      摘要:AIM:To study the preparation process of extraction and purification for analgesic and hemostatic bioactive portion from Lamiophlomis rotata (Benth.) Kudo.METHOD:The quantitative analysis was performed by HPLC and dimethylaminobenzaldehyde method.By Using single factor and orthogonal experiments,the best extraction and purification process with macroporous resin was optimized.RESULTS:The best extraction was established.solvent:60% ethanol.soak:36 hours.elution volume:10 times of materials.percolation rate: 5ml.min-1kg-1.Pufification process:The ethanolic extract should be separated by polyamide.Then separate the water elution with XDA-7 macroporous resin and collect the 30% ethanolic extract.CONCLUSION:XDA-7 macroporous resin has better ability of absorption and anti-absorption for iridoid glycosides and 30% ethanol was the best solvent for anti-absorption.

      摘要:The objective of the current study was to determine the optimum ultrasonic extraction conditions for hypericin from Hypericum perforatum L. The optimum conditions were evaluated with 2-level fractional factorial design and optimized using response surface methodology. High yields of hypericin were observed from Hypericum perforatum L. by ultrasonic extraction technology. The extraction time, acetone concentration and the ratio of solvent to material were the most significant parameters on hypericin extraction as evaluated through FFD. The extraction conditions were further investigated with central composite design. The results showed that the optimal parameters for this method were: extraction time of 36.15 min, acetone concentration of 68.14(Vol) of acetone/ethanol solvent, and ratio of solvent to material of 11.87 mL?g-1. Under the optimized condition, the maximum productivity of hypericin is 1.44%. The yield of hypericin under the optimized extraction conditions were great higher than that obtained from conventional solvent extraction. The present study demonstrates that ultrasonic is a great efficient tool for the fast extraction of hypericin from Hypericum perforatum L.

      摘要:[Abstrct] Objective: To optimize the extraction technology for Jianshen Pian. Method: The shape of Radix Astragalus was studied on the content of astragaloside Ⅳ,and orthogonal test was used for studying the effect of water volume , extraction time and times on the content of Icariin,astragaloside Ⅳ and solid.Result: The optimal extraction process is as follows: with 8 times of water extracted twice, each time for 1.5 h, and processing method of Radix Astragali is cutting for about 5~10mm . Conclusion: The extraction process is reasonable and feasible. The method of content determination is accurate and reliable.

      摘要:Objective To study the absorption characteristics of Notoginsenoside R1 influenced by density, energy and p-glycoprotein with rat non-everted gut sac system. Methods Different concentration of Notoginsenoside R1 was immersed into Rat non-everted gut sac, and the absorption difference was compared. 2-nitropenol, a metabolic inhibitor, was used to study the influence of energy on the absorption of Notoginsenoside R1. Verapamil, a p-glycoprotein inhibitor, was used to study the influence of p-glycoprotein on the absorption of Notoginsenoside R1. Results The Notoginsenoside R1 concentration in the sac was increasing according to the time. It is obvious that 2-nitropenol affect the absorption of Notoginsenoside R1. Verapamil had no effect on the absorption of Notoginsenoside R1 in sac system. Conclusion The absorption mechanism of Notoginsenoside R1 might be passive diffusion. Notoginsenoside R1 might not be the substrate of p-glycoprotein.

      摘要:[Abstract]: Objective: To choose the optimal extracting process of cordyceps militaris by multi-index synthetic evaluation method. Methods: Using Cordycepin, adenosine, uridineas, contents of polysaccharides and paste rates as detecting index, orthogonal test method was used for studying with four extracting factors (the solid liquid ratio, alcohol concentration, the extracting time, the times of extracting) on the extracting process. Results: The optimum condition of cordyceps militaris were eight times of water, extracting one hour, two times. Conclusion: The study optimized extraction technology of cordyceps militaris and provide the experimental basis for it’s further use and product development.

      摘要:Objective: To study the safety and solubilization of excipients solubilizing excipients used in injections. Method: Solubilization was measured by HPLC and hemolysis and irritation were used for the safety evaluation of excipients. Result: Safety and solubilization of solubilizing excipients were different. Conclusion: As more objective screening for injection solubilizing excipients, it should be researched into the preparation.

      摘要:Objective: To research Yinhuang decoction and its five stage membrane filter liquor, to analyze the content change of baicalin and other two kinds effective composition and in vitro effect of bacteriostasis. Method: Multi-stage membrane separation by molecular size was used to separate Yinhuang decoction. Ingredients were determined by HPLC. Staphylococcus aureus and Streptococcus pneumoniae were used to test the minimum inhibitory concentration (MIC) of source liquid and all levels filter liquor. Result: In vitro effect of bacteriostasis from high to low order was four, three, five, two, source, one level filter liquor. Conclusion: Membrane separation method can be used to isolate antimicrobial activity substances by section. The antibacterial effect of membrane filter liquor after microfiltration is stronger than source decoction.

      摘要:Objective: To reveal the impact of process on dissolution rate of calcium of traditional Tibetan medicine Nanhanshuishi by determining various processed Nanhanshuishi decoctions. Method: The calcium content in various Nanhanshuishi (crude, water quenched, liquor quenched,and yoghurt quenched) was determined by EDTA titration. Result: The dissolution rate of calcium of traditional Tibetan medicine Nanhanshuishi was raised after being processed, the order of dissolution rate of calcium of processed Nanhanshuishi was the yoghurt quenched>the liquor quenched>the water quenched. Conclusion: The study has provided basis for evaluating quality, clarifying the processing mechanism and regulating the processing technology of traditional Tibetan medicine Nanhanshuishi.

      摘要:Objective: To study the preparation process of extraction and purification for analgesic and hemostatic bioactive portion from Lamiophlomis rotata. Method: Determination of the total iridoid glycosides and 8-epideoxyloganic acid were performed by dimethylaminobenzaldehyde method and HPLC respectively. By Using single factor and orthogonal experiments, the best extraction and purification process with macroporous resin was optimized. Result: The best extraction was established as follows: solvent of 60% ethanol, soaking 36 hours, elution volume was 10 times of materials, percolation rate of 5mL ·min-1kg-1. The purification process was as follows: the ethanolic extract should be separated by polyamide. The water elution was separated with XDA-7 macroporous resin and collecting the 30% ethanolic extract. Conclusion: XDA-7 macroporous resin has better ability of absorption and anti-absorption for iridoid glycosides and 30% ethanol was the best solvent for anti-absorption.

      摘要:Objective: To study the in vitro percutaneous absorption and in vitro release state of the main effective element colchicine in Fengling gelatin. Method: The Franz diffusion cells were chosen with excised SD rat skin and dialyzer as the in vitro permeability barrier; the content of colchicine was determined by HPLC, the cumulative permeation amount per unit area and release amount of colchicine were calculated, and the percutaneous absorption velocity and release velocity were compared. Result: The in vitro release and penetration process of colchicine in Fengling gelatin conformed to Higuchi kinetic equation process within 8h; the release amount of colchicine in Fengling gelatin was higher than cumulative permeation amount, and the release velocity was higher than percutaneous absorption velocity. Conclusion: Skin was the main percutaneous absorption barrier of colchicine in Fengling gelatin. The active constituent could be released steadily, which showed that Fengling gelatin was a prolonged action lente liberantes praeparatum.

      摘要:Objective: To screen the optimal formula for total flavonoids dispersible tablets of hawthorn leaves. Method: Different kinds of excipients in the prescription were selected and their amount was optimized by using disintegration time as parameters.The optimal composition of the prescription was determined by orthogonal design experiment. Result: The optimal formulation was as follows:16% total flavonoids extracts of hawthorn leaves,55% MCC,8% cCMC-Na,18% starch and 1% magnesium stearate.The disintegration time of dispersible tablet was (66±5)s. Conclusion: The dispersible tablet had reasonable prescription and its dissolution rate was much better than the common tablet of hawthorn leaves total flavonoids in the market.It would be a product worthy of exploitation and utilization.

      摘要:Objective: To study the preparation method for baicalin microcapsule using gelatin gum as sealing materials. Method: Baicalin microcapsule was prepared by simple coacervation method. The uniform design was adopted to optimize the preparation procedure with drug loading as evaluation indexes. Result: The optimum preparation technique of baicalin microcapsule was: the ratio of core material to coating material was 1 ∶2; the temperature of encapsulation was at 45 ℃ and the stirring speed was 550 r ·min-1. Conclusion: The results showed that baicalin microcapsule had appropriate diameter, distribution of microcapsule particles size and the dissolution meets the requirements.

      摘要:Objective: To optimize the processing technology of Panacis Quinquefolii Radix. Method: On the basis of actual production, four factors in the rinsing and softening process were examined, that is,rinsing time, rinsing interval, rinsing water amount and softening time.The three levels were set in every factor,and the experiments were arranged with orthogonal design.The optimal rinsing and softening conditions were screened on the basis of qualified product percentage and the total saponin content. Result : The optimal processing conditions are as follows:the rinsing time is 3 hours,the rinsing interval is 30 minutes,the amount of water used is four times of weight of the medicinal materials,and the softening time is 50 seconds with 2 450 MHz microwave. Conclusion :The Panacis Quinquefolii Radix qualified product percentage and the saponin content is high with the aptimal processing.

      摘要:Objective: To study the chemical constituents in Trachelospermi jasminoides. Method: The compounds were isolated and purified by recrystallization and column chromatography with silica gel,ODS and Sephadex LH-20.Their structures were elucidated on the basis of physicochemical properties and spectral analysis. Result: Ten compounds were identified as trachelogenin(Ⅰ),arctigenin(Ⅱ),nortrachelogenin(Ⅲ),arctiin(Ⅳ),tracheloside(Ⅴ),matairesinoside(Ⅵ),nortracheloside(Ⅶ),nortrachelogenin-5’-C-β-glucoside(Ⅷ),nortrachelogenin-8’-O-β-glucoside(Ⅸ) and tanegoside A(Ⅹ) respectively. Conclusion: Compound Ⅹ was isolated from Trachelospermum jasminoides for the first time.

      摘要:Objective: The volatile oils of Fructus Aurantii Immaturus are analyzed to verify whether the chemical constitutes are various in different species. Method: The Fructus Aurantii Immaturus samples of Citrus aurantium and Citrus sinensis are collected from Chongqing,and each species have two batches.The volatile oil which obtained by the method of water steam distillation are analyzed by GC-MS,then the chemical constitutes of volatile oil are identified by Data Base Retrieval and by comparing with the relative standard mass spectrum data,and the relative amount of each constitute is calculated by area normalization method. Result: Twenty-nine constitutes are identified in two batches of C. aurantium,and account for 97.4% in total oil.Thirty-eight constitutes are identified in two batches of C.sinensis, and account for 99.07% in total oil.Among them,twenty-four constitutes are common in the two species,and nineteen constitutes are different.Meanwhile,the content of each constitute exhibit some different between the two species.In the same species of Fructus Aurantii Immaturus,the chemical constitutes are no different in two batches. Conclusion: The chemical constitutes are obviously different between C.aurantium and C.sinensis.

      摘要:Objective: To determine the content of puerarin in the concentrating process of Xiaoke pills rapidly by near-infrared spectroscopy. Method: Near-infrared transmissionspectroscopy technology and fiber optic probe were used to collect NIR spectra of Xiaoke pills concentration liquid.With HPLC analysis results as reference,the fast determination method of puerarin was established with partial least squares (PLS). Result: The R2,RMSECV and RPD of the calibration model for puerarin were 0.955 9,0.241 and 4.36,respectively.The average relative deviation of the predication set was 1.8%. Conclusion: The method is accurate,fast and simple,which could be generalized to the on-line quality control of Xiaoke pills concentrating process.

      摘要:Objective: To develop a RP-HPLC method for determination of mangiferin and spinosin in Ziziphus Jujuba Mill granules. Method: Samples were purified by Liquid-Liquid extraction.Hypersil C18(4.6 mm×250 mm,5 μm) column was used.The mobile phase consisted of acetonitrile and water with 1% glacial acetic acid;the flow rate was 1.0 mL ·min-1.Detection wavelengths were set at 258 nm,and switched to 335 nm at 25 min. Result: The linear response ranges were 4.320×10-3-5.200×102g ·L-1 (r=0.999 4) and 5.200×10-3-1.040×10-1g ·L-1 (r=0.999 7)for mangiferin and spinosin respectively.The average recoveries (n=9)of these compounds wre 94.6% (RSD 1.9%) and 95.1%(RSD 2.3%)respectively. Conclusion: A simple,accurate and reproducible method is provided for simultaneous determination of mangiferin and spinosin in Ziziphus Jujuba Mill granules by RP-HPLC.

      摘要:Objective: To establish an HPLC-ELSD method to determine the content of astragaloside I,II and acetylastragaloside I in Astragalus extract. Method: The HPLC-ELSD system consisted of Welc C18 column (4.6 mm×250 mm,5 μm) with the mobile phase consisting of acetonitrile-water using a gradient elution.The flow rate was 1 mL ·min-1 and the column temperature was at 30 ℃.The drift tube temperature of ELSD was 80 ℃ with a carrier gas (air) pressure of 50 psi. Result: The average recoveries of astragaloside I,II and acetylastragaloside I were 101.16%,99.34% and 100.43% respectively,with RSD of 2.76%,2.28%, 2.55%.The content of astragaloside I,II and acetylastragaloside I was 4.01%,1.70%, 3.67% respectively. Conclusion: It was proved that the newly-established HPLC-ELSD method was simple,reliable,well-separated and precise,which could be used for quantitative analysis of saponins in Astragalus extract.

      摘要:Objective: To study pattern recognition of the HPLC fingerprint of Rhizoma Panacis Majoris. Method: Twelve batches of samples,Rhizoma Panacis Majoris,come from seven provinces were carried out to gain fingerprints by HPLC,The pattern recognition of the characteristic fingerprint of Rhizoma Panacis Majoris,known as cluster analysis and principal component analysis formed. Result: The fingerprint of 12 batches of Rhizoma Panacis Majoris showed 8 characteristic peaks,in which 2 common peaks were confirmed.the samples were grouped into 2 types. Conclusion: Different geographical environment and climaties lead to differences between the quality of Rhizoma Panacis Majoris,the pattern recognition of the fingerprint provides the experimental basis for manufacture and quality control of Rhizoma Panacis Majoris.

      摘要:Objective: To determine the content of dendrobine in Dendrobium nobil from different collection periods,to discover the inherent rule about the content change owing to different collection periods. Method :Determinate the content of dendrobine in D. nobil from different collection periods by gas chromatography. Result: The content of dendrobine varies in different harvest time,with 0.239% at the lowest point in September and 0.559% at the highest point in January. The dendrobine content of the annuals is 0.546%,which is the largest among that of different harvest years. Conclusion: This offered evidence to determine the most appropriate harvest time for D. nobil .

      摘要:Objective: The aim is to establish chromatographic fingerprint of Morinda officinalis in different harvest period by HPLC in order to control its quality. Method: A gradient elution mode was applied on a column ODS-C18 (4.6 mm×250 mm,5 μm) at 30 ℃ with methanol-0.4% phosphoric acid as mobile phase.The detection wavelength was at 360 nm.The flow rate was 1.0 mL ·min-1. Result: Samples from different harvest period were high similarity,but fingerprint graphics showed evident difference. Conclusion: The results of peak areas were in accordance with the request of fingerprint.The established fingerprint can be used for the quality control M.officinalis in different harvest period.But their ratios between each other are difference.

      摘要:Objective: To establish a method for determining 6-gingerol in Antiemetic compound particles. Method: 6-gingerol in decoction of Antiemetic compound particles was determined by HPLC.The column was Agilent TC C18 (4.6 mm×250 mm,5 μm) with methanol-water (65 ∶35) as mobile phase.The flow rate was 0.8 mL·min-1,and the column temperature was at 30 ℃.The detection wavelength was set at 280 nm;injection volume was 10 μL. Result: 6-gingerol was linear in the range of 0.113 5-2.270 μg and the regression equation was Y=540 646X-1 188 (r=0.999 9,n=6).The average recovery was 99.90%,RSD 1.14%. Conclusion: The method is simple and reproducible for the determination of 6-gingerol in decoction of Antiemetic compound particles by HPLC.

      摘要:Objective: To determine gentiopicrin in Fufang Yinchen Syrup by HPLC. Method: Samples were determined on a Diamond C18 (4.6 mm×150 mm,5 μm),the mobile phase consisted of methonal-water (25 ∶75)with flow rate at 1 mL·min-1,UV detection wavelength at 270 nm,column temperature at 25 ℃ and sample size at 10 μL. Result: The linear ranges of gentiopicrin was 1.257 5-12.575 μg (r=0.999 8),the average recovery were 99.24% (RSD 1.84%). Conclution: The method is sensitive,simple,accurate,and it can be used for the quality control of Fufang Yinchen Syrup.

      摘要:Objective: To establish an HPLC method for content determination of total scopoletin in wild and cultivated Cortex Lycii from different areas. Method: The separation was performed on a column C18, the mobile phase was acetonitrile-1% acetic acid (15 ∶85);the flow rate was 0.8 mL ·min-1;the column temperature was at 30 ℃; the detective wavelength was at 345 nm. Result: The linear range of scopoletin was from 6 to 48 mg ·L-1.The average recovery was 97.19% (RSD 1.77%). Conclusion: The method is reliable,accurate and has a good specificity.

      摘要:Objective: To establish the fingerprint chromatogram of Lonicera dasystyla,and to probe and research the establishment method and application to the quantitative fingerprint chromatogram. Method: RP-HPLC was used.The samples were separated on a C18ODS2 column (4.6 mm×250 mm,5 μm) and detected at 328 nm with methanol,acetonitrile and 0.05% phosphoric acid as mobile phase by gradient elution.The column temperature was set at 25 ℃.The flow rate was 0.5 mL ·min-1. Result: The fingerprint chromatogram based on the data of 12 batches of Lonicera dasystyla was set up. Conclusion: The fingerprint chromatogram of Lonicera dasystyla can be used to recognize and access other samples as a standard.

      摘要:Objective: To establish a RP-HPLC method to determine the content of aesculetin in Qingre Kasen Keli. Method: The RP-HPLC method was used.The analytical column was KromasilMC18 (4.6 mm×250 mm,5 μm),The mobile phase was acetonitrile-0.1% phosphoric acid (10 ∶90).The flow rate was 1.0 mL ·min-1.The detector wavelength was at 346 nm;injection volume was 10 μL. Result: The linear range was within 0.010 1-0.253 2 μg (r=0.999 9).The average recovery was 99.1% (RSD 1.14%). Conclusion: The method is simple,accurate and sensitive,and can be used for quality control of Qingre Kasen Keli.

      摘要:Objective: A gel permeation chromatographic method was developed for the extraction of 7 pesticides from Chinese wolfberry. Method: Quantity and quality determination method were developed by gas chromatography with mass spectrometry. Result: In the linear range 0.05-1.00 mg ·L-1 of each pesticide,the correlation coefficient was higher than 0.99.The average recoveries were from 79.3% to 109.3% with the RSD 2.39%-10.33% (n=6).The limits of detection (LOD) for these pesticides were from 2.1-36.7 μg ·L-1. Conclusion: The method developed is reliable,accurate and can be applied for quantitative and qualitative analysis of 7 pesticides from Chinese wolfberry.

      摘要:Objective: To establish a method for simultaneously determining geniposide and baicalin in Niuhuangqingwei pills. Method: A double wavelength RP-HPLC method was developed.The analysis was performed on a Hibar C18 column (4.6 mm×150 mm,5 μm) with gradient elution using 0.2% phosphoric acid (A) and acetonitrile (B) in the order of 0-9.00 min 15%B and 9.00-10.00 min 15%-25%B, after 10.00 min,25%B,the flow rate was 1.0 mL ·min-1.Geniposide and baicalin were detected at 240 nm and 278 nm respectively.The column temperature was at 30 ℃. Result: The calibration curves of geniposide and baicalin showed good linearity in the ranges of 0.16-0.80 μg and 0.69-1.61 μg.The average recoveries were 98.62% and 96.93% with RSD of 1.97% and 2.12% respectively. Conclusion: The method is simple,accurate and reproducible for simultaneous determination of geniposide and baicalin in Niuhuang Qingwei pills.

      摘要:Objective: To analyze the difference of the saponins in the different processed products of Radix Polygalae(crude Radix Polygalae,licorice And Honey Processed Radix Polygalae). Method: The content of polygalacic acid and senegenin in the different processed products of Radix Polygalae was determine by HPLC using an Agilent ZORBAX SB-C18(4.6 mm×250 mm,5 μm) with acetonitrile-0.1% phosphoric acid (41 ∶59) as the mobile phase; the flow rate was 1.0 mL ·min-1; temperatures was 25 ℃ and UV detection wavelength at 210 nm. Result: The order of the content of polygalacic acid and senegenin (from higher to lower) was:crude Radix Polygalae, licorice processed Radix Polygalae,honey processed Radix Polygalae. Conclusion: The different process method has some effects on the content of saponins in Radix Polygalae. The content of polygalacic acid and senegenin in honey processed Radix Polygalae was the lowest. The LD50 experiment result suggests that honey processed can reduce the toxic composition of crude Radix Polygalae .

      摘要:Objective: To study the chemical constitutes of Smilax china and the cytotoxic activities of isolated compounds. Method: Various chromatographic techniques were used to separate and purify the constituents.Their structures were established on the basis of their physico-chemical properties and spectral data.The isolated compounds were evaluated for their cytotoxic activities by MTT method. Result: Six steroidal saponins were isolated from the dried rhizomes of S.china. Their structures were established on the basis of spectroscopic evidence (NMR,MS) as borassoside B (1),dioscin(2),protodioscin (3),22-O-methyl protodioscin(4),isonarthogenin 3-O-α-L-rhamnopyranosyl (1-2)-O- -β-D-glucopyranoside (5),diosgenin-3-O- -β-D-glucopyranoside(6).Compounds 2 and 6 showed significant cytotoxic activities against K-562 carcinoma cell line at the concentration of 5 mg ·L-1.The inhibitory percentage of compounds 2 and 6 on K-562 cell growth were 89.1% and 90.1% at concentration of 5 mg ·L-1. Conclusion: Compounds 1 and 6 were isolated from this plant and Smilax for the first time.Compounds 2 and 6 showed cytotoxic activities against K-562 carcinoma cell line in vitro.

      摘要:Objective: The volatile compounds from the leaves of Hypericum perforatum were studied. Method: Solid-phase micro-extraction (SPME) was used to extract the volatiles.The chemical constituents were analyzed by gas chromatography-mass spectrometry (GC-MS) coupled with Kovats index.A quantitative analysis in percent was performed by peak area normalization measurements. Result: Forty-one compounds (97.03%) were identified from the volatile constituents in the leaves of H.perforatum. Conclusion: The main constituents were γ-muurolene (12.70%),α-curcumene (11.36%),δ-cadinene (9.09%) and β-trans-farnesene(6.95%).

      摘要:Objective: To analyze the appropriate producing area of Capparis spinosa. Method :The TCMGIS(suitability evaluation geography information system of traditional Chinese medicine producing area)was used to analyze the appropriate producing area of C. spinosa basing on the optimum ecological factors of Turpan,east Xinjiang autonomous region and South Xinjiang autonomous region,the traditional roducing areas of C. spinosa. Result: The results showed that the suitable producing areas of C. spinosa included 183 counties of 5 autonomous regions and provinces,i.e. Xinjiang,Inner-Mongolia,Ningxia autonomous regions,and Gansu as well as Qinghai provinces. The total area was 1 251 361 km2 and distributed mainly in eastern and southern part of Xinjiang,Aalashan of Inner-Mongolia,northern part of Gansu and in the middle of Ningxia. Conclusion: The suitable producing areas based on TCMGIS covered all areas registered in the 3 rd national investigation of Chinese traditional medicine resource. And the results were also corresponding to the traditional producing area of C. spinosa recorded in ancient literature and the successful cultivation areas nowadays. It has a important reference value for cultivation and science division of C. spinosa.

      摘要:Objective: To identify the microscopic identification of dry mature seeds of Prunus persica(L.) Batsch and Prunus davidiana(Carr.) Franch. Method: Research the features of tissue and powder of P. persica and P. davidiana using the method of microscopic identification. Result: In tissue slices of peach seed, the seed coat cell lateral contains a lot of stone cells, the endosperm cell layer is relatively narrow; In powder slices there contains some features about the epidermal cells of seed coat, cotyledons cells, catheter cell and so on. Conclusion: The stone cells,the epidermal cells of seed coat, the endosperm and the cotyledons cells, the catheter cells, the aleuronic grain could be used as identification characters through the research. Provide scientific basis for the identification, quality evaluation, development and utilization of the peach seed.

      摘要:Objective: To synchronously investigate the effects of Shouwu Fang on the pharmacokinetics of levodopa (L-DOPA) in blood and in extracellular fluids of striatum in rat with Parkinson’s disease (PD),by simultaneous blood-brain microdialysis in freely moving rats. Method: SD rats were randomly divided into four groups:control group,model group,L-DOPA group (ip L-DOPA 24 mg ·kg-1and benserazide 6 mg ·kg-1),SWF+L-DOPA group (ip L-DOPA pretreated with ig SWF).The rat model of Parkinson’s disease was induced by injecting 6-hydrodopamine (6-OHDA) into striatum of rats,blood-brain microdialysis technique was used for simultaneously sampling.High performance liquid chromatography-fluorescence detector (HPLC-FLD) and high performance liquid chromatography-electrochemical detector (HPLC-ED) were used to detect the concentration of L-DOPA in blood and striatum.The data were analyzed with DAS program. Result: Compared with L-DOPA group,the blood L-DOPA concentration of SWF+L-DOPA group was increased significantly at 6 time points,the concentration in striatum was decreased at 15 min after drug administration.The blood pharmacokinetic parameters of L-DOPA group and SWF+L-DOPA group were as follows:AUC(1 627.7±420.57) mg ·L-1 ·min-1,(2 626.44±980.6) mg ·L-1.min-1,Tmax:30 min,(37.5±8.22) min,MRT(0-t);(71.98±3.19) min,(83.44±9.53) min,striatum pharmacokinetic parameters:Tmax:(50±15.49) min,(81±27.25) min,MRT(0-t);(68.65±15.39) min,(107.91±26.66) min.Compared with L-DOPA group,SWF+L-DOPA group blood AUC was increased (P<0.05),Tmaxand MRT(0-t) were delayed (P<0.05);striatum Tmaxand MRT(0-t) were delayed too (P<0.05). Conclusion: Shouwu Fang delayed L-DOPA elimination,increased its absorption in blood,and reduced fluctuation of concentration in striatum.

      摘要:Objective: To study the intestinal absorption of the different match-pair of Coptis-Evodia,to compare the intestinal absorption rate and absorption coefficient. Method: Single pass perfusion models in rats and HPLC were used to study the concentration and absorption coefficients of match-pair of Coptis-Evodia,to study the absorption and transport in the small intestine. Result: The absorption rate of Coptis-Evodia 6 ∶6 were the largest in jejunum as Coptis-Evodia 6 ∶1 were the largest in Jejunum and Ileum,the difference was significant (P<0.05).The Ka of Coptis-Evodia 1 ∶1 in jejunum,kleum and colon was larger than that in Duodenum as the Kapp was the largest in ileum;the absorption rate of Coptis-Evodia 2 ∶1 and 6 ∶1 in ileum were larger than the other intestine,the difference was significant (P<0.05).The absorption rate of Coptis-Evodia 2 ∶1 in lleum were larger than the other match-pair,the difference was significant (P<0.05). Conclusion: Coptis-Evodia 1 ∶1 and 6 ∶1 were better absorpted in jejunum and ileum,Coptis-Evodia 2 ∶1 was better absorpted in ileum.

      摘要:Objective: To investigate pharmacokinetics of Xiongbing Microemulsion in rabbit blood. Method: The plasma concentration of tetramethylpyrazine (TMP) was determined by HPLC after i.v.The data were processed by DAS 2.1 software to calculate the pharmacokinetic parameters. Result: The calibration curve was linear in the range of 0.062-12.4 mg ·L-1 (R2=0.998).The measured pharmacokinetics of free TMP concentration fitted open two compartment model after i.v.The main pharmacokinetic parameters for TMP were as follows:t1/2α was 15.617 min;t1/2was 49.171 min;and V was 0.509 L ·kg-1;CL was 0.014 L ·min-1 ·kg-1;AUC(0-∞)was 222 063.235 μg ·min ·L-1;MRT (0-∞)was 30.841 min. Conclusion: With good linearity,precision and recovery,the method is sensitive and simple,and suitable for pharmacokinetic study and the research of TMP.The pharmacokinetic characteristics of TMP in rabbits were understood.

      摘要:Objective: To study the time-quantity relationship of index components of Huanglian Jiedu(HLJD) and its connection with the effect in ischemic brain injury in rats with syndrome of fire-heat after oral of Huanglian Jiedu decoction. Method: By electrocoagulation established model of focal cerebral ischemia with syndrome of fire-heat, after treatment group were orally administered Huanglian Jiedu decoction, catch blood at different time are as followes, 0.083, 0.25, 0.5, 0.75,1, 2, 3, 4, 6, 8, 12, 24 h, determination of index components in rats draw the time-quantity curve; Catched brains at the points mentioned above, determinated the changes of IL-6, TNF-α, and draw the time-effect curve. Combined with time-quantity curve and time-effect curve, evaluate the function of index components in Huanglian Jiedu decoction treat ischemic disease with Syndrome of fire-heat. Result: Pharmacokinetics of index components of HLJD are different, which have a effect on its pharmacodynamics. Geniposide made a marked effect at prophase times after administration,while baicalin and baicalein made a lasting effect. Conclusion: Huanglian Jiedu decoction to play anti-ischemic brain with syndrome of fire-heat injury, the function connected with index components relevant content in the body, different components producted various time bar.

      摘要:Objective: To compare pharmacokinetics of matrine between colon target pellets of Sophora Flavescens and solution,make clear the pharmacokinetic character of the targeted agents in vivo. Method: RP-HPLC was applied to determine the content of matrine in plasma and colon contents after the administration and DAS pharmacokinetic software was used for calculating the acquired PK parameters. Result: The parameters of matrine contained in pellets in plasma and colon contents were all differed from those in solution,such as tmax,Cmax,AUC ad.There was obviously delay effect on absorbability of matrine in pellet,and most of that was not absorbed into systemic circulation,and then high concentration distribution was found in colon. Conclusion: The pellets of Sophora Flavescens prepared show good colon-specific distribution characteristics,and achieve the expected design goal.

      摘要:Objective: To study the influence of cream processing on metabolism and tissue distribution of bitter almond in rat. Method :Amygdalin was set as control group given by mainline,bitter almond and its cream product were given to SD rats by oral,HPLC was employed for the determination of amygdalin and its metabolite in blood,heart,liver,spleen,lung and kidney,and LC-MSn was used for further identification of the metabolite structure. Result: Amygdalin was distributed in the tissues as prototype after given by mainline,while in the groups of oral administration,it was distributed as the secondary metabolite prunasin;and compared to bitter almond crude material,the time of prunasin distribution in the tissues was prolonged after cream processing,and showed two peaks phenomenon. Conclusion: Cream processing have a significant effect on the metabolism and tissue distribution of bitter almond,this provides information for the mechanism of bitter almond’s processing.

      摘要:Objective: Analysis the difference of urine metabolites between hypertension patients with the syndrome of ascendant hyperactivity of liver yang and healthy volunteers,search the possible biomarker,reveal the nature of the syndrome of ascendant hyperactivity of liver yang. Method: According to the clinical diagnostic criteria of the syndrome of ascendant hyperactivity of liver yang,select typical cases,select nine typical cases and eleven healthy volunteers,analysis the components in the urine derivatives of hypertension patients with the syndrome of ascendant hyperactivity of liver yang and healthy volunteers by gas chromatography mass spectrometry,match and align the data,select the same features,make the PCA and PLS-DA by the software Simca-P,track biomarkers and explain the possible relevant metabolic pathways. Result: The data from the analysis by PCA and PLS-DA of the urine samples shows that:two sets of data from GC-MS can be classified in the score plot.Some significantly changed metabolites like stearic acid,inositol,D-fructose,phthalic acid have been identified between the two groups in terms of relative metabolism of body. Conclusion: The metabonomics method,gas chromatography-mass spectrometry combined with pattern recognition,has advantages in studying pathophysiological changes of the body under the complicated conditions.The study provided a scientific basis for the the essence of traditional chinese medicine syndrome theory and clinical diagnosis of the syndrome of ascendant hyperactivity of liver yang.

      摘要:Objective: To study the absorption characteristics of notoginsenoside R1 influenced by density,energy and p-glycoprotein with rat non-everted gut sac system. Method: Different concentration of notoginsenoside R1 was immersed into rat non-everted gut sac,and the absorption difference was compared.2-nitropenol,a metabolic inhibitor,was used to study the influence of energy on the absorption of notoginsenoside R1.Verapamil,a p-glycoprotein inhibitor,was used to study the influence of p-glycoprotein on the absorption of notoginsenoside R1. Result: The notoginsenoside R1 concentration in the sac was increasing according to the time.It was obvious that 2-nitropenol affected the absorption of notoginsenoside R1.Verapamil had no effect on the absorption of notoginsenoside R1 in sac system. Conclusion: The absorption mechanism of notoginsenoside R1 might be passive diffusion.Notoginsenoside R1 might not be the substrate of p-glycoprotein.

      摘要:Objective: To investigate the effect and mechanism of anti-hypoxia components from Saussurea involucrata. Method: The BALB/c mouse models of normobaric hypoxia,decompression hypoxia and chemistry poisoning hypoxia were developed to evaluate the survival time of mice in hypoxia,and the biochemical indicators,including superoxide dismutase (SOD),malondialdehyde (MDA),lactic acid (LD) and lactate dehydrogenase correlated with anti-hypoxia were measured. Result: Compared to the model group,the ethanol extract of S.involucrata at the dosage of 500, 250 mg ·kg-1 dose-dependently enhanced the survival time of mice in different hypoxia models.At the same time,they could increase SOD (126.31±19.18), (120.45±13.78) U ·mL-1 and lactate dehydrogenase LDH activities (11 334±212.3), (10 882±173.5) U ·L-1,and decrease the content of MDA (3.21±0.52), (3.52±0.33) μmol ·L-1, and LD (2.37±0.44), (2.92±0.36) nmol ·L-1 in blood plasma of mice under a simulated decompression and hypoxia conditions. Conclusion: These results demonstrated that the ethanol extract of S.involucrata possessed notable anti-hypoxic activity.The mechanism may be related to decrease the level of LD,promote the process of anaerobic glycolysis and improve the metabolism of oxygen radicals to the body.

      摘要:Objective: To establish a stable pentobarbital sodium-induced model of acute cardiac insufficiency in the rats and compare the effects of different cardiotonics. Method: Male Wistar rats weighing 220~260 g were randomized into three groups,namely,modeling group,dobutamine group and deslanoside group.The acute cardiac insufficiency model was established in the rats by the intravenous administration of pentobarbital sodium.In dobutamine deslanoside and Astragalus mongholicus groups the rats with cardiac insufficiency were treated with intravenous injection of with dobutamine,deslanoside and radix astragali separately.The left ventricular pressure,heart rate and dp ·dtmax-1 were measured using physiological recording system. Result: Pentobarbital sodium injection established a stable model of acute cardiac insufficiency.The cardiac function in the rats with the cardiac insufficiency was significantly improved by the injection of dobutamine and radix astragali.However,no significant alteration was observed in the rats treated with deslanoside injection. Conclusion: A stable model of acute cardiac insufficiency could be established in the rats by administration of pentobarbital sodium.The model is applicable to dobutamine or radix astragali related studies,but not suitable to the studies involving deslanoside.

      摘要:Objective: To compare the haemostatic mechanism of carbonized and raw Scutellariae Radix, screen effective haemostatic component from the carbonized herb by fevered and bleeding rat models. Method: SD rats were randomly divided into blank control group, model compared group, Yunnan white drug group, raw Scutellariae Radix group, carbonized Scutellariae Radix group and chloroform Scutellariae Radix group.Experimental material was used to ig dose of 20 mL ·g-1 once a day, for 7 days. Prothrombin time (PT), thrombin time (TT), activated partial thromboplastin time (APTT), fibrinogen content (FIB) and hemorheological parameters were examined to study the effects of experimental material on SD rats. Result: The model group had significantly difference with blank control, prolonged PT, TT, APTT, HBV, MBV and LBV to (17.26±1.73) s, (23.15±1.83) s, (22.43±1.93) s, (5.72±0.55) mPa ·s, (7.44±0.78) mPa ·s and (6.59±1.10) mPa ·s. Carbonized Scutellariae Radix and chloroform Scutellariae Radix could significantly shorten the PT, TT and APTT of the fevered and bleeding model rats.PT, TT and APTT of carbonized Scutellariae Radix group and chloroform Scutellariae Radix group was respectively that (14.53±1.45) s, (13.24±1.46) s;(20.91±1.15) s, (18.70±1.44) s and (18.07±0.75) s, (15.59±1.74) s.Compared with the model compared group, P<0.05 or P<0.01.Otherwise, there was no effect on FIB. Conclusion: The chloroform extract is the effective component for hemostasis, and plays roles by ways of endogenous thrombin, extrinsic thrombin and the both, but has no relation with fibrin.The improvement of hemorheological parameters is a basis for hemostasis.

      摘要:Objective: Preparing rabbit serum containing Clastrus orbiculatus extracts (COE),to investigate preliminarily the effects on proliferation and vascular endothelial growth factor-c (VEGF-c) expression in hepatoma cells of mice (Hepa1-6). Method: The New Zealand white rabbits received COE with dose of 20 mg ·kg-1 ·d-1 by gavage.The medicated rabbit serum was obtained using serum phamacological approach.Hepa1-6 cells were cultured in media with different dosages of COE-containing serum (the low,medium,high dose groups containing 10%,20%,30% medicated serum respectively).At the same time,the negative control group and 5-fluorouracil (5-Fu) positive control group were set up.The proliferation inhibition effect of medieated serum containing COE on Hepa1-6 cells was detected by microculture tetrazolium (MTT) assay and the expression level of VEGF-c of Hepa1-6 cells was tested by enzyme linked immunosorbent (ELISA). Result: The typical morphological changes of apoptosis were observed after incubation with COE-containing serum of each dose.The medium and high dose COE-containing serum could significantly inhibit the proliferation of Hepa1-6 cells time-dependently.The VEGF-c expression level of Hepa1-6 cells treating with medium and high dose COE-containing serum for 24 hours were significantly decreased compared with that of the negative control group (P<0.05). Conclusion: COE-containing serum can significantly inhibit proliferation and VEGF-c expression of Hepa1-6 cells in vitro.

      摘要:Objective: To observe the effect for Xuanfu Daizhe Decoction (XFDZD) on motilin (MTL),gastrin (GAS) and vasoactive intestinal peptide (VIP) content in blood and tissue in rats with low gastric motility. Method: Seventy Wistar rats were divided into normal group (10 rats) and model group (60 rats) at random.Each rat of model group was irritated stomach with liquorice decoction (10 g ·kg-1).After stomach irritation,model group were divided into model group of 3 days,model group of 7 days,low,medium,high dosage groups of XFDZD,and the group of domperidone.The stomach with low gastric motility was irritated with domperidone,and the treatment groups were treated with XFDZD at different doses.Then the content of MTL,GAS,VIP in blood and gastric antrum was tested with radioimmunopercipitation. Result: Liquorice decoction could make rat model of low gastric motility.XFDZD could make MTL,GAS content increase in rat’s blood and organization,at the same time make VIP content reduce,campared with model groups,there were all significance difference (P<0.05 or P<0.01). Conclusion: XFDZD could treat disease of gastrointestinal dismotility by regulating brain-gut peptides content in rat's blood or gastric antrum.

      摘要:Objective: To evaluate the anti-inflammatory and analgesic effects of ligustilide. Method: The pharmacodynamic effects were observed in 2 analgesic tests and 4 inflammatory models in experimental animals,including:hot plate test in mice, writhing reaction induced by acetic acid in mice, increased permeability of abdominal cavity induced by acetic acid in mice, auricular edema induced by dimethy benzene in mice, paw swelling caused by carrageenan in rats, cotton-pellet granuloma in rats. Result: Ligustilide (100,60,20 mg ·kg-1) could delay the latency of licking of the hind paw in hot plate test.And it could reduce the times of writhe response actions,and the inhibition rates were 61.21%,48.12%,43.61% accordingly.Ligustilide could distinctively inhibit the increase in permeability of abdominal cavity induced by acetic acid in mice,and the inhibition rates were 64.84%,57.85%,49.67% respectively.Ligustilide could significantly inhibit the auricular edema induced by dimethy benzene,and the inhibition rates were 52.06%,41.51%,34.21%.Ligustilide could significantly inhibit the paw swelling caused by carrageenan in rats,and the inhibition rates were more than 42% from 1 to 3 hour.Ligustilide could significantly inhibit the cotton-pellet granuloma in rats,and the inhibition rates were 52.64%,50.32%,45.33%. Conclusion: Ligustilide has an analgestic effect and can significant inhibit acute and chronic inflammation in animals.

      摘要:Objective: To study hypoglcemic effect of total polyphenols from seeds of Toona sinensis (STP)on diabetic mice model. Method: Diabetic model was induced by once intraperitoneal injection of STZ.Diabetic mice were randomly divided into model group,Metformin group and high-,middle-and low-doses of STP groups (100,80,60 mg ·kg-1).All groups were fed for 14 consecutive days,and then the blood glucose were detected by glucose oxidizes on the fifteenth day.The organ index was also detected. Result: The blood glucose level of the diabetic mice was significantly decreased.STP could increase liver index significantly(P<0.05).STP could decrease MDA(P<0.05) and increase SOD (P<0.05) in liver homogenate of the diabetic mice significantly. Conclusion: STP can significantly decrease the blood glucose level in mice with diabetes induced by STZ.

      摘要:Objective: To investigate the effect of Jingluoshu Capsule for activating blood circulation to remove blood stasis (ABCRBS),and its analgestic and anti-inflammatory action in diabetic rats. Method: High,medium and low dose groups of Jingluoshu Capsule,1.4,0.7,0.35 g ·kg-1in rats and 2.8,1.4,0.7 g ·kg-1in mice accordingly,were established.,Positive medicine (Tangmaikang Grannules) was ig given once daily at the dose of 2.5 g ·kg-1.The diabetic rat model was established by intraperitoneal injection of streptozotocin (STZ) in Wistar rats.The animals were randomly divided into normal control group,model control group,positive medicine control group,positive control group of Chinese medicine,high,medium and low dose groups of.Jingluoshu Capsule.of the efficacy of the drugs was evaluated with hemorrheological parameters,anti-inflammation and analgestic tests. Result: Jingluoshu Capsule could decrease the whole blood viscosity at high shear rate,show inhibitory action on the increase in capillary permeability induced by histamine.The capsule significantly inhibited the formation of granuloma induced with cotton ball.After 30 and 60 minutes of administration,it could prolong the time of first licking feet in the hot-plate test in mice and increase the pain threshold.The capsule could also significantly decrease the number of twisting body induced by acetic acid in mice,showing a significant analgesic effect. Conclusion: Jingluoshu Capsule has significant effect of activating blood circulation to remove blood stasis,and anti-inflammatory,analgestic actions in diabetic rats.

      摘要:Objective: To observe the effects of Danqing Soft Capsule (DSC) on rat with non-alcoholic fatty liver (NAFLD). Method: The SD rats were randomly divided into control group,model group,DSC high dose group,medium dose group,low dose group,and polyene phosphatidyl choline (PPC) group.Non-alcoholic fatty liver disease model in SD rats was established with high fat diet.DSC high dose group,medium dose group and low dose group were intervened with different concentrations of DSC,which are respectively equivalent to petroleum ether extract of salvia 0.456,0.228,0.114 g ·kg-1.PPC group was intervened with PPC of 0.159 g ·kg-1ig.Control group and model group were administered the same volume (1.2 mL ·kg-1,ig) of distilled water.After treatment for 4 weeks,rat liver pathological changes,TG,TC level,and blood lipids levels were observed. Result: The levels of TC,TG,LDL-C in serum and the levels of TG and TC in liver of control group,traditional Chinese medicine (TCM) groups and western medicine (WM) group were significantly lower than those of model group (P<0.01,P<0.05).Compared with those in model group,the degrees of fatty degeneration of liver cells in TCM groups and WM group were significantly improved (P<0.01). Conclusion: DSC can reduce the level of TG,TC,LDL in serum as well as the level of TG and TC in liver.It can improve the steatosis of liver in rats with NAFLD.

      摘要:Objective: To investigate the protective effect of Taraxaci Herba on alcohol-induced liver injury. Method: The paper studied alcohol-induced liver injury model in mice. The model mice were randomly divided into five groups, including Taraxaci Herba low dose group,Taraxaci Herba medium dose group,Taraxaci Herba high dose group,bifendate group and model group. Every group was respectively treated for 6 weeks with Taraxaci Herba, bifendate or normal saline accordingly. The activities of aspartate transaminase (AST),alanine transaminase (ALT) in serum,superoxide dismutase(SOD),glutathione perioxidase (GSH-Px);and the content of malondialdehyde (MDA),nitrogen monoxide(NO), TNF-α in liver were investigated. Result: Taraxaci Herba could remarkably resist the increase of ALT,AST in serum( P <0.05-P<0.01),and decrease MDA,NO,TNF-α level( P <0.05-P<0.01),and improve SOD activity and GSH-Px level ( P <0.05-P<0.01),and the differences were statically significant. Conclusion: Taraxact Herba could change inflammatory mediators and cytokines significantly, thereby reducing or preventing related injury for the anti-inflammatory purpose.

      摘要:Objective: To observe the effect of Tangzhiping granules on NO,NOS and morphological changes of kidney tissue in diabetes mellitus (DM) Wistar rats with nenopathy. Method: The diabetic nenopathy model of rats with was successfully made by high adipose diet and injection of streptozocin (STZ) in abdominal cavity.Then the rats were intervened withTangmaikang granules and Tangzhiping granules and the contents of NO and NOS were detected.The morphological changes in kidney tissue wereobserved. Result: After treatment,and the contents of NO and NOS in kidney in the treatment groups were increased significantly compared with that of the model (P<0.05),The contents of NO and NOS in the Tangzhiping high dose group were increased significantly compared with those in the positive drug group and in the Tangzhiping low dose group (P<0.05).Morphological slides showed that the Tangzhiping high and low doses and the positive control drug had protective effect on injury caused by diabetic nenopathy.The protection come from the Tangzhiping high dose was the most. Conclusion: Tangzhiping granules can increase NO and NOS in rats with diabetic nenopathy and promote the kidney tissue repairs at the same time.

      摘要:Objective: To study the protection of Ruyi Dingchuan pill on emphysema rat,and provides theoretical basis for its clinical application. Method: Sixty SD rats were randomly divided into normal group,model group,Ruyi Dingchuan pill of high,medium and low dose groups,Guilong Kechuanning capsule control group.Rat model of emphysema was established by intratracheal instillation of lipopolysaccharide (LPS) twice and exposed to cigarette smoke daily.After 3 weeks,the rats were administrated by gastric lavage of corresponding drugs for 14 days.Artery blood gas analysis and pathological changes in the lungs were observed.The expression of IL-10 and TNF-α was examined by ELISA. Result: Pathological changes with obvious emphysema and inflammation cell infiltrates were found after modeling.Average alveolar area and the levels of TNF-α in the model group were higher than those in the normal group(P<0.05).The arterial oxygen content and the levels of IL-10 in the model group were lower than those in the normal group(P<0.05).Ruyi Dingchuan pill could obviously lessen the infiltrations of inflammatory cells and formation of emphysema,increase arterial oxygen content,and significantly reduce the expression of TNF-α (119.80±5.53) ng ·L-1 and increase the expression of IL-10 (33.59±7.92) ng ·L-1 in the blood serum(P<0.05).Its effect was statistically insignificant compared with the positive drug. Conclusion: Ruyi Dingchuan pill had apparent protective effect for emphysema rats.

      摘要:Objective: To study the experimental anti-arrhythmia and anti-hypertensive effects of extract from Menispermum dahuricum (MD). Method: The experimental arrhythmia model in rat was caused by aconitine and BaCl2.The blood pressure was measured.The occurrence of VA,VT,VF caused by aconitine in rats was recorded,and arrhythmias caused by BaCl2 in rats was recorded.Blood pressure was monitored in rats before and after the treatment of extract from MD. Result: MD (40 mg ·kg-1,iv) could significantly increase the amount of aconitine for inducing VP and VT in rats;MD (40,20 mg ·kg-1,iv) could increase amounts of aconitine for inducing VF in rats.MD (40,20 mg ·kg-1,iv) could delay the occurrence of arrhythmia induced by BaCl2 and shorten the duration of arrhythmia in rats.MD (40 mg ·kg-1) showed obvious anti-hypertensive effect,the anti-hypertensive duration was lasted significantly. Conclusion: MD has significant protective effect to the experimental arrhythmia induced by drugs.MD has lasting anti-hypertensive effect.The anti-arrhythmic mechanism is likely related to inhibition of transmembrane movement of Ca2+ and Na+.

      摘要:Objective: To study the pharmacological effects of Coptidis Rhizoma and Caryophylli Flos on anti-inflammatory,analgesic and for treatment of oral ulcers with external use. Method: In the anti-inflammatory experiments 60 rats were randomly divided into 5 groups.Their right feet plantar were intradermally injected with 1% carrageenan and given Coptidis Rhizoma extract externally,the paw circumference was measured with soft rule.In analgesia experiments,60 mice were randomly divided into 5 groups.Their tails were given Caryophylli Flos extract externally,tail tenderness threshold was measured with tenderness instrument.In the oral ulcer experiment,96 rats were randomly divided into 8 groups.After the model of oral ulcers,the rats were given Coptidis Rhizoma or Caryophylli Flos extract externally,the ulcer healing rate was then evaluated with computer calculating. Result: ① The anti-inflammatory experiments showed that berberine inhibited topical carrageenan induced rat paw edema,there was significant difference between the dose group and control group(P<0.05).② Analgesic experiments showed that clove \ could significantly elevate the tenderness mechanical pain threshold.there was significant difference between the large,medium dose groups and control group(P<0.05).③ Experiments showed that treatment of oral ulcers induced by phenol with berberine and Caryophylli Flos could gain significant effect,there was significant difference between the large,medium dose groups and control group(P<0.05);also there was significant difference between the medium dose berberine group and the medium dose Caryophylli Flos group (P<0.05),the effect of Caryophylli Flos was better. Conclusion: External use of Coptidis Rhizoma extract has significant anti-inflammatory effects.External use of Caryophylli Flos extract shows effect of analgesia.External use of Coptidis Rhizoma and Caryophylli Flos together can treat oral ulcers effectively.

      摘要:Objective: By observing the effects of Toutongning capsule on the expression of c-fos and c-jun in periaqueductalgrey (PAG) and thalamencephalon in rats with migraine to discuss the therapeutic mechanism of Toutongning capsule. Method: The SD rats were evenly divided into 6 groups:normal group,model group,Flunarizine(1.04 mg ·kg-1)intervention group,high-dose,mid-dose and low-dose Toutongning capsule groups(760,380,190 mg ·kg-1).Migraine model in rats was established by NTG subcutaneous injection.After NTG subcutaneous injection the specimen at 4 hours and 6 hours was obtained.The expression of c-fos and c-jun were detected by immunohistochemistry. Result: The expression of c-fos and c-jun in PAG and thalamencephalon were increased in the model group compared with that in the normal group (all P<0.01).At 4 hours after NTG subcutaneous injection,there was much more expression of c-fos and c-jun compared with those at 6 hours after NTG subcutaneous injection.The expressions of c-fos and c-jun in PAG and thalamencephalon were decreased in the high-dose Toutongning capsule intervention group,mid-dose Toutongning capsule intervention group and Flunarizine intervention group compared with those in low-dose Toutongning capsule group and in the model group (all P<0.01).There was no significant difference between model group and low dose intervention group. Conclusion: By reducing the expression of c-fos and c-jun and restrainning the transmission of pain information in PAG and thalamencephalon,Toutongning capsule could treat migraine.And the effect of high-dose Toutongning capsule and mid-dose Toutongning capsule were much better.

      摘要:Objective: To study the effect of Crotonis Fructus alkaloid (CA) on apoptosis and the expression of Bcl-2 and Bax in human hepatoma SMMC-7721 cells. Method: After treatment with CA for 24-48 h,MTT assay was used to determine the cell growth inhibitory rate in vitro,flow cytometry was used to detect the apoptosis of SMMC-7721 cells before and after CA treatment.Western blot was used to detect the expression of Bcl-2 and Bax. Result: CA inhibited the growth of SMMC-7721 cells in a dose-and time-dependent manner.After treatment with 50-200 mg ·L-1 CA for 24 h,the number of apoptosis cells was increased dose-dependently from (0.26±0.14)% in the control group to (5.85±0.47)%,(17.70±1.23)%,(33.25±2.08)% and (49.52±1.85)% in CA groups,respectively.By Western blot analysis,we found the level of Bax was up-regulated,whereas,the level of Bcl-2 down-regulated. Conclusion: CA can efficiently inhibit the proliferation and induce apoptosis in SMMC-7721 cells.Its mechanism is likely related to the down-regulation of Bcl-2 and up-regulation of Bax.

      摘要:Objective: To study the analgesic and anti-inflammatory effects,as well as acute toxicity of the Wuling Jianzhou pellet. Method: Writhing method and formalin test were used in mice to evaluate the analgesic effect.Xylene induced ear swelling in mice and formation of cotton ball granuloma in rats were adopted to investigate the anti-inflammatory effect.LD50was assessed in mice. Result: At the dose of 3.8,7.6 g ·kg-1,Wuling Jianzhou pellet significantly reduced the number of writhing induced by acetic acid (P<0.05),as well as mouse ear swelling induced by xylene (P<0.01).In formalin test,pain of phase I but not phase Ⅱ,was reduced after administration of Wuling Jianzhou pellet at 3.8,11.4 g ·kg-1 (P<0.01).The formation of cotton ball granuloma was inhibited by Wuling Jianzhou pellet at 7.6 g ·kg-1 (P<0.05).The LD50of Wuling Jianzhou pellet was 148.4 g ·kg-1. Conclusion: Wuling Jianzhou pellet has significant analgesic and anti-inflammatory effects,and exhibits low toxicity.

      摘要:Objective: To study the impacts of culture conditions on the Caco-2 cell model,to optimize the appropriate conditions for a stable,reproducible Caco-2 model. Mehtod: The environment of small intestine was in vitro simulated,and rat tail collagen was coated on three types of Transwell monolayerly and compared.Morphological features were observed by microscope,the integrity,polarity and permeability of monolayer were measured by the resistance,alkaline phosphatase,the amount of sodium fluorescein. Result: After culture of 21 days,the integrity,polarity of Caco-2 monolayer cultured on 24-well Transwell were higher than that on 12-well Transwell,but the permeability were the opposite.The TEER of the monolayer coated with rat tail collagen was increased rapidly in early stages,and was no significant effect at the middle and late stages. Conclusion: Three types of Transwell can be successfully used to culture Caco-2 cell monolayer.24-well Transwell is suitable for small samples with high sensitivity and high-throughput screening of a large number of samples.12-well Transwell has higher volume of sample and permeability,is suitable for routine laboratory operations and equipment.

      摘要:Objective: To investigate the effect of metabolism and bio-transformation of GRg1 by intestinal enzyme and microflora in rats. Method: The stability of GRg1 in the artificial gastric juice and intestinal juice was tested.The metabolism and bio-transformation of GRg1 administrated orally in rats were studied. Result: GRg1 was degraded totally within 2 hours in artificial gastric juice.Meanwhile,GRg1 was stable in artificial intestinal juice.GRg1 was metabolized by intestinal enzyme microflora in rats. Conclusion: GRg1 should be administrated from other routes instead of oral route in order to improve the bioavailability.

      摘要:Objective: To investigate the protective effect of acetylpuerarin on cerebral ischemia-reperfusion injury in rats and its mechanism. Method: Wistar rat was used as a model of focal cerebral ischemia and reperfusion.The positive reacted cells of apoptosis were counted and immunohistochemical method was used to measure the number of PI3-K/ Akt positive cells in rat’s cerebral cortex of sham-operated group,model group,citicoline group and acetylpuerarin groups(10,50,250 mg ·kg-1). Result: Acetylpuerarin could significantly increase the expression of the PI3-K and Akt,and reduce the number of cells of apoptosis(P<0.01).The higher expression and the fewer cells of apoptosis in the medium dose acetylpuerarin group (P<0.05)and the high dose acetylpuerarin group was better than the citicoline group(P<0.05). Conclusion: Acetylpuerarin has obviously protective function for ischemia-reperfusion in rat.

      摘要:Objective: To observe the effect of Jingyuankang capsule on the expression of Bcl-2,Bax mRNA in bone marrow cells of model mice of chemotherapy-induced myelosuppression and make a further study of the mechanism of the medicine. Method: Measuring the expression of Bcl-2,Bax mRNA in bone marrow cells of model mice of myelosuppression with real-time fluorescence quantitative PCR. Result: The Jingyuankang group had the highest expression of Bcl-2 mRNA and lowest expression of Bax mRNA,compared with the model group,the difference was significant,while compared with the control group,there was no significant differences. Conclusion: These results confirmed preliminarily that Jingyuankang capsule could increase the expression of the anti-apoptotic gene Bcl-2 and decrease the expression of proapoptotic gene Bax,through affecting the apoptosis-related pathway to prevent the blood cells from apoptosis caused by chemotherapy,this may be an important mechanism.of Jingyuankang capsule treating the chemotherapy-induced myelosuppression.

      摘要:Objective: To explore the mechanism of the domestic β-aescin treating chronic venous insufficiency through observing its actions on the isolated canine saphenous venous tension,venous pressure,venous return and lymphatic drainage. Method: The isolated canine spiral saphenous uenous tension test was performed to detect the activity of the β-aescin.Furthermore,in the comdition of constant canine femoral artery perfusion kept in the extracorporeal circulation,we measured the changes in the canine femoral artery pressure,femoral artery artery flow and the lymphatic drainage flow after intravenous injection of the drug. Result: When β-aescin was administrated at the dose between 5.0×10-5-5.25×10-6g ·mL-1,it could obviously increase the contractile tension of the venous to norepinephrine in a dose-dependent manner.With canine femoral artery perfusion kept constant,β-aescin,whose doses were 50 mg and 100 mg,could reinforce intently the canine femoral venous tension,accelerate the rise of the venous return and lymphatic drainage. Conclusion: The domestic β-aescin extracted from shorea has some therapeutical effect on chronic venous insufficiency by strengthening the venous tension,increasing the venous pressure and promoting venous return and lymphatic drainage.

      摘要:Objective: To investigate the pathological change of H22 hepatoma induced by radioactive particle125I combine with cisplatin(DDP). Method: 125I,Cisplatin (DDP) and 125I combined with DDP were implanted into the mice H22 hepatoma,individually.After 28 days,the tumor growth was observed,and the pathological change in the cells were investigated by optical microscope and electron microscope. Result: After125I,DDP were implanted into the mice H22 hepatoma,the tumor growth showed fast at first phase and then became slow;125I combined with DDP made the tumor growth slower with cell degeneration and necrosis.Under the optical microscope,cells of125I group and DDP group had a irregular shape,part of cells showed necrosis and surrounding tissue cells had lots of lymphocytes.But the characteristics of125I combined with DDP was cells necrosis.Under the electron microscope,the nuclear membrane was complete,chromatin distributed uniform,nucleolus was concentrated or fragmentated and the apoptotic bodies were form.The configuration of rough endoplasmic reticulum was loosening and degranulated,with increasing endoplasmic reticulum,which had more apoptosis of cancer cells,and had more apoptotic cells aggregated in the125I group and DDP group.But the125I combined with DDP group was with necrosis or incomplete necrotic cells mainly,which also had the apoptosis cancer cells. Conclusion: The interstitial implant of radioactive seed125I induces hepatoma cells apoptosis. 125I combined with DDP induces apoptosis,cell degeneration and necrosis.

      摘要:Objective: To investigate the interventions of taurine and vitamin in atherosclerosis (AS) induced by hyperhomoaysteine(HHcy) in rabbits. Method: The model groups with HHcy were respectively fed with taurine,folic acid,vitamin B6 and vitamin B12-a high L-methionine diet.Compared with the control group(NC),the serum of the model groups were tested respectively in the 4th week and 8th week to determine the content of Hcy,MDA,TC,TG,NO,TXB2 and the activity of SOD and the changes in abdominal artery disease by vascular ultrasound and histologyical observation. Result: The contents of Hcy,MDA,ET and TXB2 in the serum of the rabbits having received methionine increased a lot(P<0.01,P<0.05) in the 4 week and 8 week than that of the control group,but the content of NO and the activity of SOD were much lower(P<0.01),compared with the NC group in the same period.Hcy in the rabbits having received taurine were much higher,but Hcy in the rabbits having received vitamin displayed on change in the 8 week.And the contents of MDA,ET and TXB2 in both model groups were much lower (P<0.01) than the group with methionine,but the contents of NO and the activity of SOD were much higher(P<0.01).In the group with methionine Hcy could obviously induce abdominal arterial lesions with intimal thickening,smooth muscle cell hyperplasia,elastic fiber fracture,sort disorder and fatty plaque.Compared with the NC group,the rabbits received taurine and those received vitamin displayed no arterial lesions,no fatty plaque with neat structure. Conclusion: Taurine can resist the destrucion induced by Hcy.Vitamin can promote Hcy metabolism,inhibit vascular damage induced by Hcy and delay or prevent AS.

      摘要:Objective: To study the anti-tumor mechanism of Xiaoai Jiedu prescription in vivo. Method :The mice bearing tumors(estabilished by subcutaneous inoculation of H22 cells)were fed with Xiaoai Jiedu prescription.Morphology of the tissue of tumor under light microscope and tumor cells under electron microscope were observed. The matrix metall-oproteinase(MMP-2) were assayed. Result: The medium dose group of Xiaoai Jiedu prescription has the largest amount of apoptosis. There were many pinocytosises except the general morphological change in each Xiaoai Jiedu prescription group. The activity of MMP-2 had significantly decreased in the low-dose group of Xiaoai Jiedu prescription(P<0.05). Conclusion: Xiaoai Jiedu prescription has the anti-tumor effect in vivo possibly through the mechanism of inducing tumor cell apoptosis and inhibitting the activity of MMP-2.

      摘要:Objective: To study the mechanisms by which APS ameliorates insulin resistance,we examined whether treatment with APS improves insulin sensitivity in insulin-resistant rats and whether this is associated with an increase of adiponectin and reduction of tumor necrosis alpha(TNF-α) and free fatty acides(FFA) in serum. Method: Seventy male SD rats were randomly divided into two groups:normal control group(NC group,n=12) and high fat-diet-induced model group(HFM group,n=60).Rats in NC group were fed with ordinary diet while those in HFM group were fed with high fat diet for six weeks.Then the rats in HFM group were randomly divided into five groups:high fat-diet-induced control group(HFC group),pioglitazone group(Pio group),low APS group(Laps group),medium APS group(Maps group) and high APS group(Haps group),with 12 rats in each.Normal saline,20 mg ·kg-1 ·d-1 pioglitazone and APS of different dosages (200,400,800 mg ·kg-1 ·d-1)were lavaged respectively for eight weeks.Changes of fasting blood glucose(FBG),fasting insulin(FINS),adiponectin,TNF-α and FFA in one half rats of each group were routinely measured,meanwhile glucose infusion rate(GIR) of another half was evaluated by hyperinsulinaemic-euglycaemic clamp technique. Result: Compared with those in NC group,the FFA,FINS and TNF-α of the rats in HFC group were significantly higher(P<0.05),while adiponectin and GIR significantly lower(P<0.05).Compared with those in HFC group,the above mentioned indexes in the low and medium APS groups were improved significantly(P<0.05). Conclusion: These results indicate that moderate APS can regulate part of the insulin signaling in insulin-resistant serum and that APS could be a potential insulin sensitizer for the treatment of insulin resistance.

      摘要:Objective: The aim of this study is to develop the effective therapy on Dingxin capsule for D-dimer(D-D)of atrial fibrillation. To reveal the mechanism of Chinese medicine with the effects of nourishing Qi and Yin and activating blood in the prevention and treatment of Atrial Fibrillation(AF). Method :Forty rats were randomized to 4 groups:normal group,model group,Dingxin capsule curatio group,verpamil intervention group. The level of D-D of AF rats were measured. Result: With the comparison of treating group and model group,the level of D-D in treating group lowed obviously(P<0.05). Both of them had statistic significance.Both Dingxin capsule group and Verpamil intervention group could low the level of D-D,but Dingxin capsule group is better. Conclusion: Dingxin capsule could correct the level of D-D in AF rats. D-D played important role in the pathogenesis of AF.

      摘要:Objective: To investigate the clinical effects of Diane-35 with metformin on treating women with polycystic ovary syndrome(PCOS). Method: Thirty-eight patients took orally Diane-35 and metformin for 3 consecutive courses of treatment.Clinical symptoms,luteinizing hormone(LH),follicle stimulating hormone(FSH),T,Leptin,HOMA-IR,SHBG,BMI and the ovarian volume were kept under observation before and after taking Diane-35 and metformin.Reproductive hormones,serum glucose and insulin level were compared to that before therapy. Result: The content of LH/FSH,androgen,HOMA-IR were less than that before the treatmennt.The content of SHBG was more than that before taking medicine,and there were highly significant difference (P<0.01).The content of BMI was less than that before taking medicine,the size of ovaries was reduced and there were significant difference (P<0.05). Conclusion: Diane-35 and metformin are effective to reduce insulin resistance and androgen and to recover ovulation in women with PCOS.And the treatment was shown to be safe and effective.

      摘要:Objective: To explore the impact of Xuesaitong United Batroxobin on serum hsCRP and plasma vWF levels in patients with acute cerebral infarction. Method: Eighty-two patients with acute cerebral infarction were divided into control group (n=41) and treatment group (n=41).Control group patients were treated with Xuesaitong iv injection 20 mL with 250 mL saline once daity for (d 1-d 4).Based on the application of the control group,the treatment group patients were additionally treated with Batroxobin 20 mL in 250 mL normal saline intravenously for d 1,d 3,d 5. The neurological deficit score and plasma vWF levels,serum hsCRP level were observed in this two groups,before treatment and after treatment 7,14 d. Result: The neurological deficit scores were significantly lower in the treatment group and control group,after 7,14 d (P<0.01).Compared with the control group,treatment group decreased more significantly (P<0.01).Compared with that before treatment,plasma vWF and serum hsCRP levels were significantly increased after 7 d (P<0.01),compared with the control group,treatment group increased more significantly (P<0.01);14 d plasma vWF levels and serum hsCRP levels were significantly lower (P<0.01),the treatment group decreased more significantly (P<0.01). Conclusion: Xuesaitong United Batroxobin appears to be better than the treatment of acute cerebral infarction by Xuesaitong alone.

      摘要:Objective: To observe the effect of Fructus Bruceae oil emulsion (FBE) combined with chemotherapy on immune function in patients with advanced non-small-cell lung cancer (NSCLC). Method: One hundred and fifty-six patients with advanced NSCLC were randomly assigned to two groups,the 74 patients in the control group were only treated with chemotherapy of GP regimen,84 in the treatment group with the chemotherapy of the same regimen and combined with FBE.The cellular immune(CD3+,CD4+,CD8+,CD16+ CD56+)and humoral immunity(IgG,IgA,IgM,C3,C4)were tested by immunocytochemistry method and biochemistry method respectively.The treatment effects on immune were compared between before and after chemotherapy. Result: The response rates were 40.3% in the treatment group and 36.5% in the control group, there was no statistical difference between them.Compare before chemotherapy,the cellular immune function in the control group was decreased,which was increased in the treatment group.The cellular immune function in the treatment group was better than that in the control group after chemotherapy(P<0.05 or P<0.01).No visible change in humoral immune was found in each group. Conclusion: FBE combined with chemotherapy of GP regimen can improve the cellular immune function in patients with advanced NSCLC.

      摘要:Objective: To reveal prognostic value of combined detection of NT-proBNP,LA and CRP in the plasma with acute myocardial infarction(AMI). Method: Analysis of cardiac adverse events with 92 cases of AMI in the 2008-2010 year,comparison the NT-proBNP, LA and CRP between the groups of occurred adverse cardiac events and did not occur adverse cardiac events. Result: The level of NT-proBNP,LA,and CRP in the adverse cardiac events occurred group were all higher than those without adverse cardiac events group,the differences were statistically significant (P<0.05). Conclusion: Combined detection of NT-proBNP,LA and CRP in patients with AMI maybe a good early-warning function of occurred adverse cardiac events.

      摘要:Paridis Rhizoma is the dry rhizome of Paris polyphylla Smith var. yunnanensis(Franch.) Hand.-Mazz. or Paris polyphylla Smith var. chinensis(Franch.) Hara. This paper summarizes the chemical composition, pharmacologic actions and clinical use of the herb in recent years. It mainly contains steroidal saponins, sterol, mineral element and free amino acids, and steroidal saponin is its main ingredient. It has effects of antitumor, antiphlogistic, hemostatic, anti-oxidation and uterine contraction. Paridis Rhizoma is being used in analgesia and hemostasis, anticancer by oral administration, and being used in anti-infection and anti-inflammatory and analgesic by external application. However, no innovative drugs with good activity have been developed from Paridis Rhizoma yet. Therefore, we should speed up the researches on Paridis Rhizoma, and make a further research progress for its innovation.

      摘要:Recurrent oral ulcer (ROU) represents a very common but poorly understood mucosal disorder. It can occur in men and women of all ages, races and geographic regions. It is estimated that at least 1 in 5 individuals has at least once been afflicted with recurrent oral ulcer. Attacks may be precipitated by local trauma, stress, food intake, drugs, hormonal changes and vitamin and trace element deficiencies. However, to date, no principal cause has been discovered. There is no curative therapy to prevent the recurrence of ulcers, and all available therapies can only reduce the frequency or severity of the lesions. In order to provide a valuable reference for clinical treatment, in this article we collected and analyzed the reports about recurrent oral ulceration recently, and make a review of treatments for recurrent oral ulceration.

      摘要:The dysfunction of nervous system caused by cerebral infarction is difficult for medical study.Recently,the study mainly focused on ischemic areas,but there is little emphasis on secondary damage.Nogo-A conducts the inhibitory signals through the neural pathways of Nogo/NgR→Rho/Rock to hurt axons and participate in the secondary damage in cerebral infarction.It is the important reasons for the dysfunction of nervous system caused by acute cerebral infarction.The author will discuss the relationship between Nogo-A and secondary damage in cerebral infarction.

      摘要:This paper presented a review of chemical and pharmacological of Ardisia crenata in nearly ten years on the basis of consulting plenty of domestic and foreign literature. The research indicated that A.crenata contains triterpenoid saponins, coumarins, volatile oil, phenols, quinone, cardiac glycoside, organic acid, saccharide etc. A. crenata was reported to possess relieving cough and asthma, anti-inflammatory, antibacterial, antineoplastic, antifertility, anticoagulant, lower blood pressure, triggering uterine contractions, cAMP phosphodiesterase inhibitory effect. The purpose of this study was to elaborate the research of chemical composition and biological activity of A. crenata, The in-depth research on A. crenata appears to have highly worth development and apply for the therapy of diseases.

      摘要:In this paper, the literature are consulted and the different effects of single herbs in vitro antibacterial activity in China are reviewed at home and abroad over the past five years,summarized possible mechanism and antibacterial composition of inhibiting pathogenic bacteria and pathogens of it,including heat-clearing drugs, superficies-resolving drugs, purgation drugs, desiccating drugs,carminative drugs,astringent drugs,tonic drugs,cough and asthma drugs,eliminating dampness and diuresis drugs,promoting blood circulation and expelling blood stasi drugs,and other drugs.In order to provide a reference for the use of our abundant resources of Chinese medicine and the development of antibacterial agent with Chinese medicine,which have fewer side effects and better clinical efficacy. It provides a basis and new ideas for the antibacterial medicine.

      摘要:Based on the previous edition, standard of pharmaceutical excipients in 《Chinese Pharmacopoeia》 2010 Edition has a larger remodel. Although the collection contains a greater variety and projects to enrich and perfect, there are still some deficiencies. In this paper, the actual situation of the domestic pharmaceutical industry, foreign Pharmacopoeia contained situation, and the standard of pharmaceutical excipients was discussed. This article mainly studies the varieties, series, monographs project, expressing aspects of 《Chinese pharmacopoeia》 2010 edition pharmaceutical excipients standard, in order to provide the reference for the further improvement of pharmaceutical excipients standards, better meet the domestic pharmaceutical preparations and medicinal materials industry needs.
