      摘要:Objective:To analyze the composing experience of professor YAN Zheng-hua's prescriptions for treating stomachache by using traditional Chinese medicine(TCM)inheritance support system. Method: The prescriptions for stomachache composed by YAN Zheng-hua were collected,then the data were input in the TCM inheritance support system and the composing principles were analyzed by using data-mining methods, such as revised mutual information,complex system entropy clustering and unsupervised hierarchical clustering. Result: Based on the analysis of 150 prescriptions, the frequency of each herb and the association rules among the herbs were used, 49 core combinations and 29 new prescriptions are mined from the database. Conclusion: Professor YAN Zheng-hua is good at soothing the liver and regulating qi to relieve stomachache. TCM inheritance support system can be used to analyze clinical experiences of senior TCM doctors.

      摘要:Objective:To study dissolution regular pattern of ingredients from traditional Chinese medicine compound by solvent with different solubility parameter, and provide new ideas and theoretical basis for chromatographic pharmacodynamics sampling. Method: With different solubility parameters of solvent to dissolve Buyang Huanwu decoction extract,fingerprint chromatogram of ingredients from obtained lysate were determined by HPLC,and combining with total statistical moment theory,difference of solvent with different solubility parameter on component solution from total prescription was evaluated by total statistical moment similarity method. Result: RSD of AUCT, MCRTT, VCRTT from samples were 58.2%,12.8%,24.8%,respectively.It showed that chemical composition and total amount had significant difference,which was dissolved from solvent with different solubility parameter,its range of total statistical moment standard similarity 0.370-0.998.Sample similarity of S4, S5, S6 (solubility parameter of 18.33-20.60 J1/2·cm-3/2) compared to other samples, S7 and S8 (solubility parameter 21.73-22.87 J1/2·cm-3/2) compared to other samples except adjacent samples, these were less than 0.9,it indicated that constituent ratio of solvent on total prescription dissolution composition was different. Conclusion: Solubility parameter sampling method could make composition ratio of traditional Chinese medicine compound with significant difference,dissolution of solvent with different solubility parameter on total prescription had obvious law, it could provide an entirely new and referenced ideas for in vitro sampling with chromatographic pharmacodynamics, and could lay foundation for establishing a complete theoretical guidance of chromatographic pharmacodynamics sampling.

      摘要:Objective:To investigate influence of chitosan flocculation sedimentation method on flavonoids and alkaloids from Gegen Qinlian decoction. Method: With puerarin, baicalin, berberine hydrochloride and extract rate as indexes,orthogonal design method was used to study effect of three factors including liquid concentrated degree,pH and the amount of chitosan on refining flavonoids and alkaloids from Gegen Qinlian decoction by chitosan flocculation sedimentation method,and compared with ethanol precipitation,centrifugal sedimentation and filtration method. Result: Optimum purification process of chitosan flocculation sedimentation method was:the mass concentration of liquid 0.78 g·mL-1,the addition amount of chitosan 10 %,pH 6,by comparing with different precipitation method,sedimentation effect of chitosan was the best. Conclusion: Chitosan could effectively retain active ingredients in liquid and reduce extract rate,but this method could change composition proportion of ingredients in liquid, so it should be cautious to choose this method.

      摘要:Objective:To study on wall-broken technology of Ganoderma lucidum and increase broken rate of G. lucidum powder. Method: According to index of moisture content and breaking hardness of G. lucidum powder, effect of embrittlement temperature, vacuum drying temperature and vacuum drying time on embrittlement conditions with different temperature and pressure were studied by L9(34) orthogonal experiment.And embrittlement ratio of prepared embrittlement sample and superfine grinding sample were compared. Result: It was found that optimum embrittlement conditions were:embrittlement temperature, vacuum drying temperature and vacuum drying time were 100 ℃,80 ℃ and 1 h,respectively.Under these conditions,the moisture content and breaking hardness were 3.0% and 3.7 kg.After embrittled and dried,G. lucidum powder was broken 50 min,broken rate of G. lucidum was up to 99.5%. Conclusion: Bbroken rate of G. lucidum could enhance by combining difference temperature and pressure embrittlement and superfine grinding technology.

      摘要:Objective:To optimize alcohol extraction technology of total flavonoids from Zhichen Weili tablet. Method: With content and pharmacodynamic effect of total flavonoid as indexes,extraction solvent was determined by single factor test;With the content of naringin,hesperidin and neohesperidin as comprehensive scoring index,the concentration and amount of ethanol,extraction time and extraction times were chosen as factors,alcohol extraction technology of total flavonoids from Zhichen Weili tablet was optimized by orthogonal test. Result: Optimized alcohol extraction conditions were:reflux extracted 3 times with 7,6,6 times the amount of 70% ethanol,extracted 2 h per time. Conclusion: This extraction process was reasonable and simple,it could provide a reference for production process of total flavonoids from Zhichen Weili tablet.

      摘要:Objective:To optimize mechanical boiling technology of Sini decoction. Method: With the contents of benzoylmesaconine, liquiritin, glycyrrhizic acid and 6-gingerol as indexes,the content of indicator ingredients were determined by HPLC,mechanical boiling technology of Sini decoction was optimized by single factor test and orthogonal test with the amount of water,soaking time,boiling time and boiling times as factors. Result: Optimum mechanical boiling technology was as follows:soaked 60 min with 10 times the amount of water,boiled 2 times with 60 min per time. Conclusion: This optimized technology was rational and feasible, it could provide experimental basis for improving quality and clinical efficacy of Sini decoction.

      摘要:Objective:To optimize water extraction technology of Yudan capsule. Method: With comprehensive retention rate of loganin, salvianolic acid B and chlorogenic acid as index,extraction times was optimized by single factor test;the amount of water,soaking time and boiling time were chosen as factors,water extraction technology of Yudan capsule was optimized by orthogonal design. Result: Optimal extraction technology was as follows:soaked 0.5 h with 8 times the amount of water,decocted for 1 h,filtered,residues added 6 folds the amount of water and decocted for 1 h. Conclusion: This optimized extraction technology was stable and reasonable, which provided experimental basis for production of Yudan capsule.

      摘要:Objective:To optimize prescription ratio of tetrahydropalmatine gel. Method: With 24 h accumulative permeation amount as index,the amount of carbomer,azone,propylene glycol,glycerine was chosen as investigating factors,prescription of tetrahydropalmatine gel was optimized by orthogonal design,and to examine in vitro drug release property of this gel. Result: Optimum formulation contained 1% tetrahydropalmatine,0.5% carbomer-940,3% azone,15% propylene glycol,10% glycerol. Conclusion: This optimized formulation was reasonable,preparation technology was stable and simple,its in vitro penetration was in line with zero order kinetic equation.

      摘要:Objective:To compare differences of pressurized microwave-assisted extraction method(PMAE), focused microwave-assisted extraction method (FMAE) and post microwave irradiated-reflux extraction method(PMIRE) in extracting alkaloids from Sophora flavescens,and investigate extraction process and microwave mechanism of these 3 kinds of microwave assisted extraction methods. Method: With oxymatrine and matrine as target alkaloids, which were determined by HPLC,thermodynamic functions were calculated by optimizing microwave power, extraction temperature,extraction time,solid-liquid ratio,medicinal granule and other factors,surface structural change of herbs was observed by electronic scanning microscope. Result: Compared with conventional solvent reflux extraction(SRE),extraction efficiency of PMAE,FMAE and PMIRE increased significantly,ΔH°and ΔS°of alkaloids increased obviously,but ΔG°became smaller.Microwave irradiation could enable cell wall of herbs surface structure to rupture,and strengthen extraction process. Conclusion: Extraction efficiency of alkaloids from S. flavescens with microwave assisted extraction was superior to SRE;Extraction efficiency had large differences by different microwave assisted manners,irradiation degree and extraction process;PMAE had shortest extraction time in the closed system;Because of strong microwave direct role,PMIRE had the best cell-rupture and extraction yield,and its equipment was simple and easy-operating.

      摘要:Objective:To optimize matrix prescription and preparation technology of compound gallnut gel ointment. Method: With skin following,film residue,matrix uniformity,ductility,viscosity,hardness and others as multi-sensory indexes,matrix prescription was optimized by synthetic weighted mark method and single factor test. Result: Optimum preparation technology was as follow:drug-PVPK90-CMCNa-PAASNP700-glycerol-1,2-propanediol-EDTA(10.5∶1.4∶0.6∶5∶18∶5∶1). Conclusion: Matrix of these prepared compound gallnut gel ointment had great drug loading capacity,good adhesion and better properties.

      摘要:Objective:To investigate equilibrium solubility and apparent oil-water partition coefficient of matrine at different pH values,and provide basis for development design of matrine liver targeting preparation. Method: Equilibrium solubility and apparent oil-water partition coefficient of matrine at pH 2~11 phosphate buffer were determined respectively,apparent solubility was determined by saturated solution method and shake-flask method,oil-water partition coefficient was calculated through mass concentration ratio of oil phase and water phase after balanced of matrine distribution,the concentration of sample liquid was determined by HPLC. Result: Matrine could dissolve in different pH solution,oil-water partition coefficient had tendency to increase with increasing of pH,it increased significantly in organic solvent,and solubility increased with polarity increasing.Solubility in water 70.26 g·L-1,oil-water partition coefficient of matrxine (lgPapp) was greater than 1.0 when pH>7.00,it could be easy to drug absorption. Conclusion: Matrine had hydrophilic and lipophilic,lipophilic was better under alkaline conditions.

      摘要:Objective:To optimize extraction technology of some herbs from compound Yangshen Yizhi tablet. Method: Reflux ethanol extraction was used,with the content of ferulic acid as index,which was determined by HPLC,L9(34) orthogonal design was used to optimize extraction technology with the concentration of ethanol, the amount of extraction solvent and extraction times as factors. Result: Optimum extraction technology was:extracted 2 times with 8,6 times the amount of 70% ethanol,1.5 h per time. Conclusion: This optimized extraction technology of compound Yangshen Yizhi tablet was stable and reliable.

      摘要:Objective:To synthesize and characterize hyaluronic acid grafted octadecyl (HA-C18) amphiphilic polymer. Method: HA-C18 polymer was prepared by acylation reaction with carboxyl group of hyaluronic acid and amine group of octadecyl,chemical structure of(HA-C18 polymer were determined by FTIR analysis.Self-aggregation behavior of HA-C18 polymer was evaluated,and critical aggregation concentration was determined;With intravenous accessories of Tween 80 and Cremophor EL as references,hemolysis of HA-C18 polymer was evaluated. Result: The critical aggregation concentration of HA-C18 copolymer was 10.0 mg·L-1.Hemolysis of HA-C18 micelle was far lower than Tween 80;But hemolysis of Cremophor EL was slightly lower than HA-C18 micelles,but their difference was not obvious. Conclusion: HA-C18 copolymer could be used as a novel injectable drug carrier for poor water soluble drugs.

      摘要:Objective:To optimize matrix formulation of Geranium wilfordii cataplasm. Method: With adhesive force, shapability and general sensory as comprehensive evaluation index,ratio of NP700,tartaric acid,PVP-K90 and dihydroxy alumimun aminoacetate were chosen as investigated factors,L9(34) orthogonal experiment was used to evaluate properties of cataplasm with different materials ratio. Result: Optimum matrix formulation of cataplasm was NP700-tartaric acid-PVP K90-dihydroxy alumimun aminoacetate 7.5∶0.4∶3∶0.05. Conclusion: This optimized formulation was feasible and stable with better performance.

      摘要:Objective:To optimize refining process of total saponins from extracts of Astragalus membranaceus. Method: With total saponins from A. membranaceus as index,the content of it was determined by UV spectrophotometry,adsorption capacity,kind and its amount of elution solvent were investigated by single factor test; Sample quantity,adsorption time and elution flow velocity were chosen as factors,elution conditions were optimized by orthogonal test. Result: optimized refining process was:mixed with 5 mL sample solution and 10 mL water,on column,adsorption time 1.0 h,eluted with 10 times the amount of water and 5 times the amount of 70% ethanol,collected eluent,elution flow velocity 2 mL·min-1.Eluent was decompressed recovery to dried,purity of total saponins was up to 87.63% after drying at 105 ℃. Conclusion: This optimized technology was simple and feasible,it could be better to purify total saponins from A. membranaceus.

      摘要:Objective:To optimize extraction process parameters of Sophorae flavescens. Method: With total alkaloids from S. flavescens and dry extract as indexes,extraction solvent and pH of solvent were investigated by single factor test;With total weight of the content of total alkaloids and solid as indexes,Orthogonal design was used to investigate effect of solvent consumption,extraction time,extraction times on extraction process. Result: Optimal extraction process was:extracted 3 times with 6 times the amount of 0.2% hydrochloric acid,2 hours each time. Conclusion: This optimized technology was stable and feasible,it could be used to industrial extraction of S. flavescens.

      摘要:Objective:To study on chemical constituents of the ethyl acetate extract part of Stelmatocrypton khasianum. Method: Ethyl acetate extract part were isolated and pured by silica gel column chromatography,MCI and Sephadex LH-20,Their chemical structures were elucidated by spectral data analysis(1H-NMR and13C-NMR). Result: Five compounds were isolated from the ethyl acetate extract part,they were identified as syringaresinol (Ⅰ),diasyringaresinol (Ⅱ),episyringaresinol (Ⅲ),justiciresinol (Ⅳ), threo-4', 4″, 7″, 9″-tetrahydroxy-3, 3', 3″, 5-tetramethoxy-4, 8″-oxy-7,9'∶7',9-diepoxylignan(Ⅴ). Conclusion: Five kinds of compounds were isolated from S. khasianum for the first time.

      摘要:Objective:To develop a HPLC method for the simultaneous determination of β-elemene in lyophilized powder for injection of β-elemene-polybutylcyanoacrylate nanoparticles. Method: The analysis was performed on DiamonsilTMC18 (4.6 mm×200 mm, 5 μm). The mobile phase was MeOH-water (95∶5),The wavelength of the detector was set at 210 nm. Result: β-Elemene presented a good linear correlation in range of 0.224-1.792 μg (r=0.999 6). The average recovery was 97.0%(RSD 1.3%). Conclusion: The method is simple, rapid and accurate, and suitable for the quality control of lyophilized powder for injection of β-ELE-PBCA-NP.

      摘要:Objective:To establish a new method to identify the peeled and non-peeled pieces of Platyeodon grandiflorum. Method: TLC and HPLC chromatography were used to identification. Result: There have significant differences in two kinds of pieces. The non-peeled pieces have three extra spots more than the peeled pieces and the HPLC chromatography showed five extra peaks expect thirteen common peaks. Conclusion: The established method was simple, it could be used for evaluation of different pieces of P. grandiflorum more effectively. Meanwhile, it also enriches the quality evaluation method.

      摘要:Objective:To determine rapidly the content of water-extract in Salvia miltiorrhiza by near-infrared spectroscopy and the chemometrics method. Method: A calibration model of near-infrared spectroscopy was built up for determination of the content of water-extract in S. miltiorrhiza by partial least squares (PLS) method. Result: The best spectral pretreatment method is MSC, the best spectral collection range is 7 038.39-4 418.78cm-1,and main fraction is 6,correlation coefficient of calibration cross validation (R2)was 0.984 3,root mean square error of cross validation (RMSECV) was 0.857, and external prediction deviation RMSEP of 11 samples was 0.787.Predicted the average recovery was 100.03%. Conclusion: It was feasible to determine the water-extract content in S. miltiorrhiza. This method is fast and simple with consequence accuracy,which should be applied more extensively in traditional Chinese medicine system.

      摘要:Objective:To develop a HPLC method for determination of the content of luteolin and diosmetin in Buchnera cruciata. Method: Samples were extracted for 90 minutes with methanol,a Agilent Extend-C18 column(4.6 mm×250 mm,5 μm) was used for composition separation and the mobile phase consisted of acetonitrile-0.7%acetic acid (30∶70) with a flow rate of 1.0mL·min-1.The detection wavelength was at 350 nm, column temperature was at 35 ℃. Sample volume is 10 μL, The content of luteolin and diosmetin in B. cruciata was determined by external standard method. Result: The linear of luteolin and diosmetin concentrations were within 1.126-112.6 mg·L-1(r=0.999 7),0.886-88.6μg·ml-1(r=0.999 5), the average recoveries of luteolin and diosmetin were 101.3%(RSD 1.2%,n=6), 99.4%(RSD 2.2%, n=6). Conclusion: The method is simple, accurate and reproducible with good stability, can be used as quality control methods for B. cruciata medicinal materials.

      摘要:Objective:To determine the content of chlorogenic acid, 3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid in Chrysanthemi Indici Flos of different areas. Method: HPLC was used. The chromatographic separation was performed on a Diamonsil C18(4.6 mm × 250 mm,5 μm) column eluted with gradient mobile phase of methanol-0.1%H3PO4,the detection wavelength was set at 326 nm and column temperature was set at 25 ℃. Result: Nice linear relation between the area and content when the amount was within 1.00-20.00,4.64-92.80,2.233-44.66 mg·L-1.The recoveries were 98.97%,99.14% and 99.39% respectively,RSD was 1.81%,1.66% and 1.36% (n=6) respectively. Conclusion: The method is accurate,reproducible and suitable to determine these active components in Chrysanthemi Indici Flos. It can be used for quality control of Chrysanthemi Indici Flos.

      摘要:Objective:Comprehensively compare and evaluate quality of Guizhou Lysionotus pauciflorus. Method: HPLC method and UV method were used on different orgin of Guizhou, different harvest time, different processing method for determination of the origin of L. pauciflorus, for two kinds of chemical composition method,by using principal component analysis and comprehensive quality evaluation comparative study. Result: When the L. pauciflorus products in the Yongzheng, cold blunt village and reaps in July and is dryed, its comprehensive quality was best. Conclusion: Different origin, different harvest time, different processing method of the origin of L. pauciflorus affect the quality deeply. The study results provide certain scientific basis for the comprehensive quality of L. pauciflorus and variety breeding, base construction of GAP, the rational development and utilization of medicinal materials.

      摘要:Objective:To establish the methods for the quantification and quantitation of Kudouzi tablets. Method: Sophora alopecuroides was identified by TLC.The content of matrine was determined by HPLC. HPLC method was performed on a Phenomenex-Pack Gemini-NX 5U C18 110A(4.6 mm×250 mm,5 μm)column with a mobile phase of acetonitrile-0.01 mol·L-1ammonium carbonate water(20∶80).The flow rate was 1 mL·min-1.The wavelength of detector was 220 nm and the column temperature was set at 30 ℃. Result: Characteristics of identification were very obviously.The linear range of matrine was 4.37-26.22 mg·L-1(r=0.999 5) with an average recovery of 102.59%(RSD 0.75%,n=6). Conclusion: The method can be used for the quality and quantity control of Kudouzi tablets.

      摘要:Objective:To found HPLC method for the determination of nomadic gallic acid in diyu tannin extract, then offer some references for the extraction process and quality standard study of diyu tannin. Method: The chromatographic condition was investigated. The methods of preparing sample solution, specialization, linearity and linear range, precision, stability, recovery and the content of samples was determined. Result: The content of nomadic gallic acid was determined at 272 nm detection wavelength with methyl alcohol-0.05% phosphoric acid aqueous solution(5∶95)as mobile phase, the flow rate was 1.0 mL·min-1, and the column temperature was at 30 ℃. The methods of preparing sample solution,specialization, linearity and linear range, precision, stability,recovery of this method all met the relative requirements. The content of gallic acid in the preparing sample ranged from 3.21% to 4.05%, the average was 3.71%. Conclusion: The results indicated that above-mentioned method was not only scientific, reasonable, feasible, but simple, rapid and accurate as well; what follows next, it can provide the basis for the relative study of tannins.

      摘要:Objective:To investigate the application of isolated heart perfusion technique on evaluating the bioactivity of Danshen Injection, and lay the foundation for further improving the standards of quality control. Method: Isolated guinea pig heart perfusion was performed with Langendorff method to measure the Danshen Injection if it had any effects on coronary flow and then use the percentage increase in coronary flow as an index to investigate its biological activity. After inspection of dose-effect relationship and reproducibility studies, the biological activity would be tested from 12 batches of Danshen Injection from the four manufacturers. Result: Danshen Injection has a significant effect on increasing the coronary flow. Under a certain condition, Danshen Injection showed some dose-effect relationship in the range of 0.1 to 0.6 mL, and in the dose of 0.3 mL, repeatability of the activity was good both in the same or different isolated heart within a certain time. Conclusion: It may be of great possibility that Langendorff isolated heart perfusion technique will become a good method for evaluating the bioactivity of Danshen Injection after further improvement and exploration.

      摘要:Objective:Determining the plasma protein binding rate of flavonoids in Compound Hongcao freeze-dried powder for injection to provide a reference for clinical safe medication and the method for the plasma protein binding. Method: The ultra-high performance liquid chromatography-mass spectrometry (UPLC-ESI-MS/MS) as a detection tool, using equilibrium dialysis method and ultrafiltration to study the plasma protein binding rate of orientin, vitexin, cynaroside and quercitrin, and compare the differences between the results. Result: When the drug-plasma concentrations were from 5-100 mg·L-1, the results showed high plasma binding ability of these compounds with two method and the binding rates were independent of the investigated concentrations. However, two method determination of vitexin and cynaroside showed statistical difference(P<0.01). Conclusion: The established methods as equilibrium dialysis and ultrafiltration both meet the require of determination the plasma protein binding and the two method had showed difference in some compounds.

      摘要:Objective:A HPLC method was established for the determination of berberine hydrochloride,baicalin and wogonin in Yankening Pian. Method: Shim-pack VP-ODS(4.6 mm×250 mm,5 μm) column was used with the mobile phase of methanol-acetonitrile-water,which was adjusted to pH3.0 by 0.1%phosphoric acid solution and triethylamine by gradient elution. The flow rate was 0.8 mL·min-1. The detection wavelength was set at 265 nm. Result: The HPLC method showed a good linear relationship in the range of 3.838-115.1 mg·L-1(r=0.999 8)for berberine hydrochloride,15.51-465.2 mg·L-1(r=1.000 0)for baicalin,7.005-210.1 mg·L-1(r=1.000 0)for wogonin. Recoveries for the three components were all above 98%. Conclusion: The methods is simple,accurate and repeatable,which can be used for the quality control of the tablets.

      摘要:Objective:To develop a method to determine naringin, hesperidin and neohesperidin in Citri Reticulatae Pericarpium and Aurantii Fructus. Method: HPLC was used to determine directly naringin, hesperidin and neohesperidin of Citri Reticulatae Pericarpium and Aurantii Fructus. Phenomsil C18(4.0 mm×250 mm,5 μm)as the chromatographic column, and acetonitrile-10% aqueous phosphoric acid (24∶76)as the mobile phase, the flow rate of 1.0 mL·min-1,the column temperature was at 40 ℃. Result: The linearity of verbascoside was good in the range of 6.05-60.5,4.34-43.4 and 7.14-71.40 mg·L-1(r>0.999 5).The recovery of the added sample was 98.65%,98.66% and 100.78% with RSD of 2.01%, 1.52% and 2.11%. Conclusion: The method is simple, accurate and can be used for the determination of naringin,hesperidin and neohesperidin in Tangerine peel and Aurantii Fructus extract.

      摘要:Objective:To establish an HPLC method for the determinetion of naphthoquine in antimalaria drug Artemisinine Naphthoquine Primaquine Tablets. Method: The assay was performed by a Diamonsil ODS C18 columm(4.6 mm×250 mm,5 μm)and a mobile phase of acetonitrile-0.25% diethylamine with radio of 23∶77(pH 2.5,adjusted with phosphoric acid),and the flow rate was 0.8 mL·min-1.The wavelength of detector was set at 342 nm and the column temperature was maintained at 25 ℃. Result: Naphthoquine has a good linearity in the rang of 0.16-0.8 μg (r=0.999 9),and the average recovery was 99.56%with RSD of 0.32%(n=9). Conclusion: The method was simple, accurate and suitable for the determination of naphtoquine in Artemisinine Naphthoquine Primaquine Tablets.

      摘要:Objective:To establish a high performance liquid chromatographic method (HPLC) for determination of baicalin in relieving itching cream intermediate products. Method: Agilent Eclipse Plus C18 column was used to separate the analyte with a mobile phase consisting of methyl alcohol and 0.37% phosphoric acid at a flow rate of 1 mL·min-1. The detection wavelength was set at 280 nm. Result: Baicalin showed good linearity (r2=0.999 9) in the range of the tested concentration (0.0792-0.792 μg); the average recovery of the method was 98.87% with RSD 1.14% (n=6). This method was used to successfully determine 3 batches relieving itching cream intermediate products, the content of baicalin in aqueous extract of Scutellaria Baicalensis was 5.75, 6.08, 5.94 g·L-1. Conclusion: This is a more stable, sensitive and precise method for monitoring relieving itching cream intermediate products, which could also be used as the quality control method for the process of the relieving itching cream.

      摘要:Objective:To establish the quality standards of Fengliao Changweikang dispersible tablets. Method: Daphniphyllum calycinum and Polygonum hydropiper were indentified by TLC, and the content rudin was determined by HPLC. Result: The characteristic identification by TLC was distinct and highly specific. Rudin showed a good liner relationship at a range of 0.8-1.6 μg (r=0.999 2), and the average recovery rate was 98.2% with RSD of 0.7%. Conclusion: The method is reliable,accurate and specific, and it can be used for quality control of this preparation.

      摘要:Objective:To establish the quality standard for Qing'e Wan. Method: (+)-Pinoresinol di-O-β-D-glucopyranoside(PDG) was determined by HPLC. Result: The results of quantitative evaluation showed that (+)-pinoresinol di-O-β-D-glucopyranoside (PDG) concentration was linear in the range of 4.43-443 mg·L-1 (r=0.999 9), the average recovery rate was 100.99% (RSD 2.17%). This method met the analysis requirement. Conclusion: The established quantitative methods were simple, accurate, reliable and reproducible, which can be effectively used for the quality control of Qing'e Wan.

      摘要:Objective:To study the constituent changes of Polygonati Rhizoma before and after processing. Method: Polygonati Rhizoma was prepared with wine by stewing for different time. Headspace injection-GC-MS was used and the structures of compounds were identified by Data Base Retrieval and compared with the retention indexes of n-alkanes standards. The relative content of every constituent was obtained from the peak area normalization method of chromatographic peaks. Result: There are 12 common compounds between raw products and processed products, yet the relative content was different. Compared with raw products, 20 compounds produced and 7 disappeared after different processed for different time. Conclusion: The chemical constituents and content have changed in the root of Polygonati Rhizoma before and after processing.

      摘要:Objective:To establish a RP-LC/ESI-MS combined with HILIC/ESI-MS method for the research of metabonomics. Method: For RP-LC/ESI-MS, the assay was conducted on a Prevail C18 (4.6 mm×250 mm,5 μm). The mobile phase A was 10 mmol·L-1 ammonium formate (adjusted to pH 4 with formic acid) in water, B was acetonitrile-formic acid (99.9∶0.1). A gradient elution mode composed of A and B was used as the mobile phase. For HILIC/ESI-MS, the assay was conducted on a Alltima HP HILIC (2.1 mm×150 mm,3 μm).The mobile phase A was 10 mmol·L-1 ammonium formate (adjusted to pH 4 with formic acid) in water, B was acetonitrile-formic acid (99.9∶0.1). A gradient elution mode composed of A and B was also used as the mobile phase. Electrospray ionization (ESI) in the positive and negative ion mode was used as the ionization source. Nitrogen was used as the nebulizer gas and was maintained at a flow of 11.0 L·min-1 with a nebulizer pressure of 50.0 psi. The fragmentor voltage was set at 275.8 V. Mass spectra were recorded covering m/z 15-2200. Result: Endogenous compounds with different polarity in rat urine could be detected by using RP-LC/ESI-MS and HILIC/ESI-MS. Conclusion: This method could offer a reference for metabonomics study.

      摘要:Objective:To study the effective fraction and compounds from Honeysuckle on inhibition of bacterial biofilms. Method: The micro-plate method was used as bioassay-guided and compounds were isolated from effective fraction by repeated silica gel column chromatography. Research their effect of bacterial biofilms. Result: The ethanol extract fraction was active site and 15 compounds were obtained from it. Soya-cerebrosideⅡ ( 1 ), 5-o-cafffeoyl quinic acid butyl ester ( 2 ), ginnol ( 3 ), sweroside ( 4 ), 7-epi-vogeloside ( 5 ), stearyl ( 6 ), octacoylalcohol ( 7 ), protocatechuic aldehyde ( 8 ), protocatechuic ( 9 ), cinnamic acid ( 10 ), caffeic acid ( 11 ), ursolic acid ( 12 ), quercetin ( 13 ), oleanolic acid ( 14 ), ferulic acid ( 15 ). When the concentration was 1 g·L-1, the inhibition rates of compound 4, 5, 12 were 36.16%, 37.16% and 46.18% separately. Conclusion: Compound 1 isolated from Honeysuckle for the first time. The compound 4, 5, 12 had certain inhibition on bacterial biofilms of Escherichia coli biofilms.

      摘要:Objective:To establish GC method for the determination of sixteen residual organic solvents in Lianju Ganmao capsule. Method: The sample was directly injected into AT-624 wide-bore capillary column and analyzed with FID detector using butyl acetate as the internal standard and dichloromethane as the solvent. Nitrogen was used as carrier gas. Result: n-hexane, toluene,ethylbenzene, m-xylene, styrene, cumene,n-decane,n-butylbenzene, n-hendecane, divinylbenze, n-dodecane,naphthalene, n-tridecane,n-tetradecane,n-pentadecane showed a good linear relationship over the rang of 1-32 mg·L-1 and Benzene was linear at range of 0.2-6.4 mg·L-1(r≥0.998 4). The average recovery was in range of 87.2%-116.9%.Each of the relative standard deviations was no more than 10% (n=9). Conclusion: The method is convenient, rapid, accurate, high sensitivity and can be used for the simultaneous determination of sixteen residual organic solvents in Lianju Ganmao capsule.

      摘要:Objective:To analyze the chemical constituents of volatile oil from the leaves of Camellia chrysantha. Method: By extracting C. chrysantha with 75% ethanol, the filtrate was concentrated and extracted with ether to get the ether parts, and the chemical composition was identified and analyzed by GC-MS method. Combining with computer retrieval technology and search of national institute of standards and technology(NIST) compound spectral-library for identification, their relative content was determined by the area normalization method. Result: Sixty-one kinds of chemical constituents were isolated from the ether parts of C. chrysantha. The relative content is 78.41% in the total content, the main chemical ingredients were 2 -( oxo-18 alkyl) ethanol, two pentadecane, 1-iodo-hexadecane, and so on. Conclusion: The present GC-MS method is fast, simple and accurate to analyze the composition of ether parts of C. chrysantha. It initially revealed the chemical composition of the medicine, and provided the experimental basis for further study of its chemical composition and the medicinal quality evaluation.

      摘要:Objective:To study the chemical constituents of the butanol portion of Ailanthus altissima. Method: The butanol portion was isolated and purified by silica gel column chromatography, HP-20 and Sephadex LH-20. Their chemical structures were elucidated by spectral data (1H-NMR,13C-NMR). Result: Five compounds were isolated from the butanol portion. They were identified as betulin ( 1 ), uvaol ( 2 ), betulinic acid ( 3 ), phaseic acid ( 4 ), 1-O-(6-O-α-L-rhamnopyranosyl-β-D-glucopyranosyl)-4-allylbenzene( 5 ). Conclusion: Compounds 1-4 were isolated from this plant for the first time.

      摘要:Objective:Inducing callus and adventitious root from Scrophulariae Radix;isolating endophytes;and comparing Hapagoside yield of the different parts of Scrophulariae Radix. Method: Tender leafs of Scrophulariae Radix were cut into small pieces and sterilized to inoculate on MS,N6 mediums with different plant hormones to induce callus; Liquid medium were used in adventitious root inducing and medium transformed according to growing phase during the culture process;general method was used in endophytes isolated. Hapagoside detected with UV spectrophotometry at the wave length 255 nm. Result: Moderate density callus were induced with MS+NAA 0.05 or 0.2 or 2 mg·L-1+6-BA 2 mg·L-1 medium;mass adventitious roots obtained after 30 d’s vibrate cultivation; 4 strains of endophytes produce hapagoside were isolated from Radix Scrophulariae root; hapagoside content of callus, adventitious root and fermentation liquor of the four endophyte straints in orderly are 0.411, 0.099 5, 0.451, 0.444, 0.489, 0.440 g·L-1. Conclusion: Content of hapagoside in callus is 4 times than that of adventitious root;endophytes can yield almost the same amount hapagoside as callus, and have great potential in producing secondary metabolites.

      摘要:Objective:To identify the metabolites of isorhamnetin-3-O-β-D-rutinoside bio-transformed by rat intestinal flora in vitro. Method: Isorhamnetin-3-O-β-D-rutinoside was incubated with rat anaerobic intestinal flora in vitro with HPLC monitoring the biotransformation course, and a LC-MS method based on the combined use of ultra-performance liquid chromatography, electro-spray ionization, collision-induced dissociation and tandem mass spectrometry has been applied to an investigation of the structural characterization of biotransformation products. The metabolites were identified by accurate molecular weight, CID-MS-MS fragmentation information, combined with reference substance and literature data review. Result: Five metabolites-quercetin, isorhamnetin, kaempferol, quercetin-3-O-glucoside and isorhamnetin-3-O-glucoside were identified though the above method, and another compound, maybe, 5,7,3',4'-tetramethoxy flavonoid-3-O-glucoside, need more information to identify. Conclusion: During the biotransformation by rat intestinal flora in vitro, isorhamnetin-3-O-β-D-rutinoside usually lose its glycosyl ligands,-OH or be adding-CH3, and which lead to their lipophilicity and chemical diversity increasing.

      摘要:Objective:To study the relationship between the gallic acid content in Polygonum capitatum and soil fertility in different adaptation areas in Guizhou.The aim is to provide reference for the cultivation of P. capitatum. Method: The gallic acid contents and soil fertility indices on roots of P. capitatum from 41 adaptation areas were determined, and the data was analyzed by SPSS 16.0. Result: The content of gallic acid in almost 70% different populations was 0.15 %-0.25 %.The content of available ammonium nitrogen and organic matter in the soil is low; P. capitatum needs relatively more potassium and phosphorus; PH 5.65-7.73, suitable soil is slightly acidic or neutral. The correlation analysis shows that soil texture and fertility did not obviously effect the accumulation of the gallic acid content. Conclusion: During the cultivation of P. capitatum, it is recommended to choose a neutral or subacidic sandy loam, relatively fertile and convenient drainage-irrigation land, meanwhile, pay attention to maintain potassium and available phosphorus not less than 60.0, 100.0 mg·kg-1 respectively in its growth process.

      摘要:Objective:To investigate the pharmacokinetic properties of cimicifugoside H-2 in rats, and the absolute bioavailability of cimicifugoside H-2 in vivo were also scarcely reported. Method: SD rats were administrated an intravenous dose of cimicifugoside H-2 (5 mg·kg-1) and oral dose of cimicifugoside H-2 (5 mg·kg-1) or Cimicifuga foetida extract (50 mg·kg), respectively. At different time points, the concentrations of cimicifugoside H-2 in rat plasma were determined by LC-MS-MS method. Main pharmacokinetics parameters were estimated by non-compartmental analysis using the DAS 2.0 software. Result: The curves showed excellent linear regressions within the range of tested concentrations. The intra-and inter-day variations were below 11% in terms of RSD. The recoveries were 87.0%-91.6% for spiked cimicifugoside H-2 samples. The PK parameters after iv administration of individual cimicifugoside were respectively: elimination half-life 1.03 h-1; clearance 691 mL·h-1·kg-1. PK parameters following oral administration of the extract and cimicifugoside H-2 was respectively: Tmax 0.5 and 4.0 h. Absolute oral bioavailability (F) was 21.2% and 36.7%. Conclusion: This method proved to be specific, accurate and sensitive to be applied to the pharmacokinetics studies of H-2 in rats. Comparison of the pharmacokinetics of cimicifugoside H-2 given alone and together in rat suggest that administration of herbal preparations of Cimicifuga for clinical use may provide longer duration of action than administration of single isomers.

      摘要:Objective:To prepare Saussurea involucrata microemulsion,investigate relationship between retention volume and time of of S. involucrata microemulsion in abdominal skin of mice,investigate its pharmacokinetics of local transdermal drug delivery,and investigate its skin absorption effect. Method: Characteristics of prepared S. involucrata microemulsion were determined including polydispersity index,size distribution and Zeta potential and other physical and chemical properties.With chlorogenic acid and rutin as indexes, biological retention volume of chlorogenic acid and rutin in administration skin with different time was determined by HPLC,and pharmacokinetic parameters were fitted by DAS software. Result: Characteristics of prepared S. involucrata microemulsion including mean particle size,polydispersity index and Zeta potential were 54.51 nm, 0.281 and-0.687 mV.Pharmacokinetic process of microemulsion in rats' abdominal skin was fitted with two compartment model. Conclusion: Active ingredients of S. involucrata microemulsion could quickly permeate into skin,and accumulate long time in local skin.

      摘要:Objective:To study the effect of total saponins from Entada phaseoloides (TSEP) on insulin resistance in type 2 diabetes rats and exploring the molecular mechanism using HepG-2 insulin resistance hepatocytes. Method: ①Type 2 diabetic rat model was established by high-fat diet and low-dose streptozotocin. Insulin tolerance test was performed to determine the effect of TSEP on insulin resistance. ②Insuin resistance in HepG-2 cells was induced by 0.25 mmol·L-1 of palmitic acid. The expression levels of protein tyrosine phosphatase-1B (PTP-1B) among different treatment groups were compared by Western blot. Result: Compared with the model group, the insulin sensitivity in the TSEP treated rats was increased. In normal HepG-2 cells, the protein levels of PTP-1B were decreased after insulin treatment. However, its expression level could not be ameliorated by TSEP. Conclusion: TSEP can improve the insulin resistance in type 2 diabetes mellitus rats. A possible mechanism underlying the insulin resistanceby TSEP in hepatocytes may be related to modulating PTP-1B expression.

      摘要:Objective:To investigate the effects of Buyang Huanwu decocotion combined with Liuwei Dihuang Wan on learning memory,amyloid precursor protein (APP), growth associated protein 43 (GAP-43) and synaptophysin SYP expression in permanent cerebral ischemia model in rats. Method: To establish the permanent middle cerebral artery model in rats. Male SD rats were randomly divided into five groups: normal group, model group, Buyang Huanwu decocotion group, Liuwei Dihuang Wan group and combination group. After treatment for 30 days, Morris water maze was used to measure the learning memory, and the technology of Western blot was adopted to examine the expression of APP, SYN, GAP-43. Result: After the ischemia model was established for 30 days, the spatial memory was damaged obviously in model group. Buyang Huanwu decoction and Liuwei Dihuang Wan as well as the combination therapy could improve the spatial memory ability in different degrees. The escape latencies of the combination decoction were decreased significantly compared to the model group (P<0.05) on the second day of position navigation test. On the fourth day of position navigation test, the escape latencies in Buyang Huanwu decocotion group and the combination decoction group was decreased obviously (P<0.05). In the spatial probe testing, the piercing number in Buyang Huanwu decocotion group and the combination decoction group were increased significantly compared to the model group, the first time through the original plat of the combination decoction group was shorted remarkably compared to the model group (P<0.01). Compared to sham group, the APP expression of the model group was increased significantly and the GAP-43 and SYN expression was descended obviously (P<0.01, P<0.05) while Buyang Huanwu decoction group and the combination decoction could decrease the APP expression and increase the GAP-43, SYN expression remarkably compared to the model group (P<0.05). There was no statistical significance inj the APP expression between Liuwei Dihuang Wan group and model group. The GAP-43 and SYN expression was increased obviously (P<0.05). Conclusion: Buyang Huanwu decoction combined with Liuwei Dihuang Wan can alleviate the cerebral ischemia injury and reduce the abnormal APP accumulation. In addition, the combination decoction can reduce GAP-43 and SYN synthesis and promote regeneration of neurite, and promote the recovery of learning memory.

      摘要:Objective:To study the effects of Naomai Shukang capsule on transient ischemic attack in mice. Method: Kunming mice were divided into Naomai Shukang capsule group (1.8,0.9,0.45 g·kg-1), the model group, sham operation control group,and the positive control group (Xueshuantong 1.125 g·kg-1,nimodipine 0.03 g·kg-1), mice were fed Naomai Shukang capsule for 7 d, on the 8 d, after 1 h of administration, both sides of the common carotid artery were ligatured for 30 min and reperfusion for 15 min to make transien cerebral ischemia model; superoxide dismutase(SOD)and malonaldehyde(MDA) content were determinated; section of brain tissue was homogenated, Na+-K+-adenosine triphosphatase(ATPase), Mg2+-ATPase, Ca2+-Mg2+-ATP,Ca2+-ATPase activity of brain homogenate were measured, part of brain tissue was calculated after weighing for brain water content. Result: Naomai Shukang capsule could enhance Na+-K+-ATPase, Mg2+-ATPase, Ca2+-Mg2+-ATP,Ca2+-ATPase activity in brain,and decrease MDA content, increase SOD activity, brain edema was relieve induced by cerebral ischemia. Conclusion: Naomai Shukang capsule can improve ATP energy of brain, scaveng oxygen free radicals, relieve cerebral edema, protect brain cells, reduce brain structure and function damage.

      摘要:Objective:To observe the effect of Huoxue Jiedu formula on angiotensin Ⅱ(Ang Ⅱ) and endothelin-1(ET-1) in retina of diabetic rats. Method: A single intraperitoneal injection of streptozotocin(65 mg·kg-1)was used to induce diabetic rats. Model rats were randomly divided into diabetes mellitus(DM)group,Huoxue Jiedu formula groups(15.4,7.70,3.85 g·kg-1) and Doxium group(0.167 g·kg-1).The normal group and model group were given normal water by intragastric administration. All rats were killed after 20 weeks. The serum AngⅡ and ET-1 levels were measured. Immunological histochemistry assay was used to observe the express of ET-1.The expressive abundance of ET-1mRNA in rat's retina were detected by real-time quantitative PCR. Result: The levels of AngⅡ and ET-1 in serum of model group were (417.95±156.11),(330.12±81.59) ng·L-1, which was higher than control group.The retinal ET-1 protein expression and ET-1mRNA were significantly higher in DM rats than in control group(P<0.01).The levels of ET-1 in serum of Huoxue Jiedu formula groups were lower than those in model group(P<0.01).Compared with the model group, the expression of ET-1 in Huoxue Jiedu formula groups depressed evidently(P<0.01)and the expression of ET-1mRNA were reduced than those in model groups(P<0.01). Conclusion: Huoxue Jiedu formula medicine can reduce the expression of ET-1 in retina of diabetic rats.

      摘要:Objective:To study the effect of Wendan decoction on connexin 43(Cx43) mRNA expression in brain tissue of rats with schizophrenia. Method: Fifty SD rats were divided five groups by random, including normal group(A),model group(B),Wendan decoction high-dosage group(C),medium-dosage group(D) and low-dosage group(E),respectively. The A and B groups were given normal saline and the C,D, E groups were given Wendan decoction(40,20,10 g·kg-1) every day, which lasted for 21 days. Dizocilpine maleate(MK-801) 0.6 mg·kg-1 was ip given to establishthe rat schizophrenic model. Then rat behaviors were observed for 3 days all rats were sacrificed. Then RT-PCR method was used to detect Cx43 mRNA expression in the nervecells of the every group rats. Result: The Cx43 mRNA expression of group B was lower than that of group A (P<0.05).And the Cx43 mRNA expression of group C,D and E was rose compared with that of group B(P<0.05). Conclusion: Wendan decoction can reinforce the expression of Cx43 mRNA in the brain tissue of rats with schizophrenia.

      摘要:Objective:To study the mechanism of Linggui Zhugan decoction (LGZGD)interference on ventricular remodeling (VR) in rats after acute myocardial infarction (AMI), through observing the expression of nuclear factor-κB(NF-κB)and NF-κB mRNA of myocardial tissue and the content of NF-κB in ventricular remodeling rats with AMI. Method: AMI model was produced by ligation of coronary artery. 2 weeks after modeling,rats were randomly classified into model, captopril, and low (2.1 g·kg-1), middle(4.2 g·kg-1) and high (8.4 g·kg-1)dosage of LGZGD group. Control group and the other 5 groups were administered medications intragastrically for 4 consecutive weeks. The content of NF-κB in myocardial tissue and serum was detected by ELISA and the expression of NF-κB was assayed by RT-PCR and the expression of NF-κB mRNA by Western blot. Result: For sham-operation, model, low, middle and large dosage of LGZGD group and captopril group, the content of NF-κB of myocardial tissue was 0.190±0.011,0.772±0.026,0.366±0.059,0.295±0.033,0.235±0.013,0.341±0.023.The relative expression of NF-κB mRNA were 1.000, 26.875, 6.574, 4.340, 1.194, 5.540.The content of serum NF-κB were (125.85±14.76),(196.98±17.79),(163.89±20.08),(131.73±10.47),(141.93±10.33),(133.93±9.27)ng·L-1.The content of NF-κB in serum and the expression of NF-κB, NF-κB mRNA of myocardial tissue of model group increased significantly compared with control group (P<0.01). Low, middle and high dosage of LGZGD and captopril group could decrease the content of NF-κB in serum and inhibited the expression of NF-κB,NF-κB mRNA in myocardial tissue of model rats significantly compared with model group (P<0.01 or P<0.05). Conclusion: The mechanism of LGZGD interference VR of post-AMI seems to be related to the inhibition of NF-κB.

      摘要:Objective:To observe the effect of salvianolic acid B(Sal B) on the angiogenesis in ischemic myocardium of the rats with myocardial ischemia. Method: The left coronary of SD rats were blocked to make the models of acute myocardial ischemia. Sixty rats were randomized into model group, Sal B groups (1.6,3.2,6.4 mg·kg-1) and vascular endothelial cell growth factor(VEGF) group. A sham operation group was set up. Ten rats were included in each group. Two weeks after administration, HE staining was used to detect the myocardial infarct size; using kits to detect the contents of nitrogen oxide(NO), NO synthase(NOS) and VEGF in myocardial tissue; immunohistochemical method was employed to detect microvessel counts(MVC) in ischemic myocardium. Result: The infarct size of myocardium in Sal B 3.2,6.4 mg·kg-1 groups (7.18±3.15)%,(5.74±2.02)% was lower than that in the model group(14.84±5.40); Sal B could increase the contents of NO,NOS and VEGF in myocardial tissue. The number of MVC in the marginal zone of myocardial infarct in Sal B 3.2,6.4 mg·kg-1 groups (28.6±5.86),(30.20±5.07) was higher than that in the model group(15.60±4.62). Conclusion: Sal B may promote angiogenesis of ischemic myocardium in the rats with myocardial ischemia. Its mechanism might be related to promoting the contents of VEGF and NO.

      摘要:Objective:To study the compatibility of San'ao decoction. Method: Based on orthogonal design, mice was divided into 16 groups, 16 in each group according to L16(43) table. Subcutaneous sensitization was undertaken on day 1 and 8, 5% OVA atomization was applied to stimulate sensitization from day 15 to 28, before which San'ao Decoction with the corresponding compatibility proportion was given. Airway reactivity, the number of eosinophils (EOS) in blood, ESO in BALF, and white blood cells, pathological change of the lung and trachea were detected on the 29th day. To confirm the experiments, 48 mice were divided into 3 groups: blank group, model group and optimum compatibility group with 16 in each. The above mentioned methods and indicators were applied and detected. Result: Based on synthetic sores, the optimum of San'ao decoction was as follows: 6 g Ephedrae Herba; 9 g Armeniacae Semen Amarum; and 3 g Radix et Rhizoma Glycyrrhizae. The sequence of priority was like: Ephedrae Herba>Armeniacae Semen Amarum>Radix et Rhizoma Glycyrrhizae. The confirmed experiments showed significant difference(P<0.01) in airway reactivity, the number of EOS in blood, ESO in BALF, and white blood cells, pathological change of the lung and trachea compared with model(A1B1C1). Conclusion: The optimum compatibility proportion of San'ao decoction is similar to that used in clinical practice; and their sequence of priority confirms the compatibility principle of San'ao decoction.

      摘要:Objective:In order to elaborate the traditional Chinese medicine(TCM) theory 'Cuzhi Rugan’scientifically, we estimated vinegar-steamed Schisandra sphenanthera based on anti-oxidation in vivo and in vitro. Method: We estimated the difference in the aspect of anti-oxidation between vinegar-steamed and non-vinegar-steamed S. sphenanthera by (DPPH) radicals scavenging in vitro through ultraviolet spectrophotometry at 517 nm, ascorbic acid as the positive control. Meanwhile we measured the difference in the aspect of liver tissue (T-AOC), between vinegar-steamed and non-vinegar-steamed S. sphenanthera, intragastric administration with high(5 g·kg-1), medium(2.5 g·kg-1) and low(1.25 g·kg-1) doses per day and lasted ten days by rat acute liver injury model caused by CCl4 in vivo. Liver was collected 24 h after modeling. Finally, we comprehensively evaluated the anti-oxidation ability between vinegar-steamed and non-vinegar-steamed S. sphenanthera in vitro and in vivo. Result: The in vitro results showed that the ability of DPPH radicals scavenging of vinegar-steamed S. sphenanthera(IC50 1.887 1 g·L-1)was better than the non-vinegar-steamed one(IC50 4.571 2 g·L-1). The in vivo results showed that the ability of T-AOC in the rat liver tissue of vinegar-steamed S. sphenanthera and non-vinegar-steamed one were much higher than the positive control one in every doses(P<0.05). And the ability of T-AOC in the rat liver tissue of medium and low doses vinegar-steamed S.sphenanthera are much higher than the non-vinegar-steamed one(P<0.05). Conclusion: The ability of radicals scavenging by S. sphenanthera is enhanced by vinegar-steamed which is a frequently-used TCM prepared method, and we also try to elaborate the 'Cuzhi Rugan’in science.

      摘要:Objective:To observation the distribution of isorhynchophyline in liver tissues before and after compatibility with gastrodin and isorhynchophylline in rats. Method: HPLC was employed to determinate the isorhynchophyline in liver tissuesin rats before and after compatibility with gastrodia and unicaria;analyticalcolumn was WatersTM(4.6 mm×250 mm, RP 18.5 μm); the mobile phase consisted of methanol-water (70∶30), pH 8.0, fluid rate was 0.8 mL·min-1; the column temperature was 30 ℃,the detector wave length was set at 254 nm,the sample amount is 20 μL. Reslut: Isorhynchophyline in liver and kidney tissues was linear in the range of 0.16-4.0 mg·L-1. There were no significant differences in liver tissues of SD rats before and after compatibility. Conclusion: Gastrodin has a certain effects reduction in the liver tissues distribution.

      摘要:Objective:To investigate the anti-inflammatory effect of total flavones of Trollius chinensis Bge (TFTL). Method: The experiments of carrageenin-induced paw edema in rats, histamine-induced pigment oozing from skin vessel in rats and cotton pellet granuloma in rats were used to investigate the effect of TFTL (0.1,0.2,0.4 g·kg-1) on acute and chronic inflammation. Aspirin (Asp) and Jinlianhua tablets (JLHT) were used as positive controls. Result: In 0.2,0.4 g·kg-1 of TFTL groups, the intradermal area of pigment infiltration and pigment content in rats, the paw edema degree in rats and the weight of cotton pellet granuloma in rats were lower than those in the model groups(P<0.05,P<0.01). In 0.4 g·kg-1 of TFTL group, the intradermal pigment content in rats, the paw edema degree of rats and the weight of cotton pellet granuloma in rats were lower than those in JLHT 2.4 g·kg-1 group(P<0.05). In 0.4 g·kg-1 of TFTL group, the paw edema degree was lower than that of Asp 200 mg·kg-1 group(P<0.05). In the test of c arrageenin-induced paw edema in rats, the duration of anti-inflammatory effect in 0.2 g·kg-1 and 0.4 g·kg-1 of TFTL groups were 5-6 h, while the duration of anti-inflammatory effect in 0.2 g·kg-1 of Asp group were 3 h. Conclusion: TFTL showed significant anti-inflammatory effects on acute and chronic inflammation in a dose-dependent manner. Anti-inflammatory effect of TFTL was stronger than that of JLHT. The duration of anti-inflammatory effect of TFTL was longer than that of Asp.

      摘要:Objective:To investigate the effect of icariin on osteogenic differentiation and adipogenic differentiation of rat bone marrow stromal stem cells (rBMSCs). Method: rBMSCs were incubated with final concentrations of 1×10-5 mol·L-1 icariin under the conditions of osteogenic induction culture. The alkalin phosphatase(ALP) activity was determined at day of 3, 6, 9, 12 days and mineralized nodules were stained at the 15th days; Total RNA was isolated and the gene expressions of bone morphogenetic protein 2(BMP-2) and Runx-2 were investgated by Real time PCR. rBMSCs adipogenic differentiation were treatment by icariin with the concentration 1×10-5mol·L-1, triglycerides content was determined at day of 5, 10, 15 and 20,rathonum red staining was made at day of 21.Total RNA was isolated at 12, 24, 48 and 72 h to investigate the gene expression of PPARγ and C/EBPα mRNA by Real Time PCR. Result: Icariin significantly enhanced the ALP activity of the rBMSCs and the number of calcified nodules, and increased the gene expression of osteogenic factor BMP-2 and Runx-2.Icariin significantly reduced triglyceride levels and the formation of lipid droplets, and decreased gene expression levels of the adipogenic transcription factors PPARγ and C/EBPα. Conclusion: Icariin can regulate rBMSCs differentiation,and promote rBMSCs osteogenic differentiation and at the same time icariin can exert suppression of rBMSCs adipogenic differentiation.

      摘要:Objective:To observe the therapeutic effects of Polyrhachis vicina Roger on gout. Method: Kunming mice and Wistar rats were randomly divided into control group,model group,positive control group,P. vicina low,medium and high dose groups(1,2,4 g·kg-1), respectively. The anti-gout effects of P. vicina were studied by observing its effects on the level of blood uric acid in hyperuricacidemia mice induced by hypoxanthine and nomal mice, and rat’s foot swelling induced by microcrystalline sodium urate (MSU) after 15 days treatment. Result: Compared to the level of blood uric acid in control group(131.09±45.78) μmol·L-1and model group(198.63±47.27)μmol·L-1, medium(2 g·kg-1) and high dose groups(4 g·kg-1)of P. vicina alcohol extract,and high dose group(4 g·kg-1)water extract could reduce the blood uric acid in model mice, and satisfactory inhibited acute gouty arthritis,relieved rat’s foot welling in model rats induced by MSU in 3 hours.The effects of P. vicina alcohol extract on gout was better than that in water extract. Conclusion: P. vicina could inhibit the level of blood uric acid in hyperuricacidemia mice induced by hypoxanthine and inflammation induced by MSU,which indicates that P. vicina has therapeutic effect on gout.

      摘要:Objective:To study the effects of gingerol on the contents of liver microsomal cytochrome P450 and the activities of CYP2E2 and CYP3A in mice. Method: Gingerol(200,100,50 mg·kg-1·d-1) were ig given to male mice for 5 days, after the liver microsomes were prepared by calcium isolation, the effects of gingerol on the contents of mice liver heavy, microsomal protein, CYP450 and CYPb5 were determined and the activities of CYP2E2 and CYP3A were tested by using erythromycin and aminopyrine. Result: The gingerol had no effects on the contents of liver heavy and microsomal protein in mice treated with gingerol(200,100,50 mg·kg-1·d-1) but could markedly decrease and increase the contents of CYP450 and CYPb5,respectively(P<0.01); the activity of CYP2E1 was inhibited in mice treated with gingerol(200,100,50 mg·kg-1·d-1) (P<0.05) and the effect of inhibition became stronger with the dose increasing of gingerol. The activity of CYP3A was inhibited in mice treated with gingerol(200 mg·kg-1·d-1), (P<0.05). Conclusion: Gingerol has the effects of inhibiting the content of CYP450 and the activities of CYP2E1 and CYP3A, the degree of inhibition is related to dose.

      摘要:Objective:To observe the curative effect of Feixianfang on pulmonary fibrosis of rat model induced by bleocamicina. Method: One hundred and forty four healthy rats were randomly divided into six groups: normal group, model group, positive control group, Feixainfang high dose, middle dose and low dose group, 24 rats each group. The model were made with Szapiel's method, and the next day all rats were given the corresponding drugs with intragastric administration. The three treatment groups were given to high, middle and low dose of Feixianfang (3.2, 1.6,0.8 g·kg-1),positive control group was given to prednisone acetate (6 mg·kg-1). And normal group and model group were given normal saline, once a day, and the treatment lasted 21 days. Lungs were collected at the 7th day(10 rats),14th day(4 rats), and 21st day (10 rats)respectively. Result: Compared with model group, high dose treatment group showed significant difference in improving lung coefficient, increasing glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD) and decreasing malondialdehyde(MDA), nitrogen oxide(NO),transforming growth factor-β1(TGF-β1), platelet derived growth factor(PDGF) and tumor necrosis factor-α(TNF-α) (P<0.05 or P<0.01). Middle and low dose treatment group had the same effect as positive control group. And positive control group was better firstly than treatment group, but worse in intermediate stage and later stage. This directed that high dose treatment group had better effect than positive control group markedly, and the long-term effect was superior to prednisone acetate. Conclusion: Feixianfang is an effective formula in treating and preventing pulmonary fibrosis. It can intervene in the injury caused by bleocamicina with dose-effect relationship.

      摘要:Objective:To study the effects of Fufang Ejiao Jiang (FFEJJ) against 5-fluorouracil (5-FU) chemotherapy in hepatoma H22-hearing mice. Method: The SPF-Kunming mice were used to establish mouse model of hepatoma H22.The mice were randomly divided into 5 groups: tumor model group, 5-FU group (20 mg·kg-1, ip), 5-FU combined FFEJJ group. FFEJJ were orally administrated at a daily dose of 3, 1.5, 0.75 g·kg-1, respectively corresponding to high, middle and low dose. The normal control group was set up and each group had 10 mice. After continuous administration for 7 d, the peripheral white blood cell count (WBC), tumor weight, inhibition rate, spleen index and thymus index were measured. The peripheral blood lymphocyte transformation rate and the phagocytotic activity of peritoneal macrophages were detected. Result: The high-dose FFEJJ significantly reduced the tumor weight, elevated the spleen index, thymus index and WBC counts compared with 5-FU group (P<0.05). 5-FU decreased the lymphocyte transformation rate and peritoneal macrophage phagocytosis rate and the phagocytic index(P<0.01). These index in FFEJJ group were significantly higher than that of 5-FU group (P<0.05), and reached to (35.9±9.2)%, (41.9±6.3)%, and (0.74±0.17) in high dose FFEJJ group respectively. Conclusion: The results suggest that FFEJJ could enhance efficacy and reduce toxicity in hepatoma treatment as a supplement of 5-FU.

      摘要:Objective:To explore the effect of blood-activating and stasis-resolving medicines on type 2 diabetic rats. Method: Type 2 diabetic rats model was established by feeding high calorie-diet and injection of small-dose streptozotocin(STZ).The selected rats were randomly divided into six groups:normal control group,model group,diamicron group and Suxiao Jiuxin pills group,Diao Xinxuekang group,Compound Danshen dripping pills group. Fasting blood glucose was detected during the administration on 0 week,2th week and 4th week.After 4 week’s administration,oral glucose tolerance test,free fatty acid(FFA),triglyceride(TG),total cholesterol(TC),leptin,glycated serum protein fructose ammonia(GSP) were determined,pancreas and liver were examined. Result: After treatment of 4 weeks,the blood suger in each group was as follows::normal control(5.32±0.69)mmol·L-1,model fgroup(20.40±2.75)mmol·L-1; diamicron group(9.18±3.91)mmol·L-1,and Suxiao Jiuxin pills group(10.72±2.60)mmol·L-1,Diao Xinxuekang group(12.09±3.85)mmol·L-1,Compound Danshen dripping pills group(14.76±2.57)mmol·L-1. The TG in each group was as follows: normal control(0.50±0.08)mmol·L-1,model group(1.00±0.20)mmol·L-1;diamicron group(0.80±0.21)mmol·L-1,and Suxiao Jiuxin pills group(0.70±0.24)mmol·L-1,Diao Xinxuekang group(0.72±0.21)mmol·L-1,Compound Danshen dripping pills group(0.81±0.13)mmol·L-1. Adjust lipid metabolism level,reduce the level of atrophy lesion of pancrea and liver. Conclusion: The blood-activating and stasis-resolving medicines have therapeutic effects on type 2 diabetic rats.

      摘要:Objective:To study the effects of total alkaloids from Pinellia ternate (TAPT) on learning and memory ability in aging mice induced by D-galactose and its preliminary mechanism. Method: One hundred mice were randomly assigned into control group, model group, TAPT low, middle and high dose group(7.5, 15, 30 mg·kg-1), 20 mice in each group. The TAPT and the model groups were given 120 mg·kg-1 D-galactose (sc, for 8 weeks) to induce aging-mouse model, and the control group was given the same volume of saline water; meanwhile, the TAPT groups were given appropriate liquid (ig, for 8 weeks), while the control and the model groups were given the same volume of distilled water. The step-down test and the Y-maze test were adopted to reflect the learning and memory of mice; and the colorimetric method was used to determine the cellular content of malondialdehyde (MDA), the enzyme activity of superoxide dismutase (SOD) and acetylcholinesterase (AChE). Result: Compared to the model group, the TAPT treatment brought mice to shorten the time to reach the final in the Y-maze training at (21.9±2.9), (20.8±3.2) ,( 19.7±2.8) times respectively, and also reduce the mistakes in memory test at (3.8±1.4), (3.2±1.7),(1.7±1.1) times respectively. SOD activity in brain tissues of aging model mice reduced evidently at(28.98±13.08) U·mg-1, while MDA and AChE contents increased evidently at (2.78±0.21)nmol·mg-1 ,( 1.05±0.16)U·mg-1, while TAPT treatment could evidently decrease MDA(P<0.01) and AChE contents(P<0.01),and increase SOD activity(P<0.01). Conclusion: TAPT treatment could help aging model mice induced by D-galactose evidently to enhance their learning and memory, which might be related to its antioxidation and its inhibitory activity to AChE.

      摘要:Objective:To evaluate the effect of 4-methyl-cyclopentadekanone on learning and memory in vascular dementia rats. Method: Ninty rats were divided randomly into positive control group(6.1 mg·kg-1), model group, sham operation group, 4-methyl-cyclopentadekanone low, medium and high dose group(8.1, 2.7,0.9 mg·kg-1). The model was indueced by arteria carotis ligation, and after 7 days these rats were administrated orally once a day for 28 days and given a administration dose at 10 mL·kg-1.The model group and sham operation group were administrated normal saline. Related biochemical indicators and praxiology in each group were observed. Result: Avoiding dark counts and total cholinesterase(TChE) activity degrade obviously(P<0.05)while the intragastric administration was 8.1, 2.7 mg·kg-1, meanwhile the SOD activity increase at a dose of 8.1 mg·kg-1. Conclusion: 4-methyl-cyclopentadekanone can enhance learning and memory in vascular dementia rats.

      摘要:Objective:To explore effect and its mechanism of Didang decoction on chronic prostatitis. Method: Fifty rats were randomly divided into control group (distilled water 10 mL·kg-1), model group (distilled water 10 mL·kg-1), Didang decoction high-dose group (16.2 g·kg-1), low-dose group (8.1 g·kg-1),positive control group (Qianliekang 0.5 g·kg-1). Chronic prostatitis model was induced by Xiaozhiling injection 0.1 mL. The corresponding drugs were given. The plasma level of thromboxane B2 (TXB2), 6-keto prostaglandin F1α (6-keto-PGF1α), the tumor necrosis factor-α(TNF-α) was measured. Prostate tissue was collected to observe prostate tissue gland secretion and lumen, interstitial inflammatory cells and fibrous tissue. Result: Compared with model group, plasma TXB level (819±224)μg·L-1, Didang decoction high-dose group, low-dose group and positive control group (915±195),(1 068±64),(1 101±317) μg·L-1, is significantly descreased (P<0.05). Compared with pathological model group 6-keto-PGF1α(576±187)μg·L-1, Didang decoction high-dose group,low-dose group and positive control group (1 071±197),(838±197),(911±199) μg·L-1 is significantly increased (P<0.05). Compared with pathological model group T/K(2.89±1.04), Didang decoction high-dose group, low-dose group and positive control group(0.88±0.23),(1.34±0.31),(1.28±0.49) is significantly descreased (P<0.05). Compared with pathological model group TNF-α(410±129)μg·L-1, Didang decoction high-dose group, low-dose group and positive control group(144±35),(312±69),(238±84)μg·L-1 is significantly descreased (P<0.05). Pathological results show that Didang decoction high-dose group, prostate gland organization lumen, gland secretion, interstitial inflammatory cells, fibre hyperplasia with lower scores, with significant difference were. Conclusion: Didang decoction can lower TXB2, rise 6-keto-PGF1α level, make T/K ratio greatly reduced, at the same time for chronic prostatitis rats inflammation in the blood of the TNF-α possess the inhibition.

      摘要:Objective:To study the effect of small interfering RNAs(siRNAs) combined with Brucea javanica oil emulsion targeted ERCC1 Gene on reverting platinum resistance of human lung adenocarcinoma cell line A549/DDP. Method: The siRNAs which targeted the same sequences of ERCC1 gene were designed and synthesized.Human lung adenocarcinoma cell line A549/DDP was cultured and tranfected with siRNAs by Lipofectin2000.Experimental cell grouping:A549/DDP(Control),Brucea-treated group,siRNA-treated group,Brucea and siRNA treated group. ERCC1mRNA was assayed by RT-PCT,and the protein expression was detected by Western blotting.The efficiency of A549/DDP to cisplatin was determined by MTT method. Result: The expression of ERCC1mRNA and protein decreased significantly after transfection of siRNAs,B. javanica oil and B. javanica oil+siRNAs.The efficiency of A549/DDP to cisplatin reversed significantly. The drug-resistant multiple droped back to 9.72,8.89,7.59,6.83.The reversing effect increased significantly by siRNAs combined with B. javanica oil. Conclusion: Both siRNAs and B. javanica oil can reverse platinum resistance of human lung adenocarcinoma cell line A549/DDP.The inhibition effect increases significantly by siRNAs combined with B. javanica oil emulsion.

      摘要:Objective: To observe the effect of botulinum toxin type A (BTX-A),electrical field stimulation(EFS)and acetylcholine(ACh) on spontaneous contractility in gastric body and gastric fundus smooth muscle. Method: Muscle strips in gastric body and gastric fundus were prepared, and subdivided randomly into control group, EFS group, BTX-A(10 U·mL-1)group, BTX-A(10 U·mL-1)+ EFS group, ACh(100 μmol·L-1)group, ACh(100 μmol·L-1)+BTX-A(10 U·mL-1)group, Ach(100 μmol·L-1)+Atropine(1 μmol·L-1)group. The data were recorded by physiological experimental system of BL-420. Result: EFS enhanced the tension(P<0.05)and amplitude(P<0.01)in gastric body contractility, and similar results was observed in gastric fundus contractility; BTX-A decreased spontaneous contractile tension and amplitude(P<0.01)in gastric body and tension(P<0.05)in gastric fundus; BTX-A inhibited EFS-induced smooth muscle contractility including tension and amplitude(P<0.01)in gastric body,tension(P<0.01) and amplitude(P<0.05)in gastric fundus, BTX-A inhibited ACh-induced smooth muscle contractility including tension and amplitude(P<0.01)in gastric body and gastric fundus. Conclusion: EFS enhances smooth muscle spontaneous contractility in gastric body and gastric fundus; BTX-A inhibits gastric body and gastric fundus smooth muscle spontaneous contractility; BTX-A inhibits EFS and ACh-induced smooth muscle contractility in gastric body and gastric fundus.

      摘要:Objective:To study the antifatigue effect of Pingwei San(PWS) on rats of the spleen and stomach being stranded by dampness. Method: Rat models of the spleen and stomach being stranded by dampness was made. The rats were divided randomly into five groups: the control group, the model group and three groups administered with high, mediate and low dose of respectively. at the eighth day after modeling, the control group and the model group were administrated with physiological saline solution, the others three groups were administered with 1.5, 3, 6 g·kg-1 PRFS respectively. After final administrating, the behavioral changes of rats were observed and the exhaustive swimming time of rats was recorded. At the end of the experiment, the levels of blood plasma lactic acid(LA), serum lactate dehydrogenass(SLDH), serum urea nitrogen(SUN), hepatic glycogen, and hemoglobin(Hb) were determined. Result: The rat behaviors in the three groups with PWS were remarkably improved. The last exhaustive swimming time increased significantly, hepatic glycogen level of high-dose group rats was obviously increased(P<0.01), and SUN level of high-dose and mediate-dose group rats were decreased significantly(P<0.05). The levels of Hb, LA and SLDH in all animals were not different obviously. Conclusion: Decreasing disintegration of protein and amino acids,and increasing the level of hepatic glycogen in exercise load were possibly antifatigue mechanism of PWS.

      摘要:Objective:To study the protective effect of polysaccharides from Niuxin persimmon leaf on pancreas injury induced by streptozotocin (STZ) in diabetic mouse. Method: Diabetic mice model was established by injection of STZ. The model mice were randomly divided into 5 groups: model group, dimethyldiguanide group (32 mg·kg-1), low-, medium-and high-dosage groups of Niu Xin persimmon leaf polysaccharides (30, 60, 120 mg·kg-1). The drugs were given to mice daily for 15 consecutive days. Fasting 12 h at the end of 15th day, the blood samples were collected and centrifuged to obtain serum for test. Glucose oxidase (GOD) and radioimmunoassay (RIA) were used to examine the index of glycemia and insulin for fasting blood glucose(FBG) and fasting insulin(FINS); the levels of tumor necrosis factor(TNF), interleukin-8(IL-8), interleukin-6(IL-6) in serum were tested by ELISA. Pathological change was observed by HE staining for mouse pancreas specimens; Expression of NF-κB in pancreas was determined using Western blot analysis. Result: Compared to model control group, Niuxin persimmon polysaccharides significantly reduced the levels of blood-sugar and restored insulin levels in diabetic mice,with significant difference (P<0.05). Inflammatory cytokine levels in serum were reduced by the polysaccharides with s significant difference (P<0.05). The polysaccharides also showed protective effect against the pancreatic injury; as well as down-regulation of NF-κB protein in pancreatic tissue. Conclusion: Niuxin persimmon leaf polysaccharides can play the role of hypoglycemic activity and regulation of insulin. The mechanism may be associated with restoring islet function and inhibition of the inflammatory pathway in the pancreas.

      摘要:Objective:To study the effects of extracts from T. triquetrum on blood glucose and blood lipid and insulin(INS) in diabetic mice induced by streptozotocin. Method: T. triquetrum was extracted and separated by reflux extraction with different solvent, petroleum ether fraction, chloroform fraction, ethyl acetate fraction, n-butanol fraction and 60% ethanol fraction were obtained respectively.Type 2 diabete mice models were established by intraperitoneal injection of streptozotocin (STZ)150 mg·kg-1.The experimental mice were randomly divided into eight groups, control group, model group, metformin group(100 mg·kg-1), petroleum ether group, chloroform group, ethyl acetate group, n-butanol group and 60% ethanol group, the dose were 200 mg·kg-1. Control group and model group were given normal saline. After the experimental mice were administrated with drugs for 20 days, the fasting blood glucose was determined by method of glucose oxidase, fasting insulin was determined by the radioimmunoassay, and blood lipid were determined by chemical method. Result: Compared to model group, ethyl acetate fraction, n-butanol fraction and 60% ethanol fraction could significantly reduce fasting blood glucose level in diabetic mice induced by STZ(P<0.05 or P<0.01). Of all the fractions, 60% ethanol parts had the most significant hypoglycemic effect. hypoglycemic rate was 23.61%.It also decreased insulin resistance index (IR), contents of TG, TC and LDL-C and increased the insulin sensitivity index (ISI) and HDL-C content in the serum significantly (P<0.05 or P<0.01). Chloroform fraction and petroleum ether fraction had no significant effect on diabetic mice(P>0.05). Conclusion: The effective constituent of antidiabetics from Tadehagi triquetrum are mainly exist in the n-butanol fraction and 60% ethanol fraction, they can significantly reduce fasting blood glucose level in diabetic mice induced by STZ, improve insulin resistance and adjust metabolic disturbance.

      摘要:Objective:To study the effects of Polysaccharides from Galli Gigerii Endothelium Corneum (PECG) on blood lipid, blood glucose and immune function in diabetes mellitus rats with hyperlipidemia. Method: Wistar rats were feed with the high fat and sugar feedfor 30 d,thenthe rats was injected with streptozotocin intraperitoneally,continue to feed with high fat and sugar feed, meanwhile, administered orallywith 80,20 mg·kg-1 dosages of PECG for 40 d. Blood lipid, blood glucose and immune function in rats of different groups was observed.The index thymus and spleen were calculated. Result: The levels of total cholesterol (TC),triaglyceride (TG),low density lipoprotein cholesterol (LDL-C) and blood glucose decreased significantly (P<0.05) and those of high density lipoprotein cholesterol (HDL-C),the index thymus and spleen increased greatly(P<0.05) and SI increased obviously in rats of high dose PECG group (P<0.05). Conclusion: PECG can decrease the level of fasting blood lipid and blood glucose in diabetes mellitus rats with hyperlipidemiaand improved cellular immune function significantly.

      摘要:Objective:The present study was designed to observe the effect of Paeornla Licore Tang (PR/L) and its disassembled formula Shaoyao (PR),Ganchao (L) on damage of rat IEC-6 cells induced by H2O2 , and also to investigate the potential mechanism. Method: IEC-6 cells were cultured with blank control medium, PR/L, PR, or L of the fixed dosages previously for 24 h, and then stimulated with 200 μmol·L-1 H2O2 for 2 h. Cell survival was measured by MTT assay. Cell apoptosis was evaluated by using flow cytometry with Annexin V / PI staining. Expression of Caspase-3 mRNA was detected by RT-qPCR. Result: Compared with the controls, the H2O2 model showed decrease in survival and expression of Caspase-3 mRNA, and increase in apoptosis. Compared with the H2O2 model, after treatment of PR 200 μg·mL-1 or 100 μg·mL-1, Gancao of 100 μg·mL-1 or 50 μg·mL-1, or both PR and L for 24 h, cell damage and cell apoptosis were reduced significantly, and expression of Caspase-3 mRNA and protein was both increased. Conclusion: Shaoyao,Gancao, and Shaoyao Ganchao Tang can protect IEC-6 cells from damage by H2O2, which may be due to the decrease in Caspase-3 expression.

      摘要:Objective:To study the effect and pharmacological mechanism of Morus alba for trating non-insulin-dependent diabetes mellitus. Method: The KKAy mice were randomly divided into 5 groups: model group, Morus alba high, middle and low dose groups (3, 1.5, 0.75 g·kg-1) and metformin group (0.4 g·kg-1). Model group and control group animals were administrated with the same volume of tap water. Four weeks later, the contents of fasting plasma glucose (FPG), fasting plasma insulin (FIns) were observed, and the insulin sensitivity of mice was analyzed. Total RNA was extracted from island cells and the content of TLR2 and TLR4 was evaluated with Real time-PCR method. Result: Morus alba of different doses could obviously decrease the amount of FPG(P<0.05 or P<0.01), decrease the content of insulin, reinforce the sensitivity of rat to exogenous insulin (P<0.05 or P<0.01) and decrease the expression of TLR2, TLR4 of KKAy mice islet cells. Conclusion: Morus alba had significant effect to decrease FPG and improve the sensitivity to exogenous insulin, its effects may be related to the decrease in expression of TLR2, TLR4 and inhibition of inflammation.

      摘要:Objective:To investigate the anti-ulcer recurrence mechanism of Kuiyangping through establishing experimental chronic ulcer model. Method: Okeba modified method was used to establish gastric ulcer model in rats by administering intragastrically with aspirin once a day. Kuiyangping of 5.60 g·kg-1 was administrated for 14 days. The RT-PCR method was used to detect the expressions of vascular endothelial growth factor(VEGF) and its receptor Flt-1 mRNA in the gastric mucosa surrounding ulcer. Result: Kuiyangping could increase the expression of VEGF and Flt-1 mRNA in the gastric mucosa surrounding ulcer (compared with recurrence group, P<0.01). Conclusion: Kuiyangping could decrease the gastric ulcer recurrence induced by aspirin. The effective mechanisms might be related to the enhancement of the expression of VEGF and Flt-1 mRNA, the increase in microvascular permeability, and the promotion of vasculogenesis in the gastric mucosa surrounding ulcer.

      摘要:Objective:To research the effect of polysaccharides of Radix Rosa laevigata and Caulis Rosa laevigata on intestinal flora of mouse. Method: Intestinal microbe dysbiosis in mice were constructed by intragastric administration of lincomycin (15 g·kg-1,bid, 3 d). The mice were given polysaccharides of Radix Rosa laevigata and Caulis Rosa laevigata (100 g·kg-1 of crude drug for 6 days,) and then normal intestinal flora of fresh fecal was taken on the 10th day to observe the changes in normal intestinal microflora in the first 10 days. The result was expressed as lg10n·g-1.The nomal control, positive control and spontaneous recovery groups were set up. Result: Intestinal microbe dysbiosis in mice was successfully established. After the intestinal microbial flora in mice was disturbed and then treatment by polysaccharides of Radix Rosa laevigata and Caulis Rosa laevigata for 6 days, alteration of intestinal flora in polysaccharides of Radix Rosa laevigata was basically regulated. For polysaccharides of Caulis Rosae laevigatae, comparing with the modle groups, only the quantity of Enterobacteriaceae and Bifidobacterrium was increased significantly (P<0.01). Conclusion: The polysaccharides of Radix Rosa laevigata and Caulis Rosa laevigata have the function of regulating alteration of intestinal flora. Comparing with Caulis Rosae laevigatae, Radix Rosa laevigata has the better curative effect.

      摘要:Objective:To investigate the differences in nature of the properties of Radix Curcumae and Rhizoma Curcumas Longae,we compared their pharmacological activities. Method: The rat model of cold-damp jaundice was established by gavaging with phenyl isothiocyanate to induce liver injury combined with rhubarb feeding and cold-damp environmental stress. The aqueous extracts of Rhizoma Curcumas Longae and Radix Curcumae were gavaged both at a dose of 10 g·kg-1 for 16 days. The jaundice index including aspartate aminotransferase and alanine amiotransferase and total bilirubin in serum, the antioxidant indexes including superoxide dismutase and malondialdehyde in hepatic mitochondria, the energy metabolism indexes including succinate dehydrogenase and sodium-potassium-adenosine triphosphate and calcium-magnesium-adenosine triphosphate in hepatic mitochondria, redox capacity of hepatic mitochondria, the pituitary-adrenal, pituitary-thyroid, pituitary-gonadal function indexes including adrenocorticotropin and luteinizing hormone and thyroid stimulating hormone in serum and liver pathological changes were observed. Result: In jaundice effect and pathological changes including portal area bile duct pathological change and mesenchymal inflammatory cell infiltration and liver parenchyma pathological change, the aqueous extract of Rhizoma Curcumas Longae was better than the aqueous extract of Radix Curcumae. In cold-damp jaundice model group, hepatic mitochondria energy metabolism and redox capacity and superoxide dismutase and the pituitary-adrenal, pituitary-thyroid, pituitary-gonadal function were low. The aqueous extract of Rhizoma Curcumas Longae could enhance those function and the aqueous extract of Radix Curcumae was not obvious. Conclusion: The aqueous extract of Radix Curcumae showed cold in nature and the aqueous extract of Rhizoma Curcumas Longae showed warm in nature.

      摘要:Objective:To study the therapeutic effect of Baiyu Lianyu mixture (BYLY) on diabetes. Method: The mice model of diabetes was induced by intravenous injection of streptozotocin.The diabetes mice were divided into model group,BYLY high dose group (6.0 g·kg-1), BYLY middle dose group(3.0 g·kg-1),BYLY low dose group(1.5 g·kg-1,ig) and phenformin group. The drugs were given orally. Blood sugar,glucose tolerance and oxygen-derived free radicals were observed. Result: Compared to the model group,the concentration of fasting blood sugar of the BYLY groups was decreased obviously (P<0.05,P<0.01), the glucose tolerance increased obviously (P<0.05,P<0.01).The activity of superoxide dismutase (SOD) and glutathione peroxidase(GSH-Px) increased (P<0.01), malondialdehyde (MDA) decreased obviously (P<0.05,P<0.01). Conclusion: BYLY shows the effects of decreasing blood sugar,raising glucose tolerance and improving metabolism turbulence of oxygen-derived free radicals on diabetes.

      摘要:Objective:To observe theinfluence of the joint medication of Ampelopsis japonica root and Aconitum carmichaelii root on the function and pathological morphology of the heart, liver and kidney in rats. Method: Rats were administrated with water decoction of A. japonica and A. carmichaelii for successive 14 days. At thirty minutes after the last administration, the serum levels of total serum protein(TP), alburnin(Alb), alanine aminotransferase(ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN), creatinine (Cr)and lactate dehydrogenase (LDH) were measured. Autopsy was conducted after the death of the rats and the heart, liver, kidney were harvested for pathological examination.The index of viscera was calculated. Result: The levelof ALT and AST (P<0.05)was increased in the A. japonica group; the level of AST (P<0.05) was increased in the aconitum group. When treated with A. japonica and A. carmichaelii jointly, the level of ALT,AST and LDH (P<0.01 or P<0.05)all increased. The examination of organ index showed that the liver index increased(P<0.05) in A. japonica groupo and all the indexes (P<0.01 or P<0.05)of liver and kidney increased when A. japonica and aconitum A. carmichaelii were administrated jointly. Autopsy and histopathological examination indicated that the liver and kidney of the rats changed significantly when A. japonica and A. carmichaelii were administrated jointly. Conclusion: The toxicity of the joint medication of A. carmichaelii japonica and A. carmichaelii in rats would be strengthened.

      摘要:Objective:To observe the therapeutic effects of Guizhi Fuling decoction(GFD) and Huanglian Jiedu decoction(HJD) on patients with unstable angina(UA). Method: Seventy-two UA patients were randomly assigned to the treatment group(36 cases) and the control(36 cases). All were treated by the regular UA treatment, but to patients in the treatment group, GFD and HJD was given additionally. The course of treatment was three weeks. Before and after the treatments, changes of clinical symptoms, 24-hour ambulatory electrocardiogram(AECG), blood lipids and serum levels of high-sensitivity C-reactive protein(hs-CRP) were observed. Result: Compared with before treatment, obvious improvement of clinical symptom and experiment index was obtained in two groups. After treatment, the frequency of angina attacks, myocardial ischemia times in 24 h, the longest duration of a single myocardial ischemia, the total duration of myocardial ischemia in 24 h and hs-CRP were reduced and high density lipoprotein cholesterol(HDL-C) was increased further in the treatment group(P<0.05). Conclusion: GFD and HJD showing definite effects on UA.

      摘要:Objective:To study clinical curative effect of Maren Runchang pill combined with catgut implantation at acupoint for treatment of chronic functional constipation (FC) in patients. Method: One hundred and ten cases of FC were divided into the treatment group and control group (n=55 each). The treatment group was applied with Maren Runchang pill (1-2 g, bid, orally)combined with catgut implantation at acupoint for 6 weeks, and the control group was treated by Maren Runchang pill alone. Difficulty of defecate, Bristol score, defecate time, stool frequency and abdominal distention and other symptoms were evaluated. Result: After treatment, the two groups showed decreases in symptom scores significantly (P<0.01). The treatment group symptom scores were lower than that of the control group (P<0.01). The total effectiveness in the treatment group was 98.18%, better than that of 85.45% in the control group (P<0.05). Conclusion: Maren Runchang pill combined with catgut implantation at acupoint for treatment of FC can reduce the symptom scores, improve the clinical effect. The therapy is worthwhile to be applied clinically.

      摘要:Objective:To observe the clinical efficacy of Zishen-Shugan-Ningxin prescription combind with acupuncture to treat menopausal depression, in order to provide more safe and effective method for the treatment of menopausal depression. Method: Choosing 96 patients with depression from February 2010 to June 2011 in our hospital as the samples and divided them into three groups, they were given the Zishen-Shugan-Ningxin, acupuncture,and combination treatment. Then using the Hamilton Depression Rating Scale (HAMD) and menopausal index (KMI) scale to evaluate the therapeutic effect of the three groups. Result: After the treatment of 2,4,8 week, the difference of HAMD score were significant, prescription + acupuncture group was significantly lower than the other two groups; but there were no significance between simple prescription group and acupuncture group (81.25% vs 78.13% after treatment. (χ2=0.097, P= 0.527), while the total effective rate of prescription + acupuncture treatment was 96.88 percent, what were significant with prescription group and acupuncture group, respectively (χ2=4.010, P= 0.046,χ2=5.143, P=0.046). Conclusion: Using Zishen-Shugan-Ningxin prescription with acupuncture to treat menopausal depression were significantly of the effect, not only can effectively prevent a variety of side effects of western medicine, but the results are stable and less recurrence, the patient can easy to accept this treatment, and it is indeed a safe and effective treatment.

      摘要:Objective:To observe clinical efficacy and adverse reaction of Cornu Antelopes capsule as adjunctive therapy for partial seizures. Method: The treatment period was six months,89 patients with partial Seizures were chosen that were divided into refractory partial epilepsy (35)and not refractory partial epilepsy(54). Result: Cornu Antelopes capsule produced statistically significant median reduction in seizure frequency as compared with the pre-cornu antelopes capsule period (3 months baseline), the efficiency was 53.9% for total seizure counts. The efficiency and control rate is 78.8% and 21.2% for not refractory partial epilepsy compared to 18.9% and 0 for refractory partial epilepsy. Conclusion: Cornu Antelopes capsule adjunctive therapy for Partial epilepsy is an effective and safe treatment.

      摘要:Objective:To explore the effects of Xuebijing injection on blood gas analysis and cardiac function in patients with acute exacerbation of chronic pulmonary and heart disease. Method: Seventy-six cases with acute exacerbation of chronic pulmonary and heart disease were randomly divided into control group and treatment group. Each group contained 38 cases. Patients in control group were received with conventional therapy, while the cases in treatment group were given with conventional therapy plus Xuebijing injection for iv infusion (50 mL, bid) for four weeks. Result: The levels of PaO2, PaCO2, hemorheological parameters, mean pulmonary artery pressure and cardiac output were obviously improved, compared with those of control group after the treatment (P<0.05). The effective rate of treatment group was 86.84%, which was higher than that of control group (60.53%, P<0.05). Conclusion: Treatment with Xuebijing injection could improve the blood gas analysis and cardiac function in patients with acute exacerbation of chronic pulmonary and heart disease.

      摘要:Objective:To investigate the clinical efficacy of the integrative Chinese and western medicine treatment for acute exacerbations of chronic obstructive pulmonary disease (AECOPD) with symptom of mental confusion due to phlegm. Method: Sixty cases of AECOPD with symptom of mental confusion due to phlegm were randomly divided into observation group and control group (n=30, each). The control group was only received norms Western medicine treatment, the observation group on the basis of the control group was given Ditan decoction, one dose each day. The clinical efficacy was evaluated after 10 days of continuous treatment. Result: After treatment, the total effective rate in observation group was 83.3%, better than 60% of the control group (P<0.05); blood SaO2, PaO2 and PaCO2, blood gas analysis in the observation group were better than those in the control group (P<0.05 or P <0.01). Conclusion: Clinical efficacy of the integrative Chinese and western medicine treatment for AECOPD is confirmed.

      摘要:Objective:To observe clinical efficacy of Mailuoning oral liquid combined with rehabilitation training for patients with limb dysfunction after stroke. Method: One hundred and one patients with limb dysfunction after cerebral apoplexy were randomly divided into treatment group (n=52) and control group (n=49). The control group was adopted to Bobath rehabilitation therapy, and based on the rehabilitation the treatment group was combined with Mailuoning oral liquid,20 mL, tid. Both groups were treated for 8 weeks. According to the modified Barthel index scale(BI), activities of daily living (ADL) were evaluated. Fugl-Meyer movement scale evaluation was carried out for lower extremity function. Breg balance scale (BBS) assessment was used for balance ability. Holden walking functional rating scale (FAC) assessment was selected for walking ability. Result: The two groups could significantly increase the BI, BBS, Fugl-Meyer and FAC score, and the treatment group was better than the control group (P<0.01). For improving the ability of independent walking and reducing the disability rate, the treatment was superior to the control group (P<0.05). Conclusion: Mailuoning oral liquid combined with rehabilitation training can improve ADL for patients with limb dysfunction after stroke.

      摘要:Objective:To observe the clinical efficacy of Fuzheng Xiao'ai decoction (FX) treatment for esophageal cancer after chemotherapy. Method: Seventy-six cases of esophageal cancer after surgery were randomly divided into treatment and control groups. Control group of 37 cases was treated with 5-fluorouracil (5-FU), cisplatin (DDP), and with the necessary symptomatic treatment. On the treatment basis of the control group, the treatment group (n=39) was postoperatively started FX for 12 weeks. The adverse reactions, KPS score, and serum immunoglobulin were observed. Result: The incidences of adverse reactions in treatment group were lower than those in the control group (P<0.05, P<0.01) including the delayed gastric emptying, diarrhea, loss of appetite and the weight loss> 5%. After 1 month of treatment, at the end of chemotherapy and two weeks after the end of chemotherapy, KPS score were higher in the treatment group (P<0.01). In the treatment group, IgG, IgA, and IgM levels were higher than those in the controls by the end of the treatment (P<0.05, P<0.01). However, the total effective rates in the treatment group and the control group were 79.48% and 70.27% accordingly. Conclusion: FX can reduce the adverse reactions caused by esophageal cancer surgery and chemotherapy to improve patients' quality of life, improve immune function.

      摘要:The complex reaction processes of traditional Chinese medicine(TCM) applications restrict the exhaustive analysis about mechanism of detoxicity by compatibility, which is the key to solve the problems of TCM toxicity. Efficacy and toxicity of drug determines the characteristics of toxic herbs with 'Efficacy-toxicity intersection’. With network pharmacology appeared, new methods on research about discovering toxic components in TCM, toxic mechanism and reducing toxicity will be provided. Through reviewing the practice of TCM toxicity, the article will discuss the application strategy of network pharmacology used in the research on TCM systematically and its possibility (Example: Aconiti Lateralis Radix).

      摘要:To summarize the research progress on chemical components and pharmacological actions of Clerodendron trichotomum and provide useful references for the further research.All papers about chemical components and pharmacological actions in CNKI, MEDLINE and CA databases etc were referred to and summarized. The chemical components isolated from C. trichotomum. include mainly essential oil, flavone, phenylpropanoids, glucoside and alkaloids; Pharmacological references show that it has depressurization, anti-inflammation, anti-oxidation, analgesia, mitigate, anti-cell multiplication and anti-HIV. The studies about big heteropolarity components which related with chemical and pharmacological have been reported more, however, the moderate and small heteropolarity components were researched lessly, we should carry out further studies on moderate and small heteropolarity components, to establish foundation for C. trichotomum’quality criteria and deep development.

      摘要:This paper is aimed to summarize the anti-cancer effects and mechanism of Juglans mandshurica. Nearly 40 documents, which were contributed by both domestic and foreign researchers, where found in the databasses sush as cqvip, sciFinder and other ones. The anti-tumor effect of the J. mandshurica can be summarized to the following 6 aspects: affecting cell cycle, inhibiting of DNA topoisomerase, inducing apoptosis of tumor cells, improving the immune system, damaging the mitochondrial structure, changing the structure and biochemical of cell membrane. J. mandshurica. has an bright future as a anti-cancer drug and it is worthy further studies to obtain a more efficient product with lower toxicity.

In this paper, the research of cerebrosides in traditional Chinese midicine and it’s pharmacological activity in recent ten years has been summarized. The review has provided some information for the study, development and utilization of cerebrosides in future.

But on the basis of effect and production place of wild or planted ‘Pingzhu’in Chengkou county, it is similar with Rhizoma atractylodis. Conclusion: ‘Pingzhu’produced in Chengkou county is Atractylodes lancea(Thunb.)DC. in essence.

      摘要:As a famous traditional Chinese medicine, Angong Niuhuang Wan is used for treatment of various diseases, such as encephalitis, apoplexy, cerebral hemorrhage, ischemic cerebral injury and so on. It also has unique effect on coma, hyperpyrexia induced by cerebral diseases. Pharmacology studies on Angong Niuhuang Wan reveal its versatile activities on encephalopathy, like cerebral hemorrhage, ischemia injury, sepsis and so on in animals. The safe application of Angong Niuhuang Wan appears to be utmost important due to the contained realgar (As) and cinnabar (Hg). This article describes the progress in studies of Angong Niuhuang Wan, including pharmacologic actions and clinical applications, to provide a new thinking and reference to the further utilization of Angong Niuhuang Wan.

The author found it is more cormon in the gardenoside name mix,combine to the Commonly used in traditional Chinese medicine effective ingredient nomenclature, put forward ‘zhizigan' to gardenoside is to provide a reference for the for scholar’s further exchanges.
