      摘要:Objective: To investigate cytotoxicity and cellular uptake of brucine-loaded solid lipid nanoparticles (B-SLN) on HepG2 cells.Method: 3-(4, 5-Dimethylthiazole-2-yl)-2, 5-diphenyltetrazolium-bromide (MTT) assay was used to detect the cytotoxicity of B-SLN on HepG2 cells, the cellular uptake experiments were observed by fluorescence microscopy, flow cytometry was used to detect the cellular uptake of cells under different conditions.Result: Both brucine solution group and B-SLN group had an inhibitory effect on the proliferation of HepG2 cells, and the inhibition rate increased with increasing of the time and the concentration of brucine, half-inhibitory concentration (IC50) of brucine group in 48, 72 h were 208.5, 78.5 mg·L-1, IC50 of B-SLN group in 48, 72 h were 563.3, 114.9 mg·L-1.When the concentration of brucine was in the range of 125-500 mg·L-1, with increasing of the drug concentration, the cellular uptake increased from 50.2% to 71.2%;when the temperature was under the condition of 4℃ and 37℃, the cellular uptake increased from 43.0% to 55.2%;within uptake time of 30-240 min, with the incubation time increasing, the cellular uptake amount increased from 9.7% to 56.4%.Conclusion: SLN administration system can significantly improve the activity of brucine against cancer cells, B-SLN has the ability to increase cellular uptake of drugs.

      摘要:Objective: To discuss the change law of chemical constituents in Phellodendri Amurensis Cortex before and after being processed, and to provide the basis for the quality standard of its decoction pieces.Method: HPLC was performed by taking acetonitrile-0.4 mol·L-1 ammonium chloride solution as mobile phase, column temperture at 35℃, the flow rate of 1.0 mL·min-1 and the detection wavelength of 215 nm.Chemical constituents in Phellodendri Amurensis Cortex before and after being processed was analyzed and 7 main constituents were measured.Result: Among 10 batches of Phellodendri Amurensis Cortex decoction pieces, the contents of magnoflorine, phellodendrine, jatrorrhizine, palmatine, berberine, obaculactone and obakunone were 2.468-6.783, 1.947-4.197, 0.209-0.506, 3.049-4.557, 5.869-13.168, 5.141-7.337, 1.798-4.975 mg·g-1.After being processed, in addition to a sample, all the constituents reduced, among which alkaloids like berberine and palmatine, almost disappeard, however, obakunone and obaculactone existed.Among 9 batches of Phellodendri Amurensis Cortex decoction pieces, the contents of obaculactone and obakunone were 3.164-6.483, 0.281-1.614 mg·g-1.Conclusion: The appearance and internal components of Phellodendri Amurensis Cortex after being processed obviously change.The quality standard, which applies to Phellodendri Amurensis Cortex, can't apply to charred Phellodendri Amurensis Cortex decoction pieces.So the process degree of samples is different, the process degree should be controlled according to the laws.

      摘要:Objective: Taking betaxolol hydrochloride as model drug, to prepare drug loading montmorillonite-chitosan nanoparticles by ionic gelation of chitosan with sodium polyphosphate (TPP), which is known simply as Mt-BH/CS NPs.Method: Mt-BH/CS NPs was prepared by ionic gelation, orthogonal test was adopted to optimize its formulation process by taking encapsulation efficiency and drug loading as indexes.Encapsulation efficiency, drug loading and in vitro release properties of Mt-BH/CS NPs were examined by UV method and its biological properties of nanoparticles suspension were investigated by mucoadhesive test and in vitro cytotoxicity test.Result: The best formulation process was as following:chitosan concentration of 1.5 g·L-1, TPP concentration of 1.2 g·L-1, chitosan-TPP (10:1), betaxolol hydrochloride concentration of 2 g·L-1.The average encapsulation efficiency of five batches of Mt-BH/CS NPs was 36.13% and its average drug loading was 14.50% with RSD<1.0% by dialysis method.The cumulative release rate of Mt-BH/CS NPs was 82.23% in 10 h.In the mucoadhesive test, after mixing with mucous membrane, the reduction of Zeta potential value of Mt-BH/CS NPs indicated that there was mucoadhesive property of Mt-BH/CS NPs.The cell viability of human immortalized corneal epithelial cells after being exposed to betaxolol hydrochloride solution and Mt-BH/CS NPs showed that cytotoxicity of betaxolol hydrochloride solution was higher than that of Mt-BH/CS NPs suspension.Conclusion: Mt-BH/CS NPs has a high drug loading, slow in vitro release, low toxicity and great mucoadhesive property, which may hold promise to be an attractive novel nanosystem for the treatment of ocular diseases.The new drug delivery system can improve the capability of release properties and enhance the interaction with the eye, so as to improve the ocular bioavailability of drugs and reduce their adverse reactions.

      摘要:Objective: To study on in vitro release and transdermal absorption properties of Huoxue Zhitong microemulsion gels.Method: Improved Franz diffusion cell was adopted to investigate in vitro release in semipermeable membrane and transdermal absorption in mouse skin of Huoxue Zhitong microemulsion gels.The contents of paeonol, eugenol and methyl salicylate in receptor fluid were determined by UPLC.Result: In 6 hours, the cumulative release rates of paeonol, eugenol and methyl salicylate were 70.35%, 59.40% and 54.64%, the transmission rates of them were 1.138, 11.57, 73.54 μg·cm-2·h-1, respectively.In 6 hours, the accumulative permeation rates of paeonol, eugenol and methyl salicylate were 40.34%, 36.34% and 41.44%, the release rates of them were 0.657, 7.127, 56.04 μg·cm-2·h-1, respectively.Conclusion: Huoxue Zhitong microemulsion gels has a good in vitro release and transdermal properties, the release laws of all indexes are in accordance with Higuchi equation and the transdermal actions are consistent with zero-order kinetic equation.

      摘要:Objective: To explore the exclusive methods for quality control of Lonicerae Japonicae Flos extract.Method: HPLC was employed with mobile phase of acetonitrile-0.4% phosphoric acid solution for gradient elution and detection wavelength at 327 nm.The quality of Lonicerae Japonicae Flos extract in Yinhuang preparations was analyzed, the content of chlorogenic acid and specific chromatogram of Lonicerae Japonicae Flos extract were compared with the extract and original medical herb of Lonicerae Flos and Eucommiae Folium.Result: The highest contents of chlorogenic acid in the original medical herbs and its extracts were Lonicerae Flos, mass fractions were 3.56%-5.09% and 4.62%-6.12%;there was no difference between Lonicerae Japonicae Flos extract and Eucommiae Folium extract, the content of chlorogenic acid was about 4%.After acidification, the contents of chlorogenic acid in Lonicerae Japonicae Flos extract, Lonicerae Flos extract and Eucommiae Folium extract were all increased.Specific chromatogram of extract and acid extract of Lonicerae Flos and Eucommiae Folium were the same as the standard chromatogram of Lonicerae Japonicae Flos extract.Some of Yinhuang preparations had been checked geniposidic acid from Eucommiae Folium and macranthoidin B from Lonicerae Flos.Conclusion: Lonicerae Japonicae Flos extract may be added to Lonicerae Flos and Eucommiae Folium, its quality control should be strengthened.

      摘要:Objective: To optimize the extraction process of volatile oil from Angelicae Sinensis Radix and fit its extraction kinetic model equation, and then to detect chemical components in volatile oil and aromatic water.Method: The improved extractor of volatile oil was adopted, extraction process of volatile oil from Angelicae Sinensis Radix was optimized by response surface methodology, then the extraction rate of volatile oil was fitted and the regression equations were calculated.Components in light oil, heavy oil and emulsified aromatic water during the extraction process of volatile oil were determined by GC-MS, in order to find out the chemical material base for formation of emulsification.Result: Optimal extraction process was as follows:extracting time of 8.55 h, solid-liquid ratio of 1:10, soaked 1.82 h;extraction rate of volatile oil was(0.70±0.01)%.Kinetic equations of optimal technology were V=0.72(1-e-0.000 74t) (R2=0.996 7) and ln(0.72-V)=-0.014 1t-0.349 3(R2=0.998 7).Components in aromatic water were different from those in volatile oil from Angelicae Sinensis Radix, especially for 2-vinyl-4-methoxyphenol.Conclusion: This optimized extraction process is feasible and stable, two extraction dynamics equations can be fitted to its extraction process, components in aromatic water have a larger difference with those in volatile oil.

      摘要:Objective: To optimize the extraction process of Xiao'er Wenli Zhixie plaster by supercritical CO2 fluid extraction technology and its matrix prescription.Method: Extracting amount of cinnamaldehyde was selected as the indictor, L9(34) orthogonal test was used to optimize supercritical CO2 fluid extraction technology of Xiao'er Wenli Zhixie plaster.Initial viscosity, peel strength and cohesive strength were selected as indictors, L9(34) orthogonal test was used to optimize matrix prescription.The uniformity of Xiao'er Wenli Zhixie plaster was investigated by the uniformity test.Result: Optimum extraction parameters were as follows:extraction temperature at 45℃, extraction pressure of 50 MPa, extraction time of 2.5 h, flow rate of CO2 of 20 kg·h-1.Optimum matrix prescription was as follow:polyvinyl alcohol-sodium polyacrylate-glycerol-carboxy methyl cellulose sodium (0.6:2.5:1.0:1.0).This prescription showed satisfactory performance of initial viscosity, peel strength and cohesive strength;its uniformity was 15, which was in line with regulation in the 2015 edition of Chinese Pharmacopoeia.Conclusion: The optimized extraction process is stable and available.The optimum matrix prescription of is simple and stable, which can be applied to industrial production of Xiao'er Wenli Zhixie plaster.

      摘要:Objective: To comprehensively evaluate the genetic diversity and medicinal quality of Belamcandae Rhizoma from different provenances and provide a theoretical basis for rational utilization of germplasm resources and fine variety breeding of Belamcandae Rhizoma.Method: The genetic diversity of 9 provenances of Belamcandae Rhizoma were analyzed by ISSR molecular markers, and the content of irisflorentin in the rhizome was determined by HPLC.Result: The contents of irisflorentin in 9 provenances of Belamcandae Rhizoma were among 0.12%-0.36%; the percentage of polymorphic loci (PPB) were 55.42%-90.36%; the coefficient of gene differentiation (GST) was 0.204 1 and the gene flow (Nm) was 1.951 5.Conclusion: The content of secondary metabolites of Belamcandae Rhizoma had little correlation to genetic diversity, but was more subjected to growing environments, so in the process of variety breeding for Belamcandae Rhizoma, more work shall be done to deal with germplasm selection, cultivation environment and cultivation methods synthetically.

      摘要:Objective: To explore the dynamic change characteristics of phylloplane microbe from Aconitum carmichaeli and its correlation with Aconitum downy mildew, with those cultivated in Jiangyou Sichuan as the research subjects.Method: The leaves with downy mildew disease at different levels were collected. Foliar microbes were extracted by combined shock shaker and ultrasound method. The count of bacteria and fungi were measured by dilution plate count method.Result: Aconitum downy mildew was associated with environmental temperature. The count of phylloplane bacteria and fungi in the leaves suffering from downy mildew of A. carmichaeli was increased as compared to healthy one, and the count of other fungi showed a more regular upward trend with the increasing degree of downy mildew disease of A. carmichaeli.Conclusion: Aconitum downy mildew from A. carmichaeli had rapid onset in its growth period at the end of March to the end of May, and the daily average temperature between 15℃ to 20℃ was more appropriate for the growth and reproduction of Aconitum downy mildew pathogens. Changes in count of phylloplane fungi from A. carmichaeli were positively correlated with the degree of Aconitum downy mildew disease.

      摘要:Objective: To study the effective hemostatic components of Loropetalum chinense.Method: With blood coagulation/bleeding time in mice as the primary outcome, the different extract parts (water, 30%, 60%, 80%, and 95% ethanol extracts) were tested for hemostatic activity. The compounds were isolated and purified by various chromatographic methods such as silica gel, ODS, Sephadex LH-20.Their structures were identified according to the physical and chemical properties and spectral data.Result: The results showed that 60% ethanol extracts had the shortest blood coagulation/bleeding time, with strongest anticoagulant effect. Nine compounds were isolated from 60% ethanol extracts and identified as tiliroside (1), 3-O-coumaroylquinic acid(2), kaempferol-3-O-β-galactopyranosyl-(1→6)-β-D-glucopyranoside(3), kaempferol-3-O-β-galactopyranoside (4), 5-O-coumaroylquinic acid methyl ester(5), 5-O-coumaroylquinic acid(6), 3, 5-O-diocaffeoylquinic acid(7), 4, 5-O-diocaffeoylquinic acid(8), and 3, 4-O-diocaffeoylquinic acid(9). All of the compounds except 1 and 4 were isolated from the plant for the first time.Conclusion: The 60%ethanol extracts of L.chinense have strong anticoagulant effect, and phenolic acids act as the main effective hemostatic component.

      摘要:Objective: To develop an HPLC method for quantitative determination of chlorogenic acid, scopoletin, resveratrol and morin in Mori Cortex.Method: An Eclipse-C18 column (4.6 mm×250 mm, 5 μm) was adopted with acetonitrile-0.01% phosphoric acid as the mobile phase for gradient elution. The flow rate was 1.0 mL·min-1. The detection wavelength was set at 285 nm and the column temperature was 30℃.Result: The calibration curves showed good linear relationship in the ranges of 6.4-76.8 mg·L-1 (r=0.999 9) for chlorogenic acid, 5.15-61.8 mg·L-1(r=0.999 6) for scopoletin, 1.11-13.32 mg·L-1 (r=0.999 6) for resveratrol and 2.76-33.12 mg·L-1(r=0.999 7) for morin. The average recoveries were between 98.4% and 99.4%(RSD≤2%). The sample solution was steady within 12 h, and this method showed good repeatability. Through the determination of 5 batches of Mori Cortex samples, results showed that the contents of chlorogenic acid, scopoletin and morin were higher in Henan samples (batch number 140401); and the content of resveratrol was higher in Hubei samples (batch number D5090202).Conclusion: The method is precise and accurate for determination of four ingredients in Mori Cortex.

      摘要:Objective: To study the correlation between color trait of Menispermi Rhizoma powder and contents of total alkaloids.Method: The value of ΔL*, Δa*, Δb* and ΔE* of Menispermi Rhizoma powder were determined by the colorimeter, and the contents of four chemical components (sinomenine, daurisoline, dauricine and tetrandrine) were detected by HPLC, then the data were analyzed by SPSS 15.0 software to explore the correlation.Result: The brightness value of ΔL*, total chromatic aberration value of ΔE* and the content of dauricine showed significant correlation (P<0.01); there was no significant correlation between all color indexes and contents of sinomenine, daurisoline and tetrandrine. However, color indexes of Δa*, Δb*, ΔE* had significant positive correlation with the total contents of daurisoline and dauricine and four kinds of alkaloids (P<0.05).Conclusion: The deeper orange red of Menispermi Rhizoma power and bigger total color difference, the greater total contents of daurisoline and dauricine and four kinds of alkaloids.

      摘要:Objective: To evaluate the medicinal quality of different cultivars (strains) of Angelica Sinensis Radix by TOPSIS and provide reference for controlling quality of Angelica Sinensis Radix.Method: With volatile oils, ferulic acid, butene phthalide, ligustilide, European angelica lactones A, polysaccharides, and flavonoids extract contents as the evaluation indexes, TOPSIS method was used to establish the comprehensive evaluation model-TOPSIS model for angelica medicinal quality based on the multiple index evaluation system.Result: Mingui No. 3, Mingui No. 4 and wild angelica herbs had the best quality, followed by angelica produced in Min county and Zhang county, as well as cultivated strain 2000-01 and Mingui No. 2, Mingui No. 5, and Mingui No. 1.The angelica produced in Qinghai had the worst quality. The content of volatile oil in Mingui No.4 was 1.48 times of that in Qinghai angelica, while ligustilide content was 1.69 times of that in Qinghai angelica.Conclusion: Through this research, it is suggested that the contents determination of butene phthalide and ligustilide, and characteristic fingerprint can be used as quality evaluation indexes of Angelica Sinensis Radix.

      摘要:Objective: To study the volatile oil and fat-soluble components of Alectoria jubata.Method: The volatile oil and fat-soluble components were analyzed by gas chromatography-mass spectrometry (GC-MS).Result: The 34 volatile oil compounds were identified in the extracts of A. jubata, accounting for 17.93% of the total constituents. Terpenes (6.71%), ketones (4.19%), alcohols (3.27%) were the main compound types in the volatile oil. Its major components included 4-penten-2-one(4.19%), cis-7-tetradecen-1-ol (3.03%), 3, 7-dimethyl-1-octene (1.30%), neohexane (1.28%) and germacrene D (1.01%). Meanwhile 33 fat-soluble components were identified in the extracts of A. jubata, accounting for 89.63% of the total constituents. Fatty acids (81.65%), alkanes (4.49%) and alcohols (1.67%) were the main compound types in the fat-soluble components. Its major components included linolelaidic acid methyl ester (34.98%), oleic acid methyl ester (10.94%), pentadecanoic acid 14-methyl-, methyl ester (7.40%) and octadecanoic acid methyl ester (4.13%).Conclusion: The volatile oil and fat-soluble components were extracted and the compounds were identified for the first time. This research could provide a scientific basis for further research and development of A. jubata.

      摘要:Objective: To develop a method for quantitative analysis of multiple-components by single marker (QAMS) for simultaneous determination of lovastatin and lovastatin acid in Fermentum Rubrum and its preparations.Method: Lovastatin was used as the internal reference substance, and the relative correction factor (RCF) of lovastatin acid was determined by RP-HPLC. Then the QAMS method was established for simultaneous determination of the two components. The durability of the established method was verified with varying HPLC instrument and chromatographic columns. Then the accuracy of the QAMS method was evaluated by comparing the contents of lovastatin acid with those obtained by the external standard method.Result: The RCF value showed good durability for both lovastatin and lovastatin acid. There were no significant differences between the quantitative results of QAMS method and external standard method.Conclusion: An accurate, rapid and highly durable QAMS method has been developed for simultaneous determination the contents and locations of lovastatin and lovastatin acid, so as to provide reliable basis for the application of QAMS method in quality control of Fermentum Rubrum and its preparations.

      摘要:Objective: To establish the HPLC fingerprint of Rosae Laevigatae Fructus from Guangxi and make cluster analysis and principal component analysis in order to provide a basis for its quality control.Method: ODS column was used with methanol -0.2% phosphoric acid solution as the mobile phase for gradient elution. The column temperature was 25℃; the flow rate of the mobile phase was 0.8 mL·min-1; the detection wavelength was 255 nm, and the injection volume was 10 μL. Similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine (2012.1 version) was used to confirm the common peaks in 10 batches of samples and evaluate the similarity. SPSS 22.0 statistical software was used to make cluster analysis and principal component analysis for the HPLC fingerprint.Result: A total of thirteen common chromatographic peaks were identified in the software of chromatographical evaluation, and ten peaks of them with good stability, strong absorption peak, and obvious characteristics as common peaks. By comparing with control products, three chemical components were identified as catechin, hesperidin and rutin. The fingerprint similarity was greater than 0.96 for 10 batches of Rosae Laevigatae Fructus samples. 10 batches of samples from different areas were classified into three groups based on cluster analysis, and the accumulative variance contribution was 82.98% for two principal components.Conclusion: The study can provide data support for improving quality standards of Rosae Laevigatae Fructus from Guangxi, providing basis for quality evaluation and exploitation.

      摘要:Objective: To study the components of essential oil from Zanthoxylum bungeanum and Z. schinifolium, evaluate the internal quality differences of the samples from different regions and sources, as well as provide scientific basis for the quality standarand research and quality assessment.Method: Essential oil was extracted by steam-stilling. The chemical components of volatile oil were detected by gas chromatography-mass spectrometry-automated mass spectral deconvolution & identification system (GC-MS-AMDIS) combined with Kováts retention index (RI).Result: A total of 62 components in the essential oils of 4 batches of Z. bungeanum were analyzed by GC-MS, and the top five components with high contents from high to low were linalool, linalyl acetate, α-terpineol, (-)-terpineol and limonene. A total of 32 components in the essential oils of the 4 batches of Z. schinifolium were identified, and the top five components with high contents from high to low were estragole, germacrene D, limonene, linalool, and caryophyllene.Conclusion: There were certain differeces in volatile oil components from differe regions and sources of pepper medicinal herbs, and particularly evident between different sources.

      摘要:Objective: To explore the effects of modified Zexietang on treatment of non-alcoholic fatty liver disease (NAFLD) in rats.Method: NAFLD rat model was established by high fat feeding (10% or 45% high fat feed) for 12 weeks and divided into 4 groups:control group, model group, modified Zexietang group and polyene phosphatidyl choline group. After the administration for four weeks, the corresponding volume of double stilled water was given for four weeks. Western blot was used to detect the expression of Toll like receptor 4 (TLR4), myeloid differentiation factor 88 (MyD88), nuclear factor-kappaB p65 (NF-κB p65), phosphorylated p65 (p-p65), p38 mitogen activated protein kinase (p38 MAPK), p-p38 (phosphorylated p38), c-Jun-NH2-terminal kinase (JNK) and cysteinyl aspartate-specificproteases-1 (Caspase-1) in liver.Result: Compared with control group, the above proteins in model group were significantly increased (P<0.05); and proteins in the modified Zexietang group were significantly decreased compared with model group (P<0.01).Conclusion: Modified Zexietang could inhibit liver inflammation and alleviate liver damage. Its mechanism may be related to the inhibition of the expression of protein in TLR4/NF-κB and MAPK signal pathways.

      摘要:Objective: To study the action mechanism of Yiqi Yangyin recipe with supplementing Qi and nourishing Yin)in treating leukemia by observing the effects of Yiqi Yangyin recipe and its separated recipes on expressions of Cytochrome C(Cyt-C), apoptotic protease activating factor 1(Apaf-1), the second mitochondria-derived activator of caspase/direct IAP-blinding protein with low PI(Smac/Diablo), apoptosis-inducing factors(AIF) in acute myeloid leukemic cells.Method: NOD/SCID mice were taken as the research platform and injected with KG-1a cells through tail vein to establish the model of human acute myeloid leukemia. The model mice were randomly divided into Quanfang group, Fuzheng group, Quxie group and control group. The spleen cell suspensions of NOD/SCID mice were prepared to measure the cell apoptosis rates by flow cytometry. The immunohistochemistry was used to detect the expression of the mitochondrial-related apoptosis factors in bone marrow of mice, including Cyt-C, Apaf-1, Smac/Diablo and AIF.Result: After treatment, the survival time of treatment groups were significantly longer than that of control group(P<0.01), the survival time of Quanfang group mice were longer than that of Fuzheng group and Quxie group(P<0.01), the survival time of Quxie group mice were longer than that of Fuzheng group mice(P<0.01). The cell apoptosis rate in all treatment groups was significantly higher than that of control group(P<0.01), the cell apoptosis rate of Quanfang group were higher than that of Fuzheng group and Quxie group(P<0.01), and the cell apoptosis rate of Quxie group were higher than that of Fuzheng group(P<0.01). With respect to the expressions of Cyt-C, Apaf-1, Smac/Diablo and AIF in bone marrow of mice, the treatment groups were higher than control group(P<0.05), particularly Cyt-C and Apaf-1(P<0.01). All of the above expressions of Quanfang group were higher than those of Fuzheng group and Quxie group(P<0.01). Quxie group was higher than Fuzheng group(P<0.05), particularly Cyt-C, Apaf-1 and Smac/Diablo(P<0.01). In terms of the effect on each tested indicator, Quanfang group was better than Fuzheng group and Quxie group, and Quxie group was better than Fuzheng group.Conclusion: Yiqi Yangyin recipe can increase the cell apoptosis rate in NOD/SCID, improve the expression of the mitochondrial-related apoptosis factors Cyt-C, Apaf-1, Smac/Diablo and AIF, induce apoptosis of leukemia cells and inhibit the proliferation of leukemia cells, which may be associated with the mitochondrial apoptosis pathway.

      摘要:Objective: To study the mechanism of Breviscapine on cerebral ischemia reperfusion injury of rats by cytochrome (CYP) 4A1, CYP4A3, CYP4A8 pathways.Method: Totally 193 male SD rats were selected to establish the model of middle cerebral artery occlusion (MCAO). After cerebral ischemia for 2 h and reperfusion, they were injected with Breviscapine at the doses of 12, 6, 3 mg·kg-1, qd, for 7 days. After the final medication, tissue fluids isolated from brain tissues were prepared. Genes expressions of CYP4A1, CYP4A3 and CYP4A8 in brain tissues of rats were examined by Real-time PCR assays. The content of endothelial nitric oxide synthase (eNOS) was examined by enzyme linked immunosorbent assay(ELISA).Result: Compared with shame group, model group showed significant increase in motility (P<0.01), volume of cerebral infarction and behavioral score (P<0.01) at 168 h, and up-regulation in the expressions of CYP4A3, CYP4A8 (P<0.01). The content of eNOS was increased in brain tissues and serum. Compared with model group, Bre 12 mg·kg-1 group and Bre 6 mg·kg-1 group showed significant reduction in motility (P<0.05), volume of cerebral infarction (P<0.01) at 168 h, behavioral score (P<0.01) after 24 h, and down-regulation in the expressions of CYP4A3, CYP4A8 (P<0.05, P<0.01) and the content of eNOS (P<0.01) in brain tissues and serum. Bre 3 mg·kg-1 group showed significant reduction in the behavioral score (P<0.01) at 120 h and the content of eNOS (P<0.01), and down-regulation in the expression of CYP4A3, CYP4A8 (P<0.01) in brain tissues.Conclusion: Motility, volume of cerebral infarction and behavioral score were increased by cerebral ischemia reperfusion injury, the expression of CYP4A3, CYP4A8 were up-regulated, and the content of eNOS was increased in brain tissues. Breviscapine injection could cure cerebral ischemia reperfusion injury of rats by reducing the volume of cerebral infarction, behavioral score, which may be correlated with down-regulation in the expressions of CYP4A3, CYP4A8 in brain tissues.

      摘要:Objective: To investigate the effect of combination of Glycyrrhizae Radix et Rhizoma and Genkwa Flos with different proportions in resisting hepatocellular carcinoma (HCC) ascites in mice.Method: A total of 135 male Kunming mice were randomly divided into normal control group, model group, Genkwa Flos and Glycyrrhizae Radix et Rhizoma combination groups(ratios of 1:0.25, 1:0.5, 1:1, 1:2, 1:4), pure Genkwa Flos group and pure Glycyrrhizae Radix et Rhizoma group. HCC ascites model was established by inoculating H22 cells intraperitoneally in all groups except for the normal group. HCC ascites mice in drug-treatment groups were intragastrically administrated with the corresponding drugs, while the normal and model groups were given the equal volume of normal saline. At 12 d, ascites amount, abdominal girth, weight, and liver and kidney indicators were measured respectively. Expressions of aquaporin-2 (AQP2) and renal vasopressin receptors-2 (V2R) in the kidney tissues were detected by Western blot. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN) and creatinine (Cr) were determined by the biochemistry analyzer.Result: The model group showed significant higher weight, abdominal girth, ascites, liver index, ALT, AST, BUN, Cr and lower kidney index than normal control group (P<0.01), with notable rise in AQP2 and V2R protein expressions. Compared with the normal group, the pure Genkwa Flos group and Genkwa Flos and Glycyrrhizae Radix et Rhizoma groups showed significant reduction in weight, abdominal girth, ascites and expressions of aquaporin-2 and renal vasopressin receptors-2 in the kidney tissues (P<0.05). Genkwa Flos and Glycyrrhizae Radix et Rhizoma group (1:1) also showed dramatic increases in weight, abdominal girth, ascites and expressions of aquaporin-2 and renal vasopressin receptors-2 in the kidney tissues (P<0.05).Conclusion: The combination of Genkwa Flos and Glycyrrhizae Radix et Rhizoma with different proportions could increase or decrease the effect of Genkwa Flos in resisting HCC ascites in mice. The underlying mechanism might be correlated with the effective regulation of aquaporin-2 and renal vasopressin receptors-2 protein expression.

      摘要:Objective: To study the regulatory effect of ultra-filtration extract from Angelice Sinensis Radix and Hedysari Radix(UFEASH) on transforming growth factor-β1(TGF-β1) and Smad signaling pathway of kidney tissue of rat with diabetic nephropathy (DN), and explore its possible mechanism.Method: The rat model with DN was induced by one-time high-dose (60 mg·kg-1) intraperitoneal injection with streptozotocin (STZ). All experimental rats were divided into blank group, model group, irbesartan group and low, median, high-dose UFEASH groups, and then intragastrically administrated once a day. Fasting blood glucose, urea nitrogen (BUN), serum creatinine (SCr), 24 h urinary protein were detected after 8 weeks. The changes in renal tissue structure were observed under light microscope. TGF-β1 and smad2/3 content of the rats were tested by ELISA. The expression of TGF-β1, Smad2/3 protein and mRNA expressions in renal tissues were detected by immunohistochemistry and Real-time PCR, respectively.Result: Compared with the normal group, the model group showed significant increases in FBG, BUN, SCr and 24 h urinary protein; ELISA showed notable increases in TGF-β1 and Smad2/3 content, remarkable renal pathological structural abnormality and notable increase in TGF-β1 and Smad2/3 protein expressions (P<0.05, P<0.01). Compared with the model group, UFEASH groups significantly reduced the level of FBG, BUN, SCr and 24 h urine protein of rats with DN (P<0.05). The levels of TGF-β1 and Smad2/3 mRNA expressions were also decreased (P<0.05). The abnormal structure of renal tissue was alleviated. The expression of TGF-β1 and Smad2/3 were reduced (P<0.05, P<0.01).Conclusion: UFEASH can improve renal function in rats with DN and delay the chronic pathological progress of DN, which may be related to the inhibition of TGF-β1/Smad signaling pathway.

      摘要:Objective: To investigate the effects of Zhengan Xifeng Tang on the irritable degree and conjunctival congestion in Parkinson's disease (PD) rats with syndrome of hyperactivity of liver-yang, and the possible mechanisms.Method: A total of 105 male Wistar rats were randomly divided into the normal control group, sham-operated group, model group, selegiline group (0.9 mg·kg-1·d-1), and high, medium and low-dose Zhengan Xifeng decoction groups (32, 16, 8 g·kg-1), with 15 in each group. Rats were given 6-hydroxydopamine (6-OHDA) through intra-substantia nigra injection and Fuzitang through intragastric administration for four weeks to prepare rat model of PD with syndrome of hyperactivity of liver-yang. Caspase-3 activities were measured by using spectrophotometry. Real-time PCR and Western blot were used to analyze tyrosine hydroxylase (TH) and NADPH quinone oxidoreductase-1(NQO-1) mRNA and protein expressions.Result: Compared with sham-operated group, model group showed significant increases in irritable degree and conjunctival congestion, and NQO-1 mRNA and protein expressions in midbrain tissues, and decrease in TH protein expression (P<0.05). Compared with the model group, Zhengan Xifeng Tang group showed significant decreases in irritable degree and conjunctival congestion and increases in TH protein expression and NQO-1 mRNA and protein expressions (P<0.05). However, the above-mentioned markers were unaffected in selegiline group.Conclusion: Effect of liver-wind-tranquilizing of Zhengan Xifeng Tang may be correlated with up-regulation in gene expression of TH and NQO-1 in the midbrain of PD rats with syndrome of hyperactivity of liver-Yang.

      摘要:Objective: To compare the effects of Yinchenhao Tang compound decoction dispensing granules, single dispensing granules and traditional decoction on carbon tetrachloride induced liver injury in mice.Method: Ninety mice were randomly divided into blank group, model group, positive group, Yinchenhao Tang compound decoction dispensing granules high, medium and low dose groups (17.16, 8.58 and 4.29 g·kg-1), single dispensing granules high and medium dose groups (17.16 and 8.58 g·kg-1), and traditional decoction group (4.29 g·kg-1). The mice in blank group and model group received water by intragastric administration everyday, and the mice in other seven groups were treated with prophylactic administration for 7 days. After intragastric administration at 7th day, all the groups except blank group received 0.05% carbon tetrachloride by intraperitoneal injection to establish acute liver injury models. The activities or contents of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), total protein (TP), albumin (ALB) and total bilirubin (TBIL) via eye socket were detected after 18 hours. Mice livers were taken for HE staining to observe the histopathological changes; liver homogenate was taken to test superoxide dismutase (SOD) and malondialdehyde(MDA) contents. The changes of main active ingredients among three different preparation methods were simply compared.Result: All of Yinchenhao Tang compound decoction dispensing granules, single dispensing granules and traditional decoction can reduce the degree of CCl4-induced liver injury; while the effects of Yinchenhao Tang compound decoction dispensing granules high dose group were superior to the single dispensing granules and traditional decoction groups(P<0.05, P<0.01).Conclusion: Yinchenhao Tang can protect the carbon tetrachloride induced liver injury. Effect of compound decoction was better than single dispensing granules possibly due to the different active principle dissolution rate in combined frying process. As compared with traditional decoction, the compound decoction had better effect possibly due to higher effective composition content with optimization process. Its mechanism for protecting liver may be related to scavenging free radicals and inhibiting lipid peroxidation.

      摘要:Objective: To investigate the effects of Guizhi Fuzitang on mitogen-activated protein kinase (MAPK) signaling pathway in the synovial tissue of rats with collagen-induced arthritis (CIA).Method: The 60 SD rats were randomly divided into normal group, model group, Guizhi Fuzitang group (1.5, 4.5, 13.5 g·kg-1) and methotrexate group. The rats were injected subcutaneously with chicken typeⅡcollagen (CCⅡ) through tail root to establish CIA model. Since the 14th day after modeling, the arthritic index of all groups at different time points were observed and measured. The expressions of JNK, ERK and p38 proteins in MAPK signal transduction pathway as well as the expression of corresponding phosphorylated proteins p-JNK, p-ERK and p-p38 were detected by western blot.Result: The swelling degree and arthritis index of the CIA rats were higher than the normal group at 7 th, 14 th, 21 th, 28 th, 35 th, 42 th time, and were significantly reduced at d14 after being given Guizhi Fuzitang. Guizhi Fuzitang can obviously alleviate the synovial hyperplasia and the infiltration of inflammatory cell, and reduce the inflammatory cells. Compared with the normal group (p-JNK, p-ERK, p-p38), the protein expression level (p-JNK, p-ERK, p-p38) of the CIA group significantly increased, with significant differences between the two groups (P<0.01). Compared with the model group, the protein expression level (p-JNK, p-ERK, p-p38) of the methotrexate group significantly decreased, with significant differences between the two groups (P<0.01). Compared with the model group, CIA rats showed significantly lower protein expression levels of p-JNK, p-ERK, p-p38 in a dose-dependent manner.Conclusion: Guizhi Fuzitang could inhibit the activation of MAPK signal transduction pathway to alleviate the symptoms of inflammation in CIA rats.

      摘要:Objective: To investigate the effect of pre-treatment with Buyang Huanwu Tang (BHT) on Src, Akt and p38 MAPK proteins and their phosphorylation expressions in vivo platelet activation signaling pathways of rats with cerebral ischemia-reperfusion (I/R) injury.Method: SD rats were randomly divided into blank control group, sham-operation group, model group, Clopidogrel Hydrogen Sulfate group (7.81 mg·kg-1) and high-dose, medium-dose and low-dose BHT group (29.69, 14.84, 7.42 g·kg-1). After oral administration with drugs for 14 d, the middle cerebral artery occlusion (MCAO) method was used to replicate the model of cerebral I/R injury, neurological deficit scores were measured. Besides, 2, 3, 5-Triphenyl tetrazolium chloride (TTC) staining method was used to calculate the volume of cerebral infarction; Western blot was used to measure platelet activation signaling pathways Src, Akt and p38 MAPK protein and their phosphorylation expressions. All the data were gathered after 14 days of continuous ig administration.Result: Compared with the model group, BHT at different doses and Clopidogrel Hydrogen reduced neurological deficit score and infarct volume percentage significantly in rats with cerebral I/R injury (P<0.05). BHT at different doses could significantly decrease Src, Akt and p38 MAPK phosphorylation levels (P<0.05). Clopidogrel Hydrogen dramatically reduced Src and Akt phosphorylation levels (P<0.05). Each group showed no statistical difference in total protein expression of Src, Akt and p38 MAPK.Conclusion: Pre-treatment with BHT had a cerebral protective effect on rats with cerebral I/R injury, which is partially attributed to its anti-platelet activation effects, and may be related with the inhibition of Src family kinases, PI3K-Akt and p38 MAPK signaling pathway.

      摘要:Objective: To investigate the effect of triterpene from Chaenomelis Fructus on the expression levels of Akt, NF-κB and inflammatory factors in synovial tissues of joint in adjuvant arthritis (AA) rats, and explore its possible mechanism.Method: Totally 80 male SD rats were randomly divided into normal group, model group, triterpene from Chaenomelis Fructus high and middle dose groups (50, 100 mg·kg-1). The models were established by injection of adjuvant on right hind paws of the rats. Triterpene from Chaenomelis Fructus (50, 100 mg·kg-1) was given by ig administration, and foot volume method was used to determine paw swelling and joint scores. The pathological changes of synovium were observed under optical microscope; and the levels of tumor necrosis factor-α (TNF-α), interleukin 1β (IL-1β), interleukin 6 (IL-6) and interleukin 8(IL-8) in joint synovial tissues were measured by using ELISA. The protein expression levels of protein kinase B (Akt), phospho-Akt (p-Akt) and nuclear transcription factor-κB p65 (NF-κB p65) in joint synovial tissues were measured by western blotting.Result: After 14days of joint inflammation, paw swelling reached the peak as compared with the normal group. The joint synovial hyperplasia and cell infiltration were seen obviously in model rats; the levels of TNF-α, IL-1β, IL-6 and IL-8 in joint synovial tissues were significantly increased; the NF-κBp65 and p-Akt protein expression levels were increased (P<0.01). As compared with the model group, triterpene from Chaenomelis Fructus (50 and 100 mg·kg-1) significantly decreased arthritis score, alleviated paw swelling, decreased the levels of TNF-α, IL-1β, IL-6 and IL-8 in joint synovial tissues; and significantly down-regulated the protein expression levels of p-Akt and NF-κB p65 (P<0.05, P<0.01).Conclusion: Triterpene from Chaenomelis Fructus has a significant anti-inflammatory effect. The mechanism maybe related to inhibiting Akt/NF-κB signaling pathway and activating the pro-inflammatory cytokines production.

      摘要:Objective: To study the effect of compound Shihu mixture(CSM) single-dose administration on insulin signaling in db/db mice.Method: According of the fasting blood glucose and body weight, 30 male db/db mice of 14-16 weeks were randomly divided into model group, Chinese medicine group 1 and Chinese medicine group 2; another 10 db/m mice were selected as normal group. After single-dose administration of CSM1 and CSM2, glucose tolerance(OGTT) levels were determined from the overall aspect. The changes in levels of 6-phosphofructokinase-1 (PFK), pyruvate dehydrogenase (PDH), and expression levels of insulin receptor (InsR) and Akt phosphorylation were determined at 30 min and 60 min of administration.Result: As compared with the normal group, the blood glucose OGTT levels were significantly increased in model group at 0, 30, 60, 120 min; activities of PFK and PDH in hepatic cells were significantly decreased; the protein expression levels of InsR and Akt were significantly reduced at 30 min and 60 min (P<0.01). As compared with the model group, the single-dose administration of both CSM1 and CSM2 could significantly reduce blood glucose OGTT levels at 60, 120 min (P<0.05, P<0.01); increase the activities of PFK and PDH in hepatic cells, and promote the phosphorylation levels of InsR Tyr1150/1151, Akt Ser473 and Thr308 in a time-dependent manner(P<0.05, P<0.01).Conclusion: Single-dose administration of both CSM1 and CSM2 could improve the glucose tolerance in db/db mice and increase the activities of PFK and PDH in hepatic cells. The mechanism may be associated with improving insulin signal transduction and promoting the phosphorylation of InsR Tyr150/1151, Akt Ser473 and Thr308.

      摘要:Objective: To discuss the effect of Sijunzi Tang on the expression of myocardial proteins in rats with chronic heart failure.Method: Establishing Among 42 SD male rats, 6 rats were randomly selected as normal group, and the rest 36 rats were used to establish chronic heart failure rat models. After successful modeling, they were divided into Sijunzi Tang high-dose group and low-dose group (11.88, 2.97 g·kg-1·d-1) and Captopril group (100 mg·kg-1·d-1). Rats in blank group and model group received administration of the same volume of normal saline, continuous for 1 month. Left ventricular tissues were taken respectively from various groups, and two dimensional gel electrophoresis was used to isolate proteins of cardiac muscle, analyze the differential protein spots and conduct in-gel digestion and mass spectrum identification for the differential protein spots.Result: Compared with the model group, 31 proteins had up-regulated expression and 28 proteins had down-regulated expression in Sijunzi Tang group. 8 proteinswhich displayed significant difference were respectably selected in each group (Volume≥1.5) for mass spectrum identification and 4 proteins were identified as follows after integration and duplicate removal:Suclg2 protein and pyruvate dehydrogenase complex related to energy metabolism; myosin light chain 2 related to myocardial contraction function; and serine protease inhibitor A3N related to myocardial cell reconstruction.Conclusion: Sijunzi Tang could regulate the expression of a variety of myocardial proteins in chronic heart failure rats, probably by regulating the energy metabolism, reducing cardiac remodeling and improving systolic function and heart function.

      摘要:Objective: To investigate the inhibitory effect of atractylenolide Ⅱ on Lovo cells and its mechanism.Method: Lovo cells were cultured in vitro, and treated with respectively atractylenolide Ⅱ at different concentrations to incubate for 24 h. The proliferation ability of Lovo cells was detected by 3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-H-tetrazolium bromide (MTT) assay. The apoptosis of Lovo cells impacted by atractylenolide Ⅱ were measured by flow cytometry (FCM), and the effect of atractylenolide Ⅱ on the expression of poly ADP-ribose polymerase1 (PARP1) and cysteinyl aspartate specific proteinase-3 (Caspase-3) were detected by the Western blot.Result: Compared with the blank control group, at the dose of 150 mg·L-1, atractylenolide Ⅱ significantly inhibited the proliferation of Lovo cells (P<0.05). With the increase of the concentration of atractylenolide Ⅱ, cell survival rate was gradually decreasing. At the concentration of more than 300 mg·L-1, cell survival rate dropped to less than half. High-dose (300 mg·L-1) atractylenolide Ⅱ can significantly promote the apoptosis of Lovo cells (P<0.01). At the dose of 100 mg·L-1, atractylenolide Ⅱ can cause shear in PARP1 and Caspase-3. With the increase of drug concentration, the shearing effect was significantly enhanced. Atractylenolide Ⅱ can inhibit the growth and proliferation of Lovo cells, regulate the expression of PARP1 and Caspase-3, and then promote the apoptosis of cells.Conclusion: Atractylenolide Ⅱ can significantly inhibit the proliferation of Lovo cells and induce their apoptosis, its molecular mechanism may be related to the regulation of the expression of PARP1 and Caspase-3.

      摘要:Objective: To investigate the effect of Tetrastigma Hemsleyani Radix flavone (THRF) on proliferation, migration, invasion and Notch1 in esophageal cancer (EC) 9706 cells, and discuss its possible anti-esophageal cancer mechanism.Method: The EC9706 cells were cultured in vitro and treated with THRF with different concentrations(0.5~20 g·L-1), with no THRF in blank group, and with MW 167 in positive control group. The 50% inhibitory concentration (IC50) was measured by cell counting kit-8 (CCK-8). The effect of THRF on adhesion rate was detected by the cell adhesion assay. The cell migration ability were evaluated by the wound healing assay. The cell invasion ability were evaluated by the transwell chamber assay. The expression of Notch1 and Notch1 mRNA expressions were detected by immunocytochemical analysis and Real-time PCR assay.Result: THRF can significantly increase the inhibition rate of EC9706 cells, with statistically significant difference compared with blank group (P<0.05, P<0.01). THRF can significantly inhibit the adhesion rate, the migration ability, the invasion cells, Notch1 positive cell rate and Notch1 mRNA expressions, with statistically significant difference compared with blank group (P<0.05, P<0.01).Conclusion: THRF can inhibit the EC9706 cells viability, migration and invasion ability, its mechanism for anti-esophageal cancer may be correlated with down-regulation of Notch1.

      摘要:Objective: To observe the effect of modified Chenping Tang for the treatment of senile community acquired pneumonia with the syndrome of phlegm dampness obstructing lung and blood stasis.Method: A total of 104 patients were randomly divided into treatment group and control group by random number table, with 52 cases in each group. Patients in treatment group received routine western medicine combined with modified Chenping decoction, while patients in control group only received pure western medicine. Clinical symptom and sign scores, curative effect of traditional Chinese medicine (TCM) syndrome, peripheral blood inflammatory markers (white blood cell count, neutrophil count, rapid C reactive protein, erythrocyte sedimentation rate and procalcitonin), inflammatory absorption of chest CT and adverse drug reactions were evaluated and compared.Result: Compared with control group, treatment group showed better clinical symptom and sign scores, curative effect of TCM syndrome, peripheral blood inflammatory markers, and inflammatory absorption of the chest CT among patients with senile community acquired pneumonia with the syndrome of phlegm dampness obstructing lung and blood stasis (P<0.01). Mild adverse reactions, such as phlebitis, gastrointestinal reaction and liver damage occurred in a few patients in the process of trial, but the adverse reaction rates between two groups were not statistically significant.Conclusion: Modified Chenping Tang combined with routine western medicine shows a better efficacy in treating patients with senile community acquired pneumonia with the syndrome of phlegm dampness obstructing lung and blood stasis, and so is worth clinical promotion and application.

      摘要:Objective: To investigate the effect of Wutou Chishizhi Tang on tumor necrosis factor-α (TNF-α), B-type natriuretic peptide (BNP) and homocysteine (Hcy) levels in serum of patients with Senile Ischemic Cardiomyopathy and heart failure.Method: The 94 patients with senile ischemic cardiomyopathy and heart failure were selected randomly divided into 2 groups. 47 cases in control group were treated with basic treatment, and 47 cases in treatment group were treated with Wutou Chishizhi Tang based on the treatment in control group. Ventricular remodeling and cardiac function, serum, quality of life, clinical efficacy and adverse reactions were compared after the treatment.Result: The effective rate in control group (74.47%) was lower than that in treatment group (91.49%), with significant difference (P<0.05). As compared with those before treatment, the left ventricular remodeling and cardiac function, serology, quality of life were changed in both groups, the left ventricular end-diastolic diameter (LVEDD), left ventricular end systolic diameter (LVESD) in treatment group were lower than those in control group, while the left ventricular ejection fraction (LVEF) and output per minute (CO) indexes were higher than those in control group, and the levels of serum BNP, Troponin I (cTnI), high-sensitivity C-reactive protein (hs-CRP), TNF-α and Hcy levels were lower than those in control group after treatment (P<0.05), in addition, the improvement on life quality in treatment group was superior to that in control group after treatment (P<0.05), with no serious adverse reactions in both groups during treatment.Conclusion: Wutou Chishizhi Tang has clear efficacy in the treatment of Senile Ischemic Cardiomyopathy and heart failure, as it can reduce the levels of serum TNF-α, BNP and Hcy levels, and improve the quality of life.

      摘要:Objective: To investigate the efficacy and compatibility mechanism of Lonicerae Japonicae Flos-Weeping Forsythia herbal pairs.Method: All of chemical components related to the two traditional Chinese medicines were researched from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP), in order to explore potential targets related to candidate compounds. The network pharmacological method was adopted to analyze its efficacy and compatibility mechanism.Result: According to the oral bioavailability (OB) and drug likeness (DL) conditions, 16 candidate active molecules, 64 corresponding target and 166 kinds of related diseases were screened out. The value of the first three candidate compounds were bicuculline (37), turpentine monomethyl ether (30), and 5-hydroxy-7-methoxy-2-3, 4, 5-trimethoxyphenyl chromone (30). The five target proteins with the degree value equal to 13 were SLC6A3, DPP4, AR, NOS2, PTGS2.The related diseases with high degree value were non-specific cancer, Alzheimer's disease, breast cancer, pain, cardiovascular disease, inflammationin the order from high to low.Conclusion: The result of the study preliminarily verifies the basic pharmacological effects of Lonicerae Japonicae Flos-Weeping Forsythia herbal pairand its related mechanisms, and lays a solid foundation for further studies on the mechanism of action.

      摘要:Objective: To investigate the efficacy material basis and the compatibility mechanism of Platycodonis Radix-Glycyrrhizae Radix et Rhizoma.Method: All the chemical components related to the two traditional Chinese herbs from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) were searched. The oral bioavailability (OB)≥50% and drug likeness (DL)≥0.18 were used as the screening conditions for molecular compounds. The potential targets related to candidate compounds were found through the TCMSP, and then the drug-target interaction network as well as target-disease interaction network were constructed.Result: The 43 candidateactive molecules, 98 corresponding targets and 224 related diseases were obtained through screening according to OB and DL conditions. The top three compounds were arenaringenin (48), formononetin (41) and shinpterocarpin (36). The top three target proteins were PTGS2 (32), PPAR-γ (21) and ERS1 (18). Related diseases of higher degree values were non-specific cancer, inflammation, pain, breast cancer and Alzheimer's disease and so on.Conclusion: The results of the study preliminarily verified the basic pharmacological effects and related mechanisms of Platycodonis Radix-Glycyrrhizae Radix et Rhizoma, and laid a good foundation for further study on the mechanism of action.

      摘要:Objective: To predict the active compounds, therapy targets of Liuwei Dihuangtang (LWDHT) as well as possible diseases, and explore the molecular mechanism of its compatibility and its efficacy on diseases under the guidance of ‘treating different diseases with the same method’.Method: Oral bioavailability (OB) and drug-likeness (DL) based on TCMSP database were used to screen the active compounds of LWDHT and its related diseases. Therapy targets were obtained from the Drugbank database, and then the Chinese herbal medicine (CHM) compound-targets-disease network was constructed by using cytoscape software. The network topology analysis, target function and signaling pathway analysis were also performed.Result: The 42 of 507 compounds in LWDHT were screened out (OB≥30%, drug-likeness index≥0.18).The 221 related targets were found and 9 categories of diseases involving 56 diseases were enriched, among which tumor, cardiovascular and respiratory diseases were most closely correlated with the formula. The compatibility of LWDHT, ‘Sanbu’ with reinforcing effect and ‘Sanbu’ with reducing effect can regulate cellular apoptosis, metabolism, proliferation, and vascular smooth muscle contraction. Besides, LWDHT also can regulate protein synthesis modification and secretion. The ‘Sanbu’ with reinforcing effect can regulate transmembrane transport activity of ion, blood pressure, the metabolic process of prostaglandin and retinoic acid. The ‘Sanbu’ with reducing effect can regulate cell signaling and biosynthetic process.Conclusion: In this study, 42 compounds were screened out as the potential effective compounds of LWDHT, and 221 targets and 56 diseases were predicted to be associated with the formula. The compatibility of ‘Sanbu’ and ‘Sanbu’ made 1+1>2 synergistic effect mechanism. LWDHT owned multiple targets and multiple pathways to treat various diseases under the guidance of ‘treating different diseases with the same method’.

      摘要:Objective: Present study aims to screen the active ingredients of traditional Chinese medicine for treatment of acute myelocytic leukemia.Method: Herbs and compounds for acute myelocytic leukemia (AML) were collected, and the systematic pharmacology approach was used to predict the nature of medicines, potential targets and potential thiol consumption characteristics. shikonin with higher oral Bioavailability (OB), drug-likeness (DL) and thiol consumption activities were selected as the verification molecule. The potential targets of screened compounds were predicted to build potential compound-target and potential target-disease networks. Key targets and potential active compounds were screened according to topological characteristics. Glutathione (GSH) content was detected by GSH detection kit and OPA fluorescence method after shikonin treatment. Digital gene expression profile sequencing analysis was used to verify the accuracy of the predicted targets after 100 μg·L-1 shikonin treatment on HL-60 cells for 48 h.Result: After in silico screening, we identified 72 active compounds with appropriate OB, DL and thiol consumption characteristics among 4 024 compounds in 40 herbs for acute myelocytic leukemia. Then 10 potential active compounds were obtained which could act on AML related targets. The results verified that shikonin depleted GSH after shikonin (100 μg·L-1) treatment on HL-60 cells and chemical system. The predicted targets could regulate the expression of varied genes in gene chip (100 μg·L-1, 48 h).Conclusion: According to the study, systematic pharmacology method and experimental verification were helpful to excavate the potential active ingredients for treatment of acute myelocytic leukemia.

      摘要:Pulsatillae Radix, also known as Pulsatilla Chinesis Regel, has a long history of medication, and was found in Sheng Nong's Herbal Classic. In Eastern Han Dynasty, ‘Medical Sage’ Zhang Zhong-jing wrote in Treatise on Febrile and Miscellaneous Diseases, ‘Pulsatillae Radix Tang is an antipyretic and purgative medicine’. Ancient herbal literatures and records are different in descriptions of ‘white velvet nearby root’ and ‘white velvet nearby head’. Through the study of herbal medicine of Pulsatillae Radix, we reviewed and analyzed previous herbal literatures, studied its basic, origin and efficacy, and summarized and compared Chinese Pulsatilla sect samples. According to taxonomic studies for Chinese Pulsatilla sect, we made a textual research for the basis of Chinese herb Pulsatilla sect and provide the basis for further development and utilization of Pulsatillae Radix. Through the analysis of ancient herbal researches and literatures, we only identified it as a Ranunculaceae Pulsatilla sect plant, but could not identify it as P. chinensis. Because the medication of Pulsatillae Radix greatly varies in different places in China, we could not identify it based on characteristics. Worse still, there are many confused and counterfeit products in the market, which seriously affects people medication safety. With the characteristics of accuracy, simplicity and rapidity, modern molecular biological DNA bar code technology provides a new method for the identification of plant species and the discovery of new species and reliable basis for identifying the authenticity of botanicals. DNA bar code technology can help identify the wide variety of Pulsatilla sect plants. A large number of experiments show that Pulsatilla sect plants (such as P. cernua, P. dahurica) have similar chemical components and index components with P. Chinensis, and their medicinal value shall be determined through preparation and pharmacology-based analytical techniques.

      摘要:Intestinal flora homeostasis in maintaining normal physiological function, regulation of body's immunity and antagonistic potential against pathogenic microorganisms has important physiological significance.Intestinal flora and the traditional Chinese medicine (TCM) theory of spleen is closely related in the physiological function, intestinal dysbacteriosis and TCM spleen deficiency causal each other, use the spleen prescription treated from the spleen to restore the balance of intestinal flora steady has important theoretical and practical significance.This article summarized the research status of two aspects from the connection between the spleen deficiency and the intestinal dysbacteriosis, spleen and prescription of TCM regulating intestinal flora, in order to further research from the angle of microecology of the mechanisms of spleen and prescription of TCM.

      摘要:On the basis of a comprehensive consulting, sorting and analyzing of the ancient and modern medical literature, this paper summarizes the general situation of the historical development and modern research of Arcae Concha.Taking the book A Summary of Chinese Medicine Processing Data Through History as clue, the author refers to 30 classical medical works written from the Three Kingdoms period to the Qing dynasty, 9 versions of Chinese Pharmacopoeia, the 1988 edition of The National Chinese Medicine Processing Criterion, and 22 books of local Chinese medicine processing criterion.After that, the author summerizes all the processing methods of Arcae Concha in ancient and modern literature.Arcae Concha processing has a long history with processing methods including pulverization, the fire burnt, calcined quenching, calcined vinegar, vinegar boiling, calcined brine, etc.At present, China mainly use the method of calcining.Summerizing the predecessors' experience will provide beneficial reference to further study of Arcae Concha.

      摘要:Objective: Recently, Chinese medicine has been found playing an effective and positive role in prevention and control of airborne diseases, especially acute infectious diseases, such as severe acute respiratory syndrome (SARS) and H1N1 subtype avian influenza, etc.The efficacies, clinical and pharmacological effects of some traditional Chinese medicine (TCM) for the treatment of such diseases have been explored and many studies have been carried out all over the world.Most of the present results showed that some herbs and complex prescriptions originated from TCM might inhibit virus proliferation, suppress bacteria, restrain inflammatory reaction and improve immunity in the prevention and control of airborne disease.This analysis covered the previous work in this field in order to provide some hints for the future studies and applications, which indicated that many single Chinese herb and formula exert exact therapeutic effects on SARS and H1N1, while some of them increase the effects or reduce side-effects when combined with chemical drugs.Furthermore, the basic idea of TCM on treating and preventing diseases, as well as the regulations on applications and composition of TCM, should be closely connected with relative studies, since patient is the focus of all efforts during practice.

      摘要:Tripterygii Radix et Rhizoma is a kind of commonly used traditional Chinese medicine (TCM) in treating rheumatoid arthritis, with multiple pharmacological effects, including heat-clearing and detoxifying, eliminating wind and dampness, detumescence and analgesia, parasite destroy and itching relieving. According the development of modern pharmacological research, it shows anti-inflammatory, analgesic, anti-tumor and immune regulatory effects, and has been clinically applied to treat immune diseases, renal disease and dermatosis disease, particularly for rheumatoid arthritis. However, due to its multi-organ toxicity, especially hepatotoxicity and nephrotoxicity, it has been restricted from being widely used in clinic, which has become a hot research topic in recent years. Our research group focused on the attenuation and synergy effect of Tripterygii Radix et Rhizoma through transdermal drug delivery, in order to promote application of Tripterygii Radix et Rhizoma. This paper collects and analyzes relevant literatures from 2006 to 2016, including traditional processing for attenuation, combined administration for attenuation, modern preparations for attenuation, structural modification for attenuation, biological technology for attenuation and their combined applications. By analysis of the current conditions and difficulties in developing the attenuation and synergy of Tripterygii Radix et Rhizoma and comparing the differences with the existing attenuation modes, this study provides ideas for further studies on attenuation and synergy of Tripterygii Radix et Rhizoma, promote the rational development and safe clinical application, and offers a reference for the rational application of other toxic TCMs.
