      摘要:Objective: To optimize purification technology of total polysaccharides from Liuwei Dihuang biological preparation. Method: With the amount of crude polysaccharides as index,orthogonal test was adopted to optimize alcohol precipitation process.Based on single factor tests,taking composite score of deproteinization rate and retention rate of total polysaccharides as index,orthogonal test was adopted to optimize deproteinization process.With composite score of decolorization rate and retention rate of total polysaccharides as index,orthogonal test was adopted to optimize decolorization process. Result: Optimum alcohol precipitation process was as follows:alcohol precipitation concentration of 85%,alcohol sedimentation time for 6 h,relative density of solution concentration of 1.05.Optimum deproteinization process were as follows:volume ratio of total polysaccharide solution to trichloroacetic acid (TCA) of 1: 2,volume ratio of TCA-butanol of 1: 15,vibration time of 20 min,standing for 0.5 h.Optimum decolorization process was as following:decolorization temperature of 40 ℃,the amount of activated carbon 6%,decolorization time of 40 min.Quality of crude polysaccharides was 8.92 mg,deproteinization rate was 80.23%,decolorization rate was 74.84%,retention rate of total polysaccharides was 78.69%. Conclusion: These optimized extraction processes are stable and feasible,which are suitable for development of health products of total polysaccharides from Liuwei Dihuang biological preparation.

      摘要:Objective: To investigate effect of compatibility on alkaloids in Aconiti Radix Cocta from Wutou Tang. Method: HPLC-ESI-MS positive ion mode was adopted to analyze alkaloids in Aconiti Radix Cocta single decoction and Wutou Tang,parsing each component of MS2 and MS3 spectra,thus comparing alkaloids between Aconiti Radix Cocta single decoction and Wutou Tang. Result: Thirty-six common compositions were detected in Aconiti Radix Cocta single decoction and Wutou Tang,13 ingredients were detected in single decoction,which were not detected in Wutou Tang,such as dehydrared 10-OH-aconitine,dehydrated 10-OH-aconine,mesaconitine and so on. 10-OH-mesaconine in Wutou Tang was not detected in single decoction solution of Aconiti Radix Cocta. Conclusion: A lot of aconitine type alkaloids in Aconiti Radix Cocta are missing after compatibility,ingredients are clearly different.This established HPLC-ESI-MS can effectively separate various aconitine type alkaloids by providing molecular weight and structure information of components.

      摘要:Objective: To synthesize poly(ethylene glycol)-poly(lactic acid)-poly(β-amino ester) (PBAE) block polymer and prepare paclitaxel-loaded micelles to enhance its solubility in aqueous milieu. Method: PBAE was synthesized by Michael addition reaction,its structure was characterized by 1H-NMR,its dissociation contant of a base (pKb) was determined by acid-base titration,critical micelle concentration (CMC) of copolymer was measured by pyrene fluorescence spectroscopy.Paclitaxel loaded copolymer micelles were prepared by membrane hydration methods.Drug loading efficiency (DL) and entrapment efficiency (EE) of micelles were evaluated by high-performance liquid chromatography (HPLC).The size and size distribution of micelles were determined by nanoparticle size analyzer. The structure of micelles was manifested by transmission electron microscopy.In vitro release was performed in phosphate buffered saline at different pH values. Result: CMC of PELA and PBAE were 3.31,4.79 mg ·L-1,pKb values of them were 7.1 and 6.5,their DL were 12.37% and 12.05%,EE were 70.55% and 68.53%,respectively.Paclitaxel loaded micelles were stable up to one week at 4 ℃.In vitro release study showed that drug release from PELA micelles increased slightly with decrease of pH.However,drug release accelerated obviously upon acidic environment (pH 6.5 and 5.5) from pH responsive polymer micelles. Conclusion: PBAE shows high performance in increasing paclitaxel solubility,high drug loading and long stability,as well as pH controlled drug release from micelles.

      摘要:Objective: To investigate antioxidant activity of water extract and alcohol extract of Eriocauli Flos. Method: Scavenging effect of water extract and alcohol extract of Eriocauli Flos on superoxide anion free radical,DPPH radical and hydroxyl radical were determined by UV method,reduction ability of extract for Fe3+ was determined by Prussian law,and with butylated hydroxyanisole (BHA) as reference,chelating ability of extract on metal ferrous ion (Fe2+) was determined. Result: Water extract and alcohol extract of Eriocauli Flos had strong scavenging effects on three kinds of free radicals,half inhibitory concentrations (IC50) of water extract were 1.389,3.507,0.341 g ·L-1,while IC50 of alcohol extract were 0.792,5.525,0.192 g ·L-1.Extract had reduction ability on Fe3+,and had a strong chelating ability to Fe2+,effects increased with increasing of extract concentration,which showed corresponding dose-response relationship. Conclusion: Water extract and alcohol extract of Eriocauli Flos have strong antioxidant activity,and at a relatively low concentration,alcohol extract is stronger than water extract about antioxidant effect.

      摘要:Objective: To optimize deproteinization process of total polysaccharides from exocarp of unripe fruits of Juglans mandshurica. Method: With retention rate of total polysaccharides and deproteinized rate as indexes,effects of Sevage,trichloroacetic acid(TCA),papain,enzyme-Sevage and enzyme-TCA on deproteinization of total polysaccharides were compared and optimized by single factor tests and orthogonal test. Result: Effect of papain-TCA method to take off protein was the best,optimum conditions were as follows:papain dosage of 3%,hydrolysis temperature of 60 ℃, hydrolysis time of 2 h,pH of 6,the concentration of TCA 6%.Retention rate of total polysaccharides and deproteinized rate were 82.48% and 84.76%,respectively. Conclusion: Deproteinized effects on total polysaccharides from exocarp of unripe fruits of J. mandshurica have significant difference by different deproteinized methods,enzyme-TCA combination method is easy to operate with optimum deproteinized effect and short time-consuming.

      摘要:Objective: To optimize preparation process of Apocyni Veneti Folium sustained-release dropping pills for providing a reference for industrial production of this preparation. Method: Sustained-release dropping pills were prepared by solid dispersion method.On the basis of single factor tests,taking ratio of substrate to drug,ratio of polyethylene glycol 6000 (PEG6000) to stearic acid and material temperature as dependent variables,Box-Behnken design was conducted to optimize prepration process with appearance quality,pill weight difference and in vitro release as indexes. Result: Optimal preparation process was as follows:substrate-drug(6.77: 1),PEG6000-stearic acid (4.87: 1),material temperature of 66 ℃.Bias between the observed value and the predicted value were less than 3.0%. Conclusion: Box-Behnken surface response design is suitable for optimizing prepration process of Apocyni Veneti Folium sustained-release dropping pills with good predictability.These prepared dropping pills have good appearance quality,small weight difference and sustained-release effect.

      摘要:Objective: To optimize extracting process of rhynchophylline from Uncariae Ramulus Cum Uncis by enzymolysis method. Method: HPLC was adopted to determine the content of rhynchophylline with mobile phase of methanol-water (60: 40) and detection wavelength at 254 nm.With extraction rate of rhynchophylline as index,on the basis of single factor tests,effects of enzyme dosage,pH,enzymolysis temperature and time on extraction process were investigated by orthogonal test. Result: Optimal extraction conditions were as follows:dosage of cellulase 1%,pH of 4.5,enzymolysis temperature of 50 ℃,enzymolysis time of 2 h.Extraction rate of rhynchophylline was 0.54%,it increased 54.29% by comparing with heating reflux method. Conclusion: Enzymolysis method is suitable for extracting rhynchophylline from Uncariae Ramulus Cum Uncis,this optimized extraction process is stable and feasible,it is worthy of popularization and application for extraction of alkaloids from herbs.

      摘要:Objective: To optimize processing method of Polygonati Rhizoma with wine which recorded in the 2005 edition of Guizhou Province Processing Specification of Chinese Herbal Pieces,and compare with others. Method: Taking composite score of color score,water-soluble extract,ethanol-soluble extract,contents of total polysaccharides and total saponins as index,orthogonal test was adopted to optimize processing method of Polygonati Rhizoma with wine which recorded in the 2005 edition of Guizhou Province Processing Specification of Chinese Herbal Pieces,and compared with other processing methods. Result: The best processing method was steaming 3 h,moistening 3 h,repeated 4 times.This method ranked second in these five methods.Detection values of processing products among these five methods were very significant. Conclusion: This optimized method is one of the best.Different processing methods can lead to change of chemical composition and their dissolution.

      摘要:Objective: To optimize extraction process of Paozhenkang spray. Method: Taking composite score of the content of berberine hydrochloride and dry extract rate as index,orthogonal test was adopted to investigate influence of extraction time,extraction times,the amount of water on water extraction process.With composite score of the total content of aloe-emodin,rhein,emodin,chrysophanol,physcion and dry extract rate as index,orthogonal test was adopted to optimize alcohol extraction process by taking ethanol concentration,alcohol amount,extraction time and times as factors.Contents of berberine hydrochloride and five aglycones from Rhei Radix et Rhizoma were determined by HPLC. Result: Optimun water extraction process consisted of adding 12-fold of waters,decocting 3 times,90 min for each time,the content of berberine hydrochloride was 6.07%.Optimun alcohol extraction process was as following:refluxing extracted twice with 10-fold of 75% ethanol,75 min for each time;the total content of five aglycones was 1.96%. Conclusion: These optimized extraction processes are stable and feasible for industrial production of Paozhenkang spray.

      摘要:Objective: To investigate reproductive biology of Desmodii Styracifolii Herba. Method: Reproductive system,flowering habits,pollination pattern,pollen-ovule ratio (P/O),out-crossing index (OCI) and other reproductive biology characteristics of Desmodii Styracifolii Herba were measured by field observation,laboratory analysis,artificial bagging pollination and other ways. Result: Flowers of Desmodii Styracifolii Herba was bisexual,blossom peak time was from 9: 30 to 11: 00.Flowering time was about 45 days,and single one was 1 to 2 days.Numbers of daily blossom showed normal distribution patterns.Stigmas had receptivity 1 hour before blossom and after blossom in 8 hours,P/O was 8 000±1 225,OCI was 4.Artificial bagging test showed that breeding system of Desmodii Styracifolii Herba was mainly self-pollination,cross-pollination.Bees of Diptera and flies of Hymenotera were pollinators for Desmodii Styracifolii Herba. Conclusion: Breeding system of Desmodii Styracifolii Herba is mainly self-pollination,cross-pollination and pollinators are needed.

      摘要:Objective: To explore relationship between the content of total polysaccharides from Pseudostellariae Radix and altitude by determining height above sea level with the growth area in Guizhou,and provide theory foundation for quality assessment and scientific cultivation of Pseudostellariae Radix. Method: Ultraviolet speetrophotometry was used to dertermine the content of total polysaccharides,total polysaccharides from Pseudostellariae Radix was extracted by water extraction and alcohol precipitation method,sulfuric acid-anthrone was used as sprayed regents and detection wavelength was at 626 nm.Altitudes were determined by global positioning system. Result: The content of total polysaccharides in Pseudostellariae Radix varied significantly in different altitude,it was the highest of 281 mg · g-1 from Shibing county cattle big dam small toward the old,but it was the lowest of 23 mg · g-1 from Luotian,Wangzhuang,Qingzhen city.But the content of total polysaccharides had no relationship with altitude. Conclusion: The content of total polysaccharides in Pseudostellariae Radix from Guizhou is generally high,altitude has no significant effect on the content of total polysaccharides,but high altitude area should be avoided.Although Guizhou is not famous-region for Pseudostellariae Radix,it can be builded as main planting base.

      摘要:Objective: To study the chemical constituents and coagulation Factor Xa (FXa) inhibition activities of Semen Persicae. Method: Chromogenic-substrates and continuous rate methods were used to assess the FXa inhibition activities. Various chromatographic techniques were used to isolate and purify the major active compounds, and their structures were identified by spectral analysis. Result: The ethyl acetate part possessed inhibition of 60.27% to FXa, and ten compounds were isolated and identified as 9-octadecenoic acid-2,3-dihydroxypropyl ester (1), daucosterol (2), 2-hydroxy-2-phenylacetamide (3), benzoic acid (4), 2-hydroxy-9-octadecenoic acid (5), n-heptadecanoic acid (6), methyl elaidate (7), p-hydroxybenzoic acid (8), palmitic acid (9) and amygdalin (10). Conclusion: The ethyl acetate part was the major FXa inhibition active site. Compounds 1 and 3 were isolated from the family of Rosaceae for the first time, and compounds 5-8 were first reported in this plant.

      摘要:Objective: To find out the changes before and after the material basis of compatibility, and to discover the interaction among the process of compound mixing,through established the HPLC fingerprints of Yinqiao San(YQS) and Jun(J),Chen(C),Zuo(Z),Shi(S). Then investigated the relationship of compound compatibility of YQS on the whole. Method: The chromatographic condition was:Ultimate 3000 XRS ultra fast liquid chromatography, thermo C18(4.6 mm×250 mm, 5 μm) column, methanol-0.05% phosphoric acid water as the mobile phase gradient elution, the detection wavelength was 237 nm and flow rate was 1 mL · min-1. Researched and established the HPLC fingerprint of YQS and other groups, then used the fingerprint processing software to analys the data of each group HPLC fingerprint. Result: The similarities from high to low between YQS and other compatibility groups are:J, C, S (0.981)>J, C(0.959)>J, C, Z(0.953)>J, Z, S (0.806). The similarities from high to low between YQS and J, C, Z, S are:J(0.749)>C(0.573)>Z(0.136)>S(0.104). Compared with the single decoction added, the fingerprint areas of YQS decoction had 14 peaks decreased and 63 peaks increased. Conclusion: J and C were the main source of material basis of YQS. In this paper,from the perspective of the material basis composition,the datas analysed how important of J,C, Z and S in the whole prescription.This result made the traditional interpretation of prescription more persuasive.

      摘要:Objective: To established a simultaneously method to determine four kinds of tannins (gallic acid, catechin, tannic acid, ellagic acid) in leaf extracts of Sapium sebiferum. Method: Samples were detected on the ZORBAX Eclipse Plus C18 column (4.6 mm×250 mm, 5 μm), eluted with acetonitrile and 0.1% phosphoric acid as mobile phase in a gradient mode. The procedure was at column temperature of 25 ℃, detection wavelength of 270 nm and flow rate of 1 mL · min-1. Four tannins in leaf extracts of five S. sebiferum populations were then detected. Result: There were good linear relationships between the concentrations and the peak area of gallic acid, catechin, tannic acid, and ellagic acid in the ranges of 0.002-0.095 (r=0.997 1), 0.005-0.238 (r=0.999 7), 0.004-0.191 (r=0.999 2),0.001-0.048 g · L-1(r=0.999 4), respectively. The average recoveries were 97.4% with RSD of 2.0%,98.3% with RSD of 1.7%, 98.7% with RSD of 1.6%,and 98.1% with RSD of 1.7%, respectively. The mean concentrations of gallic acid, catechin, tannic acid, and ellagic acid in leaf extracts of five S. sebiferum populations were 0.112, 3.380 7, 17.139 3, 1.354 7 g · L-1, respectively. Conclusion: We found a simple, rapid and accurate method to simultaneously determine the concentrations of gallic acid, catechin, tannic acid, and ellagic acid in S. sebiferum leaves extracts within 30 minutes, and may provide methodological basis to the exploration of other plant resources that are rich in tannins.

      摘要:Objective: To analyze the component correlation of oleanolic acid and ursolic acid between Diospyros kaki and its eleven kinds of Taxilli Herba. Method: Oleanolic acid and ursolic acid were extracted with ethanol by ultrasound for 30 minutes, and determined by RP-HPLC. Agilent Eclipse XDB C18column (4.6 mm×250 mm, 5 μm) was used for the separation, with a flow rate of 1.0 mL · min-1 and column temperature of 25 ℃. The mobile phase were methanol-acetonitrile-0.05% solution of ammonium acetate (12:67:21) and detection wavelength was 210 nm. Result: There were good linearity in the range of 2.34-375 mg · L-1(r=0.999 9) and 5.21-834 mg · L-1(r=0.999 9) for oleanolic acid and ursolic acid, respectively. The average recovery rates were 99.57% and 99.72% for oleanolic acid and ursolic acid, respectively. In D. kaki, the content of oleanolic acid was 0.382 6-0.541 1 mg · g-1(branches) and 1.773 6-4.274 6 mg ·g-1 (leaves), and ursolic acid was 0.649 0-1.625 8 mg · g-1 (branches) and 6.023 7-9.398 0 mg · g-1(leaves). Ursolic acid was not detected in the eleven kinds of Taxilli Herba, and oleanolic acid was found only in Viscum ovalifolium and V. diospyrosicolum. The contents of oleanolic acid were 1.221 4 mg · g-1(in branches of V. ovalifolium), 10.815 8 mg · g-1 (in leaves of V. ovalifolium), and 7.158 4 mg · g-1 (in V. diospyrosicolum), respectively. Conclusion: The oleanolic acid and ursolic acid were accumulated in the branches and leaves of D. kaki. Oleanolic acid was accumulated in V. ovalifolium and V. diospyrosicolum, but ursolic acid was not accumulated. The oleanolic acid and ursolic acid were not accumulated in Macrosolen cochinchinensis, M. tricolor, Helixanthera parasitica, H. Sampsoni, D. pentandra, S. parasitica, S. parasitica var. graciliflora, Taxillus chinensis and T. maclurei.

      摘要:Objective: Determine the content of mangiferin in Anemarrhenae Rhizoma by near-infrared spectroscopy(NIRS). Method: Use the content of mangiferin in samples determined by HPLC as reference value. The quantitative model of mangiferin in Anemarrhenae Rhizoma was established by NIRS combining with partial least squares(PLS). Result: The correlation coefficients (R2) was 0.963 61, the root mean square error of calibration (RMSEC) was 0.061 6, the root mean square error of predication (RMSEP) was 0.088 1. Conclusion: The method is accurate, fast, simple. It can be used as a rapid detection method for Anemarrhenae Rhizoma.

      摘要:Objective: Study the change regulation on FTIR spectra of Codonopsis Radix before and after sulfur-fumigated process, and provide a basis for quality control of Codonopsis Radix. Method: Different quantities of sulfur were used to fumigate Codonopsis Radix. The powder, alcoholic and aqueous extracts of Codonopsis Radix before and after sulfur-fumigated processes were analyzed by FTIR, the further analysis was used by second derivative infrared spectroscopy. Result: Before and after sulfur-fumigated process, the difference of FTIR spectra was not obvious, while through high resolution of second derivative infrared spectroscopy the distinction was showed. The messages of infrared spectra are as follows:for powder of Codonopsis Radix, the height of absorption peaks in the range of 1 500-1 600 had obvious difference. And for alcoholic extract and aqueous extract, the same feature had appearance in the range of 1 500-1 600 cm-1, 1 715 cm-1. The height of absorption peaks of powder of Codonopsis Radix had a regular change along with the quantity of sulfur. Conclusion: The change regulation on FTIR spectra before and after sulfur-fumigated process could be utilized to distinguish the Codonopsis Radix whether or not to be sulfur-fumigated, and provide a basis for quality control.

      摘要:Objective: This research intends to study and indentify the chemical constituents of Caesalpinia minax. Method: On the basis of the regulation function of C. minax to interleukin-6(IL-6) and interleukin-8(IL-8), which is the key factors for periodontitis. Take the refluxing extraction and gradient extraction for extraction and separation. The compounds were isolated and purified by repeated chromatography on silica gel, thin layer chromatography and recrystallization. The structures were determined by MS, NMR, and physical and chemical data analysis. Result: 8 compounds were obtained. They were identified as stigmasterol (1), caesalmin C (2), bonducellpin F (3), β-sitosterol (4), β-amyrin (5), n-hexacosane (6), n-triacontane (7), simiarenol (8). Conclusion: Compounds 3 and 6-8 were isolated for the first time from C. minax. Bonducellpin F was a new cassane furanoditerpenoid. Simiarenol was triterpenoid.

      摘要:Objective: To explore the best total genomic DNA extraction method from the dried and fresh Corni Fructus and to compare the DNA quality as a foundation for future research. Method: The new modified CTAB method, SDS method were used to get DNA, the content of nucleic acid and agarose gel electrophoresis were tested. Result: The OD260/OD280 ratios of DNA extracted by using the new modified CTAB method A were between 1.7-1.9 and the agarose gel electrophoresis results showed that the bands of the DNA were both tight and distinct. Compared with the DNA samples from dried herbs, results showed that DNA quality from fresh herbs was better. Conclusion: New modified CTAB is a simple and fast method for extract the DNA in high quantity and quality from herbs, the obtained DNA by this method is suitable for further studies of molecular biology.

      摘要:Objective: To extract and separate polysaccharide from Melastoma dodecandrum and to analyze the monosaccharide composition. Method: The polysaccharide was extracted by the water and precipitated by alcohol, and then separated and purified by DEAE-cellulose column chromatography. Water-soluble neutral polysaccharide and acidic polysaccharide were obtained. The monosaccharide composition of the purified polysaccharides was determined by HPLC method with precolumn derivatization. Result: The neutral polysaccharide was consisted of glucose, galactose, mannose, arabinose, and rhamnose, with molar ratio of 123:84:27:19:5, also contained a small amount of xylose. The acidic polysaccharide was consisted of galactose, galacturonic acid, glucose, arabinose, mannose, glucuronic acid, and rhamnose, with molar ratio of 14:10:7:6:4:1:1, also contained a small amount of xylose. Conclusion: To develop an analysis method for extracting polysaccharide from M. dodecandrum and determining it's composition, the method is simple, convenient and feasible.

      摘要:Objective: The contents of baicalin and chlorogenic acid in Lidan tablets were simultaneously determined by HPLC. Method: The sample were separated on Capcell Pak C18 (4.6 mm×150 mm, 5 μm) column, which eluted with acetonitrile (A) and 0.3% phosphoric acid (B). The ratio of acetonitrile and water increased from 87:13 to 67:33 in 20 min with detected wavelength at 315 nm, flow rate at 1 mL · min-1, column temperature at 30 ℃ and injection volume was 10 μL. Result: The result showed that two compounds reach the baseline separation, the linear range of baicalin and chlorogenic acid was 0.001 0-0.049 9 g · L-1 (r=0.999 9) and 0.005 3-0.026 7 g · L-1 (r=0.999 3), respectively, and the average recovery of baicalin and chlorogenic acid was 98.62% (RSD 1.1%) and 98.98% (RSD 0.3%), respectively. Conclusion: The method is rapid, precise and practical that it can be used to determine the two components in Lidan tablets.

      摘要:Objective: To set up an content determination method for oleanolic acid and ursolic acid in Lycopi Herba by HPLC, and to find the relationship between altitude and medicinal material content. Method: AgilentTC C18 column(4.6 mm×250 mm,5 μm) was selected, the mobile phase was methanol-acetonitrile-0.2% glacial acetic acid solution(78:10:12), the flow rate was 0.8 mL · min-1. The column temperature performed at 40 ℃, and the target component was detected at 210 nm. Result: Oleanolic acid showed a linear ranges at 0.164-2.46 μg(r=0.999 8) and ursolic acid was 0.325 6-4.884 μg(r=0.999 8). The average recovery of the added sample were 99.03% (RSD 1.7%) and 98.83% (RSD 1.4%), respectively. Altitude and herbal content showed a certain degree of positive correlation. Conclusion: The method is simple and rapid, accurate and reliable, therefore available for the elevation of Lycopi Herba. Relationship between altitude and quality can be a preferably guidance for planting area.

      摘要:Objective: To investigate pharmacokinetics and tissue distribution of gossypol nanosuspensions(GP-NS) in mice. Method: HPLC was established to determine the concentration of gossypol in serum and different tissues of mice.After tail administration of GP-NS and gossypol solution,their pharmacokinetics and tissue distribution differences were compared. Result: After administration of GP-NS,area under plasma concentration time curve (AUC0-∞) was (18 792.56±2 304.43) μg · L-1 · h-1,in vivo residence time (MRT0-∞) was (21.82±1.60)h,apparent volume of distribution (V) was (0.75±0.16) L · kg-1,clearance(CL) was (1.37±0.37) L · kg-1 · h-1and maximum plasma concentration (Cmax) was (17 589.81±3 034.14) μg · L-1.Values of relative uptake rate (Re)in the liver,heart,spleen,lung and kidneywere 1.44,3.21,12.19,1.88 and 6.54,respectively. Conclusion: GP-NS can significantly increase the level of gossypol concentration in mice and improve its bioavailability.GP-NS is helpful for gossypol to reach a long circulation time and is hopeful to be its novel drug delivery system.

      摘要:Objective: The aim of this study was to investigate the steady-state brain-plasma concentration ratio of morroniside in focal cerebral ischemia-reperfusion model rats. Method: The in vivo rat model was established. The middle cerebral artery occlusion was induced by the intraluminal filament technique. The concentrations of morroniside in the plasma and brain of rats were determined from normal control group, sham-operated group and focal cerebral ischemia- reperfusion model group, and the blood brain partitioning(lg BB) of morroniside was calculated while the plasma concentration of morroniside was in the steady-state via intravenous drip (55 min) by LC-MS/MS. Result: Morroniside almost could not pass through the blood brain barrier in normal rats. The lg BB of morroniside in the left and right brain of focal cerebral ischemia-reperfusion model rats were (1.36±0.44) and (2.26±0.13), respectively, which were significantly higher than the normal control group and sham-operated group rats. Conclusion: The results demonstrated that morroniside could pass through the blood brain barrier in rats in focal cerebral ischemia-reperfusion model group more easily than normal control and sham-operated group.

      摘要:Objective: The aim of this article was to study the pharmacokinetics of chrysophanol solid dispersion in rats. Method: Male SD rat (n=20) were randomly divided into two groups (n=10). The rats were administrated chrysophanol and chrysophanol solid dispersions respectively, the two groups were fed in the same dosage of chrysophanol (40 mg · kg-1). The concentration of chrysophanol in plasma were determined by high performance liquid chromatography (HPLC).The software of DAS1.0 was used to process the pharmacokinetic parameters. Result: The concentration-time profiles of chrysophanol and its solid dispersions conformed to two-compartment model. Their AUC, Cmax, and Tmax in chrysophanol group were (226.7±18.62) μg · h-1 · mL-1,(1.37±0.14) mg · L-1 and(68.99±5.24) h, respectively, while in chrysophanol solid dispersions group they were (1 210.0±56.32) μg · h-1 · mL-1,(8.17±0.94) mg · L-1 and(38.42±2.78) h, respectively. Statistical results indicated that both of AUC and Cmax showed significant difference in the two groups (P<0.05). Conclusion: The preparation of chrysophanol solid dispersion with polyethylene glycol as the carrier increases the bioavailability of chrysophanol in rats.

      摘要:Objective: Effects of yuanhuacine on inflammatory related factors has been studied in cell supernatants. Method: Yuanhuacine(0,1.62,6.48 mg · L-1) was acted on RAW264.7 cells for 48h, the levels of inflammation-related mediators like arachidonic acid(AA)prostaglandin G2(PGG2), prostaglandin F2α(PGF2α), leukotriene B4(LTB4),15-deoxy-δ-12,14-prostaglandin J2(15d-PGJ2), 5,15-dihydroxyeicosatetraenoic acid(5,15-DiHETE) were determined by UPLC-MS/MS. Result: Compared with the control group,the relative contents of AA,PGF2α,PGG2 and 5,15 DiHETE were markedly increased,and the relative content of LTB4 was also increased at the high dose of Yuanhuacine while the relative content of 15d-PGJ2 was decreased. Conclusion: Yuanhuacine can play a role in regulating both the proinflammatory and anti-inflammatory pathways.

      摘要:Objective: To observe the protective effects of Buxin Ruanmai granules on acute myocardial infarction in anaesthetized dogs. Method: A total of 36 dogs were divided into the Buxin Ruanmai granules high dose, middle dose,low dose groups,Xinyuan capsule group,model group and sham group.The hemodynamics and myocardial oxygen metabolism, the myocardial infarct size,and the changes of the enzymes in serum were determined in the acute myocardial infarction model induced by ligation of left anterior descending (LAD) in the anaesthetized dogs;nitro blue tetrazolium (NBT) staining was used to observe infarct size;blood gas analyzer was used to measure oxygen content. Result: Except for increasing coronary blood flow,the cardium oxygen consumption, myocardial infarct size, the activity of serum creatine kinase (CK) and lactate dehydrogenase (LDH) were decreased in acute myocardial infarction dogs treated with the Buxin Ruanmai granules(P<0.05, P<0.01). Buxin Ruanmai high dose had significant difference(P<0.01). Conclusion: The Buxin Ruanmai granules have protective effects on myocardial ischemia in anaesthetized dogs.

      摘要:Objective: The aim of the study was to investigate effects and mechanism of Danhong Huayu Koufuye (DHK) on diabetic retinopathy and diabetic nephropathy based on the aspect of thrombus formation. Method: Effects of DHK on the clotting time and bleeding time were observed by capillary glass tube method and transecting the tip of the tail, respectively. Antithrombotic effects of DHK were examined with the ligation model of inferior vena cava, carotid artery-carotid vein bypass and ferric chloride-induced arterial thrombosis in rats. The animals in the study of the clotting time, bleeding time, the ligation model of inferior vena cava and carotid artery-carotid vein bypass model were randomly divided into 4 groups:model group, low dosage of DHK(0.60, 2.25, 1.60, 1.60 g · kg-1 · d-1),high dosage of DHK(1.20, 4.50, 3.20, 3.20 g · kg-1 · d-1),aspirin group(2.88, 11.26, 7.81, 7.81 mg · kg-1 · d-1), respectively. Rats in ferric chloride-induced arterial thrombosis experiments were divided into 5 groups:sham group,model group,low and high dosage of DHK(1.60,3.20 g · kg-1 ·d-1), and clopidogrel group(30 mg · kg-1 · d-1). Clotting time was assayed after single and multiple administration for 5 days. Bleeding time and the thrombus weight were detected after administered for 5 days. Result: Compared with vehicle control, DHK significantly prolonged the clotting time in New Zealand rabbits (P<0.05) and bleeding time in mice (P<0.05). High dosage of DHK significantly inhibited thrombus formation. Inhibition rate of thrombus dry weight in high dosage DHK group was 73.8% in ligation model of inferior vena cava, and 45.1% in carotid artery-the carotid vein bypass model. Thrombus percentage of high dosage DHK group was (52.1±17.7)% (P<0.05; vs model group) in ferric chloride-induced arterial thrombosis in rats. Conclusion: DHK has activities of promoting blood circulation to remove blood stasis and prevent thrombus formation.

      摘要:Objective: To explore the effect of compatibility of astragalus flavonoids and kudzu flavonoids on glucose and lipid metabolism in diabetes mellitus(DM)rats. Method: Sixty-six SD male rats were randomly divided into six groups according to blood glucose, which was normal group, model group, positive control group, astragalus flavonoids group, kudzu flavonoids group, and astragalus flavonoids and kudzu flavonoids group,11 rats in each group. In addition to the normal group, the other groups were induced by streptozotocin (STZ, 46 mg · kg-1) to make the DM model. At the same time, the drugs were ig given. Astragalus flavonoid group, kudzu flavonoid group, astragalus flavonoid and pueraria flavonoid group were respectively given at dose of 0.039, 0.036, 0.075 g · kg-1;positive control group was given Jinqi Jiangtang tablets at dose of 1.47 g · kg-1;normal group and model group were given the same amount of distilled water(10 mL · kg-1). After administration for 30 days, the levels of blood glucose, serum cholesterol (CHO) and triglyceride (TG) were tested, and hepatic insulin (INS), glucose transporter 4 (GLUT4), adiponectin (ADPN), and leptin (LEP) were tested as well. One-way ANOVA was used to analyze the single effect of astragalus flavonoids and kudzu flavonoids on glucose and lipid metabolism, and factorial design analysis of variance was used to analyze their interactions as well. Result: In DM rats, the contents of blood glucose, CHO and TG were statistically increase (P<0.05), and the concentrations of hepatic INS, GLUT4, ADPN and LEP were statistically decreased (P<0.05). Astragalus flavonoids could reduce blood glucose level in rats effectively (P<0.05), while the effect of kudzu flavonoids was not obvious, and there was no synergistic effect between the two drugs. Both astragalus flavonoids and kudzu flavonoids could reduce serum TG and CHO, but the combined effect was not obvious compared to any single drug, and there were antagonistic effects between the two drugs. Both astragalus flavonoids and kudzu flavonoids could raise the level of hepatic GLUT4(P<0.05).In terms of hepatic INS, GLUT4 and LEP levels, the combined effects were better than any single drug, it showed that there were synergistic effects between the two drugs on them. Conclusion: Compatibility of astragalus flavonoids and kudzu flavonoids could improve the glucose and lipid metabolism by regulating INS, GLUT4 and LEP levels synergistically.

      摘要:Objective: To investigate the effects of microRNA-520b(miR-520b) associated with blood stasis syndrome on interleukin-8(IL-8) and the intervention of tanshinone ⅡA(Tan ⅡA). Method: Synthetic miR-520b mimic(50 nmol · L-1) or inhibitor (100 nmol · L-1) were transfected into 1.6×104 cells/well human umbilical venous endothelial cells (HUVECs) 24, 36, 48 h by lipofectamineTM 2000 and then, 5, 10, 20, 40 mg ·L-1 Tan ⅡA intervented miR520b-transfected HUVECs 24, 48 h. The proliferation(n=5) and nitric oxide(NO), endothelial protein C receptor(EPCR),Von Willebrand factor(vWF),thrombomodulin(TM) and endothelin(ET)' s concentration of HUVECs were analyzed by MTT assay, nitrate reduction assay and ELISA assay respectively(n=3). Transfection efficiency of miR-520b was detected by the fluorescence microscope and IL-8 mRNA and protein were also detected(n=3). Result: Compared with normal group, miR-520b inhibited the proliferation and NO concentration, and promoted the EPCR, vWF, and ET concentration of HUVECs in the miR-520b group. After 48 hours, the expression of IL-8 mRNA and protein were significantly decreased in miR-520b group. Tan ⅡA increased significantly the proliferation and NO concentration, and decreased significantly the EPCR, vWF, TM and ET concentration of HUVECs. Additionally, the expression of IL-8 mRNA and protein were significantly increased in the Tan ⅡA group, in comparison to the miR-520b group. Conclusion: MicroRNA-520b may promote the development of blood stasis syndrome in HUVECs by inhibiting the expression of IL-8. This process may be reversed by Tan ⅡA.

      摘要:Objective: This study was aimed to investigate the inhibitory effect of extract of Antrodia camphorata (ANCA-E-D) in human hepatocellular carcinoma cells SMMC-7721, and to explore its possible mechanism. Method: In this study, we used the exponential phase cells. The MTT method was used to detected the inhibitory effect of ANCA-E-D in various concernation on proliferation of SMMC-7721 cells which were cultured in vitro after 24,48, 72 hours. Cell morphological changes were observed after Wright's stain. The flow cytometry was adopted to detected the change of cell cycle and the detection of apoptosis in vitro. Result: The concentration of 20,40,60 mg · L-1 ANCA-E-D could markedly inhibit the proliferation of SMMC-7721 cells after 24,48,72 h. The 50% inhibiting concentration IC50 of SMMC-7721 cells for 24, 48,72 hours were 31.21, 24.12,21.48 mg · L-1, respectively. The SMMC-7721 cells displayed distinctive apoptotic character by Wright's staining with ANCA-E-D.Flow cytometry indicated that the different concentrations of ANCA-E-D could induce SMMC-7721 cells apoptosis, that the late apoptosis is dominant. Conclusion: ANCA-E-D can inhibit the proliferation of human hepatocellular carcinoma cells SMMC-7721, and the possible mechanism may relate to the induction of apoptosis.

      摘要:Objective: To explore the effects of extracts from Ginseng Radix et Rhizoma, Notoginseng Radix et Rhizoma and Chuanxiong Rhizoma on vascular endothelial cells and cadherin in replicative senescence of human cardiac microvascular endothelial cells (HCMECs) model. Method: The replicative senescence HCMECs model was induced in HCMECs by generation cultivating to the eighth cells. The in vitro cultured HCMECs were divided into 5 groups:the eighth generation cell-senescence untreated group (old group), the resveratrol group (10 μmol · L-1), the low-, intermediate-and high-dose herbal treated groups (50, 100, 200 mg · L-1). The intervention time of drug was 24 h. Changes of HUVECs aging were observed by the senescence-associated β-galactosidase staining (SA-β-gal), cell proliferation was detected by using cell counting kit-8 (CCK-8), and cell cycle was analyzed by using flow cytometry. The morphological changes of VE-cadherin were observed by using laser confocal microscopy. The protein expression of VE-cadherin was observed by using Western blot. Result: Compared with the old group, the extracts from Ginseng Radix et Rhizoma, Notoginseng Radix et Rhizoma and Chanxiong Rhizoma could decrease the proportion of aging stained, promote cell proliferation, reduce the proportion of G0/G1 phase, increase the proportion of G2/M phase, maintain the integrity of VE-cadherin, and promote the expression of VE-cadherin protein (P<0.05, P<0.01). Conclusion: The extracts from Ginseng Radix et Rhizoma, Notoginseng Radix et Rhizoma and Chuanxiong Rhizoma could delay aging, promote the proliferation and mitosis, maintain the integrity of VE-cadherin and promote the expression of VE-cadherin protein in replicative senescence of HCMEC model, which may contribute to the delay of in vitro HCMEC senescence for Chinese herbal medicines of nourishing qi and resolving blood stasis.

      摘要:Objective: To detect the expression levels of cytokine interleukin 1β (IL-1β), interleukin 2 (IL-2) and tumor necrosis factor alpha (TNF-α) as well as the mRNA expression in hippocampus and colon tissues of diarrhea predominant irritable bowel syndrome (IBS-D) rat model, and to make a preliminary analysis of the pathogenesis of brain-gut axis in IBS-D. Method: Thirty Sprague Dawley rats were divided into the normal group (n=10) and the model group (n=20). The IBS-D rat model was replicated in the model group, and the expression levels of IL-1β, IL-2 and TNF-α were detected by using immunohistochemical analysis, the mRNA expressions of IL-1β, IL-2 and TNF-α were detected by using fluorescence quantitative PCR method. The relationship of the different expression in hippocampus and colon tissues was analyzed by using SPSS 13.0. Result: Compared with the normal group, the levels of IL-1β, IL-2 and TNF-α in the hippocampus and colon tissue of increased significantly in the model group, and the results showed some degree of correlation between the hippocampus and colon tissue. The mRNA expression of IL-1β in the hippocampus of IBS-D rats increased significantly, the mRNA expression of TNF-α in the hippocampus and colon tissues increased significantly(P<0.05), but there were no significant changes in the mRNA expression level of IL-2. Conclusion: As the mediators of inflammation, the expressions of IL-1β, IL-2 and TNF-α,were abnormal in the hippocampus and colon tissues of the IBS-D rats, suggesting that the occurrence and development of IBS-D may be contributed by the effect of nerve endocrine immune network and the realization of brain-gut axis function changes.

      摘要:Objective: To investigate the hypoglycemic effects of Averrhoa carambola root extract (YTG) in diabetic mice induced by streptozotocin (STZ). Method: Diabetic mice were induced by intravenous injection of STZ. Mice were divided into the normal group, the model group, the metformin group, the high-, medium-and low-dose YTG groups (1.2, 0.6, 0.3 g · kg-1 · d-1) of 10 mice each. Drugs were intragastrically administered to mice for 4 weeks. Fasting blood glucose (FBG) was determined at the 2nd and 4th week. After 4 weeks, oral glucose tolerance test (OGTT), liver glycogen, activities of hexokinase (HK), pyruvate kinase (PK), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malonyldialdehyde (MDA) were measured. Result: Compared with normal group, FBG, the area under curve of blood glucose, and MDA content increased significantly, the liver glycogen content and the activity of HK, PK, SOD, and GSH-Px decreased significantly in the model group (P<0.01).Compared with the model group, FBG, the area under curve of blood glucose, and MDA content decreased significantly, while the liver glycogen content and the activity of HK, PK, SOD, and GSH-Px increased significantly in YTG group (P<0.05, P<0.01). Conclusion: YTG could reduce the blood glucose level of diabetic mice, improve the glucose tolerance. Its mechanism may be related to enhancing the activities of HK and PK and improving the liver antioxidant ability.

      摘要:Objective: To investigate the protective effects of Cassia nomame against acute liver injury induced by carbon tetrachloride(CCl4) and ethanol in mice. Method: Sixty mice were randomly divided into six groups:the control group, the model group, the high-, medium-, low-dose C. nomame groups (0.9, 0.6, 0.3 g · kg-1), and the bifendate group (0.15 g · kg-1). The mouse models of acute liver injury were established by CCl4 and ethanol. After intragastrical administration of the corresponding medicines once daily for 3 days, the effects on serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), the liver index, the contents of malonaldehyde (MDA), the activities of superoxide dismutase (SOD), the levels of glutathione (GSH) in liver tissues were measured. The protective effects on liver tissues were evaluated by using microscopic examination of hematoxylin-eosin (HE) staining. Result: Compared with the normal group, the levels of ALT, AST, MDA and the liver index increased, levels of SOD and GSH decreased in the model group (P<0.01). Compared with model group, the levels of ALT, AST, MDA and the liver index decreased, levels of SOD and GSH increased in the C. nomame groups (P<0.05). Moreover, the alleviated pathological changes in liver tissues were observed in the C. nomame groups. Conclusion: C. nomame showed a protective effect against acute liver injury induced by CCl4 and ethanol in mice.

      摘要:Objective: To investigate the effect of Cassia nomame on obesity and insulin resistance in high-fat diet-induced C57BL/6J Mice. Method: The experimental animals were randomly divided into six groups:the normal group, the model group, the low-, middle- and high-dose Cassia nomame groups(0.1,0.3,0.9 g ·kg-1), and the rosiglitazone group(0.75 mg ·kg-1) of 15 rats each. All mice received intragastric administration of the corresponding medicines, the body weight of mice was measured every week. After 12-hour fast but water-drinking, the fasting plasma glucose (FBG) of mice was tested at the end of the eighth week. After all experimental animals were executed, body length was measured, the adipose tissue around stomach, testis and kidney were weighed, and the stomach adipose tissue was stained by HE staining. Fasting insulin (FINS) was measured and the insulin resistance index (IRI) was calculated. The levels of total cholesterol (TCH), triglyceride (TG), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in serum were measured. Result: Compared with normal diet group, the body weight, the levels of FBG, FINS, TCH, TG, IL-6 and TNF-α increased in the model group (P<0.01). Compared with the model group, the body weight, and the levels of FBG, FINS, TCH, TG, IL-6 and TNF-α decreased in the C. nomame group (P<0.05, P<0.01). HE staining showed that the fat content and cell volume decreased in the C. nomame group. Conclusion: Cassia nomame could effectively inhibit obesity and insulin resistance in high-fat diet-induced C57BL/6J mice.

      摘要:Objective: To explore the effect of duanteng Yimu decoction (DTYM) on anti-inflammation and counteracting bone destruction, and to investigate its mechanism of counteracting bone destruction of adjuvant arthritis in rats. Method: Seventy-two SD rats were divided into nine groups;the normal group, the model group, the methotrexate group (MTX, 1.06 mg · kg-1), the high-, medium-, low-dose celastrus orbiculatus groups (47, 23.5, 4.7 g · kg-1) and the high-, medium-, low-dose DTYM groups (94, 47, 9.4 g · kg-1). The rats except the normal group were induced by injecting complete Freund's adjuvant to establish adjuvant arthritis rat model. After intragastrical administration of the corresponding medicines once daily for 36 days, the feet swelling degrees, ankle X-ray, C-terminal telopeptide of type I collagen (CTX-I) and N-terminal propeptide of type I collagen (PINP) serum concentration, and ankle pathological were observed. Result: Compared with the normal group, the feet swelling degrees, CTX-I and ankle X-ray score increased significantly in the model group (P<0.01). Compared with the model group, the feet swelling degrees, CTX-I and ankle X-ray score decreased significantly in all treatment groups (P<0.01). No statistically significant differences in PINP were found between any two groups. Pathological study showed that there were less synovial cell hyperplasia, inflammatory cell infiltrations, pannus formation and cartilage destruction in all treatment groups. Moreover, the above results were better in the DTYM groups. Conclusion: The mechanism of DTYM decoction in improving arthritis and bone destruction may be related to inhibiting the level of CTX-I.

      摘要:Objective: To study the effect of Baoxin pill on liver function in the rabbit with dilated cardiomyopathy (DCM). Method: Seventy-two rabbits were randomly divided into the normal group, the model group, the high-, medium-, low-dose Baoxin pill groups (2.7, 1.35, 0.675 g · kg-1), and the captopril group (2.5 mg · kg-1). The DCM model was induced in rabbits by injecting doxorubicin (0.75 mg · kg-1) via the ear vein twice weekly for 4 weeks. The heart rate, aortic dimension, left ventricular and diastolic diameter, left ventricular and systolic diameter were measured, and the left ventricular ejection fraction (LVEF) and the stroke volume (SV) were calculated at the fourth week. After continuous intragastric administration of the corresponding medicines for 30 days, the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), γ-glutamyltransferase (γ-GT), total protein (TP), albumin (ALB), creatine kinase (CK) were measured at the eighth week. The pathological changes of heart tissue were observed by HE staining. Result: Compared with normal group, the LVEF and SV decreased at the fourth week, the levels of ALT, AST, γ-GT, ALB, TP, CK increased, the level of TP decreased at eighth weeks in the model group (P<0.05, P<0.01). Compared with model group, the levels of ALT, AST, γ-GT, ALB, TP, CK decreased, the level of TP increased at eighth weeks in the Baoxin pill groups (P<0.05, P<0.01). Moreover, the alleviated pathological changes in heart tissues were observed in the Baoxin pill groups. Conclusion: Baoxin pill has a certain protective effect on liver function in rabbits with dilated cardiomyopathy induced by adriamycin.

      摘要:Objective: To explore the inhibiting effect and mechanism of Bushen Shugan formula on A549 cells proliferation and metastasis by PTEN-PI3K-AKT pathway. Method: Sixty rats were randomly divided into 6 groups: the normal saline group (2 mL), the cisplatin group (8 mg · kg-1), the low-, high-dose Bushen Shugan formula groups(15, 30 g · kg-1), and the low-dose Bushen Shugan formula plus cisplatin group (15 g · kg-1+8 mg · kg-1), the high-dose Bushen Shugan formula plus cisplatin group (30 g · kg-1+8 mg · kg-1) of 10 rats each. The corresponding medicines were intragastrically administered to rats twice daily for 5 days and the drug serums were prepared. The cell proliferation ability of each group was evaluated by MTT. The capillary structure in each group was observed by three-dimensional cell culture and the number was counted. The mRNA expressions of AKT-1, CyclinD1 and NF-κB were detected by using RT-PCR. The protein expressions of PTEN, p-PI3K and p-AKT were detected by using Western blot. Result: Low-dose drug-contained serum of Bushen Shugan formula could inhibit the proliferation of A549 in a time-dependent manner. The inhibiting rates were lower after 48, 72 and 96 h culturing, the numbers of tuber-shaped structure were lower, the protein expression of PTEN was higher, the protein expressions of p-PI3K and p-AKT were lower, and the mRNA expressions of Akt1, CyclinD1, NF-κB were lower in the Bushen Shugan formula plus cisplatin groups than the other group (P<0.05, P<0.01). There were certain synergies between Bushen Shugan formula and cisplatin. Conclusion: Drug-contained serum of Bushen Shugan formula could inhibit the cell proliferation and metastasis of A549 cells by PTEN-PI3K-AKT signaling pathway.

      摘要:Objective: To establish the chronic hepatitis B duck model and study the protective effect of Millettia pulchra aqueous extract on chronic hepatitis B duck. Method: One-day old Guangxi shelducks were infected with 0.2 mL duck hepatitis B virus (DHBV). The infected ducks, detected by real-time PCR 7 days after infection, were divided into model group, M. pulchra high-dose and low-dose group (20,10 g · kg-1) and colchicine group(0.02 g · kg-1), and the uninfected ducks were served as normal group. In addition to the normal group, the rest of ducks were vaccinated with 0.2 mL DHBV strong positive serum per week, until the end of the experiment. The corresponding drugs were successively administrated for 5 weeks after 25 weeks of DHBV attack. Blood and liver tissue were collected 24 hours after the last administration. The activity of alanine aminotransferase (ALT) and aspartate amino transferase (AST),and the content of duck hepatitis B virus surface antigen (DHBsAg) and duck hepatitis B virus DNA(DHBV-DNA) in serum were measured. The activity of superoxide dismutase (SOD) and glutathion peroxidase (GSH-Px), and the level of malondialdehyde (MDA) and hydroxyproline (Hyp) in liver homogenate were determined.Hematoxylin-eosin (HE) staining was used to observe the damage degree of liver cells. Result: In model group, the levels of ALT and AST, and the content of DHBsAg and DHBV-DNA in serum were significantly increased;the activity of SOD and GSH-Px was significantly decreased, and the levels of MDA and Hyp were significantly increased, compared with normal group. M. pulchra aqueous extract (20,10 g · kg-1) significantly reduced the activity of ALT, and the content of DHBV-DNA in serum, increased the activity of GSH-Px, and decreased the levels of MDA and Hyp in liver. M. pulchra aqueous extract (20 g · kg-1) significantly reduced the activity of AST, increased the activity of SOD. HE staining showed that M. pulchra aqueous extract could significantly decrease the damage degree of liver cells. Conclusion: Millettia pulchra aqueous extract has certain protective effect on chronic hepatitis B duck caused by DHBV.

      摘要:Objective: To observe the hypoglycemia effect of polysaccharide from Atractylodes macrocephala (PAM) in spontaneous type 2 diabetic db/db mice. Method: Type 2 diabetic db/db mice were divided into model group, positive control group (metformin hydrochloride 250 mg · kg-1), three dose of PAM (300,90,30 mg · kg-1) and administrated orally once daily for 4 weeks. Body weight and general physical signs were observed during the treatment. The level of fast blood glucose(FBG) was determined at the second and the fourth week and the oral glucose tolerance test (OGTT), fast plasma insulin(Ins) and lipid level were determined after administration for 4 weeks. The insulin sensitivity index (ISI) was calculated based on insulin level. Result: Compared with the model group, db/db mice treated by PAM were in better station, smooth hair, as well as lower food and water intakes. PAM (300,90 mg · kg-1)could significantly decrease the levels of FBG, Ins, and increase the ISI;meanwhile PAM could decrease the serum glucose levels in 0.5,2 h after glucose administration and lower the area under curve of glucose. Conclusion: PAM could significantly decrease the blood glucose and improve the glucose tolerance. The hypoglycemic effect of PAM may be related to decreasing the plasma insulin and improving the insulin sensitivity.

      摘要:Objective: To investigate effect and mechanism of Begonia grandis sinensis through studying effective actions of two extracts on chronic pelvic inflammatory disease (CPID) rats. Method: CPID rats were separated to 7 groups and 8 rats in each group.Sham operation group only accepted surgery no modeling,while other rats were treated by mechanical damage and mixed infection and divided into the mode group,butanol high and low groups,ethyl acetate high and low groups and positive group.After treatment of administration for 14 days (sham operation group and model group only treated by water as negative control),uterine swelling degree,uterine specimen,level of interleukin-2(IL-2) and IL-6 of all the groups were detected. Result: These two extracts of B. grandis sinensis could inhibit uterine swelling,improve degree of inflammation in the uterus,restore concentrations of IL-2 and IL-6 to normal level in CPID rats. Conclusion: B. grandis sinensis can effectively reduce uterine inflammation of rats,its mechanism is relative with inhibition of inflammatory factors.

      摘要:Objective: To investigate the effect of Shenmai injection atomization inhalation on serum C- reactive protein(CRP)in patients with radiation pneumonitis effect changes and humoral immune function. Method: Fifty cases of radiation pneumonia were selected and randomly divided into experimental group and control group with 25 cases in each group. The experimental group in the same period of treatment on inhalationof Shenmai injection, the control group without drug. Each of the 50 patients before and after treatment the blood sampling time, concentration and determination of CRP and humoralimmune serum IgG, IgA, IgM, C3. According to the rating of acute radiation injury(RTOG) grading standard of radiation pneumonia 50 cases patients were graded. Result: CRP levels in two groups of patients showed varying degrees of decline,while the experimental group after inhalation of Shenmai injection of serum CRP decreased more significantly, the difference was statistically significant (P < 0.05). After the treatment,control group and experimental group serum CRP value by comparison has the remarkable difference (P<0.05). The acute radioactive lung injury classification stadard(RTOG) grading standards of radiation pneumonia will be gradedpatients, know the value of serum CRP and radioactive pneumonia was positively correlated with the severity of (r=0.72, P<0.05). Comparing the two groups before and after treatment in patients with the concentration of IgG, IgA, IgM, the difference was statistically significant(P<0.05), the experimental group after the treatment, the concentration of complement C3 increased significantly in control group did not change significantly, the differences between these two groups by comparison with statistical significance (P<0.05). Conclusion: Inhalation of Shenmai injection can significantly reduce the clinical symptoms,reduce the inflammation reaction in patients with pneumonia level, improve body immune state of patients, enhance the immune function of organism.

      摘要:Objective: This clinical study aimed to evaluate the efficacy of the therapy of warming the lung to reduce watery phlegm for treating non-small cell lung cancer(NSCLC) with malignant pleural effusion(MPE). Method: In this study, 63 NSCLC patients with MPE were randomly divided into test group and control group, with 36 patients in test group and 27 patients in control group. The test group received the Chinese medicine therapy of warming the lung to reduce watery phlegm combined with chemotherapy with cisplatin treatment through pleural effusion. The control group received single chemotherapy with cisplatin treatment through pleural effusion. The primary observation index was disease stabilization rate (DSR) of MPE, the secondary observation indexes included karnofsky(KPS), traditional symptoms, toxicities and median survival time(MST). Result: We evaluated all the 63 patients. The DSR of MPE was 94.44%(34 patients) in the test group and 77.78% (21 patients) in the control group(P<0.05). Compared to the control were, the KPS and traditional symptoms were both better. No unexpected toxicities were observed. The MST was 11.0 months in the test group and 7.5 months in the control group (hazard ratio[HR=0.21,95%CI(0.12,0.37),P<0.05]. Conclusion: The Chinese medicine therapy of warming the lung to reduce watery phlegm combined with chemotherapy with cisplatin treatment through pleural effusion is effective to NSCLC with MPE.

      摘要:Objective: The aim of this study was to observe the efficacy of combination of traditional Chinese medicine(TCM) periodic therapy and western medicine clomiphene and progynovain treating polycystic ovarian syndrome(PCOS) causing infertility. Method: In our study, 58 patients with PCOS causing sterility were randomly divided into the control group (n=29) and the treated group (n=29).The treated group received the combination therapy and the control group was treated by the same dosage of clomiphene and progynova. Both groups received continuous treatment for 3 menstrual cycles. During treatment the endometrial thickness and type, the ovulation rate,the pregnancy rate,luteinized unruptured follicle syndrome(LUFS) rate,adverse reaction to clomiphene citrate and ovarian hyperstimulation syndrome(OHSS) rate were observed. Result: The periodic ovulation rate in the treated group was 80.28%,while in the control group it was 64.38%,with significant difference between two groups (P<0.05).The percentage of A type endometrium in the treated group was higher than that in the control group, the difference was significant (P<0.05). The total pregnancy rate is higher than that in the control group, but with no significant difference. Comparison of the occurrence of LUFS, adverse reaction to clomiphene citrate and OHSS between the two groups showed no significant difference. Conclusion: The combination therapy of clomiphene,progynova and TCM periodic therapy has a better therapeutic effect in treating PCOS causing infertility than that which only uses western medicine clomiphene and progynova.

      摘要:Objective: The aim of this study was to evaluate the efficacy of triple combination therapy of vinegar Kansui Radix, Fufang Dachengqi Tang and somatostatin in the treatment of paralytic ileus. Method: The clinical data of 70 patients with paralytic ileus, who were treated at affiliated hospital of Gansu university of traditional Chinese medicine(TCM), were analyzed. They were divided into treatment group (30 cases) and control group (40 cases) according to the treatment. The control group was received combination treatment of routine treatment and Fufang Dachengqi Tang retention-enema. The treatment group was received combination therapy of vinegar Kansui Radix and somatostatin on the basis of the control group. Clinical symptoms, signs, gastrointestinal decompression amount, improvement time of clinical symptom, recovery time of body temperature and leukocyte number and clinical efficacy were observed and compared between the two groups. Result: The post-treatment remission rate of abdominal pain, abdominal distension, flatus and defecation were higher in treatment group than in control group (P<0.05). The post-treatment remission rate of nausea and vomiting showed no statistically difference in both two groups. The post-treatment gastrointestinal decompression amount in treatment group was lower than in control group (P<0.01). The improvement time of clinical symptom including abdominal pain, abdominal distension, vomiting, intestinal sound recovery, flatus, defecation and eating in treatment group were shorter than control group (P<0.01). The recovery time of body temperature and leukocyte number in treatment group were shorter than that in control group (P<0.01). The clinical curative rate and total effective rate in treatment group were higher than in control group (73.3% VS 22.5%,96.7% VS 87.5%,P<0.05). Conclusion: Combination therapy of vinegar Kansui Radix,Fufang Dachengqi Tang and somatostatin can improve the efficacy of paralytic ileus, with the advantage of fast improvement of clinical symptom, quick recovery of gastrointestinal function and easy control of infection.

      摘要:Objective: The purpose of this study was to observe the efficacy of Chaihu Shugan San(CHSGS) in the treatment of subacute thyroiditis in plateau area. Method: In this study, one hundred and sixteen cases of patients were included and randomly divided into the traditional Chinese medicine (TCM) group and the western medicine group by odd and even numbers, with 58 cases in each group. The patients in the Western medicine group were treated with conventional western medicine, and the patients in the TCM group were given CHSGS on the basis of conventional western medicine. The efficacy was observed after treatment, and the recurrence were observed in the following six months. Result: After one month, the total efficiency in the TCM group was 96.55%, while it was 81.03% in the western medicine group, and the difference was statistically significant (P<0.05). The FT3,FT4,C-reactive protein(CRP), ESR in the TCM group decreased significantly, and thyroid stimulating hormone(TSH) was increased and the difference was statistically significant compared with the Western medicine group(P<0.05). In the TCM group, there were 5 recurrence cases after six months, and the recurrence rate was 8.62%, while in the western medicine group there were 26 recurrence cases with a recurrence rate of 44.83%, the difference between two groups was statistically significant (P<0.05).There was no adverse reaction in both two groups. Conclusion: CHSGS combined with the western medicine was curative in the treatment of subacute thyroiditis, which can significantly reduce the relapse rate, and be worth of promoting in clinical.

      摘要:Objective: To observe the efficacy and safety of Bushen Qiangdu Zhilou decoction combined du-moxibustion in treating ankylosing spondylitis of kidney deficiency and cold syndrome. Method: Fourty patients were selected from the Rheumatism clinic and randomly divided into the combination group (traditional Chinese medicine combined du-moxibustion treatment) and the control group (Chinese medicine treatment). All patients received 12 weeks of treatment. Before and after treatment, the integral score of Chinese medical syndrome, the Bath ankylosing spondylitis activity index (BASDAI), the Bath ankylosing spondylitis functional index (BASFI), patients' overall evaluation (PGA), spinal pain VAS, refers to the distance, pillow wall distance, thoracic mobility, lumbar motion changes (Schober test), blood sedimentation (ESR) and c-reactive protein (CRP) level were measured. At the same time, the adverse reactions like liver function test, kidney function test, blood routine examination were recorded during the treatment. Result: The total effectiveness of symptomatic efficacy in the combination group was 94.4%, which was superior to that of 70% in the control group (P<0.05). The cases of ASAS20, ASAS50 and BASDAI50 in the combination group were more than those in the control group (P<0.05).The improvements of the refers to the distance spinal pain, spinal inflammation, the BASDAI, score of Chinese medical symptom were better in the treatment group as compared with the control group (P<0.05). Levels of ESR and CRP decreased in the two groups after treatment (P<0.05, P<0.01), but there was no statistically significant difference between the two groups. Unusual liver and kidney function, and blood routine tests were found during the treatment. However, no serious adverse reactions were found. Conclusion: Bushen Qiangdu Zhilou decoction combined du-moxibustion could enhance the clinical curative effect and safety greatly in treating ankylosing spondylitis, and it has wide application prospect.

      摘要:Objective: To discuss the effectiveness and safety of Shenmai injection in treating patients with acute ST-segment elevation myocardial infarction (STEMI). Method: One hundred and twenty STEMI patients were selected from our hospital and randomly divided into the control group and the experimental group of 60 patients each. The patients in the control group received reteplase for injection. On the basis of the control group, the patients in the experimental group added Shenmai injection. The left ventricular ejection fraction (LVEF) and left ventricular end-diastolic diameter (LVEDD), the incidence of myocardial in farction recurrent, stable angina, congestive heart-failure, cardiac death, arrhythmia, peak time and recovery time of creatine kinase (CK) and CK isoenzyme-MB (CK-MB), recovery situation and time of ecg ST segment, length of hospital stay and injection adverse reaction data of patients in both two groups were observed. Result: The clinical effect in the experiment group was superior to that in the control group (P<0.01). After treatment, LVEF in the experiment group was higher than that in the control group (P<0.05). Reperfusion rates of coronary in two groups were 78.33% and 88.33%, respectively, while there was no significant difference between two groups. The cumulative incidence rate of adverse cardiovascular events in the experiment group was 95%, which was higher than that of 63.33% in the control group (P<0.01). The average ST-segment elevation resolution was higher, recovery time of ST-segment was shorter, peak time and recovery time of CK and CK-MB were shorter in the experiment group than those in the experiment group (P<0.01). Conclusion: Shenmai injection combined with reteplase could improve cardiac function, relieve ischemia-reperfusion injury, ameliorate blood flow in the heart muscle, protect heart muscle and reduce the happening of bad cardiovascular event in treating STEMI.

      摘要:Objective: To discuss the effect of modified Geqi powder combined concurrent chemoradiotherapy on quality of life and immunoregulation in treating patients with middle and terminal esophageal cancer. Method: One hundred and fifteen patients with middle and terminal esophageal cancer were randomly divided into the control group (57 cases) and the observation group (58 cases) by a random number table. Patients in the control group received three dimensional conformal radiation therapy and synchronous chemotherapy. Based on the treatment of the control group, patients in the observation group added modified Geqi powder. All patients in two groups received 12 weeks of treatment. The detectable lesion was detected by CT or MRI. Before and after treatment, symptoms and signs were recorded. Patients' life of quality was evaluated by esophageal cancer specific scale (QLQ-OES18). Levels of CD3+, CD4+ and CD4+/CD8+ of T cell subpopulations and natural killer cell (NK) were detected. Safety was monitored during the treatment. Result: The efficient of solid tumor in the observation group was 70.69%, which was higher than that of 56.14% in the control group, while there was no statistically significant difference between two groups. After treatment, scores of dysphagia, difficulty in feeding, pain, drool, swallowing obstruction, dry mouth and appetite were lower, levels of CD3+, CD4+ and NK were higher in the observation group than those in the control group (P<0.01). The occurrence degree of acute radiation injury (P<0.05) and nausea and vomit (P<0.01) were lower than those in the control group (P<0.01). Conclusion: Modified Geqi powder could ameliorate clinical symptoms and signs, relieve toxic and side effect of radiotherapy and chemotherapy, and improve bodies' immune function and quality of life, and it has an obvious short-term effect.

      摘要:Objective: To observe the clinical effect of modified Jianpi Xiaozhi decoction in treating diabetic gastric paralyze (DGP), and to investigate its influence on motilin and gastrin. Method: Ninety-six patients were randomly divided into the control group (47 cases) and the observation group (49 cases) by a random number table. Patients in the control group received 5 mg cisapride tablets thrice daily 30 min before a meal. Patients in the observation received 1 dose modified Jianpi Xiaozhi decoction in twice daily. All patients in both two groups received 4 weeks of treatment. Before and after treatment, scores of the main symptoms and signs were recorded. Gastric emptying test was conducted. Levels of motilin and gastrin were detected. Result: The total efficient rate in the observation group was 91.8%, which was superior to that of 74.4% in the control group (P<0.05). After treatment, stomach row space in the observation group was lower than that in the control group (P<0.01). Levels of plasma motilin and gastrin in the observation group were lower than those in the control group (P<0.01). Scores of fullness degree, fullness, duration, early satiety, nausea and vomiting, heating amount and defecate in the observation group were inferior to those in the control group (P<0.01). Conclusion: Modified Jianpi Xiaozhi decoction could improve clinical symptoms and promote gastric emptying in treating DGP. The clinical curative effect of therapy is better than that of cisapride. Its mechanism may be related to regulating the levels of motilin and gastrin.

      摘要:Objective: To discuss the amelioration effect of maintenance treatment of traditional Chinese medicine on quality of life and survival rate in treating patients with advanced primary hepatocarcinoma (APH). Method: Eighty-five patients with APH were randomly divided into the control group (34 cases) and the observation group (51 cases) according to the different therapy methods. Patients in the control group received symptomatic relief and supportive treatment. Based on the treatment of the control group, patients in observation group received ① 250 mL Shenqi Fuzheng injection, ②20-100 mL Shenmai injection for 2 weeks separated a week, ③ 50-100 mL aidi injection, ④ 4 brucea javanica oil soft capsules thrice daily for 4-weeks period separated a week between two weeks, ⑤ Traditional Chinese medicine for oral administration according to their different condition. All patients in both two groups received 6 months of treatment until their deaths. Karnofsky (KPS) grades of quality of life and grades of the main symptoms and signs were evaluated every month. Child-pugh classification scores of liver function and ECOG scores of physical condition were made before treatment and after 3-, 6-month treatment. Levels of liver function including alanine aminotransferase (ALT) and aspartate aminotransferase (AST), total bilirubin (TBil) and serum alpha-fetoprotein (AFP) were detected. Patients' medium survival time and survival rate of half a year were evaluated. Result: KPS grades were higher after 4-, 5-and 6-month treatment in the observation group than those in the control group (P<0.05). Scores of the main symptoms and signs increased, while the results were lower after 2-, 4-and 6-month treatment as compared to the control group (P<0.01). Scores of Child-Pugh and ECOG were lower after 3-and 6-month treatment in the observation group than those in the control group (P<0.01). Levels of ALT and AST were lower after 3-month treatment in the observation group than those in the control group (P<0.01). Levels of ALT, AST and TBil were lower after 6-month treatment in the observation group than those in the control group (P<0.01). Level of AFP after 3-and 6-month treatment in the observation group was lower than that in the control group (P<0.01). Survival rate of half a year in the observation group was 50.98%, which was higher than that of 37.14% in the control group (P<0.05). The medium survival time in observation group was 132 days, which was higher than that of 86 days in the control group (P<0.01). Conclusion: Maintenance treatment of traditional Chinese medicine could relieve clinical symptoms and signs, ameliorate patients' quality of life and physical condition in treating APH. Moreover, it could prolong patients' medium survival time and survival rate of half a year.

      摘要:Objective: To discuss the curative efficacy of Yiqi Xiaoji decoction for oral administration and Jiedu Huazheng decoction for enema in treating chronic pelvic inflammatory mass of damp heat and blood stasis syndrome, and to discuss their influence on blood rheology, T lymphocyte subgroup, complement component C3 and C4. Method: One hundred and five patients were randomly divided into the Western medicine group (51 cases) and the combination group (54 cases) by a random number table. Patients in the Western medicine group received comprehensive therapy including cefotaxime, tinidazole injection and physiotherapy. Based on the treatment of the Western medicine group, the patients in the combination group added Yiqi Xiaoji decoction and Jiedu Huazheng decoction. Diameter size of bag piece was detected via type-B ultrasound. The main signs and symptoms scores were conducted before and after treatment. Levels of hemorheology index, T lymphocyte subsets (CD3+, CD4+, CD8+, CD4+/CD8+), C3 and C4 were tested. Result: The clinical efficacy of the total effective rate in the combination group was 92.5%, which was superior to that of 76.4% in the Western medicine group (P<0.05). After treatment, the diameter of the masses in the combination group was lower, scores of symptoms and signs were less than those in the Western medicine group (P<0.01). Blood viscosity (low shear and high shear), plasma viscosity, fibrinogen and platelet aggregation rate were lower, levels of CD3+, CD4+and CD4+/CD8+were higher, and levels of CD8+ was lower in the combination group than those in the Western medicine group (P<0.01). Compared with before, levels of C3 and C4 increased after treatment, and the results were higher in the combination group than those in the Western medicine group (P<0.01). Conclusion: Based on the Western medicine treatment, Yiqi Xiaoji decoction and Jiedu Huazheng decoction could reduce masses, relieve clinical symptoms and signs in treating chronic pelvic inflammatory mass, and its effect is better than Western medicine alone. The mechanism of action may be related to ameliorating microcirculation and improving bodies immune function.

      摘要:Objective: The aim of this article was to discuss the composition rules of Tibetan medicinal formulas used to treat heat illnesses of the liver. Method: Formulas described for treating heat illnesses of the liver (mchin tshad) were selected from the rGyud bZhi. Frequency and association rules were analyzed by the statistical computing CRAN R software. Result: In this study, 67 Tibetan medicinal formulas including 95 ingredients were found for treating liver heat illnesses. Among these formulas, the top 10 ingredients frequently used are:StigmaCroci, Calciosinti, Herba Swertiae Chirayitae, Bovis Calculus, Chebulae Fructus, Brag Zhun, Adhatoda Vasica, Herba Lagotis, Santali Albi Lignum and Caryophylli Flos. These 10 ingredients comprised 46.26% of the extracted formulas.A total of 204 association rules were found with a support value higher than 1 and a confidence higher than 0.7.Totally, 6 most frequently cited association groups were derived, as the following:Chebulae Fructus-Fructus Termlnallae Billericae-Fructus Phyllanthi;Calciosinti-Bovis Calculus-Stigma Croci-Herba Lagotis-Herba Swertiae Chirayitae-Herba Aconiti Tangutici;Adhatoda Vasica-Calciosinti-Stigma Croci-Herba Swertiae Chirayitae-Bovis Calculu;Calciosinti-Stigma Croci-Santali Albi Lignum-Herba Lagotis;Fel Ursi-Stigma Croci-Brag Zhun; Bovis Calculus-Stigma Croci-Santali Albi Lignum-Calciosinti. Conclusion: The most common characteristic of the ingredients among the formulas derived is their cooling activity, as they are the common compounds used to treat general heat illnesses.Stigma Croci, the most frequently used compound has the effect of removing pathogenic heat and protecting the liver.Stigma Croci is also commonly used to enhance the efficacy of other formulas for treating the liver. The most commonly occurring compound groups derived by the current study all contain compounds which can protect liver and treat heat illnesses generally. Their taste composition is primarily sweet and bitter, which are the tastes described to treat heat illnesses in general.The target activity of such groups is described to increase the cooling and dulling quality within the bodily constituents such that the hot and sharp qualities of mkhris pa of heat illnesses are pacified.This study demonstrated that the composition rules of Tibetan medicinal formulas in the rGyud bZhi indicated for treating heat illnesses of the liver is consistent with the Tibetan medicinal compounding theory.As such, the compounding rules of Tibetan medicinal formula should be applicable to direct clinical practice.

      摘要:Objective: The aim of this study was to evaluate the safety and efficacy of Huoxue Huayu method of Chinese herbal medicine in the treatment of ulcerative colitis(UC). Method: All the randomized controlled trials (RCTs) on activating blood circulation to remove blood stasis method of Chinese herbal medicine in the treatment of UC before June 2014 were searched in the CNKI, VIP,CBM and Wangfang database. The quality of RCTs meeting inclusion criteria was assessed and the related data were extracted;Meta-analysis was performed by Stata12.0 software. Result: Totally, 20 RCTs involving 1 897 patients were included. Meta-analysis showed high effective rate [the total effective rate RR=1.248, 95%CI(1.187,1.313),Z=8.66,P<0.01] and safety. Sensitivity analysis showed that the evaluation result was stable. Funnel plot,Egger's test and Begg's test suggested no evidence of substantial publication bias. Conclusion: Huoxue Huayu method is more effective in the treatment of UC than the western medicine with higher safety. Because of the low quality of the included RCTs, the improvement of clinical research on the disease in high quality need to be confirmed.

      摘要:Objective: Osteoarthritis (OA) is a common chronic joint disease in the elderly, characterized by degenerative changes, destruction of articular cartilage and secondary bone hyperplasia, with the main clinical manifestations of repeated episodes of pain, joint swelling, joint deformity and movement disorder. And its pathogenesis is still not clear.Traditional Chinese medicine believes that OA belongs to the bone rheumatism category, for the deficiency of the Ganshenxukui, Qizhixueyu, Mailuoizu as the representation, Chinese medicine treatment also from start to pathogenesis, invigorating the kidney and promoting blood circulation mainly, and concurrently to Qushi analgesic. Each generation of physicians summarizes prescription many treatment of Bi syndrome based on clinical experience, effectively delay the disease process of OA. In addition, the clinical diagnosis of osteoarthritis mainly relies on symptoms and imaging examination, but the diagnosis method is not sensitive to early patients, which hampers the timely treatment of early patients with osteoarthritis. With the development of molecular biology, more and more studies show that various cytokines play an important role in the occurrence and development of osteoarthritis, which are closely related to the articular cartilage, synovial membrane and the subchondral bone function. There are many cytokines and proteins involved in the pathological changes of OA, such as interleukin and tumor necrosis factor alpha, transforming growth factor, insulin-like growth factor, matrix metalloproteinase and bone bridge bone protein, etc. At the same time, some specific biomarkers have been discovered in OA patients with body fluids, such as cartilage metabolism markers, markers of bone metabolism and markers of synovial metabolism substance, which can reflect early articular cartilage degeneration to assist to evaluate the early diagnosis, treatment and prognosis of osteoarthritis. At present, the research on cytokines have been probed into the mechanism level, such as the combination of cytokines and receptors on the target cells, cytokines activating intracellular signal transduction pathway. In biomarker research, given the many indicators lacking specificity, it is necessary to develop biomarkers with high sensitivity and specificity, and develop a simple and accurate method to reflect the early articular cartilage degeneration and other relevant pathological changes.

      摘要:Bone marrow is an important hematopoietic and immune organ. It contains hematopoiesis stem cells, mesenchymal stem cells, stromal cells, which provide a specific microenvironment with various growth factors and chemotactic factors. However, it causes the primary obstacle of successful eradication of bone marrow-associated diseases. Under the pathological condition, the bone marrow microenvironment supports the metastasis and growth of malignant cells. In addition, the delivery of therapeutic drugs has been hampered by its unique anatomical structure and bone marrow blood barrier. With the progress on the study on the bone marrow microenvironment and the mechanism of the particle uptake from the blood circulation, macrophage and E-selectin receptor on endothelial cells have been discovered as targets for improving the bone marrow-targeting ability of drugs used for therapy. Combined with nanoparticles technology, passive or active bone marrow-targeting ability of drugs are further improving by controlling the size or modifying the surface of nanoparticles. Herein, the physiological structure and biologic functions of bone marrow are analyzed, and the bone marrow targeting nanoparticles including liposomes and polymer micelles are summarized, which provides the favorable methods for the diagnosis and therapy of bone marrow-related diseases.
