      摘要:Objective: To optimize simple sequence repeat-polymerase chain reaction(SSR-PCR) system for providing a reference to genetic diversity and phylogenetic analysis among germplasms of Eucommiae Cortex. Method: Orthogonal design and single factor tests were adopted to optimize SSR-PCR system of Eucommiae Cortex with Taq DNA polymerase,template DNA,Mg2+,primers and dNTP concentration as factors,test data were analyzed by SPSS software. Result: Effects of each factors were in the order of Mg2+>Taq DNA polymerase>primers>dNTP>DNA template.Optimal reaction system was as follows:10×PCR buffer of 2 μL,Taq DNA polymerase of 0.5 U,Mg2+ of 1.25 mmol · L-1,dNTP concentration of 0.2 mmol · L-1,primers of 0.3 μmol · L-1 and DNA template of 60 ng in 20 μL reaction solution. Conclusion: Bands are bright and clear with good stability and reproducibility by amplification of this optimized SSR-PCR system,it is suitable for genetic diversity and phylogenetic analysis among germplasms of Eucommiae Cortex.

      摘要:Objective: To establish extraction method of total DNA from Aquilariae Lignum Resinatum and its rDNA ITS2 PCR amplified system. Method: Modified CTAB method,DNA extraction kit method,modified CTAB method combined with DNA extraction kit were employed to extract total DNA from Aquilariae Lignum Resinatum.Fragment of rDNA ITS2 barcode amplified with universal primers of ITS2P1/ITS2P2,PCR amplified conditions were optimized by orthogonal test,rDNA ITS2 barcode was characterized by sequencing and blasting in GenBank. Result: A260 nm/A280 nm of total DNA from Aquilariae Lignum Resinatum was 1.79 which extracted by modified CTAB method combined with DNA extraction kit and the concentration of total DNA was 350 mg · L-1,PCR amplified fragment was highest.Effects of different level factors about ITS2-PCR reaction from large to small successively was template DNA>primers>Taq DNA polymerase>dNTPs.Optimum PCR(20 μL)reaction system was template DNA of 40 ng,primers of 0.2 mmol · L-1,dNTP of 0.5 mmol · L-1 and Taq DNA polymerase of 1.5 U through the intuitive analysis.Both the length of two sample sequences were 230 bp,identities were 100% by comparing with Aquilariae Lignum Resinatum in GenBank. Conclusion: Total DNA from Aquilariae Lignum Resinatum with high quality can receive by modified CTAB method combined DNA kit method,ITS2 barcode can be quickly obtain by this optimized process,this study provides basis for molecular identification and phylogenetic analysis of Aquilariae Lignum Resinatum.

      摘要:Objective: By cluster analysis of plant morphology,medicinal properties and volatile oil production among different germplasms of Curcuma kwangsiensis to provide reference for breeding and classification of this variety. Method: Original plant identification was adopted to measure growth period data of C. kwangsiensis.After being harvested,according to 2010 edition of Chinese Pharmacopoeia,data of medicinal properties were recorded.Materials of C. kwangsiensis were adopted to extract and determine volatile oil whose weight was heavier than 150 g.Cluster analysis was employed to select fine varieties with good plant growth,high production of herb and volatile oil. Result: With plant morphology as index,samples clustered into four categories.With medicinal properties and yield as index,samples clustered into four categories.Taking volatile oil of C. kwangsiensis as an indicator,according to yield and color of volatile oil,samples could be divided into three categories. Conclusion: Integrated three classification can confirm classification for original uncertain varieties of C. kwangsiensis.System clustering method is effective for identification and classification of C. kwangsiensis,this study provides theoretical basis for germplasm genetic diversity and breeding excellent varieties of this herb.

      摘要:Objective: To optimize alkali extraction and acid precipitation process of water-soluble protein from Schisandrae Chinensis Fructus. Method: With extraction rate of protein as index,single factor tests and orthogonal design were adopted to optimize extraction process of water-soluble protein by taking solid-liquid ratio,pH of extract,extraction temperature and time as factors,isoelectric point and molecular components of water-soluble protein were analyzed.Coomassie brilliant blue Bradfrod was employed to determine protein content in extract. Result: Isoelectric point of water-soluble protein was 3.4.Optimum extraction conditions was as following:solid-liquid ratio of 1:35,pH of extract 9.5,extraction temperature at 35 ℃,extraction time of 2 h.Extraction rate of water-soluble protein was up to 43.62%.SDS-PAGE analysis showed that majority subunits of water-soluble protein were 50,40,38,21 kDa,and a small amount of 18,16 kDa. Conclusion: This optimized extraction process is stable and feasible,this study provides a reference for development of functional health food by water-soluble protein from Schisandrae Chinensis Fructus.

      摘要:Objective: To optimize ethanol extraction technology of Chaihu Xiaoying granules which could guide clinical medication of this preparation. Method: With composite score of yield of dry extract and mass percents of paeoniflorin,hesperidin,saikosaponin a and saikosaponin d as index,orthogonal design was taken for investigating effects of the concentration of ethanol,the amount of ethanol,extraction time and times on extraction technology.Contents were determined by HPLC,molile phase of saikosaponin consisted of acetonitrile(A)-0.1% phosphoric acid solution with gradient elution program of (0-10 min,10%-25%A;10-25 min,25%-35%A;25-33 min,35%-45%A;33-50 min,45%A;50-55 min,45%-10%A),detection wavelength was set at 210 nm;molile phase of paeoniflorin and hesperidin consisted of acetonitrile-0.2% acetic acid solution(15: 85),detection wavelengths were set at 230 nm and 283 nm,respectively. Result: Optimal extraction condition was to reflux extract twice with 6 times the amount of 60% ethanol for 1.5 hours per time.Yield of dry extract was 35.98%,mass percents of paeoniflorin,hesperidin,saikosaponin a and saikosaponin d were 0.944%,1.543%,0.089%,respectively. Conclusion: This optimized technology is scientific,reasonable,stable and feasible,which can provide a reference for industrial production of Chaihu Xiaoying granules.

      摘要:Objective: To investigate ultrafine grinding rules of Angelicae Dahuricae Radix pieces. Method: Fine powder and ultrafine powder Ⅰ,Ⅱ,Ⅲ,Ⅳ of Angelicae Dahuricae Radix were prepared,size distribution,wettability,infrared spectrum and moisture absorption were used to evaluate these powders.HPLC was employed to determine the content of imperatorin and evaluate dissolution with methanol-water(70: 30) as mobile phase and detection wavelength at 251 nm. Result: Particle size of ordinary powder was the maximum and ultrafine powder Ⅳ was the minimum.Wettability of ordinary powder was the worst,but it improved significantly after being ultrafine ground.Wettability of ultrafine powder Ⅳ was the best and its contact angle approached zero.Infrared spectrum and moisture absorption of these five kinds of powders were extremely similar,dissolution of ultrafine powder Ⅲ was better than others. Conclusion: Chemical compositions in four kinds of ultrafine powders do not change by comparing with ordinary powder,while their micromeritic properties are difference,it shows that ultrafine grinding of herbs should choose appropriate particle size acorrding to specific application.

      摘要:Objective: To optimize flash extraction process of puerarin from Puerariae Lobatae Radix. Method: Based on single tests,taking extracting rate of puerarin as index,orthogonal test was adopted to optimize flash extraction process with ethanol concentration,solid-liquid ratio,extraction voltage and time as factors.HPLC was employed to determine the content of puerarin with mobile phase of methanol-water(25: 75) and detection wavelength at 250 nm. Result: Optimum extraction technology was as following:extracted 2 min with 15 times the amount of 50% ethanol at 90 V.Under these conditions,extracting rate of puerarin was 37.42 mg · g-1 and its transfer rate was 88.89%. Conclusion: This optimized process is stable and reproducible with short extracting time,it can be used as process parameters for industrial production of puerarin from Puerariae Lobatae Radix.

      摘要:Objective: To investigate effects of different penetration enhancers on transdermal permeation of tetrahydropalmatine from Baijiezi San patches and provide a reference for development of this preparation. Method: Modified Franz diffusion cell was adopted,with excised male rabbits skin as model,HPLC was adopted to determine the content of tetrahydropalmatine with mobile phase of methanol-0.1% phosphoric acid(adjusting pH to 6.0 with triethylamine) (60:40) and detection wavelength at 280 nm.Accumulative permeation quantity of tetrahydropalmatine was fitted,in order to investigate effects of different penetration enhancers on transdermal permeation of Baijiezi San patches. Result: Accumulative permeation quantity of Baijiezi San patches with 2% azone as penetration enhancers was highest.When combination of penetration enhancers,transdermal permeation of 2% azone combined with 5% propanediol was better,but its effects was weaker than 2% azone alone. Conclusion: Azone can significantly improve transdermal absorption of Baijiezi San patches.

      摘要:Objective: By comparing contents of gallic acid and total flavonoids from different processed products of Acalypha australis to explore influence of processing methods on the content of active ingredients in A. australis. Method: HPLC was adopted to determine the content of gallic acid with mobile phase of acetonitrile-0.05% phosphoric acid(4: 96) and detection wavelength at 270 nm.UV was employed to determine the content of total flavonoids with rutin as index components and detection wavelength ar 515 nm.Compared content differences of these two ingredients from different processed products of A. australis. Result: There were changes after concocted on contents of gallic acid and total flavonoids in A. australis.Order of the content of gallic acid in each product was fried yellow product>wine sunburn product>vinegar sunburn product>fried coke product>raw product product>fried carbon product,the content of total flavonoids was in the order of vinegar sunburn product>wine sunburn product>fried coke product>fried yellow product>raw product>fried carbon product. Conclusion: Processing methods have a certain influence in contents of gallic acid and total flavonoids,the minimum contents of these two ingredients are both in fried carbon product.

      摘要:Objective: A method of HPLC-DAD was developed to evaluate the quality of Angelicae Dahuricae Radix through multi-target ingredient determination and finger printing analysis technologies. Method: The optimal conditions of separation and detection were achieved on a Platisil DOS C18(4.6 mm×250 mm, 5 μm) with linear gradient elution of acetonitrile-water. The detection wavelength was 254 nm. Collect 10 batches from different origins in China to assess the similarities according to similarity evaluation for ‘chromatographic fingerprint of traditional Chinese medicine’(2004A). Result: In quantitative analysis, the two components showed good regression with linear ranges, and their recoveries were 102.52% and 98.23%. Conclusion: This study indicated that the combination of quantitative and chromatographic fingerprint analysis can be readily utilized as a quality control method for Angelicae Dahuricae Radix.

      摘要:Objective: To prepare chemical constituents and establish an HPLC method for determining these constituents in Gardeniae Fructus. Method: The chromatographic separations were obtained on a Kromasil C18 column (4.6 mm×250 mm, 5 μm) with the mobile phase consisting of acetonitrile and 0.1% phosphoric acid aqueous solution (18: 82) at a flow rate of 1.0 mL · min-1. The detection wavelength was set at 327 nm, and the column temperature was 40 ℃. Result: 6"-p-coumaroyl genipin gentiobioside(1) and 3,5-di-O-caffeoyl-4-O-(3-hydroxy-3-methyl) glutaroyl quinic acid(2) were isolated from Gardeniae Fructus. These two constituents showed good linear relationship in range of 0.118 8-1.188 μg and 0.122 8-1.228 μg, respectively. The average recoveries were 99.23%(RSD 2.4%) and 100.4%(RSD 2.3%). Harvesting time has significant effect on the contents of the two constituents. Conclusion: The tests reflect the variation of the constituents in different harvesting time and the methods established are suitable for the quality improvement of Gardeniae Fructus.

      摘要:Objective: To study materiomic of release kinetics and dissolution characteristics of Liuwei Dihuang pills and correlation of them. Method: Choosing Liuwei Dihuang pills from three manufacturers and different batches produced by manufacturer A. According to Chinese Pharmacopia, using the evaluation methods of release kinetics of traditional Chinese Medicine to determining the materiomics release kinetics and the release characteristic of Liuwei Dihuang pills by the paddle method, determining the dissolution time, and analyzing the correlation between the characteristic parameter of drug release and dissolution of Liuwei Dihuang pills. Result: T50 of Liuwei Dihuang pills of the three manufacturers are 107.01,134.33,and 13.01 min, respectively while Td are 234.11,385.42,and 206.17 min. The materiomic release kinetics had significant difference among three different manufacturers (P<0.01);T50 of Liuwei Dihuang pills of three different batches produced by manufacturer A are 107.01, 125.93, 194.44 min and Td of them are 234.11, 385.42, 206.17 min. The materiomic release kinetics had significant difference among different batches produced by manufacturer A (P<0.01). Their disintegration time are 32, 43, 37, 35, 21 min, there was high lever of linear correlation between the release curve fitting parameter as T50, Td and disintegration time trials result (P<0.05). Conclusion: There were significant difference among Liuwei Dihuang pills of different manufacturers or different batches produced by manufacturer A under the evaluation methods of materiomic release kinetics. And the release rate has correlation with the disintegration time.

      摘要:Objective: To study the anti-tumor triterpenes from the pericarps of Juglans regia. Method: The chemical constituents isolated and purified by silica gel, Sephadex LH-20 columns and recrystallization. The structures of isolated compounds were elucidated on the basis of NMR spectral data and physicochemical properties. Result: Ten compounds were separated and identified as 20(S)-protopanaxadiol(1), 20(S)-protopanaxadiol-3-one(2), 20(S)-hydroxyldammar-24-en-3-one(3), 20(S),24(R)-dihydroxydammar-25-en-3-one(4), 1β,12β,20(S)-trihydroxydammar-24-en-3-one(5), 2α-hydroxyloleanolic acid(6), 2α,23-dihydroxyoleanolic acid(7), 2α-hydroxyursolic acid(8), 23-hydroxyursolic acid(9), 3-epikatonic acid(10). Conclusion: Compounds 1, 3, and 5-10 were isolated from J.regia for the first time.

      摘要:Objective: To study acute toxicity, anti-inflammatory and anti-nociceptive effects of 70% ethanol extract (E70%EtOH) and water extract (Ewater) of Oxalis corniculata, HPLC was used to analyze figure information and determine the best extrat. Method: The maximum dosage test was used to evaluate safety of E70%EtOH and Ewater. Permeability increasion of celiac capillary induced by glacial acetic acid in mice was performed to investigate the anti-inflammatory effect, writhing test induced by glacial acetic acid in mice was used to observe the anti-nociceptive activity. Analyze different extracts by HPLC. Result: E70%EtOHand Ewater didn't show toxicity in the range of 35 to 1 120 mg · kg-1, MTD>1 120 mg · kg-1. Compare with Ewater, E70%EtOH could remarkable inhibit the permeability increasion of celiac capillary and the writhing reaction in mice, HPLC peak information is richer and the common peak ratio is higher than that of Ewater. Conclusion: Lethal dose of E70%EtOH and Ewater may be very high and toxicity of E70%EtOH and Ewater may be very low. The effects of E70%EtOH is better than that of Ewater, E70%EtOH can more fully the enrichment of active component.

      摘要:Objective: To investigate the dissolution of L-epicatechin from the ultra-micro and traditional powder of Xanthoceras sorbifolia. Method: HPLC technique was used to detect the content of L-epicatechin. Chromatographic conditions were as follows:Agilent Eclipse-C18 column (4.6 mm×250 mm, 5 μm), detection wavelength 280 nm, mobile phase of acetonitrile-0.04% H3PO4 (15:85), flow rate 1.0 mL·min-1, injection volume 20 μL. Dissolution amount and the dissolution rate of L-epicatechin from the ultra-micro and traditional powder of X. sorbifolia were determined, and exoteric dissolution behavior was analyzed. Result: Dissolution amount of L-epicatechin from ultra-micro and traditional powder were 5.868 mg·g-1 and 8.440 mg·g-1, respectively. Exoteric dissolution curves were in line with Weibull distribution. T50, Td, T70, T80, T90 of traditional powder were 0.476, 1.952, 2.939, 5.488, 9.961 min, respectively, but these of ultra-micro powder were 0.180, 0.348, 0.469, 0.768, 1.249 min. The dissolution rate of ultra-micro powder of X. sorbifolia was increased obviously (P<0.01). Per unit of time (1 h) dissolution amount increased by about 44% compared with traditional powder. Conclusion: Superfine grinding can promote the dissolution of L-epicatechin in X. sorbifolia, and application of superfine grinding is feasible and necessary.

      摘要:Objective: To investigate the antioxidant activity of Arenaria kansuensis extracts in vitro. Method: The air-dried and powderded herb of A. kansuensis was extracted over 2 hours with 10 times of 95% alcohol refluxing for 2 times and the combined alcohol extracts were concentrated under vacuum drying. The resultant residue was partitioned in petroleum ether, dichloromethane, ethyl acetate and n-butanol orderly. The four extracts were configured to the appropriate solution. The antioxidant activity of four extracts were analyzed by 1,1-diphenyl-2-pieryl-2-picrylhydrazyl (DPPH) radical scavenging, 2,2-amino-di(3-ethyl-benzothiazoline sulphonic acid-6) ammonium salt (ABTS) radical scavenging, and ferric reducing antioxidant power (FRAP) assay. Result: The ethyl acetate and n-butanol extract of A. kansuensis showed stronger scavenging activity against DPPH ·, ABTS+ · and reducing antioxidant power of 2,4,6-tris (2-pyridyl) -s-triazine(TPTZ) than other extracts, but showed lower capacity than positive control of vitamin C. Conclusion: The scavenging effects showed a dose-response relationship at the selected concentration with the extracts of A. kansuensis. The ethyl acetate and n-butanol extract have the strongest free radical scavenging activity.

      摘要:Objective: To screen the polypenols with high content and strong antioxidant capacity, and to encourage the development of Fagaceae resources. Method: The total content of polypenols from 8 kinds of Fagaceae plants were determined by Folin-phenol method, and their antioxidant capacities were evaluated by four antioxidant methods, including 1,1-diphenyl-2-pieryl-2-picrylhydrazyl(DPPH) scavenging, 2,2-amino-di(3-ethyl-benzothiazoline sulphonic acid-6) ammonium salt (ABTS) radical scavenging, Fe2+ chelating, and Fe3+ reducing assays. The relationship between the total polypenols content and antioxidant capacity was analyzed. Result: The Castanopsis hystrix had the highest total polypenols content (10.61%), and followed by C. fissa (10.09%). Comprehensive evaluation of antioxidant capacity test results showed that all plants had good antioxidant capacity, and the C. hystrix had the strongest antioxidant capacity. A dose-effect relationship between the antioxidant capacity and the total polypenols content of C. hystrix was found. Conclusion: C. hystrix and C. fissa were the most potential plant resources for the health and skin care products with antioxidant activity.

      摘要:Objective: To study the chemical constituents from Paeonia anomala and to evaluate their antioxidant activities. Method: The compounds were isolated by chromatography on silicagel, Sephadex LH-20 column and identified on the basis of physicochemical constants and spectral analysis. Their antioxidant activities were evaluated by 1,1-diphenyl-2-pieryl-2-picrylhydrazyl(DPPH) and 2,2-amino-di(3-ethyl-benzothiazoline sulphonic acid-6) ammonium salt (ABTS) radical scavenging methods. Result: Five compounds were isolated and their structures were identified as ethyl gallate (1), methyl gallate (2), oleanic acid (3), syringate (4) and β-sitosterol(5). The results showed that the DPPH radical scavenging activity of compounds 1 and 2 were lower than the positive control of Vitamin C with IC50values (35.13±0.28), (44.64±0.37) μmol · L-1. Their ABTS radical scavenging activity were lower than Vitamin C with IC50values of (25.13±0.68), (35.76±1.48) μmol · L-1, respectively. Conclusion: Compounds 3, 4 are isolated from this plant for the first time. Compounds 1, 2 have powerful antioxidant activies, and the others have little antioxidant activies.

      摘要:Objective: In Xintongning spray quality standard, it had the determination of requirements on the ferulic acid and cinnamic acid content, but for two sets of different methods.The method established a HPLC method for the determination of ferulic acid, cinnamic acid in Xintongning spray. Method: The method used high performance liquid chromatography separation and the effective components of the calculation of its content.The analysis was performed on a Phenomenex Luna C18 column(4.6 mm×150 mm,5 μm), mobile phase was acetonitrile -0.05% phosphoric acid for gradient elution,the detection wavelength was 290 mm,the flow rate was 1.0 mL · min-1 and column temperature at 30 ℃. Selected the 1 batch of Xintongning spray samples for method validation test, and the 4 batch of Xintongning spray samples according to the method of content determination. Result: Blank interference test, precision test, stability test, repeatability test, linear relation test, recovery test method validation tests were in line with the relevant requirements, RSD was less than 3%.The linearities were in the ranges of 0.025 16-0.276 76 mg · L-1 for fetulic acid(r =1), 0.010 4-0.114 4 mg · L-1 for cinnamic acid(r =0.999 5),respectively. The two components were able to achieve baseline separation, separation degree greater than 2.The average recoveries were 100.35%, 101.93% for ferulic acid, cinnamic acid. Conclusion: The method is simple, accurate, reproducible, can be used for quality control of this preparation,and can be saved testing time and the testing cost objective. The method for the quality standard of Chinese traditional patent medicine multi group method for the determination of content of the establishment has reference significance.

      摘要:Objective: To develop a method for determining liquiriti,naringin,hesperidin,neohesperidin and glycyrrhizic acid ammonium salt in Jianpi Shuwei gel. Method: The mobile phase consisted of acetonitrile-0.05% phosphoric acid with gradient elution at the flow rate of 1.0 mL · min-1.The detection wavelength was 237 nm(liquiriti,glycyrrhizic acid ammonium salt),283 nm(naringin,hesperidin,neohesperidin). Result: The liquiriti,naringin,hesperidin,neohesperidin and glycyrrhizic acid ammonium salt all had good linear relationship in the ranges of 0.128 3-0.641 6,1.105 2-5.526,0.225 2-1.126 0,1.092-5.460,0.314 9-1.574 4 μg.The average recoveries were 101.47%,99.22%,100.02%,101.64% and 98.11%(RSD<3%),respectively. Conclusion: The method is simple,accurate,reproducible and nice to the separation,can be used for the quality evaluation of Jianpi Shuwei gel.

      摘要:Objective: To investigate the interaction and changing trend of chemical composition in eighteen incompatible medicaments, Genkwa Flos and Glycyrrhizae Radix, with different compatibility using HPLC fingerprints. Method: The fingerprints of Genkwa Flos and Glycyrhizae Radix with different compatibility proportion were analyzed by HPLC. Characteristic peaks were identified preliminarily based on the MS spectra and literature data. Result: The dissolution of liquiritin apioside, apigenin-5-O-β-D-glucoside, isoliquiritin apioside, hydroxy-genkwanin and glycyrrhizic acid were reduced with increase the proportion of Glycyrhizae Radix. The dissolution of apigenin-7-O-β-D-glucuronide was reached the maximum value at 1:1 compatibility proportion. Conclusion: The dissolution of Genkwa Flos and Glycyrrhizae Radix reduced, thus may reduce its pharmacological action, provide the mentality and the method of evaluation of traditional Chinese medicine incompatibility from chemical angle.

      摘要:Objective: To establish an HPLC method for determination of gallic acid, loganin, puerarin, ferulic acid, emodin and physcion in Gengnianshu pills. Method: HPLC was applied on an Agilent C18 column (Zorbax SB-AQ 4.6mm×250 mm, 5 μm) with acetonitrile-0.2% formic acid water as mobile phase by gradient elution. The flow rate was 1.0 mL·min-1. Detection wavelengthes were set at 238 nm (loganin), 270 nm (gallic acid, puerarin, emodin and physcion), and 320 nm (ferulic acid). Result: The methodology of 6 components met the requirements, the blank test showed no interference, the average recovery rates were 98.2%-103.2%, and RSD was 1.4%-1.9%. Conclusion: This method is easy, accurate with high specificity. This study provides scientific basis for the quality evaluation of Gennianshu pills.

      摘要:Objective: To study on the HPLC fingerprint of Scutellaria indica, establishing a method for the content determination of scutellarin, wogonside, apigenin and wogonin, simultaneously. Method: The separation was carried out on an Agilent Eclipse XDB-C18 column (4.6 mm × 250 mm, 5 μm) with the mixture of 0.6% acetic acid and acetonitrile and tetrahydrofuran as mobile phase in a gradient elution model (flow rate 1.0 mL · min-1, detection wavelength were 365 nm from 0 to 40 minute and 275 nm from 40 to 65 minute, column temperature 30 ℃, sample size 10 μL). 16 batches of S. indica were analyzed with the developed HPLC fingerprint and determination methods, the data calculation was performed with similarity evaluation system for chromatographic fingerprint of TCM (Version 2004A). Result: The linear range of scutellarin, wogonside, apigenin and wogonin were 5.125-82.00(r=0.999 9), 7.375-118.0 (r=0.999 9), 1.550-24.80(r=0.999 9), 7.188-115.0(r=0.999 9) mg · L-1, respectively. The average recovery rates of scutellarin, wogonside, apigenin and wogonin were 103.37%(RSD 1.8%), 100.63%(RSD 1.3%), 104.03%(RSD 0.8%), 98.39%(RSD 2.5%), respectively. In the fingerprint, 28 common peaks of S. indica were marked and 4 peaks were determined. Conclusion: This method provides a scientific basis for controlling the quality of S. indica.

      摘要:Objective: To simultaneous determine the contents of rhein, emodin and chrysophanol in Wenshen Jiangzhuo powder by HPLC. Method: An external standard method by HPLC with ANGLA Promosil C18 column (4.6 mm×250 mm, 5 μm) was used with the column temperature was 30 ℃;the detection wavelength was 224 nm, and the injection volume was 10 μL. The flow rate was set at 1.0 mL · min-1 and gradient elution with acetonitrile and H2O (included 0.1% phosphoric acid). Result: Rhein, emodin and chrysophanol were well separated by this method. Linearities of rhein, emodin and chrysophanol were good in the ranges of 0.757 2-3.786 0 μg(r=0.999 4), 0.347 6-1.738 0 μg(r=0.999 3), and 1.731-8.656 μg(r=0.999 2), respectively. The average recoveries of rhein, emodin and chrysophanol were 101.7%, 102.6% and 101.1%, RSD were 3.0%,2.3%,1.9%. Conclusion: The method is simple, accurate and reproducible, and can be used for quality control of Wenshen Jiangzhuo powder.

      摘要:Objective: To investigate the chemical constituents in the volatile oil from the leaves of Callicarpa nudiflora in Baisha, Hainan. Method: The essential oil was extracted by water steam distillation and its chemical constituents were separated, determined and identified by gas chromatography-mass spectrometry combined with computer retrieval in compound databases. The relative percentage content of each compound was calculated by peak area normalization method. Result: The 76 compounds were separated and 70 of them were identified, which covered 98.69% of the total oil. The major constituents with higher relative content were β-pinene (20.70%), α-pinene (9.41%), caryophyllene oxide (6.90%), caryophyllene (6.65%), o-cymene (6.62%), trans-4-thujanol (5.85%) and myrtenol (5.61%). Conclusion: The major chemical constituents in the volatile oil from the leaves of C. nudiflora in Baisha, Hainan are monoterpenes and their oxygenated derivatives.

      摘要:Objective: To established an UPLC-MS method for quantitative determination of six active ingredients in Shenqi Yiqi Guben tablet. Method: The UPLC separation was carried out with an Acquility UPLC BEH C18(2.1 mm×100 mm, 1.7 μm) using gradient elution with a mobile phase consisting of acetonitrile-water (included 0.1% formic acid) at a constant flow rate of 0.5 mL · min-1. The MS detector set as follows:negative ion mode detection, source spray voltage (IS)-5 000 V, auxiliary heating gas temperature 450 ℃, atomization gas 50 psi, auxiliary gas 40 psi, curtain gas 30 psi, deaggregation voltage -55 V, collection methods TOF-MS/MS. Result: Six components:paeoniflorin, ginsenoside Re, ginsenoside Rg1, forsythin, ginsenosides Rb1, and astragaloside were in a good linearity, r greater than 0.999, recoveries were 98%-102%, RSD were less than 4.5%. Conclusion: The established method for the determination of the six components of Shenqi Yiqi Guben tablet is accuracy and precision that can be used for quality control of the preparation. The UPLC-MS method can increased resolution, lower production costs of drugs compared with HPLC-MS. It provides a reference to set standards for other Chinese medicine formulations containing these ingredients.

      摘要:Objective: To investigate influence of different dose of Chuanxiong Rhizoma(LC) on pharmacokinetics of gastrodin(GAS) and gastrodigenin(HBA) from Gastrodiae Rhizoma(GE) in migraine model rats. Method: Rats were randomly divided into LC-GE(1: 3) group(doses of LC and GE extracts were 0.163,0.484 g · kg-1),LC-GE(2:3) group(doses of LC and GE extracts were 0.326,0.484 g · kg-1) and LC-GE(4:3) group(doses of LC and GE extracts were 0.652,0.484 g · kg-1),plasma and brain tissue was collected at different times after drug administration.Concentrations of GAS and HBA were determined by HPLC,a mixture of acetonitrile-0.01% phosphoric acid(2: 98) was employed as mobile phase for determination of GAS and HBA in rats plasma with temperature of column oven was maintained at 35 ℃.While mobile phase of acetonitrile-water(3: 97) was used for quantification of HBA in rats brain,column temperature was heated at 30 ℃.DAS 2.0 software was used to calculate pharmacokinetic parameters. Result: Area under curve[AUC(0-t)] of GAS in rats plasma from these three groups were (1 984.41±89.91),(1 708.74±108.96),(1 862.76±82.11) mg · L-1 · min,their mean residence time[MRT(0-t)] were (63.62±2.04),(61.69±2.40),(75.04±4.29) min,their clearance(CL) were (0.034 3±0.002 4),(0.039 9±0.002 3),(0.031 6±0.003 2) L · min-1 · kg-1,respectively.AUC(0-t) of HBA in rats brain from these three groups were (72 891.70±12 075.24),(64 201.77±5 921.88),(97 046.82±11 002.49) μg · L-1 · min,their MRT(0-t) were (82.15±15.34),(68.33±11.22),(96.68±8.87) min,their CL were (0.395 5±0.072 8),(0.450 7±0.049 2),(0.301 2±0.036 7) L · min-1 · kg-1,respectively. Conclusion: These findings demonstrate LC-GE(4: 3) group(equivalent to crude herbs ratio of 4: 1) can enhance bioavailability,prolong MRT and reduce elimination speed of GAS in plasma as well as HBA in brain by comparing with other compatibility groups,which is consistent with usual dose in ancient prescriptions.This study demonstrates compatibility connotation at a right dose-ratio of LC and GE through angle of pharmacokinetics.

      摘要:Objective: To investigate therapeutic effects of Nostoc commune powder on blood glucose of type 2 diabetic rats. Method: Type 2 diabetic rats model was established by intraperitoneal injection of streptozotocin(STZ,35 mg · kg-1) on SD rats after rats were fed with high sugar and high fat for 4 weeks.These rats were divided into 6 models,included low dose group(N. commune powder 25 mg · kg-1),middle dose group(N. commune powder 50 mg · kg-1),high dose group(N. commune powder 100 mg · kg-1),positive group(metformin hydrochloride 200 mg · kg-1),model group(physiological saline 200 mg · kg-1) and normal group.After 6 weeks of administration,indexes of body weight,fasting blood glucose,total quality of micturition and defecation and water intake were measured. Result: After 6 weeks of administration,compared with the model group,body weight of diabetic rats were significantly increased in low,middle and high dose group,blood glucose and total quality of micturition and defecation,water intake levels of three dose groups of diabetic rats were significantly decreased;these indexes had significant differences among these three dose groups.Effects between positive control groups and three N. commune powder groups were statistically nonsignificant on these four indexes. Conclusion: N. commune powder can reduce blood glucose levels of experimental rats with type 2 diabetes mellitus by intraperitoneal injection of STZ,and effect of hypoglycemic is positively correlated by N. commune powder concentrations.

      摘要:Objective: To observe the effect of Chaihu Shugan decoction (CHSGD) on the content of serotonin(5-HT), interleukin-1beta(IL-1β),interleukin-6(IL-6)mRNA expression of hippocampus in chronic temporal lobe epilepsy and depression model rats. Method: The intraperitoneal injection of lithium chloride-pilocarpine was used to make epileptic-depression model,the modeling rats were randomly divided into model group, citalopram group, CHSGD-high,medium,low dose group and normal group. The drug intervention group was given citalopram 10 mg · kg-1 · d-1, CHSGD-H,M,L at dose of 10,5,2.5 g · kg-1 · d-1 orally, normal group and model group were given equal salineby continuous lavage for 4 weeks, twice each day. By forced swim test (FST) and the changes of sucrose consumption the depressive behavior was assessed, video was usec to monitor seizures, hippocampal 5-HT number was determined by liquid chromatography mass spectrometry. Hippocampal IL-1β, IL-6 mRNA expression was detected by real-time fluorescence quantitative PCR detection. Result: Compared with normal group, model group rats FST total fixed time extended obviously, the sugar water consumption decreased significantly, IL-1β mRNA expression quantity increased obviously (P<0.01);5-HT content reduced, IL-6 mRNA expression increased (P<0.05). Compared with model group, drug intervention after 4 weeks, citalopram, CHSGT-H,M dose group of seizures in rats reduced significantly, FST total fixed time shortened significantly, sugar water consumption increased significantly, the hippocampus IL-1β, IL-6 mRNA expression lowered significantly (P<0.01), the content of 5-HT increase (P<0.05). Citalopram group and CHSGD-H,M dose group of comparison, there was no statistical significance. Conclusion: High, medium dose of Chaihu Shugan decoction can inhibit epilepsy comorbidity of depression in hippocampus of modle rats IL-1β, IL-6 mRNA expression, promote the expression of 5-HT, reduce the number of seizures and improve depressive behavior in rats.

      摘要:Objective: To observe the effect of 12-deoxyphorbol-13-palmitate (DP) which isolated from the roots of Euphorbia fischeriana on the vascular endothelial growth factor (VEGF) expression, and whether it shows anti-VEGF effects via Von Hippel-Lindau(VHL)/hypoxia induced 1 alphal(HIF-1α) signaling pathway. Method: The cells were divided into normoxia control group, normoxia experimental groups (10, 20, 40 μmol · L-1 DP), hypoxia control group, hypoxia experimental groups (10, 20, 40 μmol · L-1 DP). The hypoxia control group and experimental groups were pretreated with CoCl2(150 μmol · L-1). The viability of MCF-7 cells was measured by MTT method at a cell density of 70%-80% and at 6, 12, 24 h. The secretion of VEGF was investigated by enzyme-linked irnmuno sorbent assay(ELISA).The mRNA level of VEGF was detected by RT-qPCR. The protein expressions of VEGF, HIF-1α and VHL were determined by Western blot. The localization and expression of HIF-1α were detected by immunofluorescence. Result: MTT assay showed that DP significantly decreased the cell viability at 12, 24 h (P<0.05) and at DP concentration of 20 μmol · L-1 and 40 μmol · L-1. Under hypoxia condition, the expression of VEGF and HIF-1α was increased compared with that under normoxia condition. But the up-regulation was reversed (P<0.05) by the addition of DP. The VHL level was down-regulated under hypoxia condition. However after treatment of DP, the expression of VHL was increasing significantly (P<0.05). Conclusion: DP could down-regulate VEGF by affecting VHL/HIF-1α signaling pathway.

      摘要:Objective: To investigate the protective effect of snake venom tripeptide pENW on the injury of human umbilical vein endothelial cells (HUVECs) induced by lipopolysaccharide (LPS) in vitro and to explore its underlying mechanism. Method: HUVECs were treated with pENW (10-4, 10-5, 10-6 mol · L-1) and nitric oxide synthase(NOS) inhibitor L-NMMA (5×10-4 mol · L-1), then induced with LPS (1 mg · L-1) for 24 h. The cell viability was evaluated by MTT assay. Nitric oxide (NO) content was measured by Griess Reagent. The nitric-oxide synthase (NOS) activity was assessed by commercially available kits. The NOS expression was measured by Western blot. Result: Compared with normal control group, vascular endothelial cells were obviously damaged and NO release increased by LPS, which could be protected by pENW in a concentration-dependent manner (P<0.05, P<0.01). L-NMMA and pENW could increase the cell viability rates and decrease NO production (P<0.05, P<0.01). Meanwhile, iNOS activity and protein expression were inhibited by pENW(P<0.05, P<0.01). However, there was no significant effect on eNOS activity and protein expression. Conclusion: The results showe that pENW has protective effect on HUVECs induced by LPS. Its mechanism may be related to the suppression of iNOS expression along with decreasing levels of NO and iNOS activity.

      摘要:Objective: To observe the effect of Zhali Nusi recipe (ZLNS) on regulating blood-brain barrier (BBB) permeability and promoting the grafting of bone marrow mesenchymal stem cells (BMSCs) from vascular to brain tissue after the transplantation. Method: SD rats were randomly divided into sham-operated group, model group, ZLNS, BMSCs transplantation group and ZLNS combined with BMSCs group. Middle cerebral artery occlusion (MCAO) model was duplicated with nylon thread. BMSCs were cultured and amplified by the whole bone marrow adherence method. The rats were intragastrically administrated with ZLNS(14.6 g · kg-1 ·d-1), BMSCs suspension solution were transplanted into brain through carotid artery (2×106/200 μL). Brain of rats was taken out at 1, 3, 7, 14 days after transplantation. Brain water content (BWC), brain infarct area ratio (BIAR) and Evans Blue (EB) leakage were detected by dry-weight assay, TTC and fluorescence spectrophotometry, respectively. Result: The BWC, BIAR and EB in model group increased obviously than the sham-operated group (P<0.01). Compared with the model group, the BWC at 3, 7, 14 days decreased obviously, the EB leakage decreased at 3, 7 days in ZLNS, transplantation and combination groups (P<0.01), the BIAR decreased in all groups (P<0.01, P<0.05). Compared with the transplantation group, the BWC at 1 day and EB at 1, 7, 14 days in ZLNS groups decreased (P<0.01, P<0.05), all above indicators in combination groups decreased and the results were obvious at 1, 14 days (P<0.01). Compared with the ZLNS groups, all above indicators decreased in combination group and the results were obvious at 7, 14 days (P<0.01, P<0.05). All changes were remarkably serious at 7 days and obviously improved at 14 days being increased firstly and then decreased after cerebral ischemia in each groups (P<0.01). Conclusion: The injury at 7 days after cerebral ischemia reperfusion was serious than that at 1, 3 days, and there was some improvement at 14 days in rats after cerebral ischemia reperfusion. The ZLNS could regulate the permeability of BBB, promote the grafting of BMSCs from vascular to brain tissue after the transplantation. It would play a better role in brain protection by the combination ZLNS and BMSCs transplantation.

      摘要:Objective: To investigate the effects and potential mechanisms of Xihuang Wan on cell migration and invasion in SW480 human colorectal cancer cells. Method: SW480 human colorectal cancer cells were used as the cell model. Wound healing and transwell assay were used to determine the inhibitory effects of serum of rats containing Xihuang Wan on cell migration and invasion in SW480 cells, respectively. Western blot was used to measure the protein expression levels of ERK and phospho-ERK, and the qPCR assay was used to evaluate the mRNA expression levels of E-cadherin, PARP-1, Vimentin and Snail in SW480 cells after serum containing Xihuang Wan treatment. Result: The abilities of cells migration and invasion showed dose-dependent reducing in SW480 cells after treatments with the serums containing Xihuang Wan. The qPCR results showed that the serum containing Xihuang Wan enhanced E-cadherin, while reduced PARP-1, Vimentin and Snail mRNA expression levels. In addition, Western blot results showed the decreasing phosphorylation of ERK after treatments with serum containing Xihuang pill. Conclusion: The serums containing Xihuang Wan inhibited cells migration and invasion, affected the mRNA expression levels of epithelial-mesenchymal transiton (EMT) related markers including E-cadherin, PARP-1, Vimentin and Snail in SW480 cells. The mechanism may be related to the ERK/MAPK pathway. Our studies provide further pharmacological basis for developing Xihuang Wan as a preventive/chemotherapeutic agent of metastasis in colorectal cancer.

      摘要:Objective: To investigate the protective effect and mechanism of buckwheat flavones on diabetic nephropathy rats. Method: The model of diabetic nephropathy rats was made by intraperitoneally injecting alloxan once per week for 3 times. The rats were randomly divided into the model control group, the buckwheat flavones groups (50, 100, 200 mg · kg-1), the irbesartan group (25 mg · kg-1) and the normal group. The rats were intragastrically administrated of the corresponding medicines once daily for 12 weeks. The fasting blood glucose (FBG) and 24-h urine protein were determined every 3 weeks. The levels of serum creatinine (SCr), blood urea nitrogen (BUN), triglyceride (TG), total cholesterol (TC) in serum were tested by auto analyzer, and the levels of fructosamine (FTS), advanced glycation end products (AGEs), superoxide dismutase (SOD), and methane dicarboxylic aldehyde (MDA) in kidney tissues were tested after the treatment. Result: Compared with the normal group, FBG and 24-h urine protein increased continuously, the levels of SCr, BUN, TG, TC in serum and FTS, AGEs, MDA in kidney tissues decreased, the SOD level increased (P<0.05,P<0.01), and kidney tissues were injured remarkably in model control group. Compared with the model control group, the buckwheat flavones could improve the above indexes (P<0.05, P<0.01). Conclusion: The buckwheat flavones have protective effects on diabetic nephropathy rats, and its mechanisms are in part associated with reducing blood glucose, blood lipid, and decreasing non-enzymatic glycosylation and oxidative stress.

      摘要:Objective: To discuss the protective effect of paeonol on ventricular remodeling in acute myocardial infarction (AMI) rats and to investigate its influence on Smad2, Smad3, Smad7 mRNA expression of TGF-β/Smads signaling pathway. Method: In vivo model of AMI was made using the left anterior descending coronary branch ligation. The rats were randomly divided into the model group, the sham group, the low-, medium-and high-dose paeonol groups (8, 12, 16 mg · kg-1), and the captopril group (10 mg · kg-1) based on ECG evaluation. The heart weight index and infarct size were measure after 4 weeks. The mRNA expression levels of Smad2, Smad3, Smad7 in myocardial tissues were detected using real-time quantitative (RT-qPCR) technique. Result: Compared with the model group, the myocardial infarct size was significantly reduced (P<0.05), the heart weight index decreased (P<0.05) in all dose paeonol and captopril groups. Compared with the sham group, Smad2 and Smad3 mRNA expression levels were significantly higher, Smad7 expression level was lower in the model group (P<0.01). While, Smad2 and Smad3 mRNA expression levels decreased significantly, Smad7 expression level increased significantly in all dose paeonol groups (P<0.01). Conclusion: Paeonol has anti-myocardial fibrosis effect in AMI rats. Its mechanism may be related to increasing the Smad7 mRNA expression of TGF-β1 inhibitory signaling protein, and down-regulating the Smad2 and Smad7 mRNA expressions of TGF-β receptor signaling protein.

      摘要:Objective: To determine the protective effects of Maikang tablets on blood vessel in the experimental hyperlipidemic rats. Method: The rats were randomly divided into 5 groups according to their baseline total cholesterol (TC) level, including the control group, the hyperlipidemia model group, the low-middle-and high-dose Maikang tablets groups (0.47, 0.94, 1.88 g · kg-1). The rats in the control group were fed with basal diet, the rats in the hyperlipidemia model and Maikang tablets groups were fed with high-fat diet, and the rats in Maikang tablets groups added Maikang tablets. All rats were intragastrically administrated of the corresponding medicines for 30 days. The level of blood lipid and the contents of malonaldehyde (MDA), nitric oxide (NO), endothelin (ET) -1, calcitonin generelated peptide (CGRP), prostacyclin (PGI2), endothelium-derived hyperpolarizing factor (EDHF) were determined and the activity of nitric oxide synthase (NOS), superoxide dismutase (SOD) of the rats were tested. Result: Compared with the normal group, the body weight of rats in the model group had little change. The contents of hyperlipidemia TC, triglyceride(TG), low density lipoprotein cholesterol (LDL-C), MDA, ET-1,CGRP, PGI2, EDHF, NO increased obviously (P<0.05, P<0.01), the activity of NOS increased significantly (P<0.01), the content of high density lipoprotein cholesterol (HDL-C) and the activity of SOD decreased significantly (P<0.01) in the hyperlipidemia modle group. Maikang tablets also had no effect on body weight, while it could improve the above indicators significantly (P<0.05, P<0.01). Conclusion: Maikang tablets have good effects on blood vessel protection and vascular repair function in rats with hyperlipidemia.

      摘要:Objective: To prepare the dragon's blood gels and to investigate its effect of wound tissue repair in mice. Method: The orthogonal experiment was designed with carbomer concentration, propylene glycol volume and pH value as factors, the appearance, irritation, thermostability, chill stability and centrifug stabiity as evaluation index to optimize the best formula of dragon's blood gels. Thirty mice were randomly divided into 3 groups, the model group (gels base), the observation group (dragon's blood gels) and the positive drug group (clindamycin lidocaine gels). The trauma was made by cuting a size of the diameter of 1.2 cm wound in the back of the mice. The wound changes at different time points were observed and the histopathological score of wound biopsy was evaluated. Result: The preparation of dragon's blood gel was optimized as follows:0.5% Carbomer-940, 2.5 mL Propylene glycol and adjusting pH to 8.0.The product displayed a predominant spread property with uniform appearance and color. No changes of product were observed after the thermostability, chill stability and eentrifug stabiity tests. The mice tests showed that the wound surface was more smooth and neat, and the heal time was shorter in the dragon's blood gels and positive drug groups than those in the model group with significiant difference (P<0.05). Conclusion: The preparation technology of dragon's blood gels is simple and the gel has good stability. Morever, the dragon's blood gels could promote the tissue repair in mice.

      摘要:Objective: To observe the effect of Lantana camara on ulcerative colitis rats. Method: Forty\|eight rats were randomly divided into 6 groups:normal group,model group,salazosulfapyridine(SASP,0.3 g · kg-1)group,L. camara low,middle and high dose groups(12,6,3 g · kg-1). Ulcerative colitis model was copied by 2, 4, 6-trinitrobenzenesulfonic acid(TNBS). After 10 days' treatment,the changes of colonic mucosal damage index (CMDI), histopathological score (HS), serum levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-4 (IL-4) and interleukin-10 (IL-10) were observed. Result: Compared with normal group,the CMDI and HS of model group were significantly increased(P<0.01);compared with model group,the CMDI and HS of middle-and high-dose L.camara groups and SASP group decreased significantly(P<0.05 or P<0.01);there was no significant difference in CMDI and HS among L.camara groups and SASP group. Compared to normal group,the serum TNF-α and IL-1β levels of model group increased significantly(P<0.01),while the IL-4 and IL-10 levels decreased significantly(P<0.01).The serum TNF-α and IL-1β levels level of treatment groups decreased significantly(P<0.01). Compared with model group,while the IL-4 and IL-10 levels increased significantly(P<0.01).Compared with SASP group,the serum TNF-α level of low-dose L.camara group increased significantly (P<0.01);the serum IL-1β level of high and middle dose L.camara groups decreased significantly(P<0.01),while the serum IL-10 level of these two groups increased significantly (P<0.05);the IL-4 level of low-dose L. camara group decreased significantly(P<0.05). Conclusion: L.camara treated UC rats effectively, probably by regulating the balance of pro-and anti-inflammatory cytokines.

      摘要:Objective: To observe the effects of Xuefu Zhuyu Tang on cardiac function, cardiac muscle and blood vessel in rabbits with coronary heart disease at stage of blood stasis syndrome. Method: Thirty-six healthy rabbits were randomly divided into normal group, sham-operation group,model group, and Xuefu Zhuyu Tang low-dose,middle-dose,high-dose groups six groups with six rabbits in each group. Left coronary artery implanted constrictor ring to establish the model of coronary heart disease at the stage of blood stasis syndrome. The sham-operation group was just opened the chest but not implanted the constrictor ring. On the basis of the model group from the first day of the fourth week after operation the low-dose,middle-dose,high-dose groups were respectively intragastric administrated(ig) with Xuefu Zhuyu Tang according to 10,20,40 g · kg-1,once a day for continuous five weeks while the control group, the model group and the sham-operation group were ig the same volume normal saline. Then to detect the cardiac function,heart index,the levels of vasoactive factors and observe pathological change pictures of cardiac muscle and blood vessel. Result: Compared with the normal group,in the model group cardiac function declined, heart index increased, the levels of angiotensis Ⅱ(AngⅡ),tumor necrosis factor-α(TNF-α),reactive oxygen species(ROS) while the level of nithic oxide(NO) reduced(P<0.05, P<0.01),meantime, in the model group there were pathological changes of cardiac muscle and blood vessel. Compared with the model group, all of the low-dose,middle-dose,high-dose groups could improve cardiac function,inhibit the pathological changes of cardiac muscle and blood vessel and restore the normal levels of vasoactive factors, which had a statistical significance (P<0.05, P<0.01). Conclusion: It was included that Xuefu Zhuyu Tang improved the pathological changes of cardiac muscle and blood vessel and restored the cardiac function by alleviating the oxidative injury of coronary artery.

      摘要:Objective: To investigate the effect of beta-asarone on the expression of circadian gene clock in depression model rats. Method: Male SD rats were randomly divided into five groups:normal group, model group, agomelatine (melatonin agonist group drug), beta-asarone low-dose group, high-dose group with 10 rats in each group,chronic unpredictable stimulation (CMUS) was given for 28 days to copy the rat model of depression. From the beginning of the eighth day model group, agomelatine group, beta-asarone low-dose groups, beta-asarone high-dose group were given normal saline, agomelatin(40 mg · kg-1 · d-1), beta-asarone(12.5,25 mg · kg-1 ·d-1), 5 mL · kg-1 gavage. The body weight,horizontal movement and sucrose preference were detected for assessing rat ethological changes on the beginning of the experiment and the 28th day. Then they were sacrificed and, taken the rats brain out on the ice. The expression of circadian gene were detected by using Q-PCR, Western blotting. Result: On behaviors,after 28 days the increases of the model rat body weight were significantly lower than normal group and drug group.On expression of the circadian gene clock, compared with normal group, the expression of circadian gene clock in model group was significantly higher than that in normal group. There was no significant difference among the agomelatine group, beta-asarone two doses group, and the normal group on the expression of the circadian gene clock;compared with the model group, the expression of the gene in agomelatonin group, beta-asarone two doses group were significantly lower than the model group;compared with agomelatine group, no significant difference in the expression of clock among the beta-asarone low dose group, the beta-asarone high dose group and the agomelatine group. Conclusion: beta-asarone may improve the depressive state in depression by affecting the expression of the depression in circadian clock genes clock rats.

      摘要:Objective: To observe the total flavone of hawthorn leaf (TFHL) on apoptosis of cerebral ischemia reperfusion injury in rats and c-fos, c-jun the effects of gene expression. Method: Eighty SD rats were randomly divided into sham group, model group, TFHL 70 mg · kg-1 group, 140 mg · kg-1group and Yinxingye tablet 3.5 mg · kg-1 group, and each group of 16. Four blood vessel blocking method to establish the whole cerebral ischemia reperfusion injury model of shuttle box test to detect the ability of learning and memory;spectrophotometric assay superoxide dismutase (SOD)activity and malondialdehyde(MDA) content;enzyme-linked immunosorbent assay (ELISA) method to detect the content of caspase-3;immunohistochemical method to detect c-jun and the expression of c-fos. Result: Compared with sham group, model group rats learning and memory ability, active rats escape response rate significantly decreased (P<0.01), Brain tissue apoptosis cells increased significantly (P<0.01);c-fos and c-jun gene expression obviously increased (P<0.01). Compared with model group, TFHL active escape response rate and Yinxingye tablet group rats increased (P<0.05), SOD activity was significantly increased (P<0.05), MDA content was significantly lower(P<0.05),apoptotic cells decreased significantly (P<0.05),c-fos and c-jun gene expression decreased significantly (P<0.05). Conclusion: TFHL can reducing cerebral ischemia reperfusion injury of brain tissue c-fos and c-jun gene expression, inhibition of neural cell apoptosis, improving cerebral ischemia-reperfusion injury in rats of nerve function, improve the learning and memory ability of rats.

      摘要:Objective: To investigate the effectiveness of water molecules transportation of salivary gland in Sjǒgrenǒs syndrome mice treated by different Chinese medical herbs. Method: The model of Sjǒgren's syndrome was made. The experiment was divided into normal group and model group, namely Spreading Hedyotis Herb group,Arnebiae radix group,Artemisiae Annuae Herba group,Taraxaci Herba group,Isatidis Folium group,Isatidis Radix group,Scutellariae Barbatae Herba group,Scrophulariae Radix group,Centellae Herba group,Rhizoma Paridis group. The mice were given drugs and normal saline from fourteenth day, once daily, for 4 weeks;the average water intakeand salivery secreatery were observed. Result: Compared with the normal group, the increased amount of water intake of the model group was significant(P<0.05). After treatment, compared with the model group, the amount of water intake of the treatment group reduced from 2 weeks the difference between the model group and spreading hedyotis herb group,artemisiae annuae herba group,isatidis folium group,scutellariae barbatae herba group was remarkable(P<0.05)after four weeks. At sixth week, the amount of water intake was reduced compared with the model group,in Spreading Hedyotis Herb group,Artemisiae Annuae Herba group,Taraxaci Herba group,Isatidis Folium group,Isatidis Radix group,Scutellariae Barbatae Herba group,Scrophulariae Radix group,Centellae Herba group. Compared with the normal group, the salivary flow of the model group reduced significantly(P<0.05)after treated two and four weeks. The salivary flow of model group and Artemisiae Annuae Herba group,Isatidis Folium group,Isatidis Radix group,Scutellariae Barbatae Herba group,Centellae Herba group was remarkably different(P<0.05) at fourth, sixth weeks,all the treatment group increased significantly (P<0.05). Compared with the normal group, the expression of AQP5 reduced significantly(P<0.05)and the AQP5 of all treatment groups was improved after treatment compared with the model group, and Artemistiae Annuae Herba group,Isatidis Folium group,Sculellaria Barbata group were improved significantly(P<0.05). Conclusion: The different Chinese medical herbs with heat clearing-detoxication are effective in reducing average water quantity,improving saliary flow and AQP5 of the Sjǒgren's syndrome mice.

      摘要:Objective: To investigate the effect of Danshao Huaxian capsule (DSHX) on the expressions of Gremlin and bone morphogenetic protein 7 (BMP7) in rat hepatic stellate cell (HSC) and to explore its possible mechanism in treating hepatic fibrosis. Method: A total of 20 male Wistar rats were randomly divided into control group, Fuzheng Huayu capsule group (0.8 g ·kg-1), low-and high-dose DSHX treated group (0.5, 1.0 g · kg-1) (n=5). Rats in each group were intragastrically administered with the corresponding medicines, respectively, twice per day for 5 days. Then, the drug-containing serums were collected and added to the cultured HSC-T6.HSC proliferation was detected using MTT colorimetric assay. Collagen (Col) Ⅰ, Col Ⅲ and α-smooth muscle actin (SMA) of culture supernatant were detected by ELISA. The expression of transforming growth factor (TGF) -β1 in HSC was measured by immunocytochemistry staining, and the expressions of Gremlin and BMP7 were determined by RT-PCR and Western blot, respectively. Result: Compared with the control group, the HSC proliferation was significantly inhibited (P<0.05), the concentrations of Col Ⅰ, Col Ⅲ, α-SMA and the expression levels of TGF-β1 were decreased (P<0.01, P<0.05), the mRNA and protein expressions of Gremlin were decreased (P<0.01), and the mRNA and protein expression of BMP7 were elevated (P<0.01) in the low-and high-dose DSHX serum groups. The above results in DSHX serum groups were the same as the positive control serum group. Conclusion: DSHX has potential effect in treating hepatic fibrosis in rats. It could inhibit the proliferation and activation of HSC and reduce the synthesis of extra cellular matrix, which are partly achieved by down-regulating gremlin and up-regulating BMP7.

      摘要:Objective: To investigate the inhibitory effects of capsaicin on pancreatic cancer SW1990 cells and explore its potential mechanism of action. Method: The pancreatic cancer SW1990 cells were treated with various concentrations of capsaicin (100, 150, 200, 250, 300 μmol · L-1), followed by 12, 24, 48 h cell culture. Effects on cell proliferation were determined by a CCK-8 Kit. SW1990 cells were treated with capsaicin (150, 200, 250 μmol · L-1) for 24 h. The cell cycle distribution and the percentage of apoptosis were determined by flow cytometer. Caspase-3 activity was measured by spectrophotometry and the expressions of p38 and p53 were examined by Western blotting analysis. Result: Capsaicin inhibited the growth of pancreatic cancer SW1990 cells in a dose-and time-dependent manner. Flow cytometer results showed that the cell cycle was arrested at G1/G0 phase and the apoptosis rate was increased in a dose-dependent manner. Pro-caspase-3 could be activated. Moreover, the expressions of p-p38 and p-p53 were up-regulated. Conclusion: Capsaicin could inhibit the growth of SW1990 cells by inducing G1/G0 phase cell cycle arrest and apoptosis. The molecular mechanism may be realized through up-regulating p38 and its downstream p53, and activating the pro-caspase-3, then promoting cell apoptosis in pancreatic cancer.

      摘要:Objective: To investigate the protective effect and its mechanism of different effective extracts of Mori Folium on the infrared(IR) model of HepG2 cells. Method: High glucose and insulin in HepG2 cells to form a cell model of insulin resistance, glucose oxidase detect the expression of glucose consumption, and using the real-time fluorescent quantitative polymerase chain reaction(PCR) technique to observe the expression level of C-Jun amino termind kinose(JNK) pathway related genes in different effective extracts of Mori Folium on the IR model of HepG2 cells. Result: High sugar and high insulin culture environment capable of inducing cells to produce insulin resistance.All of the groups can increase the glucose consumption(P<0.01)and has the influence on the expression of JNK related genes. Conclusion: The alkaloids, flavonoids and polysaccharides of mulberry leves can increase the glucose consumption on the HepG2 cells, which may be related to regulating JNK signaling pathways.

      摘要:Objective: To observe the effect of Yishen Huoxue herbs combined losartan on endogenous hydrogen sulfide and inflammatory cytokines in patients with maintenance hemodialysis. Method: Ninety patients with maintenance hemodialysis were selected and randomly divided into 3 groups (Yishen Huoxue herbs group, losartan group and Yishen Huoxue herbs combined losartan group). The endothelium-dependent flow-mediated dilatation (FMD) of brachial artery was determined using ultrasound, and the levels of total cholesterol (TC), triglyceride (TC), low-density lipoprotein cholesterol (LDL-C), homocysteine (Hcy), high-sensitivity C-reactive protein (hs-CRP), hydrogen sulfide (H2S), sulfhemoglobin (SHb), endothelin-1 (ET-1), nitric oxide (NO), tumor necrosis factor (TNF-α), interleukin-6 (IL-6) were measured before and after treatment. Result: After treatment, all patients in the combination group showed that the TC (3.68±0.85) mmol · L-1, LDL-C (2.07±1.61) mmol · L-1, Hcy (31.76±17.23) μmol · L-1 was decreased than before [(5.29±1.28) mmol · L-1, (3.45±1.84) mmol · L-1, (35.97±18.23) μmol · L-1, respectively] (P<0.05);the ET-1 (20.77±10.01)pg · mmol-1, TNF-α (39.6±6.53) ng · L-1, IL-6 (37.6±3.51) ng · L-1 was decreased than before [(25.39±14.26) ng · mol-1, (42.4±7.61)ng · L-1, (40.6±4.62) ng · L-1, respectively] (P<0.01). The FMD in the combination group was increased [(10.05±3.56)% vs (6.67±1.94)%] (P<0.01) after treatment. Conclusion: The treatment of combination Yishen Huoxue herbs and losartan could improve the recovery of vascular endothelial function in patients with maintenance hemodialysis effectively.

      摘要:Objective: To discuss the curative efficacy of Jiedu Yukui decoction cooperated with conventional western medicine therapy in treating erosive oral lichen planus (OLP) and to investigate its influence on serous tumor necrosis factor-α (TNF-α), and interleukin-2, 4, 10 (IL-2, 4, 10). Method: Eighty eligible patients were randomly divided into control group (40 cases) and traditional Chinese combined western medicine group (40 cases) according to the order of being in hospital. Patients in control group received compound chlorhexidine gargle thrice daily for 2 weeks, 1 mL triamcinolone acetonide combined with 2 mL 2% lidocaine submucosally local injection for 4 weeks, and vitamin A, C orally for 12 weeks. Based on the treatment of control group, patients in traditional Chinese combined western medicine group added dialectical modified Jiedu Yukui decoction for 12 weeks. The objective index and signs, subjective symptoms and quality of life were evaluated before and after 4 weeks of treatment. The levels of TNF-α and IL-2, 4, 10 were detected before and after 12 weeks of treatment. Result: The clinical effect in traditional Chinese combined western medicine group was superior to that in control group (P<0.05). After 4 weeks, scores of objective index and signs, subjective symptoms and quality of life were decreased, and the change was more obvious in traditional Chinese combined western medicine group as compared with before (P<0.01). Levels of TNF-α and IL-2, 4, 10 in traditional Chinese combined western medicine group were less than those in control group after 12 weeks of treatment (P<0.01). Recurrence rate was 22.5% in traditional Chinese combined western medicine group, which was lower than 45% in control group (P<0.05). Conclusion: Jiedu Yukui decoction cooperated with conventional western medicine therapy could relieve the symptoms of patients, improve their quality of lives and clinical effect, and reduce the recurrence in treating erosive oral lichen planus. Its mechanism of action may be related to regulating the inflammatory response induced by T cell.

      摘要:Objective: To observe the influence of Fuzheng Shugan decoction combined group psychotherapy in treating patients with stomach cancer after therapy. Method: Eighty patients were randomly divided into control group (40 cases) and observation group (40 cases) by random number table. Patients in control group received 20 mg fluoxetine capsules and 10-20 mg escitalopram citalopram tablets once daily. Patients in observation group received Fuzheng Shugan decoction combined group psychotherapy. Patients in both groups received 8 weeks of treatment. The self-rating anxiety scale (SAS), self-rating depression scale (SDS), Hamilton's depression scale (HAMD), Herth hopes list (HHI) and cartesian function scale (KPS) were scored before and after treatment. The quality of life was evaluated using QLQ-STO22 questionnaire of European organization for research and treatment of cancer (EORTC) once before and after treatment. Result: The amelioration of quality of life in observation group was superior to that in control group after treatment (P<0.05), while there was no statistic significance difference for antidepressant action between two groups. Scores of SAS and SDS in observation group were lower than those in control group (P<0.01), while there was no statistic significance difference for the score of HAMD in two groups. Score of KPS was decreased in control group, while it was increased and the result was higher in observation group (P<0.01). Scores of dysphagia, stomachache, reflux hiccup, constrained diet, anxiety, dry mouth and taste from QLQ-STO22 in observation group were lower than those in control group (P<0.01). Scores of the total average of hope, positive attitude to present and future and taking positive action and keeping healthy relationships with others from HHI in observation group were higher than those in control group (P<0.01). Conclusion: Fuzheng Shugan decoction combined group psychotherapy treatment has a comparative effect as compared with fluoxetine capsules in treating depression of stomach cancer after therapy. It can improve the hope of life, stabilize and improve the quality of life and realize the higher-quality life of patients.

      摘要:Objective: To discuss the clinical effect of nebulizer of Fufang Zhongjiefeng asrosol combined with proton pump inhibitors and gastric motility-promoting drugs in treating gastroesophageal reflux laryngitis (GERL). Method: Eighty patients with GERL were randomly divided into control group (40 cases) and observation group (40 cases) by random number table. Patients in both groups took 20 mg omeprazole magnesium enteric-coated tablets orally once daily and 5 mg mosapride citrate tablets orally thrice daily. Patients in control group added dexamethasone injection and nebulizer of gentamycin injection once daily for 8 weeks. And patients in observation group added nebulizer of Fufang Zhongjiefeng asrosol once daily for 8 weeks. Before and after treatment, scores of reflux symptom index (RSI) and reflux finding score (RFS) were graded, 24-hour pH of esophagus was detected. Result: The clinical effect in observation group was superior to that in control group (P<0.05). The scores of swallowing difficulty or breathing difficulties/suffocate attack and heartburn, acid reflux, chest pain, and the total scores of RSI in observation group were less than those in control group (P<0.01). Besides, the joint hypertrophy and granuloma/granulation tissue score, and the total RSI score in observation group were less than those in control group (P<0.01). After treatment, the total times of acid reflux and number>5 min, the percentage of the total of pH<4 and DeMeester scores in both groups were decrease as compared with the results before. But there was no statistically significant difference between the groups. Conclusion: Nebulizer of Fufang Zhongjiefeng asrosol combined with proton pump inhibitors and gastric motility-promoting drugs could ameliorate the symptom and sign of gastroesophageal reflux laryngitis, and there is obvious clinical effect.

      摘要:Objective: To discuss the clinical effects of Yichang Zhixie decoction on diarrhea predominant irritable bowel syndrome (IBS-D) and to investigate its influence on brain-gut peptide factors including serum 5-hydroxytryptamine (5-HT), substance P (SP), vasoactive intestinal peptide (VIP), somatostatin (SS), etc. Method: One hundred patients with IBS-D were randomly divided into the control group and the observation group (50 cases in each group). Patients in control group took orally 1 piece of flupentixol and melitracen tablet each morning and noon, and 2 g live combined bifidobacterium thrice daily. Based on the treatment of control group, patients in the observation group added 1 does of Yichang Zhixie decoction. Patients in both groups received a 4-week period of treatment. The digestive main symptom and the special quality of life scale (IBS-QOL) were scored before and after treatment. The prevalence and incidence situation of patients were recorded during 3-month period of follow-up. The levels of serum 5-HT, SP, VIP and SS were detected. Result: The clinical curative effect of observation group was superior to that of control group (P<0.05). The main digestive symptoms scores (abdominal pain, abdominal distension, defecation times, stool shape and mucous stool) in observation group were less than those in control group (P<0.01). IBS-QOL score of observation group was higher than that of control group after treatment and during follow-up period (P<0.01). The recurrence rate of observation group was 45.8%, which was inferior to 69.2% of control group during follow-up period (P<0.05). Levels of 5-HT, SP, VIP and SS in observation group were lower than those in control group (P<0.01). Conclusion: Yichang Zhixie decoction could reduce clinical symptoms, improve quality life and reduce long-term recurrence rate of IBS-QOL patients. Its action mechanism may be related to adjusting the levels of 5-HT, SP, VIP, and SS, although the exact mechanism deserves further study.

      摘要:Objective: To observe the curative efficacy of Huadu Yuyang decoction in treating mild and and moderate ulcerative colitis (UC) and to investigate its influence on interleukin-1, 4, 10(IL-1, 4, 10) and tumor necrosis factor-α (TNF-α). Method: Eighty-five patients with mild and and moderate UC in active stage were randomly divided into control group (42 cases) and observation group (43 cases) by random number table, and another 15 patients were assigned as healthy control. Patients in both groups received orally sulfasalazine enteric-coated tablets and prednisone. Patients in control group received mesalazine enemas, patients in observation group received retention enema of Huadu Yuyang decoction for 5 days in a week. Patients in two groups received 8 weeks of treatment. Scores of syndome of dampness-heat in large intestine and disease activity were graded, colonoscopy test was detected, and levels of serous IL-1, 4, 10 and TNF-α were detected before and after treatment. Result: The clinical effect in observation group was superior to that in control group (P<0.05). The syndrome of dampness-heat in large intestine, colonoscopy scores and the Mayo scores in observation group were less than those in control group (P<0.01).Levels of serum TNF-α and IL-1 were obviously higher (P<0.01), IL-4 and IL-10 levels were significantly lower in two groups than those in healthy control groups before treatment (P<0.01). After treatment, levels of serum TNF-α and IL-1 in observation group were lower than those in control group (P<0.01). There was no statistically significant difference for TNF-α level between observation group and healthy control group. IL-4 and IL-10 levels in observation group were superior to those in control group (P<0.01). Conclusion: Huadu Yuyang decoction retention enema could improve clinical symptoms, control activities, and promote the healing of mucosa ulcer in UC patients at mild and moderate stage. Its mechanism may be related to promoting the inflammatory cytokines/suppression of inflammatory cytokines balance and reducing intestinal inflammation of UC.

      摘要:Objective: In order to explore the single medicine and medicine suits of using rules in Synopsis of Golden Chamber, and merger with Treatise on Febrile Disease to find the using rules in Theory of Typhoid Miscellaneous Disease, eventually, we can find drug laws used by Zhang Zhongjing. Method: We use strong bidirectional association rules in Synopsis of Golden Chamber and Theory of Typhoid Miscellaneous Disease respectively(exception of unilateral), and design four group to statistical the single medicine and medicine suits' support and confidence, then find out the single support above 0.1 medicine and medicine on support and confidence is higher than 0.05. Result: The first six high-frequency single-agent with Glycyrrhizae Radix et Rhizoma, Cinnamomi Ramulus, Zingiberis Rhizoma Recens, Jujubae Fructus,Pinelliae Rhizoma, Pulsatillae Radix in 1, 3 groups; the first six high-frequency medicine suits are Cinnamomi Ramulus, Pinelliae Rhizoma, Pulsatillae Radix, Poria, Zingiberis Rhizoma, Ginseng Radix et Rhizoma in 2, 4 groups. Table 4 before six high-frequency medicine with Cinnamomi Ramulus, Pulsatillae Radix; Pinelliae Rhizoma and Zingiberis Rhizoma; Cinnamomi Ramulus, Poria; Cinnamomi Ramulus, Atractylodis Macerocephalae Rhizoma; Ginseng Radix et Rhizoma, Cinnamomi Ramulus; Ginseng Radix et Rhizoma, Pinelliae Rhizoma; Cinnamomi Ramulus, Pulsatillae Radix; Ginseng Radix et Rhizoma, Pinelliae Rhizoma;Ginseng Radix et Rhizoma, Cinnamomi Ramulus; Ginseng Radix et Rhizoma, Zingiberis Rhizoma; Cinnamomi Ramulus, Poria; Cinnamomi Ramulus, Ephedrae Herba. Conclusion: The medication in Golden is warm, assits the Yang spirit;Supporting Zhang Zhongjing treatment internal medicine miscellaneous disease, focusing on the tonify deficiency, and cure these diseases, bias exorcism, nourish the commonly used method;always protect spleen and stomach, the postnatal weight;the consumer medicinal preparation and have the obvious core tendency, Zhang Zhongjing prescription has the obvious core tendency in the medicine use aspect, forms take the core medicine as hen main body, take often applies drugs to as the link, take the base medicinal preparation as central kind of side system. Looking from uses the core tendency, the medicine used in Synopsis of Golden Chamber and Treatise on Febrile Disease is roughly consistent, but the medicine uses in Synopsis is wider than Febrile.

      摘要:Objective: To analyze the compatible principles of the prescriptions for depression disease by using traditional Chinese medicine inheritance support system (TCMISS). Method: The prescriptions for depression disease in China national knowledge infrastructure (CNKI) were collected and analyzed using data mining method. Result: Based on the analysis of 252 cases of prescriptions, 30 of 239 herbs (frequency≥25) were found to be commonly used in clinic, and 142 or 17 couplet medicines (support≥25 or 50, respectively), 10 association rules (support≥25 and confidence≥0.9), 26 core combinations and 5 new prescriptions were obtained from the database. Conclusion: TCMISS is a helpful tool for the research of traditional Chinese medicine. The prescriptions for prevention and cure of depression disease are mainly composed of Bupleurum, supplemented by herbs to activate blood circulation and herbs to dissipate phlegm for resuscitation, and so on.

      摘要:The intestine is an important place for oral drug metabolism in the body, a large number of bacteria in the gut has played an inevitable role in drug biological conversion. Thus, the intestinal flora biotransformation of the active ingredients of traditional Chinese medicine (TCM) has become the focus of attention in recent years. In order to further explore the regularization of biotransformation, the relevant literatures of biotransformation on TCM including a single active ingredient and compound ingredients were collected. Related literature searches were performed in CNKI, CMCC, VIP, CBM WanFang and PubMed Data in the past 20 years. The results implied that the glycosides could be hydrolyzed to aglycone;alkaloids could be hydrolysised and dehydrated easily;diester could be converted to the corresponding mono-type alkaloids and lipids, which reduced the toxicity;lactones structure may cause breakage or demethylation reactions. In vivo biotransformations were occurred in many types of active ingredients of TCM via intestinal flora, which caused changes in the structure or the formation of new active ingredients. Intestinal flora also played an important role in the metabolism of TCM, such as for the confirmation of the rationality and efficiency of herbal compatibility.
