      摘要:Objective: In order to improve the quality of Chinese medicine in the production,knowledge organization method was applied to the identification of critical process parameters (CPPs) of Chinese medicine under the guidance of quality by design (QbD). Method: Taking literatures related to ethanol extraction process of Salviae Miltiorrhizae Radix et Rhizoma as research object,rank-sum ratio (RSR) method was used to achieve CPPs recognition in ethanol extraction process of Salviae Miltiorrhizae Radix et Rhizoma,containing knowledge extracting,knowledge restructuring and result analyzing. Result: By using knowledge organization method and RSR method,the importance of CPPs in the research object was recognized successfully.Ethanol concentration and ethanol consumption were in the first level,their RSR values were 0.319 3 and 0.252 1,respectively.Followed by number of extraction,extraction time and extraction temperature,their RSR values were 0.180 7,0.205 9,0.067 23,respectively. Conclusion: Within the QbD framework,knowledge organization method can identify CPPs effectively and provide guidance for quality control in production process of Chinese medicine.

      摘要:Objective: To prepare solid dispersions of total quinones from Rubiae Radix et Rhizoma and evaluate its in vitro dissolution ability. Method: This solid dispersions was prepared by solvent-melting method and solvent method.Effect of different matrix on preparation technology were investigated,with solubility and in vitro dissolution performance as indexes,optimal formulation was confirmed. Result: This solid dispersions was prepared by solvent method,its optimal formulation was ratio of polyvinyl pyrrolidone (PVP) S-630 and total quinones extract of Rubiae Radix et Rhizoma of 5:1.Solubility of this solid dispersions in 1%sodium dodecyl sulfate (SDS) were increased to 285.39 mg·L-1,in vitro cumulative dissolution within 60 min was 66.37%. Conclusion: The prepared product has good characteristics,which is confirmed to enhance the solubility of total quinones extract of Rubiae Radix et Rhizoma.It can provide a reference for the development of solid preparation originated from Rubiae Radix et Rhizoma.

      摘要:Objective: To establish a purification process for W2 [3-O-α-L-Rha-(1→6)-β-D-Glu-(1→4)-β-D-Glu-(1→4)-β-D-Rib-(1→3)-α-L-Rha-(1→2)-α-L-Ara oleanolic saponin]in Clematis manshurica by macroporous resin. Method: The content of W2 was determined by HPLC,and it was used as index.Type of macroporous adsorption resin was screened by inspecting the adsorption and desorption ability of nine candidate macroporous resins.The static adsorption kinetics and dynamic gradient elution of the best type of macroporous resin was investigated,single factor tests were adopted to optimize purification process. Result: Optimum purification process was as following:the concentration of sample solution of 0.4 g·mL-1,adsorbed by LX-17 macroporous resin column with a flow rate of 1.5 BV·h-1,washed by 4 BV of water and 5 BV of 40%ethanol for removing impurity with a flow rate of 3 BV·h-1,taking 1 BV of 80%ethanol to remove the solution in column,and eluted by 6 BV of 80%ethanol for enrichment of W2 with a flow rate of 1.5 BV·h-1.Under these conditions,the purity of W2 in dry extract was 43.14%. Conclusion: LX-17 macroporous resin is suitable for separation and purification of W2.This optimized process is reasonable,stable and feasible,which can obtain W2 sample with high purity.

      摘要:Objective: To optimize processing technology of Zhangbang Coptidis Rhizoma processed by ginger juice. Method: HPLC was employed to determine contents of berberine hydrochloride,palmatine hydrochloride and coptisine with mobile phase of acetonitrile-0.05 mol·L-1 potassium dihydrogen phosphate (28: 72) and detection wavelength at 345 nm.Taking composite score of contents of berberine hydrochloride,palmatine hydrochloride,coptisine and yield of dry extract as index,orthogonal test was adopted to optimize processing tehcnology. Result: Optimal processing technology was ginger juice amount of 20%,frying time of 12 min and processing temperature at 140℃.Mass fractions of berberine hydrochloride,palmatine hydrochloride,coptisine and yield of dry extract were 14.15%,1.89%,1.96%and 28.80%. Conclusion: This optimized processing technology is reliable,it can provide a reference for industrial production of Zhangbang Coptidis Rhizoma processed by ginger juice.

      摘要:Objective: To investigate effect of polysorbate-80 on in vitro dissolution of salvianolic acid B in Penyankang suppositories. Method: With dissolution time and dissolution rate as indexes,HPLC was used to calculate cumulative dissolution rate of salvianolic acid B in Penyankang suppositories with different amount of polysorbate-80,mobile phase was 1% formic acid-methanol-acetonitrile (66:20:14),flow rate was 1 mL·min-1,column temperature was set at 30℃,detection wavelength was 286 nm. Result: Within 120 min,cumulative dissolution rates of salvianolic acid B from Penyankang suppositories with polysorbate-80 addition amount of 1.0%,1.5% and 2.0% all can reach more than 80%. Conclusion: Polysorbate-80 can promote dissolution of salvianolic acid B in Penyankang suppositories.In order to guarantee quality of this preparation,the amount of polysorbate-80 chooses 1.0%.

      摘要:Objective: To establish a high performance liquid chromatography (HPLC) method for content determination of 5-hydroxymethylfurfural (5-HMF) in Schisandra Chinensis Fructus (SCF), determine the content of 5-HMF in commercial SCF and Schisandra processed with vinegar (SPV) decoction pieces, and carry out the correlative analysis between 5-HMF content and its storage and color. Method: HPLC-PDA method was performed on ODS column (4.6 mm×250 mm, 5 μm) with methanol-water (5: 95) as the mobile phase at a flow rate of 1 mL·min-1. The detective wavelength was set at 284 nm and the column temperature was maintained at 30℃. Result: The HPLC method for content determination of 5-HMF in SCF and SPV was established. The contents of 5-HMF in SCF and SPV were 3.6-70.2, 3 719.0-14 486.0 μg·g-1, respectively. Conclusion: The content of 5-HMF in SPV is significantly higher than that in SCF. With the extension of storage time of SCF, the color gradually deepens and the content of 5-HMF increases.

      摘要:Objective: This study was based on data of the transcriptome sequencing of Ganoderma,GLPRMT1,GLPRMT2 and GLPRMT3 were completed in detail by using bioinformatics method. Method: GLPRMT1,GLPRMT2 and GLPRMT3 were analyzed and predicted by the tools of bioinformatics in the following aspects:physical and chemical characteristics of amino acid sequences,function domain,hyophobicity or hydrophilicity,secondary structure and tertiary structure of protein,molecular phylogenetic evolution,and so on;expression levels of GLPRMT1,GLPRMT2 and GLPRMT3 were identified by FPKM from the transcriptome data. Result: Open reading frame finder indicated that GLPRMT1 and GLPRMT2 genes were full-length,while GLPRMT3 was partial;structural domain of GLPRMT1,GLPRMT2 and GLPRMT3 showed high homology with AdoMet-MTases domains;GLPRMT1,GLPRMT2 and GLPRMT3 had relative close relationship to PRMT3 family,PRMT1 family and PRMT5 family of fungus,respectively.From transcriptome data,transcripted level of GLPRMT2 was significantly higher than GLPRMT1 and GLPRMT3,and transcripted level of these three genes have increased significantly in growing periods of Ganoderma. Conclusion: These results provide basis for further understand the function of arginine methyltransferase and epigenetic regulation of Ganoderma.

      摘要:Objective: To quickly determine the moisture content in Amomi Fructus by using near infrared spectroscopy(NIRS) technology. Method: Vacuum drying method was used for determination of moisture content in samples of Amomi Fructus.Both standard normalization method and the second derivative method were used for the spectra pretreatment, combined with partial least squares(PLS) to establish near infrared quantitative analysis models for the moisture content of Amomi Fructus. Result: The internal cross-validation correlation coefficient (R2) in the calibration set of the established models, root mean square error of calibration (RMSEC), root mean square error of prediction (RMSEP) and the performance index (PI) were 0.986 7, 0.242, 0.285 and 92.5. Conclusion: The method is simple, rapid, accurate, and non-destructive, and can be used for rapid determination of moisture content in Amomi Fructus.

      摘要:Objective: To conduct qualitative and quantitative analysis of Chrysanthemi Indici Flos from different origins by using near-infrared spectroscopy(NIRS) and TQ software. Method: Near-infrared spectra (NIR) of Chrysanthemi Indici Flos from different origins were collected by near-infrared diffuse reflectance spectroscopy technique, and discriminant analysis of TQ software was uesd to distinguish the origin of Chrysanthemi Indici Flos. The content of chlorogenic acid was determined by using HPLC, and the partial least squares (PLS) method was used to quantitatively predict the chlorogenic acid in the Chrysanthemi Indici Flos from different origins. The qualitative and quantitative analysis models were established and verified respectively. Result: The established qualitative model can quickly and accurately identify the Chrysanthemi Indici Flos from different origins, and the number of wrong discrimination cases in verification set was 0; the correlation coefficients (R2) of the quantitative analysis model was 0.995 38; the root mean square error of calibration (RMSEC) was 0.008 12; and the root mean square error of predication (RMSEP) was 0.012 8. Conclusion: The qualitative and quantitative models could be used to accurately analyze the Chrysanthemi Indici Flos from different origins. This method is simple, accurate and could be used for the rapid quality analysis of Chrysanthemi Indici Flos.

      摘要:Objective: To analyze the chemical constituents of 95%ethanol extract of Semiaquilegiae Radix by using HPLC-ESI-Q-TOF-MS. Method: C18 column (4.6 mm×150 mm, 5 μm) was adopted, with acetonitrile-0.1% formic acid as the mobile phase for gradient elution; ESI ion source was used as mass spectra, and the data were collected in the positive mode. The mass scanning range was between m/z 50-1 500. Result: Eleven components were identified from 95%ethanol extract of Semiaquilegiae Radix by analyzing mass spectra information, and comparing with data from relevant literatures. Conclusion: Based on HPLC-ESI-Q-TOF-MS technology, relative molecular mass and spectrometry pyrolysis data were determined, and chemical components of Semiaquilegiae Radix were determined by literature analysis and retrieval. This study provides powerful evidence for further studying the pharmacodynamics material basis of Semiaquilegiae Radix.

      摘要:Objective: To study the major chemical constituents form Arnebiae Radix. Method: The compounds were isolated with chromatographic techniques, such as silica gel, polyamide and Sephadex LH-20.The structures of isolated compounds were identified by physiochemical properties and spectral analysis. Result: Ten compounds were obtained form Arnebiae Radix and identified as ursolic acid (1), gallic acid (2), naringenin (3), deoxyshikonin (4), shikonin (5), 5-hydroxynaphthalene-2-ethoxy-1,4-dione (6), 4,4'-dihydroxy-3,5-dimethoxybibenzyl (7), 2',3-dihydroxy-4,5-dimethoxy bibenzyl (8), quercetin-3-O-β-D-glucopyrano-side (9), kaempferol-4'-O-methylether (10). Conclusion: Eight compounds 1-3, 6-10 are reported from the plants for the first time.

      摘要:Objective: To study the constituents of the high polar fraction of the leaves of Artocarpus heterophyllus, determine the antihyperlipidemia effective products in of the high polar fraction except polysaccharides, and provide a scientific basis for their subsequentant research and utilization. Method: The 10 kg rough power of the leaves of A. heterophyllu was extracted with 60%ethanol twice. The extract was combined and concentrated under reduced pressure at 4℃, standed for 24 h and filtered. The filtrate was concentrated as the high polar fraction. The high polar fraction was isolated with macroporous resin and sephadex column chromatography, and preparative HPLC. The obtained monomeric compounds were identificated by UV, MS, H-NMR and C-NMR. Result: Three compounds were separated and purified from the high polar fraction of the leaves of A. heterophyllus, and identified as vitexin-2″-O-xylosiden (compound 1, 80.5 mg), a flavone C-glycoside with two monosaccharides, molecular formula C26H28O14, relative molecular mass 564; isoquercitrin (compound 2, 4.8 mg), molecular formula C21H20O12, relative molecular mass 464; samsesquinoside (compound 3, 12.6 mg), a lignin grape glycoside with one monosaccharide, molecular formula C37H46O16, relative molecular mass 746. Conclusion: Compound 1 and 3 are discovered from A.heterophyllus for the first time. Our previous research work showed that the high polar fraction except polysaccharides had favorable hypolipidemic activity. The content of vitexin-2″-O-xylosiden in the high polar fraction of A.heterophyllus is relatively high and is the one of the antihyperlipidemia effective constituents probably.

      摘要:Objective: To provide technical support for the utilization and development of Salvia deserta by establishing the quality standards. Method: Ten batches of crude drugs of S. deserta were collected. The morphology, inspection, identification and the content of target components in these samples were studied. With rosmarinic acid as the reference substance, TLC method was used for qualitative identification. The contents of rosmarinic acid were determined by HPLC. Result: Nature and microscopic characteristics of S. deserta were described. According to the measurement results of 10 batches of samples, moisture in the crude drug was not more than 10%; the content of total ash was not more than 14%; the content of acid-insolubal ash was not more than 7%. The extracts were determined by hot dipping method; the content of ethanol extracts was tentatively determined not less than 13%and content of water extracts was not less than 15%. The spots in TLC were clear and well-separated. The sample size of rosmarinic acid was 1.001-6.006 μg (r=0.999 8), showing a good linear relationship with peak area integral value. The average recovery rate was 100.54%, RSD 1.9%. The content of rosmarinic acid was not less than 2.0%. Conclusion: The newly established standard can be used for quality control of S. deserta.

      摘要:Objective: To establish an ultra-high performance liquid chromatography-triple quadrupole mass spectrometry (UPLC-MS-MS) method for simultaneous determination of four terpene lactones in ginkgo folium extract from different manufacturers. Method: Chromatographic separation was carried out on ACQUITY UPLC BEH C18 column (2.1 mm×100 mm, 1.7 μm) with water-methanol (70: 30) as the mobile phase, at a flow rate of 0.4 mL·min-1. The column temperature was set at 30℃; Waters XevoTM TQ worked in multiple reaction monitoring (MRM) mode for negative ion detection. Result: The tested constituents were separated well within 5 min, and showed good linear relationship with the peak areas within corresponding concentration range (r>0.992 3). The average recoveries ranged from 98.13%to 102.08%with RSD value below 3.0%. There was difference in contents of total terpene lactones in Ginkgo Folium extract from different manufacturers, but all the contents complied with the requirements of Pharmacopoeia of the People's Republic of China. Conclusion: This method was simple and fast with high precision, sensitivity and repeatability, which can be used for rapid quantitative analysis of terpene lactones in Ginkgo Folium extract.

      摘要:Objective: To quickly analyze and identify the chemical constituents in Danshen-chuanxiongqin injection by using UPLC-LTQ-Orbitrap mass spectrometer. Method: The separation was performed on ACQUITY UPLC BEH C18 column (2.1 mm×50 mm, 1.7 μm) with acetonitrile (A) and 0.1%formic acid solution (B) as the mobile phase for gradient elution. The sample size was 5 μL. Electrospray ion source (ESI) was applied for qualitative analysis under the negative ion mode. The flow rate was 300 μL·min-1 and mass scan range was m/z 100-1 000. Result: Fifteen chemical compounds were identified from Danshen-chuanxiongqin injection. Conclusion: UPLC-LTQ-Orbitrap method be used to quickly identify the chemical components of Danshen-chuanxiongqin injection, laying a foundation for further studying the pharmacodynamics substances of Danshen-chuanxiongqin injection.

      摘要:Objective: To develop and validate a high performance liquid chromatography coupled with evaporative light scattering detector (HPLC-ELSD) method for the quantitative determination and fingerprint analysis of 7 active steroidal components in Xiaoaiping injection. Method: Chromatographic separation was performed on an Eclipse XDB-C18column (4.6 mm×250 mm, 5 μm) by gradient elution using acetic acid in acetonitrile solution at a flow rate of 0.8 mL·min-1. The drift tube temperature of ELSD was set to 60℃ and nebulizer gas pressure was 0.4 MPa. Method validation was performed to assure its linearity, limits of detection and quantification, precision, repeatability, stability and accuracy. Result: Fingerprint analysis obtained 13 common peaks in Xiaoaiping injection, and the similarity was higher than 0.94. The HPLC-ELSD method allowed the quantification of 7 compounds (17β-tenacigenin B, tenacigenin B, tenacigenin A, tenacigenoside A, marsdenoside I and tenacissoside F), and was successfully applied to fingerprint analysis for 11 batches of Xiaoaiping injection. Conclusion: It was the first time to apply HPLC-ELSD for simultaneous determination of 7 active ingredients and fingerprint analysis of Xiaoaiping injection. The results proved the reliability and comprehensiveness of the combination of quantitative analysis for marker ingredients and chemical fingerprint for TCM formula.

      摘要:Objective: To establish the quality standard of Aeschynanthus bracteatus, and provide scrientific basis for its quality control. Method: Empirical and microscopic identification methods were adopted to observe morphological characteristics,microscopic characteristics, miosture content, ash content and ethanol-soluble extracts were determined by empirical method and referring to the method in Chinese Pharmacopoeia (2015). TLC and HPLC were used for qualitative identification and quantitative analysis of 5,7-dihydroxy-6,4'-methoxyfldvone in A. bracteatus. Result: The macroseopical identification, microscopic identification and TLC method had the specificity. The sample size of 5,7-dihydroxy-6,4'-methoxyfldvone was between 0.108 2 μg to 1.082 μg (r=0.999 9) with an average recovery rate of 99.7% (RSD 1.1%). The content of 5,7-dihydroxy-6,4'-methoxyfldvone in 6 batches of samples was between 0.62% to 0.88%. Moisture content in A. bracteatus was between 8.9% to 11.0%; total ash was between 3.9% to 7.4%; ethanol-soluble extracts content was between 11.0% to 15.9%. Conclusion: The established method can be used for quality control of Aeschynanthus bracteatus.

      摘要:Objective: To analyze in vivo metabolic pathways and products of isorhynchophylline in rats. Method: UPLC-Q-TOF-MS with positive ion mode was established and applied to identify the metabolites of isorhynchophylline in rat plasma after intragastric administration of 37.5 mg·kg-1,mobile phase was 0.1%formic acid and acetonitrile for gradient elution. Result: A total of 13 compounds,including 1 prototype and 12 metabolites (11 of phase Ⅰ and 1 of phaseⅡ) were identified by compared with the blank biological samples and the fragmentation mechanism of MS spectra of reference substances. Conclusion: The metabolic pathway of isorhynchophylline in rats is summarized as oxidation,stereoisomerism,hydroxylation,dehydrogenation,hydrolyzation and glucuronidation.This paper enriches ESI-MS fragmentation rules of indole alkaloids and provides reference for further mass spectrometry analysis of such compounds.

      摘要:Objective: To investigate effect of Scutellariae Radix on endogenous metabolites in the serum and liver of normal mice based on metabolomics. Method: 1H-NMR based on metabolomics,coupled with multivariate and univariate statistical analysis,was used to compare differential endogenous metabolites in the serum and liver between the normal mice and those treated by Scutellariae Radix. Result: Endogenous metabolism of mice treated by Scutellariae Radix had significant changes.Compared with the blank group,contents of 10 kinds of metabolites in serum had changed,contents of 3-hydroxybutyric acid,citric acid and choline increased,levels of α-glucose and β-glucose decreased in mice serum of Scutellariae Radix group;19 kinds of metabolites in the mice liver of Scutellariae Radix group also changed,levels of glutamic acid,isoleucine and leucine increased,levels of xanthine decreased.Changes of these metabolites in mice of Scutellariae Radix group involved in glycometabolism,lipid metabolism and amino acid metabolism. Conclusion: Impact of Scutellariae Radix on metabolism of mice is related with the cold property of Scutellariae Radix,its anti-oxidative,hepatoprotective and hypoglycemic effect and other pharmacological action.This study can provide a reference for the further research of property and pharmacological mechanism of Scutellariae Radix.

      摘要:Objective: To develop the study on pharmacokinetics of Tougu Xiaotong capsules after single and multiple oral administration in rats,to discuss relevance between plasma concentration and pharmacokinetic parameters in different ways of administration,which provides basis for the clinical rational use of traditional Chinese medicine compound. Method: SD rats were randomly divided into 2 groups,including single and multiple administration group,respectively.After intragastrical administration of Tougu Xiaotong capsules,according to the area under absorbance-wavelength curve (AUAWC) being proportional to the total concentration of total solid overall composition,AUAWC was used to measure overall composition content of the capsules.The blood drug concentration data were processed in DAS 2.0 pharmacokinetic software;after single and multiple oral administration,pharmacokinetic parameter and the steady-state plasma concentration were calculated by compartment model. Result: After single oral administration,Cmax was (10.200±0.586) mg·L-1,tmax was (0.515±0.054) h,t1/2 was (1.318±0.049) h and AUC0-24 h was (13.054±0.422) mg·h·L-1,respectively.Steady-state plasma concentration was achieved after 6 d in the multiple oral administration group,Cav was (4.073±0.282) mg·L-1,DF was 1.166±0.175 and AUCss was (48.878±1.931) mg·h·L-1,respectively. Conclusion: Change of plasma concentration after multiple oral administration of Tougu Xiaotong capsules is in a good steady-state process,it is consistent with in vivo behaviour of the single oral administration group,which shows rationality in the clinical dosage of Tougu Xiaotong capsules.

      摘要:Objective: In order to establish the plasma HPLC fingerprints of Buyanghuanwu Tang(BYHWT) and to further explore its pharmacodynamic material basis. Method: The prototype components and metabolites in rat plasma were identified through comparing the fingerprints of BYHWT, rat plasma-containing active components and blank plasma. Result: After the ig administration of BYHWT, 30 main components into blood were marked in the plasma fingerprint,10 came from the original compound of rat plasma, one peak was the solvent peak. Other 19 were the drug-induced compositions, 4 of which were prototype constituents, other 15 were metabolites,such as 2 metabolites from Astragali Radix, 1 metabolite from Semen Persicae. Conclusion: The method established in this study was accurate, reliable, and can be used to research the BYHWT plasma fingerprint and efficacy material foundation.

      摘要:Objective: To observe the effect of compound Tufuling granules (CTG) on physical and chemical indexes and protein levels in hyperuricemia (HUA) rats with syndrome of phlegm-dampness. Method: Forty-eight rats were randomly divided into blank group, model group, western medicine group(febuxostat, 4 mg·kg-1), CTG low-dose group, CTG mid-dose group and CTG high-dose group(1,2,4 g·kg-1). The rats in all groups except blank group received high-fat diets combined with hypoxanthine by intragastric administration and potassium oxygen by intraperitoneal injecting to establish hyperuricemia (HUA) rat models with syndrome of phlegm-dampness, and then the rats were given with appropriate treatment. Thirty days later, the changes of behavioristics and serum uric acid (UA), alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), low density lipoprotein (LDL), protein levels of renal urate reabsorption transporter 1 (mURAT1) and liver pathology of all the rats were detected. Result: In model group, the levels of UA, ALT, AST, TG, LDL and mURAT1 protein levels were significantly higher than those in blank group (P<0.05). In CTG high-dose group, the levels of UA, ALT, AST, TG, LDL and mURAT1 protein levels were significantly lower than those in model group (P<0.05), and the scores of syndrome of phlegm-dampness were significantly improved (P<0.05). The difference was not significant in levels of AST, TG, and LDL between CTG mid-dose group and model group, but the effects on other levels in mid-dose group were similar with those in CTG high-dose group. There was no significant difference between CTG low-dose group and model group. In western medicine group, only the level of UA was significantly lower than that of model group (P<0.05), and there was no significant difference in the level of UA between western medicine group and CTG high-dose group. Liver damages were observed in liver pathology of model group. Conclusion: CTG may reduce the level of serum uric UA in HUA rats with phlegm-dampness, improve the constitution of phlegm-dampness, liver function and blood lipids as compared with the western medicine, and the mechanism may be associated with down-regulating the protein levels of renal mURAT1.

      摘要:Objective: To investigate the anti-proliferation effect of licochalcone B (LCB) on the breast cancer MCF-7 cells, and discuss its action mechanism. Method: MCF-7 cells in logarithmic growth phase were inoculated into 6-well plates at a density of 6×104 cells/well; then Trypan blue staining was performed to obtain the cell growth curve and calculate cell doubling time. MCF-7 cells in logarithmic growth phase were inoculated into 96-well plates at a density of 1×104 cells/well; then MTT assay was used to detect the anti-proliferation effect of LCB (0, 10, 20, 40, 60, 80 μmol·L-1) after treatment for 24, 48, 72 h. MCF-7 cells in logarithmic growth phase were inoculated into 6-well plates at a density of 2×105 cells/well; then the morphological changes of MCF-7 cells were observed under microscope after 48 h treatment by LCB (0, 20, 40, 80 μmol·L-1). MCF-7 cells in logarithmic growth phase were inoculated into 6-well plates at a density of 2×105 cells/well; then the morphology of apoptosis was observed by Hoechst 33258 staining method. MCF-7 cells in logarithmic growth phase were inoculated into cell culture flask at a density of 9×105 cells/flask; then after 48 h treatment by LCB (0, 20, 40, 80 μmol·L-1), apoptosis rate was determined by flow cytometry, and real-time fluorescence quantitative polymerase chain reaction (qPCR) method was used to detect the changes in Bcl-XL, Bax, Caspase-3 and Caspase-9 transcriptional levels. Result: As compared with 0 μmol·L-1 group,LCB effectively inhibited MCF-7 cell lines proliferation in a concentration-dependent and time-dependent manner; LCB treatment increased apoptosis rate of MCF-7 with obvious morphological changes, exhibiting chromosomes margination, nuclear condensation, nuclear fragmentation and other morphological features. Cell apoptosis induced by licochalcone B exhibited a concentration-dependent manner; licochalcone B significantly down-regulated the expression levels of Bcl-XL and up-regulated the expression levels of Bax, Caspase-3 and Caspase-9 (P<0.05, P<0.01). Conclusion: LCB could significantly inhibit proliferation and induce apoptosis of breast cancer MCF-7 cells, and the mechanism may be associated with up-regulating Bax,down-regulating Bcl-XL expression levels, and activating Caspase-3/Caspase-9-mediated mitochondrial apoptosis signaling pathways.

      摘要:Objective: To observe the effect of the aqueous extract of Rhei Radix et Rhizoma on superoxide dismutase (SOD) activity, catalase (CAT) activity, the contents of malondialdehyde (MDA) and hydrogen peroxide (H2O2) in rats of endotoxemia, and to preliminarily explore its protective effect on oxidative stress in endotoxemia rats. Method: Totally 50 SD rats were randomly divided into the control group, the model group, the dexamethasone group (7.5×10-4 g·kg-1), the high and low-dose Rhei Radix et Rhizoma groups (3.0, 1.5 g·kg-1).All animals were administered continuously twice daily for 7 times, once in the morning and the other in the evening.Then 0.5 h after the last administration, rats were received an intravenous injection of 5 mg·kg-1 lipopdysaccharide(LPS) to replicate the endotoxemia model.The rectal temperature of rats was tested once every 0.5 h.After 4 h, the animals were anesthetized, and the blood was collected from the femoral artery, then spleen, thymus, adrenal gland and other tissues were removed.The organ coefficient was calculated.The activities of SOD, CAT and the content of MDA, H2O2 in serum were measured by using a colorimetric indicator. Result: Compared with the model group, the aqueous extract of Rhei Radix et Rhizoma at dosage of 1.5 g·kg-1 can notably reduce the index of temperature (TRI4) in endotoxemia rats (P<0.01, P<0.05), and apparently decrease MDA, H2O2 levels in serum (P<0.05), and significantly increased SOD, CAT vitality (P<0.05). Conclusion: Oxidative stress is an important link in the endotoxemia induced by LPS, Rhei Radix et Rhizoma play a certain protective effect on endotoxemia by increasing the activity of SOD and CAT to clear free radicals excessive accumulated in the body, and intervening the signal transduction of oxidative stress injury in endotoxemia rats.

      摘要:Objective: To investigate the anti-inflammatory effect of modified Zhuye Shigao decoction and its relevant mechanisms, in order to clarify the scientific basis for superior efficiency of traditional Chinese medicines in the clinical treatment of GA (damp heat accumulation) according to Wu School's therapy for gout. Method: Totally 120 C57BL/6 mice were selected.The models of peritonitis and acute gouty arthritis were induced with monosodium urate (MSU) crystals, with 50 C57BL/6 mice for each model.Both the mice models were randomly divided into different groups, namely the model group, the positive control group (colchicine, 0.65 mg·kg-1), the modified Zhuye Shigao decoction group (11.48, 5.74, 2.87 g·kg-1), and the normal control group, with 10 mice in each group.After the intervention with different medicines, serums of both inflammation models were collected to detect the inflammatory mediators interleukin-1 (IL-1β) and Caspase-1 with enzyme-linked immunosorbent assay, and observe the effect of medicines for the acute inflammation response.The joint inflammatory reaction was induced by injecting MSU crystal suspension inside the ankle joint of the right hind limb.The arthritis was scored at 6 h after modeling; joint tissues (synovial membrane, cartilage and bone) were collected at 48 h to prepare paraffin sections, and analyze the pathological damage degree of acute gouty arthritis in mice with histological examination (HE) assay. Result: Serum IL-1β and Caspase-1 of the model group were significantly higher than the normal group (P<0.01), with more obvious inflammatory infiltration and arthritis symptom in the model group; serum IL-1β and Caspase-1 of mice in the modified Zhuye Shigao decoction groups were lower than the model group (P<0.05, P<0.01), with remarkable alleviation in inflammatory cell infiltration and symptoms of arthritis in mice. Conclusion: Modified Zhuye Shigao decoction has an anti-inflammatory effect, whose mechanism may be related to inhibition of inflammatory cytokines IL-1β, Caspase-1.

      摘要:Objective: To preliminarily investigate the protective effects of quercetin on the survival rate of H2O2-induced oxidative injury in H9C2 cardiomyocytes, and such key factors as reactive oxygen species (ROS), nicotinamide adenosine denucleotide hydro-phosphoric acid(NADPH) oxidase and apoptosis, in order to clarify the underlying mechanism. Method: A model of hydrogen peroxide H2O2-induced H9C2 cells oxidative injury was established in vitro, and divided into the normal group, the model group and the quercetin group. Cell viability was examined with an methylthiazolyldiphenyl-tetrazolium bromide(MTT) assay to determine the available concentrations of H2O2 and quercetin. The effects of quercetin on ROS level and apoptosis rate were detected by 2', 7'-Dichloroflurescin diacetate (DCFH-DA) and flow-cytometry, respectively. We also used Real-time PCR and Western blot to examine the effect of quercetin on oxidative stress and apoptosis mechanism-related molecules (NOX-4, p22phox, Bcl-2, Bax and Caspase-8)genes and protein expression level. Result: Quercetin of 10 μmol·L-1 has significant anti-oxidative and anti-apoptotic effect. Cell survival rate increased obviously in quercetin group compared with H2O2-model group (P<0.05), ROS level and apoptosis rate significantly decreased (P<0.05, P<0.01). Moreover, quercetin had an up-regulation on PI3K, Bcl-2 and NOX4 (P<0.05) and down-regulation of p22phox, Bax and Caspase-8 (P<0.05). Meanwhile, quercetin can inhibit Bax, p22phox protein expressions (P<0.05), and up-regulate PI3K, Bcl-2 protein expressions (P<0.05). Conclusion: Quercetin protects H9C2 cells from H2O2-induced oxidative injury and inhibit myocardial cell apoptosis by regulating NOX-4 p22phox, reducing intracellular ROS level, activating PI3K and increasing Bcl-2.

      摘要:Objective: To investigate the inhibitive effect of Gehua Jiecheng decoction on tumor growth of transplanted H22 in BALB/c mice and expressions of adenomatous polyposis coli (APC) protein in tumor tissues. Method: Thirty-four male BALB/c mice were randomly divided into four groups, namely blank group, model group and Gehua Jiecheng decoction of high and low dose(40,20 g·kg-1) groups. Except for the blank control group, the other groups were included in the preparation of the transplanted liver cancer mouse model. At the 2nd day after the modeling, the mice were given drugs, the blank control group and the model group were given NS, ig, and Gehua Jiecheng decoction high and low dose groups were given the same volume of decoction, ig, with the dosage 0.2 mL for each. After 12 days, all mice were sacrificed. Their tumor lumps were collect to weigh and calculate the inhibition rate and make pathological sections. Spleen and thymus were collected to calculate the spleen index and thymus index, enzyme-linked immunosorbent assay (ELISA) was used to examine interleukin-1 (IL-1) and tumor necrosis factor-α (TNF-α) content in peripheral blood after blood was collected and centrifuged. Immunohistochemistry was used to examine the expression of APC, Western blot method was adopted to detect the expression of APC protein expression. Result: Compared with the model control group, the inhibition rate of Gehua Jiecheng decoction high and low dose groups were 37.05%and 16.51%(P<0.05, P<0.01), the model group showed high expression of APC protein, Gehua Jiecheng decoction high and low dose groups showed significantly lower APC expressions than model group (P<0.05, P<0.01). Conclusion: Under a certain dose range, Gehua Jiecheng decoction can inhibit the growth of liver cancer in mice tumor blocks in a dose-effect manner, and regulate and control cancer suppressor gene APC, which may one of Gehua Jiecheng decoction's mechanisms for inhibiting liver cancer.

      摘要:Objective: To investigate the synergistic effect of Qige San and cisplatin in inhibiting proliferation of esophageal carcinoma cells in hypoxia, in order to explore the mechanism from the perspective of miR-21 and its target genes. Method: Drug-containing serum was prepared. The single or combined effects of Qige San and cisplatin on cells activation were tested by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromid (MTT) assays. Apoptosis and cell cycle arrest induced by Qige San and cisplatin were detected by flow cytometry. MiR-21, programmed cell death 4 (PDCD4), phosphatase and tensin homolog (PTEN) mRNA expressions were detected by Real-time PCR, while PDCD4 and PTEN protein expression were detected by Western blot. Result: Qige San in concentrations of 8%and 10%combined with 1 mg·L-1 cisplatin has a synergistic effect in inhibiting proliferation of esophageal carcinoma cells, with the coefficients of drug in interaction (CDI) of 0.90, 0.94, respectively. The expressions of miRNA-21 in cisplatin high concentration group, and combination high and low concentration group were significantly lower than that in the control group(P<0.05). The expression of miRNA-21 in combination high and low concentration group was significantly lower than that of cisplatin groups with the same concentration (P<0.01), and the low concentration group (P<0.05). The expression of PDCD4 mRNA was significantly lower in the cisplatin high concentration group and the Qige San group than in control group (P<0.05), whereas the expression of PTEN and PDCD4 mRNA in combination high and low concentration group was significantly higher than that in control group (P<0.05), and PDCD4, PTEN mRNA expressions in combination high and low concentration group was significantly higher than that of cisplatin groups with the same concentration (P<0.01), and PTEN mRNA in high concentration combination group was higher than that in lower concentration combination group (P<0.01). Compared with the control group, the expression of PTEN protein was significantly higher in combination high and low concentration group(P<0.05), and that in combination high and low concentration group was significantly higher than that in the same concentration of cisplatin group (P<0.01). PDCD4 was significantly higher in cisplatin high and low concentration group and combination high and low concentration group that in control group (P<0.05), and combination high concentration group was significantly higher than that in cisplatin group and combination low concentration group (P<0.05, P<0.01). Conclusion: Qige San and cisplatin have a synergistic effect in inhibiting proliferation of esophageal carcinoma cell line EC9706, and the mechanism may be related to increase the expression of PDCD4 and PTEN by inhibiting miRNA-21 expression.

      摘要:Objective: To study the effect of apigenin in treatment of rats osteosarcoma apoptosis and B-cell lymphoma 2(Bcl-2), Bcl-2 assaciated X protein(Bax), Caspase-3, Caspase-9 and nuclear factor-κB(NF-κB) protein expressions. Method: Totally 50 rats were randomly divided into model group, cisplatin group (10 mg·kg-1), apigenin groups (1, 2, 4 mg·kg-1), with 10 rats in each group. The rat osteosarcoma model was made by operation. All groups were given drups for 30 days. Oteosarcoma was collected and weighed 24 h later after the last administration. TUNEL assay was adopted for in situ labeling of apoptosis of osteosarcoma cells. Immunohistochemistry was used to detect expressions of Bax, Bcl-2 in osteosarcoma. Caspase-3, Caspase-9 and cytochrome C(Cyt-C) expressions in osteosarcoma were tested using Western blot assay. Result: Compared with model group, apigenin-treated groups showed decrease in osteosarcoma weight, significant increase in the number of apoptotic cells, obvious up-regulation in Bax, Caspase-3, Caspase-9 and Cyt-C protein, and down-regulation in the expression of Bcl-2. Conclusion: Apigenin can increase Bax, Caspase-3, Caspase-9, the expression of Cyt-C, down-regulate the expression of Bcl-2, inhibit the proliferation of osteosarcoma cells, and induce apoptosis in osteosarcoma cells, so as to show a therapeutic and inhibitive effect on osteosarcoma.

      摘要:Objective: To observe the effects of total saponins from Albiziae Cortex(SA) on CD4+ and CD8+T lymphocyte subsets, levels of interleukin(IL)-2 and tumor necrosis factor (TNF)-α, apoptosis inducing factor (Fas), apoptosis-induced ligand (FasL) and proliferating cell nuclear antigen (PCNA) protein expressions in tumor-bearing mice, and to explore its intervention mechanism of cellular immune functions. Method: Totally 60 Kunming mice were randomly divided into normal group, model group, cyclophosphamide (CTX) group, SA high, medium and low dose groups. The H22 tumor bearing mice models were established. CTX (20 mg·kg -1·d-1), SA (4, 2, 1 mg·kg -1·d-1) were given to the mice of experimental groups by intraperitoneal injection respectively. After continuous treatment for 11 days, the tumor inhibition rate, the levels of IL-2 and TNF-α in serum were detected, and the expressions of Fas, FasL and PCNA protein expressions in tumor cells were determined by immunohistochemical staining; tumor tissue morphology was observed by htoxylineosin(HE) staining. Result: As compared with the model group, the average weight of transplanted tumor in various dose groups of SA reduced significantly (P<0.05,P<0.01). The number of CD4+ and CD8+ T cells was increased obviously in medium dose group(P<0.05,P<0.01), but CD4+/CD8+ ratio was not affected. The levels of IL-2 in medium and high dose groups were significantly increased(P<0.05), and the levels of TNF-α had no change. The FasL and PCNA protein expressions were significantly decreased in various dose groups of SA (P<0.01), and Fas expression level in medium and high dose groups was significantly elevated (P<0.01). Conclusion: SA have immunomodulatory effects on cellular immunity. The mechanisms may be associated with up-regulating CD4+ and CD8+ T cell subsets, promoting IL-2 secretion, increasing Fas protein expressions and inhibiting FasL and PCNA protein expressions.

      摘要:Objective: To observe the protection effect of extract from Xinjiang mulberry on carbon tetrachloride (CCl4) and alcohol-induced acute liver injury in mice. Method: Totally 60 Kunming mice (30 males and 30 females) were randomly divided into 6 groups:normal group and model group with intragastric administration of distilled water,positive group (bifendate pills,0.40 g·kg-1),Xinjiang mulberry extract low dose group,middle dose group and high dose group (1.525,3.050, 6.100 g·kg-1). All the other mice except those in normal group received CCl4 and alcohol to establish acute liver injury models in mice. After successful modeling,the mice in all groups received intragastric administration for continuous 7 days. The liver coefficient in mice was determined after drug administration,and colorimetric method was used to determine the activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum of CCl4 liver injury mice,and the contents of triglyceride (TG),malondiadehyde (MDA) and glutathione (GSH) in liver homogenate of alcoholic liver injury mice. HE staining method was used to observe the pathological section of liver tissues in mice. Result: As compared with the normal group,the activities of ALT and AST in serum of CCl4 liver injury mice in model group were significantly increased (P<0.01);the contents of MDA and TG in liver tissues of alcoholic liver injury mice in model group were significantly increased while the content of GSH was significantly reduced (P<0.05,P<0.01);the liver injury was more obvious in the model group during the pathological observation. As compared with the model group,the extract from Xinjiang mulberry could reduce the activities of ALT and AST in serum,significantly reduce the contents of TG and MDA,and significantly increase the content of GSH,with statistically significant differences (P<0.05,P<0.01). The pathological examination results indicated that the extract from Xinjiang mulberry had obvious liver-protecting effect. Conclusion: The extract from Xinjiang mulberry has notable protective effect for the CCl4 and alcohol-induced liver injury in mice.

      摘要:Objective: To study the tumor cytotoxicity of different extracts and different extraction methods of Periplaneta americana. Method: Cell counting kit-8 (CCK-8) method was used to compare the anti-tumor activities of different extracts (ethanol extract, petroleum ether fraction, ethyl acetate fraction and n-butanol fraction) and different extraction methods (percolation method, ultrasonic method, and reflux method) on three kinds of tumor cells (human liver cancer SMMC-7721 cells, human erythroleukemia K562 cells, and human colon cancer cell line HCT116). Result: The ethanol extract and ethyl acetate fraction of P. americana had inhibitory effect on the proliferation of above tumor cells in varying degrees, and the activity was strongest for ethanol extract; P. americana extracted by three extraction methods showed antitumor activity in vitro in varying degrees, and the percolation method had the strongest inhibitory effect on three kinds of tumor cells with minimal IC50 values. Conclusion: Ethanol extract of P. americana had strongest in vitro antitumor activity, followed by ethyl acetate fraction, and percolation method shall be used for extraction.

      摘要:Objective: To investigate the effect and mechanisms of epigallocatechin-3-gallate (EGCG) on the human gastric adenocarcinoma cell lines MGC-803. Method: MGC-803 cells in logarithmic growth phase were treated with different doses of EGCG (25, 50, 100 μmol·L-1), and another blank group was established. After 48 h cells culturing, methyl thiazolyl tetrazolium (MTT) assay was used to evaluate the proliferation of cells, and the apoptosis of cells was determined by flow cytometric analysis. The protein expression levels of p-p38, p38, Bax, Bcl-2, and Cleavage-Caspase-3 were detected by Western blot. Result: As compared with the blank grouo, EGCG treatment could induce the proliferation of MGC-803 cells and inhibit their apoptosis in a concentration-dependent manner, significantly up-regulate the protein expression levels of Cleavage-Caspase-3 and Bax, and significantly down-regulate the protein expression levels of p-p38 and Bcl-2, with statistically significant differences (P<0.05, P<0.01), but EGCG had no obvious effects on the expression of p38. Conclusion: EGCG can induce proliferation of MGC-803 cells and inhibit their apoptosis by suppressing p38 signaling activation.

      摘要:Objective: To investigate the clinical curative of Quyidi prescription in patients with type 2 diabetes mellitus (T2DM) in a hypercoagulable state. Method: Totally 60 patients were randomly divided into two groups by using the digital table method. The traditional Chinese medicine(TCM) group was treated with TCM decoction, 2 times per day, the control group was orally treated with oral sodium phosphate ligustrazine hydrochloride, 50 mg/time, tid. After 4 weeks, their tissue factor (TF), blood rheology, coagulation function, platelet aggregation rate, blood lipid, blood glucose and other related indicators of TCM symptom scores were observed for a statistical analysis and comparison for efficacies. Result: In the aspect of tissue factors, the treatment group was superior to the control group (P<0.01), which indicated that the TF level of the treatment group was significantly higher than that of the control group. After treatment, the blood rheological indexes of both groups were reduced (P<0.05), and the treatment group was superior to the control group (P<0.05). In terms of blood coagulation function, and blood lipid and blood glucose levels, both groups showed obvious alleviations(P<0.05, P<0.01), and the treatment group was superior to the control group. Meanwhile, the treatment group was significantly better than the control group in the efficacy of TCM syndrome. There was no adverse event and reaction during the trial period. Conclusion: The Qi supplementing, Yin nourishing, promoting blood activating and phlegm expelling therapy can effectively decrease the plasma levels of TF in T2DM patients, and alleviate blood lipid and blood coagulation indexes, at the same time, it can alleviate glucose and lipid metabolic disorder, and TCM syndrome scores among T2DM patients, in order to improve the quality of life.

      摘要:Objective: To evaluate the clinical effect of renal failure recipe on renal function and fibrosis in patients with early and middle-term chronic renal failure. Method: Totally 60 cases of patients with three and four-stage chronic kidney disease (spleen and kidney deficiency, blood stasis type) were selected and randomly divided into control group and treatment group. The former received losartan in addition to western medicine integration treatment, while the latter was given renal failure recipe in addition to the former's therapy. After 6 months of treatment, we observed the changes in traditional Chinese medicine (TCM) syndrome scores and curative effect, therapeutic efficacy, hemoglobin, renal function, 24-hour urine protein, connective tissue growth factor(CTGF) and bone morphogenetic protein-7 (BMP-7) in the two groups of patients. Result: The curative effect of TCM syndrome and curative effect of treatment group were obviously superior to that of control group (P<0.01). Treatment group was superior to control group in reduction in 24-hour urine protein and creatinine and increase in glomerular filtration rate (P<0.05, P<0.01). The two groups showed significant reduction in serum CTGF levels (P<0.05), and increase in blood bone morphogenetic protein-7 (BMP-7) level (P<0.05). Conclusion: Renal failure recipe shows an obvious clinical curative effect in treating patients with early and middle-term chronic renal failure compared with the control group, and can reduce proteinuria, alleviate renal function, and prevent renal fibrosis. Its mechanism may be related to the down-regulation of serum CTGF and upregulation of BMP-7 expression.

      摘要:Objective: To observe the effects of Shuanghe Zaoren granule on sleep electroencephalogram (EEG) and its clinical curative effects on patients with primary insomnia. Method: Totally 120 patients with primary insomnia were randomly divided into observation group and control group. The observation group (60 cases) was treated with Shuanghe Zaoren granule, 1 pack/time, tid. The control group (60 cases) was treated with Estazolam tablets, 1 mg, bedtime, qd. The therapeutic course was 2 weeks for both groups. The Pittsburgh Sleep Quality Index (PSQI), traditional Chinese medicine (TCM) symptom score and clinical efficacy, and key parameters of sleep EEG, such as sleep latency (SL), total sleep duration (TSA), awakening frequency (AT), awakening duration (ASA), awakening sleep ratio (A/TSA), S1, S2, S3, S4, paradoxical sleep (REM) and sleep efficiency (SE) were observed and evaluated for both groups. The clinical curative effects of the two groups were recorded. Result: The total effective rate in the observation group was 90%, which was much higher than 73%in the control group (P<0.05). Compared with before treatment, a shortened sleep latency, awakening frequency and awakening duration, prolonged deep sleep, increased sleep efficiency, shortened light sleep and paradoxical sleep (REM) were observed in the sleep EEG parameters in two groups (P<0.01). However, the sleep electroencephalogram parameters in the observation group were better than control group (P<0.05, P<0.01). The scores of PSQI in observation group and control group were 16 points and 17 points, respectively before treatment, after treatment, the PSQI score was significantly decreased to 5 points and 8 points in two groups (P<0.01). The PSQI score of observation group was significantly lower than the control group (P<0.01). Score of TCM clinical symptoms after treatment were significantly lower than that before treatment in each group (P<0.01). In the observation group, TCM clinical symptom score reduced by 3 times of the control group (P<0.01). Conclusion: Shuanghe Zaoren granule could shorten the time for falling asleep, decrease awakening frequency and awakening duration, prolong the deep sleep, improve the structure and quality of sleep, with an especially better clinical curative effect for patients with fire excess from yin deficiency and heart-spleen deficiency. Therefore, it is a safe and effective drug to treat primary insomnia.

      摘要:Objective: To observe the clinical effect of traditional Chinese medicine(TCM) leg balneotherapy in the treatment of knee osteoarthritis with wind-cold-dampness arthralgia, in order to explore a new TCM method for the treatment of osteoarthritis. Method: In a retrospective study, 102 cases of patients with knee osteoarthritis treated in South Area of Guang'anmen Hospital of China Academy of Chinese Medicine Sciences in recent years were selected and randomly divided into the treatment group and the control group. The 51 patients in treatment group was given TCM Tuiyu prescription or leg balneotherapy, and the 51 patients in control group was treated with hot compress with Guyouling liniment for two weeks. At the first and second weeks before and after the treatment, VAS was used to score knee joint functions and TCM syndrome scores, so as to evaluate the clinical effect of the therapy. Result: At the first and second weeks after the treatment, VAS scores significantly reduced in the treatment group (P<0.05). Western Ontario McMaster Universities (WOMAC) scores in both groups were significantly decreased, and the treatment group was superior to the control group (P<0.05). At the second week after the treatment, TCM symptom scores were obviously reduced (P<0.05), and the treatment group showed a higher reduction than the control group. There was no pain and adverse event during the treatment. Conclusion: TCM leg balneotherapy is effective and simple, and worthy of promotion.

      摘要:Objective: To evaluate the clinical efficacy of Qinggan Qushi Huoxue decoction in treatment of patients with nonalcoholic fatty liver. Method: Totally 112 cases of nonalcoholic fatty liver patients were randomly divided into control group (57 cases) and treatment group (55 cases). Control group was given polyene phosphatidylcholine capsules, treatment group was treated with Qinggan Qushi Huoxue decoction. The treatment lasted for two months. Before and after the treatment, patients' digestive system ultrasound, liver function, blood lipid, insulin resistance index (HOMA-IR), weight index (BMI), waist circumference and symptom scores were observed. Result: The treatment group showed a total effective rate of 100%, which was higher than control group (P<0.05), compared with before treatment, two groups of patients showed significant decreases in alanine aminotrans-ferase (ALT), aspirate aminotrans-ferase (AST), total cholesterol (TC), triglyeride (TG), low density lipoprotein cholesterol (LDL-C), HOMA-IR, BMI and waist circumference after treatment (P<0.05,P<0.01), and treatment group was better than control group (P<0.05). Conclusion: Qinggan Qushi Huoxue decoction can significantly alleviate the clinical symptoms of patients with alcoholic fatty liver, reduce their weight index and waistline, alliviate liver function and insulin resistance, and regulate lipid metabolism.

      摘要:Objective: To investigate the efficacy of Tiaochong Jiangya decoction combined with acupuncture in treating menopausal hypertension with syndrome of disturbance of Chong and Ren, and observe its effect on neurotransmitter and levels of sex hormone. Method: One hundred and sixty patients with menopausal hypertension were selected and divided into western medicine group and traditional Chinese medicine (TCM) group according to random number table. Patients in western medicine group took metoprolol succinate sustained-release tablets, 47.5 mg/time and qd. The patients in TCM group were treated with Tiaochong Jiangya decoction combined with acupuncture (Bahui, Hegu, Taichong,Quchi acupoints). The treatment course was 8 weeks for both groups. The blood pressure levels were detected for 3 times before and after treatment, to get mean value. Chong-Ren symptom scores and Kupperman scores were compared between two groups pre-post treatment. The levels of neuropeptide Y (NPY), neurotensin (NT), serum estradiol (E2), testosterone (T), progesterone (P) and sex hormone binding globulin (SHBG), were detected in two groups both before and after treatment. Result: The clinical effective rate was 91.89%for blood pressure in TCM group, higher than 82.67%in western medicine group, but there was no statistically significant difference. The total effective rate was 93.24%in TCM group, higher than 72%in western medicine group (P<0.01). After treatment, there was no statistically significant difference in SBP and DBP levels between two groups. After treatment, Chong-Ren disturbance symptom scores and Kupperman scores in TCM group were lower than those in western medicine group (P<0.01). After treatment, the levels of NPY, NT, P, and T in TCM group were lower than those in western medicine group, but levels of E2 and SHGB were higher than those in western medicine group (P<0.01). Conclusion: Tiaochong Jiangya decoction combined with acupuncture has equivalent efficacy with metoprolol succinate sustained-release tablets in treating menopausal hypertension with disturbance of Chong and Ren, and the mechanism of action may be associated with regulating neurotransmitter and levels of sex hormone.

      摘要:Objective: To discuss the clinical efficacy of Danqing granules in treating acute intrahepatic cholestasis (IC) with liver-gallbladder dampness-heat syndrome, and investigate the action mechanism from inflammatory reaction and oxidative stress. Method: Ninety-nine patients were randomly divided into control group (49 cases) and observation group (50 cases) by random number table. Patients in control group received conventional liver protection treatment, enzyme reducing treatment and other symptomatic treatment, orally took ursodeoxycholic acid tablets, 250 mg/time, 3 time/day. Based on the treatment of control group, patients in observation group received additional Danqing granules, 10 g/time, 3 time/day. The treatment course was 4 weeks for both groups. Before and after treatment, levels of total bilirubin (TBIL), alkaline phosphatase (ALP), gamma-glutamgtranspeptidaser (GGT), total bile acid (TBA), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) were detected. Scores of liver-gallbladder dampness-heat syndrome were graded every week. The levels of tumour necrosises factor-α (TNF-α), interleukin-6 (IL-6), interleukin-1 (IL-1), total superoxide dismutase (T-SOD), and malondialdehyde (MDA) were also detected both before and after treatment. Result: The total effective rate was 92%in observation group, higher than 75.5%in control group (P<0.05). At the second, third and forth week after treatment, scores of liver-gallbladder dampness-heat syndrome in observation group were lower than those in control group (P<0.01). After treatment, the levels of TBIL, ALP, GGT, TBA, ALT, AST, TNF-α, IL-6 and IL-1 were lower than those in control group (P<0.01). The levels of SOD were increased in both groups after treatment, and the level in observation group was higher than that in control group(P<0.01). Levels of MDA were decreased in both groups after treatment, and the level in observation group was lower than that in control group (P<0.01). Conclusion: Danqing granules based on the western medicine group can relieve clinical symptoms, protect liver function, and improve effect, and its mechanism of action may relieve inflammatory injury and inhibit oxidative stress.

      摘要:Objective: To observe the efficacy of Weijin Xiaoyong decoction in the adjuvant therapy of bronchiectasis at acute exacerbation stage with syndrome of phlegm-heat obstructing lung, and investigate its effects on levels of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in serum and sputum. Method: Ninty-eight patients with acute exacerbation stage bronchiectasis were randomly divided into treatment group (49 cases) and control group (49 cases) according to random number table. Non-pharmaceutical interventions were given according to ‘Expert Consensus on Diagnosis and Treatment of Adult Bronchiectasis Disease’ in both groups. In the control group, 0.4 g ofloxacin hydrochloride injection was added to 400 mL normal saline for anti-infection (200 mL/time, bid), 0.6 g ofloxacin hydrochloride injection was added to 600 mL normal saline for pseudomonas aeruginosa infection (300 mL/time, bid), 30 mg ambroxol injection was added to 100 mL normal saline for dissipating phlegm, qd. The patients in treatment group also received Weijin Xiaoyong decoction (one dose/day, boiled and taken orally twice) based on the treatment in control group. The treatment course was 2 weeks for both groups. Forced expiratory volume in one second (FEV1), FEV1/forced vital capacity (FVC), blood oxygen partial pressure (PaO2), CO2 partial pressure (PaCO2), blood oxygen saturation (SaO2) and clinical symptom scores were compared between two groups. The clinical efficacy was analyzed in two groups. The levels of MMP-9 and TIMP-1 in serum and sputum were detected in both groups. Result: After treatment, FEV1, FEV1/FVC, PaO2 and SaO2 levels in treatment group were significantly higher than those in control group, while clinical scores such as PaCO2 and cough were significantly lower than those in control one (P<0.01). The total effective rate was 95.92%in treatment group, significantly higher than 79.59%in control group (P<0.05). After treatment, the levels of MMP-9 in serum and sputum in treatment group were significantly lower than those in control group, while TIMP-1 levels were higher than those of control group (P<0.01). Conclusion: Weijin Xiaoyong decoction based on the routine western medicine, has an exact effect in the adjuvant therapy of bronchiectasis at acute exacerbation stage with syndrome of phlegm-heat obstructing lung, and the mechanism may be associated with down-regulating MMP-9 levels in serum and sputum, and up-regulating TIMP-1 level.

      摘要:Objective: To explore the molecular mechanism of Shen's antihypertensive Siwu decoction treating hypertension. Method: By reference to Chinese Medicine Dictionary (second edition), active ingredients of the four herbs (Uncariae Ramulus Cum Uncis, ALismatis Rhizoma, Chuanxiong Rhizoma, Raphani Semen) in Shen's antihypertensive Siwu decoction were obtained. The corresponding target proteins of the ingredients were retrieved in PubChem database. Meanwhile, hypertension genes were also retrieved in PubMed gene database. The protein and gene data were imported into the molecular network analysis platform to respectively build the corresponding molecular networks and biological pathways. Through online comparison, some targets and molecular network mechanisms for Shen's antihypertensive Siwu decoction treating hypertension can be found. Result: Shen's antihypertensive Siwu decoction showed a treatment effect on hypertension by regulating 3 molecules [nuclear factor-κB(NF-κB), vascular endothelial growth factor(VEGF) and extracellular signal-regulated kinase 1/2(ERK1/2)] and 2 pathways (G-protein coupled receptor signaling and VEGF signaling). Conclusion: Shen's antihypertensive Siwu decoction is an effective prescription to treat hypertension. Molecular network analysis can predict the molecular mechanisms of traditional Chinese medicine compounds.

      摘要:Objective: To investigate the prescription principles of traditional Chinese medicine (TCM) for pulmonary fibrosis, in order to provdie reference for clinical TCM treatment and research and development of new medicine for PF. Method: Literatures with titles of ‘pulmonary fibrosis’ and ‘treatment’, and the publishing time of ‘2005-2014’ were retrieved in the databases of CNKI, CBM and TCM, and analyzed with the methods of ‘frequency analysis’ and ‘cluster analysis’. Result: Totally 382 articles were retrieved, and 129 articles were selected, involving 182 prescriptions. The most frequently used traditional Chinese medicines for the treatment of PF were Astragali Radix and Salviae Miltiorrhizae Radix. Among the herbs used for more than 26 times, Buqi herbs accounted for 52%, and Huoxue herbs accounted for 48%. The accumulative frequency of tonic herbs, Huatan herbs, Zhike herbs, Qingre herbs and Huoxue herbs reached 77.39%. As to the Pathogenesis, Qi deficiency and blood stasis took up the largest proportion of 40.58%. Conclusion: Pathogenesis of PF is the most closely related to deficiency, phlegm and blood stasis. Main treatment principles are supplementing Qi and nourishing Yin, nourishing the lungs and clearing the phlegm, promoting blood circulation to remove meridian obstruction, and nourishing the lungs and clearing the heat. Medicines for supplementing Qi, promoting blood circulation and clearing the heat have a higher use frequency.

      摘要:Objective: To make the systematic review and Meta-analysis for the effect of post-laparoscopic oral administration with Danzhu Xiaoyi decoction on the pregnancy, abortion and recurrence of treating patients with endometriosis (EMT). Method: Randomized controlled trials (RCTs) and controlled trials (CCTs) related to the effects of post-laparoscopic oral administration with Danzhu Xiaoyi decoction for EMT were collected in CNKI, CBM, VIP Wanfang database and PubMed (before August 2015). The quality of the RCTs and CCTs were assessed using Cochrane systematic review, involving selection bias, performance bias, measurement bias, attrition bias, and reporting bias. The overall effect of multiple findings was studied by using RevMan 5.3 software through a Meta-analysis for a fixed effects model, and then heterogeneity evaluation and sensitivity analysis were conducted. Result: A total of 4 literatures were included. According to the results of the Meta-analysis, compared with pure laparoscopic surgery, post-laparoscopic oral administration with Danzhu Xiaoyi decoction for EMT had a significantly higher pregnancy (I2=0%), lower heterogeneity [RR=1.47, 95%CI (1.13, 1.90)], with significance test Z=4.96 (P<0.000 01). At the same time, it had an obviously lower the incidence of spontaneous abortion (I2=0%), lower heterogeneity [RR=0.29,95%CI (0.09, 0.92)], with significance test Z=2.28 (P=0.02). In addition, there were a significantly lower incidence of recurrence (I2=0%), lower heterogeneity [RR=0.23,95%CI(0.07, 0.78)],with significance test Z=2.36 (P=0.02). Conclusion: Post-laparoscopic oral administration with Danzhu Xiaoyi decoction could improve the pregnancy rate and reduce the incidence of spontaneous abortion as well as recurrence. However,it is still necessary to carry out high-quality three-blinded RCT with a large sample size.

      摘要:Objective: To evaluate the efficacy and safety of combined treatment of Wenxin granules and bisoprolol in treating arrhythmias induced by heart failure (HF). Method: Literatures were searched in PubMed, EMbase, CNKI,Cochrane Central Register of Controlled Trials, Wanfang and VIP database (since the establishment to May 10, 2015) to select qualified RCTs. RevMan 5.3 software was used to make a Meta-analysis. Result: Totally 279 relevant literatures, involving 6 RCTs (527 patients), were selected and included in this study. According to the results of the meta-analysis, the cardiac failure-arrhythmias group showed a higher rate in improving arrhythmia frequency than the control group [OR=3.85, 95%CI (2.39, 6.20), P<0.000 01], and in improving palpitations than the control group [OR=4.48, 95%CI (2.42, 8.30), P<0.000 01]. The combined treatment group (Wenxin granules + bisoprolol) showed a lower rate of adverse events than the control group [OR 0.42, 95%CI (0.18, 1.01), P<0.05]. Conclusion: Combined treatment with Wenxin granules and bisoprolol is more effective and causing less adverse events than the use of only bisoprolol for treating HF-induced ventricular arrhythmias.

      摘要:Objective: To investigate the application of the grey screening method in the research of the prescriptions treating colorectal cancer (CRC). Method: According to the inclusion criteria, 132 cases of prescriptions were collected, involving 260 herbal medicines, with the cumulative frequency of 2 363 times. Through the self-built ‘syndrome-herbal properties-frequency database’, the grey screening method was adopted to analyze the rules of traditional Chinese medicine (TCM) prescriptions for CRC. Result: In prescribers for TCM in treatment of CRC, doctors gave priority to strengthening and consolidating body resistance, and also consideration to clearing heat-toxicity, promoting blood circulation for removing blood stasis, and regulating Qi-flowing for hemostasis. Moreover, strengthening vital Qi and eliminating pathogenic factor should be taken into account, which reflects TCM's holism of and treatment based on syndrome differentiation. The analysis on core drugs shall be made to determine that the treatment of CRC medicines often had effects in tonifying deficiency, clearing heat, promoting blood circulation for removing blood stasis. The top 5 herbs with the hugest grey correlation degree are Saposhnikoviae Radix,Scutellariae Radix,Sanguisorbae Radix, Rhei Radix et Rhizoma,Rehmanniae Radix, and evolved into 5 basic prescriptions. Conclusion: The grey theory, in the combination of the grey screening method for multivariate analysis, conforms to the thought of ‘treatment based on syndrome differentiation’ in TCM, and provides new ideas for clinical medications.

      摘要:DNA gene identification is used as a more important molecular biological technology to identify traditional Chinese medicine in recent years. It can identify good herbs at the genetic level, with a high accuracy. DNA gene identification includes DNA molecule genetic markers, DNA hybridization and DNA barcoding molecular identification method, particularly the fastest growing one is DNA barcoding molecular identification method. DNA barcoding molecular identification method is a molecular biological technique to identify species using an admittedly short DNA sequence in genome, and an effective complement to traditional morphological identification methods. Because DNA sequences of different species were composed of four bases, namely adenine (A), guanine (G), cytosine (C) and thymine (T), analysis on a specific sequence of DNA fragments can distinguish different species. Hebert, proposed the animal species identification method by measuring mitochondrial cytochrome C oxidase subunit I (COI) gene sequences in 2003.CHEN Shi-lin proposed to identify genes with COI gene-dominated and ITS2 gene-supplemented animal medicine DNA barcoding system on the basis of a large number of experiments on animals. A large number of experiments have showed that, DNA barcoding is accurate, feasible, simple and general to identify animals, and provides a new method for identifying animal species, finding new species and authenticating animal medicines. However, because the technology is to identify by gene, it has higher requirements for experimental materials and extraction methods. Particularly, animal medicines prepared many years ago have a severe DNA degradation, which sets higher requirements for the DNA extraction technology. In this case, the technology can only identify the exact source, but not its medicinal parts, which has to be identified in the combination with other identification methods. In this article, studies on DNA barcodes at home and abroad are summarized to provide a theoretical basis for future studies and applications.
