      摘要:Biomechanopharmacology opens a new approach and brings new topics for researches of activating blood circulation to remove blood stasis.To investigate bioresponses come from flow shear stress and Chinese herbal medicines and to determine the dose-response patterns in anti-atherosclerosis,anti-thrombosis and thrombolysis gained from joint effects of shear stress and Chinese herbal medicines,will play active roles in prevention and treatment for atherosclerosis and thrombosis and in preventive medicine generally.

      摘要:Objective: To investigate intestinal absorption characteristics of index components from Huanglian Jiedu Tang (HLJDT) in normal and cerebral ischemia states,and discuss rationality of HLJDT used in clinical to treat cerebral ischemia,basing on theory of correspondence between syndrome and prescription. Method: Taking geniposide,berberine hydrochloride and baicalin in HLJDT as indexes,in situ single pass perfusion model was established to investigate intestinal absorption characteristics of HLJDT with different concentrations in rats with normal and cerebral ischemia. Result: Compared with the normal group,absorption of geniposide,berberine hydrochloride and baicalin from HLJDT with different concentrations in all model group increased to some extent in different intestine segments.Berberine hydrochloride increased significantly in duodenum and jejunum.Baicalin increased significantly in the whole intestine.Intestinal absorption of geniposide increased significantly in jejunum,ileum and colon. Conclusion: Absorption of main active ingredients of HLJDT in the cerebral ischemia model group is better than that in the normal group,which supports rationality of widely use of HLJDT in the clinical application to exerting therapeutic effect and confirms theory of correspondence between syndrome and prescription.

      摘要:Objective: To illuminate effects of viscosity on release of sustained-release preparations after compatibility of homemade slow-release improved accessory I and others. Method: With Chuanping extract as a model drug and dextrin as a filler,different sustained-release tablets(prescription A,B,C) with different dosage of modified accessory I(40%,45% and 50%) were prepared;sustained-release tablets(prescription D,E,F,G) with the best proportion of modified accessory I and other accessories[lactose,dextin-sugar(1: 1),starch-sugar(1: 1),microcrystalline cellulose] were prepared.HPLC was adopted to measure release rates of ephedrine,pseudoephedrine and scopolamine in Chuanping sustained-release tablets,balanced release of different component were determined by similarity factor method. Result: Torque of accessory I was less than 10%,viscosity of improved accessory I was 218.3×10-3 Pa·s.Viscosities of prescriptions A,B,C were 105×10-3,265×10-3,540×10-3 Pa·s;release finished at 6,8,10,respectively.Finished-release time of prescription C met demand,release of ephedrine,pseudoephedrine and scopolamine at 10 h were 97.21%,96.79%,94.21%,releasing factor(f2) values were 75.08,71.42,61.88,balanced release rates were obtained well.Viscosities of prescription D,E,F,G were 400×10-3,435×10-3,475×10-3,808.3×10-3 Pa·s,finished-release times were 8,>10,10,2 h,respectively.Prescription F met requirement,release degrees of ephedrine,pseudoephedrine and scopolamine at 10 h were 98.40%,97.92%,97.99%,f2 values were 74.78,74.65,74.70,balanced release rates were obtained well. Conclusion: Improved accessory I can meet demand of balanced release well.Compatibility of different accessories can affect balanced release of Chuanping sustained-release tablets.When different dosages of improved accessory I combine with dextrin,viscosity and release are positively correlated;when improved accessory I is compatible with different filler,viscosity and release are negatively correlated.

      摘要:Objective: To provide a reference for application of total polysaccharides in Coicis Semen by investigating effect of key parameters on high temperature α-amylase enzymolysis for starch in Coicis Semen. Method: Take the content of reducing sugar as a quantitative indicator and color change after iodine solution testing as a qualitative indicator,based on single factor tests,orthogonal test was adopted to investigate effects of the amount of enzyme,hydrolysis temperature,hydrolysis pH and reaction time on amylohydrolysis. Result: Optimal parameters were as follows:the amount of enzyme 600 U·g-1,enzymolysis temperature 75 ℃,enzymatic pH of 6.0,enzymolysis time of 120 min.The content of reducing sugar in extract of total polysaccharides in Coicis Semen was about 25%,meanwhile,color of samples did not alter after iodine solution testing.Purity of total polysaccharides in Coicis Semen was 76.63%,which showed a great improvement by comparing with 54.41% of samples without removing starch. Conclusion: High-temperature α-amylase can remove starch from Coicis Semen readily and rapidly,it is conducive to extraction of total polysaccharides.

      摘要:Objective: To optimize formulation of Ginkgo Folium bi-layer osmotic pump controlled-release tablets. Method: With accumulative release rate of total flavonol glycosides as index,single-factor tests were adopted to screen formulations of drug-layer,push-layer and coating solution,drug release behavior was evaluated by similarity factor method.Taking the amount of PEG4000 in coating solution,coating weight and the amount of polyoxyethylene in push-layer as independent variables,linearity of release curve and accumulative drug release in 16 h as dependent variables,central composite design-response surface methodology was applied to optimize formulation. Result: Optimum formula was as follows:polyoxyethylene amount of 120 mg,the amount of PEG4000 of 1.5 g,coating weight of 11%;accumulative release of total flavonol glycosides in 16 h was above 90%. Conclusion: Release curve of this preparation meets requirements with correlation coefficient of 0.996 4.This optimized formulation is stable and suitable for industrial production of Ginkgo Folium bi-layer osmotic pump controlled-release tablets.

      摘要:Objective: To optimize clarification process of Gastrodiae Rhizoma extract with chitosan flocculating method. Method: HPLC was employed to determine the content of gastrodin with mobile phase of acetonitrile-water(3: 97) and detection wavelength at 220 nm.Taking clarification effect and solids content as indexes,on the basis of single factor tests,orthogonal design was used to optimize clarification process with the adding amount of chitosan,liquid concentration ratio,flocculation temperature and standing time as factors. Result: Optimal clarification procedure was obtained under these conditions:the adding amount of chitosan 0.15%,liquid concentration ratio 1.13 g·mL-1,flocculation temperature at 25 ℃,standing time of 10 h.The concentration of gastrodin was 65.93 g·L-1,solids content was 3.157%. Conclusion: This optimized process is feasible and economical with good clarification effect,which is suitable for purification of Chinese medicine extract.

      摘要:Objective: To encapsulate ginsenoside Rh2 within glycyrrhetinic acid modified liposomes and investigate its physicochemical properties and in vitro inhibitory effect on SMMC-7721 cells. Method: Ginsenoside Rh2 liposomes(Rh2-L) and ginsenoside Rh2 liposomes surface-modified with glycyrrhetinic acid(GA-Rh2-L) were prepared by film dispersion method,UPLC was employed to determine the content of ginsenoside Rh2 with mobile phase of acetonitrile-water(28: 72),flow rate of 0.3 mL·min-1 and detection wavelength at 203 nm.Encapsulation efficiency(EE),particle size,Zeta potential and in vitro release were measured,MTT experiment was adopted to evaluate inhibitory effect of ginsenoside Rh2 solution,Rh2-L and GA-Rh2-L on SMMC-7721 cells. Result: Modification of glycyrrhetinic acid showed no effect on physicochemical properties of Rh2-L,such as EE,particle size,Zeta potential and in vitro release.EE of Rh2-L and GA-Rh2-L were (91.67±1.05)% and (95.54±2.23)%,average particle size of them were (138.6±45.8) nm and (146.5±48.9) nm,Zeta potential of them were -(12.75±0.34) mV and -(14.79±0.67) mV,respectively.In vitro release of ginsenoside Rh2 from Rh2-L and GA-Rh2-L within 72 h were (84.67±7.23)% and (89.03±8.61)%,while ginsenoside Rh2 solution released (91.23±5.17)% within 12 h.In vitro cytotoxicities(IC50) of GA-Rh2-L on SMMC-7721 cells was increased by 0.524 fold compared with ginsenoside Rh2 solution and 0.596 fold compared with Rh2-L.Ginsenoside Rh2 solution,Rh2-L and GA-Rh2-L all inhibited proliferation of SMMC-7721 cells in dose-and-time-dependent manners. Conclusion: Glycyrrhetinic acid modification can not affect physicochemical properties of Rh2-L,and it can increase targeting efficiency of liposomes on hepatocellular carcinoma,thus resulting in higher cytotoxicities in comparison with Rh2-L.This study provides new ideas for targeted-therapy to hepatocellular carcinoma.

      摘要:Objective: To optimzie preparation process of pomegranate peel polyphenols liposomes by using supercritical CO2 fluid precipitation technology. Method: With entrapment efficiency as index,main factors were screened by Plackett-Burman test,preparation process of pomegranate peel polyphenols liposomes was optimized by steepest ascent test and Box-Behnken test. Result: Ratio of raw materials and membrane material,reaction temperature and pressure were significant factors,optimal preparation conditions was as following:ratio of raw materials and membrane material of 1: 21.15,reaction temperature at 48.3 ℃,reaction pressure of 23.88 MPa.The measured value and the predicted value of entrapment efficiency were 80.295% and 81.379%,effective size was 168.5 nm,Zeta potential was -73.8 mV. Conclusion: This test model meets requirements.In vitro release of pomegranate peel polyphenols liposomes is slow,its sustained release effect is obvious.

      摘要:Objective: To optimize fermentation conditions of total polysaccharides from Astragali Radix by probiotic N-14. Method: UV was adopted to determine the content of total polysaccharides with detection wavelength at 620 nm.Based on single factor tests,response surface methodology(RSM) was employed to optimize fermentation process of total polysaccharides from Astragali Radix with yield of total polysaccharides as response value and sterilization time,fermentation time,the amount of substrate as independent variables. Result: Optimum fermentation conditions were as follows:the amount of substrate 4 g,fermentation time of 24 h,sterilization time of 15 min,pH of 7,inoculation of probiotic 1%.Yield of total polysaccharides was up to 6.56%,which was lower than the predictive value of 6.69%,but 57.31% higher than yield of total polysaccharides(4.17%) by water extraction and alcohol precipitation process. Conclusion: RSM is suited to optimize fermentation process of Astragali Radix with probiotic N-14,this optimized process is stable and feasible,it provides a reference for utilization of this herb.

      摘要:Objective: To optimize extraction technology of total saponins from Ziziphi Spinosae Semen and provide a reference for industrial production of this component. Method: HPLC-UV method was employed to detect contents of jujuboside A and jujuboside B with mobile phase of acetonitrile-water(34: 66) and detection wavelength at 204 nm.On the basis of single factor tests,effects of ethanol concentration,liquid-sodid ratio,extraction time and extraction times on extraction process were investigated by orthogonal test. Result: Linear ranges of jujuboside A and jujuboside B were 0.24-2.40,0.12-1.20 μg,average recovery of them were 100.39% and 99.40%.The best extraction technology of total saponins from Ziziphi Spinosae Semen was as following:extracted 2 times with 12 times the amount of 70% ethanol for 1.5 h per time;average contents of jujuboside A and jujuboside B were 0.606,0.228 mg·g-1,respectively. Conclusion: This established HPLC method is accurate and reliable to detect contents of jujuboside A and jujuboside B.At the same time,this optimized extraction technology of total saponins from Ziziphi Spinosae Semen is stable and feasible with advantages of energy-saving,time-saving and so on,so it is suitable for industrial production.

      摘要:Objective: To optimize extraction technology of Qizhu granules. Method: With contents of astragaloside and paeoniflorin as comprehensive evaluation index,orthogonal test was adopted to optimize water extracting technology.Taking composite score of extracting amounts of specnuezhenide and berberine hydrochloride as index,orthogonal test was adopted to optimize ethanol extraction process.The content of astragaloside was determined by HPLC-ELSD,mobile phase consisted of acetonitrile-water (34: 66).HPLC-DAD was employed to determine contents of paeoniflorin,specnuezhenide and berberine hydrochloride with mobile phases of methanol-0.05 mol·L-1 potassium dihydrogen phosphate solution (30: 70),methanol-water (32: 68),0.05 mol·L-1potassium dihydrogen phosphate solution-acetonitrile(50: 50) and detection wavelengths of 230,224,345 nm,respectively. Result: The best water extraction process was:extracted twice with 12,10 times the amount of water for 2 h per time.Optimum ethanol extraction process was as following:extracted thrice with 6 times the amount of 60% ethanol for 2 h each time. Conclusion: These optimized technologies are stable,feasible and suitable for standardized production of Qizhu granules.

      摘要:Objective: To optimize extraction process of volatile oil from Ferula assafoetida by steam distillation and analyze chemical compositions in volatile oil. Method: With extraction rate of volatile oil as index,conditions for extraction of volatile oil from F. assafoetida were optimized by uniform design.Effect of extraction time on chemical compositions in volatile oil were investigated by GC,conditions were as following:high purity nitrogen as carrier gas,flow rate of carrier gas 1 mL·min-1,injection volume of 0.5 μL,split ratio of 50: 1,total flow of 54.0 mL·min-1,inlet temperature of 250 ℃,detector temperature of 250 ℃.Temperature-rising program was as following:the initial temperature of 60 ℃,kept for 2 min,then temperature rise to 80 ℃ with heating rate of 4 ℃·min-1,kept for 5 min,temperature rise to 180 ℃ with heating rate of 10 ℃·min-1,kept for 5 min,temperature rise to 200 ℃ with heating rate was 4 ℃·min-1,kept 2 min.Extraction process of volatile oil from F. assafoetida was determined by GC and chromatogram similarity. Result: Some compounds in volatile oil were unstable thermally.Optimum extraction process of volatile oil from F. assafoetida was as following:added 6.5 times amount of water and extracted 4 hours.Average extraction rate of volatile oil was 8.32%. Conclusion: This optimized extraction process of volatile oil is reproducible,reasonable and practicable,it can guarantee stability of compositions in volatile oil from F. assafoetida.

      摘要:Objective: To optimize extraction process of burning Drynariae Rhizoma formula granules and establish determination of naringin in this preparation. Method: Taking extracting amount of naringin and yield of dry extract as a comprehensive evaluation index,orthogonal test was adopted to optimize extraction process with the amount of water,extraction time and extraction times as factors.Naringin in this formula granules was determinated by HPLC,mobile phase was methanol-acetic acid-water(35: 4: 65) and detection wavelength was 283 nm. Result: The best extraction technique was as follows:soaked 0.5 h,extracted 2 times with 10,8 times the amount of water for 0.5 h per time.The mass fraction of naringin was 0.77% and yield of dry extract was 12.6%.Calibration curve of naringin was linear in the range of 0.063-1.575 μg,average recovery was 100.70% with RSD of 1.2%.Tentatively per 1 g of this formula particle contained ≥5 mg of naringin. Conclusion: This extraction process and determination are stable and feasible with high extraction efficiency of naringin,which can be scientific basis for production and quality control of burning Drynariae Rhizoma formula granules.

      摘要:Objective: To establish a sensitive and specific HPLC method for processed products of ginger and analyse the chemical composition of dried ginger, ginger peel and charry ginger. Method: HPLC-UV analysis was carried out on an Agilent TC-C18(2) column(4.6 mm×250 mm, 5 μm) with binary solvent delivery gradient elution, photodiode-array-detector. The detection wavelength was set at 280 nm and the column temperature was kept at 30 ℃. Ten batches of fresh ginger were collected from different habitats in China, and the processed products were analyzed with the established method. The data were analyzed with ‘similarity evaluation system for chromatographic specific chromatogram of traditional Chinese medicine (Version2004A)’ Result: Four main peaks were designated as 6-gingerol, 8-gingerol, 6-shaogol and 10-gingerol. Compared with dried ginger, the contents of 6-gingerol were gradually decreased while the 6-shaogol increased in charry ginger, and at the same time, some new elements were found in the later. Conclusion: The good reproducibility and stability are shown in experiment results. The fingerprint can be used for the comparison of died ginger, ginger peel, and charry ginger.

      摘要:Objective: To establish an HPLC method for simultaneous determination of five nucleosides, uridine 5'-monophosphate, uridine, adenosine 5'-monophosphate monohydrate, guanosine, adenosine in Bailing capsules. Method: The separation was developed on an Ultimate AQ C18 column (4.6 mm×250 mm, 5 μm) by gradient elution with methanol-0.05 mol·L-1 potassium dihydrogen phosphate solution, with flow rate at 1.0 mL·min-1 and column temperature at 25 ℃. The detection wavelength was at 260 nm. Result: The baseline separation of uridine 5'-monophosphate, uridine, adenosine 5'-monophosphate monohydrate, guanosine, and adenosine was successfully achieved. The results showed that these five ingredients had good linearity within the test ranges of 35.25-528.8(r=1.000), 38.07-571.1(r=1.000), 34.96-524.5(r=0.999 8), 38.34-575.1(r=1.000) and 39.84-597.6 ng(r=1.000), respectively. The recovery were 101.2% (RSD 1.9%), 103.1% (RSD 1.1%), 96.7% (RSD 1.3%), 101.4% (RSD 2.7%) and 103.2% (RSD 1.6%), respectively. Conclusion: The developed method of the HPLC-fingerprint and quantitative analysis showed good specificity, precision, reproducibility, stability and could be used for the quality control of Bailing capsules.

      摘要:Objective: To establish a quantitative method of three main saponins and two major active flavonoids in Sanjie Zhentong capsules by high-performance liquid chromatography coupled with ultraviolet detectors and evaporative light scattering detectors (HPLC-UV-ELSD), and compare the contents of notogin-senoside R1, ginsenoside Rg1,ginsenoside Rb1,loureirin A, loureirin B among 15 batches of the drugs. Method: HPLC-UV-ELSD was conducted on a Waters Symmetry@C18(4.6 mm×250 mm, 5 μm) with gradient elution, the mobile phase was ACN and H2O, the temperature of drift tube was 45 ℃ and the nebulizing gas flow rate was 3.7 L·min-1. Result: Five compounds were simultaneously detected with one injection, good linear relationships were found(0.999 5-0.999 7), and the average recovery rates were 99.1%-104.2%. Conclusion: The HPLC-UV-ELSD method is simple and reproducible, which can be used for quality control of Sanjie Zhentong capsules.

      摘要:Objective: To explore the bioactive components of Lonicerae Japonicae Caulis and to study the phenolic acids of the plant. Method: Compounds were isolated and purified by solvent extraction ethanol extraction, repeated column chromatography on silicagel and Sephadex LH-20.Their structures were elucidated on the basis of physical characteristics and spectral data. Result: Six phenolic compounds were obtained and identified as methyl p-hydroxycinnamate (1), ethyl caffeate (2), methyl caffeate (3), ferulic acid (4), caffeic acid (5), and chlorogenic acid (6). Conclusion: Compounds 1, 3 and 4 were isolated from Lonicerae Japonicae Caulis for the first time.

      摘要:Objective: HPLC was adopted to determine the baicalin content of the Scutellariae Radix and Scutellariae Radix powder by using irradiation sterilization, together with herbs without the sterilization. Compared the content changes before and after mirradiation sterilization. Method: Accurasil-C18 chromatographic column(4.6 mm×250 mm,5 μm), methanol-water-phosphoric acid (47: 53: 0.2) as mobile phase, the wavelength detection was 280 nm with flow rate of 1.0 mL·min-1, column temperature 30 ℃, and the injection volume of 10 μL. Result: Baicalin had good linear relationship in the range of 0.163 2-0.816 0 μg. The average recovery was 99.44% with the RSD of 1.6%. Content baicalin in Radix had significant changes before and after sterilization. Conclusion: It indicates that Scutellariae Radix is not suitable for applications of irradiation sterilization,which takes baicalin as index.

      摘要:Objective: To establish a TLC fingerprint for identifying Gastrodiae Rhizoma and compare fingerprint of Gastrodiae Rhizoma from different habitats. Method: A TLC method for identification of Gastrodiae Rhizoma was established with silica gel GF254 TLC plate and adding methanol for pre-deployment,methanol was evaporated to dryness,activation for 30 min at 110 ℃,with chloroform-ethyl acetate-methanol-water(2.5: 1: 1: 0.1) as developing agent,deployment temperature was 20-25 ℃,relative humidity was 60%-70%,sprayed with 10% sulfuric acid in ethanol at 105 ℃,viewing and imaging under UV light(366 nm),fingerprint software generated a total system solution model.Differences of 30 batches of Gastrodiae Rhizoma were investigated through cluster analysis and principal component analysis. Result: This established TLC had a clear spot color.Cluster analysis of thirty batches of Gastrodiae Rhizoma was divided into five categories,principal component analysis was divided into six categories,it could distinguish Gastrodiae Rhizoma from different habitats. Conclusion: Diversity differentiation of Gastrodiae Rhizoma may be due to geography,spatial distance,temperature and humidity,in terms of chemical compositions shows some differences.This established TLC fingerprint can be used to control quality of Gastrodiae Rhizoma from different sources.

      摘要:Objective: Study on the content of several inorganic elements in Dipsaci Radix before and after stir frying with wine, and analyzed by grey relational grade analysis. Method: The content of inorganic elements were determined by atomic absorption spectroscopy and atomic fluorescence spectrometry. Result: The content of inorganic elements, Fe, Mn, Cu, Zn, Pb, Cd, As, and Hg, are different in the Dipsaci Radix before and after stir frying with wine, and with no overall upward or downward trend. Conclusion: The content and distribution of inorganic elements in Dipsaci Radix has changed after processed. The content of Fe, Mn and Zn has increase, especially the content of Mn. This may be one reason why wine fried Dipsaci Radix with the activity of nourishing liver and kidney.

      摘要:Objective: Analysis and comparison of Hubei Mao herb fried gluten, rice water broiled, fried and other three kinds of different processing methods in the chemical composition of volatile oil. Method: Application of steam distillation to extract three different processing methods of Hubei lancea volatile constituents by GC-MS analysis and identification of the chemical composition, and the use of peak area normalization method to calculate the relative quality of each component scores. Result: From three kinds of 88 kinds of volatile oil components were separated and identified 87 kinds of volatile chemical composition, their common ingredient 51 kinds, identified the chemical composition of each sample on average accounted for 94.61% of total volatile oil. Conclusion: There are some differences between three kinds of Hubei lancea volatile chemical composition and content of the different processing methods.

      摘要:Objective: To establish GC fingerprints of volatile oil from Chuanxiong Rhizoma. To compare the consistency of fingerprints of volatile oil from Chuanxiong Rhizoma by distillation coupling solvent extraction and traditional distillation extraction. Method: The volatile oil was extracted from Chuanxiong Rhizoma by distillation coupling solvent extraction.The chromatography fingerprints was established by AT-5 fused-silica capillary (0.32 mm×30 m,0.25 μm)column,injector temperature of 250 ℃, FID detector with temperature at 250 ℃.The similarity was calculated with "TCM fingerprint aided similarity evaluation software". Result: Established GC fingerprints of volatile oil from Chuanxiong Rhizoma.The reproducibility,accuracy and stability of the method were very good in the tests (all RSD<3%).The common components of volatile oil from Ligusticum chuanxiong by distillation coupling solvent extraction and traditional distillation extraction>98%.The similarity of common components was more than 0.998. Conclusion: The ligustilide from Chuanxiong Rhizoma by distillation coupling solvent extraction is 83.46 percent higher than traditional distillation extraction.The fingerprints of volatile oil from Chuanxiong Rhizoma by distillation coupling solvent extraction and traditional distillation extraction is consistent.

      摘要:Objective: To optimize sample solution preparation method and establish assay method of astragaloside IV in Astragali Radix. Method: UPLC-ELSD was used to determine the content of astragaloside IV with mobile phase of acetonitrile-water(32: 68),column temperature of 30 ℃,flow rate of 0.5 mL·min-1,temperature of ELSD at 45 ℃,carrier gas was dry compressed air with pressure of 0.5 kPa,gain value was 7.Ammonia-methanol mixture was used to ultrasonic extract of astragaloside IV as well as n-butanol extraction.Preparation method of test solution was optimized through single-factor tests. Result: Optimal preparation method was as following:added methanol 80 mL and concentrated ammonia 40 mL,ultrasonic extraction for 30 min,purification by subsequent extraction with n-butanol for four times every 40 mL in sample preparation.The content of astragaloside Ⅳ from 11 batches of Astragali Radix by this optimizing method were significantly higher than the result determined by HPLC method in 2010 edition of Chinese Pharmacopoeia. Conclusion: This optimized sample preparation method is simple with less loss of effective constituents.UPLC analysis method is accurate and reliable with high sensitivity.

      摘要:Objective: To establish an HPLC method for simultaneously determining the contents of calycosin-7-O-β-D-glucoside, hypericin, rutin, isoquercetin, myricetin, quercetin, loganin, morroniide in compound Huangkui granules. Method: The determination was performed on an Aglient C18(4.6 mm×250 mm,5 μm) chromatographic column,the mobile phase was acetonitrile-0.05% formic acid aqueous solution with gradient elution at a flow rate of 1.0 mL·min-1;the detection wavelength was 240,260 and 360 nm, column temperature was 30 ℃. Result: The calibration curves of calycosin-7-O-β-D-glucoside, hypericin, rutin, isoquercetin, myricetin, quercetin, loganin, morroniide were linear in the ranges of 3.5-35.0(r=0.999 6),32.09-300.9(r=0.999 7),2.21-22.12(r=0.999 7),18.68-186.9(r=0.999 8),6.32-63.20(r=0.999 7),3.54-35.44(r=0.999 6),12.39-123.9(r=0.999 6), 12.88-128.8 mg·L-1(r=0.999 6),respectively. The average recoveries were 98.1%(RSD 1.7%),99.7%(RSD 0.6%),98.7%(RSD 2.5%),100.1%(RSD 1.4%),99.7%(RSD 1.0%),99.9%(RSD 2.7%),99.0%(RSD 1.2%),99.7%(RSD 1.3%),respectively. Conclusion: The method established is simple, accurate and reproducible, which can be used for the quality control of Huangkui granules.

      摘要:Objective: The aim of this study was to express the odor and taste of Chinese Hawthorn objectively by using electronic nose and electronic tongue technology to, then to distinguish the samples collected from different areas and species. Method: The odors and taste response values of Chinese Hawthorn samples were obtained by using electronic nose and electronic tongue. The data was analyzed through principal component analysis (PCA), discriminant factor analysis (DFA) and statistical quality control (SQC) analysis. Result: Chinese Hawthorn samples from both different areas and species can be discriminated by electronic nose, and electronic tongue can effectively distinguish samples from different species. Conclusion: Electronic nose and electronic tongue can be used to distinguish Chinese Hawthorn samples from different areas and species, and in combination with chemometric models they could be applied to identify the odor and taste of Chinese Hawthorn as a new method.

      摘要:Objective: To optimize the concentration of ethanol to extract total flavonoids from Bauhinia championii, and purified using silica gel column chromatography. Quantitative determine the content of two major flavonoids in total flavonoids. Method: The optimized concentration of ethanol was selected based on anti-inflammatory effects of extract from B. chaampioni on knee osteoarthritis model in rats. The total flavonoids were enriched by silica gel column chromatography. The contents of two flavonoids, 3',4'-methylenedioxy-5',5,6,7-tetramethoxyflavone and 3',4',5',5,7-pentamethoxyflavone was determined by HPLC. Result: The 70% ethanol extract of B. chaampionihad the strongest anti-inflammatory effect. The solvent system used in silica gel column chromatography was dichloromethane-ethyl acetate (8:1 to 4:1). The content of 3',4'-methylenedioxy-5',5,6,7-tetramethoxyflavone and 3',4',5',5,7-pentamethoxyflavone in total flavonoids were 21.3% and 46.7%, respectively. Conclusion: The established route to obtain total flavonoid from B. chaampioni was feasible.

      摘要:Objective: To develop an HPLC method for simultaneous quantification of paeonol and rhein in the tissue of rats. Method: The adopted chromatographic column was Phenomsil BDS C18(4.6 mm×250 mm, 5 μm), the mobile phase was acetonitrile-0.1% phosphoric acid aqueous solution (37: 63), the flow rate was 1.0 mL·min-1, and detection wavelength was set at 274 nm. Methyl alcohol was used to precipitate the protein of the plasma samples and the aurantio-obtusin was used as an internal standard. Result: The linear range of paeonol was Y=-0.055+0.03X(r=0.993 2). The intra-day RSD and inter-day RSD were less than 4.1%. For rhein, the linear range was Y=-0.152+0.014X(r=0.993 5), the intra-day RSD and inter-day RSD were less than 4.9%.The stability of the plasma sample were well acceptable. Conclusion: The developed method for the quantification of paeonol and rhein in rat plasma is suitable for the distribution studies of drug-couple Radix Paeoniae Rubra and Rhei Radix et Rhizoma in the tissue of rats.

      摘要:Objective: To observe the effect of Yishen Huazhuo decoction on express of brain derived neurotrophic factor(BDNF) and tyrosine kinase keceptor B(TrkB) in the hippocampus of experimental rat model with Alzheimer's disease and explore the mechanism of Yishen Huazhuo decoction. Method: Male SD rats were randomly divided into 5 groups, sham operation group, model group, low, middle and high dose group of Yishen Huazhuo decoction. According to the brain stereotaxic atlas of rats, animal model of Alzheimer's disease was established by injecting Aβ25-35 into the left lateral ventricle. Treatment groups were given three different doses of Yishen Huazhuo decoction (2.8,5.6,11.2 g·kg-1) within the next 4 weeks, once daily. The Morris Water Maze was employed to investigate the function of learning and memory in model rats. The effect of protein expression of BDNF and its TrkB receptor in the hippocampus were detected by means of Western blot. Result: Compared with sham operation group, models’ learning and memory ability and the protein expression of BDNF and its TrkB receptor in the hippocampus were significantly lower (P<0.05). Yishen Huazhuo decoction could significantly enhance the learning and memory ability and the protein expression of BDNF as well as its TrkB receptor in the hippocampus of the rats compared with the model group (P<0.05). Conclusion: Yishen Huazhuo decoction could promote the protein expression of BDNF and its TrkB receptor in the hippocampus, and improve the learning and memory ability of the AD rats.

      摘要:Objective: To observe the effects and mechanisms of Yizhi capsule on beta-amyloid protein 1-42 (Aβ1-42)induced learning memory disorders and cholinergic system in rat Alzheimer's disease(AD) model. Method: The SD rats were randomly divided into seven groups, single dose normal saline(NS, 5 μL) was given via right hippocampus injection to sham group, single dose Aβ1-42(2 g·L-1) 5 μL was given via right hippocampus injection to model group and the treatment groups were orally given Yizhi capsules for one month. After 21 days of administration, Morris water maze was used to measure learning and memory performance.The rats of treatment groups were given Yizhi caspase for 10 days. After the watermaze experiment, biochemical detection choline acetyltransferase (ChAT) and acetylcholinesterase (AchE) activity were measured.The protein expressions of Aβ,P-tau(S262) were determined by Western blot method. Result: Compared with normal group, and in the total distance and escape latency sham group had no difference. Compared with sham group, in model group the total distance and escape latency were significantly prolonged, and differences was statistically significant(P<0.01). Compared with model group,in other groups the total distance and escape latency were significantly shorten(P<0.05,P<0.01), the AchE of enzymeactivity was significantly lower and increased ChAT enzyme activity,the phosphorylation of Aβ1-42and Tau at Ser262 were decreased in other groups. Conclusion: Yizhi capsule can attenuate the learning and memory impairment induced by Aβ1-42, which maybe related to increasing the cholinergic system of the central nervous system and suppressing Tau protein.

      摘要:Objective: To investigate the effects of Gegen Qinlian Tang-containing serum (GGQLT-CS) on glycometabolism in insulin-resistance HepG2 cell line. Method: GGQLT-CS was prepared.MTT was used to detect the effects of GGQLT-CS on cell proliferation;the model of insulin resistance was established,the effect of GGQLT-CS on the glucose consumption of HepG2 cells was observed in insulin resistance through the method of glucose oxidase, detecting phosphoenolpyruvate carboxykinase (PEPCK) activity was detected with the method of the lactate dehydrogenase coupled assay, and the anthrone method was used to detect intracellular glycogen content. Result: GGQLT-CS could promote glucose consumption of HepG2 cell in insulin resistance, reduce PEPCK activity, and increase hepatic glycogen content. Conclusion: GGQLT-CS can regulate glucose metabolism of HepG2 cells, and improvehepatic insulin resistance.

      摘要:Objective: To study the effect of berberine(Ber) on plaque area quantification, visfatin and inflammatory cytokines expression in ApoE knockout (ApoE-/-) mice. Method: Fifty male 8-week-old ApoE-/- mice were fed with a high fat diet and randomly assigned into five groups:model group, low-dose Ber(2.5 mg·kg-1·d-1) group, medium-dose Ber(5 mg·kg-1·d-1) group, high-dose Ber(7.5 mg·kg-1·d-1) group and simvastatin (5 mg·kg-1·d-1) group. Ten C57BL/6J mice fed with a common diet were served as control group. Serum lipids and visfatin, plasma interleukin-6(IL-6) and tumor necrosis factor-α (TNF-α) levels were determined after 16 weeks. Atherosclerotic lesions in the aortic root were examined by H&E staining. Visfatin expression in the aorta and distribution in atherosclerotic plaques were identified by Western blotting and by immunohistochemistry. Result: The add weight value, serum visfatin, triglyceride(TG), total cholesterol(TC) and low densty lipoprotein chdesterol(LDL-C), plasma IL-6 and TNF-α, visfatin expression in the aorta of model group were statistically increased compared with control group and each treatment group (P<0.05). Atherosclerosis plaque lesions in aorta of simvastatin group, high-dose Ber group and medium-dose Ber group were statistically decreased compared with model group (P<0.05). Conclusion: Ber can inhibit the development of atherosclerosis by reducing serum lipids, atherosclerosis plaque, plasma IL-6 and TNF-α levels in ApoE-/- mice, the mechanism is related to down-regulating visfatin expression.

      摘要:Objective: To investigate the effect of paeonol on ischemic arrhythmia and the expression of miRNA-1 of rats. Method: SPF SD rats were randomly divided into the normal group, the model group, the positive group (propranolol hydrochloride injection 2 mL·kg-1) and the paeonol group (6, 3, 1 mg·kg-1). The ischemic arrhythmia models were induced by injecting 5 μg·kg-1dose of aconitine intravenously. ECG changes of each group were recorded, premature beat (VPB) frequency and time for the first time, and ventricular tachycardia (VT) and ventricular fibrillation (VF) frequency and duration were recorded. Hearts were taken out on the end of the experiment, myocardial infarct sizes were measured, and the expression of miRNA-1 in myocardial cells was measured by RT-qPCR method. Result: Compared with the normal group, the ischemic arrhythmia was occurred, the frequencies of VPB, VT and VF increased, the VPB time for the first time shortened, the VT and VF durations increased, myocardial infarct sizes and miRNA-1 expression increased in the model group (P<0.05, P<0.01).The above indicators had good improvement in the paeonol group(P<0.05,P<0.01). Conclusion: Paeonol has an obvious effect in treating ischemic arrhythmias.

      摘要:Objective: To investigate the regulatory effect on nuclear factor-κB(NF-κB) signaling pathways of Shenxin Maihe particles based on treating coronary heart disease (CHD) in ischemia-reper fusion injury model rats. Method: Sixty SD rats were randomly divided into the sham operation group, the model group, the diltiazem group (36 mg·kg-1), the low-, moderate- and high-dose Shenxin Maihe particles groups (0.17, 0.51, 1.53 g·kg-1). All rats received the corresponding medicines by intragastric adiminstration once daily for 7 days. Contents of interleukin (IL) -6 and IL-8 in the rats serum were detected by ELISA, the mRNA expressions of NF-κB, tumor necrosis factor-α(TNF-α), inducible nitric oxide synthase (iNOS), intercellular surface adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecules-1 (VCAM-1) in the tissue of rats were detected by RT-PCR, the protein expressions of vascular endothelial growth factor (VEGF), heat shock protein 70 (HSP 70) and cyclooxygenase-2 (COX-2) were detected by Western blotting. Result: Compared with the sham operation group, the levels of IL-6 and IL-8 increased, the mRNA expressions of NF-κB, TNF-α,iNOS, ICAM-1 and VCAM-1 increased, the protein expression of COX-2 increased, the protein expressions of VEGF and HSP 70 decreased in the model group (P<0.01). Compared to the model group, Shenxin Maihe particles could improve the above indicators significantly (P<0.01). Conclusion: Shenxin Maihe particles has an evident effect against myocardial ischemia injury. Its mechanism may be related to the inhibition of inflammatory cytokines in the NF-κB signaling pathways and activating blood vessel protection factor synergistically. The results provide relevant theory basis for popularization and application of Shenxin Maihe particles in treating CHD.

      摘要:Objective: To study the protective effect and mechanisms of Sini Tang (SNT) and its components compatibility on chronic heart failure (CHF) induced by doxorubicin in rats. Method: The CHF rat model was induced by intraperitoneal injection of doxorubicin (ADR, 2.5 mg·kg-1) once a week for 6 weeks. The successful model rats were randomly divided into 6 groups:the model group, the Aconiti Lateralis Radix Praeparata group (ALRP, 1.5 g·kg-1), the ALRP-Glycyrrhizae Radix et Rhizoma group (ALRP-GRR, 3 g·kg-1), the ALRP-Zingiberis Rhizoma group (ALRP-ZR, 2.5 g·kg-1), the SNT group (4 g·kg-1) and the captopril group (6.25 mg·kg-1). Meanwhile, another 10 normal rats were assigned to the normal group. The rats in the treatment groups receive the corresponding medicines by intragastric administration for 4 weeks and the rats in the normal and the model group received the same volume of distilled water. After the last administration, left ventricular weight index (LVWI) and heart weight index (HWI) were measured. The pathological changes in myocardial muscle of rats were observed using HE staining. B-type natriretic peptide (BNP), angiotensin Ⅱ (angⅡ) and aldosterone (ALD) were assayed by ELISA. Result: Compared with the normal group, the LVWI and HWI increased obviously (P<0.05), the BNP, AngⅡ and ALD levels were significantly higher (P<0.01) in the model group. Compared with the model group, HWI was reduced in the SNT group (P<0.05), the BNP, AngⅡ and ALD level were lower in all treatment groups (P<0.05). Furthermore, the results of SNT were better than ALRP in reducing the level of BNP and ALD in serum. Conclusion: SNT and its components compatibility have protective effect on CHF induced by ADR in rats. The mechanism may be related to attenuating cardiac remodeling and inhibiting the excessive activation of neuroendocrine factor. SNT has obvious effect of anti-heart failure. ALRP-ZR group could partly improve the cardiac function of CHF. ALRP-GRR group has superior cardiotonic effect to the application of ALRP alone.

      摘要:Objective: To study the aglycone from Rhei Radix et Rhizoma(AR) on liver microsomal P450 isozymes/cytochrome P450 (CYP450) CYP1A2 activity in rats by enzyme assays. Method: Sixty SD rats were randomly divided into the control group, the phenobarbital sodium (PB) group (75 mg·kg-1 0.5% sodium carboxyl methyl cellulose, CMC-Na), the dexamethasone (DEX) group (80 mg·kg-1), the beta-naphthoflavone(β-NF) group (75 mg·kg-1), the ketoconazole(KET) group (82.5 mg·kg-1) and the different doses of AR groups (7.08, 14.40, 30.82, 46.55, 51.63, 61.63, 100.07 mg·kg-1). The experiment was performed in the liver microsome and the phenacetin was taken as the probe drug. The acetaminophen concentrations were determined by HPLC and the relationship between CYP1A2 activity and the dosage was conducted. Result: Compared with CMC-Na group and the KET group, the production of acetaminophen was very different in the AR groups at 61.63, 30.82, 14.40 mg·kg-1 (P<0.05). The induce effect was much weaker than three kinds of induction agents, and it was enhanced with the increasing of the dosage. Conclusion: The AR could induce the activity of CYP1A2 in a dose-dependent manner.

      摘要:Objective: To observe the effect of Guanxin V mixture on acute myocardial infarction (AMI) after the ventricular remodeling, and investigate the mechanism of Guanxin V improving ventricular remodeling. Method: The model of acute myocardial infarction was established by left anterior descending coronary artery ligation. All of the rats were treated with Guanxin V mixture and Xuezhikang capsule for 4 weeks,The heart color measurement of ejection fractions (EF) value, fractional shortening (FS) value, calculate the left ventricular mass index and left ventricular surface area were observed,Western blot technique was used to detect the expression of Toll like receptor 4 (TLR4), myeloid differentiation factor 88(MyD88), nuclear factor-kappa B/p65 (NF-κB/p65) proteins. Result: Compared with the normal group and sham operation group,the EF,FS and expression of TLR4,MyD88,NF-κB/p65 in model rats were up-regulated, left ventricular mass index(LVMI) was decreased,the difference was statistically significant(P<0.05),Compared with the model group,the EF,FS and expression of TLR4,MyD88,NF-κB/p65 in model rats were decreased,LVMI was increased,the difference was statistically significant(P<0.05).Expression of Guanxin V mixture was inversely proportional to the concentration of protein,the difference was statistically significant(P<0.05). Conclusion: Guanxin V mixture can inhibit TLR4/MyD88/NF-κB p65 signal pathway,this may be one of the mechanisms of improving ventricular remodeling.

      摘要:Objective: To study the effects of the total alkali Sophora alopecuroids(TASa) on gastro antrum smooth muscle strips in diabetic gastroparesis rats and the possible mechanisms. Method: Forty wister rats were randomly divided into the control group(10 rats) and model group(30 rats).Rats in the model group were given intraperitoneal injection of streptozotoc to induce type I diabetic model. Weight, concentration of blood glucose, residual rate of gastric pigment and intestinal conduction velocity were detected. The isolated perfusion method was used to record amplitude, duration, area and contraction frequency of the gastro antrum smooth muscle in vitro. The strips of model group were divided into six groups:TASa group, TASa+1×10-7 mg·L-1 verapamil group, TASa+1×10-6 mg·L-1 phentolamine group, TASa+1×10-6 mg·L-1 benzhydramine group, TASa+1×10-6 mg·L-1 indomethacin group, TASa+1×10-6 mg·L-1 atropine group. The contraction effects of TASa were investigated. Result: TASa could increase gastro antrum smooth muscle's contraction amplitude, contraction duration and contraction area, had no effect on contraction frequency. Verapamil could inhibit the gastro antrum smooth muscle contraction of TASa, but atropine, phetolamine, benzhydramine and indomethacin had no contraction effect. Conclusion: TASa may activate calcium channel, increase calcium flow to the interior of the cell and increase gastro antrum smooth muscle's contraction.

      摘要:Objective: To study the effect of Yaobitong capsule(YBT)on inflammatory cytokines and pain in the rat lumbar disc herniation model (RLDHM). Method: The model of lumbar disc herniation(LDH) was established by compressing silica gel pieces on the L5 nerve root of rat. All the rats were randomized into model group,positive group(Yaotongning capsules,0.16 g·kg-1)and YBT treatment groups(0.17,0.34,0.68 g·kg-1). After administration for 30 d,the mechanical pain thresholds of left hind limb,serum tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6)levels and pathological change in the nerve root of rats were detected respectively. Result: The model of LDH in rats was successfully established. Compared with normal control group,the mechanical pain thresholds of the model rats were decreased significantly(P<0.05),the serum TNF-α,IL-1β,IL-6 levels were increased significantly(P<0.01),and the obvious lesion was observed in nerve root. In the 0.34, 0.68 g·kg-1 dose of YBT groups,the mechanical pain thresholds were increased significantly,the serum TNF-α,IL-1β,IL-6 levels reduced significantly(P<0.05,P<0.01),and the pathological change was improved obviously in the nerve root. Conclusion: YBT could improve the symptoms and nerve root lesions in rats with LDH,which may be related with inhibition of inflammatory cytokines.

      摘要:Objective: To observe the effect of New Bitongling (NBTL) on collagen-induced arthritis (CIA) rats and to investigate its influence on the expression of high mobility group box 1(HMGB1) mRNA in the synovial tissues. Method: NBTL extractum was made by a series of processing technology. 12 of 72 SD rats were assigned to the normal group. The CIA models were induced in other rats, then they were divided into the model group, the low-, moderate-, high-dose NBTL groups (3.2,1.6,0.8 g·kg-1·d-1) and tripterygium wilfordii Hook F (twHF) group (0.008 g·kg-1·d-1). All rats received the corresponding drugs at the day of secondary immune and continuously dosed for 21 days by intragastric administration. The joint volumes were measured, the pathological tissues were observed and HMGB1mRNA in the synovial tissues were detected by quantitative real time PCR (RTq-PCR). Result: Compared with the normal group, the joints swelling was more obvious (P<0.01) and the expression of HMGB1mRNA was higher in the model group (P<0.01). Compared with the model group, the joints swelling was improved at the 14th day (P<0.05) and the 21th day (P<0.01) and the expression of HMGB1mRNA in the synovial tissues decreased (P<0.01) in the NBTL group at high dose. The pathological section of joints showed the lesions were improved in the all doses of NBTL and twHF groups as compared with the model group. Conclusion: NBTL could reduce joints swelling of CIA rats and inhibite the expression of HMGB1mRNA in the synovial tissues. The mechanism for therapy on rheumatoid arthritis of NBTL may be related to inhibiting the activation of inflammatory pathway through decreasing HMGB.

      摘要:Objective: To investigate the effect of Mori Folium effective parts (total polysaccharides, flavonoids and total alkaloids) on high insulin-induced insulin resistance of human hepG2 cells by nuclear factor-κB(NF-κB) pathway. Method: The insulin resistance cell model was induced by insulin stimulated human HepG2 cells for 48 h. The experimental groups were the control group, the model group (10 μg·mL-1 insulin), total polysaccharide (MFP) group, total flavonoid (MFF) group, the total alkaloid group (MFA), parthenolide (PTL) group. Residual glucose content of liquid culture was detected using glucose oxidase-peroxidase method (GOD-POD). Expressions of NF-κB, I-κB kinase β(IκKβ) and inhibitor κB protein α(IκBα) mRNA were detected using RT-qPCR. The relative expressions of protein NF-κB, IκKβ and IκBαwere detected using Western blot. Result: The optimal doses against insulin resistance for MFP, MFF, MFA and PTL were 200, 100, 10, 20 μmol·L-1, respectively. After insulin stimulation, NF-κB, IκKβ activity were enhanced significantly (P<0.01), NF-κB binding protein IκBα decreased significantly (P<0.01), which indicated that NF-κB pathway had been activated. After intervention of the mulberry effective parts, only flavonoids could lower NF-κB, IκKβ activity significantly (P<0.05), inhibite the degradation of IκBα (P<0.05). The results was not significant in polysaccharide group and alkaloid group. Conclusion: After high insulin-induced insulin resistance in HepG2 cells, NF-κB pathway was activated to some extent, suggesting that there is a certain relationship between insulin resistance and NF-κB inflammatory pathways. Meanwhile, mulberry flavonoids could improve the insulin resistance by NF-κB pathway.

      摘要:Objective: To study chemical compositions changes of different extracting parts of Jiuwei Tongqiao decoctions by gradient extraction with four kinds of organic solvents,screen effective extracting parts by in vitro pharmacodynamic,select the best effective parts on proliferation effect of anti-glioma cells,and provide experimental basis for further study on mechanism of Jiuwei Tongqiao decoctions. Method: After alcohol extract enrichment of Jiuwei Tongqiao decoctions,gradient extraction with petroleum ether,chloroform,ethyl acetate and water saturated n-butyl alcohol was to get petroleum ether extract(part Ⅰ),chloroform extract(part Ⅱ),ethyl acetate extract(part Ⅲ),water saturated n-butyl alcohol extract(part Ⅳ) and the residual liquid water extract(part Ⅴ).HPLC was employed to determined four index components,including calycosin-7-O-β-D-glucoside,formononetin-7-O-β-D-glucoside,calycosin and formononetin,mobile phase was consisted of (acetonitrile+0.05% formic acid)(A)-(water+0.05% formic acid)(B) for gradient elution (0-50 min,5%-55%A;50-70 min,55%-60%A;70-90 min, 60%-85%A;90-100 min,85%-100%A) and detection wavelength was 254 nm.U251 cells of anthropogenic brain glioma was selected for tracking,MTT was applied to determine effect of extraction parts for cell survival. Result: The content ratio of calycosin glucoside-ononin-calycosin in part Ⅲ was 1.0: 10.8: 8.2,its half inhibition rate(IC50) was 46.89 g·L-1.In comparison with other parts,index ingredients in part Ⅲ were more comprehensive and concentration of them were higher,inhibitory effect on U251 cells was more obvious and appeared a certain concentration-dependent manner. Conclusion: Part Ⅲ is the most effective parts in Jiuwei Tongqiao decoctions for anti-proliferative effect of U251 glioma cells.

      摘要:Objective: To investigate the effects of theaflavin aganist ischemic brain injuries in rats. Method: Sixty-four SD rats were randomly divided into 4 groups as following:sham operation group (Sham), model group (Model), theaflavin low and high dose group, which was administrated with theaflavin 20, 40 mg·kg-1 respectively. Rats of Sham and Model group were administrated with equal-volume normal saline. Rats were treated with continued 7 days, and then ischemic brain injuries in rats were reproduced by middle cerebral artery occlusion(MCAO). After operation 24 h, rats were observed behavior and neurological behavior score by Longa method. Then, water content of brain tissue in rats was calculated in 6 rats selected from each group. Finally, another 10 rats in each group were determined superoxide dismutase(SOD), malondialdehyde(MDA) and nitric oxide(NO) level in ischemic brain tissue by colorimetric methods. Result: Compared with Sham group, there were obvious behavioral disorders displayed in Model group, simultaneously water content of brain tissue, MDA and NO content were elevated, however, SOD activity was reduced. Compared with Model group, behavioral score and SOD activity in theaflavins groups were significantly ameliorated, yet water contents of brain tissue and MDA content were attenuated significantly. Besides, NO content was elevated significantly in theaflavins high dose group. Conclusion: Theaflavins can provent against ischemic brain injuries in rats.

      摘要:Objective: To analyze the toxicity source of Curculiginis Rhizoma and Epimedii Folium and to study the correlation between their toxicity and the content of curculigoside and icariin with several compatible prescriptions. Method: Under the total dosage being constant, several compatible prescriptions were obtained by uniform design with Curculiginis Rhizoma and Epimedii Folium classical proportion 1: 1 as a reference. Their extracts were obtained being by water and alcohol. Their toxicity was compared in mice. The contents of curculigoside and icariin in each compatible prescriptions were determineby HPLC, and the correlation between toxicity and content were analyzed. Result: The acute toxicity sequence is alcohol extract of Curculiginis Rhizoma-Epimedii Folium(1: 0)>alcohol extract of Curculiginis Rhizoma-Epimedii Folium(2: 1)>alcohol extract of Curculiginis Rhizoma-Epimedii Folium (1: 1)>alcohol extract of Curculiginis Rhizoma-Epimedii Folium (1: 2)>alcohol extract of Curculiginis Rhizoma-Epimedii Folium (0: 1)>water extract group. There was significantly positive correlation between the content of curculigoside with their toxicity(r=0.967, P<0.01). No correlation was found between the content of Icariin and their toxicity (r=-0.053, P>0.05). Conclusion: Toxic components of Curculiginis Rhizoma and Epimedii Folium mainly come from Curculiginis Rhizoma , and they can be easily enriched in alcohol extracts. There is positive correlation between the toxicity and the content of curculigoside.

      摘要:Objective: The aim of this article was to explore the acute toxicity of water extracts and alcohol extracts from stalks and leaves of Coix on mice so as to determine the median lethal dose (LD50) and the maximum tolerated dose (MTD). Method: Seventy-four KM mice (half male and half female) of grade SPF were divided into extract group and control group. After fasted for 16 hours before experiment, all the mice were fed by gavage. The toxic reaction and death were oberved. If the mice died, it will be dissected to observe the changes of heart, liver, spleen, lung, kidney, brain, stomach, small intestine and other organs by eyes.After the observation for 14 days, the survivals in each group were killed, and then dissected. Then we examined their main pathological changes in organs by eyes as mentioned above. Result: No obvious symptoms of poisoning were discovered on these mice, neither death. Conclusion: The maximum tolera dose of SPF KM mice to the water extracts and the alcohol extracts from leaves of Coix, and to the water extract and alcohol extracts from stalks of Coix, were 42.08,34.80,39.52, 32.64 g·kg-1, respectively, with all greater than 5 g·kg-1. According to the quarter experiment, the water and alcohol extracts from leaves and stalks of Coix are nontoxic.

      摘要:Objective: The aim of this study was to investigate the clinical effect of Jiannao Bushen pill combined with nimodipine on cognitive function and hemorheology index in elderly patients with vascular dementia (VD) and provide clinical basis for VD therapy. Method: From 2009/1 to 2013/1, we collected 90 VD cases in our hospital and divided them into 2 groups randomly, 45 cases in control group and 45 cases in observation group. The control group was treated with conventional treatment and nimodipine, while the observation group was treated with conventional therapy, and Jiannao Bushen pill combined with nimodipine. Clinical effects, the mini mental state examination(MMSE) score, hemorheology index, and the levels of inflammatory factors were observed and compared. Result: The observation group had a therapeutic efficiency ratio (TER) of 95.6.0% which was significantly higher than the control group (77.8%)(P<0.05). The MMSE score, hemorheology index, and the levels of inflammatory factors of the both groups all significantly improved after treatments. However, those of the observation group were better improved than those of the control group after treatments (P<0.05). Conclusion: Jiannao Bushen pill combined with nimodipine for VD therapy is effective, which is worthy of study and further application in clinical treatment. Our study provided new evidence for clinical therapy of VD.

      摘要:Objective: To observe the curative effect of traditional Chinese medicine for fumigating -washing and enema combined with western medicine in treating hand-foot-mouth disease. Method: Two hundred and sixty patients with a clear diagnosis of hand-food-mouth disease were randomly divided into the observation group and the control group according to their ages by a random number table of 130 patients each. The patients in the control group received ribavirin intravenous drip (10 mg·kg-1) once daily and antibiotics when needed. On the basis of the control group, the patients in the observation group began to receive traditional Chinese medicine prescription of fumigating-washing treatment twice daily and enema treatment once daily within the first 24 hours of admission. The clinical efficacy, adverse reaction and complications of the two groups were observed. Result: The total effective rate in the observation group was 94.62%, which was obviously higher than 85.38% of the control group with statistically significant difference (P<0.05). The antifebrile time, erythra regression time and hospital stays in the observation group were shorter than those in the control group with statistically significant difference (P<0.05). Incidence of adverse reactions in the observation group was 4.62%, which was lower than 11.54% in the control group with statistically significant difference (P<0.05). Conclusion: Chinese medicine for fumigating-washing and enema combined with western medicine therapy is easy to accept and good adherence. Moreover, it has good absorption, short treatment course and takes effect quickly in treating hand-foot-mouth disease of children. It is worthy of clinical popularization and application.

      摘要:Objective: To observe the additional effect of Chinese drugs for activating blood circulation and removing blood stasis (CDABCRBS) on the stability of coronary atherosclerotic plaque based on the standardized treatment with statins. Method: Totally 64 patients with stable coronary heart disease (CHD) were randomly divided into two groups (32 in each group), the control group treated with statins, and the CDABCRBS group treated with compound tablet of red sage root based on statins treatment. After the 12-month treatment, the change of coronary artery stenosis rate, coronary artery calcium (CAC) scores and plaque number were observed by 64 row spiral CT. The serum lipid level and blood stasis syndrome (BSS) scores of the patients were also tested. Result: All patients had good improvment after treatment. Compared with the control group, the change of coronary artery stenosis rate decreased significantly (P<0.05), CAC scores and plaque number decreased obviously in CDABCRBS group (P<0.05). In addition, the CDABCRBS group had superior effects in reducing the concentration of total cholesterol, increasing the concentration of high density lipoprotein cholesterol, and decreasing the atherosclerosis index, as compared to the control group (P<0.05). BSS scores in both groups decreased significantly after treatment (P<0.05), while the improvement in the BSS scores showed no significant difference between two groups. Conclusion: Standardized statins combined with CDABCRBS therapy show better effect in atherosclerosis treatment and plaque stability in stable CHD patients, which reflect the advantage of integrated traditional Chinese and western medicines.

      摘要:Objective: To discuss the curative effects of Bupi Yishen decoction in treating Alzheimer's disease and to investigate its influence on oxidative stress. Method: Sixty-six patients were randomly divided into the control group (31 cases) and the observation group (35 cases). Patients in the control group received 10 mg donepezil hydrochloride tablets once daily. Patents in the observation group received 1 does Bupi Yishen decoction, once daily. All patients in two groups received 6 months of treatment. Scores of minimum mental state examination (MMSE), Alzheimer's disease assessment scale-cognitive subscale (ADAS-cog), activity of daily living scale (ADL) and traditional Chinese medicine syndrome were conducted. Levels of oxide dismutase (SOD), malondialdehyde (MDA), total antioxidant capacity (T-AOC), 8-heterogeneous prostaglandin F2α(8-iso-PGF2α) and oxidized low density lipoprotein (ox-LDL) were detected before and after the therapy. Result: The efficacy scores of MMSE and ADAS-cog in the observation group were higher than those in the control group (P<0.05). Scores of traditional Chinese medicine syndromes, ADL, ADAS-cog in the observation group were lower than those in the control group (P<0.01), while the MMSE score was higher than that in the control group (P<0.01). After treatment, levels of MDA, 8-iso-PGF2a and ox-LDL were lower, and levels of SOD and T-AOC were higher in the observation group than those in the control group (P<0.01). Conclusion: Bupi Yishen decoction could enhance cognitive function and life self-care ability in patients with mild to moderate AD. Its curative effects are superior to donepezil hydrochloride tablets. Its mechanism may be related to antioxidant and antagonizing free radical damage.

      摘要:Objective: To discuss the clinical effects of Gushen Shengfa pills combined Wuling capsules in treating alopecia areata disease with deficiency of liver and kidney symptoms and to investigate its regulatory effect on Th1/Th2 cell factor. Method: One hundred and twenty patients were randomly divided into the control group (60 cases) and the observation group (60 cases) by digital method. Patients in the two groups received minoxidil tincture for external use. Patients in the control group added 3 pieces of compound glycyrrhizin tablets and 2 pieces of compound cystine tablets thrice daily. Patients in the observation group added 2.5 g Gushen Shengfa pills and 3 pieces of Wuling capsules thrice daily. All patients received 3 months of treatment. Another 15 healthy subjects were assigned to the healthy group. Scores of hair loss area and position, light pull hair test and hair growth were conducted before and after therapy. Besides, Pittsburgh sleep quality index (PSQI) scores were conducted. Levels of serum interferon-γ (IFN-γ), interleukin-12 (IL-12), IL-4 and IL-10 were tested. Result: The curative effect in the observation group was superior to that in the control group (P<0.05). The main symptoms scores including hair loss area and position, light pull hair test and hair growth, and the signs and PSQI scores in the observation group were lower than those in the control group after treatment (P<0.01). Compared with the healthy group, levels of IFN-γ and IL-12 were higher, the IL-4 and IL-10 levels were lower in both the two groups before treatment (P<0.01). After therapy, IFN-γ and IL-12 levels decreased, IL-4 and IL-10 levels increased in the observation group, the results were better than those in the control group (P<0.01). Conclusion: Gushen Shengfa pills combined Wuling capsules could improve the quality of sleep, control the Th1/Th2 balance, promote the growth of hair. Its clinical effect is superior to the treatment of compound glycyrrhizin tablets combined with compound cystine tablets.

      摘要:Objective: To discuss the curative effects of Shenqi Jiangtang granules, Ginkgo leaf capsules jointed with mecobalamine and α-lipoic acid in treating diabetes peripheral neuropathy (DPN) and to investigate its influence on levels of oxidative stress indicators and serum cystatin C (Cys-C). Method: One hundred and four DPN patients were randomly divided into the Western medicine group (52 cases) and the combination group (52 cases), Patients in the Western medicine group took 0.5 mg mecobalamine tablets thrice daily and 0.6 g thioctic acid capsules once daily. On the basis of therapy in the Western medicine group, patients in the combination group added 2 g Shenqi Jiangtang granules and 2 pieces of Ginkgo leaf capsules thrice daily. All patients in the two groups received 3 months of treatment. Integral scores of symptoms and Toronto (Toronto clinical scoring system, TCSS) clinical grading were conducted before and after therapy. Motor conduction velocity (MNCV) and sensory conduction velocity (SNCV) of median nerve, ulnar nerve, the common peroneal nerve, anterior tibial nerve were detected. Levels of serum total antioxidant capacity (TAOC), superoxide dismutase (SOD), malondialdehyde (MDA) and Cys-C were tested before and after treatment. Result: The curative effects in the combination group were superior to those in the western medicine group (P<0.05). After therapy, clinical symptom integral and Toronto clinical scores in the combination group were lower than those in the western medicine group (P<0.01). MNCV and SNCV of median nerve, ulnar nerve, common peroneal nerve, anterior tibial nerve in the combination group had higher improvement as compared with those in the Western medicine group (P<0.01). Levels of SOD, MDA and Cys-C in the combination group were inferior to those in the western medicine group (P<0.01), while the TAOC level was higher than that in the Western medicine group (P<0.01). Conclusion: The curative effects of Shenqi Jiangtang granules, Ginkgo leaf capsules jointed with mecobalamine and α-lipoic acid therapy is superior to using Western medicine alone in treating DPN disease. Its mechanism may be related to relieving oxidative stress injury and reducing serum Cys-C level.

      摘要:Objective: To discuss the curative efficacy of modified Xiaoyi Zhiyang decoction in treating childhood atopic dermatitis (AD) and to investigate its influence on levels of peripheral blood interleukin IL-2, IL-4, IL-17 and interferon-γ. Method: Ninety patients with AD were randomly divided into the control group(45 cases) and the observation group (45 cases) by a random number table. Patients in the control group received mometasone furoate cream twice daily and once daily when the skin lesion was controlled, 10 mL levocetirizine oral liquid for children older than 6 years and 5 mL for children 4 to 5 years old once daily. Based on the treatment of the control group, patients in the observation group added modified Xiaoyi Zhiyang decoction. All patients in the two groups received 3 months of treatment. Before and after treatment, scores of quality of life index scale (DLQI) of SCORAD and skin disease were graded. Levels of IL-2, IL-4, IL-17 and IFN-γ were detected. Result: The curative efficacy in the observation group was superior to that in the control group (P<0.05). After treatment, scores of scope of skin lesions (A), lesion severity (B) and itch and sleeping degree (C) and the total scores of SCORAD in the observation group were lower than those in the control group (P<0.01). Score of DLQI was lower than that in the control group (P<0.01). Levels of IL-4 and IL-17 were lower than those in the control group (P<0.01), while levels of IL-2 and IFN-γ were higher than those in the control group (P<0.01). Conclusion: Xiaoyi Zhiyang decoction could improve the subjective and the objective symptoms, signs, enhance quality of life in AD patients with child's stage. Its curative effect is superior to using Western medicine treatment alone and the mechanism may be related to regulating Th1/Th2 and IL-17 cytokines. The therapy deserves to further study.

      摘要:Objective: To analyze the composing principles of the prescriptions given by professor HU Guo-jun for bronchial asthma using traditional Chinese medicine (TCM) inheritance support system (V1.1). Method: Prescriptions used for bronchial asthma cases were collected. Then the data was entered into the TCM inheritance support system. The principal analysis, revised mutual information, complex system entropy cluster and unsupervised hierarchical clustering were used to analyze composing principles. Result: Based on 322 cases of prescriptions, the frequency of each herb and association rules among herbs are computed, 52 core combinations and 9 new prescriptions are mined from the database. Conclusion: Herbal rules obtained from TCM inheritance support system are in accordance with the clinical experience by Professor HU Guo-jun on treating bronchial asthma, which testifies some new values obtained from software analysis on the other side. It is worthy of further notice for promoting the heritage of TCM.

      摘要:Objective: The aim of this study was to systematically summarize the syndrome differentiation and treatment thought of professor JIN Shi in treating chronic liver disease. Method: Medcase V3.2 storehouse and medical record national physician Maian data mining system was adopted to collect the typical cases of chronic liver disease treated by professor JIN Shi. Then frequency analysis and association rule analysis were used for data mining on symptoms, syndromes, pathogenesis and therapeutic principle. Result: There were 1 915 cases of chronic liver disease included, with chronic hepatitis B viral hepatitis the most disease (43.1%). Damp-heat-blood stasis-toxin and disorder of liver-spleen were the most frequent pathogenesis, accounting for 19.7% and 10.6% respectively;noxious heat stasis syndrome and damp-heat in liver-gallbladder syndrome were the most frequent syndrome, accounting for 22.6% and 19% respectively;regulating liver and activating spleen, clearing heat and damp, were the most common therapeutic principle, accounting for 12.2% and 8.2%, respectively. Conclusion: Noxious heat stasis syndrome and damp-heat in liver-gallbladder syndrome were the most frequent syndrome for chronic liver disease treated by professor JIN Shi, who valued the pathogenesis of damp-heat-blood stasis-toxin. The common therapies for chronic liver disease were regulating liver and activating spleen, and in the assistance of syndrome differentiation based adjuvant therapy, it can be applied to clear heat and damp, detoxify and disperse blood stasis, which can be generalized to clear, soothe, activate, dissolve and tonic.

      摘要:Objective: To simulate and explore the synergistic mechanisms between Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos of Danhong injection using computer systems biology approach. Method: The combined TCMGeneDIT database and Agilent literature search (ALS) system was used to dig the protein information of Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos. The protein interaction networks were respectively built based on the protein-protein interaction extracted from the BIND, BioGRID database. The differences and similarities of the networks were compared via difference and intersection module of the Merge programme. Result: There were 934 proteins contained in the common highly-connected protein network of Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos. Four subnetworks were extracted from the common highly-connected protein network with significant difference (P<0.05) using clusterONE. Gene ontology cluster analysis demonstrated the functions of common highly-connected protein network were associated with 4 biological processes, including RNA metabolic process, nuclear factor kappa B(NF-κB) cascade, lipid homeostasis, Rho protein, Rho protein and small GTPase mediated signal transduction. The 1 431 difference protein networks extracted from the difference module analysis between Danhong injection protein and common highly-connected protein network had the relationship with the cell proliferation, migration and autophagy. Conclusion: The computer system biology approach simulated and explored the synergistic mechanisms between Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos of Danhong injection. The mechanisms might be related to the RNA metabolic process, NF-κB cascade, cell proliferation, migration and autophagy.

      摘要:Objective: To evaluate the effectiveness and safety of salvianolate injection (SI) on acute cerebral infarction (ACI) versus conventional treatment (CT). Method: Systematic literature searches were performed in PubMed, Cochrane Library, CBM, CNKI and WanFang datbase from Jan 1994 to Apr 2014.Randomized controlled trials (RCTs) on SI treating ACI were enrolled. Two reviewers screened articles according to the inclusion and exclusion criteria, extracted data and assessed the quality of the included studies independently. The Meta-analysis was performed using RevMan 5.2. Result: A total of 12 RCTs involving 1 574 patients were included. The results of Meta-analysis demonstrated that SI had higher curative rate [OR=2.78,95%CI(1.87, 4.12), P<0.000 01] on 6 RCTs, improvement evaluation of NIHSS [OR=-2.05, 95%CI (-2.51,-1.59), P<0.000 01] on 5 RCTs, and amendment many endothelia parameters as compared with the CT group. No severe adverse reactions after SI treatment were reported. Conclusion: Current evidence shows SI may be superior to CT in treating ACI patients in recovery rate, vascular endothelial cell protection and improvement hemorheology index. However, due to lack of enough high-quality studies, SI has to be further studied by conducting more strictly-designed, multicenter, large-scale and double-blind RCTs.

      摘要:Diabetic nephropathy (DN) is one of the most common microvascular complications of diabetes mellitus (DM),which is also a primary causes of end-stage renal disease (ESRD).Its pathogenic factors and pathogenesis is very complicated with various reasons. In recent years, a large number of experimental studies have shown that flavonoids of Chinese herbal medicine have significant effect in preventing and treating DN. To better understand the progress of experiments in the prevention and treatment of DN by flavonoids, the paper searched the literature nearly 10 years, summarized in terms of flavonoids classification, experimental models, the role and mechanism. The study found:in the fight against DN, the most studied is the total flavonoids, there are okra factors, breviscapine, pueraria total flavonoids, fmcl Semen flavonoids, roem leaves, green thorn fruit flavonoids, guri geraniol total flavonoids, oxytropis falcate total flavonoids, but other flavonoids is relatively small. These studies are mainly animal experiments, basically using intraperitoneal injection of STZ models or high-fat and high-carbohydrate feeding rat model to made DN model, observed indicators in blood sugar, cholesterol, kidney function-based, molecular, cellular level indicator less. Through this study, we conclude that:how to develop herbal drug flavonoids medicine is the key issue, which is under the guidance of the theoretical system of Chinese medicine, combined with modern science and technology, multi-channel, multi-level and multi-level elaborate herbal flavonoids DN material basis of prevention efficacy, mechanism of action.
