      摘要:Objective: To investigate urine metabolic profile of rats with deficiency-cold hemorrhagic disease,and discuss intervention effects of dried and charred Zingiberis Rhizoma Recens. Method: Proton nuclear magnetic resonance (1H-NMR) technique was used to analyze metabolome in urine.Partial least squares discriminant analysis (PLS-DA) was processed to analyze metabonome difference among four groups.Seven potential biomarkers were selected by variable importance in the projection (VIP).Principal component analysis (PCA) was employed to process data from the charred Zingiberis Rhizoma Recens treatment group,hemostatic effects of charred Zingiberis Rhizoma Recens were investigated through the content of potential biomarkers. Result: Charred Zingiberis Rhizoma Recens could make disorder endogenous biomarkers of the model rats to normal levels,while dried Zingiberis Rhizoma Recens had weaker efficacy.Acetoacetic acid,pyruvic acid,nitrogen oxygen trimethylamine,taurine and tryptophan were efficacy of hemostasis biomarkers in charred Zingiberis Rhizoma Recens. Conclusion: Metabolomics can reflect the whole status of metabolism in biosystems,it can be treated as a potential powerful approach in pharmacological studies for different processed products and investigation of essence of syndrome in traditional Chinese medicine.

      摘要:Objective: To optimize softening cutting processes of Morindae Officinalis Radix,in order to provide technical parameters for standard production of the pieces. Method: On the basis of single-factor test,with comprehensive score of rubiadin-1-methyl ether,monotropein and nystose as indexes,orthogonal design was adopted to detect effects of water volume,moistening time,moistening temperature and cutting length on softening and cutting processes.HPLC was used to determine the content of rubiadin-1-methyl ether,which was eluted with mobile phase of acetonitrile-0.2% aqueous phosphoric acid in gradient mode,detection wavelength was 277 nm.The content of monotropein was eluted with mobile phase of acetonitrile-0.1% aqueous phosphoric acid in gradient mode,detection wavelength was 235 nm.HPLC-CAD was used to determine the content of nystose with mobile phase of methanol-water (3 : 97). Result: Optimal softening and cutting processes were as follows:water volume of 1.0 times,moistening temperature at 50 ℃,moistening time of 9 h and cutting length of 10-15 mm.Average fractions of rubiadin-1-methyl ether,monotropein,nystose and water extract were 0.045%,5.074%,63.202% and 74.651%,respectively. Conclusion: These optimized softening and cutting processes for Morindae Officinalis Radix are rational and feasible,it can be used as technical parameters for standard production of the pieces.

      摘要:Objective: To determine the content of total flavonoids from Pilea aquarum and evaluate its antioxidative activity. Method: With the content of total flavonoids as an indicator,based on single factor experiments,response surface methodology was adopted to optimize extraction process by taking liquid-solid ratio,extraction temperature and time as factors.The content of total flavonoids in P. aquarum from different places were determined,then antioxidation of total flavonoids were evaluated against DPPH and ABTS free radical. Result: Optimum coditions were as follows:liquid-solid ratio of 1 : 19,extraction temperature at 71 ℃ and extraction time of 126 min.Yield of total flavonoids was as high as 7.08 mg · g-1 under these conditions.The content of total flavonoids in P. aquarumfrom different places was from 4.35 to 7.10 mg · g-1 with obvious regional differences.Scavenging capacity of total flavonoids were as same as vitamin C against DPPH and ABTS free radical. Conclusion: There is obvious difference of total flavonoids content in P. aquarum from different places,and total flavonoids has good antioxidative activity in a dose-dependent manner.

      摘要:Objective: To optimize molding process of Tangshenning granules. Method: With moldability,dissolubility,hygroscopicity and post-moisture absorption appearance as comprehensive indexes,single factor test was conducted to determine excipient types,central composite design-response surface methodology was adopted to optimize excipient ratio,dosage,wetting agent concentration and other molding process parameters. Result: According to optimal molding process,ratio of extract-dextrin-mannitol was 4 : 3 : 3,75% ethanol was added as softening materials,drying temperature was at 55 ℃.Average value of overall desirability (OD) was 0.786,its deviation from the theoretical prediction value (0.78) of the model equation was about 0.8%.Yield of qualified granules was about 88.44%,melting time was less than 0.5 min,critical relative humidity was about 70%,angle of repose was 34.8 degrees,and bulk density was 0.49 g · mL-1. Conclusion: This optimized molding process is stable and feasible with a high qualified rate,good dissolubility and high anti-hygroscopicity for these prepared Tangshenning granules,and it can provide basis for mass production of this preparation.

      摘要:Objective: To optimize processing technology of Plumbago zeylanica. Method: With composite score of content of plumbagin and vanillic acid as index,orthogonal test was adopted to optimize processing technology of P .zeylanica by taking processing time,the amount of liquor and ethanol concentration as factors.Contents of plumbagin and vanillic acid were determined by HPLC,mobile phase was methanol -0.15% phosphoric acid for gradient elution,detection wavelength was 260 nm. Result: The best processing technology of P. zeylanicawas:processing time of 45 min,liquor dosage of 0.2 mL · g-1,ethanol concentration of 12%.Contents of plumbagin and vanillic acid were 0.013 0% and 0.019 3%,respectively. Conclusion: This process is stable,feasible and suitable for quality control of P. zeylanica and preparation of its processed product with wine.

      摘要:Objective: To compare difference in contents of volatile components,total flavonoids and vitexicarpin of Viticis Fructus with different maturity levels. Method: Gas chromatography-mass spectrometry (GC-MS) was used to separate and identify volatile components in Viticis Fructus.Area normalization method was applied to calculate relative mass fractions of all components.In addition,ultraviolet spectrophotometry and HPLC were combined to measure contents of total flavonoids and vitexicarpin. Result: Houtian's yellow fruits and black fruits showed 35 and 36 compounds respectively,including 30 identical compounds.Duchang's yellow fruits and black fruits showed 31 and 27 compounds respectively,including 23 identical compounds.Viticis Fructus with different maturity degrees contained similar types of volatile oil,but with a great difference in their contents.The mass fraction for vitexicarpin in Houtian's yellow fruits and black fruits,Duchang's yellow fruits and black fruits were 0.268%,0.209%,0.226%,0.242%,the mass fraction of total flavonoids were 7.902%,3.540%,3.982%,2.617%,respectively. Conclusion: Different samples show a great difference in the content of total flavonoids,but with no significant difference in vitexicarpin content between yellow fruits and black fruits,suggesting a big effect of maturity on quality of Viticis Fructus.This study provides a reference for rational harvesting and utilization of Viticis Fructus.

      摘要:Objective: To analyze the volatile components of Alpiniae Oxyphylla Fructus produced in Hainan before and after being processed with salt. Method: The volatile oil was extracted from Alpiniae Oxyphylla Fructus produced in Hainan by the steam distillation, and analyzed and identified with the GC-MS technology. The relative percentage content of the compounds in the volatile oil was determined by the peak area normalization method. Result: The content of volatile oil of Alpiniae Oxyphylla Fructus produced in Hainan decreased from 1.36% to 1.12% after being processed with salt. Respectively 53 components and 48 components were identified in raw Alpiniae Oxyphylla Fructus and processed Alpiniae Oxyphylla Fructus. Specifically, 42 common components were detected in Alpiniae Oxyphylla Fructus before and after being processed with salt. After being processed, six new components were detected, 11 original components were not detected. Conclusion: According to the GC-MS analysis, the main contents in Alpiniae Oxyphylla Fructus produced in Hainan are monoterpenes and sesquiterpenes, which showed changes in their volatile oil composition and relevant content after being processed with salt. These changes will provide basis for further optimizing the processing methods and clarify the different effects of Alpiniae Oxyphylla Fructus before and after being processed with salt.

      摘要:Objective: To provide the scientific basis for the utilization and development of Origanum vulgare by formulating the quality control standard of O. vulgare. Method: Totally 10 batches of O. vulgare from different origins were collected to identify the primary plants and describe their characters. The impurities, moisture, ash, sour insoluble ash and extractum were detected based on appendix of Chinese Pharmacopoeia 2010 version (Vol). The pharmacognostic study and qualitative identification with thin layer chromatography were conducted. The HPLC method was also adopted for the content determination of rosmarinic acid in the O. vulgare. Result: The contents of impurities, moisture, ash, sour insoluble ash shall not exceed 2.0%, 10.0%, 10.0%, 2.5%.The ethanol-soluble extractum shall be no less than 19.0%. The content of rosmarinic acid shall be no less than 0.9%. Conclusion: The experimental method was accurate and reliable. The above indexes and parameters can be used as the basis for formulating the quality standards for O. vulgare and developing and utilizing O. vulgare.

      摘要:Objective: To develop methodology for significant discrimination of different groups of Gentianae Macrophyllae Radix based on 1H-NMR analysis combined with principal component analysis (PCA). Method: Five groups of Gentianae Macrophyllae Radix were extracted by the same method,1H-NMR spectra were obtained using a 400 MHz NMR spectrometer.Then data were analyzed by hierarchical cluster analysis and PCA. Result: Different groups of Gentianae Macrophyllae Radix could be discriminated by hierarchical cluster analysis.Furthermore,6 principal components were extracted in PCA,their cumulative contribution rate was 98.9%,and loading plot of PC1 and PC2 explained 76.9% of the original variance. Conclusion: 1H-NMR combined with PCA is an effective method for discrimination of Chinese medicines,its pre-treatment is convenient,therefore it can be used as a valuable means for discrimination of different groups of herbal medicines.

      摘要:Objective: To design and synthesize aderivative of oleanolic acid bearing double ‘Michael reaction acceptor’ group and to study anti-tumor activity in vitro. Method: The target compound was synthesized from oleanolic acid, via several steps including acylation, esterification, hydrolysis, oxidation and aldolcondensation. The anti-tumor activity of the compound was studied in vitro by MTT assay on Human promyelocytic leukemia cells HL-60, human hepatocellular carcinoma cells SMMC-7721, human lung adenocarcinoma cells A-549, human mammary adenocarcinomacells MCF-7, colon carcinoma cells SW480. Result: The compound was synthesized, and chemical structure was confirmed by 1H-NMR,13C-NMR and ESI-MS. Conclusion: Preliminary anti-tumor activity in vitro displays the compound had good activity in antitumors, especially inagainsting SW480 cell lines.

      摘要:Objective: To separate and identify volatile and fat-soluble components in Tibetan medicine Brag-zhun by GC-MS. Method: The water distillation method was used to extract the volatile components, and petroleum ether was used to extract fat-soluble components. GC-MS technology was used to separate and identify the components. The peak area normalization method was used to calculate the relative content of each component. Result: Forty-one chemical components were identified in volatile components of Brag-zhun, accounting for 45.67% of the total weight, there were 6 kinds of component relative content of over 2%: 1-naphthalenepropanol,a-ethenyldecahydro-a,5,5,8a-tetramethyl-2-methylene-, (aR,1S,4aS,8aS)- (11.97%),cis-verbenol(6.38%),α-copaene(2.67%),2-tetradecene,(E)-(2.56%),cedrol(2.49%), docosanoic (2.17%); forty-five chemical components were identified in fat-soluble components in Brag-zhun, taking up 92.86% of the total weight, there were 14 kinds of component relative content of over 2%: eicosane(12.83%), pentacosane(11.60%), methyl palmitate(8.88%), triacontane(6.82%), heptadecane(5.94%),5-eicosene, (E)- (5.92%),1-hexacosanol(3.50%), hentriacontane(3.43%),1-naphthalenepropanol,a-ethenyldeca-hydro-a,5,5,8a-tetramethyl-2-methylene-,(aR,1S,4aS,8aS)- (3.18%), methyl eicosanoate(3.15%), methyl stearate(2.94%), behenicacid methyl ester(2.89%), heneicosane(2.55%), tetracosanoic acid,methyl ester(2.15%). Conclusion: This study focused on volatile and fat-soluble components in Brag-zhun for the first time, in order to provide the study basis for further defining the formation mechanism of Brag-zhun.

      摘要:Objective: To establish a HPLC method for simultaneously determining three components in Zhibai Dihuang pills, i.e. morroniside, loganin and paeonol. Method: The Phenomenex C18 column (4.6 mm×250 mm, 5 μm) was adopted and eluted in gradient (0-5 min, 5%-7%A;5-26 min, 7%A;26-45 min, 7%-20%A;45-55 min, 20%-60%A;55-65 min, 60%A) with the mobile phase of acetonitrile (A)-0.3% phosphoric solution (B). The flow rate was 1 mL · min-1. The column temperature was 40 ℃. The detection wavelength was set at 240 nm for morroniside and loganin in the first 45 minutes, and then switched to 274 nm for paeonol between the 46 and 65 minutes. Result: The linear ranges of morroniside, loganin and paeonol were between 0.055 5-1.11 μg (r=0.999 9), 0.076 4-1.528 μg (r=0.999 9), 0.098 2-1.964 μg (r=0.999 9), respectively. The average recoveries of morroniside, loganin and paeonol were 100.6%, 101.6% and 101.9%, respectively, and the relative standard deviations were 2.5%, 1.8% and 1.5%, respectively. Conclusion: This method is simple, accurate and highly reproducible and so can be used to control the quality of Zhibai Dihuang pills.

      摘要:Objective: To establish a method for determination of the moisture in Polygoni Multiflori Radix Praeparata by near-infrared spectroscopy (NIRS). Method: The moisture content of 135 samples was determined by the method of drying under oven. The second derivative and the standard normal variate (SNV) were used as spectral preprocessing options, and calibration model of moisture content was established by the partial least squares regression analysis. Result: The correlation coefficients,the root-mean-square error of calibration, the root-mean-square error of predication and the root-mean-square error of cross-validation of the calibration model for moisture content were 0.990 71,0.152,0.175 and 0.482 57. Conclusion: The method is convenient, accurate, reliable and it is of high reproducibility and can predict moisture content of Polygoni Multiflori Radix Praeparata rapidly.

      摘要:Objective: To determine the content of Aconitum monoester alkaloids (AMA) in Guifu Zhitong cataplasm (GZC) by near infrared spectroscopy (NIR). Method: NIRs of 53 batches of GZC samples were determined by using TQ Analyst 8.0 software. Within the wavebands between 10 000-4 000 cm-1, one main factor was selected from the primary spectrum. The quantitative calibration determination model was established with partial least squares (PLS). Result: The established quantitative calibration model showed a good correlation coefficient (r=0.942 0), low root mean square error of calibration (RMSEC) at 0.013 5, root mean square error of prediction (RMSECV) of 0.014 4, with the average predicted recovery rate of validation set of 101.44%. Conclusion: The quantitative calibration model can predict the content of alkaloids in GZC accurately and so can be used for the quality control of aconitum monoester alkaloids in GZC.

      摘要:Objective: To establish the multi-index quantitative method for Qigu capsules. Method: The contents of echinacoside, naringin, epimedin B, epimedin C and icariin were determined simultaneously by HPLC. Platisil-C18column (4.6 mm×250 mm, 5 μm) was used and eluted with the mobile phase of acetonitrile-methanol-water in a gradient mode at the flow rate of 1 mL · min-1. The detection wavelength was 270 nm, and the column temperature was 30 ℃. Result: Echinacoside, naringin, epimedin B, epimedin C, icariin showed a good linear relationship in the ranges of 91-3 094 ng(r=0.999 8), 19-646 ng(r=0.999 7), 11-374 ng(r=0.999 5), 25-850 ng(r=0.999 5), 29-986 ng(r=0.999 6), respectively. The average recoveries were 98.9% (RSD 1.2%), 99.1% (RSD 1.3%), 99.1% (RSD 1.5%), 99.4% (RSD 1.5%), 99.3% (RSD 1.5%), respectively. Conclusion: This method is simple, accurate and highly reproducible, and so can be used to control the quality of Qigu capsules.

      摘要:Objective: To establish an HPLC method for simultaneously determining chlorogenic acid, geniposide, baicalin, baicalein and wogonin in Huangzhihua oral liquid (Lonicerae Japonicac Flos, Gardeniae Fructus, Scutellariae Radix, et al). Method: The determination was performed by HPLC on Agilent SB-C18 column (4.6 mm×250 mm, 5 μm) by gradient elution at the flow rate of 1.0 mL · min-1. The mobile phase was acetonitrile (solvent A)-0.1% phosphoric acid (solvent B). The detection wavelength was respectively set at 327 nm in the range of 0-13 min, 238 nm in the range of 13-15 min and 280 nm in the range of 15-45 min. Result: According to the regression equation, the linear range for chlorogenic acid was 4.096×10-3-1.228 8×10-1μg(r=0.999 3);the linear range for geniposide was 4.204×10-3-1.261 2×10-1μg(r=0.999 9);the linear range for baicalin was 0.117 5-3.525 μg(r=0.999 9); the linear range for baicalein was 4.152×10-3-1.245 6×10-1μg (r=0.999 9);and the linear range for wogonin was 0.425 6×10-3-1.276 8×10-2μg(r=0.999 4). The average recoveries of the above five components were 99.31%(RSD 0.8%);99.62%(RSD 0.5%);99.94%(RSD 1.3%);99.64%(RSD 0.8%);100.1%(RSD 1.2%) respectively.       摘要:Objective: To establish a method for determining ginsenoside Rg1, Re, Rb1, Rd and astragaloside Ⅳ in Yixinshu dispersible tablets. Method: By using HPLC-ELSD, Shim-pack ODS column was eluted with the mobile phase of acetonitrile-water at the flow rate of 1.0 mL · min-1. Result: According to the regression equation, ginsenoside Rg1, ginsenoside Re, ginsenoside Rb1, ginsenoside Rd and astragaloside Ⅳ showed a good linear relationship at 0.685-3.428 μg (r=0.999 5), 1.819-9.094 μg (r=0.999 0), 3.717-18.586 μg (r=1), 1.042-5.212 μg (r=0.999 5), 0.388-1.940 μg (r=0.999 8), respectively. The average recoveries were 100.05% (RSD 1.6%) for ginsenoside Rg1, 96.21% (RSD 1.1%) for ginsenoside Re, 96.20% (RSD 1.1%) for ginsenoside Rb1, 100.52% (RSD 2.5%) for ginsenoside Rd and 99.26% (RSD 1.6%) for astragaloside Ⅳ. Conclusion: This method is simple, accurate and highly reproducible and so can be used to simultaneously determine multiple components and control the quality of Yixinshu dispersible tablets.

      摘要:Objective: In order to study the chemical constituents of Cryptolepis sinensis. Method: Column chromatography on Silica gel, RP-l8 and Sephadex LH-20 were applied for the isolation and purification of the constituents. The structures were elucidated on the basis of spectral analysis, chemical evidences and by comparison with the data reported in literatures. Result: From the crude CHCl3 fraction of the EtOH extract of C. sinensis, 11 phenylpropanoids were isolated and identified as (+)-pinoresinol (1), (+)-8-hydroxy-pinoresinol (2), (+)-syringaresinol (3), horsfieldin (4), balanophonin (5), ficusesquilignan A (6), ficusesquilignan B (7), isoscopoletin (8), 7-hydroxy-6, 8-dimethoxycoumarin (9) patuletin (10) and methyl sinapate (11). Conclusion: All compounds 1-11 were isolated and identified from C. sinensis for the first time.

      摘要:Objective: To study the effect of micromolecular hydropilic extract (MHE, a P-glycoprotein inhibitor) from vingar-baked Bupleuri Radix on pharmacokinetics of methotrexate. Method: Sprague-Dawley male rats were orally administrated with methotrexate or that one combined with MHE respectively.Blood samples were collected from their eyes at different time point,treated with 20% perchloric acid protein precipitation method and then analyzed by HPLC.Pharmacokinetic parameters were evaluated by non-compartment model. Result: Compared with the group using methotrexate alone,AUC0-t,AUC0-∞, MRT0-t,t1/2of the one combined with MHE increased by 16.6%,33.8%,26.1%,120%,respectively;while CL decreased by 20.3%,but with no significant difference between their pharmacokinetic parameters. Conclusion: MHE can enhance bioavailability of methotrexate,and attention should be paid to toxic and side effects of their combined administration.

      摘要:Objective: To study pharmacokinetics and bioavailability of isochlorogenic acid A in rats with different adiminstration way. Method: A HPLC method was developed for determination of isochlorogenic acid A in rat plasma.Intravenous and intragastric doses of 32 mg ·kg-1 and 90 mg ·kg-1 were respectively administered,the concentration of isochlorogenic acid A were determined with HPLC,detection wavelength was set at 300 nm.Pharmacokinetic parameters were calculated with 3P97 software.Absolute bioavailability of isochlorogenic acid A was calculated according to AUC0-∞ and doses of isochlorogenic acid A by intravenous and intragastric administration. Result: Excellent linear relationship was obtained in the range of 0.16-110.00 mg ·L-1 with R2 of 0.998.Recoveries of isochlorogenic acid A with three concentrations (0.43,6.88,55.00 mg ·L-1) were (89.43±2.84)%,(93.16±3.95)% and (85.91±2.04)%.The intra-day RSD were 11.8%,4.0% and 4.0%.The inter-day RSD were 6.5%,5.8% and 5.8%,respectively.Metabolic processes of isochlorogenic acid A in rats fitted in with two-compartment model following intravenous and intragastric administration.Elimination half-life were (29.49±0.75) min and (44.48±0.13) min. AUC0-∞ were(355.40±32.58),(319.91±51.00) mg ·min-1 ·L-1.Its absolute bioavailability in rats was 30.71%. Conclusion: Metabolic processes of isochlorogenic acid A in rats conform to linear dynamics with fast metabolism and short half-life.

      摘要:Objective: To build an evaluation method for in vivo pharmacokinetic study on multicomponents in a complex system of traditional Chinese medicines (TCMs) in rats. Method: After mixed suspension of capsules with components from Hedyotis diffusa was orally administered to rats,blood samples were collected from eye of rats at different time points.Plasma samples were scanned by ultraviolet-visible spectrophotometer to calculate area under the absorbance-wavelength curve (AUAWC) and then produce total plasma concentration in rats at each time point.Pharmacokinetic parameters were calculated with DAS2.0 software and compared with data of every single component-quercetin,oleanolic acid and stigmasterol in plasma samples obtained by high performance liquid chromatography (HPLC),in order to determine feasibility of the above two methods. Result: With method of AUAWC,mixed total concentration of capsules conformed to one-compartment model in rats,with Tmax at 0.5 h,Cmax of 2.85 mg · L-1,AUC0-∞ of 26.81 mg · L-1 · min-1.HPLC method also showed a one-compartment model for quercetin,oleanolic acid and stigmasterol,with Tmax at 1 h,Cmax of 0.17 mg · L-1,AUC0-∞ of 0.71 mg · L-1 ·min-1 for quercetin;Tmax at 0.5 h,Cmax of 0.69 mg · L-1,AUC0-∞ of 10.69 mg · L-1 · min-1 for oleanolic acid;Tmax at 0.5 h,Cmax of 3.84 mg · L-1,AUC0-∞ of 12.49 mg · L-1 · min-1 for stigmasterol,respectively. Conclusion: AUAWC can be used to study pharmacokinetics of capsules with components from H. diffusa in rats.Its combination with HPLC can better solve synchronous test for whole and individual components in TCM preparations with a better complementarity between them.

      摘要:Objective: To investigate the inhibitory effect of paeoniflorin on isoproterenol (ISO)-induced myocardial hypertrophy in rats and its potential mechanism. Method: Totally 40 SD rats were randomly divided into five groups: the normal group, the model group and paeoniflorin low, middle and high does groups (20, 40, 80 mg · kg-1), with eight rats in each group. Except for the normal group, all of the rest groups were subcutaneously injected with isoprenaline (5 mg · kg-1) for 10 days to induce the myocardial hypertrophy model and at the same time intraperitoneally injected with PEF. At the end of drugs treatment, their hearts were collected to measure the heart weight (HW), left ventricular weight (LVW) and the ratio to body weight (BW). The changes in the size of cardiac cells were observed by hematoxylin-eosin staining. The mRNA expressions of atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP) and cardiotrophin-1 (CT-1) were measured by real-time PCR. The protein expression of CT-1 and level of reactive oxygen species (ROS) in myocardium were detected by Western blot and DCFH-DA florescent probe, respectively. Result: Compared with the normal group, the model group showed significant increases in the ratio of HW/BW and LVW/BW, cardiac myocyte cross-sectional area, mRNA expressions of ANP and BNP, CT-1 mRNA and protein expressions and ROS level at myocardial tissues (P <0.01). Compared with the model group, PEF could obviously decrease the ratio of HW/BW and LVW/BW, cardiac myocyte cross-sectional area, mRNA expressions of ANP and BNP, CT-1 mRNA and protein expressions and ROS level at myocardial tissues. Conclusion: PEF can inhibit ISO-induced myocardial hypertrophy. Its mechanism may be correlated with the down-regulation of CT-1 expression by inhibiting ROS production.

      摘要:Objective: The purpose of this study is to study the inhibition of emodin on rabbit aortic smooth muscle cell proliferation(CCC-SMC-1) and on gene expression of cathepsin D, K. Method: In a 2×104/mL density vaccination cells,the experiment was divided into seven groups, including 0.01,0.05,0.1,0.15,0.2,0.25 mmol · L-1of emodin groups, control group. Cells in each group were cultured for 24 h(n=6). The rabbit aortic smooth muscle cells was cultured, to observe emodin on rabbit aortic smooth muscle cell proliferation was determined by MTT method.The impact of emodin on gene expression of relative amount of cathepsin K (Cat K) and cathepsin D (Cat D) of rabbit aortic smooth muscle cells was assayed with PCR. Result: MTT result suggested that emodin had the inhibitition effect on rabbit aortic smooth muscle cell proliferation when the concentration was in the range of 0.05-0.25 mmol · L-1. The 50% inhibitory concentration(IC50)was 0.1 mmol · L-1. The results of PCR showed that the expression of Cat K and Cat D gene in emodin groups was significantly increased (P< 0.05) after 24 h compared with control cells. Conclusion: Emodin can inhibit the proliferation of CCC-SMC-1 cells, the mechanism of which may be related to up-regulation of the gene of Cat K and Cat D expression.

      摘要:Objective: To study effect of total alkaloids in water extraction from Stephaniae Tetrandrae Radix S.Moore(STRM) on the collagen-induced arthritis(CIA) in order to further explore the anti-rheumatism effects of alkaloidthe water-soluble alkaloids with big polarity. Method: The 732 type cation resin was used to enrich total alkaloid in water extraction from STRM. Rat CIA model was established by injection of bovine Ⅱ collagen type on toes, tail and backside in rats. After 7 days same injection was performed. Alkaloids high, middle, low dose(0.162,0.324,0.486 g · kg-1), Tripterygium wilfordii glycosides (1.5 mg ·kg-1) and dexamethasone (10.8 mg · kg-1) were administrated for 21 days. The arthritis index(AI) and toes swelling degree were observed, the level of interleukin-1(IL-1),interleukin-1β(IL-1β),interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in serum were assayed and the ankle tissue pathological changes were observed. Result: Compared with normal group,AI in model group and toes swelling degree significantly increased(P <0.01).Compared with model group,high, middle, low dose group of total alkaloid in water extraction from STRM could improve AI and toes swelling degree, and significantly decrease the level of IL-1,IL-1β,IL-6 and TNF-α (P <0.01) in serum, and reduce the pathological synovial tissue damage. Conclusion: Total alkaloid in water extraction from STRM has certain therapeutic effect on rheumatoid arthritis,and can significantly decrease the level of inflammatory factors.

      摘要:Objective: To explore the protective effects of astragaloside IV (As IV) on the dysfunction of human umbilical vein endothelial cells (HUVECs) induced by hydrogen peroxide and the underlying mechanism. Method: Human umbilical vein endothelial cells were cultured in vitro and divided into five groups including normal,model(H2O2 180 μmol · L-1), As IV(25 μmol · L-1)+H2O2(180 μmol · L-1),As Ⅳ(50 μmol · L-1)+H2O2(180 μmol · L-1),As IV(100 μmol · L-1)+H2O2 180 μmol · L-1.The cell viability was evaluated by MTT assay, endothelial superoxide anion generation was determined by staining of cells with fluorescent-labeled dihydroethidium. Western blot were used to quantify intracellular protein expression of nuclear factor-κB(NF-κB)p65,IκB-α and endothelial nitric oxide synthase (eNOS), the expression of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in the extracellular fluid by enzyme-linked immunosorbent assay (ELISA). Result: Compared with the normal group, the cell viability of model group were decreased remarkably, the intracellular superoxide anion generation were increased significantly, the protein expression of p65 was increased while the kB-α was decreased and the eNOS was increased, the production of TNF-α and IL-6 in the extracellular fluid were increased obviously(P <0.01). Comparing with the model group, the cell viability was increased, the intracellular superoxide anion generation was decreased, the protein expression of p65 was decreased, while the IκB α was increased and the eNOS was decreased, the production of TNF-α and IL-6 was decreased in 3 dose groups of As Ⅳ, and the differences showed a dose-dependent(P <0.05). Conclusion: As Ⅳ has protective effects on the dysfunction of human umbilical vein endothelial cells induced by H2O2.The mechanism may be related to reducing superoxide anion production, inhibiting eNOS uncoupling which could produce superoxide anion, thus attenuating oxidative stress and reducing the expression of NF-κB and related inflammatory cytokine. Consequently As Ⅳ could protect human umbilical vein endothelial cells from damage induced by H2O2.

      摘要:Objective: To explore the anti-diabetic effect and mechanism of components in Jinqi Jiangtang tablet in vitro. Method: A systematic anti-diabetic activity evaluation screening model in vitro was established by scavenging DPPH free radicals, reactive oxygen free radicals, and inhibiting advanced glycation end products formation, alpha-glucosidase and aldose reductase, lipase, NO production from LPS-activated mice macrophages, in order to detect the effect of four standard components—total flavonoids of Astragalus, total saponins of Astragalus, total organic acids of Honeysuckle, Berberine alkaloids in Jinqi Jiangtang tablet on the above mechanisms. Result: Total flavonoids of Astragalus, total organic acids of Honeysuckle, Berberine alkaloids showed scavenging activity on DPPH free radicals and reactive oxygen free radicals;all components showed no inhibitory activity on advanced glycation end products formation;total flavonoids of astragalus, total organic acids of Honeysuckle showed inhibitory activity of alpha-glucosidase;total flavonoids of Astragalus, total saponins of Astragalus, Total organic acids of Honeysuckle showed inhibitory activity of aldose reductase;total flavonoids of Astragalus, total saponins of Astragalus, total organic acids of Honeysuckle showed inhibitory activity of lipase;total saponins of Astragalus, Berberine alkaloids showed inhibitory effects of NO production from LPS-induced mice macrophages. Conclusion: Jinqi Jiangtang tablet has multi-component and multi-target synergistic anti-diabetic effect and achieves the mechanisms by inhibiting alpha-glucosidase, inhibiting lipase, scavenging free radicals, inhibiting aldose reductase and inhibiting the release of NO.

      摘要:Objective: To study the effect of representative Qi-tonifying and blood-activating formulae Buyang Huanwu Tang (BYHWT) on the atherosclerosis model, in order to observe the aorta Rho kinase, PAl-1 and eNOS mRNA expressions and blood lipid indicator and reveal new targets and new ways of the formulae in inhibiting atherosclerosis. Method: Totally 60 rats were randomly divided into six groups:the normal control group, the model group, BYHWT low and high dose of groups (10, 20 g · kg-1), the Simvastatin group (0.6 mg · kg-1) and the BYHWT prevention group (10 g · kg-1). Except for the normal group, the other groups were fed with VD3 and high fat diet to induce atherosclerosis and given corresponding drugs. The BYHWT prevention group was administered with drugs during the modeling. After the successful modeling and 28-day intervention, the expression level of Rho kinase, plasma tissue plasminogen activator inhibitor 1(PAl-1), endothelial nitric oxide synthase(eNOS) mRNA expression, total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterin(HDL-C), low density lipoprotein cholesterin(LDL-C) and oxidized low-density lipoprotein(ox-LDL) in atherosclerosis aorta were detected. Result: Compared with the normal group, the model group showed significantly higher Rho kinase, PAl-1 mRNA expression, liquid TC, TG, HDL-C, ox-LDL and lower eNOS mRNA expression and LDL-C, all with significant statistical differences (P <0.01);BYHWT low and high dose groups, the Simvastatin group and the BYHWT prevention group showed notably lower Rho kinase, PAl-1 mRNA expression, lipid TC, TG, HDL-C, ox-LDL and higher eNOS mRNA expression, LDL-C, with significant statistical differences (P <0.05,P <0.01). Conclusion: BYHWT has an effect in down-regulating the Rho kinase and PAl-1 mRNA expression, up-regulating the eNOS mRNA expression and blood lipid, resisting atherosclerosis, inhibiting the Rho kinase and PAl-1 mRNA expression and increasing the Rho kinase and eNOS mRNA expression, which may be related to its mechanisms.

      摘要:Objective: To investigate the effect of Jiangya Baolan tablets against left ventricular hypertrophy (LVH) in spontaneously hypertensive rats (SHR). Method: The 12-week-old male SHRs were randomly divided to the SHR group (0.5% CMC), the enalapril group (0.6 g · kg-1) and the Jiangya Baolan tablets group (0.02 g · kg-1). In addition, 10 12-week-old normotensive Wistar Kyoto (WKY) rats were included into the normal group (0.5% CMC). All of them were orally administered with drugs for 12 weeks. The rat tail sphygmomanometer was used to measure the systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean arterial pressure (MAP) of rat caudal arteries. The echocardiography was used to measure the left ventricular posterior wall thickness (LVPWT), interventricular septal thickness (IVST), left ventricular end-diastolic diameter (LVEDD), left ventricular end-systolic diameter (LVESD), left ventricular fractional shortening (LVFS), left ventricular end-systolic volume (LVESV), left ventricular end-diastolic volume (LVEDV), ejection fraction (LVEF) and stroke volume (SV). The cardiac index (CI) and left ventricular mass index (LVMI) were calculated by weighting the whole heart and left ventricle. The pathological image analysis system was used to measure the cardiomyocytes transection diameter (CTD) and observe the myocardial histopathology by hematoxylin and eosin (HE) staining. Result: Compared with the normal group, the SHR control group showed significant increases in the systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean arterial pressure (MAP) (P <0.01), obvious decreases in LVEDV, LVESV and SV (P <0.01), remarkable rises in CI, LVMI and CTD (P <0.01) and slight growths in LVST, LVPWT, LVFS and LVEF, but with no statistical significance. Compared with the normal group, the Jiangya Baolan tablets group showed notable decreases in LVFS, LVEF, CI, LVMI and CTD (P <0.05) and significant amelioration in the pathological changes of left ventricular myocardium, but with a slight antihypertensive effect that was obviously lower than that of enalapril and no significant difference with the SHR group. Conclusion: Jiangya Baolan tablets has the effect in ameliorating LVH in SHR.

      摘要:Objective: To screen active fractions of Siwu Tang for activating blood and removing stasis by comparing their effects on the blood rheology, function of blood coagulation and platelet system of the acute model rats. Method: After Siwu Tang was boiled with water, the system solvent extraction method was adopted to extract four fractions respectively. Totally 80 female SD rats were divided into eight groups:the control group, the model group, the Compound Danshen dropping pill group (0.15 g · kg-1), the Siwu Tang group (7 g · kg-1), the Petroleum ether extract group (7 g · kg-1), the Ethyl acetate extract group (7 g · kg-1), the N-butyl alcohol extract group (7 g · kg-1) and the water extract group (7 g · kg-1). Except for the normal group, the ice water bath and subcutaneous injection with adrenaline hydrochloride were applied in all of the rest groups to establish the acute blood stasis rat model, in order to compare different extracted fractions in the hemorheology of rats after the administration, prothrombin time(PT), thrombin time(TT), activate parts thrombin time(APTT), fibrinogen(FIB), platelet aggregation rate, plasma thrombox-ane B2(TXB2) and 6-keto-prostaglandin Fla(6-keto-PGFla). Result: Compared with the normal group, the model group showed significant decreases in the shear rate of whole blood viscosity, plasma viscosity, TT, PT, APTT and 6-keto-PGFla and significant increases in FIB, platelet aggregation rate, plasma TXB2.Compared with the normal group, ethyl acetate fractions of Siwu Tang can significantly reduce the shear rate of whole blood viscosity, plasma viscosity, significantly extend TT, PT, APTT, significantly reduce FIB, platelet aggregation rate, plasma TXB2, elevate 6-keto-PGFla;the Petroleum ether group can notably decrease the whole blood viscosity at a high shear rate (P <0.05), reduce TXB2 content, increase 6-keto-PGFla content (P <0.01);N-butyl alcohol fractions and water extracts can obviously reduce TXB2 and increase 6-keto-PGFla (P <0.01). Conclusion: Ethyl acetate fractions of Siwu Tang have the most effective effect in removing blood stasis.

      摘要:Objective: To investigate the anti-inflammatory and analgesic effect of essential oil from Chrysanthemi Indici Flos (EOCIF). Method: Totally 60 KM mice were divided into six groups, i.e. the normal group, the model group, the aspirin group (0.5 g · kg-1), EOCIF high, middle and low dose groups (0.8, 0.4, 0.2 g · kg-1). Auricle edema induced by dimethylbenzene, cotton pellet granuloma of the mice and foot edema induced by carrageen were used to detect the anti-inflammatory activity. Acetic acid writhing method and hot-plate method were used to study the analgesic activity. Result: Compared with the normal group, the model group showed notable auricle edema and cotton pellet granuloma and significant increase in writhing frequency, with significant statistical difference (P <0.05). Compared with the model group, EOCIF high, middle and low dose groups showed obvious inhibitory effects in dimethylbenzene-induced auricle edema, cotton pellet granuloma and carrageen-induced foot edema and significant decrease in writhing frequency, with significant statistical difference (P <0.05). Besides, after the administration, compared with the model group, EOCIF high, middle and low dose groups showed obvious increase in pain threshold in mice (P <0.05). Conclusion: The EOCIF Chrysanthemi Indici Flos shows stronger anti-inflammatory and analgesic activities.

      摘要:Objective: To observe the effect of gastrodin infused through abdominal aorta on spinal cord microcirculatory blood flow (SCMBF) during the spinal cord ischemia-reperfusion injury (SCIRI). Method: Totally 60 healthy New Zealand rabbits were randomly divided into the normal control group, the model group, gastrodin low, medium and high dose groups (25,50,100 mg · kg-1). Except for the normal group, the SCIRI model was created by clamping infrarenal aorta. The changes in SCMBF were monitored before ischemia and at 15,30,60,120 min after the reperfusion. The neurologic function score (NFS) was evaluated before ischemia and at 6, 12, 24 h after the reperfusion. The level of serum neuron-specific enolase (NSE), malondialdehyde (MDA), reactive oxygen species (ROS), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), total anti-oxidation capacity (T-AOC) and the pathological changes in spinal cord tissues were monitored before the ischemia and at 45,60, 120 min after the reperfusion. Result: Compared with the model group, the model group showed significantly higher NSE, MDA and ROS (P <0.01) and lower GSH-Px, SOD and T-AOC (P <0.01) after ischemia. NSE, MDA and ROS were significantly lower while GSH-Px, SOD and T-AOC were distinctly higher in the gastrodin group after ischemia than that in the model group (P <0.01). The SCMBF and pathological damage in spinal cord in the model group were obviously more serious than that in the gastrodin group (P <0.01),but NFS recovered rapidly in the gastrodin group during reperfusion than that in the model group (P <0.01). Conclusion: The intra-aortic gastrodin infusion could distinctly alleviate spinal cord microcirculatory dysfunction by infusing gastrodin through abdominal aorta and improving the anti-oxidation ability of spinal cord during SCIRI.

      摘要:Objective: To study the anti-aging effect and mechanism of Qing'e prescription (QEP) extract on aging mice induced by D-galactose. Method: Seventy-two KM male mice were equally divided into six groups randomly:control group, model group, vitamin E group, and low-(0.4 g · kg-1), middle-(0.8 g · kg-1), and high-(1.6 g · kg-1) doses of QEP extract groups. To establish the subacute aging model, all the groups except control were subcutaneously injected with D-galactose at the back of cervical region daily. The control group and model group were intragastrically administered with 0.5% sodium carboxymethyl cellulose solution. The vitamin E group, low-, middle-, and high-doses of QEP extract groups received consecutive Vitamin E and low-, middle-, and high-doses of QEP extract prepared in sodium carboxymethyl cellulose solution, respectively, for six weeks. After the treatment, the anti-aging and anti-fatigue effects were evaluated by conducting the rotarod test, and measuring the contents of glutathione(GSH), malondialdehyde (MDA), total antioxidant capacity (T-AOC), nitric oxide (NO) and nitric oxide synthase (NOS) in liver, urea nitrogen (BUN) and lactate dehydrogenase (LDH)in serum together with other biochemical indexes. Moreover, the expression of Manganese superoxide dismutase(MnSOD) and Peroxidase-3(Prdx-3)mRNA in hippocampus was analyzed by qPCR to evaluate the antioxidant level. Result: Rotarod test showed that model group mice was easier to fall from the rotarod (P< 0.01) in comparison with the control group. Compared with the model group, all the doses of QEP extract could significantly prolong the residence time of mice on the rod (P <0.05, P <0.01). Biochemical tests showed that serum levels of BUN, MDA, NO, NOS in model group were increased remarkably when compared with that in the control group (P <0.05). By contrast, serum LDH level was reduced (P <0.05). After treated with QEP extract, all the aforementioned indexes were improved (P <0.05, P <0.01). In liver tissues, D-galactose significantly decreased GSH and T-AOC levels (P <0.01) while elevated MDA, NO, and NOS levels (P <0.05, P <0.01) in model group mice.QEP extract could reverse those parameters markedly (P <0.05, P <0.01). Real-time quantitative PCR results displayed that the hippocampal MnSOD mRNA expression level of model group was lower than that in the control group (P <0.05). After treatment of QEP extract, both of the expressions of MnSOD and Prdx-3 mRNA were up-regulated (P <0.05,P <0.01). Conclusion: QEP extract demonstrated significant anti-aging and anti-fatigue effect by enhancement of antioxidant capacity, suggesting its possible application as a substitute for QEP.

      摘要:Objective: To explore the protective effect of Lini Semen water extract and alcohol extract on D-GaIN induced acute liver injury in mice. Method: The mice acute liver injury model was induced by D-GaIN.90 mice were divided into 9 groups, blank group, model group, Bifendate pills group, Lini Semen water extract and alcohol extracts, low, middle, high dose group. The blank group and the model group were given volume saline, the other groups were given corresponding drugs dose by gavage once daily for 10 d.At the ninth day of administration,according to the dosage of 600 mg · kg-1 intraperitoneal injection of 10% D-GaIN amine solution was given toestablish the modle. 24 h after the last administration blood samples were collected to measure serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activity.Malondialdehyde(MDA), glutathione(GSH), superoxide dismutase(SOD) in liver tissue and liver index were measured. The hepatic histological changes were observed by HE with light microscope. Result: Compared with the normal group, model group significantly increased ALT, AST, MDA levels (P <0.05, P <0.01), decreased SOD and GSH activity (P <0.01), hepatic tissue structure was not clear, dissolved necrosis of liver cells were obviously;observed.Compared with the model group, the Lini Semen water extraction high dose and alcohol extraction low dose groups could increase the activity of serum SOD (P <0.05), decrease the level of MDA (P <0.01). Lini Semen treatment group decrease serum ALT,AST and the liver index,and improved hepatic tissue structure, reduced the number of necrosis of liver cells, liver cell regeneration was observed.The high dose group improved obviously. Conclusion: Lini Semen extract has certain protective effect on acute liver injury induced by D-GaIN in mice.

      摘要:Objective: To investigate the effect of Dantian Jingshu capsule onhemorheology of vertebral artery type cervical spondylosis (CSA) model rabbits. Method: Seventy rabbits were randomly divided into 7 groups:normal group, model group, Dantian Jingshu capsule group(0.36,0.72,1.44 g · kg-1), positive control group(Jing Fukang granules 1.0 g · kg-1 and flunarizine 0.001 g · kg-1). Rabbits were injected with 775 injection(tissue sclerosing agent) in the left side of cervical vertebrae to make animal model of CSA. To observe the effects of Dantian Jingshu capsule on hemorheology[whole blood viscosity, plasma viscosity, whole blood viscosity reduction, hematocrit(HCT), erythroeyte aggregation index(EAI), red cell rigidity index(ERI), red cell deformation index (EDI)and fibrinogen] of the rabbits by intragastric administration after two weeks. Result: Compared with normal group, the hemorheology index of model group (blood viscosity, plasma viscosity, HCT, whole blood viscosity reduction, EAI) were obviously decrease. So there was significant differences between normal group and model group(P <0.01). Compared with model group, blood viscosity and aggregation of red-cells were obviously decrease and erythrocyte deformability was increased in Dantian Jingshu capsule group(P <0.01). Conclusion: Dantian Jingshu capsule could reduce the CSA model rabbits erythrocyte aggregation and reinforce the ability of erythocyte deformability so that improve its blood viscosity. Dantian Jingshu capsule can improve the CSA model rabbits' symptom of cerebral ischaemia.

      摘要:Objective: To investigate the protective effect of Ligustrazine, Shenfu injection and their combined pretreatment on heat shock protein 70 (HSP70), p38 mitogen activated protein kinase (p38 MAPK) expression and antioxidant ability at left ventricular anterior wall ischemia myocardial tissues of rats with myocardial ischemia reperfusion injury. Method: The 50 Wister rats were randomly divided into 5 groups: sham operation group (sham) and cerebral ischemia reperfusion group (IR): normal saline ip for 3 d; Ligustrazine injection-pretreated group (LI): Ligustrazine injection ip for 3 d (20 mg · kg-1 · d-1) before surgery; Shenfu injection preconditioning group (SFI): Shenfu injection ip for 3 d (10 mg · kg-1 · d-1) before surgery; Combined Ligustrazine and Shenfu Injection-pretreated group (LI+SFI): Ligustrazine (20 mg · kg-1 · d-1) + Shenfu Injection (10 mg · kg-1 · d-1) ip for 3 d before surgery. The myocardial ischemia reperfusion injury model in rat was prepared. The myocardial tissue ischemia lasted for 30 min, myocardial tissue reperfusion lasted for 120 min, myocardial tissues at left ventricular anterior wall ischemia position were collected to prepare 10% homogenate supernatant. The superoxide dismutase (SOD) activity was determined by xanthine oxidase. Glutathione peroxidase (GSH-Px) activity was determined by dithio dinitrotoluene. The immunohistochemical streptavidin peroxidase (S-P) method was used to detect heat shock protein 70 (HSP70) and p38 mitogen activated protein kinase (p38 MAPK) protein expressions at left ventricular anterior wall myocardial tissues. Result: IR group showed significantly lower SOD, GSH-Px activity(P <0.01) and higher HSP70, p38 MAPK positive expression than sham group (P <0.01). Compared with IR group, the SOD activity increased significantly (P <0.01),and the activity of GSH-Px (P <0.05), HSP70 was significantly increased (P <0.01), the positive expression of p38 MAPK protein were significantly decreased (P <0.01) in LI group. Compared with IR group, SFI group showed increases in SOD, GSH-Px, HSP70 (P <0.05), and decrease in expression of p38 MAPK (P <0.05). Compared with IR group, SOD, GSH-Px activity increased significantly (P <0.01), HSP70 was significantly increased (P <0.01), the positive expression of p38 MAPK protein were significantly decreased (P <0.01) in LI+SFI group. Compared with LI group (P <0.05), SFI group (P <0.01), LI+SFI group showed increase in SOD activity. Compared with SFI group, LI+SFI group showed increases in GSH-Px activity(P <0.05) and positive expression of HSP70 (P <0.05) and decrease in the positive expression of p38 MAPK (P <0.05). Conclusion: Pretreatment with Ligustrazine, Shenfu Injection could antagonize myocardial ischemia reperfusion injury, particularly with their combination. The mechanism may be correlated with increase activity in SOD, GSH-Px, increase expression of HSP70 and inhibition of p38 MAPK expression.

      摘要:Objective: To study the effect of extracts from Clematidis Chinensis Radix et Rhizoma in treating diabetic nephropathy in rats. Method: Totally 50 SD rats were randomly divided into the normal group, the model group, the Captopril group (10 mg · kg-1), Clematidis Chinensis Radix et Rhizoma extracts high and low dose groups (500, 250 mg · kg-1), with 10 rats in each group. Except for the normal group, the rest groups were intraperitoneally injected with streptozotocin (STZ) to prepare the diabetic rat model and then orally given corresponding drugs, qd, for 12 weeks. The effect of extracts from Clematidis Chinensis Radix et Rhizoma on urea nitrogen(BUN), serum creationine(SCr), creatimine clearance rate(CCr), urinary albumin, urinar protein excretion rate(UAER), kidney hypertrophy index, fasting blood glucose, triglyceride(TG), total cholesterol(TC), low density lipoprotein cholesterol(LDL-C), high density lipoprotein cholesterol(HDL-C), interleukin-6(IL-6), malondialdehyde(MDA), superoxide dismutase(SOD), glutathion peroxidase(GSH-Px) and Renal tissue pathomorphological changes. Result: Compared with the model group, the model group showed notably higher serum BUN, SCr, CCr, IL-6, urinary albumin, UAER, kidney hypertrophy index and fasting blood glucose, TG, TC, LDL-c and MDA and lower HDL-C, SOD and GSH-Px, with notable statistical difference (P <0.05, P <0.01). In pathological tissues, renal glomerular basement membranes and renal tubule were thickened and swollen. Compared with the model group, extracts from Clematidis Chinensis Radix et Rhizoma showed remarkably lower BUN, SCr, CCr, IL-6, urinary albumin, UAER, kidney hypertrophy index and FBG (P <0.05), TG, TC and LDL-C (P <0.05), renal MDA content (P <0.05) and renal glomerular basement membrane and renal tubule thickening and swelling and higher HDL-C (P <0.05) and SOD and GSH-Px (P <0.05). Conclusion: Extracts from Clematidis Chinensis Radix et Rhizoma has a significant inhibitory effect on renal injury induced by diabetic nephropathy.

      摘要:Objective: To study the hypoglycemic effect of wild Ligularia schmidtii(LS)picked in Changbai Mountain at different times by detecting the fasting blood glucose, insulin, liver glycogen, muscle glycogen and glucose tolerance in mice. Method: Totally 100 Kunming mice were randomly divided into the normal group, the model group, the metformin hydrochloride group (125 mg · kg-1) and LSMM groups (1.25 g · kg-1) picked at different times (including plants picked on May 15, May 30, June 15, June 30, July 15, July 30, August 15), with 10 mice in each group. Except for the normal group, all of the rest groups were intraperitoneally injected with 200 mg · kg-1 sterilized saline water and 1% alloxan solution for 72 h to establish the model. After the modeling, they were orally administered with drugs (0.01 mL · g-1). The fasting blood glucose value, serum insulin content, liver glycogen content, muscle glycogen content and glucose tolerance of mice were measured with Appropriate kits. Result: Compared with the normal group, the model group showed significant increase in the fasting blood glucose and decreases in insulin, liver glycogen and muscle glycogen content in mice at different time points, all with significant statistical differences (P <0.05). Compared with the normal group, LS picked at different times could decrease the fasting blood glucose and increase liver glycogen and muscle glycogen content in diabetic mice to varying degrees and alleviate the damaging effect of alloxan on pancreatic islet β cells in mice to some extent. Besides, compared with the normal group, it could also enhance glucose tolerance in normal mice to varying degrees, with the most significant hypoglycemic effect in LS picked in June (P <0.05) as evidenced by its blood glucose decrease by 35.66% in LS picked on June 30. Conclusion: LS picked at different times have the hypoglycemic effect on both diabetic mice and normal mice.

      摘要:Objective: To observe the curative efficacy of Busui Shengxue granule in treating chronic aplastic anemia (CAA) and its effect on protein expressions of Ras homologous members (Rho)-guanosine triphosphate (GTPase), such as cell division cycle 42 (CDC42), Ras-related C3 botulinum toxinsubstrate (Rac) and Ras homologous member A (Rho A) in bone marrow mononuclear cells (BMNC). Method: Totally 134 CAA patients were randomly divided into the test group (67 cases) and the control group (67 cases), and another 20 healthy volunteers were assigned into the normal control group. The test group was treated with Busui Shengxue granule, and the control group was treated with Zaizao Shengxue tablet;both groups were treated for 6 months. The changes in the clinical curative effect, score of traditional Chinese medicine (TCM) clinical symptoms, curative effect of TCM syndromes, peripheral blood and bone marrow hyperplasia degree were observed. The protein expressions of Rho-GTP-related enzymes (CDC42, Rho A and Rac) in BMNC of CAA patients were detected by Western-blot. Result: The test group showed better clinical curative effect and TCM syndrome curative effect than the control group (P <0.05). After the treatment, the average score of TCM clinical symptoms and peripheral blood of the two groups were significantly improved (P <0.05). The average score of TCM symptoms, white blood cell count (WBC) and hemoglobin (HGB) of the test group was better than that of the control group (P <0.05), but with no significant difference in the platelet count (PLT) between the two groups. The two groups showed no significant difference in the improvement of the bone marrow. Before the treatment, both groups showed lower relevant protein expressions of CDC42, Rho A and Rac the normal control group (P <0.05),whereas after the treatment,both groups showed significant increases in the relevant protein content of CDC42 and Rho A (P <0.05), but with no notable change in Rac expression in the test group compared with that before the treatment. After the treatment, the expressions of CDC42, Rho A and Rac in the two groups were still lower than that in the normal control group (P <0.05), and the expressions of CDC42 and Rac in the test group were better than that in the control group. Conclusion: Busui Shengxue granule has a significant effect in treating CAA, and can improve the hematopoietic function of bone marrow of patients with CAA. The abnormal expressions of Rho A, CDC42 and Rac in BMNC of CAA patients suggested that Rho-GTPases may be closely related with the pathogenesis of CAA. Busui Shengxue granule may improve hematopoietic stem cells, hematopoietic microenvironment and hematopoietic function of bone marrows by regulating the expressions of Rho-GTPases-CDC42, Rho A and Rac.

      摘要:Objective: To observe the effect of Danhong injection in relieving blood stasis and thrombosis of patients with chronic renal failure (CRF) maintenance hemodialysis. Method: Totally 130 patients with CRF maintenance hemodialysis were divided into the observation group (65 cases) and the control group (65 cases) by the random numbers table method. Patients in the control group took Dipyridamole tablets, 50 mg/time, 3 times/day for continuing administration. In addition to the therapy of the control group, patients in the observation group was given 30 mL Danhong injection and 100 mL 0.9% saline solution for slow intravenous dripping, 1 times/day. One treatment course was ten days, once a month. The total course lasted for 3 months. The blood stasis syndrome was scored at the first, second and third months before and after the treatment. The changes in blood rheology indexes, fibrinogen (FIB) level, D-dimmer (D-D), platelet granule membrane protein (CD62 P), lysosomes platelet granule membrane protein (CD63), thrombin sensitive protein (TSP), thromboxane (TXB2), 6-Ketone-prostaglandin F1α(6-Keto-PGF1α) were detected before and after the treatment. Result: The blood stasis syndrome scores in the observation group were lower than that in the control group at the second and third months after the treatment (P <0.01). The blood stasis syndrome score in the observation group declined faster than that in the control group at the second and third months after the treatment (P <0.01). After the treatment, the whole blood viscosity (high, low), plasma viscosity and platelet aggregation rate in the observation group were higher than that in the control group (P <0.01). The levels of TXB2,FIB and D-D in the observation group were lower than that in the control group (P <0.01), but with higher 6-Keto-PGF1α than that in the control group (P <0.01). The CD62 P and TSP in the observation group were less than that in the control group (P <0.05), with no statistical significance in differences in CD63 between both groups. Conclusion: Danhong injection can relieve blood stasis among patients with chronic renal failure maintenance hemodialysis, inhibit platelet activation, enhance microcirculation, reduce high blood coagulation state, and prevent the thrombosis.

      摘要:Objective: To discuss the efficacy of Bushen Tiaojing decoction in promoting ovulation among patients with polycystic ovarian syndrome (PCOS) and its effect on insulin resistance (IR) and serum leptin (LP). Method: Totally 114 patients were divided into the control group (57 cases) and the observation group (57 cases) by the random number table. Patients in the control group received ethinylestradiol and cyproterone acetate tablets and clomiphene citrate. Besides the therapy of the control group therapy, patients in the observation group were also given Bushen Tiaojing decoction. The treatment courses for both groups lasted for 3 menstrual cycles. The endometrial thickness, endometrium types (A, B, C), cervical mucus scores, number of eggs ovulated, menstruation recovery, encyesis were observed. The insulin resistance index (HOMA-IR), leptin (LP), luteinizing hormone (LH), serum testosterone (T), estradiol (E2) and follicle stimulating hormone (FSH) were detected. Result: After the treatment, the endometrial thickness and cervical mucus scores in the observation group were higher than that in the control group (P <0.01). After the treatment, type A endometrium in the observation group was higher than that in the control group (P <0.05). The menstruation recovery, number of eggs ovulated, encyesis in the observation group were higher than that in the control group (P <0.05). T, LH, HOMA-IR and LP levels in the observation group were lower than that in the control group (P <0.01). Conclusion: Combined with the western therapies, Bushen Tiaojing decoction can improve endometrial thickness and endometrial morphology, promote menstruation recovery, induce ovulation and enhance encyesis. Its mechanism may be correlated with the regulation of endocrine and metabolism.

      摘要:Objective: To observe the clinical efficacy of Mahuang Lianqiao Chixiaodou decoction combined with acupuncture and moxibustion in treating allergic rhinitis (AR) and its effect in regulating serum Thl/Th2 and IgE level. Method: Totally 121 patients with severe persistent AR were divided into the control group (60 cases) and the observation group (61 cases) according to the random number table method. Patients in the control group was given budesonide aerosol inhalation, 200-1 600 μg · d-1, once in the morning or twice separately in the morning or at night, and desloratadine tablets, 10 mg · d-1. Patients in the observation group took Mahuang Lianqiao Chixiaodou decoction combined with acupuncture and moxibustion. The treatment course for both groups lasted for 14 days. The rhinobyon, rhinocnesmus, sneezing, shed tears and other relevant accompanying symptoms were scored. Patients' living quality was evaluated by rhino conjunctivitis quality of life questionnaire (RQLQ). And levels of serum total IgE (T IgE), γ-interferon (IFN-γ), interleukin-2 (IL-2), interleukin-4 (IL-4) and Interleukin-5 (IL-5) were detected before and after the treatment. Result: The scores of rhinobyon, rhinocnesmus, sneezing, shed tears and other relevant accompanying symptoms in the observation group were less than that in the control group (P <0.05 or P <0.01). After the treatment, the total scores of non-nasocular symptoms, nasal symptoms, ocular symptoms and RQLQ in the observation group were also lower than that in the control group (P <0.05 or P <0.01). The levels of IFN-γ and IL-2 in the observation group were higher than that in the control group, but with lower IL-5 and T IgE levels than that in the control group (P <0.01). Conclusion: Mahuang Lianqiao Chixiaodou decoction combined with acupuncture and moxibustion can effectively control clinical symptoms of patients with severe persistent AR and improve their quality of life. Its mechanism of action may be achieved by regulating Thl/Th2 cytokines and reducing serum IgE level.

      摘要:Objective: To observe the clinical effect of the comprehensive therapy of intake of Jiawei Fuzi Lizhong decoction, application on Shenque acupoint and moxibustion in treating hepatocirrhosis ascites (spleen and kidney yang deficiency and water retention symptoms). Method: Totally 118 patients were randomly divided into the control group (58 cases) and the observation group (60 cases). The control group stayed in bed, took less sodium and limited liquid infusion to balance water, electrolyte and acid-base value, and received therapies for liver protection and diuresis. Based on the therapies of the control group, the observation group additionally received the comprehensive traditional Chinese medicine therapy of intake of Jiawei Fuzi Lizhong decoction, application on Shenque acupoint and moxibustion. Both groups were treated for four weeks. Before and after the treatment, the spleen and kidney yang deficiency and water retention symptoms were scored.The liver function was evaluated by Child-Pugh.The levels of glutamic-pyruvic transaminase (ALT), glutamic oxalacetic transaminase (AST), total bilirubin (TBIL) and serum albumin (ALB) were detected.Ascites was detected by B ultrasonic.And weight and abdominal perimeter were measured. Result: According to Ridit analysis, the clinical effect in the observation group was higher than that in the control group (P <0.05).The effect in TCM syndromes in the observation group was also higher than that in the control group. After the treatment, the levels of ALT, AST and TBIL in the observation group were lower than that in the control group (P <0.05 or P <0.01), but with no statistical significance in the ALB level in the inter-group comparison.The observation group showed higher spleen and kidney yang deficiency and Child-Pugh scores (P <0.01) and 24-hour urine volume (P <0.01) but lower weight and abdominal perimeter than that in the control group (P <0.05). Conclusion: Based on the conventional western medicine therapy, the comprehensive traditional Chinese medicine therapy can ameliorate the function of patients with hepatocirrhosis ascites (spleen and kidney Yang deficiency and water retention symptoms), relieve clinical symptoms and signs and promote the ascites regression, with superior comprehensive efficacy comprehensive traditional Chinese medicine syndrome score than the pure western medicine therapy.

      摘要:Objective: To observe the efficacy of Kangxian'gan decoction in treating liver fibrosis caused by chronic hepatitis B and its effect on serum leptin and transforming growth factor-β1(TGF-β1). Method: Totally 106 patients were divided into the control group (54 cases) and the observation group (52 cases) by the random number table. Both groups received Adefovir Dipivoxil tablets, 10 mg/time, 1 time/day. Patients in the control group took Fuzheng Huayu capsules, 1.5 g/time, 3 times/day. Patients in the observation group took Kangxiangan decoction, 1 dose/day. The treatment courses of both groups lasted for 48 weeks. The hyaluronic acid (HA), laminin (LN), serum levels of procollagen type III (PCIII), serum level of collagen type IV (IV-C), aspartate aminotransferase (AST), alanine aminotransferase (ALT), total bilirubin (TBIL), albumin (ALB), HBV-DNA, TGF-β1and leptin levels were detected. The HBV serological markers (HBV-M) were detected by the euzymelinked immunosorbent assay before and after the treatment. The portal vein diameter, splenic vein diameter, thickness of spleen and blood flow velocity were detected by the color Doppler ultrasound. Result: According to the Ridit analysis, the clinical efficacy in the observation group was higher than that in the control group (P< 0.05). After the treatment, the serum HA, PCⅢ, LN and IV-C levels in the observation group were less than that in the control group (P< 0.01). After the treatment, ALT, AST and TBIL levels in the observation group were lower than that in the control group (P< 0.01), but with higher ALB level than the control group (P< 0.01). The HBeAg negative conversion rate in the observation group was 29.6%, while that in the control group was 25%, with no statistical significance in their difference. The HBV-DNA, leptin and TGF-β1 levels in the observation group were lower than that in the control group after the treatment (P< 0.01). The portal vein diameter, spleen vein diameter, thickness of spleen in the observation group were lower than that in the control group, but with faster blood flow velocity than the control group (P< 0.01). Conclusion: In addition to the antiviral treatment, Kangxian'gan decoction can resist viruses, protect the liver function, improve the color doppler ultrasound of liver and spleen, reduce serum leptin and TGF-beta 1 levels. It has the inhibitory effect on the development of liver fibrosis caused by Chronic hepatitis B, with a better clinical efficacy than Fuzheng Huayu capsules.

      摘要:Objective: To observe the effect of Jinlong Guben Bufei Jianpi mixture in treating asthma in the remission stage and the asthma control test (ACT) scores. Method: Inthis study, two hundred and thirty-nine patients with bronchialasthma in the remission stage (insidious sputum in lungs andlung and spleen qi deficiencysyndromes) were collected andrandomly divided into the traditional Chinese medicine(TCM)treatment group (121 cases) and the blank control group (118 cases). The cases in the TCM treatment group took Jinlong Guben Bufei Jianpi mixture for 4 months, while the cases in the blank control group were given no medical intervention.All of them were followed up for 12 months. During the study, ACT scores, asthma ACT control, TCM syndrome scores, pulmonary ventilation functionand other indexes were observed. All data was statistically analyzed by using SAS 8.1 software. Result: According to ACT scores for the patients, the treatment group was significantly higher than the blank control group (P <0.05). There were good correlations among TCM syndrome scores,forced expiratory volume in one second(FEV1)improvement and ACT scores. Conclusion: Jinlong Guben Bufei Jianpi mixture can relieve asthma symptoms of in the remission stage, andincrease ACT scores, reduce acute attacks, with obvious curative effect.

      摘要:Objective: To investigate the protective effect of Paidu Baoshen pills and valsartan on residual renal function (RRF) of patients with peritoneal dialysis. Method: Totally seventy-six cases treated with peritoneal dialysis in our hospital between January 2012 and March 2014 were included and divided into two groups by using the hidden random number table method. The 38 cases in the control group were given valsartan and basic western treatment beside the peritoneal dialysis, while the 38 patients in the study group took Paidu Baoshen pills developed by our hospital on beside the therapies of the control group. The treatment lasted for 12 weeks (four weeks in one course of treatment). Their residual renal function indexes after the treatment, dialysis related indexes before and after the treatment and other relevant indexes of the two groups were observed,the curative effect was evaluated after three courses of treatment. Result: At the 4th, 8th and 12th week after the dialysis, both groups showed reduction in 24 h urine volume, and the study group showed significantly lower RRF and 24 h urine volume than the control group (P <0.05),at the 12th week after the treatment, both groups showed obviously lower urea nitrogen (BUN) than before the treatment, while the study group showed notably lower serum creatinine (SCr) than before the treatment (P <0.05),the study group showed significantly lower BUN and SCr than the control group (P <0.05),the study group shower significantly higher Hb and Alb before the treatment (P <0.05), with a higher rate than the control group and significant differences between groups (P <0.05),both groups showed significant reduction in the entire section of parathyroid hormone (iPTH) compared with that before the treatment (P <0.05), with no significant difference in the decrease rate between the groups (P <0.05). The study group showed a total effective rate of 71.05%, which was significantly higher than the control group (P <0.05). Conclusion: Paidu Baoshen pills combined with valsartan can delay the decreases in RRF and urine volume of patients with peritoneal dialysis and relieve anemia, with a protective effect on residual renal function of patients with peritoneal dialysis.

      摘要:Objective: To study the clinical efficacy of Zisui Shengxue capsule in the treatment of chronic aplastic anemia. Method: Totally 64 patients with chronic aplastic anemia were randomized into the control group (treated with cyclosporin A and andriol) and the observation group (given Zisui Shengxue capsule beside the therapy for the control group) according to the random number method, with 32 cases in each group. The clinical efficacy, hemoglobin (Hb), white blood cell (WBC), platelet (PLT) before and after the treatment, and the adverse reactions during the treatment in the two groups were observed and compared. Result: Compared with the control group, the total effective rate significantly increased in the observation group (81.25% VS 56.25%, P <0.05).Compared with the control group, the total effective rate for TCM syndromes significantly increased in the observation group (96.875% VS 78.125%, P <0.05).Compared with the control group, the levels of Hb (80.32±21.27 VS 69.78±20.13) g · L-1, WBC (3.92±1.11 VS 3.14±1.08)×109/L and PLT (46.37±14.32 VS 31.08±13.82)×109/L significantly increased in the observation group after the treatment (P <0.05).Compared with the control group, adverse reactions, such as hirsutism (6.25% VS 28.125%), hands shaking (12.50% VS 37.50%) and gingival hyperplasia (6.25% VS 25.00%) significantly reduced in the observation group (P <0.05). Conclusion: Zisui Shengxue capsule shows significant efficacy and less adverse reactions in treating chronic aplastic anemia, and can obviously improve the patients' hemogram indexes. It is worthy of clinical promotion.

      摘要:Objective: To systematically evaluate the efficacy and safety of sodium ozagrel combined with Dengzhan Xixin preparations in treating cerebral infarction (CI). Method: Randomized controlled trails (RCT) about sodium ozagrel combined with Dengzhan Xixin preparations in the treatment of CI were collected by retrieving from PubMed, EMbase, Cochrane Library, FMJS, CNKI, VIP and Wanfang database. The methodological quality of the included studies was evaluated, and a Meta-analysis was made for them by Rev Man 5.3.4 statistical software. Result: A total of 22 RCT were included, involving 2 198 patients. The results of Meta-analysis showed that sodium ozagrel combined with Dengzhan Xixin preparations could significantly improve the total effective rate [OR=3.76, 95% CI (2.84, 4.97), P< 0.000 01], obvious effective rate [OR=2.07, 95% CI (1.71, 2.49), P< 0.000 01] and neurological deficit scores [MD=-5.08, 95% CI (-5.73,-3.53), P< 0.000 01], with statistical significance in their differences. Conclusion: Sodium ozagrel combined with Dengzhan Xixin preparations is efficient in the treatment of CI, with no significant adverse reaction. However, restricted by the low methodological quality of the trials enrolled in the study, its efficacy shall be further confirmed by a high-quality RCT.

      摘要:Tripterygium wilfordii, as a type of Tripterygium specie of Celastraceae genus, has a long history and has been widely applied in treating rheumatoid arthritis (RA). However, T. wilfordii has a high toxicity and serious side effects in humans, such as in reproductive system, immune system and digestive system, which restricts its clinical application. Therefore, it is essential to study its attenuation and synergistic effects. In this study, literatures for T. wilfordii and its preparations in treating RA in recent years were explored to develop its attenuation process, improve formulations and define the compatibility with compound traditional Chinese medicines and the combination with western medicines and acupuncture. There had been T. wilfordii processing methods since the ancient times, mainly including cleansing, sheep blood processing and heat processing;and there are many new processing methods, such as microwave heating and bi-directional solid fermentation techniques, in recent years. All of them can achieve the attenuation and synergistic effects. There are many kinds of T. wilfordii preparations.Specifically, oral drugs have a higher toxicology through intestinal absorption, while mucosal drugs had a lower toxicology. Therefore, improved formulations can effectively reduce the irritation time in gastrointestinal tracts and increase the absorption and bioavailability. Common formulations include suppository, sustained release tablet and pill. According to the study, the combination with Glycyrrhizae can accelerate the metabolic turnover of triptolide and reduce the toxicity of T. wilfordii,the combination with Paeonia sterniana Fletcher in Journ can reduce liver injury.Besides, the combination with western medicines and acupuncture can also achieve the attenuation and synergistic effects. In this study, the T. wilfordii processing methods for the attenuation and synergetic effects were summarized in the expectation of promoting the effective application of T. wilfordii in clinical application.

      摘要:Aging refers to the reduction in the function of cells to remove metabolic wastes, which leads to the excessive accumulation of damaged proteins and organelles and then reduces the viability of organisms. Autophagy is a major metabolic pathway within cells, which can resolve damaged proteins and organelles for energy recycling and participate in various physiological and pathological processes related to aging. According to different ways of intracellular substrates in lysosomes, autophagy can be divided into macroautophagy, microautophagy and molecular chaperone-induced autophagy,and their processes are induction, nucleation, autophagy precursor extension, protein degradation and recycling. Because the signaling pathway of autophagy is partially overlapped with the regulatory effect on aging, autophagy level is closely associated with aging. The autophagy level reduces with the aging process, while the low autophagy level can accelerate the aging process,at the same time, the autophagy level can slow down the aging process. That is to say, autophagy can regulate the aging progress, which is also correlated with oxidative stress, inflammatory reaction and caloric restriction. In addition, although studies on autophagy and its regulatory effect on aging with traditional Chinese medicines are still at their infancy, they showed some conclusions in theoretical research and drug experiments and new ideas for further studies.

      摘要:In order to strengthen the research and development of Cordyceps nucleosides, multiple detection methods adopted for Cordyceps nucleosides at home and abroad were reviewed. Three detection technologies used in the analysis of Cordyceps nucleoside were selected, including high performance liquid chromatography (HPLC), capillary electrophoresis (CE) and thin layer chromatography scanning (TLCs) method. According to different detectors, HPLC was also classified and compared. The HPLC method is usually applied in the qualitative and quantitative analysis of nucleosides, with a high sensitivity, quick separation and high separation efficiency. Among them, the LC-MS method features a wider adaptability, strong separation capacity and low detection limit and can provide rich structure and molecular weight information of each component. The LC-MS method has been widely used not only in the detection of Cordyceps nucleosides, but also in intensive studies on Cordyceps nucleosides or other drugs containing nucleosides at the molecular level.

      摘要:Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease with a high incidence and disability rate.Currently,RA is deemed as one of major diseases threatening human health and life,but with no effective drug.In clinic,common drugs for RA included nonsteroidal anti-inlammatory drugs,disease modifying antirheumatic drugs and corticosteroids clinically.However,they have severe adverse reactions,which bring great pains to patients.Traditional Chinese medicine (TCM) feature multiple components and multiple-target effect,based on which TCM transdermal drug delivery preparations have advantages of low medicinal frequency,high compliance among patients,effective reduction of foods' impacts on drugs,convenience,safety and drug withdrawal at anytime.Transdermal drug delivery preparations have become a hotspot of studies because of their unique therapeutic effect and formulation superiority.In this dissertation,authors evaluated and discussed necessity,current situation and existing problems of TCM transdermal drug delivery preparations,and gave their own suggestions,such as strengthen the material base studies,ensure a good quality control of preparation,strengthen studies on the external excipients and the mechanism of external TCM preparations,strengthen the studies on the scientific use of TCM penetration enhancer,in expectation of providing a reference for research and development of TCM transdermal drug delivery system.

      摘要:Rehmanniae Radix polysaccharides are one of active ingredients from prescriptions containing Rehmanniae Radix, which are widely applied in clinic. In order to provide reference for further studies and development of Rehmanniae Radix polysaccharides and prescriptions containing Rehmanniae Radix, the advances in the chemical and pharmacological effects of Rehmanniae Radix polysaccharides in the past two decades were summarized in an all-round way. Literatures on the chemical and pharmacological effects of Rehmanniae Radix polysaccharides from domestic and foreign databases, such as CNKI, Wanfang data, PubMed, SpringerLink and ScienceDirect, were collected, analyzed and summarized. So far, seven rehmannia glutinosa polysaccharides with relative molecular weight of 103-105 were isolated from Rehamanniae Radix, and composed of such monosaccharide as rhamnose, glucose, galactose, mannose, xylose and arabinose. According to the pharmacological study, Rehmanniae Radix polysaccharides showed a good anti-oxidation ability to normal and pathological animals and further anti-aging and anti-tumor activities, and can reduce the blood glucose and lipid levels through several ways, enhance the immunity function of normal and pathological animals by influencing immune organs, immune cells and cytokines, promote the hematopoietic function of normal and pathological animals, directly or indirectly prevent and resist tumors, protect adipose-derived stem cells and promote their proliferation. Through various ways, they can induce bone marrow mesenchymal stem cells into neuron-like cells, which are very similar to normal neurons. They can also resist fatigue and anxiety. In the future, more studies shall be made on the chemical structure, toxicology, clinical pharmacology of Rehmanniae Radix polysaccharides, in order to define their mechanisms and lay a foundation for promoting their clinical application and safety.
