      摘要:Particulate matter (PM) has become one of the most serious causes of premature death in the world. A variety of diseases of respiratory system, cardiovascular system is directly related to PM. In recent years, China's environmental pollution has become more and more serious. PM2.5 was one of the main pollution sources;therefore, it is significant to study effective interventions on potential health problem caused by PM2.5. In this paper, the distribution of research teams, research units, research fields and research hotspots concerning domestic atmospheric particulates from 2003 to 2013 were studied by using the literature metrology, association rules mining and clustering analysis methods. This could provide reference information and trends about PM2.5 to researchers especially medical researchers.

      摘要:Objective: To screen technological conditions of lactose micropowder grinding modification to decrease hygroscopicity of Angelicae Dahuricae Radix(ADR) extract and analyze anti-absorption mechanism of lactose micropowder. Method: With moisture absorption rate as index, single factor test was adopted to investigate effects of kinds of lactose micropowder, ratio of raw materials to accessories and grinding time on technological conditions of lactose micropowder grinding modification, modifiers were supported by physical indexes, such as particle size distribution, microstructure and so on. Result: Optimal technological conditions of lactose micropowder grinding modification to decrease hygroscopicity of ADR extract were as follows:ARD extract-lactose micropowder(crushed 1 min) of 1:1, grinding time of three minutes.Particle sizes d0.1, d0.5 and d0.9 of modifier were 3.477, 15.078, 54.501 μm, respectively.The contact angle was (7.7±0.66) °, surface of ARD extract was coated by lactose micropowder particles. Conclusion: Available micronizing lactose can significantly decrease absolute values of initial velocity and acceleration of moisture absorption of ADR extract for reducing its hygroscopicity.Possibility of anti-absorption effects is that lactose particles are attached to surface of ADR extract after grinding modification.

      摘要:Objective: To develop an HPLC fingerprint analysis method of Phellodendri Chinensis Cortex and compare fingerprint differences of its processed products and the raw from different habitats. Method: HPLC was employed to compare fingerprint of ten batches of Phellodendri Chinensis Cortex and its processed products with mobile phase consisting acetonitril(A)-water(containing 0.3% phosphoric acid and 0.3% diethylamine, B) for gradient elution(0-10 min, 10%-14%A;10-20 min, 14%-25%A;20-45 min, 25%A;45-50 min, 25%-65%A;50-55 min, 65%-70%A;55-60 min, 70%-75%A;60-70 min, 75%-100%A), detection wavelength was set at 284 nm, column temperature was 40 ℃, flow rate was 0.8 mL·min-1 and analytic time was 70 min. Result: Its processed products had a similar fingerprint chromatogram to the raw of Phellodendri Chinensis Cortex, but existing a little difference in relative content of each component.Some of peaks in HPLC chromatogram were identified as chlorogenic acid, phellodendrine, magnoflorine, jatrorrhizine, berberrabine, palmatine chloride, berberine hydrochloride, obaculactone.Similarities of total mode peaks among ten batches of raw materials, wine and salt baked products were more than 0.9. Conclusion: This established fingerprint has good precision, reproducibility and stability, it can be used as a quality control method of Phellodendri Chinensis Cortex and its processed products.

      摘要:Objective: To research the content of heavy metals in nine kinds of traditional Chinese medicines from four professional markets, which provided a reference for establishment of special detection method for heavy metals in medicinal herbs. Method: Contents of heavy metals(Cu, Cd, Hg, Pb) in 36 batches of 9 herbs were determined by atomic absorption spectrophotometer and atomic fluorescence spectrometer in orde to compare content differences of heavy metals from different markets. Result: The content of Cu was in order of Lonicerae Japonicae Flos > Artemisiae Annuae Herba, Akebiae Caulis, Salviae Miltiorrhizae Radix et Rhizoma, Sinapis Semen, Gardeniae Fructus > Magnoliae Officinalis Cortex > Gastrodiae Rhizoma > Poria.Cd was in the order of Lonicerae Japonicae Flos, Artemisiae Annuae Herba, Sinapis Semen, Gardeniae Fructus > Akebiae Caulis, Magnoliae Officinalis Cortex > Gastrodiae Rhizoma > Salviae Miltiorrhizae Radix et Rhizoma, Poria.Hg was in the order of Lonicerae Japonicae Flos > Gardeniae Fructus > Artemisiae Annuae Herba > Magnoliae Officinalis Cortex > Sinapis Semen, Gastrodiae Rhizoma, Poria, Salviae Miltiorrhizae Radix et Rhizoma, Akebiae Caulis.Pb was in order of Magnoliae Officinalis Cortex > Sinapis Semen, Gardeniae Fructus, Lonicerae Japonicae Flos, Artemisiae Annuae Herba, Akebiae Caulis > Gastrodiae Rhizoma, Salviae Miltiorrhizae Radix et Rhizoma > Poria.In 36 batches of medicinal herbs, the content of Cu was qualified.Hg exceeded in one batche of herb, the average content of Hg was the highest in Lonicerae Japonicae Flos, over standard rate was 25%.Pb exceeded in three batches of herbs, over standard rates of Pb in the Magnoliae Officinalis Cortex and Sinapis Semen were 50% and 25%, respectively.Cd exceeded in seven batches of herbs, over standard rates of Cd in Lonicerae Japonicae Flos and Artemisiae Annuae Herba were all 75%, in Gardeniae Fructus it was 25%.But difference of heavy metals content in medicinal herbs bought from different market was not significant. Conclusion: In this investigation of four kinds of heavy metal elements, Cd pollution is most serious.Lonicerae Japonicae Flos is the most polluted medicinal herbs in 9 samples, which will be the primary object in the future research.

      摘要:Objective: To optimize kinds and amounts of penetrating enhancers in Wenjing Zhitong gel paste. Method: Improved Franz diffusion cell was employed with isolated mice abdominal skin as barrier.Kinds and amounts of penetrating enhancers were optimized through kinetic parameters of percutaneous absorption, such as cumulative permeation quanity, penetration rate and enhancement ratio in way of HPLC, mobile phase was methanol-water(80: 20), detection wavelength of ligustilide was set at 328 nm. Result: Combination of azone and propylene glycol was better than other kinds of enhancers for penetration effect, dosage of azone and propylene glycol was finally opitimized to be 4% of prescription dosage, ratio between azone and propylene glycol was 2: 1.Under these conditions, cumulative permeation quanity of ligustilide was 84.16 μg·cm-2, penetration rate of ligustilide was 3.28 μg·cm-2·h-1 and enhancement ratio was 3.33. Conclusion: Combination of azone and propylene glycol has significant penetration effect, which can be as an enhancer of Wenjing Zhitong gel paste.

      摘要:Objective: To optimize purification technology conditions of total alkaloids from Sophorae Tonkinensis Radix et Rhizoma by D101 macroporous resin. Method: Taking matrine as index component, UV was employed to determine the content of total alkaloids with detection wavelength of 415 nm.With the content of total alkaloids as index, effects of the concentration of sample solution, amount and type of eluent, elution velocity on purification process were investigated by single factor tests. Result: Optimum technology conditions were as follows:adjusted pH of sample solution to 3-4 with with hydrochloric acid, adjusted pH to 10 with 20% ammonia water, the concentration of sample solution 0.8 g·mL-1, sample flow rate of 2 BV·h-1, sample volume(equivalent as crude drug)-resin volume ratio of 1: 2.5, washed impurity with 2 BV of water, with 4 BV of 70% ethanol as eluant, elution flow rate of 4 BV·h-1.Purity of total alkaloids was 66.83%. Conclusion: D101 macroporous resin has good purification effect for total alkaloids from Sophorae Tonkinensis Radix et Rhizoma, optimized process is simple and feasible, this study provides a reference for resource utilization of Sophorae Tonkinensis Radix et Rhizoma.

      摘要:Objective: To optimize flash extraction process of total polysaccharides from Solanum nigrum. Method: Phenol-sulfuric acid method was used to determine the content of total polysaccharides with detection wavelength at 490 nm.With extracting amount of total polysaccharides and yield of extract as comprehensive index, effects of extraction voltage, extraction time, extraction times and solid-liquid ratio on flash extraction process were investigated by orthogonal test.Optimum flash extraction process was compared with hot reflux method and ultrasonic method. Result: Optimum flash extraction process was adding 30 times the amount of water at 80 V and extracting 2 times for 2 min of each time.Extracting amounts of total polysaccharides were 3.760, 3.232, 4.585 mg·g-1 by hot reflux extraction, ultrasonic and flash extraction method, respectively. Conclusion: Extraction efficiency of flash extraction method is significantly higher than the other two kinds of methods, which is suitable for extracting total polysaccharide from S. nigrum, it is feasible and rapid with good application prospects.

      摘要:Objective: To optimize purification process of total flavonoids from Pyrrosiae Folium by macroporous resin. Method: The content of total flavonoids was determined by UV-visible spectrophotometry, detection wavelength was 510 nm.With the content of total flavonoids as index, types of macroporous resin were screened by static adsorption-elution test, single factor tests were adopted to optimize purification technology with sample volume, ratio of diameter to height, the amount of eluent and so on. Result: D101 macroporous resin showed better adsorption-elution effect than others, optimum purification technology was as following:height-diameter ratio of 6-10, ratio of resin to crude drug 1: 2.73, the concentration of sample solution 0.5 g·mL-1, absorption velocity 2 BV·h-1, washed impurity with 8 BV of water, then eluted with 5 BV of 60% ethanol at 4 BV·h-1, collected eluent.Under these conditions, purity of total flavonoids was 59.06%. Conclusion: D101 macroporous resin has good purification effects on total flavonoids from Pyrrosiae Folium, this optimized purification technology is reasonable, stable and feasible.

      摘要:Objective: To study the chemical constituents of Callicarpa kwangtungensis. Method: The chemical constituents were isolated by column chromatography on silica gel, Sephadex LH-20 and MPLC. Their chemical structures were elucidated on the basis of spectral analysis. Result: Eight compounds were isolated from n-butanol portion, their structures were elucidated as 6'-β-D-apiofuranosyl cistanoside C (1), acetyl forsythoside B (2), 2'-acetylacteoside (3), acteoside (4), isoacteoside (5), forsythoside B (6), poliumoside (7), arjunglucoside Ⅱ (8). Conclusion: Compounds 1-3, 5, and 8 were isolated from this plant for the first time.

      摘要:Objective: To investigate the chemical constituents of n-butanol extract of Ginseng medicinal fungal substance, which was to clarify the basis of its efficacy substance. Method: Thin-layer chromatography, column chromatography on silica gel, Sephadex LH-20 gel chromatography, semi-preparative liquid chromatography were used for the isolation and purification of the compounds and their structures were determined by comparison of their chromatographic and spectral data with those of the authentic samples and those reported in the literature. Result: Five compounds from n-butanol extract of ginseng medicinal fungal substance were identified as 20(S)-ginsenosides-Rg2 (1), 20(R)-Ginsenosides-Rg2 (2), Ginsenosides-Rg1 (3), 20(S)-Ginsenosides-Rf (4), ginsenosides-Rb1 (5). Conclusion: All compounds are isolated from ginseng medicinal fungal substance for the first time.

      摘要:Objective: The current study aimed to identify the chemical constituents occurring in the leaves of Alpinia officinarum. Method: The collected leaves of A. officinarum were dried in an electric thermostatic drying oven at 40°C overnight. The freshly dried leaves were smashed using a universal high-speed smashing machine and then sieved manually by an 80 mesh. The resulting fine powders and residue were mixed evenly. An aliquot (0.6 g) was weighed precisely and macerated with 30 mL of methanol and then ultrasonicated three times for 30 min each. For each ultrasonication extraction, the resulting extract solutions were centrifuged at 8 000 rpm for 10 min. One micro liter of supernatant was sampled and the remaining was discarded. The residue was extracted with methanol for 2 more times. The sampled methanol extracts (3 mL) were combined and centrifuged at 13 000 rpm for 10 min to obtain the supernatant fractions that were frozen at -40 ℃ until analysis. The extract solution was appropriately diluted with methanol before analysis. Finally, a 10 μL aliquot was injected into the LC-MS/MS system, which was operated in the positive ion ESI mode with selected reaction monitoring for all the potential analytes. Result: Sixteen chemicals, including flavonoids and diarylheptanoids, were identified using LC-MS/MS with selected reaction monitoring mode(MRM). Twelve flavonoids were chrysin, pinocembrin, tectochrysin, apigenin, galangin, 3-methylgalangin, acacetin, kaempferol, kaempferide, quercetin, isorhamnetin and rutin, respectively. Four diarylheptanoids included yakuchinone A, oxyphyllacinol, hexahydrocurcumin and hannokinol. Conclusion: Sixteen chemicals were identified using LC-MS/MS and some unknown diarylheptanoids need to be identified in the future.

      摘要:Objective: To establish an HPLC method for determination of paeoniflorin and tanshinone ⅡA in Shujing Fangtong capsules. Method: Chromatographic test on paeoniflorin was performed on Luna 5u C18 column(4.6 mm×250 mm, 5 μm), mobile phase consisted of acetonitrile-0.05% monopotassium phosphate(15: 85), detection wavelength was 230 nm, column temperature was 25 ℃, injection volume was 10 μL, flow rate was 1.0 mL·min-1.Similarly, chromatographic test on tanshinone ⅡA was performed on Diamonsil C18 column (4.6 mm×250 mm, 5 μm), mobile phase was methanol-water(80: 20), detection wavelength was 270 nm, column temperature was 25 ℃, injection volume was 20 μL and flow rate was 1.0 mL·min-1. Result: Linear ranges of paeoniflorin and tanshinone ⅡA were 0.020 4-0.204, 0.043 2-0.864 μg, average recoveries of them were 99.89%(RSD 1.5%) and 100.43%(RSD 1.1%), respectively.RSDs of repeatability, stability and precidion test were less than 2%. Conclusion: This method is simple, accurate with good repeatability, which is suitable for quality control of Shujing Fangtong capsules.

      摘要:Objective: To study the chemical constituents of Macrosolen cochinchinensis. Method: The chemical constituents were isolated and purified from M.cochinchinensis by Sephadex LH-20, silica column chromatography and recrystallization. Their structures were elucidated on the basis of their physicochemical properties and spectra analysis. Result: Ten compounds were obtained and identified as lupeol (1), cycloeucalenol (2), friedelan-3β-ol (3), (23Z)-9, 19-cycloart-23-ene-3α, 25-diol (4), friedelan-3-one (5), dotriacontan-1-ol (6), β-sitosterol (7), lacceroic acid (8), rutin (9), orientin (10). Conclusion: Compounds 1-8 and 10 were isolated from this plant for the first time.

      摘要:Objective: To develop an HPLC method for simultaneous determination of five components (paeoniflorin, albiflorin, hyperin, rutin, isoquercitrin) in Yushu tablets. Method: An Agilent Zorbax SB-C18 (4.6 mm×250 mm, 5 μm) was adopted. The mobile phase was 0.1% phosphoric acid aqueous solution (A)-acetonitrile (B) with gradient elution. Analysis were performed at 25 ℃ with a flow rate of 0.8 mL·min-1, and the detection wavelength was 203 nm. Result: The method had a good linearity in the ranges of 0.360 5-0.036 0 μg (r=0.999 9) for albiflorin, 1.005 8-0.100 6 μg (r=0.999 8) for paeoniflorin, 0.150 7-0.015 1 μg (r=0.999 8) for rutin, 0.091 9-0.009 2 μg (r=0.999 8) for hyperin, 0.063 7-0.006 4 μg (r=1.000 0) for isoquercitrin. The average recoveries of five components were 100.51% (RSD 2.0%), 99.81% (RSD 2.0%), 99.09%(RSD 1.7%), 99.23%(RSD 1.8%), and 99.65%(RSD 2.0%), respectively. Conclusion: The method is dependable, simple and practical, and may be used to control the quality of Yushu tablets.

      摘要:Objective: To purify a glycopeptide named P-Ⅱ from Panax ginseng and investigated its properties. Method: The glycopeptide was extracted by 50% ethanol, precipitated with high-concentration ethanol, after decolorized by active carbon obtained crude ginseng glycopeptide, then purified by DEAE-Sepharose CL-6B column, washed successively with 0.2, 0.5 and 1.0 mol·L-1 NaCl aqueous solution. Then using Sephadex G-150 column eluted with an aqueous solution of 0.2 mol·L-1 NaCl to give P-Ⅱ. The contents of neutral carbohydrate, uronic acid and peptide were determined using glucose, galacturonic acid and bovine serum albumin as the standards. The characterization of P-Ⅱ was analyzed by cellulose acetate membrane chromatography, Sephadex G-100 column chromatography, gas chromatography, and amino acid analysis. Result: P-Ⅱ was a homogneous glycopeptide. The contents of neutral carbohydrate, uronic acid, and peptide in P-Ⅱ were 50.2%, 38.5%, and 1.9%, respectively. The molar ratio of Rha, Ara, Glc and Gal was 0.45: 1.15: 1.00: 2.36.It was consisted 16 kinds of amino acids including Asp, Fhr, and Ser. Conclusion: P-Ⅱ was a homogeneous glycopeptide, rather than a mixture of sugars and peptides, and laid the foundation for the study of structure-activity of ginseng glycopeptide.

      摘要:Objective: To establish an HPLC method for simultaneous determination of six indexes in Tongmai pill, provide reference for quality evaluation. Method: An Agilent ZORBAX SB-Aq chromatographic column (4.6 mm×250 mm, 5 μm) was used. The mobile phase was acetonitrile-methanol-0.1%H3PO4 with flow rate of 1 mL·min-1. Column temperature set at 35 ℃. The wavelengths were 260 nm for strychnine, brucine, paeoniflorin, hesperidin, and salvianolic acid B, and 316 nm for ferulic acid. Result: Strychnine, brucine, paeoniflorin, hesperidin, salvianolic acid B, and ferulic acid showed good linearity in the range of 0.037-1.097, 0.039-1.176, 0.075-2.25, 0.01-0.30, 0.113-3.384, 0.225-6.72 μg, respectively. The average recoveries were 99.69%, 100.12%, 99.56%, 99.98%, 99.52%, and 99.69%. Conclusion: The established method is simple, accurate, and reproducible. It can be used for quality control of the Tongmai pill.

      摘要:Objective: To establish a HPLC method for determination of senkyunolide and ligustilide in Chuanxiong Rhizoma and Angelicae Sinensis Radix by QAMS. Method: To measure the correction factor of ligustilide and senkyunolide under different conditions using the butylphalide as the internal reference standard, and the ligustilide and senkyunolide in Chuanxiong Rhizoma and Angelicae Sinensis Radix were determined by the internal reference standard and the relative correction factor. Result: The linear response ranges of butylphalide, senkyunolide and ligustilide were 0.199 4-3.988(r=0.999 9), 0.043 46-0.869 2 (r=0.999 9) and 0.080 67-1.613 4 μg (r=0.999 9) respectively. Relative correction factors of senkyunolide and ligustilide with reference to butylphalide were 0.490 7 and 0.226 3. Conclusion: The internal reference standard is adopted to determine senkyunolide and ligustilide in Chuanxiong Rhizoma and Angelicae Sinensis Radix. This accurate method is available and applied for quality of Chuanxiong Rhizoma and Angelicae Sinensis Radix.

      摘要:Objective: To establish the HPLC fingerprint of Archidendron clypearia leaves. Method: The chromatographic fingerprint was obtained with Odyssll C18 column (4.6 mm×250 mm, 5μm) and gradient eluted with methanol and 0.5% phosphoric acid. The column temperature was maintained at 35 ℃, and the flow rate was 1 mL·min-1. The detection wavelength was 273 nm.The injection volume was 10 μL. Result: Six chromatographic peaks among 15 common peaks were identified by UPLC/DAD/Q-TOF-MS xevo G2 Q-Tof LC-MS and be intended to be gallic acid, galuteolin, gallic acid ethyl ester, (-)-(2S)-5, 3', 4', 5'-tetrahydroxyflavan-7-gallate, 7, 4'-O-di-gallyoltricetiflavan, quercitrin. The similarities of ten batches of Archidendron clypearia leaves were above 0.97. Conclusion: The method accordance with the requirements of the methodology and can be used for the full quality evaluation of Archidendron clypearia leaves.

      摘要:Objective: To assay the volatile constituents of Callicarpa bodinieri flowers. Method: The volatiles were analyzed by head-space solid micro-extraction, coupled with GC/MS and Kovats. Result: There were 22 different kinds of volatile constituents in C. bodinieri flowers, 88.51% of the total essential constituents respectively. α-caryophyllene (30.30%), caryophyllene (10.04%), β-selinene (8.53%), 3-octanol(6.73%), β-pinene (6.35%) and β-thujene (3.99%) had higher contents. Conclusion: The major type volatile constituents of C. bodinieri flowers were terpenoids.

      摘要:Objective: Using HPLC-UV and HPLC-FLED methods to determine the content of five anthraquinones in Chinese rhubarb. Method: HPLC-UV and HPLC-FLED were used to determine the concentrations of aloe-emodin, rhein, emodin, chrysophanol, and physcion in the rhubarb. The separation was performed on a Venusil XBP-C18(4.6 mm×250 mm, 5 μm) column with methanol-0.1% phosphoric acid (80:20) as mobile phase. The flow rate was 1.0 mL·min-1. The detection wavelength for UV detector was 254 nm. The excitation wavelength and emission wavelength for fluorescence detector were 435 nm and 515 nm, respectively. Result: Aloe-emodin, rhein, emodin, chrysophanol, and physcion determined by HPLC-UV were in good linearity over the ranges of 14.6-146.4 ng (r=0.999 7), 39.0-390.4 ng (r=0.999 7), 36.8-368.0 ng (r=0.999 4), 17.0-170.4 ng (r=0.999 4), and 19.2-192.0 ng (r=0.999 4), respectively, and that determined by HPLC-FLED were in good linearity over the ranges of 3.7-292.8(r=0.999 4), 9.8-780.8(r=0.999 5), 9.2-736.0(r=0.999 4), 4.3-340.8(r=0.999 7), and 4.8-384.0 ng (r=0.999 6), respectively.Under UV detector, the limits of detection (LOD) were 0.32, 0.84, 0.61, 1.28, 2.88 ng, respectively, the limits of quantification (LOQ) were 1.05, 2.79, 2.04, 4.26, 9.60 ng, respectively.Under fluorescence detector, the limits of detection (LOD) were 0.03, 0.12, 0.18, 0.09, 0.07 ng, respectively, the limits of quantification (LOQ) were 0.09, 0.39, 0.61, 0.28, 0.24 ng, respectively. Conclusion: HPLC-FLED without interference from endogenous substances is simpler and more accurate than HPLC-UV, and it is more suitable for quantitative determination and quality control of Chinese rhubarb.

      摘要:Objective: This paper is to research the influence of covering plastic mulch on the yield and characteristics for Salvia miltiorrhiza, and the results will give a method to improve the cultivation and quality of the medicine plant. Method: The biological characteristic was investigated in the field. HPLC method was used to determine the liposoluble constituents (dihydrotanshinone, cryptotanshinone, tanshinoneⅠ, tanshinoneⅡA) and water-soluble constituents (salvianolic acid B, rosemarinic acid) of the difference cultivation mode. Result: The fresh and dry weight was increased by 18.20% and 48.62% under the cultivated condition by covering plastic mulch compared to the traditional cultivation. Meanwhile, the drying rate of the root cultivated by covering plastic mulch was increased by 23.08% compared to the condition without covering plastic mulch, while the dry rate of the up ground was decreased by 6.13%. The contents of liposoluble constituents and salvianolic acid B in the root cultivated by covering plastic mulch were increased 58.20% and 16.06%, respectively. Conclusion: The growth of S. miltiorrhiza could be improved by covering plastic mulch, meanwhile, the yield and the contents of constituents were increased significantly.

      摘要:Objective: To investigate the excretion kinetics of the effective components of ligustilide, butylphthalide and senkyunolide A in supercritical CO2 extraction part after oral administration of Xiongqiong Tang in rats. Method: The urine and feces samples of rats were collected according to different time periods after oral administration, the concentration of the above three compounds were determined by HPLC, the excretion curves were established and the excretion characteristics were studied using 3P 97 pharmacokinetic software. Result: The excretion curves showed that the urinary excretion rate of ligustilide increased gradually from 0 to 24 h, and then decreased after that, while in the feces the excretion rate of ligustilide approached the maximum from 36 h to 48 h and decreased whereafter;the urinary excretion rate of butylphthalide approached to the maximum gradually from 12 h to 48 h;the excretion rate of senkyunolide A in feces approached the maximum from 12 h to 48 h. Conclusion: The cumulative excretion-time curves and the average excretion rate-time curves of ligustilide are in accordance with butylphthalide in urine, so are the ligustilide and senkyunolide A in feces, which reflectes a kind of common excretion characteristics. These findings provide reference for investigating the integral excretion pattern of Xiongqiong Tang.

      摘要:Objective: To prepare vincamine sustained-release pellets and analyze its pharmacokinetics in Beagle dogs for providing an reference for non-pH-dependent release of sustained-release formulations of vincamine. Method: Vincamine sustained-released pellets were prepared by fluidized bed with self-made tartaric acid pills core.Beagle dogs were randomly assigned to receive sustained-release pellets and common tablets with dosage of 150 mg, then a crossover trial was conducted one week later.HPLC was employed to determine plasma drug concentrations to compare pharmacokinetic parameters of each formulation, mobile phase was 0.02 mol·L-1 potassium dihydrogen phosphate-acetonitrile(70: 30) by adjusting pH 2.3 with phosphoric acid, detection wavelength was 268 nm. Result: Pharmacokinetic parameters of sustained-release pellets and common tablets were as follows:Cmax of (1 147.40±554.50), (1 506.10±327.44) μg·L-1;Tmax of (2.38±0.25), (1.63±0.25) h;MRT of (6.75±2.44), (3.19±0.73) h;AUC0-t of (4 766.76±1740.26), (3 906.81±686.53) μg·h·L-1, respectively.Relative bioavailability of sustained-release pellets to common tablets was 122%.Dissolution test indicated that sustained-release pellets had good in in vivo and in vitro correlation. Conclusion: Compared with common tablets, vincamine sustained-release pellets show slower absorption in Beagle dogs, the average residence time is two times more than the former, relative bioavailability has also been significantly improved, indicating that this formulation can achieve non-pH-dependent release in the gastrointestinal tract.

      摘要:Objective: To screen urine biomarkers of rats after administrated aconitine and find out its related metabolic pathways. Method: Wister rats were divided into two groups which corresponded to control group(CG) and medication administration team (MAT), respectively.In each group, there were five male rats and five female rats.Biological samples were handled by liquid-liquid extraction method.Ultra high performance liquid chromatography (UPLC) coupled with time of flight mass spectrometry (TOF-MS) was applied, positive and negative ion mode was used to monitor endogenous metabolites in urine samples of rats, principal component analysis was applied to distinguish biological samples, biomarkers were selected by partial least square discriminant analysis and found out related metabolic pathways of aconitine. Result: Four potential biomarkers were identified, which were propionylglycine methyl ester, Cys-Ser-Met, pyrrole-2-carboxylic acid and 5, 8-dihydroxy-3, 4-dihydrocarbostyril. Conclusion: This study indicates that analgesia of aconitine may be associated with reduction of serotonin secretion, which can provide research basis for further study on mechanism of aconitine poisoning.

      摘要:Objective: To study the possible mechanism of Buzhong Yiqi Tang in treatment of spleen Qi deficiency syndrome amyosthenia in rats. Method: A combined method of combined techniques and overstrain were used to establish a spleen Qi deficiency model in rat. Cabbage 15-20 g was given to rats every day, 3 mL pig grease was orally given to rats in every other day for 15 days. After a successful modeling, the rats were divided into normal control group, spleen Qi deficiency group, Buzhong Yiqi Tang treatment group(high dose, middle dose, and low dose of 7.10, 3.55, 1.78 g·kg-1 respectively), and Buzhong Yiqi Tang was orally administrated for 10 days. The weight, rectal temperature, resistance to cold survival percentage and swimming time of rat were detected, and the expression of p-MLC was detected by Western blot, the value was compared before and after the treatment. Result: The weight, rectal temperature, resistance to cold survival percentage, swimming time and the expression of p-MLC in model group had a significant decrease compared to normal control group. The above mentioned indexes in middle dose and high dose groups of Buzhong Yiqi Tang had a significant increase compared to model group. Conclusion: Buzhong Yiqi Tang showed an up-regulation expression of p-MLC, which may be an mechanism for the treatment of spleen Qi deficiency syndrome amyosthenia in rats.

      摘要:Objective: To study the effects of total flavonoe of Epimedium(TFE)from Epimedium acuminatum and E. Leptorrhizum grown in Guizhou on insulin resistance (IR)-related factors in mice induced by alloxan(ALX). Method: Hundred male mice were divided into normal group and model group. The mice in model group were fed with high-fat diet for 6 weeks, then single intraperitoneal injection of 2% ALX solution (200 mg·kg-1) was made to establish type 2 diabetes models.The modeling mice were randomly divided into 6 groups with 10 in each group, including metformin group(150 mg·kg-1), E.acuminatum TFE group(100, 50 mg·kg-1), E.leptorrhizum TFE group(100, 50 mg·kg-1), respectively, once a day for 14 days. After 14 days, the animals were fasted for 12 hours, and killed. Then the level of serum insulin (FINS) and C peptide was determined.The blood sugar (FBG) and BG after injection of ins 40 min were detected. Insulin tolerance (ITT), insulin resistance index (HOMA-IR) and pancreatic index were calculated. HE staining was used to observe the morphologic changes of pancreas in mice. Result: Compared with the normal group, blood sugar of the model group increased significantly(P<0.01), C-peptide and FINS significantly increased(P<0.05), pancreatic tissue injury was serious(P<0.01), the pancreas index decreased significantly(P<0.01). Compared with model group, two TFE from E. acuminatum all obviously improved ITT(P<0.05), pancreatic index(P<0.05), reduced C-peptide, FINS and HOMA-IR(P<0.05) and obviously alleviated pancreatic tissue injury(P<0.05) in mice induced by ALX. TFE from E. acuminatum had more obvious effect(P<0.05, P<0.01). Conclusion: Two kinds of TFE from E. acuminatum have certain effect on the type 2 diabetes mice induced by ALX, and can increase insulin sensitivity, improve IR-related factors on the type 2 diabetes mice, and have a good protective effect on the pancreatic tissue injured. Especially, TFE from E.acuminatum has obvious effect. Epimedium's active ingredient on diabetes may has a greater relationship with icariin.

      摘要:Objective: This study sought to investigate whether n-butanol part of Pterocephalus hookeri (YSC-ZDC)in vitro inhibits the invasion and metastasis of human liver cancer Hep3B cells effective extraction, and its antitumor mechanisms were discussed. Method: Hep3B cell proliferation was assayed by MTT method, the fluorescent staining was used to observe the effect of YSC-ZDC on Hep3B cell apoptosis. Scratch test and Transwell were used to detect invasion and migration of Hep3B cell by YSC-ZDC, Western blot was uswed to detect related signaling pathways. Result: YSC-ZDC 50, 100, 200 mg·L-1 24, 48, 72 h after treatment could obviously inhibit proliferation. 50, 100, 200 mg·L-1 YSC-ZDC after 24 h could obviously inhibit the invasion and migration;YSC-ZDC could significantly affect the epithelial-mesenchymel transitions(EMT) related proteins. Conclusion: YSC-ZDC can significantly inhibit Hep3B cells proliferation and invasion and metastasis, and induce the Hep3B cell apoptosis. Its mechanism may be related to EMT.

      摘要:Objective: To investigate the effects of Zuogui Jiangtang Jieyu (ZJJF) prescription on expression of Bcl-2, Bax and cysteinecontaining asparate specific protease(caspase 8) in Hippocampus of diabetes mellitus rats with depression (DD). Method: DD model was established by the combination of high-fat diet, injection of streptozotocin(STZ, 38 mg·kg-1), and chronic stress.The model rats were randomly divided into three groups, the DD group, metformin(0.18 g·kg-1)combined fluoxetine(0.0018 g·kg-1)group and ZJJF(32.82 g·kg-1) groups .After 4 weeks of administration, the behavior were observed by the open-field test(OFT) test;Western-blotting was used to detect the expression of B cell lymphoma/leukemia 2(Bcl-2), Bcl-2-associated X protein(Bax)and Caspase 8 protein on hippocampal. Result: Compared with the normal group, the locomotor activity of model group were significantly reduced (P <0.01). Compared with the model group, the locomotor activity of rats in ZJJF(32.82 g·kg-1) and metformin(0.18 g·kg-1)combined fluoxetine(0.0018 g·kg-1)groups were significantly increased(P<0.01). Compared with the normal group, The Bax and Caspase 8 expression of hippocampal were significantly increased (P <0.01) and Bcl-2 was reduced(P <0.05) in model group through Western-blotting. Compared with the model group, the average level of hippocampal Bcl-2 was increased (P <0.05) and the Bax and Caspase 8 were significantly reduced (P <0.01) in positive drug group.Compared with the model group, the Bax and Caspase 8 expression were decreased (P <0.05, P <0.01) and Bcl-2 was increased(P <0.05) in ZJJF(32.82 g·kg-1). Conclusion: The depressive behavior could be improved by ZJJF, which might be correlated to the inhibition of neuronal apoptosis for protecting the hippocampal.

      摘要:Objective: To explore the influence of endothelial function and blood rheology in diabetic rats by paeonol and tanshinol. Method: Diabetic model was established by streptozotocin, the modeling 40 rats were randomly divided into model group, paeonol group(2.5 mg·kg-1, ig), tanshinol group(5 mg·kg-1, ig), paeonol combined with tanshinol group. Another 10 healthy rats were taken as normal control group, normal group and model group mice were given distilled water, the administration lasted 8 weeks. Body weight, blood sugar, the endothelial function and hemorheology index changes of rats were determined. Result: ①Compared with model group, there was no significant difference for the blood sugar in the phenol group, tanshinol group and combined group, compared with normal group, there was no significant difference. ②Compared with normal group, nitric oxide(NO), endothelin(ET-1), von Willebrand's factor(vWF) and plasminogen astivator inhibitor-1(PAI-1) were increased in model group (P<0.05), after 8 weeks treatment, compared with model group, there were no significant difference for vWF and NO in the phenol group, tanshinol group, NO in combined group significantly increased (P<0.05), vWF decreased (P<0.05);there were no significant for ET-1 in the phenol group, and ET-1 of tanshinol group and combined group significantly increased (P<0.05). There were significant difference for PAI-1 in the phenol group, tanshinol group and combined group(P<0.05);fibrinogen(Fg) in the phenol group had no significant differences, and Fg in tanshinol group and combined group had significant difference (P<0.05). ③Compared with normal group, in model group, whole blood low shear viscosity, whole blood high shear viscosity, plasma viscosity, erythrocyte and fibrin deposition pressure had significant difference (P<0.05). Compared with model group, the whole blood viscosity and high and low shear in the phenol group, tanshinol group and combined group were significantly lower (P<0.05), red blood cells were significantly lower (P<0.05), red blood cells of deposited in joint group were higher than that of group (P<0.05). Fibrous protein in tanshinol group and combined group was significantly lower than that of model group (P<0.05). Conclusion: Paeonol combined with tanshinol can improve the blood rheology of experimental diabetic rats, has certain protective effect on endothelial function of diabetic rats.

      摘要:Objective: To screen effective part of Alpiniae Oxyphyllae Fructus by comparing protective effect of different polar extract parts on diabetic nephropathy(DN). Method: Streptozotocin was adopted to establish a mice model of DN, DN mice were divided into the normal group and the model group by ig water of 10 mL·kg-1, the irbesartan group(ig by 10 mL·kg-1), Alpiniae Oxyphyllae Fructus extract groups(ig petroleum ether, chloroform, ethyl acetate, n-butanol fractions by 20 g·kg-1·d-1), various extraction sites interventions were regularly observed blood glucose and blood biochemistry, timed and measured urinary protein excretion in urine, in order to determine effective part of its anti-diabetic nephropathy. Result: Compared with the normal group, blood glucose of other groups significantly increased without significant difference among these six groups;with extension of administration time, blood glucose of each drug group did not significant decrease.Compared with the normal group, 24 h urine protein of the model group significantly increased, this index of each treatment group significantly reduced by comparing with the model group.Body weight significantly reduced and relative kidney increased singificantly of each group by comparing with the normal group, after drug intervention, each treatment group had opposite effect by comparing with the model group.After administration, serum creatinine(SCr) and blood urea nitrogen(BUN) of each group increased signigicantly by comparing with the normal group, but this index of each treatment group had oppsite effect by comparing with the moedl group.Petroleum ether part of Alpiniae Oxyphyllae Fructus showed better results than other parts, its urine protein, BUN and SCr decreased significantly.Urine protein in 24 h, SCr and BUN were (1.99±0.21) mg, (121.16±9.66) μmol·L-1 and (7.70±0.41) mmol·L-1, respectively. Conclusion: It is suggested that petroleum ether fraction of Alpiniae Oxyphyllae Fructus may be the effective fraction for anti-diabetic nephropathy.

      摘要:Objective: To discuss the effects of Jiagasong decoction (JGSD) on oxidative damaged liver cells and to explore its possible Nrf2/Bach1-ARE-antioxidant enzymes pathway mechanism. Method: The liver injury cells model was induced by 100 U·L-1 glucose oxidase (GO) and evaluated by MTT. The experiment included 9 groups:the control group, the model group, 20% JGSD contained serum group, 5 μmol·L-1 all-trans retinoic acid (ATRA) group, 80 μmol·L-1 genstein group, 80 μmol·L-1 tert-butylhydroquinone (t-BHQ) group, t-BHQ+20%JGSD contained serum group, ATRA + 20% JGSD contained serum group, genstein + 20%JGSD contained serum group. The activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), malonaldehyde (MDA), glutathione peroxidase (GSH-Px) were tested. The location in cells and nucleus protein levels of Nrf2 and Bach1 were tested by immunofluorescence and Western blot. Result: Compared to the control group, ALT, AST and MDA contents incearsed, GSH-Px activity and cell proliferation decreased, and levels of Nrf2 and Bach1 increased in the model group (P<0.01). The above indexes had good improvement in JGSD-contained groups (P<0.05, P<0.01). Moreover, there was coordinated effects between JGSY and the other three agents on Nrf2 expression (P<0.01). Meanwhile, JGSD coule promote the Bach1 export from cell nuclear (P<0.01). Conclusion: JGSD could protect GO-induced cells damage from oxidative stress, and its antioxidation mechanism is related to regulating Nrf2 and Bach1 protein levels in nuclear.

      摘要:Objective: To study the treating effect on early closed fractures using Taohong Siwu Tang (TST). Method: Sixty SD rats were randomly divided into six groups:normal group, model group, TST-treated group (18, 9, 4.5 g·kg-1) and Shenxiang Jiegu group (0.6 g·kg-1). The tibia closed fracture model rats were created using a dedicated fracture pusher. The influence on blood viscosity of TST was observed. The plasma thromboxane (TXB2) and 6-ketone prostaglandin (6-keto-PGE1α) content were detected by RIA, the serum vascular endothelial growth factor (VEGF) content was assayed by ELISA. The situation of callus microvascular and fracture healing were observed using bone biopsy. Result: Compared with the normal group, model group showed severity of fracture defects and the blood viscosity increased significantly (P<0.01). Additionally, both plasma TXB2 and TXB2/6-keto-PGF1α elevated markedly (P<0.01), and the content of VEGF increased slightly. Compared with the model group, all doses of TST groups could significantly stimulate the regeneration of blood vessel of callus in early phase and promote the healing of fracture. Meanwhile, it could reduce blood viscosity of fracture in rats, decrease the levels of plasm TXB2 and TXB2/6-keto-PGF1α significantly and increase the level of serum VEGF obviously (P<0.05, P<0.01). Especially, the effect of high dose group is close to the effect of positive drug group (P<0.01). Conclusion: TST has good stasis-removing and regeneration-promoting effect on early closing fractures, the mechanism may be related to decreasing the levels of plasm TXB2 and 6-keto-PGE1α, increasing the level of serum VEGF.

      摘要:Objective: To investigate the mechanism of Shenling Baizhu San (SLBZS) in treating dextra sulfate sodium (DSS) -induced inflammatory bowel disease (IBD) mice. Method: Eighty SPF BALB/c mice were randomly divided into control group, model group, positive group, low-, medium- and high-dose SLBZS groups. The IBD mice were fed 5% DSS in their drinking water for 7 days, and control mice received only water. The mice in SLBZS groups were given SLBZS at doses of 3, 6, 12 g·kg-1, the mice in positive group were given mesalazine sustained release granules at dose of 2 g·kg-1, and the control mice were given the same volume of normal saline by intragastric administration. The mice weight, stool occult blood in the stool, the calculation of disease activity (DAI), pathological examination of intestinal mucosal lesions integral were observed after 15 days. The expression of Claudin protein was tested by Westorn bolt. The expression of Occluding, Claudin, ZO-1 and JAM mRNA were analyzed by RT-PCR. Result: The lesion was obvious, Clandin protein decreased obviously in the model group as compared with the control group (P<0.05). Claudin protein, Occluding, Claudin, ZO-1 and JAM mRNA increased in DSS and all doses SLBZS groups as compared with the control group (P<0.05). Conclusion: SLBZS has good effect in DSS-induced IBD mice by the regulation of intestinal epithelial cell tight junctions.

      摘要:Objective: To explore the pharmacological effect and possible meachanism of Yougui Wan on Shen yang deficiency based on hypothalamus-pituitary-gonad axis. Method: The Shen yang deficiency model rats were induced by hydrocortisone. The 60 rats were randomly divided into six groups:the control group, the model group, the methyltestosterone group (0.5 g·kg-1), low-, middle-and high-dose Yougui Wan groups (0.5, 1.0, 2.5 g·kg-1). Changes of forced swimming time were tested at 15 and 30 days. Serum levels of testosterone (T) and estradiol (E2) were analyzed by immunobiochemistry, and calmodulin (CaM) mRNA in hypothalamus-pituitary-gonad axis of rats in all groups were determined via quantitative RT-PCR technique after 30 days. Result: The forced swimming time decreased, blood concentration of E2 increased, blood concentration of T decreased, and CaM mRNA levels in rat hypothalamus, pituitary and testicle increased in the model group as compared with the normal group (P<0.05). All above indexes had good improvement in methyltestosterone group and all doses Yougui Wan groups as compared with the model group (P<0.05). Conclusion: Yougui Wan have therapeutical effect on down-regulating the CaM mRNA levels of testical and hypothalamus, regulating the levels of E2 and T, and improving the symptoms of Shen yang deficiency.

      摘要:Objective: To screen the effective parts of Echinopsis Radix through bacteriostasis and anti-inflammatory activities. Method: Echinopsis Radix was firstly extracted by ethanol, and followed by petroleum ether, chloroform, ethyl acetate, and n-butanol to obtaine the different parts of extraction. Mice were randomly divided into blank group, aspirin group (0.11 g·kg-1), and high-, low-dose of different extraction parts groups. The anti-inflammatory activity of different solvent extraction parts of Echinopsis Radix was examined by xylene-induced ear swelling mice. The effects against Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Salmonella typhimurium, and Pseudomonas aeruginosa were examined by scraping line method and tube dilution method. Result: The chloroform extraction part of Echinopsis Radix showed an inhibition rate of higher than 60% in ear swelling mice, while the effect of the other extraction parts was very weak. The ethyl acetate extraction part showed the lowest MIC against S. aureus and B. subtilis (312.5 g·L-1), while the n-butanol extraction part showed the lowest MIC against S. aureus and P. aeruginosa (312.5 g·L-1). Conclusion: It shows that the chloroform extraction is the anti-inflammatory part of Echinopsis Radix, the ethyl acetate extraction and n-butanol extraction are the bacteriostatic parts. It provides the evidence for the further development and utilization of Echinopsis Radix.

      摘要:Objective: To explore the molecular mechanisms of Changweiqing in treating colon cancer invasion and metastasis. Method: The experiment included the blank group, the low-, medium-, high-dose Changweiqing groups (0.98, 1.97, 3.93 g·L-1), the oxaliplatin group(9.23 mg·L-1), and the combination group (1.97 g·L-1 Changweiqing plus 9.23 mg·L-1 oxaliplatin). The colon cancer LoVo cells were used in this study. The mRNA and protein of E-cadherin, Wint/beta chain protein signaling pathway factor β-catenin and matrix metalloproteinase-7 (MMP-7) were detected by RT-qPCR and Western blot assay. Result: Compared to the blank group, the mRNA, protein expression of E-cadherin increased, the mRNA expression of β-catenin and MMP-7 decreased in LoVo cells at all doses of Changweiqing groups, oxaliplatin group and combined group (P<0.05, P<0.01). Meanwhile, the results were better in the combination group. Conclusion: Changweiqing has synergetic effect with oxaliplatin by increasing the expression of E-cadherin and enhancing the adhesion between cells, inhibiting Wint/β-catenin signaling pathway activity, decreasing the expression of β-catenin and MMP-7.

      摘要:Objective: To observe protective effect of Gugutou Huaisiyu capsule containing serum on apoptosis in rat bone marrow mesenchymal stem cells(BMSCs). Method: The BMSCs were separated, filtrated, cultured, and high, medium and low dose Gugutou Huaisiyu capsule containing serum had harvested from mice under 0.36, 0.72, 1.44 g·kg-1 Gugutou Huaisiyu capsule gavaged, the third generation BMSCs were chose and divided into blank group (no rat serum in culture medium), normal group (with volume fraction of 20% normal rat serum in culture medium), induction apoptosis group (with volume fraction of 20% normal rat serum and all-trans retinoic acid in culture medium) and traditional Chinese medicine serum low, medium and high dose group (with volume fraction of 20% Gugutou Huaisiyu capsule containing serum of low, medium and high dose respectively, and all-trans retinoic acid in culture medium), a total of six groups.Then cell relative survival rate and cell cycle were detected by MTT method and flow cytometry, respectively. Result: Compared with normal group, the cell relative survival rate of induction apoptosis group, traditional Chinese medicine containing serum low, medium and high dose groups was increased, and G1 phase increased, G2 and S phase proportion, the differences were statistically significant (P<0.01). Compared to the induction apoptosis group, the cell relative survival rate in traditional Chinese medicine containing serum low, medium and high dose groups was declined, while the G1 phase ratio decreases, G2 and S phase proportion increase, and the difference increased obviously as the dosage increased, the differences were statistically significant (P<0.01). Conclusion: Gugutou Huaisiyu capsule containing serum could improve the survival rate of rat BMSCs, and have protective effect on apoptosis induced by all-trans retinoic acid.

      摘要:Objective: To study the effect of andrographolide on aortic micro-inflammation, atheromatous plaque formation and aortic pathology in atherosclerosis rabbit. Method: Thirty six SPF Newzealand rabbits were divided into normal group, model group, fenofibrate group(25.67 mg·kg-1), and high, medium and low dose of andrographolide group (93.34, 46.67, 23.33 mg·kg-1). Except for the normal control group, the rest animals were used to establish atherosclerosis model with bovine serum albumin and high-fat diet for 12 weeks. After the modeling, the rabbits were treated with andrographolide for 4 weeks. Result: Compared with normal group, the level of total cholesterol (TC), triglyceride(TG), low density lipoprotein cholesterol (LDL-C), interleukin-1β(IL-1β)and high-sensitivity C-reactive protein(hs-CRP), the expression of vascular cell adhesion molecule-1(VCAM-1), monocyte chemoattractant protein-1(MCP-1)and platelet derived growth factor (PDGF) of the model group were obviously increased, the level of high density lipoprotein cholesterol (HDL-C) was reduced, the difference was statistically significant(P<0.01).Compared with model group, the ratio of Interferon γ/Interleukin 4 (IFN-γ/IL-4), the level of TC, TG, LDL-C, IL-1β and hs-CRP, the expression of VCAM-1, MCP-1 and PDGF mRNA of the andrographolide group were obviously reduced, the level of HDL-C was increaed, the difference was statistically significant(P<0.01). Conclusion: Andrographolide could inhibit the inflammation process, and delay the development of atherosclerosis.

      摘要:Objective: To study the effect of cytokines of polyporus polysaccharide(PPS)on lipopolysaccharide (LPS)-stimulated J774, and to explore its anti-inflammatory mechanism. Method: J774 cells were inoculated in 6-well plates with the density of 2×105 per well.The experiment was divided into blank group, model group, LPS plus low-dose PPS group(1 mg·L-1), LPS plus middle-dose PPS group(10 mg·L-1) and LPS plus high-dose PPS group(100 mg·L-1) were set up in this study.Each group had six wells.J774 cells were polarized to inflammatory macrophage by treating with 10 mg·L-1 LPS for 3 h.Meanwhile, LPS-stimulated J774 cells were treated with PPS of 1, 10, 100 mg·L-1 for 3 h.The expressions of interleukin-1β(IL-1β), interleukin-10(IL-10), tumor necrosis factor-α(TNF-α), interferon-γ(IFN-γ) and interleukin-6(IL-6) mRNA were detected by real-time quantitative PCR (RT-PCR).And the expressions of p38 mitogen-activated protein kinase(p38), extracellular signal-regulated kinase(ERK42/44), p65 mitogen-activated protein kinase (p65) protein were determined by using Western blotting. Result: Compared to blank group, the percentages of IL-1β, IL-10, TNF-α, IFN-γ and IL-6 mRNA of model group were significantly higher(P<0.01).It meant the inflammation model was successfully established.After treated with PPS, the expressions of IL-1β, IL-10, TNF-α, IFN-γ and IL-6 mRNA were reduced.And the percentages of p38, ERK42/44, p65 protein were decreased(P<0.01, P<0.05). Conclusion: PPS could reduce inflammation in LPS-stimulated J774 cells.It might be achived through the mitogen activated protein kinase(MAPK) pathway to reduce inflammation damage.

      摘要:Objective: To explore the material basis of Banxia Xiexin Tang in regulating gastrointestinal motility. Method: The two kinds of model were established by subcutaneous injection of atropine sulfate and chronic water immersion stress respectively.Domperidone(0.01 g·kg-1), Banxia Xiexin Tang(2 g·kg-1), pungent drugs, bitter drugs and tonic with sweet drugs(1.5 g·kg-1) were orally given for 4 days. The remnant rate of stomach and progradation rate of small intestine were measured. Result: Compared with normal group, the gastric retention rate of model group increased, and the small intestinal propulsion decreased. The difference was statistically significant(P<0.05, P<0.01). When the model rats were orally given different drugs, both of the 2 indicators had different degrees of change. The banxia xiexin decoction decreased the gastric retention rate of the model rats. The ethanol precipitation of Banxia Xiexin Tang, the bitter drugs and the ethanol precipitation of bitter drugs decreased the gastric retention of normal rats and the two kinds of model rats. The difference was statistically significant (P<0.05, P<0.01). Conclusion: Banxia Xiexin Tang may be play the role of regulating gastrointestinal motility, the bitter drugs for purgation are the effective group mainly and the ethanol precipitation parts are the effective components.

      摘要:Objective: To investigate the effects of Jianpi Yangzheng Xiaozheng decoction (JYXD) on gastric cancer growth, liver metastasis situation and metastasis-related genes. Method: The human gastric cancer model in nude mice was established and the mice were randomly divided into control group, 5-Fu group, high-and low-dose JYXD groups. The inhibition rate of gastric cancer xenografts and liver metastases situation in nude mice were observed. The gene expressions of matrix metalloproteinases 2 (MMP-2), MMP-9, MMP-14, vascular endothelial growth factor (VEGF) and reversion inducing cysteine-rich protein with Kazal motifs (RECK) were detected by transcription-polymerase chain reaction (RT-PCR) assay. The protein expressions of signal transducer and activator of transcription3 (STAT3), MMP-2, MMP-9, MMP-14, VEGF and RECK were detected by Western Blot assay. Result: The average tumor weight was lower, the expressions of invasion and metastasis-related genes of MMP-2, MMP-9, MMP-14, VEGF decreased, the gene expression of RECK rised, the expression of invasion and metastasis-related proteins P-STAT3, MMP-2, MMP-9, MMP-14, VEGF decreased, and the protein expression of RECK rised in all dose JYXD groups as compard with the control group (P<0.01). Conclusion: JYXD could inhibite the growth of gastric cancer xenografts liver metastase and down-regulate the expressions of P-STAT3, MMP-2, MMP-9, MMP-14, VEGF genes or proteins and upregulate the expressions of RECK gene or protein.

      摘要:Objective: To observe the effects of total flavonoids of tartary buckwheat on palmitic acid-induced PI3K in EA.hy926 cells. Method: EA.hy 926 cells were cultured in vitro and divided into the control group, the model group, the total flavonoids of tartary buckwheat group and the metformin group. The PI3K mRNA and protein expression levels were determined by RT-PCR and immunocytochemistry, respectively. Result: Compared with the control group, the expression levels of PI3K mRNA and protein were significantly lower in the control group (P<0.01). Compared with the control group, the PI3K mRNA and protein expression increased markedly in both total flavonoids of tartary buckwheat group and metformin group (P<0.01), and there is no significant difference between two groups. Conclusion: Total flavonoids of tartary buckwheat could effectively promote the expression of PI3K mRNA and protein in endothelial cells under palmitic acid stimulation.

      摘要:Objective: To observe the effects of Butong paste (BT paste) combined with seretide on the lung function and immunologic function in patients with persistent asthma. Method: Before and after 15 days of the fortnightly periods of ‘Beginning of Winter’, 105 patients were randomly divided into 3 groups:the combination group, the BT paste group and the seretide group. Patients in the BT paste group received 10 mL BT paste, patients in the seretide group received seretide (50/250), and patients in the combination group received above two treatments each morning and evening for 3 months. Before and after treatment, asthma control test (ACT) values of three groups were observed, the levels of the first second forced expiratory volume percentage of expected value (FEV1%) and serum interferon-γ (INF-γ), interleukin 4 (IL-4) in serum were measured. Result: Compared with the other two groups, the levels of ACT value, FEV1%, INF-γ in serum were higher (P<0.05), while the levels of IL-4 were lower (P<0.05) in the combination group after treatment. Conclusion: The mechanisms of BT paste in the treatment of asthma are involved in IL-4, INF-γ and other aspects. The study also shows that the BT paste combined with seretide has synergetic effect, which could improve the lung function and regulate the immunity in treating persistent asthma. This treatment method is worthy of wide application.

      摘要:Objective: To study the efficacy and safety Yinqiao San combined with cefoperazone/sulbactam in treating community-acquired pneumonia(CAP). Method: CAP were collected as the research object and randomly divided into observation group given Yinqiao San and cefoperazone/sulbactam and control group given cefoperazone/sulbactam. Then recovery, metabolism of drugs and adverse reaction were observed. Result: Clinical pulmoncoy infection score(CPIS) score of observation group after treatment was obviously lower than before treatment from 5 d which was earlier than control group; drug concentration peak Cmax had no difference with control group, time to peak Tmax, t1/2 and adverse reaction cases were significantly less than control group. Conclusion: Yinqiao San combined with cefoperazone/sulbactam have positive clinical value for its fast metabolic, good curative effect and less adverse reaction.

      摘要:Objective: To discuss the curative efficacy and the mechanism of Langchuangshen number I decoction combined hormone and immune inhibitor in treating lupus nephritis (LN). Method: Ninety-four patients with LN were randomly divided into the western medicine group (47 cases) and the combination group (47 cases). Patients in the western medicine group received methyl prednisolone tablets and cyclophosphamide injection. Based on the treatment of western medicine group, patients in the combination group added orally Langchuangshen number I decoction. The patients in both two groups received 6-month treatment. The systemic lupus erythematosus disease activity score (SLEDAI) at the second, fourth and sixth months after treatment were recorded. The 24-h urine protein quantitation, blood urea nitrogen (BUN) and serum creatinine (SCr) were tested before and after treatment. Besides, the levels of immunoglobulin G (IgG), complement C3, complement C4, and the levels of resistance of double-stranded DNA (ds-DNA), plasma fibrinogen (FIB), D-dimer (D-D) were detected. Result: The clinical effect of proteinuria in the combination group was superior to that in western medicine group (P<0.05). Score of SLEDAI in the combination group was less than that in western medicine group at the fourth and sixth months after treatment (P<0.01). The amount of 24-h urine protein in the combination group was less than that in western medicine group after treatment (P<0.01). The number of completely relieve cases in the combination group was more than that in western medicine group during six months. But there was no statistically significant difference between two groups. The average alleviate time in the combination group was shorter than that in western medicine group (P<0.05). Levels of IgG, ds-DNA, FIB and D-D in the combination group was inferior to that in western medicine group (P<0.01). Besides, complement C3 and complement C4 levels in the combination group were higher than those in western medicine group (P<0.05, P<0.01). Conclusion: Langchuangshen number I decoction combined hormone and immune inhibitor coud control the disease activity in LN patients, improve blood high condensation state, protect renal function, reduce urinary protein and cut down the disease course. The curative effect of traditional Chiese medicine combined western medicine treatment is superior to western medicine treatment alone in treating LN.

      摘要:Objective: To discuss the curative efficacy of Qingfei Huazhuo decoction in assisted-treating acute exacerbation of chronic obstructive pulmonary disease (AECOPD) of phlegm heat obstructing lung syndrome and to investigate its influence on serum superoxide dismutase (SOD), malondialdehyde (MDA) and interleukin-10 (IL-10) and tumor necrosis factor-α (TNF-α). Method: One hundred AECOPD patients were randomly divided into the control group (50 cases) and the traditional Chinese medicine group (50 cases) by random number table. Patients in the control group received comprehensive treatment therapy including controlling oxygen therapy, infection resistance, hormone and bronchial relaxation. Based on the treatment of control group, patients in traditional Chinese medicine group added Qingfei Huazhuo decoction, 1 does daily. All patients received 2-week treatment. Scores of the main symptoms including cough up phlegm, cough, asthma and pulmonary rales were recorded before and after treatment. Quality of life through respiratory function state and difficulty in breathing questionnaire-modified (PFSDQ-M) were evaluated. The pulmonary function was appraised before and after treatment. Levels of serum SOD, MDA, IL-10 and TNF-α were tested before and after therapy. Result: The clinical effect in traditional Chinese medicine group was superior to that in control group with statistically significant difference (P<0.05). After treatment, scores of cough up phlegm, cough, asthma and pulmonary rales in traditional Chinese medicine group were less than those in control group (P<0.01). Quality of life score in traditional Chinese medicine group was inferior to that in control group (P<0.01). Forced expiratory volume in one second(FEV1) and forced expiratory volume accounted for forced vital capacity(FEV1/FVC) in control group improved, but there was no statistically significant difference. Expected value of FEV1 and FEV1/FVC in control group rose as compared with that before therapy, while the rose degree was higher than that in control group (P<0.01). Levels of serum TNF-α and MDA in traditional Chinese medicine group were lower than those in control group (P<0.01). Besides, the IL-10 and SOD levels were higher than those in control group (P<0.01). Conclusion: Qingfei Huazhuo decoction could reduce clinical symptoms of AECOPD of phlegm heat obstructing lung syndrome, improve lung function and patient's life of quality. The mechanism of action may be related to adjusting oxidation/antioxidant system and inhibiting inflammatory reaction.

      摘要:Objective: To discuss the curative efficacy of Xiaoke Tongguan decoction (XTD) combined conventional western medicine therapy in treating diabetes united coronary heart disease and to explore its influence on high mobility group protein B1 (HMGB 1) and serum retinal element 1 level. Method: Ninety-two patients were randomly divided into the control group (46 cases) and the combination group (46 cases) by random number table. Patients in the control group received western medicine comprehensive treatment including controlling blood glucose and blood pressure, aspirin enteric-coated tablets, 0.1 g, once daily, metoprolol tartrate tablets, 50 mg, once daily, simvastatin tablets, 10 mg, once daily and nitroglycerin tablets, 0.5 mg, sublingually taken when necessary. Based on the treatment in the control group, patients in the combination group added XTD, 1 does daily. All patients received 3-month treatment. Levels of fasting blood glucose (FBG), fasting insulin (FINS), glycosylated hemoglobin (HbAlc), HMGB 1 and serum visfatin 1 were tested. The insulin resistance index (HOMA-IR) was calculated and the electrocardiographic examination was performed before and after treatment. The anginal attacks and nitroglycerin dosage were recorded during observation period. The blood fat and score of Qi and Yin deficiency and blood stasis syndrome were tested. Result: After treatment, the efficient rate of the combination group was 96.65%, which was superior to 80.43% in control group (P<0.05). Levels of FINS, HOMA-IR and HbAlc in the combination group were less than those in control group (P<0.01). Cases of restoring normal with TG and HDL-C in the combination group were higher than those in control group (P<0.05). The number of angina pectoris attack each week and the dosage of nitroglycerin in the combination group were lower than those in control group (P<0.01). Nitroglycerin stopped reduction rate was 84.8% in the combination group, which was more than the rate of 65.2% in control group (P<0.05). Serum HMGB1 in both groups declined compared with the data before treatment. Besides, the degree of declining in the combination group was lower than that in control group (P<0.01). Level of serum retinal element 1 in both groups went up as compared with the data before treatment, while the risen degree in the combination group was higher than that in control group (P<0.01). Conclusion: On the basis of conventional western medicine treatment, adding XTD coud improve insulin resistance, enhance insulin sensitivity, regulate lipid metabolism, improve the symptoms of coronary heart disease. Its action mechanism may be related to reducing proinflammatory meson HMGB1 and enhancing the level of serum retinal element 1.

      摘要:Objective: To discuss the curative efficacy of compound Xuanzhu capsules combined with recombinant human tumor necrosis factor-α receptorⅡ:IgG fusion protein in treating rheumatoid arthritis (RA) patients of Shen and Qi deficiency syndrome. Method: Ninety-two patients with RA were randomly divided into the conventional treatment group, the control group and the combination group by random number table. Patients in conventional treatment group received celebrex, 0.2 g, once daily, methotrexate tablets, 10 mg, once weekly, and leflunomide tablets, 20 mg, once daily for oral administration. Based on the treatment of the conventional treatment group, patients in the control group added recombinant human tumor necrosis factor-α receptorⅡ:IgG Fc fusion protein for articular cavity injection once weekly. Based on the treatment of control group, patients in combination group added compound Xuanzhu capsules, 3 gains, thrice daily. All patients received 3-month treatment. Before and after treatment, numbers of swollen and tender joint (28 joints) were recorded and pain degree was assessed by visual analogue scales. Scores of morning stiffness, grip strength and Shen deficiency cold syndrome were recorded. Levels of erythrocyte sedimentation rate (ESR), c-reactive protein (CRP), rheumatoid factor (RF), tumor necrosis factor (TNF-α), a1-acidoglycoprotein (a1-AGP) and immune globulin (IgA, IgM, IgG) were detected. Result: After treatment, the clinical total effective rate in the conventional treatment group, the control group and the combined group were 79.31%, 90% and 96.97%, respectively. There was no statistically significant difference between any two of the three groups. The improvements of time of morning stiffness, grip strength, the number of hands rest pain, joint swelling, joint tenderness count for the control group were superior to those for the conventional treatment group (P<0.01). The improvements of hard-shelled time in the morning, grip strength, the number of hands rest pain, joint swelling, joint tenderness count for patients in the combined group were superior to those of the conventional treatment group (P<0.01). The improvements of morning stiffness time, hands grip strength, rest pain and Shen deficiency cold syndrome scores for the combined group were superior to those for the control group(P<0.05). Levels of TNF-α, RF, a1-AGP for the control group were lower than those in the conventional treatment group, while levels of ESR, CRP, TNF-α, RF and a1-AGP for the combined group were inferior to those for the conventional treatment group after treatment (P<0.05). Levels of ESR, CRP, TNF-α, RF and a1-AGP for combined group were lower than those for the conventional treatment group after treatment (P<0.05, P<0.01). IgG level for the control group was less than that in the conventional treatment group (P<0.01). IgA, IgM, IgG of the combined group were inferior to those in the conventional treatment group and the control group (P<0.01). Conclusion: Compound Xuanju capsules combined recombinant human tumor necrosis factor-α receptorⅡ:IgG Fc fusion protein could control symptoms, improve joint function, reduce syndromes and command inflammation.

      摘要:Objective: To provide evidence for further researches through systematicly reviewing and analyzing the literatures about Chinese herbal formula intervention on Alzheimer animal models. Method: Animal experiments on Alzheimer's disease using Chinese herbal formula were collected from China National Knowledge Infrastructure (CNKI), VIP, Wanfang and the Pubmed database in recent 10 years. Literature searching and screening, were conducted according to inclusion and exclusion criteria. Then the influence on outcome, pharmacologic mechanisms and resource consumption with different number herbs formulas was analysed. Result: Formulas with more kinds of herbs showed no advantage on efficacy, pharmacologic mechanisms and resource consumption than formulas with few kinds. Conclusion: It's necessary to optimize the compatibility of Chinese herbal formula in treating Alzheimer's disease, which would exhibt the overall advantages and increase the research values of Chinese herbs.

      摘要:Objective: To evaluate the efficacy of traditional Chinese medicine (TCM) of tonifying the kidney combined with western medicine in the treatment of senile dementia and to provide references for choosing suitable treatment plan in clinic. Method: The randomized controlled trials (RCTs) about TCM of tonifying the kidney combined with western medicine versus western medicine at home and abroad were comprehensively searched and collected. The data was extracted from each study and a meta-analysis was conducted with RevMan5.2.Result: Eleven studies were involved in the Meta-analysis, of which 8 studies reported on the effective cases were carried out by meta-analysis. The total RR was 1.39, 95%CI(1.25, 1.56) and the funnel plot was approximately symmetry. It was suggested that the effect of the therapy groups was better than the control groups (Z=5.77, P<0.01). Seven studies reported minimum mental state exconination(MMSE) scores were carried out by meta-analysis. The total MD was 2.13, 95%CI(0.84, 3.41) and the funnel plot was approximately symmetry. It was suggested that the therapy groups was better than the control groups on senile dementia in improving cognitive function (Z=3.24, P<0.01). Conclusion: TCM of tonifying the kidney combined with western medicine in treating senile dementia is effective. However, it still needs further study.

      摘要:Processing of Chinese materia medica is a pharmaceutical technology such as cleaning, slicing and other processing under direction of theories of traditional Chinese medicine(TCM) and in accordance with diagnosis of diseases, it is guarantee of safe medication and necessary means to improve efficacy.Processing after different methods, appearance, properties, chemical composition, pharmacological effects of Chinese materia medica will greatly changed, which can give full play to medicinal value and enhance clinical efficacy.This paper analyzed influences of processing on property of Chinese materia medica(four gas flavors, meridian, side effects, lift ups and downs), chemical compositions(alkaloids, glycosides, volatile oil, polysaccharides, tannins, organic acids, trace elements) and pharmacological effects(cough and asthma, stasis, anti-tumor, diarrhea, immunization), the overall impact of processing on Chinese materia medica was discussed, thus provided a scientific reference for rational application and modernization of Chinese materia medica.Showed in practical application of Chinese materia medica, disease needs to be combined with TCM theory and modern technology, appropriate method for processing drugs was selected in order to protect drug safety and give full play to efficacy, but processing of Chinese materia medica still exists simple processing, low productivity, vague mechanisms and other problems, it needs gradually deepening and refinement.By regulating processing standards of Chinese herbal pieces, this paper can clarify modern scientific connotation of traditional processing methods and establish an effective system of modern research as the key to further development of Chinese materia medica.

      摘要:Formulations study of sustained-release drug delivery system has made some progress for Chinese material medica, it is becoming a hotspot issue that to study synchronicity of multiple components release, but complex compositions of Chinese material medica make it difficult to study synchronicity of release.In this paper, recent related literature of studies in synchronous release of multiple components has been collected and analysed for Chinese medicine sustained-release preparations for oral administration, solid dispersion technology, layered preforming technology, multiple coating technology and osmotic pump technology are main technology to control synchronous release, synchronicity evaluation method mainly includes release similarity factor method and combination method of similarity factor and fingerprints.At this stage, control technology of synchronous release has made some achievements, but the degree of synchronous release is not high enough, synchronicity evaluation method is valuable at a certain extent, but not complete and systematic.Research on synchronous release is to absorb advantage of synchronous transport of Chinese material medica decoctions, in order to simulate original maximum effectiveness.At the same time, contrast research of synchronous release with non-synchronous release is still needed to confirm necessity and importance of synchronicity for achieving overall, synchronization and comprehensive action.

      摘要:With rapid development of economy, market competition of topical pharmaceutical preparation is becoming increasingly fierce, the focus of market competition of price and quantity has been gradually replaced by brand and product quality, many pharmaceutical production enterprises have gradually improved their product quality management to a new level, which becomes strategic direction of enterprise development.With regard to all-round improvement of production quality management level of modern production enterprise of topical pharmaceutical preparation, it is significant to a great extent that production enterprise improves awareness of GMP of their production operation staffs.The 2010 edition of good manufacturing practices(GMP) covers analysis both in terms of software and hardware, such as production areas and equipments modification, staffing and technical documentation, in order to improve the overall management so that high level GMP implementation of pharmaceutical production can be ensured.Taking topical preparation as research object, through a lot of data research and literature collection, focused on the creams as an example, in view of current market demand of pharmaceutical preparation for external use, it should apply modern product quality management model with scientific methods to all-round improve production and quality of topical pharmaceutical preparation and to cut losses caused by product quality problem, so that enterprises could enhance their competitiveness and to better cope with market competition.

      摘要:Coumarins, widely existed in Chinese herbal medicine, are important components in the natural medicine. They had shown various pharmacological effects, such as analgesic, anti-inflammatory, anti-tumor, anti-arrhythmic, etc. Coumarins can be quickly absorbed and distributed in human body, and concentrated mostly in the organs with rich blood such as heart, liver, lung, spleen and so on. The pharmacokinetic behavior of coumarins is firstly affected by delivery way in vivo. It is prior to one-compartment model when delivery way is extravascular administration such as oral, intraperitoneal injection and muscle injection and so on. When intravascular administration, such as intravenous injection and intravenous drip, the two-compartment model is primary. Second, it is affected by the distribution rate of coumarins as slow distribution phase are covered by absorption phase. So some drugs performance one-compartment model when oral administration, but it performances two rooms model when intravenous. In addition, the pharmacokinetic behavior of coumarins is also affected by animal species and individual differences in vivo, because of the CYP isoenzymes. The metabolic process of coumarins is complex, but the metabolic processes are similar because they have the same parent nuclear. A large numbers of enzymes are involved the metabolism of coumarins. When coumarins entered the body, phase Ⅰ metabolism occured firstly, such as hydroxylation, dehydrogenation, demethylation, lactone ring open-loop and so on, then phase Ⅱ metabolism reacted with glucuronic acid, sulfuric acid and glutathione etc. generated larger water-soluble compounds, and excreted finally. The author arranged documents on coumarins from recent years, reviewed the documents on biodistribution and metabolic mechanism of coumarins, which would lay the foundation for their development and clinical rational application.

      摘要:Pyrogallic acid, a widely used chemical raw material and reagent, was mostly prepared by the chemistry process of gallic acid decarboxylation. Since serious environment pollution and equipment corrosion with the above method, the article reviewed microbial strains and the related technology including mostly influence factors of yield, such as carbon sources, the concentration of substrate, pH and fermentation time from the perspective of microbial transformation. At the same time, related influence factors of the gallic acid decarboxylase (GAD) activity including pH, temperature, metal ions and additives were presented. Then, the article briefly introduced the treatment of wastewater made in the production process of gallotannin and other issues applying microorganism. Finally, some views and suggestions were put forward for the process for preparing pyrogallic acid with the new method in future, which would provide the guidance and reference for the industrial production of pyrogallic acid by microbial conversion.

      摘要:Change on pharmacological effects of Schisandrae Chinensis Fructus was decided by change on composition which was affected by of processing technologic parameters. To provide reference for scientific preparation and clinical application of Schisandrae Chinensis Fructus, a summary about the method, technology, chemical constituents, pharmacological action and analytical methods of which was reported in recent ten years was given. Through literature analysis, found that the best processing technology of Schisandrae Chinensis Fructus often factored to excipients, moistening time, processing time, but evaluation indexes were various. In addition, theoretical study of Schisandrae Chinensis Fructus prepared with wine was systemlized and the similar processing technology was given, while, there was no unified national processing technologic parameters of Schisandrae Chinensis Fructus prepared by vinegar or honey.
