      摘要:Objective: Fuzzy comprehensive evaluation method was used to evaluate subjective and objective indexes of Qingjiang orally disintegrating tablets at the same time for optimization of its formulation. Method: In order to optimize the amount of disintegrants and flavoring agent, central composite design was used, and the subjective indexes (degree of sweet and sour, gravel sense) and objective index (disintegration time) of Qingjiang orally disintegrating tablets were evaluated at the same time. Result: Optimal formulation of Qingjiang orally disintegrating tablets was Qingjiang taste-masking powder of 2 g, microcrystalline cellulose (MCC) of 1.472 g, low-substituted hydroxypropyl cellulose (L-HPC) of 0.398 g, crospolyvinylpyrrolidone (PVPP) of 0.252 g, flavoring agent of 0.301 g, lubricant of 0.06 g.Qingjiang orally disintegrating tablets had a good favor without gravel sense, disintegration time was only 48.8 s. Conclusion: Fuzzy comprehensive evaluation method can evaluate the subjective and objective indexes at the same time, and its results have good accuracy and repeatability.

      摘要:Objective: To observe effect of Psoraleae Fructus and Myristicae Semen in Ershen Wan after and before being processed on intestinal flora and expression levels of uncoupling protein 2 (UCP2) mRNA of spleen-kidney Yang deficiency diarrhea rats, to preliminarily explore its possible mechanism. Method: Rat model of spleen-kidney Yang deficiency diarrhea was made through perfusion of adenine, irregular diet and reception of ice Sennae Folium extract.Rats were averagely subdivided into four groups, such as normal group, model group, raw products group and processing group of Ershen Wan.Real-time fluorescence quantitative technique was employed to investigate effect of Ershen Wan on intestinal flora.Expression levels of UCP2 mRNA in renal tissues were assayed by immunohistochemical staining SP method. Result: Intestinal flora and expression levels of UCP2 mRNA of spleen-kidney Yang deficiency diarrhea rats changed obviously by comparing with before modeling;flora of Bifidobacterium longum and Lactobacillus was abnormally low, contents of Escherichia coli and Enterococcus faecalis increased significantly, the expression level of UCP2 mRNA in rat renal tissues of model group was significantly increased.The alteration of intestinal flora and expression levels of UCP2 mRNA recovered after receiving Ershen Wan, and the processing group was the best. Conclusion: Ershen Wan, consisting of salt-fried Psoraleae Fructus and bran-roasted Myristicae Semen, has function of promoting proliferation of intestinal available bacteria, inhibiting intestinal harmful bacteria and down-regulating expression levels of UCP2 mRNA in renal tissues, which may be related to regulation of intestinal flora and the body's basal metabolic rate.

      摘要:Objective: To prepare quercetin-poly(lactic-co-glycolic acid) block copolymer (QC-PLGA) nanoparticles freeze-dried powder and investigate its in vitro release rule. Method: QC-PLGA nanoparticles were prepared through emulsion solvent evaporation method, optimal formulation process was defined by means of orthogonal test.QC-PLGA nanoparticles mixed with the protective agent were freeze-dried, which was explored in vitro release rule through dynamic dialysis technology. Result: Optimum preparation parameters were as follows:polyvinyl alcohol (PVA) of 0.2%, PLGA of 10 g · L-1, proportion of oil and water phase of 1: 35, QC amount of 5 mg, 2% lactose as freeze-drying protective agent.Surface of QC-PLGA freeze-dried powder was smooth without shrinking subsidence, which had dense structure.After the injection water was added to sway, redispersibility was good.The rule of in vitro release was in line with Weibull equation model, which was fitted the drug release kinetics equation of ln[ln(1/1-Q)]=0.399lnt-1.503 (R2=0.973). Conclusion: The preparation technology of QC-PLGA nanoparticle freeze-dried powder is simple and feasible, which is easy to store with stable properties.Compared with quercetin, QC-PLGA nanoparticle freeze-dried powder has an obvious sustained release effect.

      摘要:Objective: To investigate the mechanism of improving dissolution of dehydrotrametenolic acid and dehydroeburicoic acid in Poriae Cutis total triterpenoids tablets by liquid-solid compacts technique. Method: Taking dissolution of dehydrotrametenolic acid and dehydroeburicoic acid as indexes, difference of dissolution between liquid-solid compressed tablets and crude drug of total triterpenoids in Poriae Cutis was compared, crude drug, liquid-solid compacts tablets powder and excipients were characterized by differential scanning calorimetry (DSC). Result: Liquid-solid compacts technique could significantly improve dissolution of dehydrotrametenolic acid and dehydroeburicoic acid.Total dissolution of these two ingredients in liquid-solid compressed tablets was 92% at 120 min, total dissolution of them in crude drug was 29% at 120 min.DSC showed that characteristic peaks of drug in liquid-solid tablets had vanished, and suggested that drugs may be present in liquid-solid compressed tablets as amorphous substance. Conclusion: Liquid-solid compacts technique can increase dissolution of total triterpenoids in Poriae Cutis and allow rapid release of poorly soluble drugs.

      摘要:Objective: To prepare compound Ganning granules, optimize its extraction process and investigate its protective effect on acute hepatic injury induced by CCl4 in mice. Method: Ethanol extraction process of Schisandrae Chinensis Fructus was optimized by Taguchi design, water extraction process of the other herbs was optimized by orthogonal test.Compound Ganning granules was prepared through wet granulation process.Mice were randomly divided into six groups, included normal group, model group, bifendate group and high, middle, low dose group of compound Ganning granules.Acute hepatic injury mice model was induced by 0.1% CCl4.Levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) in serum and malonialdehyde (MDA), superoxide dismutase (SOD) in live tissue were detected. Result: After being optimized, transfer rates of schisandrin A and sodium danshensu were more than 90%.Compared with the normal group, activities of ALT, AST and level of MDA in model group were significantly increased and activity of SOD was inhibited.Compared with the model group, activities of ALT, AST and level of MDA in compound Ganning granules groups were significantly decreased and activity of SOD was increased. Conclusion: These optimized processes are stable and feasible.Compound Ganning granules has hepatoprotective effect on acute hepatic injury induced by CCl4 in mice by up-regulation of activity of SOD and down-regulation of activities of ALT, AST and MDA.

      摘要:Objective: To compare effect of different extraction technology (water extraction of whole prescription, water extract-alcohol precipitation of Gardeniae Fructus and alcohol extraction of Oroxyli Semen and Sophorae Flavescentis Radix, alcohol extraction of whole prescription, they were referred to as process 1, 2 and 3) on anti-inflammatory, analgesic and antipyretic action of Feiduqing. Method: Anti-inflammatory effect of Feiduqing with different extraction technology was compared by inflammation test induced by xylene in mice;through glacial acetic acid writhing test to compare analgesic effect of Feiduqing with different extraction technology;by 2, 4-dinitrophenol induced rat fever to compare antipyretic effect of Feiduqing with different extraction technology. Result: Sample of process 1 could significantly inhibit inflammation induced by xylene in mice and sample of process 2 could significantly inhibit inflammation induced by xylene in mice.Sample of process 1 could significantly inhibit times of writhing caused by acetic acid;samples of process 2 and 3 could significantly inhibit times of writhing caused by acetic acid;inhibition rate of sample of process 2 was higher than sample of process 3.Sample of process 2 could significantly inhibit fever induced by 2, 4-dinitrophenol. Conclusion: Extraction process has great impact on efficacy of Feiduqing;anti-inflammatory, analgesic and antipyretic effect of sample of process 2 is the best.

      摘要:Objective: To optimize extraction process of total polysaccharides in Bupleuri Radix and compare its content difference in different processed products. Method: Phenol-sulfuric acid method was employed to determine the content of total polysaccharides at 485 nm.Single factor test and orthogonal test was adopted to optimize extraction process of total polysaccharides in Bupleuri Radix. Result: Influence of various factors on extraction process was in the order of extraction temperature>extraction times>solid-liquid ratio>extraction time.The best extraction technology conditions was as follows:extracted twice with 50 times the amount of water under 100 ℃, 2 h for each time.Under these conditions, yield of soluble polysaccharides reached 19.39%, the content of total polysaccharides in different processed products of Bupleuri Radix was 9.82%-20.82%. Conclusion: This optimized extraction process is reasonable, reliable and suitable, which can provide theory basis for research and development of total polysaccharides in Bupleuri Radix.

      摘要:Objective: To study in vitro transdermal absorption characteristics of main ingredients in Qingfei Tongluo paste (QFTLP) and investigate effect of different compatibility on transdermal characteristics of main ingredients. Method: Taking main ingredients in QFTLP as indexes, such as aloe-emodin, emodin, rhein, studies were carried out by in vitro transdermal absorption test and HPLC.In vitro transdermal properties of QFTLP was investigated, cumulative transdermal amount per unit area and transdermal rate were evaluated. Result: Cumulative transdermal amount per unit area and transdermal rate of emodin was the largest, followed by aloe-emodin, and rhein was the least.Transdermal time-lag of rhein was the longest (0.419 h), emodin was the shortest (0.006 h).Penetration curve equations of emodin, aloe-emodin and rhein in QFTLP were Q=12.144t-14.880 (r=0.97), Q=10.111t-3.886 (r=0.93), Q=1.389t-0.583 (r=0.95).In overall prescription group, accumulation permeation amounts of emodin, aloe-emodin and rhein in 8 h were (86.059±0.168), (83.079+3.303), (12.819+0.569) μg · cm-2, transdermal rates were 12.144, 10.111, 1.389 μg · cm-2 · h-1, respectively;they had significant difference (P<0.05) by comparing with main formula group. Conclusion: In vitro percutaneous penetration of QFTLP complies with zero-order kinetics.Adjuvant compatibility can promote transdermal absorption of main ingredients.

      摘要:Objective: To select proper macroporous resin for separation and purification of total flavonol glycosides in Kemin Qidan prescription and to determine purification process parameters. Method: Taking adsorption ratio and elution rate of total flavonol glycosides and calycosin-7-O-β-D-glucoside as indexes, impacts of D101, AB-8 and S-8 macroporous resin on contents of total flavonol glycosides and calycosin-7-O-β-D-glucoside were investigated, single factor tests were adopted to select the concentration of sample liquid, sample loading quantity and the amount of 70% ethanol. Result: Optimum purification process was that condensing the sample solution to a relative density of 1.05 at 60 ℃, then centrifuging;centrifugal fluid was diluted to 0.5 g · mL-1, AB-8 type macroporous resin was washed by 1 BV of water, whose diameter and height ratio was 1: 6.The flow velocity of sample loading absorption and edulcoration was 2 BV · h-1 and sample loading quantity was that each gram resin matched with solution containing 1.5 times raw medicinal material.Eventually, eluting with 4 BV of 70% ethanol by flow velocity of 4 BV · h-1 and then collecting eluent.Transfer rates of total flavonol glycosides and calycosin-7-O-β-D-glucoside were 78.22% and 43.81%. Conclusion: This optimized purification process is credible, stable and applicable to mass production of Kemin Qidan prescription.

      摘要:Objective: To choose the best RNA extraction for Crataegi Fructus from different stage. Method: In this paper, the Trizol, the modified Trizol, the cetyltrie thylammnonium bromide (CTAB) and the modified CTAB methods were compared to extract high purity and integrity RNA from Crataegi Fructus at different developmental stage. And the quality of RNA was analyzed by electrophoresis detection and the spectrophotometer. Result: According to the result, high-grade RNA, with clear 18 S and 28 S, and the value of A260 nm/A280 nm was about 2.0, was isolated by using the modified CTAB method. Furthermore, the isolated RNAs from samples at different developmental stages were successfully used for the reverse transcription- polymerase chain reaction (RT-PCR) of the ACTIN in hawthorn. Conclusion: It suggested that the modified CTAB method was useful for isolating RNA from Crataegi Fructus at different developmental stage.

      摘要:Objective: Combining an HPLC fingerprint with bioassay to make a quality evaluation of Tongtian oral liquid. Method: Fingerprint of Tongtian oral liquid was established by the HPLC method, and quantitative analysis of ten batches of Tongtian oral liquid was performed.Anti-platelet aggregation activity as biological activity indicator was used to determine the contribution of the active ingredients in Tongtian oral liquid. Result: Similarity among ten batches was more than 0.96, nineteen common peaks were obtained in the fingerprint of ten batches of Tongtian oral liquid.Compared with reference substance, No.6 peak was paeoniflorin, No.7 was prim-O-glucosylcimifugin, No.8 was liquiritin, No.10 was 4'-O-β-D-glucosyl-5-O-meth-ylvisammioside and No.11 was senkyunolide Ⅰ.Quantitative analysis showed that five ingredients had great differences among the ten batches of Tongtian oral liquid.Meanwhile, the anti-platelet aggregation activity showed that senkyunolide Ⅰ>4'-O-β-D-glucosyl-5-O-meth-ylvisammioside>prim-O-glucosylcimifugin>paeoniflorin, and senkyunolide Ⅰ was its main active ingredient(P<0.001). Conclusion: Fingerprint of Tongtian oral liquid associated with its efficacy has been established.Senkyunolide I, 4'-O-β-D-glucosyl-5-O-meth-ylvisammioside and prim-O-glucosylcimifugin are the main chemical ingredients of Tongtian oral liquid related to the anti-platelet aggregation activity, which would provide a more reasonable means for the quality assessment of Tongtian oral liquid.

      摘要:Objective: To establish the UPLC fingerprint for Bletilae Rhizoma from different origins, and provide experimental basis for effective overall quality control of Bletilae Rhizoma. Method: Fingerprint of Bletilae Rhizoma was developed by using UPLC-PDA, and the Acquity UPLC BEH C18 column (2.1 mm ×150 mm, 1.7 μm) was used with acetonitrile-water solution as the mobile phase for gradient elution. The flow rate was 0.3 mL ·min-1; the column temperature was 45 ℃, and the detection wavelength was at 280 nm. Similarity evaluation, clustering analysis and principal components analysis (PCA) were used to evaluate the quality of Bletilae Rhizoma from different origins. Result: The 20 common peaks were marked in the fingerprint of Bletilae Rhizoma, and the chemical compounds of 9 common peaks were identified. The similarity of 43 batches of samples was 0.540-0.942, and they could be classified into two clusters by clustering analysis and four clusters by PCA. Six compounds in the fingerprint were found to cause differences. Conclusion: Establishment of UPLC fingerprint of Bletilae Rhizoma and the application of chemical pattern recognition can provide more comprehensive reference for the quality control of the herb.

      摘要:Objective: To study the chemical constituents from Oldenlandia diffusa . Method: The compounds were isolated and purified by column chromatography on silica gel, MCI gel, Sephadex LH-20 and RP-semi-preparative HPLC. The structures of compounds were elucidated on the basis of physicochemical and spectral analysis. Result: Six compounds were isolated from the water extract of O. diffusa, and their structures were identified as 2'-deoxythymidine (1), 2'-O-methyl-inosine (2), inosine (3), uridine (4), guanosine (5), β-adenosine (6), respectively. Conclusion: All the isolated compounds were obtained from the Hedyotis for the first time.

      摘要:Objective: To deeply study the major chemical compounds and their bioactivities of the underground part of Isodon flavida. Method: The samples were isolated and purified by using silica gel, Sephadex LH-20, and MCI chromatographic techniques. The structures of the monomer compositions were determined based on modern spectrum technology and physicochemical properties. In addition, the antioxidant and antimalarial activities of the samples were tested by using 1, 1-diphenyl-2-picrylhydrazyl radical 2, 2-diphenyl-1-(2, 4, 6-trinitrophenyl)hydrazyl(DPPH) method and β-hematin formation inhibition method. Result: Nine compounds were obtained from n-butanol fraction of 70% acetone extract of the underground part of I. flavida, and identified as 2, 6-dimethoxybenzoquinone (1), methyl rosmarinate (2), 2α-hydroxyoleanolic acid (3), 1-methoxy-α-D-glucose (4), 1-methoxy-β-D-glucose (5), sucrose (6), adenanthoside C (7), 5α, 6β-dihydroxydaucosterol (8), and (2'R)-1-O-glyceryl-6-O-α-D-galactopyran-osyl-β-D-galactopyranoside (9). Bioactive studies indicated that compound 2 showed certain clearing effect on DPPH and inhibitory effect on β-hematin formation, with IC50 values of (124.2±2.7), (163.9±7.5) mg · L-1, respectively. Conclusion: All of the 9 compounds were isolated from the plant for the first time. Compound 2 showed certain antioxidation and antimalaria effects, which deserved further investigation. This study provided certain experimental basis for the development and utilization of Isodon plants.

      摘要:Objective: To study the chemical constituents of dichloromethane extract from the leaf of Celastrus monospermus and to clarify the basis of its efficacy substance. Method: Methanol extracts from the leaf of C. monospermus were obtained by using dichloromethane, and purified by gradient elution on silica gel column chromatography with petroleum ether-ethyl acetate system. Their structures were identified by their physical and chemical properties, as well as the spectral data. Result: Eleven compounds were isolated and identified as monosperme A (1), syringic acid (2), syringaldazin (3), vanillic acid (4), β-sitosterol (5), daucosterol(6), usu-12-en-3-one-28-methyl (7), α, β-amyrin (8), 3-oxo-olean-12-en-29-oic acid (9), tripterfrielanon A (10), and 3α, 28-dihydroxy-olean-12-en-29-oic acid (11). Conclusion: The 11 compounds were obtained. Compound 1 was a new compound and other compounds were separated from the leaf of this plant for the first time.

      摘要:Objective: To identify chemical components in volatile oil from fermentation liquid of endophytic fungus Penicillium sp. B21 from Bruguiera sexangula and the activity of antibacterial of the volatile oil. Method: Volatile oil from fermentation liquid of endophytic fungus Penicillium sp. B21 was extracted by hydrodistillation. GC-MS was adopted to analyze chemical constituents in volatile oil. Also, the antibacterial activity of the volatile components were detected with enzyme standard method. Result: Yield of volatile oil was 1.82%, 39 kinds of compounds were identified which were accounted for 93.89%of total mass of volatile oil. Antibacterial activity tests indicated that, with 20 mg · L-1 concentration, volatile oil has good antibacterial activity against Escherichia coli and Staphylococcus albus. Conclusion: The volatile oil from fermentation liquid of Endophytic Fungus Penicillium sp. B21 from B. sexangula was investigated by antimicrobial activities. It extended the research of chemical medicine and food industry areas.

      摘要:Objective: There is a rich resource of Tephrosia in China. This paper is to investigate the chemical components from the genius of Tephrosia purpurea. Method: The chemical compounds of T. purpurea branches and leaves were isolated and extracted, and their structures were identified by using several chromatographic separation methods, modern spectrum analysis method and literature comparison. Result: Twelve isoflavones were obtained from the branches and leaves of T. purpurea and identified as follows: 12a-dehydro-6-hydroxysumatrol (1), elongatin (2), tephcalostan C (3), 7, 4'-dihydroxy-3', 5'-dimethoxyisoflavone (4), 4'-demethyltoxicarol isoflavone (5), tephcalostan B (6), 5, 7-di-O-prenylbiochanin A (7), tephcalostan D (8), tephcalostan (9), 6a, 12a-dehydro-2, 3, 6-trimethoxy-8- (3', 3'-dimethylallyl)-9, 11-dihydroxyrotenone (10), 3, 4, 8, 9-dimethylenedioxypterocarpan (11), 12a-hydroxy-β-toxicarol (12). Conclusion: All the other 11 compounds except compound 4 were obtained from this plant for the first time.

      摘要:Objective: To determine the contents of free amino acids and hydrolyzed amino acids from Ligustri Lucidi Fructus by pre-column derivatization RP-HPLC. Method: The samples were derived with phenylisothiocyanate (PITC) for pre-column derivatization and HPLC method was adopted. Gradient elution was applied to determine the contents of free amino acids and hydrolyzed amino acids in Ligustri Lucidi Fructus. Result: The 21 kinds of amino acids showed a good linearity in the ranges of 0.002-0.270 mmol ·L-1 with the correlation coefficients≥0.999 5.The average recovery rate was 88.33%-99.88% with relative standard deviation of 0.6%-2.5% for free amino acids, and 90.32%-99.91% with relative standard deviation of 0.5%-2.5% for hydrolyzed amino acids. Conclusion: This method is suitable for determining the contents of amino acids in Ligustri Lucidi Fructus.

      摘要:Objective: To establish a liquid chromatography-mass spectrometry-mass spectrometry (LC-MS-MS) method to study the effects of Phellodendri Cortex (CP) on the contents of four ingredients (mangiferin, neomangiferin, timosaponin AⅢ and timosaponin BⅡ) from Anemarrhenae Rhizoma (RA). Method: Chromatographic separation was carried out on ZORBAX Eclipse XDB-C18 column (2.1 mm×150 mm, 3.5 μm), with 0.1% formic acid-acetonitrile as the mobile phase for gradient elution. The column temperature was 30 ℃ and the flow rate was 0.25 mL ·min-1. The quantification of the analytes was performed by mass spectrometry with electrospray ionization (ESI) under negative ion multiple reaction monitoring (MRM) mode. Result: Mangiferin, neomangiferin, timosaponin AⅢ and timosaponin BⅡhad good linear relationship between 5.05-404, 5.3-424, 5.05-404, 20.8-1 664 μg ·L-1 respectively (r ≥0.999). The average recovery rate was 100.6%, 99.2%, 99.1% and 98.5%, respectively. The contents of mangiferin and timosaponin AⅢ were highest in group RA-CP (1: 0). Neomangiferin content in group RA-CP (1: 1) was higher than that in other groups. Timosaponin BⅡcontent was similar in each group. Conclusion: The method is rapid, accurate and reliable, and has been successfully used to the quantification of four compounds above mentioned in RA-CP. The contents mangiferin, neomangiferin and timosaponin AⅢ can be affected by the different RA-CP compatibility.

      摘要:Objective: To establish the HPLC Phorbols part in Crotonis Fructus oil, and offer the evidence for quality evaluation of Crotonis Fructus oil accordingly. Method: To measure ten batches of Crotonis Fructus by using RP-HPLC. The separation condition was:Thermo Hypersil Gold C18 chromatographic column (4.6 mm×250 mm, 5 μm), with water as mobile phase A, and acetonitrile as mobile phase B in gradient mode, and the flow rate is 1.0 mL ·min-1, column temperature was set at 30 ℃, detection wavelength was at 235 nm. Result: The common mode of the HPLC fingerprints were setup by ensuring twelve common peaks in the chromatograms. The similar degrees to the ten batches were between 0.884-0.991. Conclusion: The method is simple, accurate and have a good repeatability. And the quality of Crotonis Fructus oil can be controlled effectively by the HPLC fingerprint.

      摘要:Objective: To isolate and identify the chemical compounds from the residues in the pulps of Persea americana, and evaluate the capacity of scavenging free radicals of phenolic compounds. Method: The chemical compounds were isolated and purified by the combined normal pressure column chromatography and high pressure liquid preparative chromatography, and their chemical structures were elucidated by spectral data analysis and comparing with the previous literature. Moreover, the capacity of scavenging free radicals of phenolic compounds was also evaluated by using 1, 1 diphenyl - 2 - trinitrobenzene hydrazine (DPPH ·) assay. Result: Seven compounds were obtained and their structures were identified as 1, 2, 4-trihydroxyheptadec-16-yne (1), 1, 2, 4-trihydroxynonadecane (2), (2R, 12Z, 15Z)-2-hydroxy-4-oxoheneicosa-12, 15-dien-1-yl acetate (3), tryptophan (4), chlorogenic acid (5), neochlorogenic acid (6), and scopolin (7) respectively. The in vitro free radicals scavenging activity experiment showed that the capacity of scavenging free radicals of compounds 5 and 6 were close to that of vitamin C. Conclusion: Compounds 4 , 6 and 7 were isolated from P.americana for the first time. In addition, phenolic compounds obtained from P. americana had good in vitro free radicals scavenging activities.

      摘要:Objective: To establish a method for simultaneously determining caffeic acid, quercetin-3-O-glucoside, kaempferol-3-O-glucoside and kaempferol in Geum aleppicum by RP-HPLC. Method: The quantitative analysis of G. aleppicum was carried out on Agilent C18 column(4.6 mm×250 mm, 5 μm), mobile phase was acetonitrile-0.5% formic acid with gradient elution, flow rate was 0.8 mL ·min-1, columntemperature was maintained at 25 ℃, and detection wavelengrt was 340 nm. Result: The linear relationships of caffeic acid, quercetin-3-O-glucoside, kaempferol-3-O-glucoside and kaempferol fell within the ranges of 0.045-1.130, 0.095-2.370, 0.105-2.635, 0.041-1.030 μg, respectively. Their average recoveries (n=6) were 99.0%, 98.5%, 97.7%, 100.7% with the relative standard deviations (RSDs) of 1.0%, 0.9%, 1.0%, 1.6%, respectively. Conclusion: This method is simple, accurate, sensitive, which can be used for quality control of G. aleppicum.

      摘要:Objective: To develop a method for determining four kinds of saponins in Shenqi compound granules. Method: Quantitative analysis was carried out on a column of Phenomenex C18 (4.6 mm×250 mm, 5 μm) by HPLC-ELSD with mobile phase of acetonitrile-water under a flow rate of 0.8 mL · min-1, column temperature was 25 ℃, flow rate of carrier gas was 2.6 L · min-1, drift tube temperature was 102 ℃. Result: Linear ranges of ginsenoside Rg1, ginsenoside Re, ginsenoside Rb1and astragaloside Ⅳ were 0.196-2.976, 0.207-3.138, 0.191-2.895, 0.190-2.879 μg;their recoveries were 99.27%, 99.33%, 99.32%, 99.27%, RSDs were 1.5%, 1.1%, 1.0% and 2.4%, respectively. Conclusion: This method is accurate, reproducible and convenient for quality control over Shenqi compound granules.

      摘要:Objective: To observe the effects of Fuzheng Kang'ai (FZKA) decoction on apoptosis of H1650 cells and discuss its molecular mechanism in apoptosis of H1650 cells. Method: Human lung adenocarcinoma cells (H1650 cells) were used as the research objects and treated for 24, 48, 72 h with 0.5, 1.0, 1.5, 2.0, 2.5, 3.0 g · L-1 FZKA decoction, and a blank group was set up, then cell proliferation was detected by 3- (4, 5-diethylthiazol-2-yl)-5- (3-carboxymethoxyphenyl)-2- (4-sulfophenyl)-2H-etrazolium, inner salt (MTS) assay. H1650 cells were treated for 24 h with 0.5, 1.0, 1.5 g · L-1 FZKA decoction and a blank group was set up, then cell apoptosis was detected by Annexin V-FITC/PI flow cytometry analysis. H1650 cells were treated for 24 h with 0.5, 1.0, 2.0 g · L-1 FZKA decoction and a blank group was set up, then vitality of Caspase-3/7 was detected by Caspase-3/7 vitality test kit. Simultaneously, effects of FZKA decoction on the expression levels of proCasapse-3, PARP and Bax were detected by Western blot assay. Result: As compared with the blank group, proliferation of H1650 cells was significantly inhibited by FZKA decoction in concentration-dependent and time-dependent manners (P<0.05). As compared with the blank group, early apoptosis of H1650 cells was significantly induced and vitality of Caspase-3/7 was increased by FZKA decoction in a concentration-dependent manner (P<0.05). As compared with the blank group, the expression levels of proCasapse-3 and PARP were significantly reduced by FZKA decoction in a concentration-dependent manner (P<0.05), and the expression of Bax was increased by FZKA decoction in a time-dependent manner (P<0.05). Conclusion: Apoptosis is induced by FZKA decoction in way of activating Caspase-3 and Bax in H1650 cells.

      摘要:Objective: To establish an in vitro blood-brain barrier, in order to study the effects of Buyang Huanwu Tang on blood-brain barrier function and associated protein expression under the condition of oxygen and glucose deprivation-reperfusion injury. Method: The in vitro oxygen and glucose deprivation-reperfusion injury model was established by culturing the isolated primary rat brain microvascular endothelial cells. The cells were divided into three groups, namely blank group, model group and Buyang Huanwu Tang group, and then were processed by following steps. Cell counting kit(CCK-8) method was used to detect cell activity; Transwell assay was used to detect the permeation of bovine serum albumin(BSA); Western blot assay was used to detect the protein expressions of Occludin, ZO-1.Real-time fluorescent quantitative PCR(qPCR) was used to detect the mRNA expressions of Occludin and ZO-1. Result: Compared with the blank group, the model group showed significant decreases in cell activity, Occludin and ZO-1 protein and their mRNA expressions, and increase in BSA permeation rate(P<0.05). In Buyang Huanwu Tang group, cell survival rate was significantly increased; bovine serum albumin (BSA) transmittance reduced; Occludin, ZO-1 protein and mRNA expression increased(P<0.05). Conclusion: Buyang Huanwu Tang can reduce the oxygen glucose deprivation-reperfusion damage in the blood-brain barrier model in vitro and up-regulate ZO-1, Occludin protein and mRNA expression.

      摘要:Objective: To study the active systemic anaphylaxis (ASA) and relevant indicators induced by Shuanghuanglian and Shengmai injection in Hartley guinea pigs and Brown Norway(BN) rats. Method: Hartley guinea pigs or BN rats were randomly divided into ovum albumin (OVA) group, Shuanghuanglian injection group, Shengmai injection group and normal group. Animals were injected with test sensitization drug liquid 3 times every other day. On day 14 of sensitization, corresponding arousal liquid was intravenously injected. The usual systemic symptoms of ASA test were observed within 30 min after the last injection. The lung tissue pathological examination was performed with hematoxylin-eosin(HE) staining. Changes of serum immunoglobulin E(IgE) levels before sensitization and after arousal, and changes of serum tryptase after sensitization and arousal were determined using radioimmunoassay in BN rats. Result: Hartley guinea pigs or BN rats in OVA group and Shuanghuanglian injection group showed systemic allergy symptoms. The pathological changes of the lung tissue with perivascular lymphocyte infiltration were observed, but the symptoms in Shuanghuanglian group were less serious. Shengmai group had no anaphylaxis or pathological abnormality of lung tissues. Compared with the normal group, the levels of serum IgE were significantly increased in OVA group, Shuanghuanglian group and Shengmai group after arousal (P<0.05, P<0.01). Compared with the animals of the same group before arousal, the total serum IgE levels after arousal were significantly increased both in OVA group and Shuanghuanglian group (P<0.05, P<0.01). Compared with the normal group, the serum tryptase levels in OVA group and Shuanghuanlian group were significantly increased after arousal (P<0.01, P<0.05), and Shengmai group had an increase trend but with no significant difference. Conclusion: Application of different animal species and increasing relevant detection indicators are useful to more comprehensively evaluate the potential ASA induced by Shuanghuanglian and Shengmai injection.

      摘要:Objective: To observe the effect of Zhuanggu Shenjin capsules (ZSJ) on bone metabolism and bone mineral density (BMD) in ovariectmized rats. Method: Totally 72 SD rats were randomly divided into sham-operated group (n=12) and modeling group (n=60). Bilateral ovaries of rats were removed to establish the modeling group, and then the rats in modeling group were further ransomly divided into model group, positive drug+estrogen group, and ZSJ high dose, middle dose and low dose groups (5.42, 2.71, 1.36 g · kg-1). Each group included 12 rats. The rats received corresponding drug by ig administration for continuous 13 weeks, 6 days/week. After the experiment, blood and bone tissues were taken to detect BMD, blood calcium (Ca), alkaline phosphatase (ALP), tartrate resistant acid phosphatase (TRAP), estradiol (E2), bone gla protein (BGP) and calcitonin in blood serum. The bone tissue pathology was observed by toluidine blue staining. Result: As compared with sham-operated group, BMD level and serum E2 level were significantly decreased, while serum ALP, TRAP and BGP levels were significantly increased in rats of model group (P<0.05, P<0.01), uterine coefficient was decreased significantly (P<0.01), the bone tissue pathological observation showed that the trabecular bone was sparse in rats of model group, with fractures and separation between the trabecular bones, gaps enlarged with arrangement disorder, loose bone lamellar structure, and the characteristic of osteoporosis was obvious. As compared with the model group, ZSJ (high dose, middle dose) could significantly increase the BMD level of the rats (P<0.01), increase the level of E2 in blood serum (P<0.05), and reduce ALP, TRAP and BGP levels (P<0.05), but had no significant effect on the level of blood Ca and CT, the bone tissue pathological observation showed that the bone tissue structure was nearly complete. Conclusion: ZSJ can increase bone mineral density and E2 levels, and improve bone metabolism, thus preventing and treating osteoporosis in rats with ovariectomy.

      摘要:Objective: To investigate the antiproliferation effect of compound Banzhilian on COLO205 human colon cancer cells in vitro and in vivo. Method: COLO205 cells were cultured in vitro and divided into blank group, cisplatin group (0.15 g · L-1), and compound Banzhilian high dose group, middle dose group and low dose group (0.60, 0.30, 0.15 g · L-1). The effect of rate of compound Banzhilian on antiproliferation rate of COLO205 cells was detected by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Western blot assay was used to detect the expression levels of apoptosis-related proteins including Bcl-2, Bax, and p53 up-regulated modulator of apoptosis (PUMA). SPF nude mice were used for in vivo experiment, and they were inoculated with human colon cancer cells COLO205 in their back skin. Tumors size would be measured after they could be seen. According to the volume of tumors, nude mice were randomly divided into compound Banzhilian high and low dose groups, paclitaxel group and blank group. The effect of compound Banzhilian in vivo growth of COLO205 cells was detected with using colon cancer transplantation tumor models in nude mice. Result: In the in vitro experiment, after being treated with compound Banzhilian, most COLO205 cells were deformed with damages of cell membrane, the number of cells decreased gradually, and eventually died. Statistical analysis showed that the antiproliferation effect on COLO205 cells was at 48 h treatment with compound Banzhilian. The inhibition rates were 68.46%, 63.11%, and 54.91% respectively in compound Banzhilian high, middle and low dose groups, the expression levels of Bax and PUMA were up-regulated in COLO205 cells treated with compound Banzhilian, while the expression level of Bcl-2 was down-regulated. The in vivo transplantation tumor experiment showed that tumor volume in treatment groups was significantly smaller than that of in blank group. Conclusion: Compound Banzhilian can inhibit COLO205 colon cancer cells proliferation both in vitro and in vivo, and its potential mechanism may be related to the induction of apoptosis.

      摘要:Objective: To investigate the effect of allicin on epithelial-mesenchymal transition (EMT) of pancreatic cancer cells. Method: The PANC-1 cells were cultured and treated with 10 μg · L-1 transforming growth factor-β1 (TGF-β1), 10 μg · L-1 TGF-β1+5 μg · L-1 allicin, 5 μg · L-1 allicin, respectively. Untreated cells were used for the normal control. Morphological changes were observed under inverted microscope. Wound scratch assay were performed to further identify if allicin could reduce the migration ability of PANC-1 cells. The expressions of E-cadherin and Vimentin in pancreatic cancer cells were detected by immunocytochemistry and Western blot. Result: The TGF-β1 can make PANC-1 cells show typical EMT morphological changes, which were accompanied by up-regulation of E-cadherin expression and down-regulation of Vimentin. Wound scratch assay indicated that allicin could reduce the migration ability of PANC-1 cells. According to the results of immunocytochemistry and Western blot, compared with the control group and the TGF-β1 group, the protein expression of E-cadherin was up-regulated in the allicin group (P<0.05), and that of aimentin was down-regulated (P<0.05). Conclusion: Allicin can inhibit the EMT of human pancreatic cancer PANC-1 cells, and may play an important role in the inhibition of tumor invasion and metastasis.

      摘要:Objective: To study Guizhou Douchi fibrinolytic kinase DCK(DCK enzyme)thrombolysis and antithrombotic function in vivo. Method: Male Kunming mice were randomly divided into model group, urokinase control group(150 U · g-1), DCK enzyme low, medium and high dose group (150, 300, 450 U · g-1), ip 7 d, once a day, each group were injected 0.8% carrageenan to build the thrombus model in tail after 30 minutes 7 days administration. Tail thrombosis was observed and the mice tail length and the length of thrombus were measured at 24, 36 h. Thrombus formation rate and thrombosis relative length were calculated. Wistar rats were randomly divided into model group, urokinase control group(100 U · g-1)and DCK enzyme low, medium and high dose group(100, 200, 300 U · g-1), ip 10 days, once a day. Rats were anesthetized by chloral hydrate solution after 30 minutes the 10 days treatment, the right common carotid artery and left external jugular vein were separated to build the right common carotid artery-left external jugular vein bypass. Thrombus wet weight were get in the bypass circulation. New Zealand white rabbits were randomly divided into model group, urokinase control group (38 U · g-1), DCK enzyme low, medium and high dose group (38, 76, 114 U · g-1)by ear vein injection administration, blood sampling of ear border vein were administered after 1 h to detect prothrombin time(PT), activated partial thromboplastin time(APTT), thrombin time(TT), human fibrinogen(FIB), euglobulin clot lysis time(ELT)and fibrin(-ogen)degradation products(FDP). Result: Compared with the model group, DCK enzyme group at 24, 36 h tail thrombosis formation relative length were reduced(P<0.01, P<0.05), DCK enzyme of high, middle dose group of bypass thrombus wet weight were reduced(P<0.01, P< 0.05). The PT, APTT, TT time extended with significant difference (P<0.01, P< 0.05), the FIB of high DCK enzyme group was reduced (P<0.05, P<0.01), high DCK enzyme and middle dose group of ELT was decreased(P< 0.05), and DCK enzyme in each experimental group FDP content were increased, the middle dose group increased(P<0.05). Conclusion: DCK enzyme has a certain thrombolysis and antithrombotic function in vivo.

      摘要:Objective: To investigate the intervention effect of Jinxin oral liquid (JOL) on the expression of suppressor of cytokine signaling1/3 (SOCS1/3) protein in lung tissues of respiratory syncytial virus(RSV) infected rats, and the possible mechanism of anti-RSV infection. Method: Totally 70 SD rats were randomly divided into the normal group, the infection model group, JOL low, medium and high dose groups (3.3, 9.9, 29.7 g · kg-1), the prevention group (given JOL 2 d in advance, ig), the ribavirin group (24.5 g · kg-1), with 10 rats in each group. Except for the normal group, the remaining groups were intranasally infected for 24 h, and treatment groups were give drugs for consecutively three days. The enzyme-linked immunosorbent assay (ELISA) was adopted to detect contents of type I-interferons IFN-α and IFN-β in RSV infected rats in vivo. The reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the changes in SOCS1 and SOCS3 mRNA expressions in lung tissues of RSV infected rats. The hematoxylin eosin (HE) staining was applied to observe pathological changes in rat lung tissues. Result: Compared with the normal group, the model group showed significant increases in IFN-α and IFN-β contents in RSV infected rats, and SOCS1 and SOCS3 mRNA expressions in lung tissues of RSV infected rats (P<0.05, P<0.01), and the model group showed more obvious pathological changes in rats lung tissues. The IFN-α expressions of prevention group and JOL middle dose group were higher than model group (P<0.05), the IFN-β expressions of prevention group, JOL middle dose group and ribavirin group were higher than model group (P<0.01); The IFN-β expression of JOL high dose group was higher than model group(P<0.05), the SOCS1 expressions of JOL middle dose group and JOL high dose group were lower than model group(P<0.05), the SOCS3 expressions of JOL middle dose group and ribavirin group were lower than model group(P<0.05) and the SOCS3 expression of JOL middle dose group was lower than model group (P<0.01). Conclusion: JOL can up-regulate the expression of I-IFN in RSV infected rats and down-regulate the expression of SOCS1/3, thus inhibiting virus infection. The SOCS1/3 may be one of JOL's mechanisms in anti-RSV infection.

      摘要:Objective: To investigate the effect of Jianpi Wenwei powder on the expression of serum tumour necrosis factor-α(TNF-α), interleukin-6(IL-6) and interleukin-1 receptor-associated kinase 4(IRAK-4) mRNA in the gastric mucosa of spleen-deficiency rats. Method: Totally 60 Wistar rats were randomly divided into normal control group, gastritis model group, positive drug control group (Colloidal Bismuth Subcitrate capsules, 0.016 g · kg-1 · d-1) and Jianpi Wenwei powder high dose group, middle dose group, low dose group (0.324, 0.259, 0.194 g · kg-1 · d-1). Except for the normal control group, all of the remaining groups were included in the spleen deficiency animal model of gastritis. The changes of anus temperature were measured in rats. At 30 d after the treatment for each group, IL-6, TNF-α in serum were determined by radioimmunoassay, histopathological changes in gastric tissues were observed by htoxylin eosin(HE) staining; reverse transcription polymerase chain reaction(RT-PCR) method was used to observe the effect of Jianpi Wenwei powder on the expression of IRAK-4 mRNA in gastric mucosa of chronic gastritis model rats. Result: Compared with the normal group, the model group showed significant decrease in the rectal temperature changes, and increases in serum IL-6, TNF-α and IRAK-4 mRNA expression in gastric tissues (P<0.01), with obvious pathological injury at gastric mucosa of rats; Compared with the model group, Jianpi Wenwei Powder can significantly alleviate the syndrome characteristics and rectal temperature changes in model rats, release the pathological morphology of gastric mucosa, significantly decrease serum IL-6, TNF-α in rats of gastritis model (P<0.01), down-regulate the IRAK-4 mRNA expression in the gastric mucosa (P<0.01). Conclusion: Jianpi Wenwei powder can recover the anus temperature of model rats to normal, alleviate the pathological morphology of gastric mucosa, reduce the content of serum IL-6 and TNF-α in the rat model of spleen deficiency chronic gastritis, and inhibit the expression of IRAK-4, which may be one of its mechanisms for treatment of gastritis.

      摘要:Objective: To observe the effects of Yougui capsule on osteoporosis in ovariectomized rats. Method: A total of 105 SPF-grade SD rats were selected, in which 15 were used in the sham-operated group, and the remaining were ovariectomized to reproduce the model of osteoporosis. The ovariectomized rats were divided into model group, Yougui capsule low, medium and high-dose groups (0.71, 1.43, 2.85 g · kg-1), estrogen group (diethylstilbestrol, 0.02 mg · kg-1) and Xianling Gubao capsule group (0.25 g · kg-1), with 15 rats in each group. The rats were given the corresponding drugs on the 7th day after surgery for six months. Orbital blood in rats was collected to determine Ca, P, alkaline phosphatase(ALP), acid phosphatase(ACP) in serum. Right and left thighbones of the rats were collected to determine bone mineral density(BMD), bone mineral(BMC) with dual energy X-ray absorptiometry. WDW electronic universal testing machine was used to determine mechanical performance indexes of the right thighbone, such as deflection, maximum force and bending strength. Result: Compared with the sham-operated group, the model group showed significant increases in Ca, P, ALP, ACP levels in serum, and decreases in BMD, BMC and bone biomechanical indexes(P<0.05, P<0.01); compared with the model group, Yougui capsule groups can significantly decrease Ca, P, ALP, ACP level in serum, and increase BMD, BMC and bone biomechanical indexes(P<0.05, P<0.01). Conclusion: Yougui capsule can improve biomechanical indexes of ovariectomized rats, and delay the rat bone mass loss, suggesting that Yougui capsule has the preventive effect on osteoporosis.

      摘要:Objective: To explore Arisaematis Rhizoma polysaccharide (ARPS) on the proliferation, apoptosis and Wnt/β-catenin signaling pathway of human renal cell carcinoma cell line GRC-1. Method: After entering into the logarithmic growth phase with RPMI-1640 culture fluid under routine condition, the GRC-1 cells were treated with different doses of ARPS (0, 20, 50, 100, 200 mg · L-1). The 0 mg · L-1 ARPS group was the blank group. The inhibition rates of proliferation at 24, 48, 72 and 96 h after the treatment were measured by viable cell counting kit assay (CCK-8). The apoptosis rate and cell cycle at 48 h after the treatment were detected by Annexin-FITC/PI double staining and PI staining methods via flow cytometry, respectively. Western blot was employed to detect the expression level of β-catenin in Wnt/β-catenin signaling pathway and its downstream target molecule C-myc and Cyclin D1 at 48 h after the treatment. Result: Compared with the blank group, ARPS within the range of 20-200 mg · L-1 presented an inhibitory effect on GRC-1 cell proliferation. The proliferation inhibition rate increased with the rise in time and concentration of ARPS in a dose-and time-dependent manner, and the above differences were statistically significant (P<0.05). Compared with the 0 μg · mL-1 ARPS, the early apoptosis rate of 50-200 mg · L-1 ARPS, the late apoptosis rate of 20-200 mg · L-1 and the total apoptosis rate were all elevated(P<0.05). There was a higher G0/G1 phase ratio than 20, 50, 100, 200 mg · L-1 ARPS groups, and lower S and G2/M phase and β-catenin, C-myc and Cyclin D1 protein levels than 0 mg · L-1 (P<0.05). Indexes within the concentration range between 20-200 mg · L-1 were statistically significant(P<0.05). Conclusion: ARPS inhibited the proliferation of renal cell carcinoma cell line GRC-1, and induced cell apoptosis and G0/G1 arrest, with the effect of inhibiting the activation of Wnt/β-catenin signaling.

      摘要:Objective: To investigate the effects of Eucommiae Cortex polysaccharide on streptozotocin-induced diabetic mice. Method: Fifty SPF Kunming mice were intraperitoneally injected with 120 mg · kg-1 streptozotocin(STZ) after being fasted over 12 hours. After 3 days, the fasting blood glucose(FBG) of mice were measured to be ≥11.1 mol · L-1, indicating the successful modeling. The diabetic mice were randomly divided into 5 groups, with 10 mice per group: model group, metformin group (32 mg · kg-1), and high, medium and low-dose Eucommiae Cortex polysaccharide groups (500, 250, 125 mg · kg-1). Another 10 healthy mice were considered as normal group. These mice were administrated intragastrically once a day for 14 days. Normal and model groups were given equal volume of saline. One hour after the last administration, the blood samples of 12-hour fasted mice were collected by drawing eyeballs and were centrifuged to separate serum for further measurement. The FBG were detected at the 14th day; the serum fasting insulin (FNS) was measured using radioimmunoassay and then the insulin sensitivity index(ISI) was calculated. Serum triglyceride(TG), total cholesterol(TC), high density lipoprotein cholesterol(HDL-C) and low density lipoprotein cholesterin(LDL-C) were analyzed using commercial kits. enzyme-linked immuno sorbent assay(ELISA) method was utilized to detect TNF-α, IL-8 and interleukin-6(IL-6) levels. The protein levels of Toll-like receptor 4(TLR4) and nuclear transcriptional factor-κB(NF-κB) in pancreas tissues were analyzed by Western blot. Result: Compared with the normal group, the model group showed significant increases in FBG, lipid TG, TC, LDL-C, serum TNF-α, IL-8 and IL-6 contents, and protein expressions of TLR4 and NF-κB in pancreas, and significant decreases in HDL-C, FNS and ISI (P<0.01); Compared with the model group, Eucommiae Cortex polysaccharide groups showed significant decreases in FBG, lipid TG, TC, LDL-C, serum TNF-α, IL-8 and IL-6 contents, and protein expressions of TLR4 and NF-κB in pancreas, and significant increases in HDL-C, FNS and ISI (P<0.05, P<0.01). Conclusion: The mechanism of Eucommiae Cortex polysaccharide on hypoglycemia and hypolipidemic may associate with improving antioxidant capacity, decreasing serum contents of inflammatory cytokines and inhibiting the expressions of TLR4, NF-κB proteins in diabetic mice.

      摘要:Objective: To investigate the effects of Tongyang Kuanxiong granule on coronary microcirculation improvement in the rat with coronary arteriosclerosis, and to explore the efficacy of the combined treatment of traditional Chinese and Western medicine for cardiovascular diseases. Method: Totally 50 Wistar rats were randomly divided into normal group, model group, Tongyang Kuanxiong granule group (50 mg · kg-1 · d-1), Simvastatin group (5 mg · kg-1 · d-1), and the mix treatment group (TY+simvastin). All the rats except those in normal group were used to prepare arteriosclerosis models. The rats in normal group were given with normal diet, and the rats in model group and treatment groups were given with high-fat diet and Vitamin D3 by ip. After 4 weeks of such feeding, the rats were treated for 8 weeks by intragastric administration. The expression levels of total cholesterol (TC), triglyceride (TG) and low density lipoprotein (LDL-C) in serum were detected by enzyme-linked immunosorbent assay (ELISA). Artery plaque area was analyzed. Aorta was taken for immunohistochemistry and Western blot to detect the expression levels of interleukin (IL)-17A, vascular cell adhesion molecule-1(VCAM)-1 and intercelluar adhesion molecule-1 (ICAM)-1. Result: As compared with the normal group, the serum artery plaque area, expression levels of TC, TG, LDL-C were significantly increased in rats of model group(P<0.05). As compared with the model group, the expression levels of TC, TG, LDL-C levels, and artery plaque area were significantly reduced in treatment groups (P<0.05, P<0.01). As compared with Tongyang Kuanxiong granule group, the improvement effect was more obvious in mixed treatment group (P<0.05). As compared with model group, the expression levels IL-17A, VCAM-1 and ICAM-1 in coronary atherosclerosis tissues were decreased in treatment groups(P<0.05), and the efficacy of mixed treatment group was better than that of Tongyang Kuanxiong granule alone group. Conclusion: Tongyang Kuanxiong granules can relieve tissue inflammation and treat coronary atherosclerotic diseases by improving blood lipid indexes, reducing the expression levels of IL-17A, VCAM-1, ICAM-1, and reducing artery plaque area, so the integrated Chinese and western medicine treatment is worthy for further promotion.

      摘要:Objective: To investigate the possible toxic reactions of Artemisinin and Piperaquine tablets in rhesus monkeys by oral administration, and evaluate its safety. Method: Approximate lethal dose method was used for the experiment. Artemisinin and Piperaquine tablet was given to 5 rhesus monkeys at the doses of 168, 377, 849, 1 910 , 4 296 mg · kg-1 respectively. Animals were observed with a period of 14 days for the toxic reactions and death. Their body weight, food intake, haematology and blood biochemistry indexes were measured, and gross pathological observation was conducted after anatomy in all rhesus monkeys, in order to calculate the range of approximate lethal dose. Result: No death appeared during 14 days observation. Vomiting and food intake reduction appeared in doses of 4 296, 1 910 mg · kg-1, and decrease in body weight was observed in monkey at the dose of 4 296 mg · kg-1. Conclusion: Approximate lethal dose of Artemisinin and Piperaquine tablets is greater than 4 296 mg · kg-1 (equivalent to 257 times of clinical dose) in rhesus monkeys by oral administration.

      摘要:Objective: To investigate the clinical efficacy of Bushen Kangshuai (BSKS) tablets for unstable angina pectoris (UAP) and observe its effect on flammatory mediator levels in serum. Method: Sixty UAP patients were randomly assigned to treatment group (30 cases) and control group (30 cases). Patients in control group received routine western medical treatment, while those in treatment group additionally took BSKS tablets. The angina symptom scores, clinical efficacy, angina frequencies, and Global Registry of Acute Coronary Events (GRACE) risk scores were compared between two groups both before and after treatment. The changes in interferon-γ(IFN-γ), interleukin (IL)-2, tumor necrosis factor-α(TNF-α), and IL-4 levels in serum were detected in both groups by enzyme-linked immunosorbent assay. Result: The angina symptom scores and angina frequency and duration were improved in both groups after treatment (P<0.05, P<0.01), and the flammatory mediator levels in serum were significantly reduced in both groups (P<0.05), the clinical efficacy in treatment group was significantly superior to that in control group (P<0.05). GRACE risk scores were highly correlated with the kidney deficiency syndrome scores of patients. Conclusion: Bushen Kangshuai tablet combined with routine western medicine treatment is an effective way to improve the clinical symptoms and lower the levels of inflammatory mediator in patients with UAP.

      摘要:Objective: To observe the effect of Jiawei Sier decoction for urinary albumin (UAL), serum superoxide dismutase (SOD), and malondialdehyde (MDA) levels in early type 2 diabetic nephropathy patients, and investigate its clinical efficacy. Method: The 90 cases of early type 2 diabetic nephropathy with traditional Chinese medicine(TCM) syndrome differentiation of Qi and Yin deficiency were randomly divided into Jiawei Sier decoction group (30 cases), gingko dipyridolum injection group (30 cases), and conventional treatment group (30 cases) according to random number table method. The patients in all 3 groups received conventional therapy for type 2 diabetes, based on the above conventional therapy, patients in modified Jiawei Sier decoction group also received Jiawei Sier decoction treatment combined with captopril therapy, patients in gingko dipyridolum injection group also received ginkgo dipyridolum injection combined with captopril therapy. All patients were treated for 4 weeks. UAL, SOD and MDA levels were detected, and the TCM symptom scores were observed both before and after treatment. Result: UAL and MDA levels in Jiawei Sier decoction group and gingko dipyridolum injection group after treatment were significantly lower than those before treatment (P<0.01), SOD level was significantly higher than that before treatment (P<0.01), and TCM symptom scores were significantly lower than those before treatment (P<0.01). As compared with conventional treatment group, UAL and MDA levels in Jiawei Sier decoction group and gingko dipyridolum injection group were significantly lower, while SOD level was higher after treatment (P<0.01), the TCM symptom scores were significantly lower in Jiawei Sier decoction group and gingko dipyridolum injection group (P<0.01). Conclusion: Jiawei Sier decoction treatment can obviously improve the clinical symptoms, reduce UAL and MDA levels and increase SOD level in serum for patients with early type 2 diabetic nephropathy, this may be related to its resistance to oxidative stress.

      摘要:Objective: To observe the effect of Yiqi Huoxue Huatan decoction on traditional Chinese medicine (TCM) syndrome scores, blood lipid indicators [total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C)], D-dimer (D-Di), C-reactive protein (CRP), intra media thickness (IMT), ankle brachial index (ABI) and brachial-ankle pulse wave velocity (baPWV) in aged patients with lower extremity atherosclerotic disease (LEAD). Method: The 66 eligible patients were randomly divided into treatment group (33 cases) and control group (33 cases). Patients in control group were treated with simvastatin tablets (40 mg, qd) and asprin tables (100 mg, qd), patients in treatment group additionally received Yiqi Huoxue Huatan decoction based on the treatment in control group. Patients were followed up for 12 weeks to observe the changes in TCM symptom scores, blood lipid indicators, D-Di, CRP, IMT, ABI and baPWV before and after treatment. In addition, the hepatic and renal functions were monitored and three routine tests (blood, urine and stool) were conducted. Result: TCM symptom scores in both groups were lower than those before treatment (P<0.05), and TCM symptom scores in treatment group were lower than those in control group after treatment (P<0.05). LDL-C, D-Di and baPWV levels in both groups were significantly lower than those before treatment (P<0.05), and the above levels in treatment group were lower than those in control group after treatment (P<0.05). Conclusion: Yiqi Huoxue Huatan decoction combined with simvastatin and asprin can effectively improve the TCM symptom scores, and reduce LDL-C, D-Di and baPWV levels in aged patients with lower extremity atherosclerotic disease (LEAD), and it may be helpful to improve arterial stiffness.

      摘要:Objective: To investigate the effect of Bushen Yizhi decoction combined with butylphthalide on glutathione peroxidase (GSH-Px), amyloid β-protein (Aβ), superoxide dismutase (SOD) and malondialdehyde (MDA) levels in patients with Alzheimer's disease. Method: The 74 patients with Alzheimer's disease were randomly divided into treatment group (37 cases) and control group (37 cases) according to the random number table method. Patients in control group were treated with oral butylphthalide soft capsule, and patients in treatment group also received Bushen Yizhi decoction based on the treatment in control group, with 12 weeks as a course of treatment. After treatment, minimum mental state examination (MMSE) scale scores, activity of daily living (ADL) scale scores, Aβ, SOD, GSH-Px and MDA levels were compared and analyzed between two groups of patients with Alzheimer's disease. Result: After treatment, the MMSE and ADL scores of both groups were significantly improved (P<0.05), the degree of improvement in treatment group was more obvious (P<0.05). After treatment, the levels of Aβ in both groups were significantly decreased (P<0.05), the extent of the reduction in the treatment group was more obvious (P<0.05), SOD and GSH-Px levels in both groups were significantly increased after treatment (P<0.05), and the extent of increase in the treatment group was more obvious (P<0.05). The level of MDA in the treatment group was significantly decreased after treatment, and it was lower than that of the control group after treatment (P<0.05). Conclusion: Bushen Yizhi decoction combined with butylphthalide can improve the cognition impairment and the function of the body's resistance to oxidative stress, thus significantly improving the self-care ability and their ability of learning and memory for the patients, so it is worthy of further promotion.

      摘要:Objective: To investigate the short-term and long-term effects of Jiaogulan Kangai decoction combined with dialectical synchronous chemoradiotherapy and maintenance treatment in treating middle and advanced stage non-small-cell lung cancer (NSCLC). Method: One hundred and seven patients with middle and advanced stage NSCLC were randomly divided into control group (52 cases) and observation group (55 cases) according to their admission sequence. Patients in control group received docetaxel plus cisplatin (TP) method. Based on the treatment of control group, patients in observation group also received dialectical Jiaogulan Kangai decoction, 1 dose/day. The observation was stopped until progressive disease appeared. Before and one month after chemotherapy, size of solid tumor was detected by CT. Karnofsky scores were graded for quality of life, and main signs and symptoms were graded. Levels of T-lymphocyte cell subsets and natural killer (NK) cells were detected. Progression free survival (PFS) was recorded and safety was evaluated. Result: The total effective rate was 72.73% in observation group, higher than 53.85% in control group (P<0.05). PFS was (8.1±1.6) months in observation group, superior to (5.4±1.5) months in control group (P<0.01). After treatment, the scores of main signs and symptoms in observation group were lower than those in control group, while scores of Karnofsky were higher than those in control group (P<0.01). The improvement of quality of life and index of stability in observation group was 87.27% in observation group, higher than 71.15% in control group (P<0.05). After treatment, levels of CD3+, CD4+, CD4+/CD8+ and NK in observation were increased as compared with the levels before treatment (P<0.01), and they were higher than those in control group (P<0.01). Level of CD8+ in observation group was decreased after treatment (P<0.01) and it was lower than that in control group (P<0.05). The incidence of Ⅲ and Ⅳ degree adverse effect in chemotherapeutic period was 21.82% in observation group, lower than 40.38% in control group (P<0.05). Conclusion: Jiaogulan Kangai decoction combined with dialectical synchronous chemoradiotherapy and maintenance treatment can shrink the size of solid tumor, relieve toxic and side effect of chemoradiotherapy, ameliorate clinical symptoms, improve patients' quality of life and immunity, and prolong progression free survival in the treatment of non-small-cell lung cancer, with obvious short-term and long-term efficacy.

      摘要:Objective: To observe the synergy and attenuation effect of Sanshen Diyu decoction as well as its effects on quality of life and lymph node metastasis in patients with radiotherapy for nasopharynx cancer. Method: One hundred and twenty patients were randomly divided into control group and observation group by random number table according to their admission sequence. Patients in control group received intensity modulated radiation therapy and Glutathione Sodium for injection. Based on the treatment of control group, patients in observation group also received Sanshen Diyu decoction. The treatment was lasted until two weeks after radiotherapy in both groups. Acute radiotherapy reactions were recorded. Before and after treatment, quality of life was evaluated by European cancer patient as well as head and neck cancer specific scale (EORTC QLQ-H&N35). Size of solid tumor was evaluated before and after treatment. Levels of epstein-barr virus NDA (EBV-DNA), vascular endothelial growth factor (VEGF), bone alkaline phosphates (B-ALP) and matrix metalloproteinase-2 (MMP-2) were detected both before and after treatment. Result: The total effective rate was 83.93% in observation group, higher than 67.27% in control group (P<0.05). Scores of acute radiotherapy reactions of mouth mucosa, ear, salivary gland, pharynx, esophagus and throat in observation group were lower than those in control group after radiotherapy (P<0.01). Scores of pain and other six symptoms by QLQ-H&N35 scale and 9 monosymptomatic scores including difficulty in opening mouth in observation group were lower than those in control group (P<0.01). Levels of EBV-DNA, B-ALP, MMP-2 and VEGF in serum of observation group were lower than those of control group after treatment (P<0.01). Conclusion: As an adjuvant treatment for radiotherapy, Sanshen Diyu decoction can relieve radioactive damages, ameliorate and stabilize quality of life, improve clinical efficacy and prevent tumor metastasis.

      摘要:Objective: To observe the efficacy of Tiaochong Shugan Tang in treating menopausal depression with kidney deficiency and liver qi stagnation syndrome and investigate its effects on levels of 5-hydroxytryptamine (5-HT) and tryptophan hydroxylase-2 (TPH-2) in serum. Method: One hundred and ten menopausal depression patients were randomly divided into control group (55 cases) and treatment group (55 cases) by random number table. The patients in control group were treated with flupentixol and melitracen tablets by orally taking (2 tablets/day, 1 tablet in the morning and one at noon). Based on the treatment in control group, the patients in treatment group also received Tiaochong Shugan Tang, 1 dose/day and bid. The patients in both groups were treated for 8 weeks. Scores of Hamilton depression (HAMD) scale and scores of Self-Rating Depression Scale(SDS) were compared between 2 groups. Clinical efficacy was analyzed after treatment for 8 weeks in 2 groups. Levels of 5-HT and TPH2 in serum were detected in 2 groups. Result: The total clinical effective rate was 96.36% in treatment group, obviously higher than 81.82% in control group, with statistical difference (P<0.05). After treatment, scores in anxiety and other factors of HAMD scale in treatment group were significantly lower than those in control one (P<0.01). After 4 and 8 weeks' treatment, scores of SDS in treatment group were significantly lower than those in control group at the same time, with statistical difference (P<0.01). After treatment, levels of 5-HT and TPH2 in serum of treatment group were significantly higher than those of control group, with statistical difference (P<0.01). Conclusion: Based on western medicine treatment, Tiaochong Shugan Tang could improve clinical symptoms, decrease scores of HAMD and SDS in treating menopausal depression with kidney Tang and liver qi stagnation syndrome, and the mechanism may be associated with down-regulating 5-HT and TPH2 levels in serum.

      摘要:Objective: To analyze the molecular network and biological pathways related to cellular immune of Baizhu Huangqi Tang (BZHQT) by using bioinformatic method, and provide ideas and basis for the immunomodulatory mechanism study of BZHQT. Method: On-line databases were searched for the chemical ingredients of BZHQT prescription. Target proteins related with each ingredient were found in PubChem. Then, these target proteins were uploaded to bioinformatic analysis software. Finally, the molecular networks of the target proteins and their biological pathways related to cellular immune were analyzed. Result: Sixty-one target proteins of the chemical ingredients were searched from BZHQT and six molecular networks of BZHQT were obtained. Multiple biological pathways related to cellular immune were obtained, and the top 3 pathways were Calcium-induced T Lymphocyte Apotosis, CCR5 Signaling in Mcrophages and IL-12 Signaling and Production in Macrophages. Target proteins of BZHQT that participated in above three signaling pathways included T cell receptor (TCR), protein kinase C (PKC), and peroxisome proliferator activated receptor gamma (PPARG). Conclusion: Complicated molecular networks and signaling pathways related to cellular immune of BZHQT were obtained. We providing ideas and basis for study of immunomodulatory mechanism of BZHQT.

      摘要:Objective: To analyze the application of Rhizoma Coptidis-Cortex Magnoliae Officinalis herbal pair compatibility, reveal its compatibility law and provide reference for clinical use. Method: Totally 475 formulae containing Rhizoma Coptidis-Cortex Magnoliae Officinalis herbal pair were retrieved from the 'Traditional Chinese Medicine Formula Database' established by Institute of Information on Traditional Chinese Medicine, China Academy of Chinese Medical Sciences. Then a new database was built to analyze the compatibility ratio, corresponding major diseases and major departments for the herbal pairs. Result: In the 475 formulae, ratio ‘1: 1’ of Rhizoma Coptidis and Cortex Magnoliae Officinalis appeared to be the most (215 times, 45.26%); the most frequent disease genus was internal medicine (329 times, 69.26%); in the formulae for common colds and flu, ratio ‘1: 1’appeared to be the most (10 times, 50%). Conclusion: The data analysis methods such as frequency statistics for the compatibility law and application rules of herbal pairs can provide a theoretical basis for clinical application.

      摘要:Objective: To evaluate the methodological quality of the systematic review/Meta-Analysis (SR/MA) of Buyang Huanwu Tang in treatment of stroke by comparing with other interventions. Method: CMCC, Wanfang China National Knowledge Infrastructure, VIP and Cocranhe databases were searched to select the literature on systematic review/Meta-analysis (SR/MA) of Buyang Huanwu Tang and other interventions in treatment of stroke. The retrieval time limit was up to May 2015.Oxman-Guyatt Overview Assessment Questionnaire (OQAQ) Scale was used to evaluate the methodological and reporting quality of the eligible SR/MA. Result: A total of 92 SR/MA literature were obtained, and 9 of them were finally included. The included Meta analysis literature had the following shortcomings: no comprehensive literature search, high selection bias, poor methodological quality of the original articles, and poor accuracy of data consolidation method and premise. The experimental design of the original articles had the following shortcomings: unclear randomizatioon methods, no grouping concealment, no blinding, no reporting of intent-to-treat (ITT) analysis, uncertain co-intervention and contamination. Conclusion: The number of domestic and overseas literature on SR/MA of Buyang Huanwu Tang in treating stroke is gradually increasing in recent years, but few of them could reach high quality SR/MA, so it is necessary to improve the methodological quality and regulate the experimental design

      摘要:Objective: To compare and systematically evaluate Tianma preparation and conventional antihypertensive western medicine in clinical efficacy and safety in the treatment of essential hypertension. Method: Systematic literature searches were perform in Cochrane library, PubMed, EMBase, CBM, CNKI, VIP database, Wanfang database and CDMD from Jan 1979 to July 2015.The quality evaluation of the included studies and data extraction were performed by using Cochrane system. The Meta-analysis was performed by using Revman 5.3.5 software. Result: A total of 27 studies involving 2 674 patients were included. The results of Meta-analysis showed that: as compared with conventional western medicine group, the antihypertensive effects and efficacy of traditional Chinese medicine syndromes were more obvious in Tianma preparation group [OR=1.67, 95%CI(1.14, 2.46), P=0.009<0.05]and [OR=3.65, 95% CI (2.42, 5.51), P<0.000 01]respectively. The safety evaluation results showed that: there was statistically significant difference in adverse reaction rate [RR=0.36, 95% CI (0.23, 0.56), P<0.000 01]. Conclusion: Tianma preparation has very good clinical efficacy and safety for the treatment of essential hypertension, but the quality of included studies is low, so it also needs further in-deep research.

      摘要:As a kind of endogenous non-coding RNA with regulation and control function, microRNA widely participates in human development, cell proliferation and lipid metabolism processes, and also plays a regulatory role in most of the life processes. In recent years, the scholars believe that vascular dementia (VD) may have a close relationship with lack of acetylcholine, brain cells apoptosis, synaptic change after cerebral ischemia and inflammation caused by cerebral ischemia. In addition, microRNA actively participates in the occurrence and development of the above-mentioned risk factors of VD, so microRNA may play a vital role in the occurrence and development of the disease. With the gradual in-depth research on the regulation mechanism of microRNA in life processes in the body, numerous studies have confirmed that microRNA expression in circulation of blood has specific changes in patients with prostate cancer, breast cancer and femoral head necrosis. As a potential new disease marker, the circulating microRNA has been recognized by a large number of scholars, and the research of circulating microRNA change in the body is supposed to be used for early diagnosis of the disease. Research progress of microRNA related to VD is reviewed in this paper, and at the same time, the effect of microRNA for early diagnosis and treatment of vascular dementia is also discussed.

      摘要:Arisaema Cum Bile is usually used in dissipating phlegm, dispelling wind and relieving convulsion.Now there are no relevant method, examination and content determination of quality standards of Arisaema Cum Bile.In the 2015 edition of Chinese Pharmacopoeia, only brief process description of Arisaema Cum Bile exists, which lead to varying quality with different manufacturers and go against development of quality standardization, and it will influence safety and effectiveness of clinical medication.Through literature survey and field research, aiming at main problems of quality standards of Arisaema Cum Bile and its process auxiliary material, we put forward corresponding suggestions and references for colleague which help to regulate and improve its quality standards.
