues and improving the mechanism of cerebral blood flow, as well as the development of Hui medicine compound prescriptions have broad prospects in the research field of Hui medicine for preventing and treating stroke.
