摘要:Objective:To develop a RP-HPLC method for the quantification of adenosine in Weigan Effervescent Tablets.Methods: The analysis was carried out on a Shimadzu C18 column(250 mm×4.6 mm,S-5 μm)and the mobile phase was a mitture of methanol-water(8∶92)with a flow rate of 1.0 mL· min-1.The detection wavelength was 260 nm,and the column temperature was 35 ℃.Results: The linear response ranges of adenosine was 4.3~33.8 μg(r=(0.999 7)),and the average recovery was 101.0%(n=9,RSD=1.4%).Conclusion: The method is quick,simple and reproducible for the determination of adenosine in Weigan Effervescent Tablets.

摘要:Objective: In order to evaluate the safety of Chinese medicine comprehensively, the development of pesticide residues determination in Chinese medicine by gas chromatography (GC) was summarized in this paper. Methods : Through analytic comparison, the developments of sample pretreatments were summarized systematically, including solid-phase extraction (SPE), gel permeation chromatography (GPC), supercritical fluid extraction (SFE), accelerated solvent extraction (ACE), matrix solid phase dispersion (MSPD), solid-phase micro-extration (SPME) and headspace analysis (HS). In addition, the comparison of gas chromatography detectors such as mass spectrometry (MS), electron-capture detector (ECD), flame photometric detector (FPD) and nitrogen–phosphorus detector (NPD) were also discussed. Results: Gas chromatography is a mature technology to accomplish pesticide residues determination in Chinese medicine. Conclusion: Gas chromatography and relevant new techniques are wildly used in safety evaluation of Chinese medicine and its preparations, which will be benefit for the international development and import of Chinese medicine.  
摘要:Objective:To develop a HPLC method for the determination of palmatine and berberine in Shulitong Granule.Methods:A Kromasil-C18(250 mm×4.6 mm,5 μm)column was used for the HPLC determination.The mobile phase consisted of acetonitrile-0.1 mol/L potassium dihydrogen phosphate(28∶72),and the flow rate was 1 mL/min.The detection wavelength was 345 nm.Results:A good linear relationship was constructed in the range of 0.016~0.080 μg(r=(0.999 8)),for Palmatine hydrochloride and in the range of 0.027~0.135 μg(r=(0.999 9)) for berberine hydrochloride. The average recovery of palmatine hydrochloride was 99.74%(RSD=2.67%,n=5),and that of berberine hydrochloride was 103.53%,RSD=1.48%(n=5).Conclusion:The developed method is simple,reproducible,specific and suitable for the quality control of Shulitong Granule.

摘要:Objective: To investigate the effect of Jianpi Huoxue Decoction on the expression of vascular endothelial growth factor (VEGF) in the femoral head of chicken with Steroid-induced Osteonecrosis of the Femoral Head(SONFH), as well as to explore the mechanism of prevention and treatment for SONFH. Methods: The animal model of SONFH was established with injection of methylprednisolone in chest muscle of chickens. 64 Leghorn chickens were randomly divided into normal control group, model group, lovastatin group and Jianpi Huoxue group, 16 in each group. At 8th and 16th weeks, blood samples were obtained to determine the level of serum total cholesterol(CHO)and triglycerid (TG), then femoral heads of both sides were taken off to observe the histopathological changes with hematoxylin and eosin staining and the expression of VEGF with immunohistochemical method. Results: At 8th and 16th weeks, in model group, relatively typical pathological changes were observed in the femoral head, the ratio of empty lacuna and fat area increased obviously, Compared with normal group, the level of serum CHO and TG increased and the expression of VEGF decreased significantly(P<0.05); Both lovastatin and Jianpi Huoxue Decoction could improve the morphological changes of steroid-induced femoral head necrosis at different levels, increased the expression of VEGF, decreased the level of serum CHO and TG(Compared with model group, P<0.05). Conclusions: Jianpi Huoxue Decoction can up-regulate the expression of VEGF, this may be one of the mechanisms for improvement in microcirculation in femoral head and the preventive and therapeutic effect for SONFH.

摘要:Objective: To observe the effects of Huannao Yicong Prescription(HYP) on learning and memory impairment,level of brain β-amyloid Protein (β-AP)and substance P(S.P)in aging rats. Methods: Morris water maze test was used to measure the spatial learning and memory of rats. 40 rats with learning and memory impairment were divided randomly into four groups (10 in each group):the model group (learning and memory impairment aged rats group),the high-dose HYP group (13.68 g crude drug/kg),the low-dose HYP group (6.84 g crude drug/kg),the positive control group (Hydrochloricdonepezil Tablets 0.45 mg/kg),besides,a normal aged rats group with 10 rats was set up,they were given tales doses of distilled water,all other groups were orally administered with drugs for 8 weeks. Then the spatial learning and memory of rats were measured again by Morris water maze. Rats were sacrificed after anesthesia and the brain homogenate was made for measuring the content of β-AP and S.P. The activity of superoxide dismutase (SOD) and the content of maleic dialdehyde (MDA) in serum was measured,the histomorphological changes in the brain were observed after hematoxylin eosin (HE) staining. Results: The latency and path length in water maze test in the model group were significantly prolonged. After 8 weeks of administration,the latency and path length in both of the HYP groups were significantly shortened than the model group,and the strategy in finding the platform was improved. Compared with the normal aged rats group,the content of MDA was significantly increased while the activity of SOD was significantly decreased in the model group. After 8 weeks of administration,compared with the model group,the level of β-AP was decreased while the S.P was increased,the activity of SOD was significantly increased(P<0.01)and the histomorphology in the brain was improved in the HYP groups. Conclusion: HYP can reduce the deposition of β-AP in the brain tissue,protect the integrity of histomorphology in the hippocampus,and thus improve the spatial learning and memory for aged rats.

摘要:Objective: To investigate the protective effects of water extract of Coptis chinensis and Evodia rutaecarpa on ethanol-induced acute gastric lesions in rats. Methods: In this study, animals were intraperitoneally pretreated with vehicle (normal saline) or Coptis chinensis and Evodia rutaecarpa water extract at 1050 (6 ∶1), 1800 (6 ∶6), 1050 (1 ∶6) mg·kg-1. Three hours later, gastric lesions were induced by topical application of 50% ethanol for 10 min. The length index of gastric lesions were determined by morphological analysis in a gastric intubatton model. The rat gastric transmucosal potential difference (PD) was recorded continuously by ex-vivo chamber technique. The pH value and H+ content of gastric perfusate were mesured by pH meter and pH-stat technique, respectively. Results: Compared with control, Coptis chinensis and Evodia rutaecarpa water extract (1 ∶6) accelerated PD recovery and reduced gastric morphologic lesions. Coptis chinensis and Evodia rutaecarpa water extract (1 ∶6) elevated H+ content and reduced pH value. Coptis chinensis and Evodia rutaecarpa water extract (6 ∶6) reduced gastric morphologic lesions. Conclusions: Coptis chinensis and E. rutaecarpa water extract (1 ∶6) showed effectively protective actions on ethanol-induced acute gastric lesions in rats, and the gastroprotective mechanisms maybe due to the strengthening action on gastric mucosal lining and the inhibition of acid backdiffusion. This investigation also indicated that E. rutaecarpa maybe have a critical role in gastroprotective effect.

Methods: The contents of Cort,ALD in blood were determined and the mRNA expression of CYP11B2 was evaluated with ELISA and RT-PCR in model group and control group. Results: Compared with control group, adenine could remarkedly decrease the contents of Cort,ALD in rat blood; and downregulate the mRNA expression of CYP11B2. Conclusion: Adenine can decrease the contents of Cort,ALD in blood and downregulate the mRNA expression of CYP11B2 in rats. It can decrease the synthesis of ALD by adjusting the ALD synthase. The results may expound the mechanism of kidney deficiency and diuresis induced by adenine from the angle of ALD synthase approach.

Conclusions: Coptis chinensis and E. rutaecarpa water extract (1 ∶6) showed effectively protective actions on ethanol-induced acute gastric lesions in rats, and the gastroprotective mechanisms maybe due to the strengthening action on gastric mucosal lining and the inhibition of acid backdiffusion. This investigation also indicated that E. rutaecarpa maybe have a critical role in gastroprotective effect.

摘要:Objective: To observe the effect of Radix Angelica and Rhizoma Ligusticum with different proportions (1 ∶0,2 ∶1,1.5 ∶1,1 ∶1,1 ∶1.5,1 ∶2,0 ∶1) extracted by different methods (by water、by ethanol、firstly by alcohol then by water) on platelet aggregation and prothrombin time (PT), thrombin time (TT). Methods: Take blood from carotid artery of rabbit and measure the rate of platelet aggregation induced by ADP and PT, TT to compare their different effects. Results: Radix Angelica and Rhizoma Ligusticum with different proportions and preparation methods showed certain effects of anti-platelet aggregation and antithrombin (P<0.01 or P<0.05). The effect on PT and TT enhanced along with the ratio of Radix Angelica gradually increases; The platelet aggregation effect was the weakest While the ratio of Danggui-Chuanxiong was 1.5 ∶1. Compared with other extracts, the solution extracted by water showed a weaker effect of platelet aggregation, while solution extracted by ethanol showed a stronger effect on PT than TT. Conclusion: The different traditional efficacy of Radix Angelica and Rhizoma Ligusticum with different proportions and preparation methods may be related with their different effects of anti-platelet aggregation and antithrombin, their intrinsic material basis and relevant mechanism are worthy of further study.

Methods: The single myocytes were enzymatically obtained from the guinea pig hearts and the action potentials were examined by whole-cell patchclamp technique. Results: 4×10-4 g/L CVB-D decreased action potential amptitude(APA)from 113.3±6.3 mV to 110.2±6.4 mV(n=6, P<0.01), prolonged action potential duration of repolarization 50%(APD50)from 616.3±68.0 ms to 657.4±79.5 ms(n=6, P<0.01),prolonged action potential duration of repolarization 90%(APD90)from 633.5±71.2 ms to 669.2±74.4 ms(n=6, P<0.01), Prolonged action potential duration of repolarization 50% to repolarization 90%(APD50-90)from 15.9±6.0 ms to 20.1±5.2 ms(n=6, P<0.01),and decreased decay slope of phase 3 (V3) from -2.6±0.8 V/s to -2.4±0.7 V/s(n=6, P<0.01).Compared with the baseline,4×10-3 g/L CVB-D makes the APD50-90 pass to increase trend gradually,but the variety trend of the V3 decrease gradually, other index sign varieties are consistent. Conclusion: CVB-D decreased APA and V3, and prolonged APD50,APD90,APD50-90,which may be its mechanism of positive inotropic effect and antiarrhythmic effect.

摘要:Objective:To study the hypoglycemic effect of Dahuang Zhechong Pill on alloxan-induced diabetic mice.Methods: We made up animal models of diabetic mice by intraperitoneal injection of 90 mg·kg-1 alloxan,the levels of glycated hemoglobin and lipids levels were measured.Mice were weighed before and after models made up.We anatomically separated the heart,liver,spleen,lung,kidney and thymus of the mice,weighed the organs and calculated the organ coefficient.Results: The level of blood glucose,glycated hemoglobin were increased,body weight and the weights of spleen,thymus were reduced after the models made up by introperitoneal injecting alloxan(90 mg·kg-1)on mice.Dahuang Zhechong Pill could reduce the level of blood glucose,glycated hemoglobin,increase level of high-density lipoprotein and improved the weight loss of the alloxan-induced diabetic mice,and increase the weights of spleen and thymus of immune organs.Conclusion: Dahuang Zhechong Pill has hypoglycemic effect on diabetic mice.

摘要:Objective: To study the hypoglycemic effect of Dahuang Zhechong Pill on alloxan-induced diabetic mice. 摘要:Objective:To study the pharmacokinetics of curcumin microcystin in rats.Methods: The microcystin was orally administrated.After carotid artery cannulation,blood samples were collected after administration of 0.08,0.25,0.5,0.75,1,2,3,4,5.5 h,and tested by HPLC.Results: The various doses of curcumin concentration-time curve is bimodal;Cmax1 and Cmax2 of each dose group are as follows: high-dose(0.90,1.20 mg/L),medium dose(0.43,0.46 mg/L),low dose(3.16,1.31 mg/L); AUC are 3.79,2.60,5.92 mg·h/L respectively.Cmax and AUC of low dose are greater than that of the high and medium doses.Conclusion: The Pharmacokinetics of curcumin were nonlinear process in rat,there may be the phenomenon of enterohepatic circulation.

摘要:Objective:To investigate the effect of Qingrehuayuziyin prescription on lymphocyte apoptosis and expression of Fas and Bcl-2 in peripheral blood in the patients with systemic lupus erythematosus(SLE).Methods: Eighty patients with SLE were selected and divided randomly into two groups,40 in the therapy group and 40 in control group respectively.Patients were got the treatment of Qingrehuayuziyin prescription combined hormone as therapy group and got hormone treating only as control group.After treating three months,the patients lymphocyte were collected and the rate of apoptosis were detected by flow cytometer.Meanwhile,the expression of Fas and Bcl-2 were analyzed by RT-PCR.Results: After 3 months treating,the rate of lymphocyte apoptosis was decreased significantly in therapy group compared with that in control group and with that in pre-treating(P<0.01).The expression of Fas and Bcl-2,compared with that in control group,were also decreased significantly(P<0.01).Conclusion: Qingrehuayuziyin prescription is useful for treating SLE by suppressing lymphocyte apoptosis,decreasing the level of Fas and Bcl-2 in peripheral blood and further modifying the immune status.

Results: The various doses of curcumin concentration-time curve is bimodal; Cmax1 and Cmax2 of each dose group are as follows: high-dose(0.90, 1.20 mg/L), medium dose(0.43, 0.46 mg/L), low dose(3.16, 1.31 mg/L); AUC are 3.79, 2.60, 5.92 mg·h/L respectively. Cmax and AUC of low dose are greater than that of the high and medium doses. Conclusion: The Pharmacokinetics of curcumin were nonlinear process in rat, there may be the phenomenon of enterohepatic circulation.  
摘要:Objective: In order to evaluate the safety of Chinese medicine comprehensively, the development of pesticide residues determination in Chinese medicine by gas chromatography (GC) was summarized in this paper. Methods : Through analytic comparison, the developments of sample pretreatments were summarized systematically, including solid-phase extraction (SPE), gel permeation chromatography (GPC), supercritical fluid extraction (SFE), accelerated solvent extraction (ACE), matrix solid phase dispersion (MSPD), solid-phase micro-extration (SPME) and headspace analysis (HS). In addition, the comparison of gas chromatography detectors such as mass spectrometry (MS), electron-capture detector (ECD), flame photometric detector (FPD) and nitrogen–phosphorus detector (NPD) were also discussed. Results: Gas chromatography is a mature technology to accomplish pesticide residues determination in Chinese medicine. Conclusion: Gas chromatography and relevant new techniques are wildly used in safety evaluation of Chinese medicine and its preparations, which will be benefit for the international development and import of Chinese medicine.
