      摘要:Objective To develop an UHPLC method of determining the characteristic chromatographic profile of Lonicera japonica Thunb for controlling the drug quality quickly and accurately. Methods The analysis was performed on an Agilent C18 column(50 mm?2.1 mm,1.8μm) with acetonitrile-0.2%phosphoric acid as mobile phase in gradient elution mode. The flow rate was 0.4 ml.min-1and the injection volume was 2.0μl with the detection wavelength of238 nm.The column temperature was set at 30℃. Results The fingerprint of Lonicera japonica Thunb was obtained within 21min and five peaks in the fingerprint were designed.14 samples were analyzedand the similarities were 0.915-0.987. Conclusion the UHPLC system enjoys the merits of a higher speed,sensitivity,and resolution, It can be used to control the quality of Lonicera japonica Thunb.

      摘要:Objective To compare the chemical compositions in the five kinds of Dendrobium. Methods Polysaccharide ,mannose and amino acid in the five Kinds of Dendrobium were determined and compared. Results The polysaccharide contents ,the amino acid contents and the mannose contents are remarkably different in the five kinds of Dendrobium. The content of polysaccharide in the Dendrobium officinnle is highedest in the five kinds of Dendrobium; The content of amino acid in the Dendrobium finbristun is higher than other four kinds of Dendrobium; the mannose contents of Dendrobium officinnle is much more higher than other kinds. Conclusion The quantity and contents of polysaccharide, amino acid and the mannose are remarkably different in the five kinds of Dendrobium. However, D1candidum officinnle has a higher quality than other four kinds of Dendrobium in terms of the same chemical compositions1.

      摘要:Objective: To use liquid chromatography-mass spectrometry techniques for qualitative analysis of alkaloids of Morus alba L. leaves and determine the content of total alkaloids in alkaloids in Morus alba L. leaves by HPLC-UV with pre-column derivatization. Method: The chromatographic separation was performed on a HiQSiL C18 column(250 mm×4.6 mm, 5 μm) using acetonitrile and 0.1% acetic acid aqueous solution under isocratic elution(50:50,V/V) at a flow rate of 0.8 mL.min-1. Morus alba L. leaves alkaloids was analyzed with 9-fluorenylmethyl chlorformate(FMOC-Cl) as the pre-column derivative agent. Detection wavelength was set at 254 nm with electrospray ionization source(ESI) and positive ion mode. And 1-deoxynojirimycin as the external standard to calculate mulberry leaf total alkaloid content. Result: Six alkaloid compounds were identified as DNJ,fagomine,3-epi-fagomine,DAB,Calystegine B2 and 2-O-β-Glc-DAB.The content of 3 batches test samples were more than 75% by external standard method. Conclusion: The method is sensitive, accurate, simple, and reliable. It can be used for quantitative analysis of Morus alba L. leaves alkaloids and preparation.

      摘要:Objective: To optimize extraction process of total flavonoids from Selaginellae Herba against hepatocarcinoma cells SMMC-7721. Method: Taking extraction time and times,the amount and concentration of ethanol as factors,result of orthogonal test was adopted to optimize weight coefficient ratio of amentoflavone and total flavonoids extraction amounts in which inhibition rate of human hepatoma SMMC-7721 cells was taken as an indicator,contents of effective components and pharmacodynamic indicators were analyzed with Pearson correlation coefficient by SPSS 17.0 software.The content of amentoflavone was determined by HPLC,mobile phase of acetonitrile-0.5% acetic acid water(33:67),detection wavelength 269 nm. Result: The best extraction conditions were as follows:reflux extracted 3 h with ten times the amount of 70% ethanol;weight coefficients of contents of amentoflaveone and total flavonoids were 0.7 and 0.3,indicating determination of effective ingredients and inhibition rate had a certain correlation. Conclusion: Taking contents of active ingredients instead of efficacy as the index of orthogonal test was scientific and reasonable;Optimized process was stable and feasible,it could provide new ideas for extraction of active ingredients from traditional Chinese medicine.

      摘要:Objective: To optimize separation and purification technology of icariin and icariside Ⅱ from Epimedii Folium by macroporous adsorption resin. Method: Taking adsorption capacity and desorption rate of icariin and icariside Ⅱ as indexes,macroporous resin models were screened by static adsorption-elution test,single factor tests were adopted to investigate adsorption and elution conditions. Result: DM301 macroporous resin was the most suitable one for purification of icariin and icariside Ⅱ,its optimum adsorption conditions were as follows:the concentration of sample solution 7.45 g·L-1,sample flow rate 3 BV·h-1;Elution conditions of icariin was eluting with 4 BV 45% ethanol at 1.5 BV·h-1;Elution conditions of icatiside Ⅱ was eluting with 5 BV 60% ethanol at 1.5 BV·h-1;Under these conditions,purities of icariin and icariside Ⅱ were 56.23% and 67.41%,respectively. Conclusion: These established process conditions could be adopted to separate and purify icariin and icariside Ⅱ at the same time with good stability,high yield and low production cost,which were suitable for industrial production.

      摘要:Objective: To optimize forming process of Dantao granules. Method: With ratio of soluble starch-dextrin,excipients consumption and ethanol concentration as independent variables,yield of qualified granules,dissolubility and critical relative humidity as dependent variables,forming process of Dantao granules was optimized by central composite design-response surface methodology and overall desirability method. Result: Optimal molding technology conditions were as follows:quality ratio of excipients-extract powder (2:1),proportion between starch-dextrin (1:1),with 75% ethanol as wetting agent;yield of qualified granules was about 85%,melting time was less than 1 min,critical relative humidity was about 58%. Conclusion: Optimized molding process was stable and reliable with high qualified rate,good dissolubility and strong anti-hygroscopicity,this research could provide basis for preparation and production environmental control of Dantao granules.

      摘要:Objective: To optimize extraction and membrane separation technology of active polypeptides in Trionycis Carapax. Method: With extract yield and extraction amount of total polypeptides as evaluation indexes,orthogonal test was employed to optimize extraction process of polypeptides in Trionycis Carapax by taking solid-liquid ratio,extraction time and times as factors.Single factor tests were adopted to investigate membrane separation technology of active polypeptides in Trionycis Carapax with membrane permeation flux or efficacy as index. Result: Optimum extraction technology was as following:smashed vinegar Trionycis Carapax through a 24 mesh sieve,extracted three times with six-fold the amount of water for 3 hours each time;Extract yield was 11.71% and extraction amount of total polypeptides was up to 2.442 g.Membrane separation conditions were as follows:The concentration of polypeptides in Trionycis Carapax extract 6%,operating pressure of ultrafiltration membrane with relative molecule weight cut off of 20 and 8 KD running at 0.18-0.21 and 0.8 MPa,respectively;Under these operating conditions,retention rate of protein was 83.67%,purity of polypeptides(relative molecular mass<8 000) in Trionycis Carapax achieved 57.62%. Conclusion: Membrane separation techniques could be used for preparation of active polypeptides in Trionycis Carapax.

      摘要:Objective: To optimize preparation process of baicalin nanocrystallines and investigate its stability. Method: Taking mean particle size and polydispersity index(PDI) as indicators,preparation process of baicalin nanosuspensions was optimized by single factor tests.Baicalin nanocrystallines was then collected by centrifugation and freeze-dried,morphology of obtained nanocrystals were analyzed,initial stability of nanosuspensions and its freeze-dried powder were also observed.Equilibrium solubility of coarse baicalin and nanocrystals was compared by shaking flask method. Result: Nanosuspensions were prepared by injecting 1 mL of baicalin solution in DMSO(50 g·L-1) into 20 mL of 0.5% lecithin solution by a high shear of 30 000 r·min-1 for 8 min under ice-water bath.These obtained nanocrystals showed an average particle size of 280.6 nm (PDI=0.047) and that of 583.6 nm (PDI=0.12) after freeze-drying,these nanocrystals were investigated as sphere-like particles;Particle size of nanosuspensions increased up to 672.0 nm after stored at room temperature for 30 d,but no significant change was found for freeze-dried powder of nanocrystals.Equilibrium solubility of nanocrystals showed as 1.64-fold of coarse baicalin. Conclusion: Precipitation method showed to be a simple and feasible approach to prepare baicalin nanosuspensions with solubility increased,but more attention should be paid to collection and drying process which affect particle size and stability of nanocrystals.

      摘要:Objective: To investigate influence of forced circulation dynamic technology upon extraction effect of Liuwei Dihuang pills. Method: With extraction ratio of loganin and dry extract ratio as indicators,under premise of extraction times without changing from the existing standard of the product,orthogonal design was used to investigate effects of the amount of adding water and extraction time on dynamic extraction technology of Liuwei Dihuang pills,and compared with traditional extraction methods.HPLC was established to determine the content of loganin with mobile phase of tetrahydrofuran-methanol-acetonitrile-0.05% phosphoric acid solution(1:4:8:87) and detection wavelength of 236 nm. Result: The amount of adding water had significant difference influence on extraction amount of loganin and dry extract ratio,but extraction time had no significant difference.Optimum dynamic extraction technology was as following:extracted twice with three times the amount of water,one hour per time;Average extraction amount of loganin was 1.068 mg·g-1,average rate of dry extract was 25.75%. Conclusion: This optimized technology was stable and feasible with characteristics of energy saving,time-saving,high extraction rate and so on,it was suitable for industrial production of Liuwei Dihuang pills.

      摘要:Objective: To optimize preparation technology of Andrographis Herba extract-β-cyclodextrin(β-CD) inclusion complex by central composite design and response surface method. Method: Andrographis Herba extract-β-CD inclusion complex was prepared by ultrasonic method.Taking inclusion rate as index,central composite design test was adopted to optimize inclusion process with mass ratio of Andrographis Herba extract and β-CD,inclusion time and inclusion temperature as factors,test data were fitted by multi-linear equation and second-order polynomial equation,optimal inclusion technology was predicted by response surface method. Result: Optimum inclusion technology were as follows:mass ratio of Andrographis Herba extract and β-CD 5.57:1,inclusion time 4.5 h,inclusion temperature 55 ℃;Deviation between the observed value and the predicted value was 1.18%. Conclusion: Central composite design and response surface method was suitable for optimizing preparation technology of Andrographis Herba extract-β-CD inclusion complex,this established mathematical model had good predictability.

      摘要:Objective: To optimize separation and purification process of total saponins from Anredera cordifolia by macroporous resin. Method: The content of total saponins was determined by UV.Model of macroporous resin was screened by static adsorption-elution test;Orthogonal test was adopted to optimize separation and purification process of total saponins from A. cordifolia by taking dynamic adsorption capacity and elution rate as indexes. Result: D101 macroporous resin was selected,its optimum separation and purification process conditions were determined as follows:the concentration of sample solution 0.1 g·mL-1,adsorption velocity 1 BV·h-1,ratio of diameter-high of cation resin column 1:10,quality proportion of crude drug-resin 0.2 g·mL-1,ethnol concentration 40%,elution velocity 2 BV·h-1,eluent volume 8 BV;Purity of total saponins from A. cordifolia under these conditions could reach above 50%. Conclusion: Optimized separation and purification process was reasonable and feasible,it could easily separate and purify total saponins from A. cordifolia.

      摘要:Objective: To optimize prescription process of loquat flowers and leaves buccal tablet(LFLBT)and investigate its anti-inflammatory,eliminating phlegm and cough pharmacological activity by pharmacodynamic trials. Method: Taking taste and disintegration time as comprehensive evaluation index,orthogonal test was adopted to optimize prescription process of LFLBT with dosages of microcrystalline cellulose,aspartame,mannitol and citric acid as factors.Phenol red excretion method and concentrated ammonia spray induced cough method and xylene-induced ear swelling in mice method were applied to verifyanti-inflammatory,eliminating phlegm and cough activity of LFLBT. Result: The best prescription process was as follows:added 70 g of MCC,5 g of mannitol,0.5% of aspartame and 1% of citric acid into 30 g of aqueousextract of loquatflowers and leaves,and granulated with 95% ethanol as binder. Compared with blank group,high dose group of LFLBT could significantly increased excretion of phenol redand significantly inhibit ear swelling caused by xylene;Middle dose group and high dose group of LFLBT couldreduce frequency of cough in three minutes and prolong latent period of cough caused by ammonia water. Conclusion: Optimized prescription process was simple and feasible,prepared LFLBT had a certain activity of moisturizing throat.

      摘要:Objective: To optimize extraction technology of modified Sijunzi decoction. Method: Taking total extraction amounts of ginsenoside as index,L9(34) orthogonal design was adopted to optimize alcohol extraction technology of Ginseng Radix et Rhizoma,Atractylodis Macrocephalae Rhizoma and Paeoniae Radix Alba with ethanol amount and concentration,extraction time and times as factors;With yield of total polysaccharides as index,L9(34) orthogonal design was used to optimize water extraction technology of Poriae Cutis,Hedyotidis Herba and Glycyrrhizae Radix et Rhizoma by taking solid-liquid ratio,extraction time and times as factors.The content of ginsenoside was determined by HPLC,mobile phase of acetonitrile-water(13:87),detection wavelength 203 nm;UV was adopted to measure the content of polysaccharides with detection wavelength at 489 nm. Result: Optimum alcohol extraction process was as following:extracted twice with 6 times the amount of 70% ethanol for 1.5 h per time;Extraction amount of ginsenoside was up to 7.69 mg·g-1.Optimum water extraction process was:extracted three times with 8 times the amount of water for 2 h each time;Yield of total polysaccharides reached 2.18%. Conclusion: This optimized extraction technology was stable and feasible,it could full extract alcohol-soluble and water-soluble active ingredients from modified Sijunzi decoction.

      摘要:Objective: To develope a near-infrared quantitative method for rapid determination of total glycosides in countercurrent extracting process of Gardeniae Fructus. Method: UV was used as a reference method to determine the content of total glycosides in Gardeniae Fructus,a rapid on-line determination method of total geniposides was established by NIR transmission method and partial least squares method.Sampling method for transmission acquisition,collection interval 4 000-10 000 cm-1,resolution 8.0 cm-1,numner of scans 64 times,empty door attenuation,gain of 1,temperature at (25±2) ℃,relative humidity 45%-50%. Result: Correlation coefficients and calibration root-mean-square error of calibration model for total glycosides in countercurrent extract of Gardeniae Fructus were 0.993 and 0.179,respectively.By external validation,prodiction root-mean-square error of calibration model was 0.145,correlatiaon coefficient of regression equation between the actual value and the predicted value was 0.991. Conclusion: NIR could be used as a kind of accurate,rapid and on-line detection method for determination of active ingredients in extracting process of traditional Chinese medicine.

      摘要:Objective: To establish a method to determine contents of astragaloside Ⅰ,astragaloside Ⅲ and astragaloside IV from Astragali Radix,then analyzed influence of honey-processed for artragaloside content. Method: HPLC-ELSD was adopted,Thermo Syncronis C18 column(4.6 mm×250 mm,5 μm),mobile phase consisting of acetonitrile-0.3% formic acid using a gradient elution,flow rate of 0.8 mL·min-1 and column temperature at 25 ℃,drifttube temprature of ELSD parameters was 110 ℃,volume flow rate of nitrogen was 3.0 L·min-1. Result: Astragaloside Ⅰ,astragaloside Ⅲ and astragaloside IV showed a good linear relationship in the range of 0.804-8.04,0.784-7.84,0.406-4.06 μg,the average recoveries of them were 98.3%,99.3%,98.9% with RSD of 2.46%,1.89%,2.38%,respectively.Contents of astragaloside Ⅰ,astragaloside Ⅲ and astragaloside IV from Astragali Radix in different places were 0.301-0.527,0.186-0.303,0.128-0.178 mg·g-1,respectively. Conclusion: This established method was simple and reliable,methodology validation met requirements.Contents of astragaloside Ⅰ and astragaloside Ⅲ from processed Astragali Radix were higher than raw materials,but astragaloside IV was opposite,indicating that honey-processed had an effect on the content of saponins.

      摘要:Objective: An analytical method was developed for simultaneously determining alisol C 23-acetate, alisol A, alisol B, alisol B 23-acetate four triterpenoids in the rhizoma of Alisma orientalis. Method: The assay was performed on a Ultimate XB-C18 column (4.6 mm×250 mm, 5 μm) with the mobile phase consisting of acetonitrile-water (65:35) at a flow rate of 1.0 mL·min-1. The column temperature was at 30 ℃ and the optimum detection wavelength of DAD was set at 245 nm. The drift tube and atomizer temperature of ELSD were 60, 55 ℃, with a carrier gas (N2) pressure of 20 psi. Result: The linear range of alisol C 23-acetate, Alisol A, alisol B and alisol B 23-acetate were 1.869-29.90 mg·L-1(r=0.999 8), 3.731-74.62 mg·L-1(r=0.999 7), 8.653-138.4 mg·L-1(r=0.999 9), 4.832-77.31 mg·L-1(r=0.999 5), respeltively. The average recoveries (n=6) were 98.78%(RSD 2.63%), 98.05%(RSD 2.72%), 98.26%(RSD 2.86%) and 97.65%(RSD 2.95%) respectively. Conclusion: The HPLC method is simple, rapid and accurate. This HPLC-DAD-ELSD verified method can be used for quantitative analysis of multi-component of Rhizoma Alismatis and provides a novel approach for evaluation of the quality of Rhizoma Alismatis comprehensively.

      摘要:Objective: To use liquid chromatography-mass spectrometry techniques for qualitative analysis of alkaloids of Mori Folium and determine the content of total alkaloids in alkaloids in Mori Folium by HPLC-UV with pre-column derivatization. Method: The chromatographic separation was performed on a HiQSiL C18 column (4.6 mm×250 mm, 5 μm) using acetonitrile and 0.1% acetic acid aqueous solution under isocratic elution(50:50) at a flow rate of 0.8 mL·min-1.Mori Folium alkaloids was analyzed with 9-fluorenylmethyl chlorformate(FMOC-Cl) as the pre-column derivative agent. Detection wavelength was set at 254 nm with electrospray ionization source(ESI) and positive ion mode. And 1-deoxynojirimycin as the external standard to calculate mulberry leaf total alkaloid content. Result: Six alkaloid compounds were identified as DNJ,fagomine,3-epi-fagomine,DAB,calystegine B2 and 2-O-β-Glc-DAB.The content of 3 batches test samples were more than 75%. Conclusion: The method is sensitive, accurate, simple, and reliable. It can be used for quantitative analysis of Mori Folium alkaloids and preparation.

      摘要:Objective: To establish a method for the determination of both oleanolic acid and hederagenin in Yanyan granule by high performance liquid chromatography and evaporative light scattering detection (HPLC-ELSD). Method: A HPLC-ELSD system was applied, using a Cosmosil PAQ-C18 (4.6 mm×250 mm,5 μm) column;mobile phase consisted of methanol (A)-water (B) under gradient elution;the flow rate was 0.9 mL·min-1. Result: As for oleanolic acid and hederagenin, the linear range was between 1.321-5.285 μg(r=0.999 3)and 0.473 6-1.894 μg(r=0.999 2), with average recovery rate to be 98.5% and 100.2%, respectively. Conclusion: The method developed is simple, rapid with reliable results, which could be applied for the determination of oleanolic acid and hederagenin in Yanyan granule.

      摘要:Objective: A method was established by HPLC for the determination of four flavonoids in Nelumbinis Folium to supply further basis to the quality control of Nelumbinis Folium. Method: Chromatography conditions were Kromasil C18 (4.6 mm×200 mm, 5 μm) column, methanol (A)-0.2% phosphric acid system as mobile phase, (0-8 min, 10%-40%A;8-23 min, 40%-70%A;23-30 min, 70%-85%A);the flow rate was 1 mL·min-1 and column temperature was set at 30 ℃. Diode array detector was used and detected at 360 nm. Result: The assay displayed good linearity;the linear ranges of quercetin-3-O-sambubioside, hyperin, isoquercitrin and quercetin were 6.10-97.60, 18.76-300.16, 8.48-135.68, 9.12-145.92 mg·L-1 respectively. The average recoveries were 100.98% (RSD 1.85%), 99.05% (RSD 1.48%), 99.29% (RSD 1.84%), 97.71% (RSD 1.78%) respectively. The content of the four flavonoids in Nelumbinis Folium from five different origins was 0.092%-0.161%, 0.949%-1.862%, 0.067%-0.133% and 0.040%-0.062%. Conclusion: The method is simple, accurate and can be used to determine quercetin-3-O-sambubioside, hyperin, isoquercitrin and quercetin in Nelumbinis Folium. The content of four flavonoids in Nelumbinis Folium from different origins varied notably and should be taken into the quality control of Nelumbinis Folium.

      摘要:Objective: To compare the chemical compositions in the five kinds of Dendrobii Caulis. Method: Polysaccharide,mannose and amino acid in the five kinds of Dendrobii Caulis were determined and compared. Result: The polysaccharide contents,the amino acid contents and the mannose contents are remarkably different in the five kinds of Dendrobii Caulis. The content of polysaccharide in the Dendrobii Caulis officinnle is highedest in the five kinds of Dendrobii Caulis;the content of amino acid in the Dendrobii Caulis finbristun is higher than other four kinds of Dendrobii Caulis;the mannose contents of Dendrobii Caulis officinnle is much more higher than other kinds. Conclusion: The quantity and contents of polysaccharide, amino acid and the mannose are remarkably different in the five kinds of Dendrobii Caulis. However,Dendrobii Caulis officinnle has a higher quality than other four kinds of Dendrobii Caulis in terms of the same chemical compositions1.

      摘要:Objective: To investigate the content of heavy metals in Sulphur fumigated and Drying Lonicera japonica Thunb. Method: The sample is prepared via microwave digestion, the processing using the method of sulphur fumigated and drying, using atomic absorption spectroscopy to determine the content of Pb,Cd,As,Hg,Cu in sulphur fumigated and drying L. japonica. Result: The content of heavy metal in 10 batches of sulphur fumigated and drying Lonicera japonica.from different areas ware determined. The recovery rate was between 96.65% and 98.97%. Conclusion: The content of Pb, Cd, As, Hg and Cu in all samples were within the limitation, the content of Pb in the sample to be low after sulphur fumigated, the content of Hg elevates, the content of Hg elevates obviously, Cu and Cd maintains invariable basically.

      摘要:Objective: To develop an UPLC method of determining the characteristic chromatographic profile of Lonicera japonica for controlling the drug quality quickly and accurately. Method: The analysis was performed on an Agilent C18 column(2.1 mm×50 mm,1.8 μm) with acetonitrile-0.2% phosphoric acid as mobile phase in gradient elution mode. The flow rate was 0.4 mL·min-1 and the injection volume was 2.0 μL with the detection wavelength of 238 nm.The column temperature was set at 30 ℃. Result: The fingerprint of Lonicera japonica was obtained within 21 min and five peaks in the fingerprint were designed.14 samples were analyzedand the similarities were 0.915-0.987. Conclusion: The UPLC system enjoys the merits of a higher speed,sensitivity,and resolution, it can be used to control the quality of L. japonica.

      摘要:Objective: To survey the quality standards of Ginseng Radix et Rhizoma Rubra slices. Method: This experiment was carried out to observe the characters, identify the quality by microscopy and TLC, and make the content determination by HPLC for the Ginseng Radix et Rhizoma Rubra slices from different areas. The chromatographic column was Lichrospher C18(4.6 mm×150 mm, 5 μm) with gradient elution of acetonitrile and water. The detection wavelength was set at 203 nm;the flow rate was at 1.0 mL·min-1and the column temperature was set at 30 ℃ respectively. Result: The appearances of samples were different when micro-characteristic was obvious and TLC spots were clear on the corresponding position of ginseng control medicine, ginsenoside Rf, Rg1, Re and Rb1.Concentration range of Rg1+Re was 0.097 0%-0.301% when concentration range of Rb1 was 0.140%-0.310% in thirteen batches of samples. Conclusion: The content of ginsenoside Rg1, Re and Rb1 was considerably different as well as the characters and micro-characteristics of slices.The study provides a reference for standards of Ginseng Radix et Rhizoma Rubra slices which need to be formulated.

      摘要:Objective: To establish the HPLC fingerprint of Arnebiae Radix smashing tissue extraction with ethanol and reflect the chemical information comprehensively and provide evidence for quality control. Method: HPLC was used on a Kromasil 100-5 C18 (4.6 mm×250 mm, 5 μm) by gradient elution with solvent system composed of water-formic acid (300:0.5)-acetonitrile, detection wavelength was 275 nm, the flow rate was 1 mL·min-1, the column temperature was set at 25 ℃. Result: HPLC fingerprint of Arnebiae Radix smashing tissue extraction with ethanol was established, which showed 13 characteristic peaks from 10 batches of crude drug. Among them, the No 4, 6, 10 peaks were respectively identified as shikonin, acetylshikonin, and β,β'-dimethylacryl alkannin. Conclusion: The method is simple and has good precision, reproducibility and stability, and can be applied to the quality control the Arnebiae Radix smashing tissue extraction with ethanol, also provide a quality assurance for further pharmacological studies and preparation research.

      摘要:Objective: To develop an HPLC method for determination of the content of platycodin D, platycodin E, praeruptorin A and praeruptorin B in Chutan Zhisouwan. Method: A Hypersil C18 column was used as the chromatographic column. The flow rate was 1.0 mL·min-1. The mobile phase consisted of acetonitrile-water (35:65) for platycodin D and platycodin E, the temperature of drift tube was set at 105 ℃;the gas flow (N2) was set at 2.6 SLPM·min-1.The mobile phase consisted of acetonitrile-methanol-water (40:30:30) for praeruptorin A and praeruptorin B, The UV detection wavelength was set at 321 nm. Result: Platycodin D and platycodin E were linear within the range of 0.041 3-0.826 0 μg (r=0.999 7) and 0.079 7-1.594 0 μg (r=0.999 8) respectively. The average recoveries were 98.32% (RSD 0.80%) and 98.94% (RSD 0.93%). Praeruptorin A and praeruptorin B were linear within the range of 0.154 9-3.098 0 μg (r=0.999 4) and 0.051 7-1.034 0 μg (r=0.999 8) respectively. The average recoveries were 98.72% (RSD 0.86%) and 99.15% (RSD 0.95%). Conclusion: The method was accurate, sensitive and reproducible, which may be used in the determination of platycodin D, platycodin E, praeruptorin A and praeruptorin B in CTZSW.

      摘要:Objective: To study the chemical constituents in the Gonostegia hirta. Method: The constituents were isolated by column chromatography and their structures were elucidated by physicochemical properties and spectroscopic analysis. Result: Nine compounds were isolated from the plant and identified as β-sitosterol (1), β-daucosteol (2), sucrose (3), epifriedelanol (4), friedelin (5), friedlane (6), oleanolic acid (7), pomolic acid (8), hederagenin (9). Conclusion: All the compounds are obtained from this plant for the first time.

      摘要:Objective: To study the chemical constituents of volatile oil from different season flowers of Erigeron annus. Method: The volatile oil was extracted by steam distillation and analyzed by GC-MS. The relative content of each component was determined by area normalization. Result: Fifty-one and thirty-two compounds were identified from the spring and autumn flowers of E. annus, representing 80.66% and 90.85% of the total content, respectively. Conclusion: Terpenes are found to be the dominant components and the chemical constituents over 2.00% are the same in the volatile oil from different season flowers of E. annus.

      摘要:Objective: To study the constituents of the whole plant of Elatostema duyunens. Method: The compounds were separated and purified by silica gel and recrystallisation. Their structures were identified by their physico-chemical properties and spectroscopic data. Result: Ten compounds were isolated and identified as kaempferol-4', 7-dimethyl-3-O-glucoside, quercetin-7-O-β-D-gluoopyranoside,kaempferol-3-O-β-D-galactopyranoside,apigenin-7-O-β-D-glucopyranoside, daucosterol, and hexadecanoic acid. Conclusion: Compounds of 1-6 were obtained from this plant for the first time.

      摘要:Objective: To analyze the physicochemical properties differences between Aconiti Laterdis Radix Praeparata and Glycyrrhizae Radix et Rhizoma decoction before and after combination. Method: Use inductivity measurer, turbid meter, viscosity meter and surface tension apparatus to measure the physicochemical properties (inductivity, salinity, turbid, viscosity, surface tension) of four kinds of decoctions;use laser diffraction to measure particle size of these four different decoctions;analyze the physicochemical properties between these decoctions. Result: There are indeed physicochemical properties differences between these decoctions;the inductivity and salinity of Aconiti Laterdis Radix Praeparata and Glycyrrhizae Radix et Rhizoma combination are less than those of other decoctions, but the particle size of combination is larger than that of other decoctions. Conclusion: Supply a research data for studying correlativity between effective chemical constituents and physicochemical properties in decotions before and after combinated.

      摘要:Objective: To provide evidences for the pharmacognostical and chemical identification and the further development of Hemiboea rubribracteata. Method: Paraffin tissue sections, powders delt by chloral hydrate before watching them and HPLC-UV. Result: Lots of stone cells existed in the cortex and the pith have large area, There are also a lot of stone cells exit in the mesophyll and parenchyma of midrib which are composed of closed collateral vascular bundles.Stone cells, starchs and solitary crystals exist in the powder of H. rubribracteata. paraboside Ⅲ and paraboside Ⅱ exist in H. rubribracteata. Conclusion: The pharmacognostic identification and chemical identification of H. rubribracteata not only can be used to identify the medicial plant but also provide evidences for the further development of it.

      摘要:Objective: To study the effects of Xiangfu Siwu decoction on the blood, nerve endocrine immune (NEI) system in female rats with Qi stagnation and blood stasis syndrome. Method: The female SD rats model with Qi stagnation and blood stasis syndrome were prepared by giving adrenaline(Adr) and chronic unpredicted stress, while the medicine groups were treated with Xiangfu Siwu decoction. ELISA and immunohistochemistry assay were employed to detecte contents of interlerkin-6(IL-6), leukotrienes 4(LTB4), estradiol(E2), progesterone(P), prostaglandin E2(PGE2), nitric oxide(NO) in the serum and β-endorphin in hypothalamus before and after the treatment. PLS-DA(Patrial least squares discriminant analysis)method was used to analyze the above factors. Result: Compared with control group, the model group had significantly higher levels on hemorheology and significantly lower levels on thrombin time(TT) and activated partial thromboplastin time(APTT), while the model group had no obvious changes on prothrombin time(PT) and fibrinogen;the model group had significantly higher levels on the content of IL-6, P in the serum, and significantly lower levels on the contents of E2, PGE2, NO in the serum, while the model group had no obvious changes on the content of LTB4.Xiangfu Siwu decoction had a significant effect on improving the model with Qi stagnation and blood stasis syndrome. Compared with control group, the medicine groups had no significant difference. Through a multi-index analysis with PLS-DA, the most important X variables were E2, PGE2, P, TT, horizontal movement, area, vertical movement, integral optical density(IOD), 30 s-1 whole blood riscosity, residence time, plasma viscosity. Conclusion: The therapeutic effects of Xiangfu Siwu decoction on many diseasesmay be mediated by modulating multi-level of blood, nerve, immune, endocrine system and keeping internal environment homeostasis.

      摘要:Objective: To observe the effects of Rhizoma Zingiberis, Cortex Cinnamomi, Syzygium aromaticum, Fructus Evodiae, Fructus Foeniculi, Rhizoma Alpiniae Officinarum, Fructus Piperis Longi, Piper cubeba, Pericarpium Zanthoxyli, Fructus Piperis Nigri, the ten herbs of warming middle-jiao and dispelling cold on the pathologic tissues of gastric mucosa in rats with stomach excess cold syndrome, and research the warming and protective effect on stomach. Method: SD male rats were divided into 15 groups:blank group, model group, Radix Aconiti Lateralis Preparatax group, Rhizoma Zingiberis group, Cortex Cinnamomi group, Syzygium aromaticum group, Fructus Evodiae group, Fructus Foeniculi group, Rhizoma Alpiniae Officinarum group, Fructus Piperis Longi group, Piper cubeba group, Pericarpium Zanthoxyli group, Fructus Piperis Nigri group, Gypsum Fibrosum group and Rhizoma Coptidis group.the stomach cold syndrome model in rats was established by water extract of Rhizoma anemarrhenae 17.5 g·kg-1,twice per day, lasting two days. the dosage of Radix Aconiti Lateralis Preparatax group was 13.5 g·kg-1,the dosage of Rhizoma Zingiberis and Fructus Piperis Nigri group was 8.1 g·kg-1, the dosage of Cortex Cinnamomi and Fructus Evodiae group was 4.1 g·kg-1, the dosage of Fructus Caryophylli and Fructus Piperis Longi, Piper cubeba group was 2.7 g·kg-1, the dosage of Fructus Foeniculi, Rhizoma Alpiniae Officinarum and Pericarpium Zanthoxyli group was 5.4 g·kg-1, the dosage of Gypsum Fibrosum group was 54 g·kg-1, the dosage of Rhizoma Coptidis group was 4.5 g·kg-1,twice per day in 3 times. The changes of the gastric mucosa were observed by naked eyes and th echanges of pathological section were observed by light microscope, and graded according score criterion of the pathologic tissues of gastric mucosa. Result: Compared with blank group, the stomach mucous membrane in model group had dark color,crimple,hemorrhagic spots and no gloss, compared with model groupthe stomach mucous membrane in the 10 treatment groups was improved.All the treatment groups could significantly decrease the imflammatory cells infiltration(P<0.05 or P<0.01)except the group of Alpinia group and reduce the glands(P<0.01). Conclusion: The ten herbs with warming middle-jiao and dispelling cold have good improvement on the pathologic tissues of gastric mucosa so as to treat stomach cold syndrome.

Consequently to rats. The serum containing drugswas used totreat HepG2 cells for 24, 48, 72 h. Then the effects of different drugs on proliferation, telomerase and apoptosis of HepG2 cells were observed by MTT, ELISA and Flow cytometry Annexin V-FITC/PI assay, respectively. Result: Compared to control group,proliferation and telomerase activities of HepG2 cells were inhibited with time-dependent manner by serum containing Sarcandrae compound significantly (P<0.05). All serum containing Sarcandrae compound could promote the apoptosis of HepG2 cells significantly. For middle dose group at 24,48,72 h the apoptosis rate was (14.2±0.9)%, (30.6±1.0)%, (61.6±1.5)%. Conclusion: Serum containing Sarcandrae compound can inhibit the proliferation and telomerase activity of HepG2 cells, and promote the apoptosis of HepG2 cells.

      摘要:Objective: To investigatet the time-effect and dose-effect of anti-hypertensive effect in spontaneously hypertensive rats (SHR) byaqueous extract and the supernatants of alcohol extracting-water precipitation of Prunella vulgaris L. Method: P. vulgaris was processed by water extraction, water extraction and alcohol precipitation.The SHR were treated with different dose of two extracts. SBP, diastolic blood pressure(DBP), mean arterial pressure(MAP), and heart rate(HR) were measured before and after oral administration for 2,5,7,10 h for determining the time-effect. Result: Compared with the model group, no significant effect were showed on MAP and HR of SHR in all the different dose of test group in every test time points. SBP of SHR decreased significantly in aqueous extract(10, 5 g·kg-1) and the supernatants of alcohol extracting-water precipitation(10, 5, 2.5 g·kg-1) of P. vulgaris,while high dose (10 g·kg-1)of the supernatants of alcohol extracting-water precipitation decreased DBP of SHR after oral administration for 5 h(P<0.05). Conclusion: The supernatants of alcohol extracting-water precipitation of P. vulgaris retain and enrich the active anti-hypertension principles when parts of other principles were removed, which can provide reference for the study of anti-hypertension material base and pharmaceutical technology in P.vulgaris.

      摘要:Objective: To investigate the inhibitory effect of combined administration bisdemethoxy curcumin (Bdmc) and adriamycin(ADR) on multidrug resistance HL-60/ADR cells. Method: HL-60/ADR cells were simultaneously treated with Bdmc within concentrations (1,2,4,8 μmol·L-1) and ADR(0.4,0.8,1.6,3.2 μmol·L-1)48 h respectively, the cell density was 2×104/hole. The inhibitory effect of both drugs on HL-60/ADR cells in vitro was evaluated by MTT. Cells were reated sequentially administration Bdmc(6,12,24,48 μmol·L-1) and ADR(0.08,0.2,0.4,1.0 μmol·L-1). The Jin's formula was used to analyze the the inhibitory effect. Result: The inhibition is 6.9%,11.8% when treated only with ADR(0.4,0.8 μmol·L-1),while simultaneously treated with Bdmc(8 μmol·L-1) it was over 94%.In simultaneous administration Bdmc (1-8 μmol·L-1) combined with ADR(0.8-3.2 μmol·L-1) revealed a simple addition or potentiation effect. In addition, sequential administration of Bdmc followed by ADR had a synergistic effect;in contrast, sequential administration of ADR followed by Bdmc exhibited an antagonistic effect. Conclusion: Simultaneous administration of Bdmc and ADR reveals a synergistic effect and sequential administration of Bdmc and ADR exhibits an unidirectional synergistic effect.

      摘要:Objective: To discuss the effects of traditional Miao national herbs Sidaxue on rheumatoid arthritis and its mechanism. Method: An animal model of adjuvant arthritis (AA) was established, rats were divided into blank control group, model control group, high dose Sidaxue treatment group, middle dose Sidaxue treatment group, low dose Sidaxue treatment groups, positive control group(glucosidorum tripterygll totorum control group). The rat paw edemas of rats was measured;Synovial cells were primarily cultured, and the 3rd generations of subculture cell were adopted for experiment. Western-blotting method was adopted to detect the protein expression level of B cell lymphoma/leukemia-2(Bcl-2), Bcl-2 associated X protein(Bax). Result: Miao national herbs Sidaxue could inhibit the swelling of paw (P < 0.05);decrease the protein expression of Bcl-2 (P < 0.05,P < 0.01), increase the protein expression of Bax in Arthritis rats' synoviocyte(P < 0.01), and decrease the rate of Bcl-2/Bax. Conclusion: Miao national herbs Sidaxue can antagonize the development of AA, the mechanism of which may be related to promoting apoptosis of synoviocyte.

      摘要:Objective: To investigate effect of licoflavone on blood glucose, glycogen, muscle glycogen expression of retinol-binding protein 4 (RBP4) of adipose tissues in rats after exhaustive exercise. Method: Fifty healty male Sprague-Dawley rats were randomized into three groups, 10 rats in each group;the quiet control group (NC), the exercise group (ME), the low. middle and high dose licoflavone combined with exercising group (MGFL, MGFM, MGFH, 4, 8, 12 g·kg-1·d-1). The rats were administrated by gavage once a day half an hour before exercising for 6 days in one week, and lasted for six weeks.The control group was administrated physiological saline. The rats were killed after 6-weeks. The contents of serum glucose, glycogen, muscle glycogen were tested by corresponding kit method. And the expression of epididymal adipose tissue RBP4 mRNA in rats was detected by reverse transcription-polymerase chain reaction (RT-PCR) method. Result: The exhaustive swimming training induced the contents of serum glucose, glycogen, muscle glycogen in the exercise group and each MGF group was significantly lower than in the quiet control group (P<0.05 or P<0.01). The content of serum glucose was respectively low(4.81±0.24), (5.31±0.12), (5.78±0.22), (5.92±0.18) mmol·L-1. The content of serum glucose in each MGF group was higher than in the exercise group. And there was a significant difference between MGFH and the exercise group (P<0.05). The content of muscle glycogen was respectively low (0.91±0.16),(0.98±0.14),(1.34±0.12),(1.46±0.23) mg·g-1. The content of muscle glycogen in each MGF group was higher than the exercise group. And there were significant difference between MGFM, MGFH and the exercise group (P<0.05). The content of glycogen was respectively decreased (6.21±1.10),(8.92±1.11),(9.18±1.13),(10.16±1.16) mg·g-1. And the content of glycogen in each MGF group was higher than that in the exercise group (P<0.05 or P<0.01). The expression of epididymal adipose tissue RBP4 mRNA in exhaustive swimming group and each MGF group increased (1.430±0.123),(1.101±0.103),(0.962±0.112),(0.926±0.101). The expression of RBP4 mRNA in the exercise group had a significant difference compared to the quiet control group (P<0.05). The expression of RBP4 mRNA in each MGF group was lower than that in the exercise group, and the expression of epididymal adipose tissue RBP4 mRNA in MGFM and MGFH groups had significant difference compared to the exercise group (P<0.05). Conclusion: The supplement of licoflavone could maintain the increase of glycogen reserve, the relatively stable blood glucose concentration and down regulate the expression of RBP4 mRNA. This is contributed to improving the athletic ability of rats.

      摘要:Objective: To observe the effect of Fuzhi capsule on learning and memory ability of rats with vascular dementia, the activity of choline acetyltransferase (CHAT), the expression of synaptophysin (P38) and hippocampal synaptic microstructure, and to explore the mechanism of Fuzhi capsule on improving the ability of learning and memory. Method: The SPF male SD rats were divided into model group, Donepezil group, Fuzhi capsule high and low dosage groups, and sham operation control group randomly.The vascular dementia model was established by permanent bilateral common carotid artery ligation (2-VO). Drugs were administered to rats by gastric perfusion once a day for 4 weeks. Then all the rats were detected the behavior by Morris water maze, the expression of CHAT and P38 in hippocampus by immunohistochemistry, the synapse microstructure of hippocampal neurons and the number of dendritic spines by Golgi staining. Result: Compared with the sham operation group, the learning and memory ability of rats in model group were decreased, the escape latency and swimming distance were significantly prolonged, the number of across the platform after remove platform was decreased, the activity of CHAT and the expression of P38 in hippocampus were decreased, the number of dendritic spines in the hippocampus neurons was reduced (P<0.05). Compared with the model group, the learning and memory ability of rats in Fuzhi capsule high and low dosage group and donepezil group were improved, the escape latency and swimming distance were shortened, the number of across the platform after remove platform was increased obviously, the activity of CHAT and the expression of P38 in hippocampus was increased, the number of dendritic spines was increased (P<0.05). Conclusion: Fuzhi capsule can improve the learning and memory ability of rats with vascular dementia by increasing the activity of CHAT and the expression of P38 in hippocampus, and promote hippocampal synaptic microstructure reconstruction.

      摘要:Objective: To investigate the mechanism of Qingre Jiedu prescription in treatment septic. Method: The septic model was induced by cecal ligation and puncture. The experimental rats were divided into five groups;control group,septic model group,Qingre Jiedu prescription treated group (24,12,6 g·kg-1). All animals were treaded via gastrogavage with distilled water and Qingre Jiedu prescription once a day for 3 days. The survival state was observed. The bacterial populations in experimental sepsis were determined by bacterial culture. And aquaporins(AQP) 2 and AQP8 were detected by western blot. Result: The fatality rate in model group was the highest (56%), meanwhile all bacteria in ascites, lymph nodes and bloods were significantly higher than that in control group. For treaded group, the mortality and the serum biochemical parameter in the high dose group were significantly decreased. The AQP2 and AQP8 expression levels in all treated groups were decreased compared with that in model group, with a dose-dependent manner. In the high dose group,the number of pathogens in lymph nodes and blood was significantly lower than that in model group. Conclusion: Qingre Jiedu prescription can decrease bacterial translocation by reducing expression of AQPs to increase excreta.

      摘要:Objective: To extract total saponins in Guizhou's Asparagi Radix (ASP) and observe the inhibitory effect of ASP on the HL-60 cells. Method: Effects of ASP on the cytoactive and apoptosis rate of HL-60 cells were investigated by MTT and flow cytometry.The expression of B cell lymphoma/leukemia-2(Bcl-2 mRNA in HL-60 cells was detected by method of Real-time PCR. Result: ASP could dose-dependently inhibit the cytoactive of HL-60 cells and improve significantly the apoptosis rate of HL-60 cells;The expression of Bcl-2 mRNA in HL-60 cells decreased significantly compared with blank group. Conclusion: ASP can decrease the expression of Bcl-2 mRNA and make HL-60 cells apoptosis,which maybe one of mechanisms of treating the human promyelocytic leukemia.

      摘要:Objective: To compare Hawthorn leaves extract niosomes and Yixintong tablet on pharmacodynamic actions of acute myocardial ischemia in rats,and provide the pharmacodynamics characteristics of Hawthorn leaves extract niosomes. Method: The rats were randomly divided into blank control group, model group, Yixintong tablet group, hawthorn leaf extract niosomes group, with 10 rats in each group. The rats of Yixintong tablet group and hawthorn leaf extract niosomes group were administered with 17.28 mg·kg-1 hawthorn leaf extract, once a day, for continuous 7 days;model group and blank control group rats were administered with equal volume of distilled water (10 mL·kg-1). On the 15th days and 16th day, except blank control group, other groups were ip 4.2 mg·kg-1 1 h after administration of isoproterenol, once a day, in order to establish the rat acute myocardial ischemia model, the rats in blank control group were ip equal volume of physiological saline. 24 h after establishing the model, 1 h after the last administration, the rats were anesthetized with 10% chloral hydrate to obtain blood samples. Some myocardium enzymes like aspartic acid transferase (AST), lactate dehydrogenase (LDH), creatine phosphokinase (CPK), malonyldialdehyde (MDA) and superoxide dismutase (SOD) were measured. Result: Compared with the blank control group, in model group the level of myocardium enzymes and MDA increased significantly (P<0.05), the level of SOD was significantly decreased (P<0.001). Compared with model group,only the LDH and MDA levels in Yixintong group were significantly decreased(P<0.05),AST, LDH, CPK, MDA levels in Hawthorn leaves extract niosomes group were significantly decreased(P<0.05 or P<0.01), the level of SOD was significantly elevated(P<0.01). Compared with the Yixintong group, the AST, LDH, CPK, MDA levels in Hawthorn leaves extract niosomes group were no significant difference, the SOD level were significantly elevated(P<0.05). Hawthorn leaves extract niosomes in rat organs had no obvious toxicity. Conclusion: Hawthorn leaves extract niosomes and Yixintong tablets in clinical equivalent dose can obviously reduce the degree of myocardial ischemia and the content of MDA in rats, which indicates a significant protective effect on myocardial ischemia induced by isoproterenol injury in rats.

      摘要:Objective: To observe the effect of the Qingre Huayu granule (QRHY) on the inositol requiring enzyme 1(IRE1) mRNA and protein after focal ischemic preconditioning in rats. Method: Eighty healthy male SD rats were randomly divided into four groups:sham-operation group, middle cerebral artery occlusion (MCAO) group,brain ischemia preconditioning(BIP) group and QRHY group.Each group was further divided into 4 subgroups according to ischemia/reperfusion(I/R) duration of 12 h,1,2, 3 d.The BIP models was established and 14 mL·kg-1 QRHY granule solution was ig given twice everyday for 3 days after preconditioning. But the MCAO and BIP group was ig given same amount physiological saline.The expression of IRE1 mRNA and protein was detected by Real time PCR and Western blot. Result: The expression both of IRE1 mRNA and its protein all increased and reached the peak at 24 h, then decreased continuously(P<0.05). BIP could increase their expression (P<0.05 or P<0.01). QRHY treatment could further induce its expression compared with BIP group(P<0.05 or P<0.01). Conclusion: BIP could upregulate the expression of IRE1 after focal ischemic injury in rats. QRHY could augment the expression of IRE1 induced by BIP.

      摘要:Objective: To investigate anti-inflammatory and analgesic effects of Lidan Zhitong capsule and its mechanism. Method: The experimental animals were randomly divided into five groups, normal saline group, low dose (mice 0.36 g·kg-1, rats 0.18 g·kg-1) group of Lidan Zhitong capsule, middle dose (mice 0.72 g·kg-1, rats 0.36 g·kg-1) group of Lidan Zhitong capsule, high dose (mice 1.44 g·kg-1, rats 0.72 g·kg-1) group of Lidan Zhitong capsule and aspirin group (mice 0.20 g·kg-1, rats 0.10 g·kg-1). The anti-inflammatory effect was evaluated by dimethylbenzene-caused ear swelling, acetic acid induced celiac capillary permeability in mice and carrageenin induced paw edema in rats. The hotplate test and the acetic acid writhing in mice were used to study the analgesic action. The anti-inflammatory mechanism was evaluated by measuring the contents of intereukin-1(IL-1), tumor necrosis factor-α(TNF-α), prostaglandin E2(PGE2) and malon dialdehyde(MDA) in the inflammatory tissue. Result: The middle and high doses of Lidan Zhitong capsule could significantly inhibit the mice ear swelling and rats paw edema(P<0.05, P<0.01), decrease celiac capillary permeability in mice(P<0.05, P<0.01). The middle and high doses of Lidan Zhitong capsule not only could remarkably improve the pain threshold of hot-plate test(P<0.05, P<0.01), but also obviously decrease writhing times injected with acetic acid in mice(P<0.05, P<0.01). Lidan Zhitong capsule could also decrease the contents of interleukin-1(IL-1), tumor necrosis factor-α(TNF-α), prostagland in E2(PGE2) and malondialdehyde(MDA) in the inflammatory tissue. Conclusion: Lidan Zhitong capsule has remarkable anti-inflammatory and analgesic effects, which may be related to the inhibition of IL-1, TNF-α, PGE2 and MDA.

      摘要:Objective: To investigate the effect of Haoqin Qingdan decoction on the content of interleukin 8(IL-8) in serum and expression of IL-8, neclear factor-κB p65(NF-κB p65) in Gastric mucosa in Helicobacter pylori-associated gastritis(HAG) mice and explore its therapeutic mechanism for Helicobacter pylori(HP) associated gastritis. Method: HP standard strains (SS1) brinell broth liquid was used to establish the HAG mice model. Sixty healthy BALB/c mice were randomly devided into six group (n=10, each), normal control group(given with equal NS), model group (given with equal NS), livzon weisanlian group(0.25 g·kg-1), Haoqin Qindan decoction (4,8,16 g·kg-1) group. The mice in every group were administrated with drugs by oral gavage once a day. All mice were killed two weeks later, the pathological changes of each group were observed by the microscope, the contents of IL-8 in serum were detected by enzyme-linked immunoassay and the expression of IL-8, NF-κB p65 in gastric mucosa tissue were detected by immunohistochemistry. Result: Compared with model group, Haoqin Qingdan decoction significantly reduced the inflammation area and average blackness of gastric mucosa in HAG mice (P<0.01);the expression of IL-8 in serum of Haoqin Qingdan decoction groups were significantly lower than that in model group (P<0.01);the expression of IL-8, NF-κB p65 in gastric mucosa of Haoqin Qingdan decoction groups were significantly lower than that in model group (P<0.05). Conclusion: Haoqin Qingdan decoction can plays a significant role in the repair of gastric mucosal injury in HAG mice, its therapeutic mechanism may be related to inhibiting the activation of NF-κB p65 and degrading the expression of IL-8 in the gastric mucosa and serum.

      摘要:Objective: To observe the effect of Huatan Tongluo therapy on gene expression of brain microvascular endothelial cells in rats with acute cerebral infarction after recombinant tissue-type plasminogen(rt-PA) thrombolysis, and provide experimental basis for preventive and therapeutict effects of Huatan Tongluo on ischemia-reperfusion injury induced by acute cerebral infarction after thrombolysis. Method: Two hundred and fouty healthy SD rats were randomly divided into six groups, sham operation group, model group, rt-PA group, rt-PA combined with Huatan Tongluo low dose group, rt-PA combined with Huatan Tongluo middle dose group, rt-PA combined with Huatan Tongluo high dose group. The middle cerebral artery occlusion(MCAO) rat model was prepared by autologous blood clots. rt-PA group and rt-PA combined with Huatan Tongluo groups used rt-PA thrombolytic therapy after thrombosis. The later were also given low, medium, high dose(3.6, 7.2, 14.4 g·kg-1) Huatan Tongluo prescription by gavage two times a day. Then observed the expression of occluding, claudin-1, ZO-1 in rat cortex and hippocampus by RT-PCR at 6, 24, 72 h and 7 days respectively. Result: In different phases of every experimental group, the expression of occluding, claudin-1, ZO-1 in different brain tissue were highest at 6 h, minimum at 24 h, increased gradually at 72 h and 7 days, and the combined with Huatan Tonglow groups increased more obviously. The expression of occluding, claudin-1, ZO-1 in rt-PA, low, middle, high dose groups at 6 h were significantly higher than that at 24, 72 h(P<0.05). Compared the model group at the same time, the expression of occluding, claudin-1 in middle dose and high dose groups of Chinese traditional medicine were significantly increasedin each phase (P<0.05). While the ZO-1 gene expression had the similar trends, but significant difference only existed at the 24, 72 h, 7 d (P<0.05). Conclusion: Huatan Tongluo combined with rt-PA therapy can regulate the expression of occluding, claudin-1, ZO-1 in brain microvascular endothelial cells. So it can protect the integration of blood brain barrier, and have protective effects on ischemia reperfusion injury induced by acute cerebral infarction after thrombolysis.

      摘要:Objective: To observe the protective effect of Panax japonicus ethanol extract on the inflammation of mice mononuclear macrophage RAW264.7 cells induced by lipopolysaccharide(LPS). Method: The RAW264.7 cells were treated with different concentration of P. japonicus ethanol extract, cell viability was determined with MTT method; the nitric oxide(NO) release of RAW264.7 cells stimulated by LPS and treated with LPS and P. japonicus ethanol extract was detected with Griess. ELISA were used to detect the expression of Tumor necrosis factor-α(TNF-α), interleukin 1β(IL-1β); nitric oxide synthase mRNA (iNOS mRNA) and neclear factor-κB(NF-κB) was tested by PCR and Western bolt respectively. Result: The safe range of ethanol extract of P. japonicus is less than 100 mg·L-1; Concentration of LPS was 1 mg·L-1;Compared with the LPS model group, P. japonicus ethanol extract 0.1, 1, 10, 40 mg·L-1 could inhibit the of NO effectively, the inhibition rates were 31.2%, 41%, 46.1%, 55.2%, and inhibit the release of TNF-α, IL-1β, with significant difference(P<0.05 or P<0.01);P. japonicus ethanol extract 10, 40 mg·L-1 could down-regulate the expression of iNOS mRNA and inhibit NF-κB nuclear translocation. Conclusion: P. japonicus ethanol extract has protective effects on RAW264.7 cells stimulated by LPS, the mechanism of P. japonicus ethanol extract may be related to it's regulation on NF-κB signal pathway, thereby inhibiting the release of NO and reducing the expression of TNF-α, IL-1β and iNOS.

      摘要:Objective: To investigate effect of of Xiaoyaosan on dopamine receptor cyclic adenosine monophosphate(cAMP)/proteinkinase A(PKA) signal transduction pathway in hyperprolactinemia(HPRL) rats. Method: The metoclopramide was injected to establish the animal model,48 SD rats were randomized into 6 groups:control group, model group, bromocriptine group, high dose modified Xiaoyaosan group, middle dose modified Xiaoyaosan group, low dose modified Xiaoyaosan group. The intervention period lasted 30 days. The immune histochemistry method to observe expression of PKA,D1R,D2R,ELISA method to observe expression of cAMP, fluorescent quantitative PCR method was used to observe expression of D1R and D2R. Result: Compared with the control group, modified Xiaoyaosan groups reduced PKA,cAMP activity(P < 0.01);dopamine D2 receptor was significantly increased(P < 0.01). Conclusion: The modified Xiaoyaosan can regulate the PRL level through dopamine receptor cAMP/PKA signal transduction pathway.

      摘要:Objective: To evaluate the efficacy and safety of Pediatric Huanglong granule treating of attention deficit hyperactivity disorder (ADHD),which belongs to hyperactivity of yang due to yin deficiency in traditional Chinese medicine(TCM). Method: Adopt a randomized, double blind, double modeling, parallel controlled with Jing Ling oral liquid as positive control medicine, multicenter clinical trial design,for a total observation completely accords with the inclusion criteria of children with ADHD belongs to hyperactivity of yang due to yin deficiency in TCM syndrome differentiation 299 cases, including test group 224 cases, control group 75 cases.Test group take Pediatric Huanglong granule according to the method of children aged 6 to 9 years take a bag once, 10 to 14 years take two bags once, two times daily, while taking the simulation agent of Jingling oral liquid ; control group take Jingling oral liquid according to the method of children aged 6 to 9 years take one once, 10 to 14 years take two once, two times daily, while taking the simulation agent of pediatric Huanglong granule,for 6 weeks. Compare the SNAP - 4 rating scale, conner scale hyperactivity index and TCM syndrome scoring before into the groups and after taking the drug 3 weeks and 6 weeks. Result: Curative effect of the disease:the control effective rate of the test group was 52.68%, the total effective rate was 83.48%; the control effective rate of the control group was 37.34%, the total effective rate was 66.67%. Curative effect of TCM syndromes:the control effective rate of test group was 54.91%, the total effective rate was 87.95%; the control effective rate of control group was 38.67%, the total effective rate was 74.67%.The diseases and TCM syndromes curative effect difference of two groups has statistical significance (P<0.01), the test group were better than the control group.The primary therapeutic effect index of two groups:SNAP-4 rating scale, conner scale hyperactivity index were tested by t-test after 6 weeks of treatment. The result showed that the difference of two groups was statistical significance (P<0.05). The t-test result of TCM syndrome score after 6 weeks of treatment showed that there were statistical significance between the two groups (P<0.01), the test group were better than the control group. Patient compliance of two groups were both well,the difference was not statistically significant. Conclusion: Pediatric Huanglong granule has the efficacy of nourishing yin for suppressing hyperactive yang and calming the nerves. It uses for treating ADHD,which belongs to hyperactivity of yang due to yin deficiency in TCM, as it can significantly improve TCM syndrome such as hyperactivity restlessness, rhembasmus, loquacity, quick-tempered, night sweating, mouth and throat feeling dry, hand and foot and heart being prone to heat, insomnia and dreamful sleep,and so on. It has good clinical efficacy and safety, and its clinical curative effect is better than the positive control medicine Jingling oral liquid.

      摘要:Objective: To know the effects of Zhali Nusi fang(ZLNSF) on the Neurological function changes and its clinical efficacy assessment in patients with acute cerebral infarction. Method: Eighty-four patients with cerebral infarction were randomly divided into treatment group (42 patients)and control group(42 patients);the treatment group were given ZLNSF, the control group were treated with nimodipine tablets;all groups in the same basic treatment and time. The clinical efficacy,neurologic impairment scores and daily living ability index changes were observed before and after treatment. Result: The two groups were compared in the treatment of 1,7,14,28 days;the treatment group before and after treatment,the differences were significant (P < 0.05),while the treatment group was significantly better than the control group (P<0.05). Compared with the control group,the clinical efficacy of the treatment group were significant increased (P < 0.05).The treatment group total effective rate was 88.09%, 69.05% of the control group, the difference was significant;neurological deficit scores and activities of daily living index in both groups improved to some extent, particularly evident 1-14 days role in the treatment group (P<0.05). Conclusion: ZLNSF can promote the recovery of defects nerve function, improve the cerebral infarction after math of adverse events and activities of daily living decline, ZLNSF is a clinically safe and effective treatment of acute cerebral infarction, that should be widely applied.

      摘要:Objective: To probe the clinical therapeutic effects of Huoxue Qufeng Tongluo decoction (HQTD) combined with acupuncture on vascular headache. Method: Ninty-two vascular headache patients were randomly divided into HQTT group (n=46) with HQTT combined with acupuncture and control group (n=46) with Nimodipine for oral administration. 2 weeks is a therapeutic course. Therapeutic effects were measured and analyzed after 2 courses. Result: The therapeutic effect of HQTT groupwas significantly higher than that in control group (P < 0.05), with the effective rates being 91.30% and 73.91%, separately. The frequency, degree, lasting time and effects in the two groups were improved considerably compared with their own basic values of pre-treatment(P < 0.01,P < 0.05). Conclusion: Huoxue Qufeng Tongluo decoction combined with acupuncture is an effective method for treatment of vascular headache, and should be widely used in the clinic.

      摘要:Objective: To investigate the changes of inflammatory cytokines, renal vascular endothelial function in early diabetic nephropathy and the effect of salvianolate on intervention.Method: Sixty-four patients with early diabetic nephropathy were selected and randomly divided into 2 groups(routine group and salvianolate group).routine group was treated with insulin;the salvianolate group was treated with 200 mg·d-1 salvianolate combined with insulin. 30 healthy as control group.The endothelium-dependent flow-mediated dilatation(FMD)of brachial artery was determined with ultrasound,and the levels of soluble intercellular adhesion molecule-1(sICAM-1),endothelin-1(ET-1), interleukin-6(IL-6),tumor necrosis factor(TNF-α)and High-sensitivity C-reactive protein(hs-CRP)were measured respectively before and after treatment. Result: All patients with early diabetic nephropathy showed that the hs-CRP,TNF-α,IL-6,ICAM-1 and ET-1 was higher than that of the controlled group(P<0.05), the FMD were lower than controlled group(P<0.05). After treatment the hs-CRP,TNF-α,IL-6,ICAM-1 and ET-1 in the salvianolate group[hs-CRP(9.24±2.16)mg·L-1, sICAM-1(257.72±16.32) μg·L-1, ET-1(103.78±12.21) ng·L-1,TNF-α(14.62±7.23) ng·L-1,IL-6(16.12±5.68) ng·L-1]was decreased more significantly than routine group[(14.58±3.32)mg·L-1, (567.43±16.24) μg·L-1, (258.5±9.78) ng·L-1, (36.72±6.23)ng·L-1, (34.62±7.01)ng·L-1](P<0.05). After treatment the FMD in the salvianolate group was increased more significantly than routine group[(10.12±3.27)% vs(7.17±0.32)%](P<0.05). Conclusion: The salvianolate can effectively reduce the bodys inflammationrecovery and recovery renal vascular endothelial function with early diabetic nephropathy.

      摘要:Objective: To observe the clinical effect and mechanism of iontophoresis of Tongluo Zhitong powder articular cavity injection of sodium hyaluronate on the early and medial stages of the knee osteoarthritis. Method: Ninty-eight patients were randomly divided into observation group( n=49) and control group (n=49). Control group were treated with articular cavity injection of sodium hyaluronate, 20 mg each time, 1 time each week, for 5 weeks. Observation group were treated with iontophoresis of Tongluo Zhitong powder on the basis of control group, 2 times each day, for 5 weeks. The symptoms and signs of patients were tested by the Western Ontario and McMaster Universities (WOMAC) scores. The level of hyaluronic aci (HA), tumor necrosis factor-α (TNF-α) and interleukin-1β(IL-1β) of knee joint synovia in pretherapy and post-treatment were detected. Result: The total effective rate of observation group (97.96%) was superior to control group (81.63%)(P<0.01). The scores of WOMAC which including pain, stiffness, swell and tenderness of observation group were lower than control group (P<0.01). The level of HA in observation group was higher than control group and the level of TNF-α and IL-1β in observation group was lower than control group (P<0.01). Conclusion: The clinical curative effect of iontophoresis of Tongluo Zhitong powder combining with articular cavity injection of sodium hyaluronate on KOA is significant. The mechanism of action may be related with increasing the concentration of HA and decreasing the level of TNF-α and IL-1β.

      摘要:Objective: The purpose is to observe the curative effect of Fuzheng Xiaoai decoction in treatment of the breast cancer endocrine syndrome caused by tamoxifen (TAM) and its influence of life quality of breast cancer patients. Method: One hundred cases of patients were randomly divided into control group (50 cases) and observation group (50 cases) according to digital method. The patients in the control group took TAM, 20 mg/time, 1 time/day, orally. Based on the control group, patients in observation group took Fuzheng Xiaoai decoction additionally, 1 dose/day, continued for 2 months. If the symptoms are improved, the frequency is changed to 1 dose every two days, continued 2 months. All the patients were followed up for 3 months. The modified Kupperman score method is used to evaluate the clinical main symptoms integral. The quality of life of patients with breast cancer-scale Functional Assessment of Cancer Therapy-Breast(FACT-B) (4.0) is used to evaluate quality of life, record the treatment and follow-up treatment cases during the continued TAM treatment period. Result: After the treatment, the modified Kupperman score of observation group significantly decreased(P<0.01) and was lower than the control group (P<0.01). The symptoms integral period of observation group was 80.0%, which is better than 12.0% of control group (P<0.01). Disease control rates (DCR) of observation group was 88.0%, which is better than 72.0% of control group (P<0.05). The physiological condition and emotional dimension score in the quality of life scale, specific entry score and total score of observation group were significantly improved (P<0.01) and were higher than the control group (P<0.01). There are 6 cases (2.0%) in observation group cannot keep TAM treatment, which is lower than 19 cases (38.0%) of control group (P<0.05). Conclusion: Fuzheng Xiaoai decoction can reduce breast cancer endocrine syndrome symptoms, improve compliance of patient in TAM treatment and improve the life quality of patients. It can make patients benefit and be worthy of clinical use.

      摘要:Objective: The purpose is to study the left heart function, traditional Chinese medicine syndrome, the quality of life, amino-terminal fragment of pro-bnp(NT-proBNP) and the influence of inflammatory cytokines of the acute myocardial infarction (AMI) patients after(PCI) postoperative that took Tongxinluo capsules. Method: Sixty cases of patients with acute myocardial infarction after PCI were randomly divided into control group (30 cases) and observation group (30 cases). Patients in the control group took aspirin, 100 mg/time, 1 time/day, orally;Isosorbide mononitrate sustained-release tablets, 30 mg/time, 1 time/day, orally;Clopidogrel 75 mg/time, 1 time/day, orally;Atorvastatin calcium 20 mg/time, 1 time/day, orally. Based on the control group, patients in the observation group additionally took Tongxinluo capsules, 4 capsules/time, 3 times/day. The treatment lasted for 3 months. The levels of left ventricular function, the detection of serum tumor necrosis factor-α (TNF-α), interleukin 6 (IL-6), hypersensitive c-reactive protein level (hs-CRP) and the NT-proBNP were monitored. The pneuma deficiency and blood stasis and Seattle angina questionnaire (SAQ) score were recorded, as well as the major adverse cardiac events happening within 6 months. Result: The improvements left ventricular end-diastolic diameter (LVEDD), left ventricular contraction at the end of the inner diameter (LVESD), left ventricular ejection fraction (LVEF) and the stroke volume (SV) of observation group are all better than those of control group (P<0.05 or P<0.01). The levels of serum NT-proBNP, TNF-α, IL-6 and hs-CRP of observation group are lower than the control group (P<0.01). The score of pneuma deficiency and blood stasis of observation group is lower than the control group (P<0.01), while the SAQ score of observation group is higher than that of control group (P<0.01). The cumulative incidence of the major adverse cardiac events of observation group is 13.3%, which is lower than 30.0% of the control group (P<0.01). Conclusion: Tongxinluo capsule can improve the cardiac function and ventricular remodeling of patients with acute myocardial infarction after PCI, relieve the clinical symptoms, improve the quality of life and reduce the incidence of adverse cardiac events. Patients can benefit from multiple aspects so that it is worthy of clinical use.

      摘要:Objective: The purpose is to study the mechanism of Baihua Danshen decoction in treatment of coronary heart disease (CHD) from the view point of endothelial function. Method: Eighty-eight cases of patients were divided into control group (44 cases) and observation group (44 cases) according to the hospital sequence. Based on 'The guide to the diagnosis and treatment of chronic stable angina', the patients in control group took conventional treatment, including anticoagulation, antihypertensive, fat adjustment and improvement of coronary blood flow. On the basis of control group, the patients in observation group took Baihua Danshen decoction, 1 dose/day. The treatment course is 12 weeks. Ultrasound is used to evaluate the function of flow-mediated dilatation (FMD). The levels plasma total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) were detected before and after treatment. Ria method is used to determine the levels of plasma endothelin-1 (ET-1), thromboxane 2 (TXB2) and 6-ketone prostaglandin F1a (6-Keto-PGF1a). The nitrate reduction method is used to evaluate nitric oxide (NO) level. The times and duration of angina are recorded. Result: During the treatment, the times and duration of angina in observation group were less than the control group (P<0.05). After the treatment, the levels of TC, TG, LDL-C of both groups were decreased, while observation group is lower than the control group (P<0.01). The level of HDL-C of two groups was increased, while the observation group was higher than control group (P<0.01). The levels of TXB2, ET-1 and ET-1/NO ratio were declined after the treatment, while the observation group are lower than the control group (P<0.01). The levels of NO and 6-Keto-PGF1a were increased after the treatment, while the observation group was higher than control group (P<0.01). The improvement of FMD in observation group is better than that of control group (P<0.01). Conclusion: Baihua Danshen decoction can improve angina pectoris of patients with CHD and regulate fat metabolism. Its mechanism may be related to adjust the imbalance of ET/NO and TXB2/6-Keto-PGF1a state and improve vascular endothelial function of patients with CHD.

      摘要:Objective: In order to provide inspiration and methods for clinical keratitis prescription,using cluster analysis of past dynasties' keratitis recipes based on data mining technology. Method: Three hundred and thirty four pieces of prescriptions treating of keratitis, Collected from the jin dynasty to the beginning of this century are entered into database for cluster analysis of their medication and prescription drug laws. Result: The drug characteristics of eliminating evil by clearing heat, dispelling wind, removing dampness, dissolve shade, detoxification, cooling blood, soothing the liver, calming wind, and strengthening body by improving eyesight, nourishing kidney, adding yin, nourishing the liver are concluded form 26 groups of keratitis drug clusters explored. Conclusion: By the use of cluster analysis, traditional Chinese drugs can be well classified according to the trend of drug action to guide clinical medication. The findings objectively reflect the keratitis drug laws and provide a guideline for keratitis standardized treatment research.

      摘要:Objective: To analyze the prescription experience of professor LI Sai-mei for treating diabetes based on the theory of structural partial-ordered attribute diagram. Method: First, we deal with the decision formal context of prescriptions in 200 medical cases about diabetes mellitus in professor LI Sai-mei's clinical working based on principle of the optimization. Then, we generate a new partial-order attribute diagram in order to present the specific c haracter. Finally, we explain properties of partial order structure graph from TCM expert's point of view based on knowledge discovery. Result: Professor Li treated diabetes concentrates on invigorating spleen and kidneys and regulating liver-qi. Conclusion: The method proposed in this paper works well in treating of the description of differentiation of syndromes and discovering new knowledge of clinical diagnosis from the known data.

      摘要:In this article,the research on chemical constituents and bioactivities of plants from Aconitum carmichaeli in recent years has been reviewed. A systematic literature survey was conducted to classify and summarize chemical compositions of A. carmichaeli,and sum up current studies on the main pharmacological activities. The main constituents of this plant are alkaloid, among which C-19 diterpenoid alkaloids are the leading compounds with aconitine-type skeleton,followed by C-20 diterpenoid alkaloids, with such types skeleton as hetisan, veatchine, napelline and denudatine. Besides,A. carmichaeli also contains flavonoid,saponin,polysaccharide,fatty acid,et al.
