      摘要:Objective: To elucidate pharmacodynamic mechanism of Pulsatillae Radix by investigating degradation dynamics of its saponins in intestinal flora. Method: HPLC-ELSD was adopted to determinate contents of pulchinenoside B3, BD, B7, B10, B11with mobile phase of methanol (A)-water (B) for gradient elution (0-30 min, 75%-90%A), drift tube temperature at 75 ℃, gas flow rate of 2.0 L·min-1.Degradation parameters were determinated through incubating total saponins from Pulsatillae Radix with rats' intestinal bacteria in vitro. Result: Saponins from Pulsatillae Radix were in accordance with first-order reaction, pulchinenoside B3, BD, B7, B10, B11 degradated rapidly in rats' intestinal flora within 48 hours with degradation rate constants were 0.079 4, 0.052 3, 0.053 9, 0.042 6, 0.046 8 h-1, half-life periods were 8.73, 13.25, 12.86, 16.27, 14.81 h, respectively. Conclusion: Intestinal flora is the most important factor on metabolism of prototype compounds of saponins from Pulsatillae Radix.

      摘要:Objective: To investigate stability of compound Hongcao freeze-dried powder for injection under strong light, high temperature and humidity conditions, and provide a reference for clinical drug safety and efficacy of this formulation. Method: According to the draft quality standards of compound Hongcao freeze-dried powder for injection, influencing factors test on its stability was carried out, changes of indexes including drug content, visible particles, particulate matter, hygroscopic weight, asepsis, pyrogen etc were determined.HPLC was employed to determine contents of isoorientin, orientin and scutellarin with mobile phase of acetonitrile-0.1% phosphoric acid (18:82) and detection wavelength at 350 nm. Result: Samples were loose-like brown substance, color was the 7th yellow standard solution, visible foreign matter, sterile, pyrogen and moisture absorption weight gain were in line with procisions.Peak areas and retention times of 17 chromatographic peaks in 10 batches of samples at high temperatures was stable with similarity of > 0.99, no vascular irritation, no hemolysis and agglomeration.Contents of isoorientin, orientin and scutellarin in samples were 5.80, 3.90, 16.50 mg, respectively.Each indicators all reached provisions of quality standards draft of compound Hongcao freeze-dried powder. Conclusion: Compound Hongcao freeze-dried powder for injection has good stability.

      摘要:Objective: To investigate effects of different processing methods on heavy metals and effective components in cut of Notoginseng Radix et Rhizoma. Method: Cut of Notoginseng Radix et Rhizoma was treated by rinsing, soaking and polishing.Contents of As, Cd, Pb and Hg were determined by inductively coupled plasma mass spectrometry (ICP-MS).HPLC was used to analyze contents of ginsenoside Rg1, ginsenoside Rb1 and notoginsenoside R1 with mobile phase of acetonitrile-0.05% phosphoric acid for gradient elution, flow rate of 1.6 mL·min-1 and detection wavelength of 203 nm. Result: Rinsing was the best way of all.After rinsing, contents of As, Cd, Pb, Hg decreased by 1.24%, 58.33%, 51.06%, 2.70%, respectively.However, contents of As and Cd increased by polishing.The content of total saponins in different processing methods ranked by rinsing > polishing > untreated > soaking. Conclusion: Rinsing can both decrease contents of heavy metals and maintain the content of saponins, so it can be used as a processing method for Notoginseng Radix et Rhizoma.

      摘要:Objective: To provide a reference for industrial production and quality control of Jiangtang Xiaozhi tablets by optimizing its molding process and establishing determination of ursolic acid in this preparation. Method: With appearance, friability, disintegration time and others as indexes, molding process of Jiangtang Xiaozhi tablets was optimized by single-factor tests.HPLC was employed to determine the content of ursolic acid in Jiangtang Xiaozhi tablets with mobile phase of methanol-0.05% phosphoric acid water (90:10), column temperature of 2 ℃ and detection wavelength at 215 nm. Result: With 15% pregelatinized starch and 5% lactose as filling agent, 92% ethanol as wetting agent, adding 3% crosslinking polyvinylpolypyrrolidone internally and externally as disintegrant, 0.3% magnesium stearate as lubricant.A good linearity of ursolic acid was in the range of 0.398-3.980 μg, the average recovery was 96.96%, the mass fraction of ursolic acid in samples was 2.22 mg·g-1. Conclusion: This optimized molding process is stable and feasible.This determination is simple, accurate, sensitive with a good reproducibility.

      摘要:Objective: To analyze main active compositions of drug-containing medium composed by monascus and Polygoni Multiflori Radix after fermentation and optimize its fermentation technology. Method: Qualitative and quantitative analysis were established by UV-visible full wavelength scanning method and ultraviolet spectrophotometry, based on single factor tests, with yields of monacolin K, free-anthraquinone, stilbene glucoside and combined-anthraquinone as indexes, orthogonal test was adopted to investigate effects of fermentation time, amounts of monascus and Polygoni Multiflori Radix on fermentation technology. Result: Contents of stilbene glucoside, combined-anthraquinone and total anthraquinone showed a downward trend, but free-anthraquinone showed a upward trend.Optimum fermentation conditions were as follows:Polygoni Multiflori Radix amount of 5%, fermentation temperature at 28 ℃, monascus amount of 6%, fermentation time of 7 d.Yields of stilbene glucoside, combined-anthraquinone, free-anthraquinone and monacolin K were 37.84, 0.89, 1.54, 0.41 mg·g-1, respectively. Conclusion: Polygoni Multiflori Radix has a promoting process on growth of monascus and its secondary metabolites of monacolin K, but monascus reduces the content of combined-anthraquinone in Polygoni Multiflori Radix, which improves medicinal value of Polygoni Multiflori Radix.

      摘要:Objective: To provide a reference for resource development of Oldenlandia diffusa by optimize extraction process of total flavonoids from this medicinal herb. Method: Extraction time, ethanol concentration and amount were selected as independent variables, overall desirability of contents of rutin, isoquercitrin and quercitrin as dependent variable, Box-Behnken design-response surface methodology was used to optimize extraction technology.HPLC-DAD was adopted to determine contents of rutin, isoquercitrin and quercitrin with mobile phase of acetonitrile (A)-0.05% phosphoric acid solution (B) for gradient elution and detection wavelength of 254 nm. Result: Optimal extraction process was as following:extracted 2 times with 14-fold the amount of 73% ethanol for 1.5 h per time.Average extracting amounts of rutin, isoquercitrin and quercitrin were 2.529, 0.435, 0.215 mg·g-1, respectively.Deviation between the measured and predicted value of overall desirability was -1.24%. Conclusion: This extraction process is stable and feasible with high prediction accuracy, which can provide a reference for preparations development of total flavonoids from O. diffusa.

      摘要:Objective: To prepare compound amino acid chelated calcium by taking Mactra veneriformis as raw materials. Method: With chelation ratio and calcium content as indexes, based on single factor tests, orthogonal test was adopted to investigate effects of pH, reaction temperature, reaction time and molar ratio of calcium to amino acid on chelating process.Infrared spectroscopy was empolyed to analyze compound amino acid chelated calcium. Result: Optimum conditions was as following:pH of 4.5, reaction temperature of 55 ℃, reaction time of 2 h, molar ratio of calcium to amino acids of 1.5:1.The content of calcium was 79.57 mg·g-1.After coordination of compound amino acid and calcium, characteristic absorption of amino shifted form 3 056.49 cm-1 to 3 245.30 cm-1, characteristic absorption of carbonyl moved from 1 573.53 cm-1 to 1 559.65 cm-1. Conclusion: This chelating process is stable and feasible, reaction product is recognized as an amino acid chelated calcium, which can provide theoretical basis for industrial production of compound amino acid chelated calcium.

      摘要:Objective: To optimize extraction process of flavonoids from Schizonepetae Spica and provide a reference for development of this medicinal herb. Method: Ethanol concentration, the amount of ethanol and extraction time were selected as independent variables, overall desirability of extracting amounts of hesperidin, luteolin and apigenin as dependeng variable, data of overall desirabilities were fitted to a second-order polynomial equation and regressed by multiple linear, response surface methodology was used to optimize extraction technology and predictive analyze.HPLC was adopted to determine contents of hesperidin, luteolin and apigenin with mobile phase of acetonitrile-0.4% phosphoric acid solution for gradient elution and detection wavelength of 283 nm. Result: Optimal extraction technology was as following:extracted 2 times with 15-fold the amount of 73% ethanol for 2 h per time;extracting amounts of hesperidin, luteolin and apigenin were 3.991, 1.307, 0.589 mg·g-1, respectively.Overall desirability was 0.925 3, whose deviation was -2.14% by comparing with the predictive value. Conclusion: This optimized extraction technology is convenient, accurate and suitable for industrial production of total flavonoids from Schizonepetae Spica.

      摘要:Objective: The aim of this study was to provide a new approach to synthesize hydroperoxylated products of 20 (S) -protopanaxadiol, and to optimize the microbial transformation conditions by Mucor racemosus AS 3.205. Method: Biotransformation conditions were optimized by altering several influencing factors. The yields of two hydroperoxylated products of 20 (S) -protopanaxadiol were evaluated. Result: Two metabolites were isolated and identified as 25, 26-en-24 (R)-hydroperoxyl-20 (S) -protopanaxadiol and 23, 24-en-25-hydroperoxyl-20 (S) -protopanaxadiol. The optimal transformation condition was as following:an inoculation amount at 10%, substrate concentration at 0.25 mmol·L-1, sample injected after inoculated 48 h, transformation time of 5 d, culture temperature at 28 ℃, and shaking speed at 150 r·min-1. Conclusion: Microbial transformation system by M. racemosus AS 3.205 can be used to obtain hydroperoxylated products of 20 (S) -protopanaxadiol. The yields of two hydroperoxylated products could be greatly increased under the optimized condition.

      摘要:Objective: An HPLC assay was established to determine and compare the contents of isoguanosine and magnoflorine in Crotonis Fructus from different areas, and used for its quality control. Method: An Accurasil C18 column (4.6 mm×250 mm, 5 μm) was used with acetonitrile (A)-0.2% phosphoric acid-0.2% triethylamine solution (B) as the mobile phase (0-10 min, 5%A;10-12 min, 5%-15%A;12-29 min, 15%A;29-30 min, 15%-5%A). The flow rate was 0.8 mL·min-1. The column temperature was kept at 30 ℃. The detection wavelength was set at 275 nm. Result: Isoguanosine had good linearity in the range of 0.078 8-0.472 8 μg (r=0.999 9). The average recovery was 99.29% with RSD of 2.5%. Magnoflorine had good linearity in the range of 0.013 7-0.082 2 μg (r=0.999 9). The average recovery was 99.28% with RSD of 2.2%. The content of isoguanosine and magnoflorine in Crotonis Fructus from different areas varied notably. The content of isoguanosine from Sichuan was the highest with 17.57 mg·g-1, while the one from Guangxi was the lowest with 7.07 mg·g-1. And the Guizhou one was the highest in the content of magnoflorine with 3.42 mg·g-1, while the Guangxi one was the lowest with 1.00 mg·g-1. Conclusion: The method is convenient, rapid and accurate, which can be applied to the quality control of Crotonis Fructus.

      摘要:Objective: To determine the total phenylethanol glycosides in water extract, water eluate and 70% ethanol eluate of Lamiophlomis Herba by first derivative ultraviolet spectrophotometry. Method: Lamiophlomis Herba was extracted with water (18 times) 3 times, 1.5 h per time. The water extract was storage at 2-4 ℃, was dried under reduced pressure at 60 ℃. Water extract were separated and enriched by polyamide and macroporous resin, to obtain the water eluate and 70% ethanol eluate Ⅰ (polyamide column), 70% ethanol eluate Ⅱ (macroporous resin column). The total phenylethanol glycosides were determined by first derivative ultraviolet spectrophotometry at 224, 252, 256, 354 nm. Result: Recovery of the total phenylethanol glycosides was 99.71%, RSD was 2.5% at 354 nm. Conclusion: This method is special, sensitive, accurate and repeatable, and can be used for the quality analysis of the total phenylethanol glycosides in Lamiophlomis Herba.

      摘要:Objective: To study the correlation between color of powder, extracting solution and pigment components' contents, such as carotenoids, rutin of Lycii Fructus expressing different degree of color-changing. Method: The colorimeter was used to determine the color of powder, fat-soluble pigments and water-soluble pigments extracting solution of Lycii Fructus. The ultraviolet-visible spectrophotometry was applied to determine carotenoid content. High performance liquid chromatography (HPLC) was applied to determine rutin content in Lycii Fructus. Result: With the appearance color deepening in Lycii Fructus, the colorimeter measured powders' brightness, reddish, yellowish extent showed a decreasing trend.The powder color was strongly correlated with water-soluble pigment extract solution color, and pigment components' contents. Conclusion: It was feasible to objectively and quantitatively measure external color of Lycii Fructus by colorimeter.The powder color was correlated with pigment components' contents, which showed that with color evaluating its inner quality is scientific.It laid the foundation for further development of rapid quality evaluation method of Lycii Fructus.

      摘要:Objective: To analysis of the volatile compounds in Scutellaria barbata using three types of fibers including PA, PDMS and PDMS-DVB. Method: A headspace solid-phase microextraction (HS-SPME) method followed by gas chromatography-mass spectrometry (GC-MS) is described for the analysis of volatile compounds in S. barbata using three types of fibers. The volatile compounds of S. barbata were extracted by PA, PDMS and PDMS-DVB fiber individually. The relative content of each component was calculated by the normalization method. Result: 90 volatile compounds were found in S. barbata., using three types of fibers. There were 41 volatile compounds (83.61%) identified in PA extract;40 volatile compounds (94.63%) identified in PDMS extract;53 volatile compounds (89.10%) identified in PDMS-DVB extract. Among them, 13 compounds were common peaksin three types of fibers. Conclusion: The results made by the three types of fibers are quite different. more alkanes and ketones can be identified for PA, more acids and ketones can be identified for PDMS, more naphthalenes, alkanes, olefinsand ketones can be identified for PDMS-DVB.

      摘要:Objective: Determination of heavy metal elements in the species of Epimedium sagittatum and semi quantitative (SQ) content. Method: Using semiquantitative ICP-MS full quality axis scanning of Epimedium sagittatum solution in whole mass acquisition between m/z 5-260. Result: Epimedium sagittatum containing Mn, Fe, Co, Ni, Cu, Zn, Ga, Ge, As, Se, Rb, Sr, Y, Zr, Nb, Mo, Cd, Sn, Sb, Cs, Ba, La, Ce, Nd, Pr, Th, Sm, Ta, Hg, Pb, Bi about 31 kinds of heavy metal concentration in the range of Mg, 1.3×10-2-93.5 mg·kg-1, wherein the iron (Fe) and manganese (Mn) contents were 93.5, 76.6 mg·kg-1, two kinds of heavy metal elements content in the highest. Conclusion: And semi quantitative heavy metal species of Epimedium sagittatum were determined in this experiment (SQ) content, semi quantitative heavy metal Yuan Suyuan Epimedium sagittatum full quantitative determination and Chinese Pharmacopoeia 2010 edition contains three other sources of Epimedium determination of reference.

      摘要:Objective: To establish an effictive quality evaluation method of Heracleum candicans roots and determine contents of two active ingredients of bergapten and imperatorin in this medicinal herb. Method: Ten batches of H. candicans roots were collected, HPLC fingerprint was performed in a gradient mode of water-methanol, detection wavelength of 302 nm.Similarity was evaluated with similarity evaluation system for chromatographic fingerprint of tradition Chinese medicine (2004A) issued by state pharmacopoeia committee. Result: HPCL fingerprint similarity among 10 batches of H. candicans roots were more than 0.89 and 11 characteristic common peaks were demarcated.Contents of bergapten and imperatorin in samples were 0.117% and 0.709%. Conclusion: This method is simple and reproducible that can be used for identification and quality evaluation of H. candicans roots.

      摘要:Objective: To establish a method for simultaneously determining liguiritin, wogonoside, baicalein and wogonin in ethyl acetate extract of Xiaochaihu Tang by HPLC. Method: The separation was performed on Diamonsil C18(2) (4.6 mm×250 mm, 5 μm). The mobile phase consisted of acetonitrile and 0.1% phosphate acid was used as gradient elution, flow rate 1 mL·min-1.The detection wavelength was set at 276 nm. The column temperature was at 30 ℃. Result: Quantitative analysis of HPLC showed that the linear ranges of liquiritin, baicalin, wogonoside, baicalein and wogonin were 0.162-1.62, 0.24-2.4, 0.206-2.06, 0.221-2.21, 0.077-0.77 μg respectively. The average recovery was 98.2%, 103.3%, 97.9%, 100.6%, 103.0%. Conclusion: The method is simple, fast, reliable, and suitable for the quality control of ethyl acetate extract of Xiao Chaihu Tang.

      摘要:Objective: To isolate neurotrophic activity compounds from the fruits of Leonurus japonicus. Method: Silica gel, Sephadex LH-20 and preparative HPLC were used for isolation and purification. The structures of compounds were elucidated by MS and NMR spectra. Neurotrophic activity was evaluated on the neurite outgrowth of PC12 cells. Result: Five compounds were obtained and identified as aurantiamide acetate (1), auraptenol (2), 1-monolinolein (3), acacetin (4), and β-sitosterol glucopyranoside (5). Compound 1 could significantly enhance the neurite outgrowth of PC12 cells. Conclusion: Compounds 1-5 were isolated for the first time from the genus Leonurus, compound 1 exhibited neurotrophic activity.

      摘要:Objective: To investigate contents of indigo and indirubin in Baphicacanthus Cusiae Rhizoma et Radix from different altitude, habitats, harvest period, and processing method. It can provide the basis for the determination of the best habitat, harvest time and processing method. Method: The HPLC conditions as follows:Agilent ZORBAX Eclipse XDB-C18 (4.6 mm×150 mm, 5 μm), mobile phase Methanol-Water, detect wavelength 300 nm, flow rate 1.0 mL·min-1, column temperature 30 ℃. Result: The linear range of indigo was 0.031-0.314 μg(r=0.999 5), recovery rate were 100.8%, RSD was 2.2%.The linear range of indirubin was 0.02-0.199 μg(r=0.999 8), recovery rate were 100.2%, RSD was 2.3%. Conclusion: The contents of indigo and indirubin in Baphicacanthis Cusiae Rhizoma et Radix of different altitude and different habitats, different harvest period, different preliminary working method in Guizhou exists a large difference.

      摘要:Objective: To establish a method for simultaneous determination of liquiritigenin, isoliquiritigenin and licochalcone A in Ganpuquban gel. Method: HPLC was applied on an Agilent HC-C18(4.6 mm×250 mm, 5 μm) column by a gradient elution using (A) 0.05% phosphoric acid and (B) acetonitrile as the mobile phase(0-4 min, 80%A;4-18 min, 80%-65%A;18-22 min, 65%-62%A;22-25 min, 62% A, 25-30 min, 62%-55%A;30-35 min, 55%A, 35-45 min, 55%-50%A;45-50 min, 50%-55%A;50-60 min, 55%-80%A). Detection wavelengths were set at 276, 368 nm. The f low rate was 0.8 mL·min-1, the column temperature was maintained at 30 ℃. Result: The linear ranges of liquiritigenin, isoliquiritigenin and licochalcone A were 0.050 4-0.403 2 μg (r=1), 0.039 4-0.315 2 μg (r=1), 1.638-8.192 μg (r=0.999 9). The average recoveries were 98.81% with RSD 1.7%, 99.16% with RSD 1.2% and 97.68% with RSD 1.0%, respectively. Conclusion: The established method is easy, accurate with high specificity, which can be used to determine the three components in Ganpuquban gel.

      摘要:Objective: To establish an HPLC method for the determination of loganic acid, gentiopicroside, swertiamarin, sweroside, shanzhiside methylester in Gentiana straminea. Method: The assay was performed on a Diamonsil C18 column (4.6 mm× 250 mm, 5 μm) eluted with acetonitrile-0.05% phosphoric. The detection wavelength was 240 nm, the flow rate was 1.0 mL·min-1, and the column temperature was set at 30 ℃. Result: The calibration curves were linear in the ranges of 0.042 1-8.420 0 μg(r=0.999 7) for loganic acid, 0.094 2-18.830 0 μg(r=1.000 0) for gentiopicroside, 0.012 4-2.471 2 μg(r=0.999 9) for swertiamarin, 0.009 9-1.971 2 μg (r=1.000 0) for sweroside, 0.011 3-2.260 0 μg(r=0.999 0) for shanzhiside methylester. The average recoveries were 95.39%-103.75%(RSD≤2.6%). Conclusion: The method is simple and accurate for the quality control of G. straminea Radix.

      摘要:Objective: To establish an HPLC method for the determination of rutin, hyperoside, quercitrin, quercetin, kaempferol and isorhamnetin in Rhododendron anthopogonoides, while using principal component analysis to evaluate its quality. Method: Samples were extracted with 75% methanol by ultrasound. The separation was performed on a C18 column (4.6 mm×250 mm, 5 μm)with acetonitrile-0.2%H3PO4 as mobile phase in a gradient elution. The flow rate was 1.0 mL·min-1, the wavelength was set at 365 nm. Result: Rutin, hyperoside, quercitrin, quercetin, kaempferol, and isorhamnetin have good linearity in the range of 0.009-0.194 9, 0.007 1-0.141 6, 0.0.188 0-0.375 2, 0.027 1-0.434 0, 0.010 8-0.216 0, 0.005 8-0.116 6 μg, respectively. The recoveries were 100%-105%, RSD were < 3%. Conclusion: The established HPLC method was simple, accurate, reproducible, and reliable. Depending on the chemical indicators, using principal component analysis can be carried out quality assessment for R. anthopogonoides.

      摘要:Objective: To make full use of limited land resources by comparing different plantling patterns of Cyathulae Radix. Method: Three plantling patterns were selected, such as rotation cropping (previous plant was Coptidis Rhizoma), continuous cropping (previous plant was Cyathulae Radix) and ordinary planting pattern (no previous plant).Emergence time, plant growth and disease of Cyathulae Radix in different plantling patterns were compared. Result: There was no significant difference between rotation cropping and normal planting patterns on emergence time, plant growth and two main kinds of disease (white rust and root knot nematode), but these two patterns were better than that of continuous cropping pattern. Conclusion: Rotation planting pattern can be used in Cyathulae Radix and Coptidis Rhizoma.

      摘要:Objective: To investigate influence of temperature on quality of Dendrobii Officinalis Caulis tissue culture seedlings and provide a reference for artificial cultivation of this medicinal herb. Method: Dendrobii Officinalis Caulis tissue culture seedlings at multiplicative and rooting stage were setted in the environment of humidity, light intensity and illumination time controlled, setting temperature were 15, 20, 25, 30, 35 ℃, growth conditions of tissue culture seedlings were observed.Pearson correlation was analyzed, variance analysis was adopted to analyze growth index difference among each temperature. Result: Tissue culture seedlings's quality was good at 25-30 ℃ and they showed good growth and reproductive capacity.Pearson correlation analysis showed that temperature with seedling number and plant height were significantly correlated at 0.01 level in proliferation stage;temperature with plant height, leaf number and root number were significantly correlated at 0.01 level in rooting stage.LSD multiple comparison showed that difference of seedling number and plant height were obvious in proliferation stage, difference of leaf number, root number and plant height were obvious in rooting stage. Conclusion: Temperature is the key factor for quality of Dendrobii Officinalis Caulis tissue culture seedlings and the most suitable temperature is 25 ℃.

      摘要:Objective: To establish a method for simultaneously determining plasma concentration of five kinds of phenolic acids from Salvia miltiorrhiza extract in rat plasma by LC-MS/MS. Method: LC-MS/MS was adopted to determine contents of lithospermic acid, rosmarinic acid and salvianolic acid A, B, C with mobile phase of 0.1% aqueous formic acid-equal volume mixture of methanol and acetonitrile for gradient eluent, flow rate of 0.6 mL·min-1.Mass spectrum was operated by an electrospray ionization probe in the negative ion mode.Scan was in multiple reaction ion monitoring mode.Ion pairs for quantitative analysis of this five phenolic acids were m/z of 537.3/295.1, 359.1/161.0, 493.3/295.1, 717.5/519.1, 491.2/293.2.Determination of this five phenolic acids could be accomplished within 7 min. Result: Linear ranges of salvianolic acid A, B and lithospermic acid were 0.78-100 μg·L-1, linear ranges of rosmarinic acid and salvianolic acid C were 0.39-50 μg·L-1.Their inter-day and intra-day precisions represented by RSD were within 15%, their accuracies were between 92%-112%. Conclusion: A sensitive, accurate and easy LC-MS/MS method has been successfully developed, which can be applied to pharmacokinetic study of five phenolic acids from S. miltiorrhiza in rat plasma.

      摘要:Objective: To investigate the biological basis of hypertension with hyperactivity of liver fire of macro characterization and behavioral tests, micro physicochemical indexes, disproof prescriptions. Method: Fifty-five SHR rats with hyperactivity of liver fire were divided into model group, Sancao Jiangya decoction (SCD) group, Sini San decoction (SND) group and nifedipine group according to Latin square method, Wistar-Kyoto (WKY) rats as control group. The macro characterization, behavioral indexes and the systolic blood pressure (SBP) were collected. Enzyme-linked immunosorbent assay(ELISA) was used to detect the serum concentration of 5-hydroxytryptamine(5-HT), angiotensin Ⅱ (ANG Ⅱ), norepinephrine (NE) after the rats were administrated for 4 and 6 weeks. Result: Compared with control group, the SBP and irritable degree increased, revolving tolerance time decreased, and serum 5-HT, ANGⅡ, NE level was higher in model group. Compared with model, the SBP in SCD group and nifedipine group reduced after treated for 4, 6 weeks, (18, 20 weeks of age), the SBP in SND group was stable and increased in the 16 weeks, and then decreased. The symptoms of liver fire hyperactivity improved, and it was not obvious in SND group, and serum 5-HT, ANG Ⅱ, NE level was lower in SCD group compared with model group. Conclusion: Fourteen-eighteen weeks is the stable time window of hyperactivity of liver fire. SHR, 5-HT, ANG Ⅱ, NE may be used as the objective index of hypertension of hyperactivity of liver fire syndrome.

      摘要:Objective: To investigate the effect of replenishing kidney-essence and removing phlegm on the ability of learning and memory and antioxidant in APP/PS1 double transgenic mice. Method: APP/PS1 double mice were randomly divided into model group, western medicine group(6.5×10-4g·kg-1·d-1), Bushen Huatan Yizhi recipe (BSHTYZR) high dose group (15.6 g·kg-1·d-1), low dose group (7.8 g·kg-1·d-1), 10 in each groups. Blank group mice (C57BL/6J) were 10 months of age with the same genetic background mice. After intragastrical administration for 3 months, the ability of learning and memory was assessed by the step down avoidance test and shuttle box test;colorimetric assay was used to measure mouse brain tissue superoxide dismutase (SOD) and malondialdehyde (MDA) content. Result: Step down avoidance test results showed that compared with model group, each group of mice significantly prolonged the incubation period and the number of errors decreased significantly (P<0.01);shuttle box test results showed that compared with model group, the number of active avoidance in treatment groups increased significantly (P<0.01, P<0.05), and active avoidance incubation period, the passive avoidance period significantly shorten (P<0.01);shuttle box test results showed that compared with the model group, in the rest of the administration group the small active avoidance significantly increased the number of rats (P<0.01, P<0.05), while the active avoidance latency and passive avoidance latency was shorter (P<0.01);in addition, in BSHTYZR high dose group and low dose group SOD activity of brain tissue increased significantly compared with the model group (P<0.01), MDA was significantly lower than the model group (P<0.01). Conclusion: BSHTYZR has a certain improvement on APP/PS1 double transgenic mice learning and memory, and may be related to the antioxidant effects.

      摘要:Objective: To investigate the effects of the ethanol extract from Schizocapsa plantaginea on the growth and angiogenesis of SMMC-7721 cell xenografts in nude mice and the antitumor mechanism of S. plantaginea. Method: S. plantaginea tubers were extracted by reflux in 80% ethanol. The human hepatocellular carcinoma SMMC-7721 cell model was established in BALB/c nude mice and the animals were randomly divided into model group, sorafenib group and high, medium, low dosage of S. plantaginea groups. The animals in S. plantaginea groups were administered 107, 71, 54 mg·kg-1·d-1 the ethanol extract and in sorafenib group were administered 30 mg·kg-1·d-1sorafenib by gavage once daily for 21 days. The animals in model group did not perform any treatment. The tumor growth was observed in xenografts model in nude mice and angiogenesis was observed by examining the expression of CD34 and vascular endothelial growth factor (VEGF) in tumor tissues by immunohistochemistry. Result: The tumor weight and the relative tumor volume in high and medium dosage of S. plantaginea groups and sorafenib group was significantly lower than that in model group. And the tumor inhibitory rates of those three groups were 47.71%, 21.27% and 51.75%. The levels of microvessel density (MVD) and VEGF in tumor tissue were also significantly lower (P<0.01) in high dosage of S. plantaginea group and sorafenib group than in model group. The MVD in medium dosage of S. plantaginea group was less (P<0.05) than that in model group. Conclusion: The ethanol extract from S. plantaginea can inhibit the growth of SMMC-7721 cell xenografts in nude mice and angiogenesis in tumor tissue, and its underlying mechanism is maybe due to its ability to reduce the expression of VEGF.

      摘要:Objective: To study effect of Danggui Shaoyao San (DSS) on aquaporin-2(AQP2) expression in the kidney of rats with cirrhosis ascites, and explore the mechanism. Method: Healthy male SD rats were randomly divided into 6 groups:normal, model, glycyrrhizin(0.052 5 g·kg-1), low, middle and high dose group of DSS(4.3, 8.6, 17.2 g·kg-1). After 1 week, except the normal group, the other rats were given 35% phenobarbital solution as drinking water, after 1 weeks normal water was recoveried, and the model was established by intraperitoneal injection of CCl4:at the 2-3 weeks 12% CCl4 oil solution was given, next two weeks 14% was given, then 16% was given until 14th week, twice a week, the volume being 10 mL·kg-1, the normal group was given equal volume of saline. At the same time, the treatment group was orally given corresponding drugs until 13th week. Then metabolic cages were used to collect urine in 24 hours for measuring the urine volume. The filter paper was used to absorb ascites.The immunohistochemistry was used to detect the expression of vasopressin Ⅱ style receptor (V2R) and AQP2 of kidney. The protein expression of V2R and AQP2 was detected by Western blotting. Result: Compared with the model group, in model group rats 24 h urine volume decreased significantly, the amount of ascites was significantly increased (P<0.05). Compared with the model group, urine volume significantly increased in 24 hours and the amount of ascites decreased differently in groups of DSS (P<0.05), the expression of V2R was significantly increased in rat kidney, kidney tissue AQP2 expression increased. Conclusion: DSS up-regulate the expression of AQP2 in the kidney of rats with cirrhosis ascites, which may be one of the mechanisms.

      摘要:Objective: To study the protective effective of specnuezhenide on glomerular mesangial cells suffering high glucose injury. Method: The effect of specnuezhenide on the cell vitality of cultured mice glomerular mesangial cells was measured with MTT method(n=6), and the toxic concentrations were identified and excluded from the subsequent treatments. The glomerular mesangial cells were stimulated with high concentration glucose(25 mmol·L-1)to mimic diabetic nephropathy, and was administrated to cells were treated using specnuezhenide at dose of 100, 50, 25 μmol·L-1 for 48 hours. The cell apoptosis rate was measured with flow cytometry (n=10), and the protein expressions of B cell lymphoma/leukmia-2(Bcl-2), Bcl-2 associated X protein(Bax), and cleaved-caspase 3 in glomerular mesangial cell were dectced with Western blot(n=3). Result: The high glucose significantly increased the apoptosis rate in glomerular mesangial cells (compared with the normal group, P<0.05), and the protein expression levels of cleaved-caspase 3 and Bax and decreased expression of Bcl-2, resulting in a significant a decline in the ratio of Bcl-2/Bax. Specnuezhenide at doses of 100, 50, 25 μmol·L-1 significantly inhibited the high glucose-induced glomerular mesangial cells apoptosis, and significantly prevented the up-regulations of cleaved-caspase 3 and Bax and the decrease of Bcl-2 expression, thus increasing the Bcl-2/Bax ratio (P<0.05). And this effect showed a concentration-dependent manner. Conclusion: Specnuezhenide has a significant protective effect on high glucose-induced glomerular mesangial cell apoptosis, which may be related to its up-regulating the expression of Bcl-2 and down-regulating with the inhibition of activation of caspase 3.

      摘要:Objective: To observe the influence of Ditan Tang on the content of β-amyloid(Aβ1-42) and expressions of presinilin(PS)-1, PS-2 in hippocampus of senile mild cognitive impairment(MCI) rats. Method: Sixty 13 months old rats were randomly divided into normal group, model group, Donepezil 0.9 mg·kg-1 group, the high dose and low dose groups(8.55, 4.28 g·kg-1) of Ditan Tang, 12 three months old rats were as young normal group. The senile MCI rats model was established by subcutaneous injection of D-galactose for 8 weeks and fed with semi-high-fat diet for 4 weeks in model group, Donepezil group, the high dose Ditan Tang group, and low dose Ditan Tang group. The learning and memory ability was tested by Morris water maze test in rats. Enzyme linked immunosorbant assay(ELISA) method was used to detect the levels of Aβ1-42in hippocampus. Levels of PS-1 and PS-2 in hippocampus were detected by immunohistochemistry. Result: Compared with normal group, the escape latency in model group rats was significantly prolonged (P<0.01), the content of Aβ1-42 in hippocampus increased significantly (P<0.01), and the expressions of PS-1 and PS-2 in hippocampus also increased (P<0.05).Compared with model group, the escape latency of Ditan Tang group(high dose and low dose) were significantly shorten (P<0.01, P<0.05). The content of Aβ1-42 in hippocampus on both high and low dose group of Ditan Tang decreased (P<0.01, P<0.05). The expressions of PS-1 and PS-2 in hippocampus on both high and low dose group decreased significantly (P<0.01). Conclusion: The Ditan Tang may decrease the contents of Aβ1-42 and the expressions of PS-1 and PS-2 in hippocampus. It might be reason of improving cognitive dysfunction in rats.

      摘要:Objective: To observe the effect of Hui medicine Zhali Nusi Fang (ZLNS) combined bone marrow mesenchymal stem cells (BMSCs) transplantation on neurons and P-glycoprotein (P-gp) expression in rats after cerebral ischemia reperfusion injury (CIRI). Method: Two hundred and fifty SD rats were randomly divided into the sham-operated group, the model group, the ZLNS group, the BMSCs transplantation group and the ZLNS combined BMSCs transplantation group. The rats except the 10 rats in the sham-operated group were subdivided into day 1, 3, 7 and 14 groups of 15 each. Middle cerebral artery occlusion (MCAO) model was duplicated by inserting a nylon thread. BMSCs were cultured and amplified by a whole bone marrow adherence method. Drugs were given to the rats by intragastric administration (14.6 g·kg-1·d-1), BMSCs suspension solution were transplanted into brain through carotid artery (2×106/200 μL). Rat brain was taken out at 1, 3, 7, 14 days after transplantation. The neuronal density (ND) in hippocampal CA1 area and brain pathological damage (BPD) were observed and determined. The expression of P-gp was measured by using an immunohistochemical method. Result: Compared with the sham-operated group, ND decreased (P<0.01), BPD increased (P<0.01, P<0.05), and the P-gp expression increased significantly (P<0.01) in the model group. Compared with the model group, the ND increased at day 3, 7 and 14 in the ZLNS, transplantation and combination groups (P<0.01, P<0.05), the P-gp expression decreased significantly in the ZLNS, transplantation and combination groups (P<0.01), the BPD decreased in all groups, and the results were obvious at day 7 and 14 (P<0.01, P<0.05). Compared with the transplantation group, the ND increased at day 1 in ZLNS group (P<0.01), decreased at day 14 in ZLNS group, and increased in combination groups (P<0.01), the BPD alleviated at day 7, 14 in the combination group (P<0.05), the P-gp expression decreased at day 1, 3 and 7 in the ZLNS groups, and the results were obvious at day 1 (P<0.05) and increased at day 14 (P<0.05), the P-gp expression decreased significantly in all combination groups (P<0.01). Compared with the ZLNS group, all the indicators in the combination groups were improved (P<0.01, P<0.05). Compared with the same time group, all changes were more obvious at day 7 and were improved at day 14. Conclusion: The ND in hippocampal CA1 area and BPD had a variable degree of damage in rats after cerebral ischemia. Both ZLNS and BMSCs transplantation could improve the amounts and condition of neurons, and the effect was superior when they were combined. Its mechanism may be related to the regulation of the dynamic expression of P-gp.

      摘要:Objective: To study the anti-gout effect and mechanism of Plantaginis Semen(PS) extract parts on the level of uric acid in hyperuricemia mice. Method: Five fractions including the petrol ether extract, the dichloromethane extract, the ethyl acetate extract, the butanol extract and the water-soluble extract were obtained by systematic solvent extraction. 90 Kunming mice were randomly divided into 9 groups:the normal group (normal saline), the hyperuricemia group (normal saline), the allopurinol group (0.01 g·kg-1), the total extract of PS(TPS, 25 g·kg-1) and the five fractions groups (25 g·kg-1).The hyperuricemia mice were induced by using oxygen oxazine acid potassium. The mice were administrated of the corresponding medicines for 7 days, and then the level of uric acid and the activity of xanthine oxidase were observed. Result: TPS and five fractions were obtained. Compared with the normal group, the level of uric acid and the activity of xanthine oxidase increased obviously in the hyperuricemia group (P<0.01). All treatment groups could reduce the level of serum uric acid obviously, and inhibit the activity of xanthine oxidase of hyperuricemia mice (P<0.05, P<0.01). Conclusion: The PS and its different extracts have good anti-gout effect in hyperuricemia mice.

      摘要:Objective: To investigate the antidepressant effects of daidzein and its possible mechanism. Method: Kunming mice were randomly divided into the normal group, the model group, the Fluoxetine group(3.0 mg·kg-1) and the daidzein groups, 10 mice in each group. Low, middle and high dose of daidzein (10, 20, 40 mg·kg-1)were administered. The depression model was established by chronic unpre dictable mild stress(CUMS), the body weight and times of grooming in the open field test were detected respectively. Mice in each group was gavaged before one hour of the daily stress. The normal group and the model group were given physiological saline for a total of 21days.The behavior changes of body mass were observed by open field test in mice, and apoptosis of hippocampal neural cell morphology and cell DG area in mice brain tissue were observed by Nissl staining and TUNEL method. Result: Compared with the normal group, the body weight of model group was reduced(P<0.01), the score of levels and vertical motion in the open field test were decreased in model group(P<0.01, P<0.05), hippocampus neural cells in DG area was showed irregularly by Nissl staining and the number of apoptotic cells were increased, the difference of statistical was significance(P<0.01).Compared with the model group, the behavior score was improved in the Fluoxetine group and each daidzein group, the body weights of mice were increased(P<0.01), the score of levels and vertical motion in the open field test were increased (P<0.01), the changes of hippocampus were reduced. Conclusion: Daidzein can improve the behavior anomalie in mice who were induced by CUMS and have neuroprotective effect. The mechanism may be related to improving the nerve cell apoptosis.

      摘要:Objective: To study the analgesic mechanisms of two extraction part of Ceratostigma willmottianum(CW)in rat pain model induced by formaldehyde. Method: Fifty-six SD rats were randomly divided into 6 groups with 8 in each group, including normal group, model group, CW alcohol extract's ethyl acetate part group(30, 15 mg·kg-1), n-butanol part group(15, 8 mg·kg-1). Each group was given corresponding drugs by intragastric injection, respectively, once a day for 7 days. At the 7th day, the animals were fasted for 12 hours, and ig 40 min later, except normal group, the other groups were given subcutaneous injection of 10% formaldehyde solution 50 μL for establishing the pain models. The impact of CW active site was observed and serum glutamate and Ca2+ were detected with biochemical kit, and the expression of c-fos and substance P(SP) was assayed with immunohistochemeistry(IHC). Result: Compared with normal group, model group showed a typical pain behavior, amounts of c-fos and SP were detected in the brain and spinal cord, and raised serum glutamic acid concentration, Ca2+ content with significant difference (P<0.05), meaning that the pain model of rats induced by formaldehyde was successful.Compared with model group, the low dose of ethyl acetate part of CW alcohol extract(15 mg·kg-1) and n-butanol part(8 mg·kg-1)can obvious inhibition to the first phase damage response(P<0.05), make serum glutamic content, brain c-fos protein expression decreased significantly(P<0.05), serum Ca2+increased significantly(P<0.05);the high dose of two parts decreased the expression of c-fos in spinal dorsal cord significantly(P<0.05);both of high and low dose of CW alcohol extract's ethyl acetate and n-butanol part decreased the expression of SP significantly(P<0.05)in central nervous system(CNS). Conclusion: Two extraction parts of CW alcohol extract have exact analgesic action in rat pain model induced by formaldehyde, and its analgesic mechanisms may be related to inhibiting the intracellular Ca2+ concentration and glutamate content in the body, reducing the expression of c-fos and SP in CNS.

      摘要:Objective: To observe the antitumor and anti-angiogenesis effect of ellagic acid against the transplantation tumor of S180 and H22 in mice, and to explore its possible mechanism by investigating the protein expressions of platelet-derived growth factor-B (PDGFB), signal transducer and activator of transcription-3 (STAT3) and p-STAT3 in vivo. Method: Mice bearing S180 and H22 cells were randomly divided into 5 groups:the model group (5% CMC), the cyclophosphamide group (20 μg·g-1·d-1), the high-, middle- and low-dose ellagic acid groups (200, 100, 50 μg·g-1·d-1). The mice were administrated with corresponding medicines for successive 10 days. The tumor inhibitory ratios, body weight, the thymus and spleen index were measured, respectively. The effect on microvascular density, PDGFB, p-STAT3 and STAT3 expressions were detected by using an immunohistochemicalassay assay. Result: The tumor inhibitory ratios of the high-, middle- and low-dose ellagic acid were 35.3%, 10.6% and 5.6% on S180-bearing mice, and 36.3%, 38.8%, 20.6% on H22-bearing mice. While, no significant influence on the body weight was found. Compared with the model group, the spleen index was higher in the high- dose group (P<0.05), and the microvascular density were lower in the high-and middle-dose groups (P<0.05), the expressions of PDGFB, p-STAT3 and STAT3 were lower in all doses of ellagic acid groups. Conclusion: Ellagic acid could inhibit the tumor growth and angiogenesis against the transplantation tumor of S180, H22 effectively. Its mechanism may be related to decreasing expression of PDGFB and further the expression and the phosphorylation of STAT3.

      摘要:Objective: To observe and screen anti-inflammatory fractions from Jiedu Huoxue Ziyin Recipe (JHZR). Method: The anti-inflammatory effects were observed by series of experiments including mouse auricular edema induced by xylen, increased vascular permeability induced by acetic acid in mice, paw edema caused by carrageen glue in mice, granuloma caused by implanted cotton ball and adjuvant arthritis (AA) in rats. Every experiment included 8 groups:the normal group, the model group, the indomethacin group (10 mg·kg-1) and the 5 groups of different fractions from JHZR (the water extract 966 mg·kg-1, the diethyl ether extract 59 mg·kg-1, the ethyl acetate extract 188 mg·kg-1, the n-butyl alcohol extract 22 mg·kg-1 and the ethyl alcohol extract 321 mg·kg-1). The effect on acute, subacute, and chronic inflammations of these extracts were observed. The effect on serum interleukin-6 (IL-6) and tumor necrosis factor-α(TNF-α) by JHZR in AA rats were observed. Result: Compared with the model group, the water and ethyl alcohol extracts of JDZR could inhibit the increase of mouse auricular edema by xylen, and the vascular permeability induced by acetic acid, and paw edema caused by carrageen glue, and granuloma caused by implanted cotton ball. Moreover, they could significantly reduce the activities of IL-6 and TNF-α in rats' serum. Conclusion: The water and ethyl alcohol fractions from JDZR have significant anti-inflammatory activity. The former fraction is effective on chronic inflammation, and the latter is effective on acute inflammation.

      摘要:Objective: To explore the chronic toxicity of Gansui Banxia Tang (GBT) on heart and kidney of rats. Method: Eighty healthy SD rats were randomly divided into four groups:the normal group, the high-, middle-, and low-dose GBT groups (37, 18.5, 9.25 g·kg-1). The corresponding medicines at 0.02 mL·g-1were intragastrically administrated to rats for 90 days. After the last dose, the blood samples of the half rats from each group were collected. The heart function indexes including creatine phosphate kinase (CK), lactic dehydrogenase (LDH) and α-hydroxybutyric dehydrogenase (α-HBDH), and the kidney function indexes including blood urea (UREA), serum creatinine (CREA) and uric acid (UA) were detected. The morphology and histology of the heart and kidney were observed by HE staining. The remaining rats in each group were observed continuously for two weeks, and the above indexes were detected again. Result: In medicine taking and during following observation, there were no significant differences for the relative function indexes and the change of morphology in heart and renal between any two groups. Conclusion: GBT has no remarkable effects on heart and renal function, and morphology of rats by long-term medication.

      摘要:Objective: To explore the chronic toxicity of Gansui Banxia Tang (GBT) on normal rat liver. Method: Eighty healthy SD rats were randomly divided into four groups:the normal group, the high-, middle-, and low-dose GBT groups (37, 18.5, 9.25 g·kg-1) of 20 in each group. The corresponding medicines at 0.02 mL·g-1were intragastrically administrated to rats for 90 days. After the last dose, blood samples of the half rats from each group were collected. The heart functions indexes including alanine aminotransferase (ALT), aspartate aminotransferase (AST), the ratio of AST/ALT, total bilirubin (TBIL), direct bilirubin (DBIL), and the blood lipid indexes including cholesterol (CHO), triglyceride (TG), high density lipoprotein (HDL), low density lipoprotein (LDL) were detected. The morphology and histology of the liver were observed by HE staining. The remaining rats in each group were observed continuously for two weeks, and the above indexes were detected again. Result: In medicine taking, compared with the control group, the ALT, AST, AST/ALT, TBIL and DBIL had significant changes in high-dose GBT group (P<0.01), the ALT, AST, DBIL (P<0.05) and AST/ALT(P<0.01) had significant changes in the middle-dose GBT group. The above indexes had no significant changes in the low-dose GBT group. The CHO, TG, HDL and LDL had no significant changes in all dose groups. The liver histopathological changes are gradually aggravated with the increase of the dose. While during the following observation, all the above indexes had no significant changes in all dose GBT groups. Conclusion: GBT has good effect on liver function and morphology of rats in a dose-dependent manner by long-term medication. Small dosage has little influence, the influence degree aggravates gradually as the increase of the dose, while the damage is reversible.

      摘要:Objective: To explore the chronic toxicity of Gansui Banxia Tang (GBT) on blood routine, blood glucose (GLU) and thyroid morphology of rats. Method: Eighty healthy SD rats were randomly divided into four groups:the normal group, the high-, middle-, and low-dose GBT groups (37, 18.5, 9.25 g·kg-1). The corresponding medicines at 0.02 mL·g-1were intragastrically administrated to rats for 90 days. After the last dose, the blood samples of the half rats from each group were collected. The blood routine indexes including red blood cell (RBC), white blood cell (WBC), hemoglobin (HGB), platelet (PLT) and GLU were detected. The morphology and histology of the thyroid were observed by HE staining. The remaining rats in each group were observed continuously for two weeks, and the above indexes were detected again. Result: In medicine taking, compared with the control group, the number of WBC significantly increased in high-dose GBT group (P<0.05), the number of WBC had no significant changes in the middle-and low-dose GBT groups, the number of RBC, HGB and PLT had no significant changes in all dose GBT groups. While in medicine taking and during the following observation, GLU showed no significant changes in all dose GBT groups. No obvious change for morphology of the thyroid was observed. Conclusion: GBT could increase the number of white blood cell reversibility by long-term, medication. But there were no effects on the number of RBC, HGB, PLT, GLU and thyroid morphology of rats.

      摘要:Objective: To explore the chronic toxicity of Gansui Banxia Tang (GBT) on the general situation, heart rate, cortisol level and adrenal gland morphology in normal rats. Method: Eighty healthy SD rats were randomly divided into four group:the normal group, the high-, middle-, and low-dose GBT groups (37, 18.5 and 9.25 g·kg-1). The corresponding medicines at 0.02 mL·g-1were intragastrically administrated to rats for 90 days.The general situation changes including action, activity, diet, sleep, color, excrement and urine were observed once daily after administration. The body weight was observed twice weekly and the heart rate was recorded once before and once after the experiment. After the last dose, the blood samples were collected and the morphology and histology of the adrenal gland were observed by HE staining. Result: In medicine taking, no significant differences for the change of body weight, heart rate and cortisol were found between any two groups. Moreover, there were no obvious changes for the general situation changes including action, activity, diet, sleep, color, excrement, urine and adrenal gland morphology. Conclusion: GBT has no remarkable effects on the general situation, heart rate, cortisol level and adrenal gland morphology in normal rats by long-term medication.

      摘要:Objective: The aim of this study was to compare the effect between Western medicine treatment combined with treatment of traditional Chinese medicine(Yiqi Yangxue Huoxue Fang) and western medicine alone in patients with refractory idiopathic thrombocytopenic purpura(rITP). Method: In this study, 35 rITP patients were divided into two groups which were treated with Western medicine combined with traditional Chinese medicine and western medicine alone, respectively. The curative effects were evaluated after 6 months. Result: Both total effective rate and complete reaction rate of rITP patients treated with combination of traditional Chinese medicine and western medicine were much higher than those treated with Western medicine alone. After treatment, platelet count(PLT) and plateletcrit(PCT) were much higher and mean platelet volume(MPV) and platelet distribution width(PDW) were much lower (P < 0.01) in the combination treatment group than those in western medicine alone treatment group. Platelet associated IgG(PAIgG) increased much higher in combination treatment group than those in western medicine alone treatment group, and so were the platelet-producing megakaryocytes in bone marrow smear. Conclusion: The combination treatment of traditional Chinese medicine (Yiqi Yangxue Huoxue Fang) with western medicine had a good curative effect in rITP patients.

      摘要:Objective: This study was undertaken to find out the polymorphism variations in the estrogen receptor-a gene PvuII (rs2234693) and XbaI (rs9340799) in the different traditional Chinese medicine(TCM) syndromes of hypomenorrhea. Method: This case-control study used the polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) analysis to find out the results. Result: ①The differences of genotype frequencies of ESR1 rs2234693 PP/Pp/pp genotype carriers were statistically significant among the different TCM syndromes of hypomenorrhea. The ESR1 rs9340799 XX/Xx/xx genotype carriers were statistically significant too (P<0.001). The study showed that the genotype frequencies of ESR1 pp and xx genotype were higher in the kidney deficiency than other TCM syndromes of hypomenorrhea.②The differences of allele frequencies of ESR1 P/p and X/x genotype carriers were statistically significant (P<0.001) among the different TCM syndromes of hypomenorrhea. The study showed that the genotype frequencies of ESR1 p and x alleles werehigher in the kidney deficiency than other TCM syndromes of hypomenorrhea. Conclusion: The results of the present study suggested that the ESR1 rs2234693 and rs9340799 gene were associated with the different TCM syndromes of hypomenorrhea.Patients with high frequencies of the p allele and x allele were more likely to respond to the hypomenorrhea with the kidney deficiency syndrome. The study showed that the ESR1 pp genotype and xx genotype were more susceptible to the kidney deficiency syndrome of hypomenorrhea.

      摘要:Objective: This study was conducted to observe the clinical efficacy of Bushen Huoxue decoction on patients with chronic heart failure(CHF), and its effect on immuno-inflammatory response. Method: In this study, sixty patients were divided into two groups by random number table, with 30 cases in the control group by routine western medicine treatment, while 30 cases in the treatment group treated by the combination of western conventional therapy with Bushen Huoxue decoction, taking a dose once for 14 days. We observed the changes of clinical symptom integrals, left ventricular end diastolic diameter(LVED), left ventricular ejection fraction (LVEF), brain natriuretic peptide (BNP), tumor necrosis factor α(TNF-α), interleukin-6(IL-6) and pentraxin-3(PTX-3) in the two groups. Result: The total effective rate of clinical symptom integrals in the treatment group (90%) was improved better than the control group (80%)(P<0.05). The level of [BNP(1 058.72±575.62) ng·L-1, IL-6(6.88±0.91) ng·L-1, TNF-α(251.54±52.72) ng·L-1, PTX-3(3.16±0.60) μg·L-1] in treatment group after treatment were improved significantly than the control group[(1 542.26±732.18) ng·L-1, (9.63±1.25) ng·L-1, (283.24±48.58) ng·L-1, (3.40±0.56) μg·L-1] (P<0.05).The improvements in LVED and LVEF in the both groups were better than those before the treatment, but the difference was not statistically significant. Conclusion: Bushen Huoxue decoction could obviously improve the CHF patient's cardiac function, reduce the contents of TNF-α, IL-6, BNP and PTX-3, restrain immuno-inflammatory response and improve prognosis.

      摘要:Objective: The aim of this investigation was to study the clinical efficacy and safety of Zhizhu Kuanzhong capsule in the treatment of functional dyspepsia and depression. Method: The Sixty four patients with functional dyspepsia and depression in our hospital from Jan 2013 to Feb 2014 were randomized into the control group (domperidone) and the observation group Zhizhu Kuanzhong capsule according to the random number, with 32 patients per group. The changes of dyspeptic symptom scores, depression scores and electrogastrogram on pre-and post-treatment, and the clinical efficacy, adverse reactions and recurrence rate in the two groups were observed and compared. Result: Compared with the control group, the dyspeptic symptom scores (3.6±1.3) VS (5.2±2.1)and depression scores (8.1±1.1) VS (15.1±1.8)were significantly decreased after treatment in the observation group (P<0.05);compared with the control group, the normal slow wave before meals (47.7±18.3) VS (40.8±15.6), normal slow wave after meals (52.3±14.0) VS (46.1±15.2), the main power ratio before meals and after meals (3.5±1.2) VS (2.6±1.0), and gastric emptying time (2.6±0.1) VS (4.4±0.2) were significantly improved after treatment in the observation group (P<0.05);compared with the control group, the cure rate (75.0% VS 50.0%)and total effective rate (93.8% VS 75.0%)were significantly increased and the recurrence rate (3.1% VS 21.9%)significantly decreased in the observation group (P<0.05). There were no obvious adverse reactions in the both groups during the treatment period. Conclusion: The treatment of Zhizhu Kuanzhong capsule can significantly improve the gastric motility, with better anti-depressant efficacy and high safety, and is worthy of wide clinical promotion.

      摘要:Objective: To observe the curative efficacy of syndrome dependent Kanglao decoction combined chemotherapy in treating multi-drug resistant tuberculosis (MDR-TB), and to investigate its influence on cellular immune regulation. Method: One hundred patients with MDR-TB were randomly divided into the control group (50 cases) and the observation group (50 cases) by a random number table. According to the different situation, patients in the control group received conventional chemotherapy. Based on the treatment of the control group, patients in the observation group added modified Kanglao decoction. All patients in two groups received 6 months of treatment. Sputum negative conversion and situation of computed tomography (CT) X-ray absorption were observed, and scores of traditional Chinese medical (TCM) syndrome were recorded. Before and after treatment, levels of T lymphocyte subsets (CD3+, CD4+, CD8+) were detected, blood routine of kidney and liver function were monitored. Adverse reaction was recorded during the treatment. Result: The comprehensive curative effect of the observation group was superior to that of the control group (P<0.05). Sputum negative conversion rate was 88% in the observation group, which was better than that of 62% in the control group during 6 months of treatment (P<0.05). The curative effect of chest CT in the observation group was 90%, which was superior to that of 64% in the control group during 6 months of treatment (P<0.05). Symptom score of TCM in the observation group was lower than that in the control group after 2, 4, 6 months of treatment (P<0.01). Levels of CD3+, CD4+ in the observation group were higher, while CD8+ standard was lower than those in the control group (P<0.01). The incidence of liver damage and leukopenia in the observation group was less than those in the control group (P<0.05). Occurrence frequency of side effects on digestion system, skin system, nerve system, etc, in the observation group were inferior to those in the control group (P<0.01). Conclusion: Kanglao decoction assisted with chemotherapy drugs could reduce sputum negative conversion, improve chest CT absorption, relieve clinical symptoms and side reaction, enhance organism cellular immunity function in treating MDR-TB. Its clinical comprehensive efficacy is superior to using Western medicine treatment alone.

      摘要:Objective: To observe the curative efficacy of Zhizhu Kuanzhong capsules combined rabeprazole sodium enteric-coated tablets in treating gastroesophageal reflux disease (GERD), and to investigate its influence on patients' quality of life. Method: One hundred patients with GERD were randomly divided into the control group (50 cases) and the observation group (50 cases) by a random number table. Patients in both two groups received 20 mg rabeprazole sodium enteric-coated tablets in the morning and evening. Patients in the control group added 5 mg mosapride citrate tablets thrice daily. Patients in the observation group added 3 Zhizhu Kuanzhong capsules thrice daily. All patients in two groups received 8 weeks of treatment. Before and after treatment, scores of GERD questionnaire (GERDQ), health inventory of SF-36 and deficiency of spleen and stagnation of Qi were graded. Result: After therapy, the curative effect of inflammation and Chinese medical syndrome in the observation group were superior to those in the control group (P<0.05). Eight dimensions scores including SF-36 scale physiological function, physiological function, body pain, general health, life energy, mental health, social function and emotional function in the observation group were better than those in the control group (P<0.05). Besides, scores of GERDQ and spleen deficiency and Qi stagnation syndrome in the observation group were inferior to those in the control group (P<0.05). Conclusion: Zhizhu Kuanzhong capsules jointed rabeprazole could reduce the clinical symptoms and signs, enhance the life quality of GERD patients. Its clinical curative effect is superior to using Western medicine treatment alone.

      摘要:Objective: To observe the curative efficacy and the mechanism of Ziren Zhixue decoction combined omeprazole in treating peptic ulcer bleeding (PUB). Method: Ninety patients with PUB were randomly divided into the control group (45 cases) and the combination group with traditional Chinese and Western medicine treatment (45 cases) by a random number table. Patients in the control group received 40 mg omeprazole injections by intravenous infusion once daily. Based on the treatment of the control group, patients in the combination group added Ziren Zhixue decoction. All patients in both two groups received 7 days of treatment. Hemostatic time and hospital stay were recorded. Before and after treatment, scores of the main symptoms and signs were graded. Levels of platelet α-granule membrane protein-140 (GMP-140), thromboxane B2(TXB2) and 6-ketone prostaglandin F1α(6-Keto-PGF1α) were detected. Result: The curative effects rate in the combination group was superior to that in the control group (P<0.05). Besides, hemostatic time and hospital stay of the combination group were shorter than those of the control group (P<0.01). The symptoms including melena, epigastric pain, lassitude, dizziness, palpitation, color of face, and the score of clinical signs in the combination group were inferior to those in the control group (P<0.01). After treatment, TXB2 level increased, 6-Keto-PGF1αlevel decreased in two groups (P<0.01). GMP-140 change in the control group was not obvious, while the GMP-140 increased in the combination group compared before the treatment (P<0.01). GMP-140 and TXB2 levels in the combination group were higher than those in the control group, while 6-Keto-PGF1α level was lower than that in the control group (P<0.01). Conclusion: Ziren Zhixue decoction combined omeprazole could reduce hemostatic time and hospital stay, improve clinical symptoms in treating patients with PUB. Its clinical curative effect is superior to using Western medicine treatment alone.

      摘要:Objective: To discuss the curative effects of Gushen Huazhuo decoction in treating renal damage during the early period in hypertension patients, and to investigate its influence on endothelin (ET-1) and nitric oxide (NO). Method: Two hundred and twenty seven patients were randomly divided into the combined group (114 cases) and the western medicine group (113 cases). Patients in the western medicine group received 4-8 mg candesartan cilexetil tablets once daily based on the treatment of calcium antagonists. Based on the treatment of the western medicine group, patients in the combined group added Gushen Huazhuo decoction. All patients in both two groups received 12 weeks of treatment. 24-hour ambulatory blood pressure was monitored before and after therapy. Levels of urine micro albumin (MA), β2-microglobulin (β2-MG), urinary N-acetyl-β-D-amino glucosaminidase (NAG), urinary transferrin (TRF) and cystatin C (CysC) were tested. Standards of serum blood urea nitrogen (BUN), creatinine (Cr), ET-1 and NO were detected before and after treatment. Result: The total comprehensive effective rate of the combined group was 86.84%, which was superior to that of 75.22% in the western group (P<0.05). After treatment, levels of 24-h average systolic blood pressure, 24-h average diastolic blood pressure, nocturnal average systolic blood pressure (nASBP) and nocturnal average diastolic blood pressure (nADBP) in the combined group were lower than those in the western group (P<0.01). Standards of MA, NAG, β2-MG, TRF and CysC in the combined group were inferior to those in the western group (P<0.01). Before and after therapy, BUN and Cr changes in two groups have no statistical significance. ET-1 standard in combined group was lower, while NO level was higher in the combined group than those in the western group (P<0.01). Conclusion: Based on conventional western medicine treatment, Gushen Huazhuo decoction could reduce blood-pressure level in hypertension patients during the early period. It has certain protective effect for hypertensive renal impairment. Besides, the therapy clinical curative effect is superior to using western medicine treatment alone. Its action mechanisms may be related to regulating vascular endothelial activity.

      摘要:Boschniakia rossica is a traditional herbal medicine in China and has many phamarcological effects, which have has been used to invigorat the kidney, strength Yang and relax bowel effect, and can also be used to cure waist cold pain, woman infecund disease and other reproductive system diseases combined with other drugs. B. rossica mainly distribute in northeast China, North Korea, Japan and Russia. In China, there is a small amount of B. rossica in Changbai Mountain District. The chemical constiuents of B. rossica are in a great variety of forms, and the aerial parts contain boschnialactone, boschnaside, boschniakine, polysaccharide, alkaloides and so on. Inorganic elements contained in B. rossica are K, Na, Ca, Mg, Fe, Zn, Mn, Cu etc. B. rossica polysaccharide has been widely studied in recent years. The polysaccharide has exhibited a wide spectrum of pharmacological effects in vitro and in vivo, involving anti-tumor, immune enhancement, anti-oxidant, liver production and so on. This review focuses on the advances in the structure, constitute and efficacy of the B. rossica polysaccharide in recent years to provide the theoretical basis for research work in future.

      摘要:Gastric ulcer is a common disease in digestive system. Although modern medicine has made great progresses in the pathogenesis and treatment of gastric ulcer, many problems still exists, such as the side-effects of drugs and the recurrence after treatment. Traditional Chinese medicine (TCM) has its advantages in gastric ulcer treatment. TCM doctors find that most of the patients with gastric ulcer in active state (GUIAS) exhibit 'toxin-heat’ syndrome. Thus in the treatment of GUIAS, they follow the principle of Qingre Jiedu Xiaoyong Shengji prescriptions. Xiaoyong Kuide Kang prescription has shown significant clinical effect on GUIAS patients. Using the Xiaoyong Kuide Kang prescription by following the principle of Qingre Jiedu Xiaoyong Shengji Prescriptions therapeutic strategy, significant clinical effect has been proved, which is also can be testified in Animal experiments. The mechanisms include inhibiting inflammation, increasing the expression of growth factors and trefoil factors, and promoting the release of prostaglandin E2 and so on. The Xiaoyong Kuide Kang consists of Astragali Radix, Coptidis Rhizoma, Taraxaci Herba, Sophorae Flavescentis Radix, Fritillariae Thunbergil Bulbus, Sepiae Endoconcha, Bletillae Rhizoma, Ginseng Radix et Rhizoma, Bupleuri Radix and Glycyrrhizae Radix et Rhizoma. Most of the single medicine extracts in the prescription have also been confirmed effective against gastric ulcer. It has been reported that liquorice and its extracts can not only kill Helicobacter pylori (Hp)directly but also inhibit Hp adhering to the stomach wall, thus relieving the infection;Coptidis Rhizoma, Taraxaci Herba and Glycyrrhizae Radix et Rhizoma can reduce inflammation associated with gastric ulcer;Bletillae Rhizoma mannan, Taraxaci Herba and saikoside can inhibit the secretion of gastric acid;some extracts have the antioxidant effect, such as dandelion ethanol extracts, phenolic compounds and polysaccharide succinic acid ester from Bletillae Rhizoma, and the total alkaloids from Coptidis Rhizoma as well;thunberg fritillary bulb and cuttlefish bone both can increase mucus secretion to protect gastric mucosa, the former can also increase the nitric oxide (NO) production;Ginseng Radix et Rhizoma root extract can protect gastric mucosa by inducing heat shock protein 27 (HSP27);Astragali Radix can promote the expression of epidermal growth factor and its receptor, accelerate the repair gastric mucosa and improve the quality of ulcer healing. Optimizing the process and elucidating the mechanism of Xiaoyong Kuide Kang would further improve the therapeutic efficacy of traditional Chinese medicine on gastric ulcer.

      摘要:Rhus genus has great potential for scientific research and resource development. It has long been used in the treatment and prevention of various diseases in many countries. The wide industrial applications and economic value prove the importance of the research on Rhus genus plants. We summarized the research progress of Rhus genus plants in chemical constituents and pharmaceutical activities by retrieving literatures at home and abroad. The compounds isolated from Rhus plants, mostly include triterpenes and their glycosides, flavonoids and their glycosides, and a small number of phenolic acids. The compounds from Rhus genus plants have various pharmacological activities, such as antiviral (anti-HIV activity), anticancer, antidiarrheal, anti-inflammatoryl, preventing decayed tooth, hepatopotective, etc. To carry out the studies related to Rhus genus plants is of great significance for discovering and developing new medicines and new medicinal resources.

      摘要:As traditional Chinese medicine, Acorus calamus exerts the function of understanding, eliminating phlegm, jianwei and educational effect. The study on its chemical composition has gained much attention. Based on the reviews by predecessors, we referred to the literatures on the chemical compositions of A. calamus by both domestic and abroad scholars, to systematically review the chemical components isolated from calamus, including terpenoids, styrene acrylic element class, flavonoids, steroid and alkaloids, with monoterpenoids and sesquiterpenoids as the most, mainly focusing on volatiles. This article systematically reviews the study on chemical components of A. calamus in the past decade, aiming to provide a reference for the further study on the chemical composition of A. calamus.

      摘要:As a traditional Chinese herbal medicine, Stigma Maydis contain a variety of active ingredients, including flavonoids, which have many important physiological activities. This paper systematically summarized the researches from domestic and abroad in the recent 35 years, to review the composition and pharmacological effects of flavonoids in Stigma Maydis, which can serve as a theoretical basis for its medicinal value. With the advances of equipments and technology, flavonoids of Stigma Maydisto gained more and more attention of scientists, the present review gives the details about the structureof flavone derivatives from Stigma Maydis in the domestic and abroad.Currently, there are 29 compounds isolated from Stigma Maydis, including flavonoids(luteolin, apigenin), flavonols (isorhamnetin), isoflavones(formononetin), flavonoid glycosides. As an active compound, the Stigma Maydis flavonoids have been found to have several pharmacological effects, such as, anti-hyperlipidemic effects, hyperglycemia reduction anti-diabetic, anti-cardiovascular, antioxidant, antitumor, antibacterial, and anti-fatigue activity. But in general, the flavonoids studied were mainly natural. It was still unclear about the pharmacological activities and mechanism of different monomer compounds. In addition, studies on pharmacological effects were just in vitro, further validation in clinical trials were required.

      摘要:The aim of this study was to explore the application of metabonomics technology in the research on the attenuation strategy by compatibility of traditional Chinese medicine (TCM). By analyzing the advantages of metabonomics used in the research on compatibility for attenuating toxicity of TCM, methods and content of compatibility for attenuating toxicity of TCM based on metabonomics was put forword. Metabolomics was a rapidly developed new technology after genomics, transcriptomics and proteomics, which is characterized by global, dynamic, comprehensive. Metabonomics studies display great potential in compatibility for attenuating toxicity of herbs as significant component of system biology. The application of metabonomics technology in the research on the attenuation strategy by compatibility of TCM should focus on four aspects, including identification of toxic components of drug metabolite group and effective ingredient, ascertainment of toxicity target organ of TCM and toxicity biomarkers, mechanism of toxicity control, and the analysis of correlation between toxic components of system modeling and attenuated activity of compatibility for attenuation synergia. Metabonomics has a vast potential in the research of attenuation strategy by TCM.

      摘要:Objective: To count toxic Chinese drugs preparations that contain aconitine, such as aconite and radix aconite praeparata, in the Compilation of National Standard for Traditional Chinese Medicines, to analyze quality problems and propose suggestions. Method: Couting and analyzing the contents of identification, detection, extractum and assay refer to toxic Chinese drugs preparations in the Compilation of National Standard for Traditional Chinese Medicines. Result: Toxic Chinese drugs preparations that contain Aconitine total 100, which mainly focused on the department of fractures and wounds, meridian and limbs system, internal medicine about kidney. Prescription decoction pieces of section preparation without inspection, content detection of sovereign drug and the toxic Chinese medicine. Conclusion: The Compilation of National Standard for Traditional Chinese Medicines published fairly early, required regularly modify because of cycle length of revised, re-evaluated and validated imperfect. Through statistical analysis of the following summarizes the standard problems in this stage, given the appropriate advice.
