      摘要:Objective: To compare in vitro permeation behavior differences between Qingfang Zhongtong external application powder and gel. Method: Taking transdermal diffusion instrument,with sinomenine as index component,abdominal skin of wistar rats containing cuticular as permeability medium,percutaneous penetration content of sinomenine in Qingfang Zhongtong external application powder and gel at seteing times were determined by HPLC,mobile phase was methanol-water-ethylene diamine (50:50:0.1),detection wavelength was 262 nm,in vitro permeation behavior of these two formulations was compared. Result: Cumulative penetration content of Qingfang Zhongtong external application powder and gel were 0.405 24,0.191 14 mg·cm-2 in 12 hours,respectively.Cumulative penetration percentage of them were 74.52% and 35.15%,respectively.Penetration behavior of sinomenine in these two formulations conformed to Higuchi equation,transdermal rate were 0.180 5,0.073 4 mg·cm-2·h-1,respectively. Conclusion: Both Qingfang Zhongtong external application powder and gel have sustained release effect.The gel releases drugs much more quickly at an early period,but during whole process of percutaneous penetration,external application powder can significantly improve both cumulative penetration content and transdermal rate of sinomenine.

      摘要:Objective: To establish a preparation technology of β-cyclodextrin (β-CD) inclusion of volatile oil from Piperis Fructus and to analysis of volatile components by GC-MS. Method: β-CD inclusion of volatile oil from Piperis Fructus was prepared by saturated aqueous solution method,with ratio of β-CD to volatile oil from Piperis Fructus,inclusion temperature and time as factors,comprehensive score of oil content in inclusion,inclusion rate of volatile oil and inclusion yield as index,inclusion conditions were optimized by uniform design.Inclusion was verified through microscopic examination and thermo-gravimetric analysis.GC-MS was employed to compare chemical compositions and percentages of samples which included volatile oil from Piperis Fructus,β-CD inclusion and redistilled volatile oil. Result: Optimum preparation conditions were as follows:β-CD to volatile oil ratio of 6:1,inclusion time of 4 h,inclusion temperature at 60 ℃.Oil content in inclusion,inclusion rate of volatile oil and inclusion yield were 11.56%,75.32% and 86.13%,respectively.These were 46,40 and 45 chemical components were identified from volatile oil,β-CD inclusion and redistilled volatile oil,respectively.But compositions in these samples were very similar,proportions of six main components in total contents of samples were more than 70%.There were some new chemicals appeared and some chemicals disappeared after inclusion and being redistilled,but the total content of these compounds was less than 1%,so that major components of each sample were almost the same. Conclusion: This inclusion process is simple and feasible for production and quality evaluation of solidified powder of volatile oil from Piperis Fructus.

      摘要:Objective: To compare contents of five kinds of oligosaccharide esters in different processed products of Polygalae Radix and to discuss its variation before and after processing. Method: Contents of sibiricose A5,sibiricose A6,glomeratose A,tenuifoliside B and 3,6'-disinapoyl sucrose in different processed products of Polygalae Radix were determined by HPLC,mobile phase of acetonitrile-0.1% formic acid solution in gradient program and detection wavelength of 330 nm. Result: Contents of five oligosaccharide esters in root bark of Polygalae Radix were all obviously higher than them in the core.Contents of these five ingredients in root bark were 0.173%,0.155%,0.071%,0.120%,0.648%,contents of them in the core were 0.090%,0.053%,0.029%,0.018%,0.205%,respectively.In honey-stir-baked products,contents of sibiricose A6 and glomeratose A were higher than them in the crude,but sibiricose A5,tenuifoliside B and 3,6'-disinapoyl sucrose were lower than them in the crude.In licorice juice boiled products,contents of sibiricose A5,sibiricose A6, tenuifoliside B and 3,6'-disinapoyl sucrose were obviously lower than them in the crude,but glomeratose A was higher than them in the crude. Conclusion: Contents of five oligosaccharide esters are quite different between root bark and the core of Polygalae Radix,processing has a certain influence on contents of oligosaccharide esters.Sibiricose A5,sibiricose A6, tenuifoliside B and 3,6'-disinapoyl sucrose in Polygalae Radix all have qualiy change after boiled with licorice juice.

      摘要:Objective: To optimize polymer materials and preparation process of puerarin sustained-release microspheres. Method: The microspheres were prepared by emulsion solvent evaporation method.Taking composite score of encapsulation efficiency,drug loading and yield as index,polymer materials and formulation process were optimized by orthogonal design.The microspheres were characterized by different techniques,such as scanning electron microscopy (SEM),thermogravimetric analysis (TGA),differential scanning calorimetry (DSC),X-ray diffraction (XRD). Result: Optimum polymer material of puerarin sustained-release microspheres was polylactic acid (PLA),optimum preparation parameters were as follows:dosage of 37.5 mg,PLA of 80 mg,polyvinyl alcohol (PVA) of 0.5%.Surface of puerarin sustained-release microspheres was smooth and round without adhesions,average particle size was 104.3 μm,drug loading was (26.20±2.24)%,encapsulation efficiency was (68.92±1.88)%,yield was (83.97±2.55)%.TGA,DSC and XRD were indicated changes in physical form of molecular spatial relationship of copolymer and puerarin in the microspheres. Conclusion: Puerarin sustained-release microspheres can be successfully prepared by emulsion solvent evaporation method,optimized process is simple and reasonable.

      摘要:Objective: To verify that acquisition time of Leontopodium franchetii was florescence and application parts were leaf and flower by investigating content differences of total phenolic acids in L. franchetii at different harvesting time. Method: UV-Vis spectrophotometry was adopted to determine the content of total phenolic acids with chlorogenic acid as standard,Folin-phenol as chromogenic agent and detection wavelength at 765 nm. Result: Linearity range of chlorogenic acid was 0-20 mg·L-1(R2=0.999 7),average recovery was 95.8% with RSD of 1.0%.The content of total phenolic acids from leaves of L. franchetii was up to 190.606 mg·g-1 in florescence.It showed that the content of total phenolic acids at florescence was significantly higher than other periods in L. franchetii,it in leaves and flowers were significantly higher than other plant parts. Conclusion: Total phenolic acids can be used as an evaluation index to confirm optimum acquisition time and effective parts of L. franchetii.It provides scientific basis for rational utilization and quality control of L. franchetii.

      摘要:Objective: To optimize bioconversion process of baicalin to baicalein in Scutellariae Radix by bacillus natto. Method: HPLC was employed to determine the content of baicalein with mobile phase of methanol (A)-acetonitrile (B)-0.2% phosphoric acid water solution (C) for gradient elution (0-28 min,20%-35%A,20%-35%B;28-32 min,35%-20%A,35%-20%B) and detection wavelength at 278 nm.Based on single factor tests,orthogonal test was adopted to optimize fermentation process by taking bacteria age,grinding degree,solid-liquid ratio and inoculation amount as factors. Result: Optimum fermentation process was as following:bacteria age of 12 h,solid-liquid ratio of 1:3,grinding degree of 40 mesh,inoculation amount of 30%,fermentation temperature of 37 ℃ and fermentation time of 7 d.Mass fraction of baicalein was 10.02% and conversion rate of baicalin was 96.5%. Conclusion: This optimized fermentation process is feasible and stable,baicalin in Scutellariae Radix basic transforms into baicalein by bacillus natto,it can upgrade medicinal value and physiological activity of Scutellariae Radix.

      摘要:Objective: To establish a method for determining dissolution of baicalin in Xiao'er Qingfei dispersible tablets and to provide a reference for quality control of this preparation. Method: HPLC was employed to determine the content of baicalin with mobile phase of methanol-water-phosphate (47:53:0.2) and detection wavelength of 280 nm.Taking dissolution of baicalin at the same time as index,single factor tests were adopted to investigate rotation rate,dissolution medium and method,difference of dissolution between Xiao'er Qingfei dispersible tablets and Xiao'er Qingfei tablets. Result: Baicalin had a good linear relationship in the range of 0.025-0.125 g·L-1 with r of 0.999 9 and average recovery of 100.41%.The paddle was adopted to determine dissolution of Xiao'er Qingfei dispersible tablets with pH 6.8 phosphate buffer as dissolution medium,rotation rate of 75 r·min-1 and sampling time of 40 min.Cumulative dissolution of Xiao'er Qingfei dispersible tablets was more than 70% in 15 min,while it was 40% for ordinary tablets. Conclusion: This method is reliable,reproducible and suitable for dissolution determination of baicalin in Xiao'er Qingfei dispersible tablets.

      摘要:Objective: To optimize extraction technology of atractylenolide I,Ⅱ,Ⅲ from Atractylodis Macrocephalae Rhizoma. Method: HPLC was adopted to determine contents of atractylenolide I,Ⅱ,Ⅲ with mobile phase of acetonitrile (A)-water (B) for gradient elution (0-16 min,60%-76%A;16-18 min,76%-100%A;18-30 min,100%A) and dual-wavelength detection of 220 nm for atractylenolide I,Ⅱ and 276 nm for atractylenolide Ⅱ.Taking extracting rates of atractylenolide I,Ⅱ,Ⅲ as indexs,extraction method was screened by single factor test,orthogonal test was adopted to optimize extraction process with extracting time,extracting times,ethanol concentration and amount as factors. Result: Alcohol warm infusion method was selected,optimum technological conditions for extracting thrice by adding 6,4,4 times the amount of 90% ethanol at 40 ℃,extracting time was 1.5,1.0,1.0 h in turn.Extracting rates of atractylenolide I,Ⅱ,Ⅲ were 92.62%,92.39%,93.06%,respectively. Conclusion: This optimized extraction process is stable and feasible with high extraction efficiency of atractylenolide I,Ⅱ,Ⅲ in Atractylodis Macrocephalae Rhizoma.

      摘要:Objective: To optimize acid enzymatic extraction process of soluble dietary fiber from Passiflora edulis peel and provide references for research and development of its functional food. Method: Based on single factor tests,with the concentration of citric acid solution,solid-liquid ratio,thermostable alpha amylase dosage,reaction temperature of complex enzyme as independent variables,extracting amount of soluble dietary fiber as dependent variable,Box-Behnken response surface methodology was used to optimize extraction conditions. Result: Optimum process conditions were as follows:solid-liquid ratio of 1:40,thermostable alpha amylase dosage of 5.0 U·g-1,the concentration of citric acid solution of 0.18% and reaction temperature of complex enzyme at 80 ℃.Yield of souble dietary fiber was 13.82%,whose relative error was -6.56% by comparing with the predictive value. Conclusion: Optimization of extraction process of souble dietary fiber from P. edulis peel by response surface methodology is accurate and reliable with some practical value.

      摘要:Objective: To optimize extraction process of four flavonoids in Scutellariae Radix and provide references for control extracts research of thin-layer chromatography of Scutellariae Radix. Method: Effects of ethanol concentration,solvent dosage and extracting time on overall desirability of yields of baicalin,wogonoside,baicalein and wogonin were investigated and forecasted by central composite design-response surface methodology.HPLC was adopted to determine contents of four flavonoids with mobile phase of acetonitrile-0.1% phosphoric acid solution and detection wavelength at 274 nm. Result: Optimal extraction process was as following:extracted twice with 12 times the amount of 54% ethanol for per time of 135 min.Yields of baicalin,wogonoside,baicalein and wogonin were 12.53%,2.77%,0.66%,0.25%,respectively.Bias between the predicted and observed value of overall desirability was 1.19%. Conclusion: This optimized extraction technology is stable and feasible with good prediction.

      摘要:Objective: To study the water-soluble chemical constituents from the stem bark of Magnolia officinalis. Method: The chemical constituents were isolated and purified by silica gel,ODS, Sephadex LH-20 and semi-preparative HPLC.Their structures were identified on the basis of ESI-MS,UV and NMR spectra. Result: Nine compounds were isolated and elucidated as (7S,8R)-syringoylglycerol(1),3,4-dihydroxy-allylbenzene-3-O-L-rhamnopyranosyl(1→2)β-D-glucopyranoside(2),3,4-dihydroxy-allylbenzene-3-O-L-rhamnopyranosyl(1→6)β-D-glucopyranoside(3)synapticaldehyde-4-O-β-D-glucopyranoside(4),syringaresinol-di-O-β-D-glucopynanoside(5),3,4,5-trimethoxyphenyl-β-D-glucopyranoside(6),magnolol(7),honokiol(8),4-O-methylhonokiol(9). Conclusion: Compounds 6 are isolated from this plant for the first time.

      摘要:Objective: To establish an UFLC-PDA method for the fingerprint of Pterocephali Herba, and compare the fingerprints of different medicinal parts. Method: The separation was performed on an Agilent Proshell 120 SB-C18 (4.6 mm×100 mm, 2.7 μm) using gradient elution with water-formic acid (100:0.2) and acetonitrile, at a flow rate of 1.0 mL·min-1. The column temperature was kept at 30 ℃, and detection wavelength was set at 238 nm. Result: The UFLC-PDA fingerprints of 17 batches were set up. The results showed that 15 chromatographic peaks were extracted as the common peaks of fingerprint of aboveground part, while in underground part peak 6 and peak 7 were nonexistent. 5 peaks were identified as chlorogenic acid, loganin, sweroside, evodia rutaecarpa glycosides and triplostoside A. The similarity degrees were above 0.9 excepted 6 batches of aboveground part. Conclusion: The method has good presicion, stability and repeatability that it could provide basis for quality control and evaluation of Pterocephali Herba

      摘要:Objective: An HPLC method for simultaneous determination of four components in Buyang Huanwu Tang extract was established. Method: The Buyang Huanwu Tang extract was separated on an Hyperil ODS2 column by gradient elution using acetonitrile(A)-0.1% phosphate (B) as the mobile phase. The flow rate was 1.0 mL·min-1. The detection wavelengths were 210 (amygdalin), 230(paeoniflorin), 250(campanulin), 315 nm (ferulic acid). Result: The linear ranges of amygdalin, paeoniflorin, campanulin and ferulic acid were 1.080-10.80(r=0.999 8), 1.735-17.35 (r=0.999 8), 0.242-2.42 (r=0.999 5), 0.106-1.06 (r=0.999 6) μg, respectively;the average recovery was 98.39% (RSD 1.9%), 98.89% (RSD 2.0%), 97.25% (RSD 3.3%), 95.39% (RSD 2.6%). Conclusion: The method is rapid and accurate for simultaneous determination of amygdalin, paeoniflorin, campanulin and ferulic acid in Buyang Huanwu Tang extract.

      摘要:Objective: To establish an HPLC method for content determination of magnoflorine, protopine, coptisine and chelidonine in Tong'an injection. Method: Gemini C18(4.6 mm×250 mm,5 μm)was used with the mobile phase of acetonitrile-0.02 mol·L-1 monopotassium phosphate at a flow rate of 1.0 mL·min-1. The detection wavelength was set at 230 nm for magnoflorine, 290 nm for protopine and chelidonine, 360 nm for coptisine, and the column temperature was maintained at 30 ℃. Result: Calibration curves of the magnoflorine, protopine, chelidonine and coptisine in Tong'an injection showed good linear relationship:10.856-108.560(r=0.999 8),24.712-247.120(r=0.999 6),3.464-34.640(r=0.999 6),39.296-392.960 mg·L-1 (r=0.999 9). The average recoveries of the components above were between 97.5%-101.8%. Conclusion: The method was feasible,accurate and stable,which could be used for determination of magnoflorine, protopine, coptisine and chelidonine in Tong'an injection.

      摘要:Objective: To study the changes of chemical constituentsin water extracts of couplet medicines Ephedrae Herba and Atractylodis Macrocephalae Rhizomain different compatibility ratio, and to search the compatibility rule of this couplet medicines. Method: The content of five alkaloids,norpseudoe phedrine, norephedrine, ephedrine, pseudoephedrineand methyle phedrine were determined by GC-MS.The content of atractylenolide Ⅰ, atractylenolide Ⅱ, and atractylenolide Ⅲ were determined by HPLC. Result: The three compatibility ratios of Ephedrae Herba and Atractylodis Macroce phalae Rhizoma were 3: 2,3: 4, and 3: 8.Compared with decoction of Ephedrae Herba,the content of five alkaloids were decreased (P <0.05)along with the increasein the proportion of Atractylodis Macrocephalae Rhizoma. And the content of atractylenolide Ⅰ, atractylenolide Ⅱ, atractylenolide Ⅲ were also decreased (P <0.01). Conclusion: Combined Ephedra Herba with Atractylodis Macrocephalae Rhizoma,the dissolution of compounds in water extracts was inhibited with each other.

      摘要:Objective: A method for rapid qualitative analysis of chemical constituents of Codonopsis pilosula by HPLC-LTQ-Orbitrap mass spectrometry was established. Mass information for different types of constituents was preliminarily approached. Method: A Phenomenex Kinetix C18(2.1 mm×100 mm, 1.7 μm) column and a gradient elution of MeCN-H2O (0.01% formic acid) were used for HPLC separation. The combination of Orbitrap mass analyzer with a linear ion trap was applied for high resolution mass spectrometry and collision-induced dissociation (CID). Result: HPLC provided high separation efficiency of chemical components of C. pilosula. Based on the high-resolution mass information and MS/MS fragmentation behaviors of chemical components, 19 compounds were identified or tentatively characterized. Conclusion: This work provided a good example for rapid analysis of different types of chemical constituents from C. pilosula. It might be helpful for further phytochemical studies and quality control of C. pilosula.

      摘要:Objective: To establish a method for determination of HPLC fingerprint of Stigma Maydis, provide reference, and basis for a quality standard of Stigma Maydis pieces. Method: Analysis was performed on an Agilent Zorbax SB-C18 (4.6 mm×250 mm, 5 μm), with the methanol-0.2% phosphoric acid as mobile phase. The gradient elution was carried out. The flow rate was 0.8 mL·min-1. The column temperature was 30 ℃. The injection volume was 10 μL. Result: HPLC fingerprint was established based on 24 batches of Stigma Maydiswere 18 common peaks, and two of them were identified asrutin(peak 11), apigenin(peak 15). Conclusion: Cluster analysis and principal component analysis should also participate in quality evaluation of Chinese herbal medicine, the HPLC fingerprin provides a reference for the quality standards for Stigma Maydis.

      摘要:Objective: To analyze and compare the volatile constituents of fruit of Eucalyptus globulus collected from Guizhou, Yunnan and Guangxi provinces, so as to provide an experimental reference for the further research and utilization of them. Method: The volatile oil was extracted by steam distillation with n-hexane as entrainer, and analyzed by GC-MS. Result: 27, 24, 17 compounds were identified from the fruit of E. globulus collected in Guizhou, Yunnan and Guangxi, respectively. There were 15 common compounds. 1, 8-cineole was verified to be the main component (Guizhou 34.861%, Yunnan 35.947%, Guangxi 35.704%). The other same components (relative content >5%) were as follows:(+)-aromadendrene, α-phellandrene, α-pinene and (-)-globulol. Conclusion: Volatile components of the fruit of E. globulus from the three areas were similar, but kinds and the contents were slightly different.

      摘要:Objective: To establish the quality standard for boiled powder of Maxing Erchen Tang. Method: The qualitative identifications of honey-fried Ephedrae Herba, Armeniacae Semen Amarum, Belamcandae Rhizoma and Citri Reticulatae Pericarpium were carried out by TLC, and the contents of belamcandin and hesperidin were determined by HPLC. Result: The results of TLC showed that the relevant spots were clear without interference in the blank reference. The liner ranges of belamcandin and hesperidin were 12.0-120.0 mg·L-1 (r=0.999 8) and 6.6-66.0 mg·L-1 (r=0.999 9), respectively. The average recovery of belamcandin was 100.00% with RSD of 1.9%. The average recovery of hesperidin was 98.16% with RSD of 1.8%. Conclusion: The established qualitative and quantitative methods are simple and accurate, reliable and reproducible, and can be effectively used for the quality control of boiled powder of Maxing Erchen Tang.

      摘要:Objective: To investigate the chemical constituents in the extract of Zhike Chuanbei Pipa dropping pills. Method: The compounds were isolated from the extract of Zhike Chuanbei Pipa dropping pills by a combination of various chromatographic techniques including column chromatography over silica gel, reversed-phase silica gel, sephadex LH-20 columns, and preparative HPLC. Their structures were identified by spectroscopic analysis including MS and NMR data. Result: Twenty-four compoundswere isolated and identified as nerolidol-3-O-{α-L-rhamnopyranosyl-(l→4)-α-L-rhamnopyranosyl-(l→2)-[α-L-rhamnopyranosyl-(l→6)]-β-D-glucopyranoside} (1), nerolidol-3-O-α-L-rhamnopyranosyl-(l→4)-α-L-rhamnopyranosyl-(l→6)-β-D-glucopyranoside (2), nerolidol-3-O-{α-L-rhamnopyranosyl-(l→4)-α-L-rhamnopyranosyl-(l→2)-[4-trans-feruloyl-α-L-rhamnopyranosyl-(l→6)]-β-D-glucopyranoside} (3), (6S,7E,9R)-6,9-dihydroxy-4,7-megastigmadien-3-one 9-O-β-D-apiofuranosyl-(l→6)-β-D-glucopyranoside (4), 4-caffeoylquinic acid (5), chlorogenic acid (6), (6S,7E,9R)-9-hydroxy-4,7-megastigmadien-3-one 9-O-β-D-glucopyranoside (7), eriobotrosideⅡ(8), 3,4-dihydroxy-benzoic acid (9), (6R,7E,9R)-9-hydroxy-4,7-megastigmadien-3-one 9-O-β-D-glucopyranoside (10), (+) lyoniresinol 9'-O-β-D-xylopyranoside (11), (-) lyoniresinol 9'-O-β-D-xylopyranoside (12), fumaric acid (13), 2,6-dimethoxy-4-(2-propenyl)phenol 1-O-β-D-glucopyranoside (14), (+)lyoniresinol (15), kaempferol (16), 4-hydroxy phenol (17), 4-hydroxy benzoic acid (18), 4-hydroxy cinnamic acid (19), (6S,7E,9R)-6,9-dihydroxy-4,7-megastigmadien-3-one (20), euscaphic acid (21), ursolic acid (22), 3-furancarboxylic acid (23), vanillic acid (24). Conclusion: Among the isolated twenty-four compounds, there are eight sesquiterpenoids (1, 2, 3, 4, 7, 8, 10, 20), four phenylpropanoids (11, 12, 14, 15), nine organic acids (5, 6, 9, 13, 17, 18, 19, 23, 24), two triterpenoids (21, 22), one flavonoid (16). These compounds were obtained from the extract of Zhike Chuanbei Pipa dropping pills for the first time.

      摘要:Objective: To explore a new method for identifying Fallopia multiflorum from its closely related species and adulterants by internal transcribed spacer 2 (ITS2) regions. Method: ITS2 regions of F. multiflora and its closely related species,adulterants were amplified,purified and sequenced.Sequences were assembled by Seqman and analysed by MEGA6.0,neighbor-joining (NJ) phylogenetic tree was constructed based on Kimura 2-parameter (K2P) model. Result: ITS2 regions of F. multiflorumexhibited obvious differences with its closely related species and adulterants.Intraspecific variation was less than interspecific genetic distance.All samples of F. multiflorum showed monophyletic on NJ tree,they could be distinguished from its closely related species and adulterants. Conclusion: ITS2 can be used as a effective marker for identifying F. multiflorum from its closely related species and adulterants,it can provide technical support for drug safety of F. multiflorum.

      摘要:Objective: To investigate inflorescence size on flower and seed quality of Carthami Flos for providing a reference for its standardized cultivation. Method: Sampling survey method was adopted to establish level of inflorescence size specifications of Carthami Flos in Henan,HPLC was employed to determine the content of hydroxysafflor yellow A in Carthami Flos with different grades of inflorescence,GC was used to determine the content of linoleic acid in seed of Carthami Flos with different grades of inflorescence. Result: Contents of hydroxysafflor yellow A in Carthami Flos with grade of I,Ⅱ,Ⅲ,Ⅳ were 2.90,2.85,2.97,2.87 mg·g-1,these four data were no significant difference.Percentage of inflorescence with grade of I,Ⅱ,Ⅲ,Ⅳ were 19.32%,37.16%,21.16%,16.36%.Inflorescence size had significant effects on thousand-seed weight,shell-kernel ratio,oil yield and the content of linoleic acid.The larger of inflorescence size had a higher thousand-seed weight and shell-kernel ratio,but oil yield was relatively low. Conclusion: Improving proportion of large inflorescence can get a better quality of flower and seed of Carthami Flos in production,inflorescence size do not have significant effect on the content of hydroxysafflor yellow A in Carthami Flos.

      摘要:Objective: In this study, we investigated the impact of combining Kansui Radix with Glycyrrhizae Radix et Rhizoma(1: 4) on the kidney in rats. On this basis, the mechanism of the reducing toxicity was also studied. Method: Male Sprague-Dawley (SD) rats were randomly assign to four groups, which were controls, Kansui Radix, Glycyrrhizae Radix et Rhizoma and Kansui Radix-Glycyrrhizae Radix et Rhizoma(1: 4).The kidney extract metabonomic analyses of Kansui Radix, Glycyrrhizae Radix et Rhizoma and Kansui Radix-Glycyrrhizae Radix et Rhizoma(1: 4) was used by NMR and OPLS-DA. Clinical biochemical and histopathological assessments were also performed. Result: All the clinical biochemical, histopathological and metabolites results were no significant change in Glycyrrhizae Radix et Rhizoma treated rats. Not only the increased glucose and decreased creatine/creatinine in plasma, but also the renal tubular epithelial cell swelling and vacuoles degeneration, glomerular blood clot were observed in Kansui Radix treated rats. Besides, decreased choline, phosphorylcholine, phenylalanine and glutamine, increased isoleucine, leucine, alanine, valine, acetic acid, creatine/creatinine was observed in Kansui Radix treated rats. In Kansui Radix-Glycyrrhizae Radix et Rhizoma(1: 4)treatment group, there was no significant change in plasma index. The histopathological changes was only the glomerular congestion and renal tubular epithelial cells expansion. The variations were only declined of choline, phosphorylcholine and phenylalanine in Kansui Radix. Conclusion: Detoxification of Kansui Radix in kidney by combining with Glycyrrhizae Radix et Rhizoma was obvious. It could be owing to the intervention of the biomarker of Kansui Radix and down regulation of the traditional indicator of kidney lesion.

      摘要:Objective: The aim of this study was to investigate the metabolites and pathways of salvianolic acid B(SAB) in rats. Method: In this study, 12 SD-rats were divided into 2 groups, with 6 rats used for collecting the blank plasma, bile, urine excretion samples, the others used for collecting the samples after intravenous administration of SAB(100 mg·kg-1). UPLC-Q-TOF method was used to implement the qualitative analysis and identification of structure of the metabolites in each sample after treatmentwere. Result: According to the mass information, 11 metabolites were discovered, with 2 metabolites including SAB ester bond hydrolysis product in plasma, 5 metabolites including danshensu, lithospermic acid (LSA), the decarboxylation and methylation metabolites of LSA in bile, 4 metabolites including SAB methylation and sulfated products in urine excretion. Conclusion: SAB could metabolize into various metabolites including danshensu. The metabolic reaction of SAB occurs mainly in the ester bond and five-member ring, via ester bond hydrolysis, molecular dehydration, decarboxylation, hydroxylation, methylation and sulfate, etc.

      摘要:Objective: To analyse characteristics of pseudoallergic reactions (PARs) of Shuanghuanglian injection (SHLI) and investigate impact of injection speed on PARs. Method: ICR mice were divided into negative group (saline),positive group (histamine of 10 mg·kg-1) and low (150 mg·kg-1),medium (300 mg·kg-1),high (600 mg·kg-1) doses of SHLI.Mice received test articles containing 0.4% Evans blue(EB) by tail intravenous injection,after 30 minutes of administration,area of blue staining and incidence of hypermeability on auricle were recorded,EB exudation of ears was quantitative measurement.ICR mice were randomly divided into negative group and SHLI group of 600 mg·kg-1,mice in different groups were given test articles without EB by tail intravenous injection.After 30 minutes,concentrations of histamine,vascular endothelial growth factor (VEGF),tumor necrosis factor-α(TNF-α) in plasma were measured by ELISA kit,lung and both ears were taken for histopathology evaluation. Result: Both reaction incidence and degree of vascular hyperpermeability were higher with increasing of SHLI dosage.Concentrations of histamine and VEGF were increased significantly after intravenous administration of SHLI,while the concentration of TNF-α was no significant difference.Histopathology evaluation showed edema,neutrophil infiltration in auricle and lung of SHLI groups.And injection speed of SHLI had influence on extent of PARs.When the same dose of SHLI were administrated by intravenous injection,injection at the lower speed (60,120 s) could cause lighter PARs by comparing with injection speed of 5 s. Conclusion: SHLI may induce vascular hyperpemeability so as to cause PARs in mice.PARs induced by SHLI may be associated with release of histamine and VEGF.SHLI-induced PARs shows a relationship with dose and injection speed.The higher dose and higher injection speed can increase incidence and degreed of PARs.Control of dose and injection speed is important to reducing PARs of SHLI.

      摘要:Objective: To investigate the effect of Xiaozhang recipe (XR) on liver fibrosis (LF) induced by carbon tetrachloride (CCl4) in rats. Method: LF model was induced by injecting CCl4 subcutaneously twice weekly for seven weeks and the rats were divided into the normal group, the model group, the colchicine group (1 mg·kg-1), the low-, middle-and high-dose XR groups (200, 400, 800 mg·kg-1). All rats were intragastrically administrated with the corresponding medicines for seven weeks. The change of body weight and the index of liver and spleen were observed. Histological changes and collagen deposition were observed by HE and Sirius red staining sections. Serum samples were collected and alanine aminotransferase (ALT), aspartate aminotransferase (AST), hyaluronidase (HA),laminin (LN), Type Ⅲ procollagen (PCⅢ) were assayed. Liver tissues were harvested and hydroxyproline (Hyp), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and maleic dialdehyde (MDA) were detected. Result: Compared with the normal group, the body weight decreased, the index of liver and spleen increased, serum levels of ALT, AST, HA, LN, PCⅢ and hepatic Hyp, MDA increased, hepatic SOD and GSH-Px decreased in the model group (P <0.01, P <0.05). Compared with the model group, serum levels of ALT, AST, HA, LN, PCⅢ and hepatic Hyp, MDA decreased, hepatic SOD and GSH-Px increased in the colchicine and XR groups (P <0.01, P <0.05). Histological detection showed serious necrosis, false lobule, inflammatory cell infiltration and collagen deposition in the liver of model group and these pathological changes could be alleviated by colchicine and XR. Conclusion: XR has preventive and therapeutic effects on liver fibrosis induced by CCl4 by inhibiting hepatic lipid peroxidation and alleviating the inflammation.

      摘要:Objective: To explore the effect of Xiaofu Tongjie Fang (XFTJF) on expressions of vasoactive intestinal peptide (VIP) and aquaporin 3 (AQP3) in colon tissue of rats with functional constipation. Method: Fifty adult male SD rats were randomly divided into the control group, the model group, the high-, medium-and low-dose XFTJF groups (380, 190, 95 g·kg-1) of 10 rats in each group. The functional constipation model was induced by using compound diphenoxylate tablets (5 mg·kg-1) orally. The rats received intragastric administration of XFTJF for 3 weeks. The general performance, the time to first black stool and fecal water content were observed. The expressions of VIP and AQP3 in rat colon tissue were detected by using immunohistochemical method. The mRNA expression levels of VIP and AQP3 in colon tissue were further detected by using RT-qPCR. Result: Compared with the control group, the time to first black stool significantly prolonged, fecal water content reduced, VIP mRNA level decreased, and AQP3 mRNA level increased in the model group (P <0.05, P <0.01). Compared with the model group, the time to first black stool shortened, fecal water content increased, VIP mRNA level increased, and AQP3 mRNA level decreased in all XFTJF groups (P <0.05, P <0.01). Conclusion: The occurrence of functional constipation in rats is probably related to the abnormal expressions of VIP and AQP3 in colon tissue. The mechanism of XFTJF in the treatment of functional constipation may be realized through regulating the expressions of VIP and AQP3 in colonic tissue.

      摘要:Objective: To observe the effect of Qiwei Baizhu San and its extract on the expressions of interferon-α (IFN-α), interleukin-4 (IL-4) and interleukin-10 (IL-10) in the small intestinal epithelial cell of enteric dysfunctions in mice. Method: The enteric dysfunctions model was induced in ICR mice by intragastric administration of ceftriaxone sodium, lincomycin and folium sennae decoction. The stool, diarrhea and fecal bacterial were observed. The mice were randomly divided into four groups:the model group (MG), the Qiwei Baizhu San group (QG,5.2 g·kg-1), the Qiwei Baizhu San extract group (QTG 0.94 g·kg-1), and the normal control group (NG).The mice in the treatment groups received 0.4 mL volume of the corresponding medicines and the mice in the MG, NG received the same volume of distilled water. The mice were sacrificed at 3, 7, 9, 11, 13 days after modeling. The positive expressions of IFN-α, IL-4 and IL-10 in small intestinal epithelial cell were observed by using immunohistochemistry assay. Result: Compared with the NG, the expression of IFN-α increased at day 3 and 7 (P <0.05), then gradually decreased at day 9, 11 13, while the results were still higher than the NG, QG and QTG (P <0.05). The levels of IL-4, IL-10 decreased in the NG, QG and QTG (P <0.05). Compared with MG, the expression of IFN-a decreased at all days (P <0.05), the levels of IL-4, IL-10 increased at day 9 (P <0.05), and these results had no significant difference between QG and QTG. Conclusion: Qiwei Baizhu San and its extracts could inhibit the intestinal inflammation and reduce immune injury by reducing expression of IFN-a, increasing the levels of IL-4, IL-10 in the small intestine.

      摘要:Objective: To investigate the effect of curcumin on the proliferation, migration, angiogenesis of human umbilical vein endothelial cells (HUVECs), and the growth of B16F10 melanoma. Method: HUVECs were treatment with different concentrations (0-15 μmol·L-1) of curcumin and the proliferation was measured by MTT. The morphological change of apoptotic HUVECs was detecte by using Hoechst 33258.The effect of curcumin on HUVECs migration ability was assayed by Transwell. The anti-angiogenesis action of curcumin was investigated by using the chick chorioallantoic membrane (CAM) assay. The expressions of vascular endothelial growth factor receptor 2 (VEGFR-2) in HUVECs and matrix metalloproteinase-2 (MMP-2) in B16F10 were measured by Western blot. B16F10 cells were inoculated into CAM and post tumor formation, the impact of curcumin on tumor growth was analyzed. The expressions of VEGFR-2 and MMP-2 in B16F10 tumor tissue were analyzed by immunohistochemistry and Western blot. Result: Curcumin (4-15 μmol·L-1) had no inhibitory effect on the proliferation of HUVECs, but it (8, 10, 15 μmol·L-1) could inhibit VEGF-induced proliferation of HUVECs (P <0.05, P <0.01). Hoechst 33258 staining showed that curcumin inhibited VEGF-induced the HUVECs, which was not dependent on the induction of HUVECs apoptosis (P <0.05, P <0.01). Transwell migration experiments showed that curcumin (8, 10, 15 μmol·L-1) could inhibit HUVECs migration, CAM assay confirmed that curcumin could reduce the number of blood vessels, inhibited angiogenesis in vivo(P <0.05, P <0.01). Curcumin could inhibit HUVECs VEGFR-2 and B1610 cells MMP-2 protein expressions (P <0.05, P <0.01). Curcumin (20 mg·L-1) inhibited the growth of melanoma B16F10 in vivo, immunohistochemistry and Western blot showed that curcumin (20 mg·L-1) inhibited the VEGFR-2 and MMP-2 protein expressions (P <0.05, P <0.01). Conclusion: Curcumin inhibited VEGF-induced HUVECs proliferation, migration and angiogenesis, inhibited the growth of melanoma. The mechanism of curcumin may be achieved by inhibiting VEGFR-2,MMP-2 protein expressions.

      摘要:Objective: To observe the effect of modified Xiongxie San on mRNA expressions of matrix metalloproteinases-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in lung tissue of cough variant asthma (CVA) model rat. Method: The 4-month old Wistar rats were randomly divided into the control group, the model group, the singulair group (1.2 mg·kg-1·d-1), and the high-, medium-, low-dose modified Xiongxie San groups (8.64, 4.32, 2.16 mg·kg-1). The CVA model was induced in rats except the rats in the control group by intramuscular injecting of 4% OVA solution of 0.5 mL from the first day, intraperitoneal injecting 2% Al(OH)3 of 0.2 mL once daily, and inhaling 1%OVA solution for 2 minutes from the fourteenth day once every other one day for 7 times. All rats were administrated intragastrically with the corresponding medicines for continuous 15 days from the fourteenth day. The cough frequency was detected. The pulmonary pathology was observed by HE staining, inflammatory cells in bronchoalveolar lavage fluid(BALF) were observed by Wright's staining. The mRNA expressions of MMP-9 and TIMP-1 in rats' lung tissue were observed by RT-qPCR. Result: Compared with the normal group, the lung injury and inflammatory cell infiltration condition were serious, the cough frequency increased, the mRNA expressions of MMP-9 and TIMP-1 in lung tissue increased in the model group (P <0.01). Compared with the model group, the lung injury and inflammatory cell infiltration condition were improved, the cough frequency decreased, and mRNA expressions of MMP-9 and TIMP-1 in lung tissue decreased in all dose of Xiongxie San groups (P <0.01, P <0.05). Specially, the pulmonary pathology improvement had better result in the high-dose Xiongxie San group (P <0.01). Conclusion: Modifed Xiongxie San could inhibit airway remodeling of CVA, and the pharmacological mechanism may be related to reducing the increased mRNA expressions of MMP-9 and TIMP-1 in lung tissue.

      摘要:Objective: To observe the effect of Bixie Chubi Tang (BXCBT) on reducing serum uric acid (SUA), anti-inflammation and analgesia, and to discuss its mechanism. Method: The 32 Kunming mice were randomly divided into four groups:the normal group, the model group, the BXCBT group (20.9 g·kg-1), and the llopurinol group (40 mg·kg-1). The mouse hyperuricemia model were induced in mice by intraperitoneally injecting allantoxanic acid potassium of 300 mg·kg-1. The levels of SUA and xanthine oxidase (XOD) were detected. The anti-inflammatory and antalgic effect was observed on the writhing test caused by glacial acetic acid and ear swelling caused by dimethylbenzene in mice. Both two experiments included the model group, the BXCBT group (20.9 g·kg-1) and the allopurinol group of 10 mice each. In another experiment, the acute gouty arthritis (AGA) model of mice was established by uric acid sodium salt. The 32 mice were randomly divided into the normal group, the model group, the BXCBT group (20.9 g·kg-1) and the indomethacin group (2 mg·kg-1). The levels of interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α) were detected, and the anti-inflammatory effects and possible mechanisms were analyzed. Result: Compared with the normal group, the levels of SUA and XOD increased, the levels of IL-1β and TNF-α increased in the model groups (P <0.05, P <0.01). Compared with model group, levels of SUA and XOD decreased, the writhing times decreased, the mouse ear swelling and ankle swelling improved, and the levels of IL-1β and TNF-α decreased in the BXCBT group (P <0.05, P <0.01). Conclusion: BXCBT has an effect on reducing SUA in mice, and the mechanism may be related to inhibiting the expression of XOD. It also has an effect on inflammation and pain, and the mechanism may be related to inhibiting the levels of IL-1β and TNF-α.

      摘要:Objective: To study the correlation between gas chromatography-mass spectrometry (GC-MS) fingerprint of the essential oil of Amomi Fructus and anti-inflammatory activity, and to provide a reference for revealing its efficacy material basis of anti-inflammatory. Method: The fingerprints of the essential oil of Amomi Fructus from different habitats (Amomum villosum from Guangxi, Wuchuan, Panlong, Chuncheng and Yongning;A. villosum, xanthioides Wu et Senjen from Yunnan and Myanmar) were established by GC-MS, and anti-inflammatory activity of the essential oil was observed by the ear swelling caused by dimethylbenzene in mice. The 90 mice were randomly divided into the model group, the dexamethasone group (12.5 mg·kg-1), the Guangxi, Wuchuan, Panlong, Chuncheng, Yongning, Yunnan and Myanmar groups(3.2 g·kg-1) of 10 mice each. The mice in the model group were intragastrically administrated with normal saline and the other mice were intragastrically administrated with the corresponding medicines once daily for 4 days. The correlation between the fingerprint and anti-inflammatory activity was analyzed by grey relational analysis. Result: The GC-MS fingerprint of the essential oil of Amomi Fructus was established from different habitats, and 13 common peaks were identified. Compared with model group, the essential oils from Guangxi and Chuncheng had no obvious anti-inflammatory activity, and the others had obvious anti-inflammatory activity as compared to the dexamethasone group (P <0.05, P <0.01). The correlation between GC-MS fingerprint and the anti-inflammatory activity was from 0.857~0.734, with ranking of borneol >linalool >camphor >α-terpineol >α-bergamotenol >D-limonene >bomyl acetate >α-santalol >myrcene >spathulenol >α-juniper alcohol >camphene >α-pinene. Conclusion: The anti-inflammatory activity of the essential oil of Amomi Fructus may be the combined effect of a variety of chemical constituents. There are definite correlations between the abundant borneol, linalool from Wuchuan, Yongning and the anti-inflammatory activity.

      摘要:Objective: To observe the effect of Zuogui Wan and Yougui Wan on bone mineral density and bone metabolism of ovariectomized rats with osteoporosis. Method: Post menopausal osteoporosis(PMOP) rats model was established by excision bilateral ovaries of 6 month old SD female rat.After successful modeling,80 modeling rats were randomly divided into model group, estradiol valerate group,Zuogui Wan groups with different doses,and Yougui Wan groups with different doses. Rats with resection of abdominal adipose tissue were divided into sham group. The changes of bone mineral density(BMD), serum bone alkaline phesphatase(BALP), serum osteocalcin(OC) and serum osteoprotegerin(OPG) deoxypyridinoline were detected respectively after 10 weeks. Result: Compared to sham operation group,BMD and serum OPG levelin of model group rats were decreased,but serum BALP, OC were increased. Compared with the model group, BMD, serum OPG of rats in Yougui Wan groups and Zuogui Wan groups were improved and serum BALP, OC content were decreased. Conclusion: Zuogui Wan and Yougui Wan can inhibit bone resorption, correct bone metabolism imbalance, which is beneficial to treatment of post menopausal osteoporosis

      摘要:Objective: To explore the effect of the Chinese caterpillar fungus isolated from natural Paecilomyces hepiali strain (Cs-4) inhibited by artificial fermentation, the Fermented-4 powder on SMMC-7721 human hepatocellular carcinoma cell growth. Method: Twenty male SD rats were randomly divided into four groups (n=5):control group and Cs-4 low, medium and high dose groups, control group was treated with distilled water, Cs-4 low, middle and high dose groups were treated with distilled water to prepare the Cs-4 (0.77,1.54,3.08 g·kg-1,ig)suspension, according to the 10 mL·kg-1 body weight orallyorally, 2 times twice a daily;after intragastric administrationfor 3 days, the serum containing drug was prepared. The liver cell suspension was adjusted to 4×104 cells/mL, each hole of 96 hole plate was added with 100 μL cell suspension culture. After 24 h, the holes were respectively added with 160 μL cells culture fluid and 40 μL blank serum and containing Cs-4.After 48 h MTT detection was carried out,the inhibition rate of proliferation of cells was calculated. The SMMC-7721 cell suspension was adjusted to 2.8×104 cells/mL, were cultured in 6 well plates (each hole 2 mL inoculation) for 24 h, the holes were added to 1.6 mL cells suspension and 0.4 mL black serum and containing drug serum were added after 48 h,Annexin V-FITC/PI staining by flow cytometry was used to detect of apoptosis;expression of vascular endothelial growth factor(VEGF), matrix metalloproteinases-2(MMP-2) were detected with ELISA method. Result: MTT showed the proliferation of SMMC-7721 cells was significantly inhibited by containing Cs-4 serum compared with the blank control group(P <0.05), the different dose inhibition rates were 12.2%, 23.2%, 38.1% in a dose-dependent manner. Apoptosis detection showed that in containing Cs-4 serum groups,the apoptosis rates were 13.9%, 23.5%, 24.9%, which was higher than those in the blank group (P <0.05). In containing Cs-4 serum groups, the concentration of VEGF in supernatant was respectively 443.5, 436.9, 411.0 ng·L-1, which was lower than that in control group (P <0.05). Conclusion: The containing Cs-4 serum can inhibit the proliferation and VEGF expression of SMMC-7721 cells,and increase the apoptosis of SMMC-7721 suggesting Cs-4 had anti-tumor metastasis action.

      摘要:Objective: To observe the effect and mechanism of total flavonoids of Lantana camara on rheumatoid arthritis(RA)in rats. Method: After establishing rat Freund's adjurant arthritis(AA) model,rats with AA were divided into four groups,model group,dexamethasone group(0.15 mg·kg-1) and total flavonoids of L.camara group(100,25 mg·kg-1).The total flavonoids of L.camara group were given for 16 days. The effects of total flavonoids of L.camara on anti-arthritic were studied through observing the changes of paw swelling,arthritis index at day 3,8,16,21 and 24, and levels of interleukin 1β(IL-1β) and tumor necrosis factor α(TNF-α)after medication.Anti-inflammatory activity was studied by mice auricular swelling test induced by xylene and rat inflammation model induced by planted cotton ball in axillary,and acetic acid-induced writhing and hot-plate test in mice were used to study the analgesic activity. Result: Compared with models,the total flavonoids of L.camara could inhibit paw edema and arthritis index in RA rats significantly(P <0.05),and the levels of serum IL-1β and TNF-α showed a marked decrease (P <0.05).The total flavonoids of L.camara could inhibit the mice ear edema induced by xylene,and could also decrease the weight of cotton ball granuloma. The total flavonoids of L.camara could increase the pain threshold value of the mice on the hot plate,and decrease the number of writhing in mice flowing ip administration of acetic acid. Conclusion: The total flavonoids of L.camara could ameliorate rheumatoid arthritis in RA rats, and have a good anti-inflammatory and analgesic action, and its effect has a close relation with the level of IL-1β and TNF-α in RA rats.

      摘要:Objective: To study the effect of paeonol (Pae) on myocardial fibrosis in rats with acute myocardial infarction (AMI) and to investigate whether its mechanism is related to the expression of transforming growth factor-β1(TGF-β1),Smad2 mRNA and protein. Method: In vivo model of AMI was made using the ligation of left anterior descending coronary branch.The successful myocardial infarction(MI) modeling was based on the following changes like electrocardiogram(ECG) ST segment level downward or upward shift ≥0.1 mV and T wave towering over with R wave of 1/2, left ventricular anterior turned pale. The rats were randomly divided into the model group, the sham group, the low-, high-dose Pae groups (8.0,12.0 mg·kg-1). The sham group and model group were given ip volume of saline daily. The left ventricular indexed were measured after 4 weeks. The pathology of myocardial tissue was observed by HE staining, the level of myocardial interstitial fibrosiswas observed by Masson staining. The mRNA expression levels of TGF-β1,Smad 2 in myocardial tissues were detected using real-time quantitative PCR(RT-PCR) technique. Result: Compared with the sham group, he left cardiac index was significantly higher(P< 0.01) in the model group. Compared with the model group, the left cardiac index was decreased(P< 0.01, P< 0.05)in all does Pae groups. HE staining showed that compared with the sham group pathological changes were significantly in the model group, there were cardiac hypertrophy, elongation and coagulation necrosis in injured myocardial hypertrophy edge part, while some loose edema occurred in left ventricular myocardial wall portion. Compared with the model group, the pathological changes were decreased. Masson staining showed that compared with the sham group, collagen fibers increased significantly in model group, and there appeared fiber epilepsy marks, and also severe myocardial fibrosis, Compared with the model group, the level of myocardial fibrosis in all dose Pae groups were decreased. Compared with the sham group, TGF-β1,Smad 2 mRNA and protein expression levels were significantly increased(P< 0.05);compared with the model group, the TGF-β1,Smad 2 mRNA and protein expression level were decreased(P< 0.01)in all dose Pae groups. Conclusion: Pae has anti-myocardial fibrosis effect in AMI rats. Its mechanism may be related to the intervention of TGF-β1/Smad2 signaling pathways.

      摘要:Objective: To discuss the lung tissue infected by mycoplasma repair mechanism by investigating the influence of Platycodon grandiflorum total saponins on pulmonary surfactant protein A(SP-A) express in lung tissue of rats infected by mycoplasma. Method: Sixty Wistar rat with half males and half females were divided randomly into 6 groups, blank group, model group, positive(azithromycin) group (45 mg·kg-1(first day),22.5 mg·kg-1(the next day to the fifth day), the high(56 mg·kg-1),middle (28 mg·kg-1) and low (14 mg·kg-1)dose group of P. grandiflorum total saponins.the model was established with the method of nasal intubation drop of Mycoplasmal pneumonia(MP). The second day after modeling, azithromycin is given for 5 d, P. grandiflorum t otal saponins is given for 10 d.the lung tissue was collected after 10 days to observe the inflammation of lung tissue by HE staining and to detection of the SP-A mRNA expression level in lung tissue. Using SPSS 17.0 software to analysis data. Result: The pathological results showed that compared with blank group, the interstitial pneumonia of model group was obvious (P <0.01). Compared with model group, the inflammation of lung tissue of positive medicine group, the high,middle and low dose group of P. grandiflorumtotal saponins significantly decreased (P <0.05). The SP-A mRNA expression level in lung tissue showed that compared with blank group, the SP-A mRNA expression of model group significantly lowered (P <0.01). Compared with model group, the SP-A mRNA expression of positive medicine group, the high and middle dose group of P. grandiflorum total saponins obviously up-regulated(P <0.05), the SP-A mRNA expression of the low dose group of P. grandiflorum total saponins had no significant difference. Conclusion: P. grandiflorum total saponins can improve the lung tissue infected by mycoplasma, and the improving effect of lung function could be realized by raising SP-A expression.

      摘要:Objective: To study the effect and mechanism of beta-elemene in vivo and in vitro. Method: The in vitro effects of beta-elemene(12.5-200 μmol·L-1) on human lung cancer SPC-A-1 cell line, A549 cells and human diploid fibroblasts MRC-5 cell proliferation were observed by MTT method;the morphological changes of apoptotic cells were detected by Hoechst 33258;the cells stained by Annexin V and propidium iodide (PI)were observed by flow cytometry for apoptosis,cell morphological changes were observed by electron microscope. Its in vivo anti-tumor effect on transplanted human SPC-A-1 tumor cells in nude mice was detected by conventional methods. Result: Fifty percent inhibition concentration(IC50) of the above-mentioned 2 kinds of lung cancer cells by beta-elemene was 126.0,134.1 μmol·L-1,but the effect on human diploid fibroblasts MRC-5 cells was promoting growth. The inhibition effects of beta-elemene on human lung cancer cell line SPC-A-1 depended on the duration and concentrations. IC50 at the 24,48,72 h was 549.7,126.0,21.2 μmol·L-1,respectively. Apoptotic body in apoptotic cells could be observed by Hoechst 33258 dying. Flow cytometry showed that beta-elemene could induce SPC-A-1 cell apoptosis distinctly at different levels.The growth inhibition rates of beta-elemene (45,135 mg·kg-1) on transplanted human lung cancer cell line SPC-A-1 cells in vivo were 34.01%,49.57%. Conclusion: Beta-elemene has an antitumor effect between 12.5-200 μmol·L-1.The mechanism of anti-tumor effect is possibly achieved by inducing apoptosis of tumor cells.

      摘要:Objective: The aim of this study was to study the impact of total alkaloids of Corydalis saxicola on human lung cancer A549 cell proliferation and apoptosis and to explore its possible mechanism. Method: The logarithmic growth phase of A549 cells were cultured in vitro and incubated with C. saxicola Banting. MTT assay was used to detect the proliferation of A549 cells after the treatment of Corydalis saxicola at different concentrations (0.1,0.05,0.01,0.005 g·L-1)for different time (24,48,72 h). Hoechst stain was applied to analyze the apoptosis effects. Flow cytometry were used to anlyze cell apotosis. The mRNA expression changes of Caspase-3 and Survivin after treatments were detected by RT-PCR. Result: C. saxicola could inhibit the proliferation of A549 cells in dose-time manner. After 48 h, inhibition of C. saxicola group in A549 cells was stronger than the control group (P <0.05).Flow cytometry showed that the apoptotic rate increased with the concentration increased (P <0.05). RT-PCR results suggested that compared with the control group, C. saxicola group up-regulated the expression of Caspase-3 mRNA and down-regulated the expression of Survivin mRNA (P <0.05). Conclusion: C. saxicola can inhibit proliferation and induce the apoposis of A549 cells in a time-and dose-dependent manner. The mechanism may be related to the up-regulation of Caspase-3 expression and down-regulation of Survivin expression.

      摘要:Objective: To investigate the neuroprotective effects and mechanism of luteolin against β-amyloid 25-35(Aβ25-35) induced neural damage. Method: Primary neurons were isolated and purified from cerebral cortex of suckling mouse of BALB/c. The experiment was assigned three groups. The control group, the model group (neurons were co-cultured with Aβ25-35 incubation for 12 h and luteolin administration (neurons were co-cultured with Aβ25-35 and luteolin incubation for 12 h). water soluble tetrazolium sait(WST-1) method was used to test cell viability. The apoptosis was examined by Hoechst 33258/propidum iodide(PI) double staining. The expression of Caspase-3 was analyzed by the Western bolt; reactive oxygen species(ROS) was examined by 2',7'-dichlorofluorescin diacetate(DCFH-DA) assay. The productions of tumor necrosis factor-α(TNF-α) and interleukin-10(IL-10) were measured using enzyme-linked immunosorbant assay (ELISA) kits. Result: WST-1 assay results displayed that the cellular viability of model group was 48.5% and had significant differences compared to control group. Cellular viability of Aβ25-35-induced cells exposed to 10,30,60,90 μmol·L-1 of luteolin was 60.8%,62.4%,71.7%,73.6% and then 60 μmol·L-1 of luteolin was choose for the next experiments. 60 μmol·L-1 of luteolin obviously increased cell viability to 71.7%;Hoechst 33258 nuclear staining and PI double staining showed that luteolin could significantly decrease apoptotic rate compared with the Aβ25-35 treated group;luteolin inhibited Caspase-3 and ROS of neurons induced by Aβ25-35. Compared with model group, luteolin also decreased the productions of TNF-α to 176.7 ng·L-1 and increased IL-10 to 634.8 ng·L-1(P <0.05). Conclusion: Luteolin possesses the neuroprotective effects on Aβ25-35 damaged neurons, which may be associated with inhibition of ROS, up-regulation of anti-inflammatory cytokine IL-10, down-regulating of Caspase-3 and TNF-α to against neurons apoptosis.

      摘要:Objective: To observe therapeutic effects of traditional Chinese medicine Yishen Tongluo decoction on diabetic nephropathy(DN). Method: One hundred and forty-two patients with diabetic nephropathy were randomLy divided into three Groups. 40 patients in control group were treated only by western medicine, 50 patients in treatment group were treated Ginkgo-dipyridolum injection(GDI) combined with western medicines;52 patients in treatment group were treated by astragalus injection and Yishen Tongluo decoction combined with western medicines,with a treatment course of 4 weeks for three groups. The changes of the therapeutic efficacy,fasting blood glucose (FBG),postprandial dlood glucose (PBG), total cholesterol(TC),triglyceride (TG),low-density lipoprotein(LDL-L),high-density lipoprotein (HDL-L),24-hour urinary protein quantity,urine albumin excretion rate(UAER),urineβ2-microgiobulin(β2-MG),blood urine nitrogen (BUN),serum creatinine (SCr),creatinine clearance rate(CCr),glycosylated hemoglobin(HbA1c),homocysteine(Hcy),intima-media thickness(IMT),et al. Result: The total effective rate in treatment group A was 68.0%(34/50 cases),and in treatment group B was 86.5%(45/52 cases),which was obviously higher than that in control group(55.5%,22/40 cases) (P< 0.05). Between in treatment groups,deffrences are statistically significant(P< 0.05). Conclusion: GDI can decrease the amount of excretingm icroalbum inuria in diabetes. astragalus injection plus Yishen Tongluo decoction can decrease significantly the amount of excretingm icroalbum inuria and protect renal function in diabetes.

      摘要:Objective: The purpose of this study was to observe the influence of effect and immune function in children with bronchial asthma after therapy of inhaled corticosteroid by treatment of Yiqi Guben Tang combined with acupoint sticking. Method: In this study, 86 children with bronchial asthma from October 2012 to August 2014 were randomly divided into two groups. The control group was given therapy of inhaled corticosteroid, on the basis of control group, the observation group was given therapy of Yiqi Guben Tang combined with acupoint sticking, then clinical curative effect, the number of asthma attacks and colds, indexes of cellular immunity and humoral immunity function before and after treatment were observed. Result: After treatment of 6 months, total effective rate in observation group was 86.05%, while total effective rate in control group was 72.09%, there was no statistically significant difference between the two groups. After treatment of 12 months, total effective rate in observation group was 95.35%, total effective rate in control group was 81.40%, with no statistically significant difference between the two groups. During the treatment, asthma attack frequency in observation group was (2.8±0.3) times, that in control group was (3.1±0.4) times, there was statistically significant difference between the two groups (P <0.05);cold frequency in observation group was (2.1±0.3) times, that in control group was (3.4±0.2) times, there was statistically significant difference between the two groups (P< 0.05). Difference of CD3+, CD4+, CD8+ before treatment between the two groups had no significant difference;after treatment, CD3+ and CD4 + of both two groups were higher than before treatment, while CD8+ was lower, with significant difference (P <0.05). CD3+ and CD4+ after treatment in observation group were higher than those in control group, while CD8+ was lower, with significant difference between both groups (P <0.05). IgA, IgM, IgE, IgG and insulin-like growth factor(IGF-I) before treatment between groups had no significant difference;after treatment, IgA, IgG, IgM and IGF-I of two groups were higher than before treatment, while IgE was lower, with significant difference (P <0.05). IgA, IgG, IgM and IGF-I after treatment in observation group were higher than those in control group, while IgE was lower, with significant difference between groups (P <0.05). Conclusion: The clinical efficacy of therapy of Yiqi Guben Tang combined with acupoint sticking in the treatment of children with bronchial asthma was significant, it can alleviate clinical symptoms of asthmatic children, reduce the recurrence of asthma in children with bronchial asthma, strengthen the function of cellular immunity and humoral immunity.

      摘要:Objective: The aim of this study was to observe and compare the curative effect of Pudilan Xiaoyan oral liquid and ribavirin in the treatment of children with hand foot and mouth disease,to obtain a better therapy. Method: In this study, two hundred and twenty cases diagnosed as hand foot and mouth disease in our hospital from 2010 May to 2013 December were selected and randomly divided into observation group and control group, with 110 patients in control group by conventional rehydration, symptomatic treatment based on the use of ribavirin treatment, and 110 cases in observation group, in the conventional treatment based on the use of Pudilan Xiaoyan oral liquid treatment. The treatment course lasted for 5 days, and the clinical treatment effects were compared between the two groups, including total efficiency, rash, bleb subsidise time, time for body temperature recovery, RV-Ag negative conversion rate and adverse circumstances etc. Result: The total effective rate of observation group was 93.3%, higher than 76.2% of the control group (P <0.05), the observation group 3 days and 5RV-Ag negative rate were 85.8% and 92.20% in the observation group, higher than 71.30%, 75.51%in the control group (P <0.05);the rash, bleb subsidise time in the observation group is (2.77±0.54) d and (2.91±0.70) d, time for body temperature recovery is (3.37±0.61) d, all were slightly lower than those of the control group (P <0.05);between the two groups, the difference in adverse reaction rate was not statistically significant. Conclusion: Pudilan Xiaoyan oral liquid and ribavirin can be used in the treatment in children with hand foot mouth disease, which can, to some extent, alleviate symptoms in children with hand foot and mouth disease, Pudilan Xiaoyan oral liquid has stronger ability to inhibit the proliferation of the virus, the symptoms subsided faster, and no obvious adverse reaction was observed. And the mechanism is related to the anti-inflammatory and antiviral synthesis effect of Pudilan Xiaoyan oral liquid. Pudilan Xiaoyan disinfection oral liquid is effective in the treatment in children with hand foot and mouth disease, and is worth of the clinical promotion.

      摘要:Objective: To observe the clinical effect of Biyankang decoction in treating chronic rhinosinusitis (CRS) of stagnated heat of gallbladder symptom, and to investigate its influence on improving patients' quality of life. Method: One hundred and twenty patients with CRS were randomly divided into the control group (60 cases) and the observation group (60 cases). Patients in the control group received 250 mg clarithromycin granules twice daily and 15 mg·kg-1 for the children according to their weight. On the basis of nasal washing therapy in the control group, patients in the observation group added Biyankang decoction orally. All patients in two groups received 4 weeks of treatment. Then patients in two groups received half does of clarithromycin granules orally for another 8 weeks. The subjective condition was evaluated by using a visual analog scale (VAS) and the objective condition was evaluated by using the Lund-Kennedy scoring method. Nasal mucociliary transport rate (MTR) was detected by using saccharin test. The quality of life was evaluated by using sinuses nasal outcome test-20 (SNOT-20) questionnaire before and after the therapy. Result: The curative effect in the observation group was superior to that in the control group (P <0.05). The VAS, Lund-Kennedy and sign scores in the observation group were less than those in the control group (P <0.01). After treatment, nasal MTR in the observation group was higher than the control group (P <0.01). The symptoms including nasal symptoms, associated symptoms, sleep disorders and emotional end, SNOT-20 total scores in the observation group were inferior to those in the control group (P <0.01). Conclusion: Biyankang decoction could improve the subjective, objective symptoms, enhance MTR and improve the quality of life in patients with CRS of stagnated heat of gallbladder symptom. Its curative effect is superior to using western medicine treatment alone.

      摘要:Objective: To observe the effect of Guifu Dihuang pills combined tamsulosin hydrochloride sustained release capsules in treating benign prostatic hyperplasia (BPH) disease of kidney-yang deficiency syndrome. Method: One hundred and sixteen BPH patients were randomly divided into the control group (56 cases) and the observation group (60 cases) via a digital method. Patients in the control group received 0.2 mg tamsulosin hydrochloride sustained release capsules orally once every night. Based on the therapy of the control group, patients in the observation group added 6 g Guifu Dihuang pills orally twice daily. All patients in two groups received 12 weeks of treatment. International prostate symptom scores (I-PSS), kidney-yang deficiency syndrome scores, and quality of life (QOL) scores caused by urination symptom were assessed. Moreover, post-void residual urine volumes (PVR), the maximal urinary flow rates(Qmax) and average urinary flow rates (Qave) were measured. Prostate size and quality were tested through type-B ultrasonic. Enuresis nocturia frequencies were recorded. Result: The total curative effect in the observation group was 91.66%, which was superior to 76.78% in the control group (P <0.05). Scores of I-PSS, kidney-YANG deficiency syndrome, QOL in the observation group were less than those in the control group (P <0.01). The PVR in the observation group was lower (P <0.01), the Qmax and Qave were higher than those in the control group (P <0.01). After therapy, prostate size and quality decreased as compared with the control group (P <0.01). Frequencies of enuresis nocturia were less than those in the control group (P <0.01). Conclusion: Guifu Dihuang pills combined tamsulosin could ameliorate lower urinery-tract syndrom, improve patients' QOL and ameliorate risk factors of clinical progression in treating BPH patients with kidney-yang syndrome, and it is worthy of further study.

      摘要:Objective: To discuss the amelioration effect of Bushen Huoxue decoction on cognition impairment of diabetes, and to investigate its influence on oxidative stress and inflammatory response. Method: One hundred and four patients were randomly divided into the control group (57 cases) and the observation group (57 cases) by a random number table. Patients in the control group received 30 mg nimodipine tablets thrice daily and 100 mg aspirin enteric-coated tablets once dialy. Based on the treatment of the control group, patients in the observation group added Bushen Huoxue decoction. All patients in both two groups received 3 months of treatment. Before and after treatment, cognitive ability was evaluated by Montreal cognitive assessment (MoCA), levels of fasting blood-glucose (FBG), glycosylated hemoglobin (HbAlc), insulin resistance index (HOMA-IR), superoxide dismutase (SOD), malondialdehyde (MDA), total antioxidant capacity (T-AOC), interleukin1β (IL-1β), tumor necrosis factor-α (TNF-α) and homocysteine (HCY) were detected. Result: After treatment, scores of visuospatial skills, physical performance, attention, language, abstract, memory, delayed memory, disorientation and MoCA scale were higher in the observation group than those in the control group (P <0.05, P <0.01). Levels of HbA1c and HOMA-IR were lower, levels of T-AOC and SOD were higher, level of MDA was lower, levels of TNF-α, IL-1β and Hcy were lower in the observation group than those in the control group (P <0.01). Conclusion: Bushen Huoxue decoction could improve the cognitive ability of patients with type 2 diabetes mellitus. Its mechanism of action may be related to ameliorating insulin resistance, oxidative stress and inflammatory response.

      摘要:Objective: To discuss the protective effect of Shenkang injection on residual renal function of patients with uremia after hemodialysis. Method: Eighty-six patients with uremia after hemodialysis were randomly divided into the control group (43 cases) and the observation group (43 cases) by a random number table. In order to control the primary diseases, patients in the control group received hemodialysis, nutritional support, treat anemia, blood glucose and blood pressure (BP) control Based on the treatment of the control group, patients in the observation group added 80 mL Shenkang injection by intravenous infusion once daily. All patients received 6-,4-week periods of treatment separated by 4 weeks without treatment each period. Before and 6-, 12-month treatment, residual renal function (RRF) and urine volume were detected. Falling speed of RRF and urine volume were calculated. Incidence rate of privary endpiont (24-hours urine volume≤400 mL) was calculated. Before and after treatment, BP, body weight (BW), blood urea nitrogen (BUN), creatinine (Cre), hemoglobin(Hb), albumin (Alb) and prealbumin (PA) were observed. Result: RRF and urine volume in the observation group were superior to those in the control group after 6-, 12-month treatment (P <0.01). The declining rates of RRF and urine volume in the observation group were lower than those in the control group during 12 months (P <0.01). Levels of systolic BP and diastolic BP in the observation group were lower than those in the control group (P <0.01). The incidence of the primary end point event was 20.9% in the observation, while which was 44.18% in the control group (P <0.05). After therapy, Hb standard in two groups went up, while the result was more obvious in the observation group (P <0.01). Alb and PA levels in the observation group were higher than those in the control group (P <0.01). After treatment, BUN and Scr standard in two groups declined, while the result was lower in the observation group (P <0.01). Conclusion: Shenkang injection has protective effect in treating residual renal function with uremia patients after hemodialysis. It could delay the deterioration speed of the disease and deserves further study.

      摘要:Objective: To discuss the curative efficacy of Bushen Shujin decoction in treating early metaphase knee osteoarthritis after oral and fumigating-washing administration, and to investigate its influence on tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) in serum and synovial fluid. Method: One hundred and sixteen patients were randomly divided into the control group (56 cases) and the observation group (56 cases) by a random number table. All patients in both two groups received sodium hyaluronate through intra-articular injection once daily for 5 weeks, and 0.2 g celecoxib capsules once daily when needed. Patients in the control group added 5 Kanggu Zengsheng capsules thrice daily. Patients in the observation group added oral and fumigating-washing administration of Bushen Shujin decoction. All patients in both two groups received 12 weeks of treatment and 6 months of follow-up. Before and after treatment, and during the follow-up, the Western Ontario and McMaster Universities (WOMAC) was evaluated, levels of TNF-α and IL-1β in serum and synovial fluid were detected. Result: The clinical curative effect in the observation group was superior to that in the control group (P <0.05). The WOMAC score of each dimension including pain, stiffness, joint function and the total score in two groups went down obviously. Besides, the scores were lower in the observation group (P <0.01). During follow-up, WOMAC score of each dimension and the total score in both groups went up compared with before (P <0.01), while the results in the observation group were inferior to those in the control group (P <0.01). Levels of TNF-α and IL-1β in the joint fluid and serum declined in both groups after therapy, while the results were lower in the observation group (P <0.01). Conclusion: Bushen Shujin decoction could reduce the clinical symptoms of early metaphase KOA in elderly patients after oral and fumigating-washing administration. The clinical curative effect is better and longer than Kanggu Zengsheng capsules and last longer. Its mechanism may be related to reducing TNF-α, IL-1β in joint fluid and serum, and inhibiting the inflammatory response.

      摘要:Objective: The aim of this study was to analyze the prescription compatibility principles of for insomnia by traditional Chinese medicine inheritance support system (TCMISS). Method: The prescriptions for insomnia from four databases (CNKI, VIP, WANGFANG and SinoMed), were collected and analyzed using data mining method after data entry into the TCMISS. Result: Based on the analysis of 126 cases of prescriptions, that reported in 84 literatures, 36 herbs (frequency≥8) were found frequently used in clinic, and 39 couplet medicines (support≥8), 10 association rules (support≥8 and confidence≥0.6), 19 core combinations and 4 new prescriptions were mined from the databases. Conclusion: TCMISS is a helpful tool for the research of traditional Chinese medicine, and prescriptions for insomnia are mainly composed of drugs with tonic effect, supplemented by herbs with mind-easing and/or antipyretic effects;furthermore, herbs for tranquilization are specific to prescriptions for insomnia.

      摘要:Objective: The aim of this study was to evaluate the efficacy and safety of Jiawei Xiaoyao San for irritable bowel syndrome. Method: We searched the VIP, CNKI, Wanfang and China biology medicine disc(CBMdisc) databases. Randomized controlled trials of clinical therapeutic studies on Jiawei Xiaoyao San for irritable bowel syndrome were included. The quality of included studies was evaluated and Meta-analysis was performed. Result: In this study, 17 trials involving 1 542 patients were included. Meta-analysis indicated that Jiawei Xiaoyao San was lower in the recurrence rate and more effective than other drugs in improving symptoms and signs of irritable bowel syndrome. The RR value (95%CI)were[RR=0.33,95%CI(0.22,0.49), P <0.000 01]and[RR=1.31,95%CI(1.24,1.38), P< 0.000 01], respectively. The most included researches did not report adverse reactions. Conclusion: Based on this review, Jiawei Xiaoyao San is effective for irritable bowel syndrome. However, the evidence is not strong enough due to the low quality of included trials. Further, large-sample and multi-center studies are needed.

      摘要:It was widely accepted, in the present studies on the exact pathogenesis of anxiety disorders, that there is injury of hippocampal neurons in brain. Overseas studies indicate that injury play an important role in the regulation of protein phosphatase 2A (protein phosphatase 2A, PP2A) and the expression of α-synuclein (α-synuclein). Therefore, we consider that the change in the activity of PP2A may induce the abnormal expression of α-syn, causing relative changes in neurons;so the phosphorylation/dephosphorylation of 129 points α-syn protein C-terminal9 might be caused through regulating the activity of PP2A, then GR activity against neuronal apoptosis are further regulated, to repair damaged hippocampus, then to work in the anti-anxiety disorder. This paper reviews the research progress on the protein phosphatase 2A, α-synuclein in the molecular mechanisms of anxiety disorders and the progress on experimental study of the mechanism of Chinese medicine compound Danzhi Xiaoyao San in the treatment of anxiety disorders in recent years.

      摘要:Near infrared spectroscopy (NIR) is a rapid, nondestructive and green analytical technology, and its quantitative analysis technique is especially rapidly developing. Determination methods with small deviation and good repeatability designated byChinese Pharmacopoeia like HPLC, GC, CE, AAS and ICP-MS were used to obtain chemical reference. Then chemometrics models like PLS and ANN were established to predict the contents of chemical compositions in herbal medicines and compound Chinese medicinal preparation. The established models were commonly evaluated by indicators of correlation coefficient (R), RMSEC (the root-mean-square error of calibration), RMSEP (the root-mean-square error of prediction) and RMSECV (the root-mean-square error of cross-validation). NIR quantitative analysis technique of has broad research and application prospects in the content determination of effective compositions, index compositions, moisture, extract and volatile oil of herbal medicines and compound Chinese medicinal preparation, effective compositions of mineral medicines, optimization of preparation technologies, and real-time monitoring of pharmaceutical process.

      摘要:The chronic obstructive pulmonary disease (COPD) is a common disease of respiratory system, its morbidity and mortality are higher. The lung function decreases gradually, the labor force and the quality of life of patients was affected seriously. It is gradually understood with knowledge of COPD in recent years,the relevant pathogenesis of COPD and research literature of traditional Chinese medicine on COPD are analyzed in this paper.Based o the results of our scientific research,pathogenesis of COPD relevant with inflammation, airway mucus secretion of excessive, oxidation/antioxidant imbalance, reconstruction were viewed. From the point of pathogenesis,the therapeutic methods are important in finding action targets for COPD, and it was of traditional Chinese medicine has certain advantages in treatment of COPD.

      摘要:Ginsenosides, as the major constituent of Ginseng Radix et Rhizoma, possess significant anti-tumor effect. However, ginsenosides exhibit poor clinical bioavailability due to thelow water solubility. This paper reviewed the development and researches on novel drug delivery systems for ginsenosides in recent years, including liposomes, nanoparticles, emulsions, microspheres, etc. These drug delivery systems could endow ginsenosides improved bioavailability as well as certain controlling release or targeting capability. In addition, micelles, fibrous membranes are also employed for ginsenosides delivery. To date, ginsenosides delivery systems are not deeply developed, the trend in the future is the combination of multi-functional systems. This review may provide certain guidance or instruction for developing novel anti-tumor drug delivery systems in the future.
